Five compounds were isolated from the ethyl acetate fraction of Semen Persicae by using various chromatographic methods, including ODS, Sephadex LH-20, HPLC and semipreparative HPLC. Their structures were identified by 1D-NMR, 2D-NMR, HR-ESI-MS, UV, IR, circular dichroism (CD) and ECD calculation techniques: (2R, 3R)-5, 7, 4′-trihydroxy-3′-methoxy-3-formylflavan-3-ol-5-O-β-D-glucopyranoside (1), (7R, 8S)-dihydrodehydrodiconiferyl 6″-benzoyl alcohol-9-O-β-D-glucopyranoside (2), (7R, 8S)-dihydrodehydrodiconiferyl alcohol-9-β-O-D-glucopyranosid (3), 2-methoxy-4-(2-propenyl)-phenyl-O-β-D-glucopyranoside (4), 2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-propane-1, 3-diol (5). Compound 1 and 2 are new compounds, and compounds 3-5 were obtained from Prunus davidiana (Carr.) Franch. for the first time.

运用ODS、Sephadex LH-20柱色谱, 结合HPLC、半制备高效液相色谱方法从山桃仁乙酸乙酯部位中分离得到5个化合物。结合1D-NMR、2D-NMR、HR-ESI-MS、UV、IR、圆二色谱(CD) 和计算ECD等方法, 鉴定其分别为(2R, 3R)-5, 7, 4′-trihydroxy-3′-methoxy-3-formylflavan-3-ol-5-O-β-D-glucopyranoside (1)、(7R, 8S)-dihydrodehydrodiconiferyl 6″-benzoyl alcohol-9-O-β-D-glucopyranoside (2)、(7R, 8S)-dihydrodehydrodiconiferyl alcohol-9-O-β-D-glucopyranosid (3)、2-methoxy-4-(2-propenyl)-phenyl-O-β-D-glucopyranoside (4)、2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-propane-1, 3-diol (5)。其中, 化合物12为新化合物, 化合物3~5为首次从该植物中分离得到。

运用ODS、Sephadex LH-20柱色谱, 结合HPLC、半制备高效液相色谱方法从山桃仁乙酸乙酯部位中分离得到5个化合物。结合1D-NMR、2D-NMR、HR-ESI-MS、UV、IR、圆二色谱(CD) 和计算ECD等方法, 鉴定其分别为(2R, 3R)-5, 7, 4′-trihydroxy-3′-methoxy-3-formylflavan-3-ol-5-O-β-D-glucopyranoside (1)、(7R, 8S)-dihydrodehydrodiconiferyl 6″-benzoyl alcohol-9-O-β-D-glucopyranoside (2)、(7R, 8S)-dihydrodehydrodiconiferyl alcohol-9-O-β-D-glucopyranosid (3)、2-methoxy-4-(2-propenyl)-phenyl-O-β-D-glucopyranoside (4)、2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-propane-1, 3-diol (5)。其中, 化合物12为新化合物, 化合物3~5为首次从该植物中分离得到。

Five compounds were isolated from the ethyl acetate fraction of Semen Persicae by using various chromatographic methods, including ODS, Sephadex LH-20, HPLC and semipreparative HPLC. Their structures were identified by 1D-NMR, 2D-NMR, HR-ESI-MS, UV, IR, circular dichroism (CD) and ECD calculation techniques: (2R, 3R)-5, 7, 4′-trihydroxy-3′-methoxy-3-formylflavan-3-ol-5-O-β-D-glucopyranoside (1), (7R, 8S)-dihydrodehydrodiconiferyl 6″-benzoyl alcohol-9-O-β-D-glucopyranoside (2), (7R, 8S)-dihydrodehydrodiconiferyl alcohol-9-β-O-D-glucopyranosid (3), 2-methoxy-4-(2-propenyl)-phenyl-O-β-D-glucopyranoside (4), 2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-propane-1, 3-diol (5). Compound 1 and 2 are new compounds, and compounds 3-5 were obtained from Prunus davidiana (Carr.) Franch. for the first time.

