In this study, plasma, urine and fecal samples were collected from rats after intragastric administration of novel insulin sensitizer Zg02 (20 mg·kg-1). The ultra-performance liquid chromatography-quadrupole-time-of-flight-tandem mass spectrometry (UPLC-Q-TOF/MSE) techniques was used to obtain the molecular ion and mass spectrometry fragment ion information of the compound, and the metabolites were quickly analyzed by combining with UNIFI metabolite software. The results showed that a total of 12 metabolites were inferred in rats after a single gavage of Zg02 (20 mg·kg-1), including 5, 7 and 11 metabolites in plasma, urine and feces (including cross-analysis), and the metabolic pathways were mainly glucuronidation and glucosylation. All animal protocols were approved by the Animal Ethics Committee of Guizhou Medical University (No. 2100856).

本研究采用超高效液相色谱-四极杆-飞行时间-串联质谱(UPLC-Q-TOF/MSE) 技术对SD大鼠经灌胃新型胰岛素增敏剂Zg02 (20 mg·kg-1) 后所收集的血浆、尿液及粪便样品进行检测, 获得化合物的分子离子及质谱碎片离子信息, 并结合UNIFI软件, 对代谢产物进行快速分析。结果表明, 单次灌胃Zg02 (20 mg·kg-1) 后在大鼠体内共推测出12个代谢产物, 其中血浆、尿液、粪便中分别有5、7、11个代谢产物(含交叉分析), 代谢途径主要为葡萄糖醛酸化、葡萄糖基化等结合反应。所有动物实验方案均获得贵州医科大学动物伦理委员会批准(编号: 2100856)。

本研究采用超高效液相色谱-四极杆-飞行时间-串联质谱(UPLC-Q-TOF/MSE) 技术对SD大鼠经灌胃新型胰岛素增敏剂Zg02 (20 mg·kg-1) 后所收集的血浆、尿液及粪便样品进行检测, 获得化合物的分子离子及质谱碎片离子信息, 并结合UNIFI软件, 对代谢产物进行快速分析。结果表明, 单次灌胃Zg02 (20 mg·kg-1) 后在大鼠体内共推测出12个代谢产物, 其中血浆、尿液、粪便中分别有5、7、11个代谢产物(含交叉分析), 代谢途径主要为葡萄糖醛酸化、葡萄糖基化等结合反应。所有动物实验方案均获得贵州医科大学动物伦理委员会批准(编号: 2100856)。

In this study, plasma, urine and fecal samples were collected from rats after intragastric administration of novel insulin sensitizer Zg02 (20 mg·kg-1). The ultra-performance liquid chromatography-quadrupole-time-of-flight-tandem mass spectrometry (UPLC-Q-TOF/MSE) techniques was used to obtain the molecular ion and mass spectrometry fragment ion information of the compound, and the metabolites were quickly analyzed by combining with UNIFI metabolite software. The results showed that a total of 12 metabolites were inferred in rats after a single gavage of Zg02 (20 mg·kg-1), including 5, 7 and 11 metabolites in plasma, urine and feces (including cross-analysis), and the metabolic pathways were mainly glucuronidation and glucosylation. All animal protocols were approved by the Animal Ethics Committee of Guizhou Medical University (No. 2100856).

