Twelve compounds were isolated from the ethyl acetate fraction of the 80% aqueous ethanol extract of the roots and stems of Dalbergia rimosa Roxb. by silica gel, MCI, Sephadex LH-20 column chromatography, and semi-preparative HPLC. Their structures were identified by spectral analysis such as UV, IR, MS, 1D/2D NMR and by comparison with literature information as dalbergiquinol A (1), dalbergiquinol B (2), R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol (3), neokhriol A (4), mucronulatol (5), (3R)-7,2′,3′-trihydroxy-4′-methoxy-isoflavane (6), isomucronulatol (7), (3S)-violanone (8), 3′-O-methylviolanone (9), eryvarin M (10), (±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone (11) and (-)-butin (12). Compound 1 and 2 are new compounds, and compounds 3-12 were isolated from this plant for the first time. Compounds 1, 2, 4, 6, 8, 11, 12 showed good scavenging effect on DPPH free radical.

运用硅胶、MCI、Sephadex LH-20等柱色谱, 结合半制备液相分离技术对多裂黄檀80%乙醇提取物的乙酸乙酯部位分离纯化得到12个化合物。通过UV、IR、MS、1D/2D NMR等波谱数据与结合文献确定化合物的结构, 分别鉴定为多裂黄檀酚A (1)、多裂黄檀酚B (2)、R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol (3)、neokhriol A (4)、mucronulatol (5)、(3R)-7,2′,3′-三羟基-4′-甲氧基异黄烷(6)、异短尖剑豆酚(7)、(3S)-violanone (8)、3′-O-methylviolanone (9)、eryvarin M (10)、(±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone (11) 和(-)-butin (12)。其中, 化合物12为新化合物, 化合物3~12为首次从该植物中分离得到。化合物124681112对DPPH自由基有良好清除作用。

运用硅胶、MCI、Sephadex LH-20等柱色谱, 结合半制备液相分离技术对多裂黄檀80%乙醇提取物的乙酸乙酯部位分离纯化得到12个化合物。通过UV、IR、MS、1D/2D NMR等波谱数据与结合文献确定化合物的结构, 分别鉴定为多裂黄檀酚A (1)、多裂黄檀酚B (2)、R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol (3)、neokhriol A (4)、mucronulatol (5)、(3R)-7,2′,3′-三羟基-4′-甲氧基异黄烷(6)、异短尖剑豆酚(7)、(3S)-violanone (8)、3′-O-methylviolanone (9)、eryvarin M (10)、(±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone (11) 和(-)-butin (12)。其中, 化合物12为新化合物, 化合物3~12为首次从该植物中分离得到。化合物124681112对DPPH自由基有良好清除作用。

Twelve compounds were isolated from the ethyl acetate fraction of the 80% aqueous ethanol extract of the roots and stems of Dalbergia rimosa Roxb. by silica gel, MCI, Sephadex LH-20 column chromatography, and semi-preparative HPLC. Their structures were identified by spectral analysis such as UV, IR, MS, 1D/2D NMR and by comparison with literature information as dalbergiquinol A (1), dalbergiquinol B (2), R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol (3), neokhriol A (4), mucronulatol (5), (3R)-7,2′,3′-trihydroxy-4′-methoxy-isoflavane (6), isomucronulatol (7), (3S)-violanone (8), 3′-O-methylviolanone (9), eryvarin M (10), (±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone (11) and (-)-butin (12). Compound 1 and 2 are new compounds, and compounds 3-12 were isolated from this plant for the first time. Compounds 1, 2, 4, 6, 8, 11, 12 showed good scavenging effect on DPPH free radical.

