A new alkaloid (1) and six known compounds (2-7) were isolated from the solid fermentation extract of the endophytic fungus Alternaria tenuissima Pas85 of Phragmites australis by silica gel column chromatography, Sephadex LH-20 column chromatography, high performance liquid chromatography and other chromatographic techniques. The structures of these compounds were determined by HREIMS, NMR, IR spectroscopy, and literature comparison, and the anti MRSA (methicillin-resistant Staphylococcus aureus) activity of 1-5 were tested. Compounds 1, 2, 3 and 4 exhibited certain inhibitory activity with MIC values of 64, 4, 32 and 32 μg·mL-1, respectively, and compound 5 showed no significant inhibitory activity.

采用硅胶柱色谱、葡聚糖凝胶Sephadex LH-20柱色谱、高效液相色谱等多种色谱技术, 从芦苇内生真菌Alternaria tenuissima Pas85固体发酵提取物中分离得到1个新的生物碱类化合物(1) 和6个已知化合物(2~7)。通过高分辨质谱、核磁共振和红外等波谱学方法及文献比对确定其结构, 并测试了化合物1~5的抗MRSA (methicillin-resistant Staphylococcus aureus) 活性。化合物1~4表现出一定的抑制活性, MIC值分别为64、4、32、32 μg·mL-1, 化合物5无明显抑制活性。

采用硅胶柱色谱、葡聚糖凝胶Sephadex LH-20柱色谱、高效液相色谱等多种色谱技术, 从芦苇内生真菌Alternaria tenuissima Pas85固体发酵提取物中分离得到1个新的生物碱类化合物(1) 和6个已知化合物(2~7)。通过高分辨质谱、核磁共振和红外等波谱学方法及文献比对确定其结构, 并测试了化合物1~5的抗MRSA (methicillin-resistant Staphylococcus aureus) 活性。化合物1~4表现出一定的抑制活性, MIC值分别为64、4、32、32 μg·mL-1, 化合物5无明显抑制活性。

A new alkaloid (1) and six known compounds (2-7) were isolated from the solid fermentation extract of the endophytic fungus Alternaria tenuissima Pas85 of Phragmites australis by silica gel column chromatography, Sephadex LH-20 column chromatography, high performance liquid chromatography and other chromatographic techniques. The structures of these compounds were determined by HREIMS, NMR, IR spectroscopy, and literature comparison, and the anti MRSA (methicillin-resistant Staphylococcus aureus) activity of 1-5 were tested. Compounds 1, 2, 3 and 4 exhibited certain inhibitory activity with MIC values of 64, 4, 32 and 32 μg·mL-1, respectively, and compound 5 showed no significant inhibitory activity.

链格孢菌(Alternaria spp) 是一种在自然界许多生态系统中普遍存在的暗色真菌, 其生存方式通常为腐生和内生[1, 2]。广泛存在于植物组织中的内生链格孢菌, 长期与宿主植物协同进化, 形成互惠互利的共生关系。链格孢菌能够产生多种次生代谢产物, 如吡喃酮、醌类、类固醇、萜类及生物碱等, 其中一些化合物具有抗炎、抗病毒、抗菌或细胞毒等生物活性[3-5]。进一步的研究表明, 内生真菌通过产生这些抗病毒、抗真菌和抗菌活性的次生代谢物来保护其寄主植物[6, 7]。近年来, 链格孢菌因其能够产生结构新颖、活性多样的次生代谢产物, 已成为微生物和天然产物领域研究的热点[8]