桃仁为蔷薇科植物桃Prunus persica (L.) Batsch或山桃Prunus davidiana (Carr.) Franch.的干燥成熟种子[1]。桃仁始载于《神农本草经》, 味苦、甘, 性平, 归肝、心、大肠经; 具有活血化瘀、润肠通便等功效; 可主治痛经、血滞经闭、产后瘀滞腹痛、跌打损伤和瘀血肿痛等病症[2]。桃仁主要含有芳香苷类、黄酮类、甾醇类和萜类等[3]成分。现代药理学研究表明, 桃仁粗提物可减轻血管及心脏损伤[4, 5]、抑制血管内皮细胞凋亡[6]、抑制血小板聚集[7]和抑制血栓形成[8]。桃仁中的单体化合物如棕榈酸、油酸可延长凝血酶时间[7], 苦杏仁苷有潜在的神经保护作用[9]。本实验以山桃仁为研究对象, 对其乙酸乙酯部位化学成分进行分离纯化, 本文报道了5个单体化合物, 包括1个黄酮苷类化合物(1)、2个木脂素苷类化合物(23) 及2个苯丙素类化合物(45), 结构见图 1。化合物12为新化合物, 化合物3~5均为首次从桃仁中分离得到。
化合物1为黄色无定形粉末。[α]-87.6 (c 0.1 MeOH), UV (MeOH) λmax (log ε): 207.0 (3.18)、227.0 (2.89)、279.0 (2.22) nm。HR-ESI-MS [M+Na]+ m/z593.163 6 (C29H30O12Na计算值593.162 9), 提示化合物1分子式为C29H30O12, 不饱和度为15。IR显示该结构中含有羟基(3 361 cm-1)和酯羰基(1 699 cm-1)特征吸收峰。在1H NMR (CD3OD, 500 MHz) 谱(表 1) 中, 显示一组ABX耦合系统的氢信号[δH 7.05 (1H, d, J = 1.8 Hz, H-2′), 6.89 (1H, dd, J = 8.2, 1.8 Hz, H-6′), 6.72 (1H, d, J = 8.2 Hz, H-5′)]; 一组AB自旋系统氢信号[δH 6.33 (1H, d, J = 2.3 Hz, H-8), 6.18 (1H, d, J = 2.3 Hz, H-6)]; 1个亚甲基氢信号[δH 3.17 (1H, dd, J = 18.0, 2.1 Hz, H-4a), 3.11 (1H, dd, J = 18.0, 4.5 Hz, H-4b)]; 2个次甲基氢信号[δH 5.70 (1H, dd, J= 3.4, 2.4 Hz, H-3), 5.12 (1H, s, H-2)] 和1个甲氧基氢信号[δH 3.57 (3H, s, 3′-OCH3)]。与之相应, 13C NMR (CD3OD, 125 MHz) 谱(表 1) 显示存在2个苯环[δC 158.5 (C-5), 98.7 (C-6), 157.3 (C-7), 97.4 (C-8), 158.5 (C-9), 101.9 (C-10), 131.6 (C-1′), 111.5 (C-2′), 148.9 (C-3′), 147.5 (C-4′), 116.1 (C-5′), 120.8 (C-6′)]、1个亚甲基[δC 27.2 (C-4)]、2个次甲基[δC 79.0 (C-2), 70.6 (C-3)] 和1个甲氧基[δC 56.3 (3′-OCH3)]信号。此外, 1H NMR和13C NMR还存在1组糖基结构片段[δH 4.87 (1H, d, J = 6.9 Hz, H-1″), 3.92 (1H, dd, J= 11.7, 1.4 Hz, H-6″a), 3.74 (1H, dd, J = 11.7, 4.2 Hz, H-6″b), 3.40~3.45 (4H, m, H-2″, 3″, 4″, 5″)]; [δC 102.6 (C-1″), 75.1 (C-2″), 78.4 (C-3″), 71.3 (C-4″), 78.4 (C-5″), 62.8 (C-6″)], 结合酸水解和衍生化后的LC-MS分析确定其为β-D-葡萄糖。HMBC谱(图 2) 显示H-1′与C-5相关, 表明葡萄糖与苷元C-5相连。以上NMR数据与已知化合物(+)-3′-O-methylcatechin 5-O-β-D-glucopyranoside相似[10], 不同之处在于化合物1多出1组苯甲酰基信号[δH 7.88 (2H, dd, J = 8.1, 1.0 Hz, H-2‴, 6‴), 7.41 (2H, t, J = 7.9 Hz, H-3‴, 5‴), 7.55 (1H, t, J = 7.5 Hz, H-4‴); δC 129.9 (C-1‴), 130.9 (C-2‴, C-6‴), 129.8 (C-3‴, C-5‴), 134.6 (C-4‴), 167.1 (C-7‴)]。HMBC谱(图 2) 显示, H-3、H-2‴、H-6‴与C-7‴相关, 可推断出苯甲酰基与黄烷醇母核的C-3位以酯基形式相连。
