Three carboline fluorescent probes F1-F3 were designed and synthesized, based on lead compound JYJ-19, an antifungal compound discovered previously by our group. The antifungal activity in vitro results showed that compound F1 had moderate antifungal activity (MIC80 = 32 μg·mL-1). The stokes shift of F1 is 70 nm. The fluorescent probe F1 has good optical properties and can be used for fluorescence imaging research. Subcellular localization experiments results showed that F1 was enriched in the mitochondria of fungal cells. The detection of intracellular reactive oxygen species levels shows that JYJ-19 enhances intracellular reactive oxygen species levels. The above results indicated that carboline compounds could exert antifungal effects by acting on fungal mitochondria.

以课题组前期发现的咔啉类抗真菌化合物JYJ-19为先导化合物, 设计合成了3个新型咔啉类荧光探针F1~F3。体外抗真菌活性结果显示, 化合物F1具有中等的抗真菌活性(MIC80 = 32 μg·mL-1); 荧光探针F1的Stokes位移为70 nm, 具有良好的光学性质, 可用于荧光成像研究; 亚细胞定位实验表明, F1在真菌细胞的线粒体富集; 细胞内活性氧水平测试结果显示, 咔啉类化合物JYJ-19作用下细胞内活性氧水平升高。上述结果表明, 咔啉类化合物可能通过作用于真菌细胞的线粒体发挥抗真菌作用。

Three carboline fluorescent probes F1-F3 were designed and synthesized, based on lead compound JYJ-19, an antifungal compound discovered previously by our group. The antifungal activity in vitro results showed that compound F1 had moderate antifungal activity (MIC80 = 32 μg·mL-1). The stokes shift of F1 is 70 nm. The fluorescent probe F1 has good optical properties and can be used for fluorescence imaging research. Subcellular localization experiments results showed that F1 was enriched in the mitochondria of fungal cells. The detection of intracellular reactive oxygen species levels shows that JYJ-19 enhances intracellular reactive oxygen species levels. The above results indicated that carboline compounds could exert antifungal effects by acting on fungal mitochondria.

