In this study, inspired by biomimetic mineralization process, we have developed imidazole-zeolite framework (ZIF-8) conjugated VNP20009, and chemotherapy drug doxorubicin hydrochloride (DOX) was encapsulated to obtain ZD@VNP. The morphology and the combination of ZIF-8 and VNP was characterized by transmission electron microscopy and laser confocal microscopy. Fluorescence spectrophotometry was used to examine the encapsulation rate and in vitro release rate of DOX. CCK-8 and FDA/PI cell viability staining experiments were used to evaluate the ability of ZD@VNP to inhibit cell proliferation. Melanoma mouse model was established to investigate the effect of ZD@VNP to inhibit tumor growth. It was shown that ZIF-8 was evenly bounded to the surface of VNP20009, and laser confocal microscopy results also confirm the combination of ZIF-8 with VNP in ZD@VNP. The encapsulation rate of DOX in ZD@VNP was calculated to be 85.7% ± 3.7%, and the release of DOX under the buffer at pH 6.0 was significantly higher than that of pH 7.4. ZD@VNP treatment resulted in a greater inhibitory effect on B16F10 cell proliferation compared to DOX treatment. Animal experiment results showed that compared with VNP+DOX, ZD@VNP treatment can significantly inhibit the growth of B16F10 tumors in C57BL/6 mice and prolonged survival (all animal experiments were approved by the Institutional Animal Care and Ethics Committee Guangdong Medical University, No. GDMU-2023-2518). In summary, ZD@VNP was prepared through a facile, one-step method, which can significantly enhance the proliferation inhibitory effect of DOX to inhibit tumor growth and prolong survival. Our results demonstrate that ZD@VNP has great application prospects in the field of drug delivery.

