Two undescribed terpene glycosides and two compounds were isolated from the n-butanol fraction of Alpiniae Oxyphyllae Fructus by using various chromatographic methods, including MCI Gel, Sephadex LH-20, ODS, silica gel and semi-preparative HPLC. The structures of the isolated compounds were identified by spectroscopy methods (1D, 2D NMR, UV, IR, MS, etc.), and the absolute configuration of the compound 1 was determined by ECD calculation and acid hydrolysis. Compounds 1 and 2 are new compound, and compounds 3 and 4 were isolated from Alpiniae Oxyphyllae Fructus for the first time.

运用MCI Gel、Sephadex LH-20、ODS和硅胶等柱色谱, 结合半制备液相、TLC等分离方法, 从益智仁正丁醇部位中分离纯化得到4个萜苷类化合物。运用现代波谱学方法(1D、2D NMR、UV、IR、MS等) 对所分离得到的化合物进行结构鉴定, 并运用计算ECD和酸水解的方法确定了新化合物的绝对构型, 其中化合物12为新化合物, 化合物34为首次从益智仁中分离得到。

Two undescribed terpene glycosides and two compounds were isolated from the n-butanol fraction of Alpiniae Oxyphyllae Fructus by using various chromatographic methods, including MCI Gel, Sephadex LH-20, ODS, silica gel and semi-preparative HPLC. The structures of the isolated compounds were identified by spectroscopy methods (1D, 2D NMR, UV, IR, MS, etc.), and the absolute configuration of the compound 1 was determined by ECD calculation and acid hydrolysis. Compounds 1 and 2 are new compound, and compounds 3 and 4 were isolated from Alpiniae Oxyphyllae Fructus for the first time.

益智仁为姜科植物益智(Alpinia oxyphylla Miq.) 的干燥成熟果实和种子[1], 是卫生部1998年公布的一种药食同源的天然植物, 主产于我国海南岛山区, 是我国四大南药之一[2]。益智仁性温味辛, 归心、脾、肾经, 有温补固摄、暖脾止泻、固涎摄唾、温肾固精缩尿的功效[3]。益智仁中主要含有萜类、二苯基庚烷类、黄酮类、甾醇类和酚酸类等化学成分[4-6]。相关药理学研究表明, 益智仁主要具有抗炎、神经保护、抗菌、抗氧化、强心、舒张血管和提高免疫力等作用[7-9]。近年来, 由于益智仁的神经保护作用再次被广泛关注, 但是关于其药效物质基础的研究不够明确, 为了进一步揭示益智仁的药效物质基础, 丰富其化学组成, 本实验对益智仁的化学成分进行了系统的分离鉴定, 从其正丁醇部位分离鉴定得到了4个萜苷类化合物, 其中化合物12为新化合物, 化合物3和4为首次从益智仁中分离得到(图 1)。