芦苇(Phragmites australis) 为禾本科芦苇属草本植物, 是中国传统中药“苇茎”和“芦根”的母本植株, 具有清肺化痰和清热利尿的功效[9, 10]Alternaria tenuissima Pas85是从芦苇健康的叶片组织中分离的一株内生真菌。前期活性导向的高通量筛选研究发现, Alternaria tenuissima Pas85的发酵提取物对MRSA的生长具有一定的抑制活性。本次实验对其固体发酵提取物进行了化学成分研究, 共分离得到7个化学成分(图 1), 其中1是一个新化合物(被命名为1-[5-(methoxymethyl)-1H-pyrrol-2-yl] ethan-1-one), 2~7是已知化合物, 分别为griseophenone C[11] (2)、6-chloro-3, 8-dihydroxy-1-methylxanthone[12] (3)、norlichexanthone[13] (4)、griseoxanthone C[14] (5)、diaporthin[15] (6)、(-)-orthosporin[16] (7)。这些化合物均为首次从芦苇内生真菌中分离发现。本文对新化合物1的结构进行了解析, 并测试了化合物1~5的抗MRSA活性。
化合物1为淡黄色油状物。高分辨质谱HREIMS显示出其分子量m/z 153.079 2 [M]+, 结合NMR数据(表 1) 确定分子式为C8H11NO2, 不饱和度为4。从IR图谱可以看到3 414、1 716和1 639 cm-1处有强吸收峰, 初步推测该化合物中含有-NH和羰基。1H NMR显示低场部分有一个活泼氢质子[δH 10.57 (1H, br s)], 两个芳香质子[δH 7.56 (1H, d, J = 3.3 Hz), 6.80 (1H, d, J = 3.3 Hz)]; 高场部分有一个单峰的氧化亚甲基[δH 4.59 (2H, s)], 一个甲氧基[δH 3.34 (3H, s)] 和一个乙酰基的质子信号[δH 2.32 (3H, s)]。13C NMR和DEPT图谱显示该分子共有8个碳, 其中有三个季碳(包括一个羰基δC 193.4, 两个不饱和季碳δC 123.7, 122.9), 两个不饱和次甲基(δC 126.4, 119.2), 一个氧取代亚甲基(δC 68.9), 一个甲氧基(δC 58.1), 一个甲基(δC 27.5)。结合不饱和度, 提示该化合物含有一个羰基、两个双键、一个甲氧基及一个环状结构。在紫外图谱中显示该化合物的一个最大吸收峰247 nm, 推测该化合物具有共轭双键结构。仔细对比发现, 该化合物NMR数据与化合物methyl 5-acetyl-1H-pyrrole-2-carboxylate[17]极其相似, 表明两者具有相似的结构, 而主要的不同之处在于该化合物多一个氧取代的亚甲基(δC 68.9/δH 4.59), 少一个酯羰基(δC 161.5)。在HMBC图谱中(图 2), H-4′ (δH 6.80)、6′-OMe (δH 3.34) 与δC 68.9均相关, 表明δC 68.9即是C-6′的碳信号。该化合物为methyl 5-acetyl-1H-pyrrole-2-carboxylate C-6′的羰基被还原成亚甲基产物。至此, 该化合物的结构得以确定, 并命名为1-[5-(methoxymethyl)-1H-pyrrol-2-yl]ethan-1-one。
对部分化合物进行抗MRSA活性测试, 结果见表 2。从表中数据可以看出目标化合物对MRSA具有不同程度的抑制活性, 化合物1~4均表现出一定的抑制活性, MIC范围为4~64 μg·mL-1, 其中化合物2具有很强的抗MRSA活性, MIC为4 μg·mL-1, 可作为抗MRSA先导化合物进行下一步研究; 新化合物1及化合物34对MRSA表现出温和的抑制活性, MIC分别为64、32、32 μg·mL-1, 化合物5并未表现出显著的抗MRSA活性(MIC > 128 μg·mL-1)。
JASCO V-650紫外可见分光光度仪(日本佳司科公司); Shimadzu IRTracer-100红外光谱仪(日本岛津公司); Rhdolph Autopol Ⅵ全自动旋光测定仪(美国鲁道夫公司); Themo Q Exactive Focus组合型四级杆Orbitrap高分辨质谱仪(美国热电公司); Bruker AVANCE (600 MHz) 核磁共振仪(德国布鲁克公司); Agilent 1260半制备高效液相色谱仪, 液相色谱柱(Agilent Zorbax SB-C18, 4.6/9.4 mm×250 mm, 5 μm) (美国安捷伦科技有限公司); ReadMax 1500型光吸收全波长酶标仪(上海闪谱生物科技有限公司); ZQZY-CG型组合式光照振荡培养箱(上海知楚仪器公司); 紫外灯ZF-1S (上海恩谊公司) 等。柱色谱硅胶(200~300目) 及薄层层析硅胶GF254 (10~40 μm, 青岛海洋化工有限公司); Sephadex LH-20 (美国通用电气公司); 分析纯石油醚、氯仿、乙酸乙酯、丙酮、甲醇和色谱甲醇、乙腈(天津科密欧试剂公司); 薄层色谱显色剂(8%硫酸-乙醇溶液)。