糖尿病(diabetic mellitus, DM), 是一种以高血糖为指征的慢性代谢紊乱性疾病[1-3]。据统计, 全球糖尿病患病率持续增大, 截至2021年, 全球IFG (空腹血糖, 6.1~6.9 mmol·L-1 [110~125 mg·dL-1]) 患病率已达5.8%, 预计2045年将增加到6.5%[4-6]。由于医疗技术的发展和降糖类创新药物的不断发现, 糖尿病的致死率呈下降趋势[7, 8], 其中口服药物治疗是临床上最常用的方法[9, 10], 即口服降糖类药物是创新药物研发的热点之一[11-13]
新型四氢咔唑衍生物Zg02 (图 1), 化学名称为6-苄氧基-9-(4-氯苯甲酰基)-1, 2, 3, 4-四氢咔唑-3-甲酸, 是课题组前期经合成工艺路线优化的具有良好胰岛素增敏活性的候选化合物[14, 15], 可通过激活AMPK信号通路降低小鼠的血糖[16]。并且在前期体外稳定性研究中发现Zg02在体外血浆和人工肠液中均较稳定, 在人工胃液和大鼠肝微粒体中不稳定[17, 18]。此外, Zg02具有半衰期长(6.46 h)、口服生物利用度低(20.2%) 的特点[19]。本研究建立UPLC-Q-TOF/MSE分析方法, 检测Zg02在SD大鼠体内的代谢产物, 获得化合物的分子离子及质谱碎片离子信息, 初步确定了Zg02的代谢规律及相关代谢产物的结构, 为接下来的成药性筛选奠定了基础, 进一步推进候选化合物药物开发的进程。
仪器  UPLC-Q-TOF (型号Xevo G2-XS, 美国Waters公司); 代谢笼(意大利Tecniplast公司); 氮气吹扫仪(型号JN300-2, 苏州吉米诺仪器有限公司); 冷冻高速离心机(型号1730R) 和高速离心机(型号X1), 香港基因有限公司; 超声波清洗器(型号KH-600E, 昆山禾创超声仪器有限公司); 十万分之一电子天平(型号FA805N, 上海菁海仪器有限公司); 超低温保存箱(型号: DW-86L486, 海尔有限公司) 等。
药品试剂  Zg02原料药(贵州医科大学药物化学重点实验室制备, 纯度 > 98%); 吲哚美辛(大连美伦生物技术有限公司, 批号J0526A5, 纯度 > 98%); 甲酸(CNW Techologies, HPLC级, 批号X075K150), 甲醇、乙腈(德国默克股份两合公司, 色谱纯); 水为屈臣氏蒸馏水; OASIS HLB固相萃取(SPE) 柱(美国Waters公司)。
实验动物  清洁级SD大鼠6只, 雌雄各半, 体质量180~240 g, 鼠龄: 8~10周, 大鼠购自贵州医科大学, 合格证号: SYXK (黔) 2018-0001。动物于动物房中饲养一周以适应环境。所有动物实验方案均获得贵州医科大学动物伦理委员会批准(编号: 2100856)。
生物样品处理方法
血浆样品的处理  取混合各时间点的大鼠血浆0.5 mL, 置于2.0 mL EP管中, 加入1 mL乙腈沉淀蛋白, 涡混震荡10 min, 于4 ℃下, 13 000 r·min-1离心10 min, 取上清液, 于37 ℃氮气下吹干。加入200 µL 50%甲醇水溶液(1∶1, v/v) 复溶样品, 涡混震荡10 min, 4 ℃下13 000 r·min-1离心10 min, 取上清液UPLC-Q-TOF/MSE进样分析。
尿液样品处理  取HLB固相萃取小柱, 加入1 mL甲醇, 再加1 mL水进行活化小柱后备用。将大鼠混合尿液1 mL上样, 依次用4倍体积水和4倍体积甲醇进行洗脱, 收集甲醇洗脱液。将其置于37 ℃氮气下吹干, 残留物用600 µL 50% 甲醇水溶液(1∶1) 复溶, 涡混震荡10 min, 4 ℃下13 000 r·min-1离心10 min, 取上清液UPLC-Q-TOF/MSE进样分析。
粪便样品处理  收集的粪便低温烘干后, 称取粪便样本适量, 用70%的甲醇水溶液(7∶3) 以1∶10 (w/v) 的比例匀化, 制备成10%的匀浆, 超声5 min, 13 000 r·min-1离心l0 min后, 取上清液1 mL上样, 取HLB固相萃取小柱(小柱预先活化平衡好备用), 依次用4倍体积水和4倍体积甲醇进行洗脱, 收集甲醇洗脱液。将其置于37 ℃氮气下吹干, 残留物用600 µL 50%甲醇水溶液复溶, 4 ℃下13 000 r·min-1离心10 min, 取上层清液UPLC-Q-TOF/MSE进样分析。
液相条件  色谱柱: Waters BEH C18 (50 mm × 2.1 mm, 1.7 μm) 柱; 流速0.40 mL·min-1; 进样量: 2 μL; 柱温: 40 ℃; 流动相: 0.01%甲酸水(A)-0.01%甲酸乙腈(B), 梯度洗脱程序: 0~2 min, 5% B; 2~6 min, 5%~60% B; 6~10 min, 60%~98% B; 10~12 min, 98% B; 12~15 min, 98%~5% B。