本研究利用仿生矿化将咪唑-沸石骨架(ZIF-8)巧妙地搭载在减毒沙门氏菌VNP20009表面, 同时包载化疗药物盐酸多柔比星(doxorubicin, DOX)得到ZD@VNP; 采用透射电镜、激光共聚焦显微镜对ZD@VNP的形貌及ZIF-8与VNP20009的结合情况进行表征。采用荧光分光光度法考察DOX的包封率及体外释放率; 通过CCK-8和细胞活/死染色FDA/PI评估ZD@VNP抑制黑色素瘤细胞(B16F10)增殖的能力, 并建立黑色素瘤小鼠模型考察ZD@VNP抑瘤效果。实验结果表明ZIF-8均匀地结合在VNP20009表面, 共聚焦结果也证实了ZD@VNP中ZIF-8与VNP的结合; ZD@VNP对DOX的包封率为85.7% ± 3.7%, 在pH 6.0的缓冲液中DOX的释放显著高于pH 7.4; 细胞实验结果表明ZD@VNP能增强DOX对B16F10细胞增殖的抑制作用; 药效实验结果表明与VNP+DOX相比, ZD@VNP治疗在C57BL/6小鼠(实验得到广东医科大学动物保护和伦理委员会批准, 编号: GDMU-2023-002518)上可显著抑制B16F10肿瘤生长, 并延长小鼠生存期。综上, 本研究借助仿生矿化通过一步法制备的ZD@VNP可显著增强DOX和VNP抑制肿瘤细胞生长的作用, 在药物递送领域具有较大的应用前景。

Preparation of Salmonella typhimurium VNP20009 conjugated with imidazole-zeolite framework and its anti-tumor effect

本研究利用仿生矿化将咪唑-沸石骨架(ZIF-8)巧妙地搭载在减毒沙门氏菌VNP20009表面, 同时包载化疗药物盐酸多柔比星(doxorubicin, DOX)得到ZD@VNP; 采用透射电镜、激光共聚焦显微镜对ZD@VNP的形貌及ZIF-8与VNP20009的结合情况进行表征。采用荧光分光光度法考察DOX的包封率及体外释放率; 通过CCK-8和细胞活/死染色FDA/PI评估ZD@VNP抑制黑色素瘤细胞(B16F10)增殖的能力, 并建立黑色素瘤小鼠模型考察ZD@VNP抑瘤效果。实验结果表明ZIF-8均匀地结合在VNP20009表面, 共聚焦结果也证实了ZD@VNP中ZIF-8与VNP的结合; ZD@VNP对DOX的包封率为85.7% ± 3.7%, 在pH 6.0的缓冲液中DOX的释放显著高于pH 7.4; 细胞实验结果表明ZD@VNP能增强DOX对B16F10细胞增殖的抑制作用; 药效实验结果表明与VNP+DOX相比, ZD@VNP治疗在C57BL/6小鼠(实验得到广东医科大学动物保护和伦理委员会批准, 编号: GDMU-2023-002518)上可显著抑制B16F10肿瘤生长, 并延长小鼠生存期。综上, 本研究借助仿生矿化通过一步法制备的ZD@VNP可显著增强DOX和VNP抑制肿瘤细胞生长的作用, 在药物递送领域具有较大的应用前景。

In this study, inspired by biomimetic mineralization process, we have developed imidazole-zeolite framework (ZIF-8) conjugated VNP20009, and chemotherapy drug doxorubicin hydrochloride (DOX) was encapsulated to obtain ZD@VNP. The morphology and the combination of ZIF-8 and VNP was characterized by transmission electron microscopy and laser confocal microscopy. Fluorescence spectrophotometry was used to examine the encapsulation rate and in vitro release rate of DOX. CCK-8 and FDA/PI cell viability staining experiments were used to evaluate the ability of ZD@VNP to inhibit cell proliferation. Melanoma mouse model was established to investigate the effect of ZD@VNP to inhibit tumor growth. It was shown that ZIF-8 was evenly bounded to the surface of VNP20009, and laser confocal microscopy results also confirm the combination of ZIF-8 with VNP in ZD@VNP. The encapsulation rate of DOX in ZD@VNP was calculated to be 85.7% ± 3.7%, and the release of DOX under the buffer at