多裂黄檀(Dalbergia rimosa Roxb.) 来源于豆科黄檀属植物多裂黄檀的干燥根茎, 分布于广西各地, 壮药名为Gengpeng (更彭), Meikouwai (美扣外), 其性味辣, 热, 具有调龙路, 除湿毒, 消肿痛的作用, 可用于治疗巧尹(头痛), 叶用于呗农(痈疮), 呗叮(疗疮)[1, 2]。目前, 国内外对多裂黄檀在木材方面研究较多, 但对其化学成分和药理作用方面的研究鲜见报道。有研究发现多裂黄檀与降香黄檀具有很高的亲缘关系[3], 二者挥发性成分相似度极高[4], 且降香黄檀的有效成分主要为挥发油和酚性成分[5]。课题组前期研究发现, 多裂黄檀中含有黄酮类、三萜类和木脂素类等成分, 在活性筛选中, 发现乙酸乙酯部位具有很好的抗氧化作用, 为了阐明多裂黄檀的药效物质基础, 寻找具有生物活性的先导化合物, 本实验综合运用各种现代色谱和光谱技术, 从多裂黄檀乙酸乙酯部位中分离纯化得到12个化合物, 分别鉴定为: (7S)-4,3′,4′-trihydroxy-2,5-dimethoxydalbergiquinol (1)、(7S)-5, 3′, 4′-trihydroxy-2, 4-dimethoxydalbergiquinol (2)、R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol (3)、neokhriol A (4)、mucronulatol (5)、(3R)-7,2′,3′-三羟基-4′-甲氧基异黄烷(6)、异短尖剑豆酚(7)、(3S)-violanone (8)、3′-O-methylviolanone (9)、eryvarin M (10)、(±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone (11)、(-)-butin (12), 见图 1。其中化合物12为新化合物, 分别命名为多裂黄檀酚A和多裂黄檀酚B, 化合物3~12为首次从该植物中分离得到。化合物124681112对DPPH自由基有良好清除作用。
化合物1  黄色油状物, 易溶于甲醇, 丙酮。HR-ESI-MS给出准分子离子峰m/z: 301.107 6 [M-H]- (Calcd for C17H17O5 301.107 6), 推测该化合物的分子式为C17H18O5, 计算其不饱和度为9。化合物1 1H NMR谱(表 1) 中, 芳香区出现5个氢质子信号: δH 6.65 (1H, d, J = 8.1 Hz), 6.57 (1H, d, J = 2.1 Hz), 6.46 (1H, dd, J = 8.1, 2.1 Hz), 6.63 (1H, s) 和6.48 (1H, s), 提示结构中存在一个1, 3, 4-三取代苯环和一个1, 2,4,5-四取代苯环, 通过HSQC谱找到它们相应的碳信号分别出现在: δC 115.4, 116.9, 120.9, 115.2和101.7; δH 6.19 (1H, ddd, J = 17.0, 10.2, 6.7 Hz), 5.09 (1H, dt, J = 10.2, 1.7 Hz), 4.84~4.80 (1H, m) 和4.87 (1H, d, J = 6.7 Hz) 提示分子中存在一个烯丙基片段, 利用HSQC谱找到它们相应的碳信号分别出现在: δC 142.7, 115.9和47.8, 通过 1H-1H COSY谱确定H-8/H-9相关进一步证实了烯丙基片段的存在。高场区出现2个甲氧基氢质子信号: δH 3.70 (3H, s) 和3.65 (3H, s), 利用HSQC谱找到它们相应的碳信号分别出现在: δC 57.4和56.7。化合物1 13C NMR谱(表 1) 中显示了17个碳信号, 其中包含7个季碳信号、7个次甲基信号、1个亚甲基碳信号和2个甲氧基碳信号。HMBC谱(图 2) 中δH 4.87和δC 124.1、153.1、115.9、142.7、136.6、120.9相关, 确定两个苯环通过烯丙基连接; δH 3.65和δC 153.1、δH 3.70和δC 142.6相关, 确定C-2、C-5分别有甲氧基取代。测得比旋光度为[α]$ {}_{\mathbf{D}}^{20} $ +22.4 (c 0.05, MeOH), 通过TDDFT ECD计算与实验ECD对比(图 3) 确定的其绝对构型为7S。综合以上信息, 确定化合物1的结构为(7S)-4,3′,4′-trihydroxy-2,5-dimethoxydalbergiquinol, 命名为多裂黄檀酚A (dalbergiquinol A)。