由化合物1 (H-2) 的峰型(br s) 确定其相对构型为2, 3-顺式构型[11]。C-2和C-3的绝对构型通过量子化学计算的方法[B3LYP/6-31G (d, p), MeOH] 确定, ECD谱结果表明, (2R, 3R)构型与化合物1的实验测试曲线趋势一致(图 3)。故确定化合物的结构为(2R, 3R)-5, 7, 4′-trihydroxy-3′-methoxy-3-formylflavan-3-ol-5-O-β-D-glucopyranoside (1), 为新化合物, 命名为persinoids A, 结构见图 1
化合物2为无色固体。[α]-17.0 (c 0.1 MeOH), UV (MeOH) λmax (log ε): 209.0 (3.26)、227.0 (3.05)、280.0 (2.47) nm。HR-ESI-MS [M+Na]+ m/z649.225 3 (C33H38O12Na计算值649.225 5), 提示化合物2分子式为C33H38O12, 不饱和度为15。IR显示该结构中含有羟基(3 393 cm-1)和酯羰基(1 705 cm-1)特征吸收峰。在1H NMR (CD3OD, 500 MHz) 谱(表 1) 中存在一组ABX耦合系统氢信号[δH 6.86 (1H, d, J = 1.9 Hz, H-2), 6.73 (1H, dd, J = 8.1, 1.9 Hz, H-6), 6.60 (1H, d, J = 8.1 Hz, H-5)]; 一组1, 3, 4, 5-四取代苯环氢信号[δH 6.59 (1H, br s, H-2′), 6.65 (1H, d, J= 1.1, H-6′)]; 2个次甲基氢信号[δH 5.52 (1H, d, J = 5.9 Hz, H-7), 3.62 (1H, dd, J = 13.5, 5.4 Hz, H-8)]; 4个亚甲基氢信号[δH 4.00 (1H, dd, J = 10.2, 5.0 Hz, H-9a), 3.74 (1H, dd, J = 10.2, 8.3 Hz, H-9b), 3.49 (2H, t, J = 6.5 Hz, H-9′), 2.52 (2H, t, J = 7.5 Hz, H-7′), 1.72 (2H, m, H-8′)] 和2个甲氧基氢信号[δH 3.79 (3H, s, 3-OCH3), 3.69 (3H, s, 3′-OCH3)]。同样, 在13C NMR (CD3OD, 125 MHz) 谱中(表 1) 也显示有2个苯环[δC 134.6 (C-1), 110.6 (C-2), 145.2 (C-3), 147.3 (C-4), 115.9 (C-5), 119.7 (C-6), 136.9 (C-1′), 118.0 (C-2′), 148.8 (C-3′), 147.5 (C-4′), 129.4 (C-5′), 114.2 (C-6′)]; 4个亚甲基[δC 72.8 (C-9), 32.8 (C-7′), 35.8 (C-8′), 62.2 (C-9′)] 和2个甲氧基[δC 56.8 (3-OCH3), 56.2 (3′-OCH3)] 信号。其中, δC 89.0 (C-7) 和δC 53.4 (C-8) 为苯骈呋喃木脂素7, 8位碳的特征信号。