质谱条件  电喷雾电离源(ESI); 正模式采集(positive mode): 毛细管电压2.0 kV, 负模式采集(negative mode): 毛细管电压1.5 kV; 离子源120 ℃, 溶剂气温度400 ℃, 脱溶剂气流量800 L·h-1, 锥孔气流量50 L·h-1; 质谱数据采集及处理软件MassLynx V4.1工作站, 扫描方式MSE Continuum模式。
通过UPLC-Q-TOF/MSE对各生物样品进行检测, MSE采集方法可在一个质谱周期内进行高、低碰撞能量全扫描, 从而简便、快速地获得组分的母离子及子离子信息, 根据得到的质谱数据结合UNIFI代谢产物预测、筛查软件, 对Zg02的代谢产物进行快速分析。在UNIFI软件中, 将给药后的样品图谱扣除空白基质图谱后, 通过原形化合物Zg02建立的专属数据库显示出样品中特有质谱信息(图 2), 在灌胃给药Zg02的大鼠血浆、尿液、粪便样品中检测出12个代谢产物, 其正、负模式下的基峰强度(base peak intensity, BPI) 色谱图分别见图 34。各代谢产物的保留时间、结构组成、碎片离子、计算m/z等见表 1, 根据代谢产物结构推测其代谢途径, 如图 5, Zg02由原型先转化为M1、M2、M3及M7, 再以M3转化为M4、M5及M6, 以M7转化为M8、M9、M10、M11及M12。图 6为主要代谢产物高、低能量质谱图, 结合图 2可分析佐证各代谢产物结构。
原形化合物Zg02在负模式扫描条件下保留时间为6.25 min, 在低能量扫描通道里存在m/z 458.117 2 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 186.092 3、323.073 0、414.126 6。
M1在负模式扫描条件下保留时间为4.62 min, 在低能量扫描通道存在m/z 622.182 5 [M+H]+的准分子离子峰, 高能量通道可见碎片离子峰m/z 93.062 8、278.157 8、322.148 2、416.140 3。根据以上准分子离子及碎片离子的结构, 可推测M1由原型化合物直接转化, 结构相当于Zg02+C6H10O5
M2在负模式扫描条件下保留时间为4.52 min, 在低能量扫描通道存在m/z 634.176 2 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 138.001 1、320.128 7、414.128 0、458.117 2。根据以上准分子离子及碎片离子的结构, 可推测M2也由原型化合物直接转化, 结构相当于Zg02+C6H8O6
M3在负模式扫描条件下保留时间为5.82 min, 在低能量扫描通道存在m/z 368.070 5 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 137.988 3、230.081 7、324.074 5。根据以上准分子离子及碎片离子的结构, 可推测M3也由原型化合物直接转化, 结构相当于Zg02-C7H6
M7在负模式扫描条件下保留时间为5.23 min, 在低能量扫描通道存在m/z 320.127 2 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 230.081 5、276.138 2。准分子离子峰m/z 320.127 2 [M-H]-推测为Zg02结构中酰胺键断裂, 再结合其碎片离子的结构, 可推测出M7的结构为Zg02-C7H3ClO。
M4在负模式扫描条件下保留时间为5.96 min, 在低能量扫描通道存在m/z 382.083 2 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 137.987 6、244.097 4、338.094 4。准分子离子峰m/z 382.083 2 [M-H]-比M3多14 Da初步推测为: M3 (Zg02-C7H6)+CH2。M3结构中酚羟基或羧酸均容易发生甲基化的结合反应, 再根据以上准分子离子及碎片离子的结构, 可以推断M4是由M3的酚羟基甲基化后形成的。
M5在负模式扫描条件下保留时间为4.16 min, 在低能量扫描通道存在m/z 532.139 5 [M+H]+的准分子离子峰, 高能量通道可见碎片离子峰m/z 139.992 3、370.085 2、394.151 2。根据以上准分子离子及碎片离子的结构, 仅能初步推断M5的结构为M3 (Zg02-C7H6)+C6H10O5
M6在负模式扫描条件下保留时间为4.03 min, 在低能量扫描通道存在m/z 544.103 5 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 368.068 2、406.114 2。碎片离子峰m/z 368.068 2 [M-H]-推测为中性丢失葡萄糖醛酸176 Da的M3, 再结合准分子离子和其余碎片离子的结构, 可推测M6的结构为M3 (Zg02-C7H6)+C6H8O6