化合物2  黄色油状物, 易溶于甲醇, 丙酮。HR-ESI-MS给出准分子离子峰m/z 301.108 1 [M-H]- (Calcd for C17H17O5 301.107 6), 推测该化合物分子式为C17H18O5, 计算其不饱和度为9。化合物2 1H NMR谱(表 1) 中, 芳香区出现5个氢质子信号: δH 6.67 (1H, d, J = 8.1 Hz), 6.61 (1H, d, J = 2.0 Hz), 6.59 (1H, s), 6.49 (1H, dd, J = 8.1, 2.0 Hz), 6.45 (1H, s), 提示结构中存在一个1, 3, 4-三取代苯环和一个1, 2,4,5-四取代苯环, 利用HSQC谱找到它们相应的碳信号分别出现在: δC 116.0, 116.9, 121.0, 116.0和102.1; δH 6.20 (1H, ddd, J = 16.9, 10.2, 6.5 Hz), 5.10 (1H, dt, J = 10.2, 1.6 Hz), 4.87~4.83 (1H, m) 和4.91 (1H, d, J = 6.5 Hz) 提示分子中存在一个烯丙基片段, 利用HSQC谱找到它们相应的碳信号分别出现在: δC 142.5, 115.5, 47.7。高场区出现2个甲氧基氢质子信号: δH 3.75 (3H, s) 和3.66 (3H, s), 利用HSQC谱找到它们相应的碳信号分别出现在: δC 56.5和57.7。化合物2 13C NMR谱(表 1) 中显示了17个碳信号, 其中包含7个季碳信号、7个次甲基碳信号、1个亚甲基碳信号和2个甲氧基碳信号。通过HMBC谱(图 4) 可知, δH 4.91和δC 123.0、149.7、116.0、136.5、121.0相关, 确定两个苯环通过烯丙基连接, δH 3.75和δC 149.7相关、δH 3.66和δC 143.4相关, 确定甲氧基取代在C-2、C-4位置上。测得比旋光度为[α]$ {}_{\mathbf{D}}^{20} $ -26.8 (c 0.05, MeOH), 通过TDDFT ECD计算与实验ECD对比(图 3) 确定的其绝对构型为7S。综合以上信息, 确定化合物2的结构为(7S)-5, 3′, 4′-trihydroxy-2, 4-dimethoxydalbergiquinol, 命名为多裂黄檀酚B (dalbergiquinol B)。
以化合物1~12对DPPH自由基的清除能力对其抗氧化活性进行评价。结果显示, 部分化合物显示出良好的抗氧化活性(表 2), 可为多裂黄檀的进一步开发利用提供参考依据。
Agilent G6230型飞行时间质谱仪(美国Agilent公司); 布鲁克AVANCE Ⅲ 500 MHz型核磁共振波谱仪(德国Bruker公司); 赛默飞世尔NICOLET iS10型中红外光谱仪、Hypersil GOLDTM C18柱(250 mm × 10 mm, 5 μm) (美国赛默飞世尔公司); 岛津LC-20AR、岛津LC-2030 PLUS型高效液相色谱仪、岛津UV2401PC型紫外可见分光光度仪(日本岛津公司); Autopol Ⅵ旋光仪(美国鲁道夫公司); V100圆二色光谱仪(英国应用光物理公司); BP211D型电子分析天平(德国赛多利斯公司); Multiskan Sky 1530型全波长酶标仪(美国Thermo Fisher Scientific公司); 旋转蒸发仪(EYELA OSB-2100、OSB-2200, 上海爱朗仪器有限公司); ZF-6型三用紫外分析仪(上海嘉鹏科技有限公司); CHP P20小孔树脂(日本三菱化学公司); LH-20羟丙基葡聚糖凝胶(Sephadex LH-20, 美国Pharmacia公司); 柱色谱硅胶(100~200目, 硅胶H, 国药集团化学试剂有限公司); 硅胶薄层板(50 mm × 100 mm等, 青岛海洋化工有限公司); 甲醇、乙腈(色谱纯, Fisher科技有限公司); L-抗坏血酸(Vitc) (纯度 > 99%, 上海麦克林生化科技有限公司); 1, 1-二苯基-2-三硝基苯(DPPH, 纯度 > 98%, 美国Sigma公司); 所用常规试剂均为分析纯(四川西陇科学有限公司)。