侵袭性真菌感染(invasive fungal infections, IFIs) 又称为深部真菌感染, 由于IFIs诊断复杂、检出率低, 发病率和死亡率高, 使其成为重症监护患者中容易被忽视的死亡原因之一[1]。全球每年的侵袭性真菌感染死亡病例中, 约80%由念珠菌、曲霉菌和隐球菌造成[2, 3]。隐球菌性脑膜炎是一种与晚期HIV相关的机会性真菌感染, 全球范围内导致HIV晚期相关死亡率的15%, 仅次于肺结核[4, 5]。隐球菌性脑膜炎通常比结核病感染更为严重, 如果不进行治疗, 其死亡率为100%, 即使采用目前推荐的治疗方法, 死亡率也常常超过40%[6]。过去的十年中, 筛查和抢先治疗早期临床感染已成为一种可行的干预措施, 有望减少隐球菌性脑膜炎对艾滋病相关死亡的影响。隐球菌抗原(CrAg) 是一种高度特异性的播散性感染生物标志物, 可在发展成脑膜炎疾病前的数周至数月内患者血液中检测到[7]
目前, 临床上只有三类药物用于治疗侵袭性隐球菌感染。5-氟胞嘧啶是第一个发现具有抗隐球菌活性的药物, 但其极易产生耐药性且具有肝毒性[8, 9]。多烯类药物两性霉素B对大多数真菌都有杀菌作用, 且较少出现耐药, 但具有严重急性肾功能衰竭、低钾血症和低镁血症等不良反应[10, 11]。两性霉素B和5-氟胞嘧啶的联合使用至今仍然是治疗隐球菌脑膜炎的金标准。三唑类抗真菌药如氟康唑、伊曲康唑、伏立康唑和泊沙康唑常用于隐球菌病的维持治疗[12]。现有药物不能满足临床需要, 亟须研发具有全新结构类型和全新作用机制的小分子抗真菌新药。
在本课题组的前期研究中, 对实验室中的化合物库进行细胞水平的筛选, 发现具有γ-咔啉骨架的化合物Hit 1对白念珠菌(Candida albicans) 和新生隐球菌(Cryptococcus neoformans) 具有中等的抗真菌活性(MIC80 C.alb = 32 μg·mL-1, MIC80 C.neo = 8 μg·mL-1), 将咔啉母核替换为四氢-γ-咔啉结构, 去掉两个手性中心, 用柔性的侧链(苯氧烷基侧链) 替换刚性和疏水较强的萘甲酰基侧链, 得到抗真菌活性提升的化合物C38 (MIC80 = 1~4 μg·mL-1), 化合物C38能够明显抑制白念珠菌生物被膜的形成, 以及酵母-菌丝态的形态学转化, 降低真菌毒力。随后对其进一步结构优化、改造得到能选择性抑制新生隐球菌生长的β-咔啉类先导化合物JYJ-19 (图 1), 其对新生隐球菌和哥特隐球菌(Cryptococcus gattii) 的MIC80为0.5~1 μg·mL-1, 对白念珠菌、光滑念珠菌(Candida glabrata)、近平滑念珠菌(Candida parapsilosis)、烟曲霉菌(Aspergillus fumigatus) 等的MIC80为1~16 μg·mL-1 (表 1), 且JYJ-19在小鼠隐球菌脑膜炎模型中表现出优秀的体内抗真菌活性, 在20 mg·kg-1的给药剂量下, JYJ-19组的小鼠脑部荷菌量CFU显著低于氟康唑组(P < 0.001)。初步的机制研究表明JYJ-19能抑制真菌生物被膜的形成、降低真菌细胞表面疏水性, 能破坏真菌细胞膜和细胞壁的形成, 可使新生隐球菌细胞周期阻滞在G2期[13, 14]。然而咔啉类化合物的抗真菌作用机制仍不明确, 还需开展进一步的研究。因此, 本研究旨在通过一种新的方法对其抗真菌的作用机制进行研究。
荧光探针是生物学研究成像的多功能工具, 具有不同的颜色组合、用于研究特定的靶位并显示活细胞中的生物学方法[15]。荧光检测可用于定量分析和定性分析标记的细胞、细菌、纳米颗粒, 还可以检测体内细菌感染[16]。例如, 抗生素类荧光探针已用于研究革兰阳性细菌的细胞分裂[17]
