Urine nontargeted metabolomics technology was developed for investigating the effect and mechanism of improving learning and memory ability in APP/PS1 mice of Psoralea corylifolia. All animal experiments were approved by the Animal Ethics Committee of Heilongjiang University of Chinese Medicine (Approval No.: 2020092502). Sixteen APP/PS1 mice were randomly divided into the model group and Psoralea corylifolia group (0.5 g·kg-1), and eight male C57BL/6J mice of the same background were selected as control group, step-through test and novel object recognition were used as evaluation indexes. Changes in urine endogenous metabolites of mice from eachgroup were determined by ultra-high performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF-MS), and differential metabolites were screened, and metabolic pathway enrichment analysis was performed. The results of pharmacodynamic investigation showed that Psoralea corylifolia can reduce the dark incubation period and number of errors in APP/PS1 mice (P < 0.01) and improve the new object recognition index of APP/PS1 mice (P < 0.01). Metabolomics analysis identified 15 differential metabolites, and 9 differential metabolites were significantly call back by Psoralea corylifolia. Metabolic pathway analysis showed that histidine metabolism, citric acid cycle, taurine and hypotaurine metabolism and glucose metabolism were the main metabolic pathways of Psoralea corylifolia in improving learning and memory ability. These studies suggest that Psoralea corylifolia improves the learning and memory ability of APP/PS1 mice, and its mechanism may be related to improving mitochondrial dysfunction, reducing peripheral histamine level, regulating energy metabolism disorders and antioxidant levels.

运用尿液非靶代谢组学探究补骨脂改善APP/PS1小鼠学习记忆能力的作用机制。所有动物实验均经过黑龙江中医药大学动物伦理委员会批准(批准号: 2020092502)。将16只3月龄雄性APP/PS1小鼠随机分为模型组和补骨脂组(0.5 g·kg-1), 另取8只同背景C57BL/6J小鼠作为空白组, 以避暗实验和新物体识别实验为评价指标, 通过超高效液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF-MS) 测定各组小鼠尿液中内源性代谢物的变化, 筛选差异代谢物, 并进行代谢通路富集分析。实验结果显示, 与模型组相比, 补骨脂可明显减少APP/PS1小鼠的避暗潜伏期及错误次数(P < 0.01), 显著提高APP/PS1小鼠的新物体识别指数(P < 0.01)。代谢组学分析鉴定出15个差异代谢物, 补骨脂能显著回调9个差异代谢物。代谢通路分析显示, 组氨酸代谢、柠檬酸循环、牛磺酸和次牛磺酸代谢和糖代谢途径是补骨脂发挥改善学习记忆能力作用的主要代谢途径。研究提示, 补骨脂改善APP/PS1小鼠的学习记忆能力的机制可能与改善线粒体功能障碍、降低外周组胺水平、调节能量代谢紊乱及抗氧化水平等相关。