pH 6.0 was significantly higher than that of pH 7.4. ZD@VNP treatment resulted in a greater inhibitory effect on B16F10 cell proliferation compared to DOX treatment. Animal experiment results showed that compared with VNP+DOX, ZD@VNP treatment can significantly inhibit the growth of B16F10 tumors in C57BL/6 mice and prolonged survival (all animal experiments were approved by the Institutional Animal Care and Ethics Committee Guangdong Medical University, No. GDMU-2023-2518). In summary, ZD@VNP was prepared through a facile, one-step method, which can significantly enhance the proliferation inhibitory effect of DOX to inhibit tumor growth and prolong survival. Our results demonstrate that ZD@VNP has great application prospects in the field of drug delivery.

实体瘤具有血管异常增生及局部组织缺氧坏死特征, 大多数药物难以到达肿瘤深部, 导致其在肿瘤部位的蓄积不足, 削弱药效[1, 2]。沙门氏菌是一类兼性厌氧的革兰氏阴性菌, 能特异性地靶向实体瘤组织的乏氧区域, 并通过竞争营养、分泌毒素及激活机体免疫应答等多种途径发挥抗肿瘤效应[3-5]。VNP20009是将鼠伤寒沙门氏菌YS14028的purI和msbB两种基因敲除后得到的减毒菌株[6]。因其安全性高、可主动靶向乏氧肿瘤而广泛应用于抗肿瘤研究[7]。然而, 单独使用VNP的临床效果并不理想, 患者肿瘤并未出现明显消退[8]。这可能是由于肿瘤微环境的复杂性, 导致单一的细菌治疗难以达到期望的抗肿瘤效果[9]。将细菌治疗与其他临床治疗手段结合或许能改善疗效。
多柔比星(doxorubicin, DOX)是小分子广谱化疗药, 对乳腺癌、肝癌等多种肿瘤都有很好的效果[10-12]。但其体内非特异性分布及心脏毒性等不良反应使患者生存期缩短, 严重限制了其应用[13]。因此需要增加DOX在肿瘤组织内分布, 达到减毒增效的目的。将DOX与细菌有机结合, 利用细菌的肿瘤靶向性提高DOX在肿瘤部位的蓄积是一个可行的方法。但这种方式对搭载药物的结构有较高要求, 且复杂的反应过程可能会降低细菌存活率[14]
纳米技术为癌症治疗提供了新的方法, 研究者们开发了多种纳米载体, 包括脂质体、胶束、无机、有机纳米粒等用于药物递送[15-17]。金属有机骨架化合物是由金属离子和有机配体通过自组装连接而成的一类具有网络结构的多孔材料。其具有较大的比表面积, 可以调节的孔径和较高的负载能力而受到研究者们的广泛关注[18, 19]。沸石咪唑骨架材料由锌离子与2-甲基咪唑配位形成, 具有高载药量和pH敏感降解等特性, 是优良的药物载体[20]
基于此, 本研究借助仿生矿化的原理, 采用一步法巧妙连接ZIF-8与VNP, 并有效包载DOX得到ZD@VNP, 期望利用VNP的肿瘤靶向性, 将DOX有效递送至肿瘤组织, 提高其在肿瘤部位的蓄积。ZIF-8的pH降解特性使其在肿瘤环境中释放DOX, 被肿瘤细胞摄取后与VNP协同实现抗肿瘤作用。作者对ZD@VNP的形貌、ZIF-8与VNP的结合状态进行了表征。在B16F10细胞上通过CCK-8及细胞活/死染色FDA/PI评估了ZD@VNP对细胞增殖能力的影响, 发现ZD@VNP能显著抑制细胞增殖。在黑色素瘤小鼠模型上的结果也表明ZD@VNP治疗可显著抑制肿瘤生长并延长小鼠生存期。本研究结果表明, 借助仿生矿化通过一步法制备的ZD@VNP可显著增强DOX和VNP抑制肿瘤细胞生长的作用, 在药物递送领域具有较大的应用前景。