此外, 1H NMR和13C NMR还存在一组糖基结构片段[δH 4.37 (1H, d, J= 7.8 Hz, H-1″), 4.60 (1H, dd, J = 11.8, 2.3 Hz, H-6″a), 4.31 (1H, dd, J= 11.8, 6.7 Hz, H-6″b), 3.21~3.56 (4H, m, H-2″, 3″, 4″, 5″)]; [δC 104.9 (C-1″), 75.3 (C-2″), 78.1 (C-3″), 72.0 (C-4″), 75.1 (C-5″), 65.3 (C-6″)], 同化合物1的方法, 通过H-1″的耦合常数7.8 Hz, 结合酸水解和衍生化后的LC-MS分析确定其为β-D-葡萄糖。结合HMBC谱(图 2), H-1″与C-9相关, 证明葡萄糖连接在苷元C-9位。以上NMR数据与已知化合物3非常相似, 区别在于化合物2多出一组苯甲酰基信号[δH 7.90 (2H, dd, J = 8.2, 1.1 Hz, H-2‴, 6‴), 7.49 (1H, t, J = 7.5 Hz, H-4‴), 7.32 (2H, t, J = 7.8 Hz, H-3‴, 5‴); δC 130.5 (C-1‴), 129.5 (C-2‴, C-6‴), 129.4 (C-3‴, C-5‴), 134.2 (C-4‴), 167.8 (C-7‴)]。HMBC谱(图 2) 显示, H-2‴、H-6‴、H-6″与C-7‴相关, 证明苯甲酰基与葡萄糖的C-6″相连。
通过对比文献[12], 由耦合常数(JH7, H8 = 5.9 Hz) 可知H-7和H-8为反式构型。绝对构型采取量子化学计算的方法[B3LYP/6-31G (d, p), MeOH], ECD谱表明, (7R, 8S) 构型与化合物2的实验测试曲线趋势一致(图 3)。故确定化合物2的结构为(7R, 8S)-dihydrodehydrodiconiferyl 6″-benzoyl alcohol-9-O-β-D-glucopyranoside, 为新化合物, 命名为persiligcoside A, 结构见图 1
TripleTOF 6600型高效液相色谱-四极杆飞行时间高分辨质谱联用仪(AB SCIEX); Bruker AM-500 MHz核磁共振谱用超导核磁共振仪(TMS内标); Thermo EVO 300紫外分光光度计、Thermo Nicolet IS10红外光谱仪(美国Thermo Scientific公司); Agilent 1260 Infinity Ⅱ LC HPLC (美国Agilent公司); EYELA CA-3310旋转蒸发仪(RIKAKIKAI, TOKYO); EYELA N-1100旋转蒸发仪(上海爱朗仪器有限公司); 赛谱锐思LC52型半制备高效液相(北京赛谱锐思科技有限公司); YMC-Pack ODS-A色谱柱(50 mm × 250 mm, 5 μm, YMC, TOKYO); Sephadex LH-20 (Parmacia Biotech公司); GF254硅胶薄层板(青岛海洋化工厂)。
桃仁于2021年4月购自于内蒙古赤峰, 经河南中医药大学董诚明教授鉴定为山桃Prunus davidiana (Carr.) Franch.的干燥成熟种子山桃仁, 干燥标本保存于河南中医药大学中药药效物质基础研究室(标本号: STR202104NM)。
山桃仁40.00 kg, 以70%乙醇在80℃下(溶剂∶溶质= 8∶1) 回流提取3次, 提取液经旋转蒸发仪减压浓缩成总浸膏, 将总浸膏依次以石油醚、乙酸乙酯等体积萃取6次, 得到石油醚部位(519.32 g)、乙酸乙酯部位(100.50 g) 与水部位(4 235.30 g) 三部分浸膏。将乙酸乙酯组分(Fr.A) 经MCI柱色谱, 以乙醇-水系统为流动相按梯度(0%、10%、20%、30%、40%、50%、60%、70%、80%、95%) 进行分离, 得9个组分Fr.A.1~Fr.A.9。Fr.A.6 (3.30 g) 经ODS色谱柱, 以甲醇-水系统为流动相梯度洗脱(5%~100%), 得到Fr.A.6.1~Fr.A.6.12。Fr.A.6.4 (307.22 mg) 经Sephadex LH-20分离得到3个组分(Fr.A.6.4.1~Fr.A.6.4.3), Fr.A.6.4.1经半制备HPLC (甲醇-水= 70∶30) 得到化合物1 (tR = 13.6 min, 12.50 mg), Fr.A.6.6 (558.30 mg) 经硅胶柱色谱和半制备HPLC (甲醇-水= 65∶35) 得到化合物2 (tR = 12.5 min, 2.30 mg)、3 (tR = 15.3 min, 22.36 mg)。