Urine metabolomics study of Psoralea corylifolia in improving learning and memory ability in APP/PS1 mice

运用尿液非靶代谢组学探究补骨脂改善APP/PS1小鼠学习记忆能力的作用机制。所有动物实验均经过黑龙江中医药大学动物伦理委员会批准(批准号: 2020092502)。将16只3月龄雄性APP/PS1小鼠随机分为模型组和补骨脂组(0.5 g·kg-1), 另取8只同背景C57BL/6J小鼠作为空白组, 以避暗实验和新物体识别实验为评价指标, 通过超高效液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF-MS) 测定各组小鼠尿液中内源性代谢物的变化, 筛选差异代谢物, 并进行代谢通路富集分析。实验结果显示, 与模型组相比, 补骨脂可明显减少APP/PS1小鼠的避暗潜伏期及错误次数(P < 0.01), 显著提高APP/PS1小鼠的新物体识别指数(P < 0.01)。代谢组学分析鉴定出15个差异代谢物, 补骨脂能显著回调9个差异代谢物。代谢通路分析显示, 组氨酸代谢、柠檬酸循环、牛磺酸和次牛磺酸代谢和糖代谢途径是补骨脂发挥改善学习记忆能力作用的主要代谢途径。研究提示, 补骨脂改善APP/PS1小鼠的学习记忆能力的机制可能与改善线粒体功能障碍、降低外周组胺水平、调节能量代谢紊乱及抗氧化水平等相关。

Urine nontargeted metabolomics technology was developed for investigating the effect and mechanism of improving learning and memory ability in APP/PS1 mice of Psoralea corylifolia. All animal experiments were approved by the Animal Ethics Committee of Heilongjiang University of Chinese Medicine (Approval No.: 2020092502). Sixteen APP/PS1 mice were randomly divided into the model group and Psoralea corylifolia group (0.5 g·kg-1), and eight male C57BL/6J mice of the same background were selected as control group, step-through test and novel object recognition were used as evaluation indexes. Changes in urine endogenous metabolites of mice from eachgroup were determined by ultra-high performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF-MS), and differential metabolites were screened, and metabolic pathway enrichment analysis was performed. The results of pharmacodynamic investigation showed that Psoralea corylifolia can reduce the dark incubation period and number of errors in APP/PS1 mice (P < 0.01) and improve the new object recognition index of APP/PS1 mice (P < 0.01). Metabolomics analysis identified 15 differential metabolites, and 9 differential metabolites were significantly call back by Psoralea corylifolia. Metabolic pathway analysis showed that histidine metabolism, citric acid cycle, taurine and