分子荧光探针的基本结构包括三个部分: 识别基团(receptor)、连接子(spacer) 和荧光基团(fluorophore)。识别基团一般是可以在细胞内与靶标结合发挥药效的化合物或药效团。化合物在设计成荧光探针之前, 利用药物化学骨架跃迁以及结构优化的方法, 找到先导化合物中发挥生物学活性的药效团作为识别基团。连接子即连接识别基团与荧光基团的连接链, 连接子一般是烷基碳链、氨基酸多肽链、乙二醇二缩水甘油醚等。连接子的长度通常也会影响整个荧光探针分子的生物活性。荧光基团是利用其光学性质在细胞内定位、检测、富集靶标的产生荧光的片段。常用的荧光片段包括蒽类(anthracene) 染料、香豆素类(coumarin)、荧光素类(fluorescein)、罗丹明类(rhodamine)、花菁类(cyanine)、硝基苯并呋喃类(NBD) 以及氟硼二吡咯(BODIPY) 类染料[18-20] (图 2)。
因此, 将具有优秀活性的抗真菌先导化合物上引入荧光基团, 设计得到新型抗真菌荧光探针, 对于病原真菌的检测及抗真菌作用机制研究具有重要意义[21, 22]
NBD荧光基团因其Stokes位移长(发射波长与激发波长差较大)、量子产率高、分子量小、可标记识别基团中裸露的氨基等多种优点, 被广泛应用于新型荧光标记基团, 在生物活性成像中发挥重要作用[23, 24]。NBD基团自身没有荧光特性, 在其4-位上引入给电子基后赋予荧光性质, 以末端氨基连接最为常见。
本研究将先导化合物JYJ-19中苯氧侧链基团替换成4-位引入氨基的荧光片段NBD, 并通过三个碳原子的碳链与咔啉药效团骨架进行连接, 设计合成了3个新型咔啉类荧光探针(图 3)。以期在评价荧光探针的光学性质以及合适的体外生物活性之后, 能够实现在真菌细胞内的成像。
以市售的荧光片段NBD-Cl (1) 为起始原料, 经一步氨解反应得到荧光侧链中间体(2), 与3-溴丙胺氢溴酸盐发生亲核取代反应得到阴性对照化合物F0。以干燥的THF为溶剂, 原料(3) 与二碳酸二叔丁酯反应得到中间体(4), 以干燥的DMF为溶剂, 中间体(4) 和1, 3-二溴丙烷在氢化钠的作用下得到中间体(5), 中间体(5) 与荧光侧链中间体(2) 发生亲核取代反应后, 在三氟乙酸酸性环境中脱去叔丁氧羰基保护基得到三种F1~F3咔啉荧光探针终产物(路线图 12)。
用微量液基稀释法对三个咔啉类荧光探针进行体外抗真菌活性测试。结果表明, 三个荧光探针中, 咔啉骨架上6位吸电子基氯原子取代的活性要优于给电子基甲氧基取代以及氢原子取代的化合物(F1 > F2、F3), 其中化合物F1的抗新生隐球菌活性较好(MIC80 = 32 μg·mL-1), 可用于抗真菌作用机制研究(表 2)。
化合物F1 (10 μmol·L-1) 在350~700 nm内的最大激发波长λex = 470 nm, 以470 nm作为激发波长记录500~700 nm的发射光谱, 最大发射波长λem = 540 nm。与文献[25]报道的NBD类荧光化合物的实验数据一致。
为了研究咔啉类化合物在新生隐球菌细胞中的分布以及可能作用的细胞器, 随即进行亚细胞定位实验的相关研究。若目标荧光探针发出的绿色荧光与特异性细胞器探针发出的红色荧光能够叠合在一起呈现出黄色, 说明化合物可能在细胞中的某细胞器处富集。将咔啉类荧光探针F1以及阴性对照化合物F0分别与新生隐球菌细胞共同培养数小时后, 再分别与特异性的细胞器染料(Mito tracker: 线粒体红色荧光探针; Lyso tracker: 溶酶体红色荧光探针; ER tracker: 内质网红色荧光探针, 购自碧云天生物科技公司) 共同培养, 在荧光共聚焦显微镜下观察颜色变化。实验结果显示, 阴性对照物F0与三种细胞器探针共定位时, 基本上无荧光产生, 说明单独的荧光片段无法进入细胞, NBD荧光片段对细胞生长无明显影响(图 4)。
咔啉类荧光探针F1与线粒体红色荧光探针在新生隐球菌H99细胞中共定位的Pearson's correlation系数为0.853 0, 高于同浓度下F1与溶酶体红色荧光探针(0.610 5)、内质网红色荧光探针(0.514 9) 共定位的实验结果(图 5)。说明咔啉类化合物是由β-咔啉骨架药效团进入细胞发挥药效作用, 上述实验结果提示咔啉类化合物可以在线粒体富集, 可能通过作用于线粒体发挥药理活性。