多裂黄檀38.0 kg, 以10倍量80%乙醇回流3次(2、1.5、1 h), 减压回收溶剂得总浸膏(1.84 kg)。取总浸膏(1.64 kg), 加适量温水悬浮后, 依次用石油醚(60~90 ℃)、乙酸乙酯、正丁醇萃取, 减压回收溶剂后得到石油醚部位浸膏(36.0 g), 乙酸乙酯部位浸膏(179.4 g), 正丁醇部位浸膏(504.9 g) 和水部位浸膏(873.0 g)。
乙酸乙酯部位浸膏145.0 g, 经硅胶柱色谱分离, 以石油醚-乙酸乙酯(100∶0、20∶1、10∶1、5∶1、2∶1、0∶1) 梯度洗脱, 得7个组分(Fr.1~Fr.7)。Fr.4 (7.5 g) 经MCI柱色谱(甲醇-水, 30∶70~100∶0) 梯度洗脱, 得9个组分Fr.4-1~Fr.4-9。Fr.4-4 (2.8 g) 以硅胶H柱色谱(氯仿-丙酮, 100∶0~5∶1) 梯度洗脱得5个组分Fr.4-4-1~Fr.4-4-5。Fr.4-4-1 (0.3 g) 经半制备高效液相(甲醇-水, 50∶50) 纯化得化合物3 (tR = 50.5 min; 29.2 mg); Fr.4-4-3 (1.0 g) 经半制备高效液相(甲醇-水, 48∶52) 纯化得化合物7 (tR = 28.3 min; 8.8 mg)。Fr.5 (21.0 g) 经MCI柱(甲醇-水, 50∶50~100∶0) 梯度洗脱, 合并得15个组分Fr.5-1~Fr.5-15。Fr.5-6 (0.6 g) 经硅胶H柱色谱(氯仿-丙酮, 50∶1~4∶1) 梯度洗脱得5个组分Fr.5-6-1~Fr.5-6-5。Fr.5-6-4经Sephadex LH-20 (氯仿-甲醇, 1∶1) 柱色谱分别得6个组分Fr.5-6-4-1~Fr.5-7-4-6。其中Fr.5-6-4-3经半制备高效液相(甲醇-0.1%乙酸, 42∶58) 分离纯化得化合物4 (tR = 40.7 min; 8.6 mg) 及半制备高效液相(乙腈-0.1%乙酸, 23∶77) 二次纯化得化合物10 (tR = 38.8 min; 2.0 mg), 化合物8 (tR = 42.7 min; 12.4 mg); Fr.5-6-4-4经半制备高效液相(乙腈-水, 25∶75) 纯化得化合物1 (tR = 48.4 min; 20.2 mg); Fr.5-6-4-5经半制备高效液相(甲醇-0.1%乙酸, 42∶58) 分离纯化得化合物6 (tR = 31.2 min; 18.6 mg)。Fr.5-7 (1.0 g) 析出白色针状结晶, 经半制备高效液相(甲醇-水, 53∶47) 纯化得化合物5 (tR = 15.3 min; 25.6 mg)。Fr.5-8 (2.0 g) 经硅胶H柱色谱(石油醚-乙酸乙酯, 3∶1~2∶3) 梯度洗脱, 依次合并得7个组分Fr.5-8-1~Fr.5-8-7。Fr.5-8-1经半制备高效液相(甲醇-水, 48∶52) 纯化得化合物9 (tR = 33.6 min; 26.0 mg)。Fr.6 (28.5 g) 经MCI柱色谱(甲醇-水, 30∶70~100∶0) 梯度洗脱, 合并得12个组分Fr.6-1~Fr.6-12。Fr.6-6经硅胶H柱色谱(氯仿-甲醇, 50∶1~15∶1) 梯度洗脱得9个组分Fr.6-6-1~Fr.6-6-9。Fr.6-6-5 (0.05 g) 经半制备高效液相(乙腈-水, 24∶76) 分离纯化得化合物2 (tR = 31.6 min; 16.7 mg)。Fr.6-6-9 (0.3 g) 经Sephadex LH-20 (氯仿-甲醇, 1∶1) 以及半制备高效液相(甲醇- 0.1%乙酸, 30∶70) 分离纯化得化合物11 (tR = 19.8 min; 1.0 mg), 化合物12 (tR = 30.3 min; 14.6 mg)。