hypotaurine metabolism and glucose metabolism were the main metabolic pathways of Psoralea corylifolia in improving learning and memory ability. These studies suggest that Psoralea corylifolia improves the learning and memory ability of APP/PS1 mice, and its mechanism may be related to improving mitochondrial dysfunction, reducing peripheral histamine level, regulating energy metabolism disorders and antioxidant levels.

阿尔茨海默病(Alzheimer's disease, AD) 是最常见的痴呆类型, 主要表现为记忆力减退和认知功能障碍[1]。目前, AD病因尚不清楚, 全球批准用于AD临床治疗的药物仅有2类, 且只能控制症状而无法治愈或阻止病情的发展[2]。中医认为肾精亏虚所致的髓海不足是AD发病的关键[3], 大量临床研究证实从“肾”论治, 补肾填精益髓法治疗AD是一种行之有效的疗法[4]。补骨脂作为经典的补肾中药, 课题组前期研究发现其有效成分补骨脂素可作用于雌激素受体β (ERβ) 介导p-ERK信号通路, 降低β淀粉样蛋白(Aβ) 对PC12细胞的损伤[5]。此外, 补骨脂提取物可通过增加ERα、ERβ表达, 调节下丘脑-垂体-性腺轴负反馈系统、调控ERβ-ERK1/2途径, 改善淀粉样前体蛋白/早老素基因1 (amyloid precursor protein/presenilin 1, APP/PS1) 双转基因小鼠的学习记忆能力[6, 7]。另外, 有研究显示补骨脂有效成分可能通过抑制caspase-3凋亡因子表达, 提高线粒体膜电位、抗炎抗氧化的作用, 发挥对神经细胞的保护作用[8, 9]
尿液代谢组学是近年来发现非侵入性生物标志物的一个重要领域, 其可以检测出针对特定疾病或治疗干预的细微代谢差异[10], 具有取样量少、实施方便、无创等优势[11]。超高效液相色谱与高分辨质谱相联用技术具有耗时少、分辨率高、特异性强和灵敏度高等特点, 能够进行更加准确可靠的分析[12]。目前尚缺少补骨脂对APP/PS1小鼠尿液代谢影响的相关研究。本研究将采用高分辨液质联用技术从非靶尿液代谢组学角度出发对补骨脂改善APP/PS1小鼠学习记忆能力的作用机制提供全新依据。
实验动物  SPF级3月龄APP/PS1小鼠16只, C57BL/6J小鼠8只, 雄性, 体重(25 ± 5) g, 购自于南京大学南京生物药研究院, 实验动物生产许可证号: SCXK (苏) 2015-0001。饲养环境温度为(21 ± 1) ℃, 相对湿度为(55 ± 1) %, 实验期间保持小鼠自由饮水进食。上述动物实验遵循黑龙江中医药大学动物伦理委员会的规定并通过动物实验伦理审查(批准号: 2020092502)。
药物与试剂  补骨脂Psoralea corylifolia Linn.购自于黑龙江中医药大学国医堂, 由黑龙江中医药大学张宁教授鉴定为正品。补骨脂提取物(由本实验室自制), 提取方法参照文献[6]。甲酸(HPLC-MS级, 货号: 540-69-2, Sigma公司)、甲醇、乙腈(HPLC-MS级, 货号分别为: W50164, W50163, Dikma公司), 水为双蒸水。
主要仪器  SCIEX ExionLC AD液相色谱仪; SCIEX 5600+Q-TOF质谱仪(Applied Biosystems Sciex公司); RD1106-SB-M避暗设备和RD1121-NR-M新物体识别设备(上海移数信息科技有限公司)。
动物分组与给药  将APP/PS1小鼠随机分为模型组和补骨脂组, C57BL/6J小鼠为空白组, 每组8只。参考《中华人民共和国药典》2020年版第一部[13], 可知补骨脂的用法用量为成人煎服每日6~10 g, 查阅其他资料[14-16]发现补骨脂在不同的病症之中用量不一, 且不同方剂中差异较大, 最终在药典的基础上适当增大剂量以保证药效, 为6~15 g。按人临床日用药量折算及提取的出膏率最终计算并选择0.5 g·kg-1的剂量灌胃给药。补骨脂组每日给予补骨脂提取物0.5 g·kg-1, 空白组与模型组每日给予等体积生理盐水, 各组均连续灌胃90天后进行行为学实验并采集尿样进行检测[6]
行为学实验  避暗实验: 设备分为明暗两室, 两室间有拱形圆洞, 明室有灯照射, 暗室底部有通电铜栅。训练时将小鼠放入明室, 小鼠进入暗室时会被电击, 训练时间为5 min。训练期间, 弃去避暗潜伏期大于180 s的小鼠。24 h后测试, 记录小鼠5 min内第一次进入暗室的时间(即为潜伏期) 和进入次数(即为错误次数)。