亚细胞定位实验结果表明, 咔啉类荧光化合物与线粒体荧光探针的相关共定位系数为0.853 0, 提示咔啉类化合物可能通过作用于线粒体部位发挥其生物学活性。细胞内活性氧水平可以反映药物作用对线粒体功能的影响。因此, 本研究采用活性氧检测试剂盒开展了细胞内活性氧水平检测实验。实验结果表明, 空白对照组真菌细胞内无荧光, 用试剂盒中阳性对照药Rosup处理后, 真菌细胞发出的荧光被共聚焦显微镜检测到, 提示细胞内活性氧水平显著提高。将咔啉类化合物JYJ-19与真菌细胞共培养后, 同样细胞发出荧光被共聚焦显微镜检测到, 提示JYJ-19可引起细胞内活性氧水平升高, 咔啉类化合物可能作用于线粒体引起功能变化(图 6)。
综上所述, 本研究基于咔啉类先导化合物JYJ-19设计合成了三个全新的小分子荧光探针, 其中咔啉类荧光探针F1保持了一定的体外抗真菌活性, 且具有良好的荧光性质。亚细胞定位实验结果显示, F1定位于线粒体中; 细胞内活性氧水平检测结果表明JYJ-19能引起细胞内活性氧水平升高。因此咔啉类化合物可能通过作用于真菌线粒体发挥抗真菌活性。
实验中所用的试剂或溶剂皆为市售分析纯或化学纯。薄层色谱(TLC) 分析使用GF254硅胶板(中国青岛海洋化工), 硅胶柱层析所用硅胶为300~400目(中国青岛海洋化工)。核磁分析使用Bruker AVANCE 300或Bruker AVANCE 600核磁仪(Bruker Company, Germany) 记录1H NMR谱图, 以TMS为内标、CDCl3或DMSO-d6为溶剂, 化学位移值(δ) 和耦合常数(J) 分别以ppm和Hz为单位。所有目标化合物的纯度均大于等于90%。
量取10 mL无水甲醇于50 mL的茄形烧瓶中, 加入市售原料1 (0.5 g, 2.5 mmol) 混匀后, 用滴管吸取25%~28%氨水溶液(4.4 mL) 逐滴加入到混合溶液中, 于室温条件下避光搅拌反应18 h, TLC板监测反应原料1基本消耗完全。反应结束后, 减压除去CH3OH溶剂, 加30 mL水超声分散反应体系, 转移至250 mL分液漏斗中, EtOAc萃取三次(50 mL×3), 合并收集三次萃取得到的EtOAc层, 依次分别用50 mL水洗涤一次, 饱和NaCl水溶液洗涤两次(50 mL×2) 促进油水相分离, 倒入适量的无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 使用抽滤漏斗过滤, 收集滤液在旋转蒸发仪下除去溶剂得到深黑绿色固体(2), 收率: 90%。1H NMR (600 MHz, DMSO-d6) δ 8.88 (s, 2H), 8.50 (d, J = 8.8 Hz, 1H), 6.39 (d, J = 8.8 Hz, 1H)。
量取15 mL干燥乙腈溶剂于50 mL的茄形烧瓶中, 称取中间体2粗品(0.36 g, 2.0 mmol, 1 equiv) 加入溶液中充分溶解后, 分别依次称取碘化钾(0.332 g, 2.0 mmol, 1 equiv)、无水碳酸钾(0.829 g, 6.0 mmol, 3 equiv)、市售3-溴丙胺氢溴酸盐(0.657 g, 3.0 mmol, 1.5 equiv) 加入上述混合溶液体系中, 于85 ℃条件下加热回流搅拌过夜, TLC板监测反应。反应结束后, 待反应体系降至室温, 减压除去MeCN溶剂, 加20 mL水超声分散反应体系, EtOAc萃取三次(20 mL×3), 合并收集三次萃取得到的EtOAc层, 依次分别用30 mL水洗涤一次, 饱和NaCl水溶液洗涤两次(30 mL×2) 促进油水相分离, 倒入适量的无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 使用抽滤漏斗过滤, 收集滤液在旋转蒸发仪下除去溶剂得到棕红色固体(F0)。收率: 64%。1H NMR (600 MHz, DMSO-d6) δ 8.43 (d, J = 9.0 Hz, 1H), 6.02 (d, J = 9.0 Hz, 1H), 4.75 (s, 2H), 4.41 (s, 2H), 2.59~2.53 (m, 2H)。