实验用芦苇(Phragmites australis) 采摘于河南省新郑市轩辕湖湿地公园, 菌株是从健康芦苇叶中分离得到的优势菌株, 送样测序后得到的18S rRNA基因序列在NCBI的GenBank基因库进行BLAST比对, 鉴定为极细链格孢菌(GenBank Accession No.: PP914110), 菌株保存于河南省水环境与健康工程技术研究中心。Alternaria tenuissima Pas85在大米培养基的发酵袋中, 30 ℃恒温培养1.5个月, 得到含次生代谢物的培养物。培养物经乙酸乙酯萃取3次、减压蒸馏去除溶剂后得粗浸膏48 g。
粗浸膏经硅胶柱色谱粗分离, 石油醚-丙酮进行梯度洗脱, 得到5段流分, 分别为Fr.1 (18 g), Fr.2 (6 g), Fr.3 (5 g), Fr.4 (4 g), Fr.5 (10 g)。Fr.2以石油醚-氯仿进行梯度洗脱得Fr.2.1~2.5, Fr.2.2经Sephadex LH-20凝胶柱色谱(氯仿-甲醇1∶1) 洗脱, 得到化合物1 (2.0 mg) 和6 (8.8 mg)。Fr.3以石油醚-乙酸乙酯进行梯度洗脱得Fr.3.1~3.4, Fr.3.2经HPLC (RP-18半制备柱, 9.2 mm × 250 mm, 5 μm; 75%乙腈-水, 流速3 mL·min-1) 分离纯化得到化合物3 (4.1 mg, tR = 5 min) 和5 (7.5 mg, tR = 8 min)。Fr.3.4经Sephadex LH-20凝胶柱色谱分离, 再经HPLC (RP-18半制备柱, 9.2 mm × 250 mm, 5 μm; 68%乙腈-水, 流速3 mL·min-1) 分离纯化得到化合物2 (8.0 mg, tR = 7 min) 和4 (3.9 mg, tR = 10 min)。Fr.4以石油醚-乙酸乙酯(1∶1) 等度洗脱后, 经Sephadex LH-20凝胶柱色谱分离, 再制备薄层色谱(氯仿-甲醇10∶1) 纯化得到化合物7 (23.5 mg)。
化合物1  淡黄色油状物, 易溶于甲醇、丙酮、氯仿。UV (MeOH) λmax (log ε) 203 (4.70), 247 (3.93) nm; IR (KBr) νmax 3 414, 2 953, 2 924, 1 716, 1 639, 1 521, 1 452, 1 407, 1 083, 950, 608 cm-1; 1H和13C NMR数据见表 1; HREIMS: m/z 153.079 2 [M]+ (C8H11NO2, 计算值153.079 0)。
化合物2  无色油状物, 易溶于甲醇, ESI-MS m/z 327 [M+Na]+, 分子式为C16H16O61H NMR (600 MHz, CD3OD) δH: 6.41 (1H, br s, H-5), 6.38 (1H, br s, H-3), 5.98 (1H, s, H-3′), 5.98 (1H, s, H-5′), 3.82 (3H, 4′-OMe), 3.67 (3H, s, 2-OMe), 2.12 (3H, s, Me-8); 13C NMR (150 MHz, CD3OD) δC: 196.6 (C-7), 167.6 (C-4′), 164.6 (C-2′), 164.6 (C-6′), 159.6 (C-4), 158.3 (C-2), 138.3 (C-6), 120.4 (C-1), 110.6 (C-5), 98.1 (C-3), 94.8 (C-3′), 94.8 (C-5′), 56.3 (2-OMe), 55.8 (4′-OMe), 19.1 (C-8)。以上数据与文献[11]对比, 确定化合物2为griseophenone C。
化合物3  白色固体粉末, 易溶于甲醇, ESI-MS m/z 299 [M+Na]+, 分子式为C14H9O4Cl。1H NMR (600 MHz, CD3OD) δH: 6.51 (1H, d, J = 2.2 Hz, H-2), 6.49 (1H, d, J = 2.2 Hz, H-4), 6.15 (1H, d, J = 2.1 Hz, H-5), 6.05 (1H, d, J = 2.1 Hz, H-7), 2.75 (3H, s, Me-10); 13C NMR (150 MHz, CD3OD) δC: 183.2 (C-9), 164.9 (C-3), 164.8 (C-8), 161.2 (C-4a), 158.8 (C-5a), 144.2 (C-1), 118.6 (C-2), 111.6 (C-1a), 103.4 (C-6), 103.4 (C-8a), 102.2 (C-4), 99.4 (C-7), 94.8 (C-5), 23.7 (C-10)。以上数据与文献[12]对比, 确定化合物3为6-chloro-3, 8-dihydroxy-1-methylxanthone。