Fr.A.5 (11.36 g) 经ODS色谱柱分离得到7个组分, Fr.A.5.5 (216.22 mg) 经凝胶柱色谱分离得到Fr.A.5.5.1~Fr.A.5.5.4, 其中Fr.A.5.5.4 (11.56 mg) 经半制备HPLC纯化(甲醇-水= 65∶35) 得到4 (tR = 16.8 min, 4.18 mg)。Fr.A.4 (7.68 g) 经ODS色谱柱分离得到12个组分(Fr.A.4.1~Fr.A.4.12), Fr.A.4.4 (525.89 mg) 经凝胶柱色谱分离得到5个部分, 主成分集中于Fr.A.4.4.2 (301.24 mg), 经制备薄层色谱得到4个部分, Fr.A.4.4.2.4 (22.36 mg) 经半制备HPLC纯化(甲醇-水= 65∶35) 得到化合物5 (tR = 14.6 min, 3.26 mg)。
称取化合物2.0 mg, 加入2 mol·L-1盐酸3 mL, 90 ℃条件下加热2.5 h。将混合物真空浓缩干燥后加1 mL蒸馏水, 用等体积乙酸乙酯萃取3次。乙酸乙酯层为苷元, 水层浓缩干燥得葡萄糖。在浓缩水层中加L-半胱氨酸甲酯1 mg、吡啶1 mL在60 ℃下加热1.5 h。向混合物中加入邻甲苯异硫氰酸酯20 μL, 在60 ℃下加热1.5 h。用LC-MS分析混合物, 通过比较, 样品与葡萄糖标准品的保留时间一致, 鉴定化合物12中葡萄糖的绝对构型为D-葡萄糖。
化合物1黄色无定形粉末, 易溶于甲醇, [α]-87.6 (c 0.1 MeOH); UV (MeOH) λmax (log ε) 207.0 (3.18)、227.0 (2.89)、279.0 (2.22) nm; IR νmax 3 361、1 602、1 271、1 072、1 026、713 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1, HR-ESI-MS m/z 593.163 6 [M+Na]+ (计算值为C29H30O12Na+, 593.162 9)。
化合物2 无色固体, 易溶于甲醇, [α]-17.0 (c 0.1 MeOH); UV (MeOH) λmax (log ε) 209.0 (3.26)、227.0 (3.05)、280.0 (2.47) nm; IR νmax 3 393、1 705、1 452、1 277、1 053、715 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1; HR-ESI-MS m/z 649.225 3 [M+Na]+ (计算值为C33H38O12Na+, 649.225 5)。
化合物3 无色油状物, 易溶于甲醇, ESI-MS m/z 523 [M+H]+, 分子式为C26H34O11。该化合物的NMR数据与文献[13]报道的(7R, 8S)-dihydrodehydrodiconiferyl alcohol-9-O-β-D-glucopyranoside基本一致。
化合物4无色油状物, 易溶于甲醇, ESI-MS m/z 327 [M+H]+, 分子式为C16H22O7。该化合物的NMR数据与文献[14]报道的2-methoxy-4-(2-propenyl)-phenyl-O-β-D-glucopyranoside基本一致。
化合物5黄色无定型粉末, 易溶于甲醇, ESI-MS m/z 257 [M+H]+, 分子式为C13H20O5。该化合物的NMR数据与文献[15]报道的2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-propane-1, 3-diol基本一致。
有研究结果表明, 桃仁中所含的芳香苷类化合物如prupersin、prupersin A-D、ethyl amygdalinate等具有抗炎活性; 含氰基官能团的芳香苷如prupersin D、苦杏仁苷和野樱苷表现出较好的抗氧化活性[16]。这将为山桃仁中获得的苷类化合物活性研究提供借鉴。