量取15 mL无水THF溶剂于50 mL的茄形烧瓶中, 称取中间体3c粗品(1.118 g) 加入溶液中充分溶解后, 逐滴加入市售的二碳酸二叔丁酯(1.205 g, 5.519 mmol, 1.2 equiv), 于室温条件下搅拌反应1.5 h。TLC板监测反应。反应结束后, 缓缓加入20 mL饱和NaHCO3水溶液淬灭反应时会有少量气体产生。转移至250 mL分液漏斗中, EtOAc萃取三次(40 mL×3), 合并收集EtOAc有机相, 依次分别用40 mL水洗涤一次, 饱和食盐水洗涤两次(40 mL×2), 倒入无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 使用抽滤漏斗过滤, 收集滤液在旋转蒸发仪下除去溶剂得到残余物粗品。粗品经硅胶拌样、硅胶柱层析(PE/EtOAc = 10∶1, v/v → PE/EtOAc = 8∶1, v/v) 得到白色固体(4c), 收率: 40%。1H NMR (600 MHz, CDCl3) δ 7.20 (d, J = 8.7 Hz, 1H), 6.94 (d, J = 1.7 Hz, 1H), 6.81 (dd, J = 8.7, 2.4 Hz, 1H), 4.63 (s, 2H), 3.86 (s, 3H), 3.77 (s, 2H), 2.77 (s, 2H), 1.51 (s, 9H)。
量取15 mL无水N, N-二甲基甲酰胺溶剂于50 mL的茄形烧瓶中, 加入中间体4c (1.11 g, 3.671 mmol, 1 equiv) 充分溶解后, 小心称取60%氢化钠(440 mg, 11.013 mmol, 3 equiv) 加入上述混合溶液体系中, 置于冰水浴环境中搅拌0.5 h后, 称取市售的1, 3-二溴丙烷(0.889 g, 4.405 mmol, 1.2 equiv) 加入反应体系中, 继续于室温条件下搅拌反应1 h。TLC板监测反应。反应结束后, 缓缓加入20 mL水淬灭剩余的氢化钠, 过程中可见黄色固体悬浮于水溶液中并放出热量, 稍冷EtOAc萃取三次(50 mL×3), 合并收集EtOAc层, 用饱和NaCl水洗涤三次(50 mL×3) 带走DMF溶剂, 倒入无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 使用抽滤漏斗过滤, 收集滤液在旋转蒸发仪下除去溶剂得到残余物粗品。粗品经硅胶拌样、硅胶柱层析(PE/EtOAc = 15∶1, v/v) 得到白色固体(5c), 收率: 14.5%。1H NMR (600 MHz, CDCl3) δ 7.24 (s, 1H), 6.94 (s, 1H), 6.85 (dd, J = 8.8, 2.4 Hz, 1H), 4.65 (s, 2H), 4.15 (t, J = 6.7 Hz, 2H), 3.86 (s, 3H), 3.75 (s, 2H), 3.37 (t, J = 6.1 Hz, 2H), 2.77 (s, 2H), 2.34~2.27 (m, 2H), 1.51 (s, 9H)。
合成方法参考化合物5c, 得近无色油状液体, 收率: 22%。1H NMR (600 MHz, CDCl3) δ 7.49 (d, J = 7.7 Hz, 1H), 7.35 (s, 1H), 7.21~7.18 (m, 1H), 7.11 (t, J = 7.5 Hz, 1H), 4.67 (s, 2H), 4.20 (t, J = 6.8 Hz, 2H), 3.76 (s, 2H), 3.38 (t, J = 6.1 Hz, 2H), 2.80 (s, 2H), 2.33 (s, 2H), 1.51 (s, 9H)。