化合物4  白色固体粉末, 易溶于甲醇, ESI-MS m/z 259 [M+H]+, 分子式为C14H10O51H NMR (600 MHz, CD3OD) δH: 6.58 (1H, br s, H-2), 6.58 (1H, br s, H-4), 6.22 (1H, d, J = 2.0 Hz, H-5), 6.11 (1H, d, J = 2.0 Hz, H-7), 2.76 (3H, s, Me-10); 13C NMR (150 MHz, CD3OD) δC: 183.4 (C-9), 165.4 (C-6), 164.9 (C-8), 164.2 (C-3), 161.0 (C-4a), 158.6 (C-5a), 144.7 (C-1), 117.6 (C-2), 112.6 (C-1a), 103.8 (C-8a), 101.9 (C-4), 99.4 (C-7), 94.8 (C-5), 23.7 (C-10)。以上数据与文献[13]对比, 确定化合物4为norlichexanthone。
化合物5  无色晶体, 易溶于丙酮, ESI-MS m/z 295 [M+Na]+, 分子式为C15H12O51H NMR (600 MHz, acetone-d6) δH: 13.38 (1H, s, 8-OH), 9.60 (1H, br s, 3-OH), 6.65 (1H, br s, H-2), 6.65 (1H, br s, H-4), 6.28 (1H, d, J = 2.0 Hz, H-5), 6.16 (1H, d, J = 2.0 Hz, H-7), 3.90 (3H, s, 6-OMe), 2.75 (3H, s, Me-10); 13C NMR (150 MHz, acetone-d6) δC: 183.1 (C-9), 165.5 (C-6), 164.9 (C-8), 163.6 (C-3), 160.4 (C-4a), 158.2 (C-5a), 144.3 (C-1), 116.9 (C-2), 112.8 (C-1a), 103.9 (C-8a), 101.6 (C-4), 98.8 (C-7), 94.1 (C-5), 57.4 (6-OMe), 23.5 (C-10)。以上数据与文献[14]对比, 确定化合物5为griseoxanthone C。
化合物6  无色晶体, 易溶于氯仿, ESI-MS m/z 273 [M+Na]+, 分子式为C13H14O51H NMR (600 MHz, CDCl3) δH: 10.95 (1H, s, 8-OH), 6.40 (1H, d, J = 2.2 Hz, H-7), 6.29 (1H, s, H-4), 6.26 (1H, m, H-5), 4.25 (1H, m, H-10), 3.85 (3H, s, 6-OMe), 2.60 (2H, m, H-9), 1.30 (3H, d, J = 5.5 Hz, Me-11); 13C NMR (150 MHz, CDCl3) δC: 166.9 (C-1), 166.4 (C-6), 163.5 (C-8), 154.8 (C-3), 139.1 (C-4a), 106.2 (C-4), 104.4 (C-5), 103.6 (C-7), 99.8 (C-8a), 65.5 (C-10), 55.8 (6-OMe), 43.1 (C-9), 23.3 (C-11)。以上数据与文献[15]对比, 确定化合物6为diaporthin。
化合物7  无色晶体, 易溶于甲醇, ESI-MS m/z 259 [M+Na]+, 分子式为C12H12O51H NMR (600 MHz, CD3OD) δH: 6.32 (1H, br s, H-7), 6.24 (2H, m, H-4, H-5), 4.14 (1H, m, H-10), 2.58 (2H, d, J = 6.3 Hz, H-9), 1.25 (3H, d, J = 6.5 Hz, Me-11); 13C NMR (150 MHz, CD3OD) δC: 169.9 (C-1), 168.2 (C-6), 165.6 (C-8), 155.9 (C-3), 141.4 (C-4a), 107.5 (C-4), 104.6 (C-5), 103.8 (C-7), 99.2 (C-8a), 66.5 (C-10), 43.9 (C-9), 23.6 (C-11)。以上数据与文献[16]对比, 确定化合物7为(-)-orthosporin。
MRSA由郑州大学第一附属医院分离提供。以万古霉素(vancomycin) 为对照药, 不加样品的DMSO作空白对照, 依据参照文献[18]采用两倍稀释法测定目标化合物的MIC。将各单体化合物分别溶解在DMSO中, 初始浓度为1 024 μg·mL-1的溶液。指示菌接种于LB培养基, 37 ℃下振荡培养至对数期, 调节菌悬液106 CFU·mL-1。96孔板的第1行加入LB液体培养基190 µL和样品10 µL, 第2至第9行每孔加入LB液体培养基100 µL。第1行混合均匀后, 取培养液100 µL到第2行, 倍比稀释至第8行。第1~8行每孔加入100 µL菌液, 单体化合物的终质量浓度依次为256、128、64、32、16、8、4、2 μg·mL-1, 每个浓度设3复孔, 实验重复3次。37 ℃恒温培养18~24 h观察实验结果, 相应澄清孔的最小浓度值为化合物的MIC值。