新物体识别实验: 将小鼠置于实验箱内自由活动5 min使其熟悉测试环境。训练时将小鼠置于箱内2个完全相同物体的中间, 物体对称放置, 小鼠自由探索5 min, 训练结束1 h后进行测试。将2个物体其中的1个用新物体取代, 记录5 min内小鼠嗅探新物体(tn) 和旧物体(tf) 的时间。计算相对识别指数[识别指数= (tn-tf)/(tn + tf)] 来评价其学习记忆能力。
样本采集与预处理  给药第90天后, 收集小鼠12 h的夜尿。所有尿液均存于接尿管中, 接尿管置于湿冰上保持低温, 将收集的尿液置于离心管中, 3 500 r·min-1离心5 min, 取上清液置于-80 ℃冰箱保存。分析前, 取出尿样室温下解冻。取尿液200 μL, 加入等量甲醇, 涡旋2 min, 4 ℃下13 000 r·min-1离心10 min后, 过0.22 μm微孔滤膜, 取5.0 μL进样。
质量控制样本的制备  每个尿样吸取10 μL, 混合均匀后作为质量控制(QC) 样本, 以用于检测UPLC-Q-TOF-MS的重复性和稳定性。QC样本的制备方法同尿样的制备。
分析条件  色谱柱: ACQUITY UPLC HSS T3柱(100 mm × 2.1 mm, 1.8 μm); 流动相0.1%甲酸水溶液(A)-0.1%甲酸乙腈(B), 洗脱程序: 0~1 min (2% B), 1~2 min (2%~5% B), 2~5 min (5%~12% B), 5~10 min (12%~20% B), 10~12 min (20%~30% B), 12~13 min (30%~50% B), 13~15 min (50%~100% B)。流速0.5 mL·min-1, 柱温40 ℃, 进样量5 μL。
质谱条件: 电喷雾离子化源(ESI), 在正、负离子扫描模式下: 扫描范围m/z 50~1 500; 喷雾电压: (+) 3.0 kV, (-) 2.9 kV, 脱溶剂气流量900 L·h-1, 脱溶剂气温度450 ℃, 锥孔反吹气流量50 L·h-1
数据处理  将所获得的原始数据导入Progenesis QI 2.0软件进行去噪音、峰匹配、峰提取及归一化处理, 通过查询HMDB、Metlin以及本地数据库和相关文献资料对差异代谢物进行鉴定。以m/z、保留时间、峰面积保存到Excel文件, 将excel文件导入MetaboAnalyst 5.0 (http://www.metaboanalyst.ca/) 在线数据库进行主成分分析(principal component analysis, PCA) 及正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis, OPLS-DA)。通过得分图比较空白组与APP/PS1之间的差异, 筛选差异代谢物(VIP > 1, P < 0.05)。通过MetaboAnalyst平台中Pathway Analysis模块对筛选出的差异代谢物进行代谢通路的富集分析。行为学数据采用SPSS 26.0统计软件进行统计分析。采用均数±标准差(x ± s) 表示。数据两组间比较, 满足正态性分布者采用独立样本t检验; 多组间比较, 满足正态性分布者采用单因素方差分析, 方差不齐者采用秩和检验, 以P < 0.05为差异有统计学意义。
与空白组比较, 模型组避暗潜伏期明显变长(P < 0.01, 图 1A), 错误次数明显增加(P < 0.01, 图 1B), 记忆消退现象较明显。与模型组比较, 补骨脂组避暗潜伏期明显缩短(P < 0.01, 图 1A), 错误次数明显减少(P < 0.01, 图 1B), 说明补骨脂组记忆消退现象减弱。以上结果说明, 补骨脂具有改善APP/PS1小鼠记忆消退的作用。
与空白组比较, 模型组新物体识别指数明显减小(P < 0.01, 图 1C), 说明模型组没有保留对旧物体的记忆, 无法辨别新旧物体, 其识别记忆能力损伤。与模型组相比, 补骨脂组新物体识别指数明显增大(P < 0.01, 图 1C), 说明补骨脂组识别记忆能力有明显的好转。以上结果说明, 补骨脂具有改善APP/PS1小鼠识别记忆损伤的作用。
各组尿样正、负离子模式下的主成分分析(PCA) 结果见图 2AB所示。从PCA得分图可见, QC样本聚为一簇, 表明所建立的代谢组学分析方法有较好的稳定性和重复性。正、负离子模式下空白组和模型组明显分离, 说明模型组小鼠的尿样已发生明显的差异代谢变化。经补骨脂提取物干预后, 模型组小鼠的尿样代谢轮廓均有明显回调趋势, 提示补骨脂对模型组小鼠代谢紊乱有一定的改善作用。进一步进行正交偏最小二乘判别分析(OPLS-DA), 结果见图 2CD所示。聚焦影响分组明显的小分子代谢产物, 正离子模式下空白组与模型组明显分离, 结果显示模型参数R2Y (表示OPLS-DA模型解释率) = 0.995, Q2 (表示OPLS-DA模型预测率) = 0.950; 负离子模式下, R2YQ2值分别为0.998、0.968, 表明拟合模型的准确度和预测能力良好。正离子模式下共筛选出8个差异代谢物, 负离子模式下筛选出7个差异代谢物, 结果列于表 1。结合代谢热图分析, 见图 3AB。图中颜色表示含量, 红色表示含量高表达, 蓝色表示含量低表达, 补骨脂给药干预后显著回调组胺、咪唑乙酸、柠檬酸、异柠檬酸、顺式-乌头酸和牛磺酸。