量取15 mL干燥乙腈溶剂于50 mL的茄形烧瓶中, 称取中间体5c (0.215 g, 0.508 mmol, 1 equiv) 加入溶液中充分溶解后, 分别依次称取碘化钾(0.084 g, 0.508 mmol, 1 equiv)、无水碳酸钾(0.211 g, 1.524 mmol, 3 equiv)、中间体2 (0.110 g, 0.609 mmol, 1.2 equiv) 加入上述混合溶液体系中, 于85 ℃条件下加热回流搅拌10 h后, TLC板监测反应原料5c以及中间体2基本消耗完全。反应结束后, 待反应体系降至室温, 减压除去MeCN溶剂, 加入20 mL水超声分散, EtOAc萃取三次(20 mL×3), 合并收集三次萃取的EtOAc层, 依次分别用20 mL水洗涤一次、饱和NaCl水溶液洗涤两次(20 mL×3) 促进油水相分层, 倒入无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 抽滤漏斗过滤, 滤液在旋转蒸发仪下除去溶剂得到棕红色固体粗品(6c), 无需纯化, 直接进行下一步反应。
量取15 mL无水二氯甲烷溶剂于50 mL的茄形烧瓶中, 将上步中间体6c粗品加入溶液中充分溶解后, 缓慢滴加5 mL三氟乙酸, 于室温条件下搅拌反应1 h后, TLC板监测反应。反应结束后, 减压除去大部分溶剂以及未反应完的三氟乙酸, 用10%氢氧化钠水溶液调节溶液体系的pH至10~11, 转移至250 mL分液漏斗中, EtOAc萃取三次(20 mL×3), 合并收集三次萃取的EtOAc层, 依次分别用20 mL水洗涤一次、饱和NaCl水溶液洗涤两次(20 mL×3) 促进油水相分离, 倒入无水Na2SO4干燥, 干燥的效果以容器内壁无水珠悬挂、容器底部的Na2SO4固体不再结成块状为佳, 使用抽滤漏斗过滤, 收集滤液在旋转蒸发仪下除去溶剂得到残余物粗品。粗品经硅胶拌样、硅胶柱层析(PE/EtOAc = 1∶1 → CH2Cl2/MeOH = 10∶1, v/v) 得到棕红色固体(F3), 合并两步收率: 16.78%。1H NMR (600 MHz, DMSO-d6) δ 8.41 (d, J = 8.5 Hz, 1H), 7.34 (d, J = 8.9 Hz, 1H), 6.83 (d, J = 2.4 Hz, 1H), 6.68 (d, J = 6.8 Hz, 1H), 6.23 (d, J = 9.3 Hz, 1H), 4.19~4.10 (m, 4H), 3.71 (s, 3H), 3.46 (s, 2H), 3.17 (t, J = 5.3 Hz, 2H), 2.69 (s, 2H), 2.10~2.04 (m, 2H)。
合成方法参考化合物F3, 得棕红色固体粉末, 收率: 22%。1H NMR (600 MHz, DMSO-d6) δ 8.44 (d, J = 7.8 Hz, 1H), 7.49 (d, J = 8.6 Hz, 1H), 7.39~7.38 (m, 1H), 7.04 (d, J = 8.1 Hz, 1H), 6.27 (d, J = 8.3 Hz, 1H), 4.21 (t, J = 6.6 Hz, 2H), 4.11 (s, 2H), 3.47 (s, 2H), 3.12 (s, 2H), 2.67 (s, 2H), 2.11~2.04 (m, 2H)。
合成方法参考化合物F3, 得棕红色固体粉末, 收率: 24%。1H NMR (600 MHz, DMSO-d6) δ 8.43 (d, J = 8.6 Hz, 1H), 7.47 (d, J = 8.2 Hz, 1H), 7.39 (d, J = 7.8 Hz, 1H), 7.10~7.05 (m, 1H), 6.98 (t, J = 6.5 Hz, 1H), 6.29 (d, J = 9.6 Hz, 1H), 4.26~4.20 (m, 4H), 3.49 (s, 2H), 3.24 (t, J = 5.7 Hz, 2H), 3.16 (s, 1H), 2.78 (t, J = 5.1 Hz, 2H), 2.12~2.07 (m, 2H), 1.89 (s, 1H)。