化合物1为无色透明液体, [α]-9.0 (c 0.1, MeOH), HR-ESI-MS给出准分子离子峰m/z 519.256 8 [M+Na]+, 计算值为519.256 4, 推测分子式为C26H40O9, 不饱和度为7。化合物11H NMR (CD3OD, 500 MHz) 谱(表 1) 显示3个烯烃特征氢信号[δH 6.60 (1H, ddd, J = 9.5, 5.4, 2.4 Hz, H-12), 5.23 (1H, br s, H-17a), 4.61 (1H, br s, H-17b)]、2个连氧次甲基信号[δH 3.94 (1H, dd, J = 10.8, 5.3 Hz, H-7), 3.25 (1H, dd, J = 12.0, 4.1 Hz, H-3)]、1个连氧亚甲基信号δH 4.38 (2H, t, J = 7.5 Hz, H-15) 和3个甲基信号[δH 1.10 (3H, s, H-18), 0.86 (3H, s, H-19), 0.75 (3H, s, H-20)]。分析其13C NMR (CD3OD, 125 MHz) 谱并结合HSQC谱发现, 化合物1中存在1个酯基信号δC 173.7 (C-16)、4个烯烃特征碳信号[δC 151.5 (C-8), 142.6 (C-12), 126.8 (C-13), 105.1 (C-17)]、2个季碳信号[δC 40.3 (C-4), 39.6 (C-10)]、4个次甲基信号[δC 90.2 (C-3), 74.1 (C-7), 55.3 (C-9), 53.5 (C-5)]、6个亚甲基信号[δC 67.2 (C-15), 38.0 (C-1), 33.8 (C-6), 27.7 (C-2), 26.4 (C-11), 26.0 (C-14)] 和3个甲基信号[δC 28.5 (C-18), 16.8 (C-19), 14.8 (C-20)], 此外化合物11H和13C NMR谱中还存在一组糖端基氢及碳信号[δH 4.33 (1H, d, J = 7.8 Hz, H-1′), δC 106.8 (C-1′)], 通过H-1′的耦合常数7.8 Hz, 结合酸水解和衍生化后的LC-MS分析确定为β-D-葡萄糖。通过比较核磁数据, 发现化合物1的苷元部分与半日花烷型二萜化合物pacovatinin B[10]相似, 为了进一步确定其结构, 进行了2D NMR实验。在1H-1H COSY谱中(图 2) 显示出H-1/H-2/H-3、H-5/H-6/H-7、H-9/H-11/H-12、H-14/H-15和H-1′/H-2′/H-3′/H-4′/H-5′/H2-6′多个质子耦合系统。在HMBC谱中(图 2), H-14、H-15/C-13、C-16的相关信号结合C-15 (δC 67.2) 较低场的化学位移推测存在一个五元内酯环片段; H-17a、H-17b/C-7、C-9的相关信号确定了C-8上存在一个末端双键; H-11/C-13、H-12/C-9、C-13和H-14/C-12的相关信号, 确定了五元内酯环片段通过C-13上的一个二碳链连接在六元环的C-9; H-1′/C-3的相关信号, 确定了葡萄糖取代在C-3。
通过NOESY谱确定了化合物1的相对构型。在NOESY谱中(图 3), H-9与H-5和H-7存在相关信号, H-5与H-3和H-7存在相关信号, 且CH3-20与H2-11存在相关信号, 证明了H-3、H-5、H-7、H-9位于环平面同侧, CH3-20位于环平面另一侧。另外, 根据H2-11与H2-14的相关信号阐明了C-12与C-13处三取代烯烃的构型为E[10]。所以确定其相对构型为(3S*, 5R*, 7S*, 9R*, 10S*, 12E)。化合物1的绝对构型是通过电子圆二色性(TDDFT ECD) 计算与实验ECD对比来确定的, 在TDDFT/B3LYP/6-31G(d, p) 水平下使用SMD模式并添加色散校正计算。结果显示实验ECD曲线与构型(3S, 5R, 7S, 9R, 10S, 12E)的曲线吻合较好(图 4), 因此, 化合物1的绝对构型确定为3S, 5R, 7S, 9R, 10S, 12E, 其结构为pacovatinin B 3-O-β-D-glucopyranoside, 命名为oxyphylloneside A。
化合物2为淡黄色透明液体, [α]-13.0 (c 0.1, MeOH), HR-ESI-MS给出准分子离子峰m/z 497.200 4 [M+Na]+, 计算值为497.199 3, 推测分子式为C22H34O11, 不饱和度为6。化合物21H NMR (CD3OD, 500 MHz) 谱(表 1) 显示一组ABX系统芳香氢信号[δH 7.01 (1H, d, J = 7.6 Hz, H-6), 6.97 (1H, d, J = 1.5 Hz, H-3), 6.78 (1H, dd, J = 7.6, 1.5 Hz, H-5)]、1个次甲基信号δH 2.83 (1H, dt, J = 13.8, 6.9 Hz, H-8) 和3个甲基信号[δH 2.21 (3H, s, H-7), 1.22 (3H, s, H-9), 1.20 (3H, s, H-10)]。