在富集通路图中, 结果如图 4。每个圆的颜色和大小是基于P值和通路影响因子决定的, 即节点颜色越深参与此通路的代谢物越多, 节点越大则它在此次机体的整体代谢轮廓中占的比重越大, 作用越明显[17]。提示补骨脂提取物主要影响组氨酸代谢、柠檬酸循环、牛磺酸和次牛磺酸代谢和糖代谢等代谢通路。
APP/PS1双转基因小鼠是目前研究AD分子机制及治疗作用的常用模型, 能够较好地模拟类似AD的病理特点, 本研究沿用课题组常用的补肾中药补骨脂干预治疗APP/PS1小鼠。根据之前的大量研究[18], APP/PS1小鼠最早可以在3月龄时出现认知行为学变化, 5月龄脑内可以出现Aβ斑块, 病理改变随年龄增长逐渐加重。因此, 选择3月龄APP/PS1小鼠作为模型动物, 可以更加直接地考察补骨脂对AD发病早期的潜在治疗途径。
研究结果显示APP/PS1小鼠的尿液中与能量相关的代谢物发生了明显变化。作者发现在APP/PS1小鼠尿液中葡萄糖含量明显升高, 这与临床AD患者尿液中葡萄糖含量变化一致, 且患者体内葡萄糖含量会随着病情的发展呈线性升高[19]。同时, 在模型组小鼠尿液中柠檬酸、异柠檬酸和顺乌头酸的浓度明显低于空白组, Yu[20]和Zheng等[21]对APP/PS1小鼠尿液的研究中也报道了其含量的降低。葡萄糖代谢在很大程度上依赖于线粒体在细胞中产生的能量, 当线粒体功能出现障碍时, 表现为葡萄糖代谢紊乱。此外, 还观察到牛磺酸含量在APP/PS1小鼠尿液中降低, Dong等[22]对APP/PS1小鼠尿液的研究中也得到了同样的结果。有研究表明, 牛磺酸可通过影响线粒体tRNA的修饰而使线粒体编码蛋白合成受阻, 影响线粒体功能[23]。同时, 牛磺酸作为抗氧化剂, 其在尿液中含量的降低可能预示着体内抗氧化水平下降。研究表明, 线粒体和生物能量变化在时间上先于Aβ稳态的可检测变化[24], 线粒体生物发生调节线粒体内Aβ的产生, 而三羧酸循环途径的下调以及牛磺酸含量的降低进一步提示线粒体功能障碍在诱导AD病理学中起着关键的上游作用。经补骨脂干预后, 尿样中代谢物水平有明显回调, 提示补骨脂显著改善APP/PS1小鼠学习记忆能力, 其机制可能与改善线粒体功能障碍, 调节体内的能量代谢紊乱及抗氧化水平有关。
组胺是炎症介质, 也是中枢神经系统中的一种重要的神经递质或神经调质, 可通过调节长时程增强(long-term potentiation, LTP) 和保护皮层海马神经元来改善AD引起的认知功能的下降[25]。组胺在APP/PS1小鼠尿液中具有明显的变化。有研究报道外周中组胺升高的原因可能是嗜碱性粒细胞、肥大细胞释放组胺, 也可能是由于携带载脂蛋白epsilon4等位基因的AD患者中存在血脑屏障渗漏的结果[26]。组氨酸是组胺的前体物质, 肠道菌群可将组氨酸降解产生组胺[27]。研究表明, 尿嘧啶水平下降预示着肠道中菌群数量降低[28]。本实验中, APP/PS1小鼠尿样中尿嘧啶水平下降, 其含量变化提示APP/PS1小鼠可能存在肠道环境的变化, 补骨脂干预后, 尿液中组胺含量显著降低, 组氨酸及尿嘧啶含量无显著变化, 提示补骨脂可能通过抗炎或是修复血脑屏障泄露, 降低外周组胺水平, 而不是通过改善肠道菌群紊乱, 发挥改善APP/PS1小鼠学习记忆能力的作用。
在补骨脂改善APP/PS1小鼠的血清代谢组学的研究中, 主要通过花生四烯酸代谢、色氨酸代谢、甘油磷脂代谢、甘油脂代谢等代谢通路发挥促进能量代谢、降低氧化损伤、减少神经炎症等作用[29]。Xu等[30]从补骨脂的乙醇提取物中共鉴定出45种化合物, 包括34种黄酮类、7种香豆素类、3种萜酚类和2种其他类, 补骨脂中不同类别化学成分具有多种药理作用。研究表明, 香豆素中的补骨脂素和异补骨脂素可使TCA循环中间产物含量显著升高[31], 且异补骨脂素还对H2O2诱导氧化损伤的线粒体超微结构有保护作用[32]。香豆素类成分中的补骨脂定, 黄酮类成分中的补骨脂宁、补骨脂二氢黄酮甲醚、异补骨脂二氢黄酮等具有良好的抗氧化作用, 其中以补骨脂定抗氧化作用最强[33]。还有萜酚类中的补骨脂酚, 补骨脂酚A和补骨脂酰脂酚F具有明显的抗炎作用[34]。除此之外, 补骨脂还具有抗菌、抗肿瘤、促进骨生长等多种药理作用。
综上所述, 补骨脂改善APP/PS1小鼠的学习记忆能力可能通过改善线粒体功能障碍, 调节能量代谢紊乱及抗氧化水平、降低外周组胺水平等方面发挥作用。今后, 可与靶向代谢组学、蛋白组学等联合进一步探究补骨脂改善APP/PS1小鼠学习记忆能力的作用机制, 更全面了解补骨脂治疗阿尔茨海默病认知障碍的潜在治疗途径。