根据美国临床实验室标准化协会(CLSI) 发布的M27-A3和M38-A2方案进行测试。将活化好的待测菌株用PBS洗3次, 再用RPMI 1640培养液配成1×103 CFU·mL-1的菌悬液。将菌悬液加到96孔板(每孔100 μL), 待测化合物以倍半稀释的方法加到菌悬液。以RPMI 1640为空白对照, 不加药菌悬液作阴性对照, 于35 ℃恒温培养箱中培养72 h, 使用酶标仪OD630进行读数, 取抑菌率≥ 80%的最小浓度即为最低抑菌浓度(MIC80)。
溶液的配制、稀释: 称取化合物F1配成2 mg·mL-1的DMSO溶液, 取定量体积的DMSO药液用无水甲醇稀释成10 μmol·L-1
波长检测: 在96孔细胞培养板上, 吸取200 μL 10 μmol·L-1化合物F1甲醇溶液, 做三复孔。全波长酶标仪检测。选择“Absorbance Scan”, 在230~800 nm检测吸光度, 最大吸光度所对应的波长作为最大激发波长, 选择“Fluorescence Scan”, 在500~800 nm检测荧光强度, 最大荧光强度所对应的波长作为最大发射波长。
从保存于4 ℃冰箱中H99菌株的SDA板上挑取单一菌落于YEPD培养液中, 在30 ℃摇床培养箱中孵育16 h, 离心收集指数生长后期的C. neoformans H99细胞, 加入1 mL PBS缓冲液清洗一遍后, 加入1 mL新鲜YEPD培养液和20 μL荧光探针化合物F1, 涡旋混匀后于37 ℃霉菌培养箱中共孵育4 h, 离心(3 000 r·min-1, 1 min) 吸弃未进入细胞内的探针分子和培养液, 用1 mL PBS洗一次, 加入1 mL新鲜YEPD培养液, 分别加入市售的线粒体、溶酶体、内质网红色荧光探针各1 μL, 于37 ℃霉菌培养箱中作用0.5~1 h后(线粒体和内质网探针需作用0.5 h, 溶酶体探针作用时间需要延长到1 h), 离心(3 000 r·min-1, 1 min) 后吸走未进入细胞内的探针分子和上清液, 1 mL生理盐水洗3、4遍后防止在显微镜下观察产生背景干扰色, 最后用200 μL生理盐水悬浮在1.5 mL灭菌离心管中, 便于取样观察(注意整个过程在避光条件下进行, 避免荧光淬灭)。取样前吹打混匀后, 取5 μL左右的菌悬液滴于载玻片中央, 沿一侧轻轻盖上盖玻片, 放置几分钟使真菌细胞固定在载玻片上后, 在荧光共聚焦显微镜(Leica TCS SP5 confocal Laser) 下观察。
荧光共聚焦显微镜参数设置: 咔啉类荧光探针化合物F0、F1的激发波长选在488 nm处, 市售的细胞器红色荧光探针的激发波长选在561 nm处。
从4 ℃冰箱中取出涂有C. neoformans H99菌株的SDA板, 挑取单一菌落于1 mL YEPD培养液中, 在30 ℃摇床中活化16 h后, 转移到1.5 mL管中, 离心弃上清培养液, 加入1 mL PBS缓冲溶液洗一遍后, 离心弃上清液, 用YEPD培养液稀释10倍后, 即1×107 CFU·mL-1的菌悬液。取若干1.5 mL离心管, 加入1 mL菌悬液, 分别加入一定体积的待测化合物, 使终浓度为4 μg·mL-1, 于37 ℃霉菌培养箱中孵育12 h后, 离心弃去上清培养液, 用PBS洗一遍后, 加入1 mL新鲜YEPD培养液和1 μL 10 mmol·L-1市售探针DCFH-DA, 继续于37 ℃霉菌培养箱中孵育20 min (每隔3~5 min颠倒混匀), 离心弃去未进入细胞内的探针分子和上清培养液, 1 mL生理盐水洗3、4遍后防止在显微镜下观察产生背景干扰色, 最后用200 μL生理盐水悬浮在1.5 mL灭菌离心管中, 便于取样观察(注意整个过程在避光条件下进行, 避免荧光淬灭)。取样前吹打混匀后, 取5 μL左右的菌悬液滴于载玻片中央, 沿一侧轻轻盖上盖玻片, 放置几分钟使真菌细胞固定在载玻片上后, 在荧光共聚焦显微镜下观察。