与之相应, 在13C NMR (CD3OD, 125 MHz) 谱中, 出现1个1, 3, 4取代苯环[δC 126.2 (C-1), 157.0 (C-2), 114.6 (C-3), 149.0 (C-4), 121.2 (C-5), 131.4 (C-6)]、1个次甲基δC 35.1 (C-8) 和3个甲基[δC 16.1 (C-7), 24.5 (C-9), 24.4 (C-10)] 信号, 此外化合物21H和13C NMR谱中还存在两组糖端基氢及碳信号[δH 4.89 (1H, d, J = 7.8 Hz, H-1′), 4.44 (1H, d, J = 7.9 Hz, H-1″), δC 102.6 (C-1′), 104.5 (C-1″)], 通过端基质子的耦合常数7.8和7.9 Hz, 结合酸水解和衍生化后的LC-MS分析确定为β-D-葡萄糖。将以上数据与文献对比, 发现化合物2与已知化合物carvacrol 2-O-β-glucopyranoside (3) 结构相似[11], 其苷元皆为香芹酚, 但是化合物2多了1个葡萄糖片段。在HMBC谱(图 2) 中H-1″/C-4′的相关信号确定了葡萄糖取代在C-4′, 同时C-4′ (δC 80.4) 向低场方向移动的化学位移也进一步证明了这个结论。故确定此化合物的结构为carvacrol 2-O-β-D-glucopyranoside-4′-O-β-D-glucopyranosyl, 命名为oxyphylloneside B。
TripleTOF 6600型高效液相色谱-高分辨质谱联用仪(AB SCIEX); 赛谱锐思LC-52型半制备液相色谱仪(赛谱锐思北京科技有限公司); Anton Paar MCP 5100型旋光仪(奥地利Anton Paar); BrukerAM-500MHz核磁共振谱用超导核磁共振仪(TMS做内标, 德国Bruker公司); Thermo EVO300紫外分光光度计和Thermo Nicolet IS10红外光谱仪(美国Thermo Scientific); RE-5210A型旋转蒸发仪(上海亚荣生化仪器厂); N-1100型旋转蒸发仪和EYELACA-3310型低温冷凝循环泵(东京理化器械株式会社); 原子型1820C型纯水机(重庆摩尔水处理设备有限公司); MCIGel CHP-20柱色谱材料(日本三菱化学公司); SephadexLH-20柱色谱材料(Parmacia Biotech公司); 正相硅胶柱色谱材料(200~300目, 青岛海洋化工厂); YMC-PackODS-A色谱柱(250 mm × 20 mm, 5 μm) 和C18 ODS柱色谱材料(50 μm) (日本YMC有限公司); 色谱纯及分析纯试剂(天津四友精细化学品有限公司)。
益智仁购自中国海南省琼中县, 经河南中医药大学董诚明教授鉴定为姜科植物益智(Alpinia oxyphylla Miq.) 的干燥成熟果实和种子, 干燥标本保存于河南中医药大学中药药效物质基础研究室(标本号: YZ202104QZ)。
将40.00 kg益智仁干燥后粉碎, 用8倍量70%乙醇回流提取2 h (3次), 提取液合并并减压浓缩得浸膏8.30 kg, 浸膏用3倍量的水混悬后依次用30 L乙酸乙酯、正丁醇进行萃取至萃取液无色后(10次以上) 浓缩得到乙酸乙酯部位(1.45 kg)、正丁醇部位(628.33 g) 和水部位(6.60 kg)。正丁醇部位上MCI柱, 用乙醇-水系统(0∶100~95∶5) 梯度洗脱后, 依次得到水部位(371.10 g)、10%乙醇部位(8.43 g)、30%乙醇部位(33.36 g)、50%乙醇部位(49.82 g)、70%乙醇部位(20.73 g) 和95%乙醇部位(105.06 g)。MCI 30%乙醇部位上凝胶柱色谱, 以甲醇-水(0∶100~95∶5) 为洗脱剂进行梯度洗脱, 依次得到Fr.A1~Fr.A10。Fr.A1 (20.50 g) 上ODS柱色谱, 以甲醇-水(10∶90~100∶0) 为洗脱剂进行梯度洗脱, 依次得到Fr.A1-1~Fr.A1-10。Fr.A1-1 (6.75 g) 过硅胶柱色谱(200~300目) 二氯甲烷-甲醇(50∶1~0∶100) 梯度洗脱, 依次得到Fr.A1-1-1~Fr.A1-1-9。Fr.A1-1-6 (440.30 mg) 经半制备高效液相(乙腈∶水= 18∶82) 纯化, 得到化合物4 (tR = 21.9 min; 3.32 mg)。Fr.A1-5 (2.88 g) 过硅胶柱色谱(200~300目) 二氯甲烷-甲醇(40∶1~0∶100) 梯度洗脱, 依次得到Fr.A1-5-1~Fr.A1-5-8。