材料与动物  黑色素瘤细胞B16F10和减毒沙门氏菌VNP20009由四川大学惠赠; 盐酸多柔比星(125745-1)购自北京华奉联博化学材料有限公司; 蛋白胨(3684237)和酵母提取物(6752383-3)购自英国Oxoid公司; NaCl (2305125)购自西陇科学股份有限公司; 硫酸卡那霉素(3673809)和透析袋购自北京兰杰柯科技有限公司; DMEM高糖培养基、青链霉素混合液(1735702)和细胞活/死染色FDA/PI染色试剂盒(6535376)购自北京索莱宝科技有限公司; Tween 80 (62573-26)购自上海麦克林生化科技股份有限公司; 胎牛血清购自美国Invitrogen公司。雄性SPF级C57BL/6小鼠(17~19 g, 6~8周龄)购自广东省医学实验动物中心, 合格证号: 0372543632, 动物实验批准编号: GDMU-2023-002518。
仪器  酶标仪(H1MF, 美国Thermo Fisher公司); 超纯水仪(Milli-Q, 北京优普超纯科技有限公司); CO2恒温培养箱(BB150-2tcs, 美国Thermo Fisher公司); 共聚焦显微镜(ZEISS Smart proof 5, 德国ZEISS公司)。
细菌培养  将复苏后的VNP划线涂布至含有50 μg mL-1卡那霉素的LB固体培养基中, 挑取单菌落隔夜培养。细菌数量通过测量600 nm处吸光值确定。将对数生长期的VNP调整至所需数量并在无菌PBS中洗涤3次。
细胞培养  使用含10% Gibco血清和1%青链霉素的DMEM高糖培养基在5% CO2恒温培养箱中培养。
ZD@VNP制备与表征  取VNP (4×108 CFU)分散于5 mL无菌PBS中, 加入0.5 mL 2-甲基咪唑(10 mg)混合均匀, 搅拌下滴加0.5 mL硝酸锌和DOX (0.5 mg)的混合溶液, 继续搅拌1 h。硝酸锌用量分别为1、5、10 mg, 将上述反应液经8 000 r·min-1离心10 min后弃去上清液, 并用PBS离心洗涤3遍得到ZD@VNP。将ZD@VNP稀释后经磷钨酸染色, 置于透射电镜下观察。将ZD@VNP置于激光共聚焦显微镜下观察DOX的荧光与VNP的明场重叠情况。
DOX包封率测定  取1 mL ZD@VNP加入葡聚糖凝胶柱中并连接紫外检测器, 收集后段滤过液用于荧光强度检测, 并根据公式(1)计算包封率:

DOX体外释放  取1 mL ZD@VNP置于透析袋中(5 000 kD), 将袋口束紧, 分别置于装有30 mL含200 g·mL-1 Tween 80的PBS (pH 6.0、7.4)的EP管中, 将其置于37 ℃、80 r·  min-1的摇床中。分别于0、2、4、8、12、24、36、48 h时取1 mL释放液, 同时补加等体积新鲜释放介质。取释放液进行荧光强度检测, 计算释放液中DOX的浓度。
CCK-8法考察B16F10细胞增殖抑制作用  取对数生长期的B16F10细胞离心得到细胞悬液, 调整细胞浓度约为每孔1×104个, 接种于96孔板中, 在37 ℃、5% CO2的培养箱中培养过夜。将培养基替换成100 μL含DOX、ZD@VNP的培养基, 同时设置VNP组、空白组和阴性对照组, 每组6个复孔。DOX质量浓度分别为0.5、1.0、2.5、5.0、7.5、10.0 μg·mL-1。在培养箱中孵育24 h后弃培养基, 每孔用100 μL PBS洗涤, 并加入含10 μL CCK-8的新鲜培养基, 于培养箱中继续孵育2 h, 用酶标仪在450 nm处检测各组吸光度, 计算细胞存活率。
细胞活/死染色FDA/PI双染色法考察B16F10细胞增殖抑制作用  取对数生长期的B16F10细胞离心得到细胞悬液, 调整细胞浓度约为每孔2×105个, 接种于12孔板中, 在37 ℃、5% CO2的培养箱中培养过夜。将培养基分别替换成500 μL含DOX、VNP、ZD@VNP的培养基, 同时设置空白对照组。在培养箱中孵育4 h后弃培养基, 每孔用100 μL PBS洗涤, 分别加入50 μL PI溶液和50 μL FDA溶液, 轻轻混匀, 通过激光共聚焦显微镜观察细胞。
ZD@VNP在B16F10肿瘤模型上的抑瘤效果  对C57BL/6小鼠背部右侧进行剃毛处理, 以每只小鼠5.0×105个B16F10细胞皮下接种。待肿瘤生长至50~80 mm3时, 随机分为5组: PBS、VNP、DOX、VNP+DOX、ZD@VNP, 每组设置8只小鼠。每只小鼠尾静脉注射100 μL以上制剂(DOX 2.5 mg·kg-1, VNP 105 CFU/只)记为第0天; 于第2、4、6天分别再次注射上述制剂, 记录小鼠体重和肿瘤大小。
统计学方法  实验结果数据均以$ \overline{x} $ ± s表示。采用one-way ANOVA进行方差分析, P < 0.05为差异有统计学意义。