参考文献[6]的方法并稍作修改, 以甲醇溶解DPPH, 配制成0.1 mmol·L-1 DPPH贮备液, 避光冷藏备用。准确称取化合物溶解于DMSO中, 稀释成系列质量浓度备用。移取50 μL一系列浓度化合物溶液和阳性对照溶液加至96孔板中, 再向各孔中加入150 μL DPPH贮备液, 混匀, 室温黑暗中放置30 min, 反应完全后在波长517 nm处测定其吸光度A1。以不加样品的相同溶剂为空白对照A0, L-抗坏血酸作为阳性对照, 每个浓度平行测定3次后取平均值, 按清除率% = (A0-A1) / A0×100%计算样品对DPPH自由基清除率, 用半数抑制浓度(IC50) 表示化合物对DPPH自由基的清除能力。

化合物3  白色粉末, ESI-MS m/z 299 [M-H]-, 分子式为C18H20O4。该化合物的1H NMR (500 MHz, acetone-d6) 和13C NMR (125 MHz, acetone-d6) 数据与文献[7]R-(-)-3′-hydroxy-2,4,5-trimethoxydalbergiquinol基本一致。
化合物4  黄色油状(甲醇), ESI-MS m/z 285 [M-H]-, 分子式为C17H18O4。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[8]的neokhriol A基本一致。
化合物5  白色针状结晶(丙酮), ESI-MS m/z 301 [M-H]-, 分子式为C17H18O5。该化合物的1H NMR (500 MHz, acetone-d6) 和13C NMR (125 MHz, acetone-d6) 数据与文献[9]的mucronulatol基本一致。
化合物6  黄色油状(甲醇), ESI-MS m/z 287 [M-H]-, 分子式为C16H16O5。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[8, 10]的(3R)-7,2′,3′-三羟基-4′-甲氧基异黄烷基本一致。
化合物7  淡黄色粉末, ESI-MS m/z 301 [M-H]-, 分子式为C17H18O5。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[11]的异短尖剑豆酚基本一致。
化合物8  白色粉末, ESI-MS m/z 315 [M-H]-, 分子式为C17H16O6。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[12]的(3S)-violanone基本一致。
化合物9  白色粉末, ESI-MS m/z 329 [M-H]-, 分子式为C18H18O6。该化合物的1H NMR (500 MHz, acetone-d6) 和13C NMR (125 MHz, acetone-d6) 数据与文献[13]的3′-O-methylviolanone基本一致。
化合物10  白色粉末, ESI-MS m/z 315 [M-H]-, 分子式为C17H16O6。该化合物的1H NMR (500 MHz, CD3OD) 和13CNMR (125 MHz, CD3OD) 数据与文献[14]的eryvarin M基本一致。
化合物11  黄色粉末, ESI-MS m/z 289 [M-H]-, 分子式为C15H14O6。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[15]的(±)-α, 3,4,2′,4′-pentahydroxydihydrochalcone基本一致。
化合物12  黄色粉末, ESI-MS m/z 271 [M-H]-, 分子式为C15H12O5。该化合物的1H NMR (500 MHz, CD3OD) 和13C NMR (125 MHz, CD3OD) 数据与文献[16]的(-)-butin基本一致。