Fr.A1-5-5 (112.30 mg) 经半制备高效液相(甲醇∶水= 50∶50) 纯化, 得到化合物1 (tR = 13.1 min; 3.40 mg)。MCI 50%乙醇部位上凝胶柱色谱, 以甲醇-水(0∶100~95∶5) 为洗脱剂进行梯度洗脱, 依次得到Fr.B1~Fr.B8。Fr.B1 (42.60 g) 上MCI柱色谱以甲醇-水(30∶70~95∶5) 为洗脱剂进行梯度洗脱, 依次得到Fr.B1-1~Fr.B1-4。Fr.B1-3 (15.38 g) 上ODS柱色谱, 以甲醇-水(10∶90~100∶0) 为洗脱剂进行梯度洗脱, 依次得到Fr.B1-3-1~Fr.B1-3-16。Fr.B1-3-12 (646.30 mg) 过硅胶柱色谱(200~300目) 石油醚-乙酸乙酯(80∶1~0∶100), 得到Fr.B1-3-12-1~Fr.B1-3-12-6。Fr.B1-3-12-5 (78.60 mg) 经半制备高效液相(乙腈∶水= 42∶58) 纯化, 得到化合物3 (tR = 17.2 min; 17.10 mg)。Fr.B1-3-12-6 (110.20 mg) 经半制备高效液相(乙腈∶水= 40∶60) 纯化, 得到化合物2 (tR = 19.6 min; 20.00 mg)。
取化合物12 (大约1 mg), 加2 mol·L-1盐酸3 mL在90℃下加热水解2.5 h。将混合物在真空下浓缩干燥后加水1.0 mL, 再加乙酸乙酯(1 mL×3) 萃取。乙酸乙酯层浓缩干燥, 得苷元; 水层浓缩干燥得葡萄糖。在浓缩后水层的样品加L-半胱氨酸甲酯盐酸盐(1 mg) 和吡啶(1 mL) 在60 ℃水浴下加热1.5 h, 再加入邻甲苯异硫氰酸酯(20 μL) 在60 ℃水浴下加热1.5 h。另取D-葡萄糖标品加L-半胱氨酸甲酯盐酸盐(1 mg) 和吡啶(1 mL) 在60 ℃水浴下加热1.5 h, 再加入邻甲苯异硫氰酸酯(20 μL) 在60 ℃水浴下加热1.5 h。将得到的葡萄糖供试品衍生物和D-葡萄糖标准品衍生物进行液质分析。液质分析条件: TripleTOF 6600 UPLC/MS (AB SCIEX); Kinetex C18 100Å(2.6 μm, 2.1 mm × 50 mm); 梯度洗脱: 0.1%甲酸水(A)-甲醇(B), 0~20 min, 10%~100% B; 柱温40 ℃, 流速0.3 mL·min-1; 质谱条件: 正离子模式。通过对比葡萄糖供试品衍生物(tR= 12.52 min, m/z 447.125 3 [M+H]+) 与D-葡萄糖标准品衍生物(tR = 12.52 min, m/z 447.125 4 [M+H]+)的保留时间和质谱数据, 从而确定了化合物12中糖的绝对构型为D-葡萄糖[12]
化合物1  无色透明液体, 易溶于甲醇, [α]-9.0 (c 0.1, MeOH); UV (MeOH) λmax (log ε) 201 (2.42)、225 (2.58) nm; IR νmax 3 421、1 735、1 674、1 206、1 077、1 014 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1, HR-ESI-MS m/z 519.256 8 [M+Na]+ (计算值为C16H20O9Na+, 519.256 4)。
化合物2  淡黄色透明液体, 易溶于甲醇, [α]-13.0 (c 0.1, MeOH); UV (MeOH) λmax (log ε) 203 (2.90) nm; IR νmax 3 338、2 926、1 677、1 420、1 201、1 075 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1, HR-ESI-MS m/z 497.200 4 [M+Na]+ (计算值为C16H20O9Na+, 497.199 3)。
化合物3  黄色油状液体, 易溶于甲醇, ESI-MS m/z 313 [M+H]+, 分子式为C16H24O6。其1H和13C NMR (CD3OCD3, 500 MHz) 数据与文献[11]报道基本一致, 确定化合物为carvacrol 2-O-β-glucopyranoside。
化合物4  黄色油状液体, 易溶于甲醇, ESI-MS m/z 329 [M+H]+, 分子式为C16H24O7。其1H和13C NMR (CD3OCD3, 500 MHz) 数据与文献[13]报道基本一致, 确定化合物为thymoquinol 2-O-β-glucopyranoside。