本文拟利用仿生矿化构建ZIF-8包覆的VNP, 其机制为VNP表面蛋白上的羧基与锌离子具有较高的亲和力, 能形成配位键从而促进ZIF-8在VNP表面的矿化[21]。作者对所制得的ZD@VNP进行透射电镜观察, 并与单独的VNP进行对比。如图 1A所示, VNP长约2 μm; 表面形态清晰可见; 当硝酸锌用量为1 mg时, VNP表面包覆很多细小的纳米粒, 但纳米粒呈聚集状态, 且细菌与细菌之间黏附(图 1B); 当硝酸锌用量为5 mg时, 观察到视野下ZIF-8的粒径约为60 nm, 均匀地包裹在VNP表面, 且未完全遮挡细菌表面结构, 有利于细菌固有性质的发挥(图 1C); 而当硝酸锌用量为10 mg时, VNP表面结合的纳米粒略有增大, 但纳米粒依然是聚集的(图 1D)。因此后续制备ZD@VNP采用10 mg 2-甲基咪唑与5 mg硝酸锌混合的处方。
将ZD@VNP置于激光共聚焦显微镜下, 作者观察到多柔比星的荧光(橙色)几乎与VNP的明场重叠, 证明ZIF-8与VNP的有效结合(图 2)。
利用DOX在激发波长(Ex) = 497 nm、发射波长(Em) = 555 nm的条件下具有最佳吸收, 以荧光强度对浓度绘制标准曲线, 并得到回归直线方程。测得包封率为85%。
图 3可知: pH 7.4的缓冲液中, DOX的释放相对平缓, 48 h内释放率约为40%, 而在模拟肿瘤微环境的pH 6.0的缓冲液下DOX释放明显加快, 24 h释放率达到55%。这可能是由于ZIF-8的pH降解特性使包载的DOX在酸性条件下释放加速导致。
CCK-8结果如图 4所示, DOX和ZD@VNP对B16F10细胞增殖的抑制作用都表现出剂量依赖性, 且ZD@VNP的细胞抑制作用显著强于DOX。当质量浓度为0.5 μg·mL-1时, DOX和ZD@VNP组细胞存活率分别约为60%和45%, 当质量浓度为10 μg·mL-1时, ZD@VNP组细胞存活率约为8%, 而VNP (105 CFU)的细胞存活率约为80%。在各个浓度梯度下, ZD@VNP的细胞存活率皆显著低于DOX组, 说明ZD@VNP增强了DOX的细胞增殖抑制作用。
细胞活/死染色结果如图 5所示, 空白组的细胞呈现大面积的绿色, VNP组有30%细胞呈红色, 证明细菌本身对于B16F10细胞具有一定的侵袭杀伤能力。DOX组有60%细胞呈红色, 而ZD@VNP组呈现大面积的红色, 说明大部分为死细胞。再次证明ZD@VNP具有较好的抑制B16F10增殖的作用。
图 6可知, PBS、VNP、DOX、VNP+DOX、ZD@VNP的中位生存期分别为16、20、20、24、37天, VNP有一定的抑制肿瘤生长的能力但弱于DOX, ZD@VNP抑瘤的能力在各组之间是最强的, 并显著强于VNP+DOX组, 说明ZD@VNP制剂中的DOX和VNP有协同抗肿瘤作用。从小鼠体重变化来看, VNP组小鼠体重无明显变化, 而DOX组则有缓慢下降趋势, 说明VNP的安全性高于DOX。ZD@VNP组小鼠体重在治疗期间无明显变化, 说明ZD@VNP具有较好的生物安全性。
VNP20009因其安全性高、可主动靶向乏氧肿瘤而广泛应用于抗肿瘤研究。然而, 在Ⅰ期临床试验中患者肿瘤并未明显消退[4]。可见, 细菌治疗须联合其他抗肿瘤策略, 才能有效治疗肿瘤。DOX是常用的化疗药物, 但其心脏毒性和体内非特异性分布严重限制了其应用[22]。利用VNP搭载DOX将提高DOX在肿瘤部位的蓄积, 与VNP协同治疗肿瘤, 在药物递送方面有着广阔的发展前景。
本研究利用仿生矿化法, 通过一步法制备了搭载ZIF-8的VNP, 并有效包载化药多柔比星得到ZD@VNP, 该制备方法快速简单、无需使用有机溶剂、安全无毒。在处方筛选过程中, 作者发现2-甲基咪唑与硝酸锌2∶1时, ZIF-8能均匀地包裹在VNP表面, 且ZIF-8粒径约为60 nm。共聚焦结果显示DOX的荧光与VNP的明场重叠, 证实了ZIF-8与VNP的结合作用。文中制备的ZD@VNP对DOX包封率达85%, 且在pH 6.0的条件下能有效释放DOX, 为其在ZD@VNP到达肿瘤组织后释放DOX被肿瘤细胞摄取提供了可能。
细胞实验结果证明ZD@VNP具有较好的抑制B16F10细胞增殖的作用, 进一步的动物实验也表明ZD@VNP具有优异的抑制肿瘤生长并延长小鼠生存期的效果, 且具有较好的生物安全性。
综上所述, 本研究采用一步法制备了搭载ZIF-8的VNP, 同时包载DOX得到ZD@VNP, 其对DOX具有较高的包封率, 且制备过程中未使用有机溶剂, 具有较高的安全性。ZD@VNP增强了对肿瘤细胞增殖抑制作用, 并在动物肿瘤模型上表现出较好的肿瘤抑制效果。文中只进行了肿瘤药效学研究, 在后续的研究中将进一步进行体内抑瘤机制的研究。