M8在负模式扫描条件下保留时间为5.42 min, 在低能量扫描通道存在m/z 362.138 0 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 91.054 8、272.092 3。根据以上准分子离子及碎片离子的结构, 可推测M8的结构为M7 (Zg02-C7H3ClO)+C2H2O。
M9在正模式扫描条件下保留时间为3.56 min, 在低能量扫描通道存在m/z 484.197 8 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 93.063 3、322.144 3、394.153 3。根据以上准分子离子及碎片离子的结构, 可推测M9的结构为M7 (Zg02-C7H3ClO)+C6H10O5
M10在负模式扫描条件下保留时间为5.28 min, 在低能量扫描通道存在m/z 322.145 2 [M-H]-的准分子离子峰, 高能量通道可见碎片离子峰m/z 91.054 4、232.097 2、278.154 9。根据以上准分子离子及碎片离子的结构, 可推测M10的结构为M7 (Zg02-C7H3ClO)+H2
M11在正模式扫描条件下保留时间为5.63 min, 在低能量扫描通道存在m/z 360.123 8 [M+H]+的准分子离子峰, 高能量通道可见碎片离子峰m/z 228.066 7、270.088 3、318.122 6。根据以上准分子离子及碎片离子的结构, 可推测M11的结构为M7 (Zg02-C7H3ClO)-2(H2)+C2H2O。
M12在正模式扫描条件下保留时间为3.43 min, 在低能量扫描通道存在m/z 522.174 4 [M+H]+的准分子离子峰, 高能量通道可见碎片离子峰m/z 228.065 2、270.087 5。准分子离子峰m/z 522.174 4 [M+H]+比M11多162 Da, 推测M12为M11葡萄糖基化后的代谢产物, 并且碎片离子峰m/z 228.065 2、270.087 5与M11中高能量通道的碎片离子峰的相似, 最终推测M12的结构为M11 (Zg02-C7H3ClO)-2(H2)+C2H2O+C6H10O5
使用UPLC-Q/TOF分析代谢产物的过程中, 根据不同结构的同位素峰的相对丰度的不同可推断未知化合物的结构, 特别是含天然核素(Cl-和Br-等) 的化合物容易通过同位素峰的丰度来判断。通常情况下含Br的化合物M+2峰的相对丰度与M峰的相对丰度一致, 含Cl的M+2峰的相对丰度是M峰的相对丰度的1/3 (此处的M是分子离子, M+1和M+2是核素离子)。分析Zg02及其代谢产物结构时, 能区别出离子中含Cl峰的M+2峰与不含Cl峰的M+1 (或M+2) 峰, 增加了代谢产物推断的可靠性。
化合物结构在质谱检测时断裂规律的推断可以使用ChemBioDraw软件来辅助验证[20]
检测代谢产物的采集方式与流程: MSE采集方法可以在一个质谱周期内, 通过高、低碰撞能量全扫描, 从而简便、快速地获得组分的母离子及子离子信息, 但其高能量采集时是没有针对性的全扫描谱图的, 对同一出峰时间的所有物质均在高能量打碎, 造成了提供的碎片离子信息与单独MS/MS采集的会有所不同, 以MSE采集方法得到的高能量图谱里面同一时间出峰的所有物质的碎片离子信息, 分析代谢产物时提升了推断代谢物碎片断裂信息的难度, 运用Waters公司开发的UNIFI软件可降低其分析难度, 该软件可以通过预先设定的原形化合物结构及原形化合物结构产生的碎片信息, 预测出代谢产物及产物断裂的碎片离子。所以初步推测受试化合物的代谢产物时, 可以采用MSE采集方法结合相应分析软件快速地获得相关信息, 在进一步研究时, 通过预测的代谢物有针对性地进行MS/MS采集(MS/MS中轰击的能量可以参考MSE采集得到的信息来调整)。通过代谢物的MS/MS信息来分析佐证前期MSE全扫描时的推断。
由于UPLC-Q-TOF/MSE检测的相关代谢产物并没有通过对照品(购买标准品或合成出相应化合物) 进样确认, 或采取一定手段分离出代谢产物对其进行UV、1H NMR、13C NMR检测以确证结构, 故对于单次灌胃给药Zg02后大鼠体内代谢产物的推断, 仅做了初步的分析推断。目前在开发候选化合物Zg02初期阶段, 后续可通过综合受试化合物其他研究的评价, 确定可以进一步深入开发该受试化合物后, 在具体确证其受试化合物的代谢产物, 代谢产物与原形化合物在体内含量的变化情况, 以期找出药效结构和毒副反应物质。
综上所述, 选用健康SD大鼠, 收集其血浆、粪便、尿液样品, 考察Zg02在大鼠体内代谢规律, 建立UPLC-Q-TOF/MSE对生物样品血浆、尿液、粪便的高分辨质谱检测方法, 15 min可完成对复杂生物样品的检测, 得到代谢产物的质谱数据。推断出Zg02单次灌胃给药(20 mg·kg-1) 后体内的12个代谢产物, 多数为Zg02原形结构和断裂结构后与葡萄糖醛酸化、葡萄糖基化结合反应, 同时对前期课题研究中体外肝微粒体代谢稳定性的结果相应[19], 推测受试物Zg02的代谢主要为结合反应。
