Two new coumarin glycosides were isolated and purified from the dichloromethane fraction of Angelica biserrata 75% ethanol extract using silica gel, Sephadex LH-20 column chromatography, and semi-preparative high-performance liquid chromatography. Through the comprehensive use of modern spectroscopic methods, their structures were identified as angelmanetin A (1) and angelmanetin B (2), respectively.

利用硅胶、Sephadex LH-20和半制备高效液相等色谱方法, 对独活(Angelica biserrata) 75%乙醇提取物的二氯甲烷部位进行分离纯化, 从独活二氯甲烷部位中分离得到2个新的香豆素苷。综合运用现代波谱学手段分别鉴定为angelmanetin A (1) 和angelmanetin B (2)。

Two new coumarin glycosides were isolated and purified from the dichloromethane fraction of Angelica biserrata 75% ethanol extract using silica gel, Sephadex LH-20 column chromatography, and semi-preparative high-performance liquid chromatography. Through the comprehensive use of modern spectroscopic methods, their structures were identified as angelmanetin A (1) and angelmanetin B (2), respectively.

独活(Angelicae Pubescentis Radix) 为伞形科植物重齿毛当归(Angelica pubescens Maxim. f. biserrata Shan et Yuan) 的干燥根, 主要分布于我国安徽、湖北、四川等地[1]。植物化学研究表明, 独活富含香豆素类、挥发油、有机酸类、多糖类化合物[2]。现代药理研究表明, 独活具有镇痛消炎、抗肿瘤、抗关节炎、预防和治疗帕金森病等疗效[3], 同时对强直性脊柱炎也有一定治疗作用[4]。本课题组前期曾对独活的化学成分进行了研究, 从中分离并鉴定了13个化合物, 其中香豆素及其苷类7个[5, 6]。为了进一步丰富独活化学成分, 并寻找新颖的天然产物, 以促进独活资源的综合开发与利用, 本实验继续对独活化学成分进行了分离鉴定, 从二氯甲烷部位得到2个新的香豆素苷(图 1)。
化合物1为白色粉末。HR-ESI-MS给出准分子离子峰m/z 559.180 9 [M+H] (计算值为559.181 6), 推测其分子式为C28H30O12, 不饱和度为14。UV光谱显示在218、263、300、329 nm处有四个吸收峰, 说明化合物可能为香豆素类化合物。IR光谱显示出明显的苯环(1 514、1 456 cm-1), 双键(1 614 cm-1), 羰基(1 714 cm-1) 和羟基(3 333 cm-1) 吸收。由化合物1 13C NMR谱(表 1) 中的9个碳信号δC 163.4 (C-2)、112.5 (C-3)、146.3 (C-4)、130.3 (C-5)、107.6 (C-6)、165.6 (C-7)、114.7 (C-8)、113.7 (C-4a)、152.6 (C-8a) 和1H NMR谱(表 1) 中对应的氢信号δH 6.15 (H-3, d, J = 9.5 Hz)、7.82 (H-4, d, J = 9.5 Hz)、7.36 (H-5, d, J = 8.3 Hz)、6.74 (H-6, d, J = 8.3 Hz) 可知化合物1为7, 8-双取代的香豆素。结合δH 3.40 (1H, m)、3.21 (1H, m) / δC 28.5, CH-9; δH 4.93 (1H, d) / δC 91.8, CH-10; δC 79.5, C-11; δH 1.35 (3H, s) / δC 22.2, CH-12; δH 1.38 (3H, s) / δC 24.3, CH-13发现与二氢欧山芹醇[7]波谱数据相似, 说明结构中含有二氢欧山芹醇片段。由δC 122.5, C-1′; δH 7.44 (1H, s) / δC 113.7, CH-2′; δC 148.9, C-3′; δC 152.9, C-4′; δH 6.72 (1H, d, J = 8.2 Hz) / δC 115.9, CH-5′; δH 7.48 (1H, d, J = 8.2 Hz) / δC 125.1, CH-6′可知结构中含有1, 2, 4-三取代苯环。另外, 从δC 167.9和56.5可知结构中还含有1个酯基和1个甲氧基。由δC 99.1、75.1、78.2、72.1、75.3、64.9推测结构中含有吡喃葡萄糖基, 结合化合物1经酸水解后, 其水溶液与D-葡萄糖对照品共薄层色谱鉴别, 且水溶液的比旋光度为$ {\left[\alpha \right]}_{\mathrm{D}}^{25} $ +48.0º(c 0.1, H2O) 可明确水解物中的糖为D-葡萄糖[8]。葡萄糖端基氢信号δH 4.66 (H-1″, d, J = 7.8 Hz) 表明其端基氢的相对构型为β[9]。在HMBC谱中, δH 6.72 (H-5′) 及δH 7.48 (H-6′) 与δC 167.9 (-COO-) 相关, 说明酯基与C-1′相连; δH 7.44 (H-2′) 及δH 6.72 (H-5′) 与C-3′相关、δH 3.83 (-OCH3) 与C-3′相关, 提示甲氧基连接在C-3′位(图 2)。由于在HMBC谱中未检测到δC 152.9 (C-4′) 与δH 4.66 (H-1″) 远程相关的信号, 但基于C-1″的化学位移δC (99.1) 及其产生的苷化位移可推测葡萄糖C-1′′连接在C-4′位。在CD光谱中, 化合物1的苷元与其类似物angelmarin[10]具有相似的正性cotton曲线(图 3)。因此化合物1的绝对构型被确定为10S, 命名为angelmanetin A。
化合物2为白色粉末, HR-ESI-MS给出准分子离子峰m/z 585.196 2 [M+H] (计算值585.197 2), 确定其分子式为C30H32O12, 不饱和度为15。UV光谱在205、326 nm处有2个吸收峰, 表明化合物可能为香豆素类化合物。IR谱中存在苯环(1 516、1 456 cm-1)、双键(1 614 cm-1)、羰基(1 714 cm-1) 和羟基(3 340 cm-1) 的吸收峰。化合物2与化合物1的核磁数据相比, 主要区别在于化合物2中多出一个反式双键信号δH 7.49 (1H, d, J = 15.9 Hz) / δC 146.9, CH-7′; δH 6.23 (1H, d, J = 15.9 Hz) / δC 115.3, CH-8′。HMBC谱中, H-7′与C-1′/C-2′ / C-6′相关, H-8′与C-1′/酯基(168.9) 相关, 表明C-7′和C-8′分别连接在C-1′位和酯基上。化合物2使用10S和10R构型通过量子化学计算得出的旋光值分别为601.5、-601.4, 与其实验值119.2比较可以推测出化合物2的绝对构型为10S, 命名为angelmanetin B。
AVANCE Ⅲ HD 400 MHz核磁共振仪(瑞士Bruker公司), 半制备高效液相色谱仪(北京赛谱锐思科技有限公司), YMC-Triart C18色谱柱(10 mm × 250 mm, 5 μm, 日本YMC公司), 圆二色谱仪(Jasco-810, 日本JASCO公司), WZZ-3自动旋光仪(上海仪电物理光学仪器有限公司), RE-2000A旋转蒸发仪(上海亚荣生化仪器厂), ZF-2型紫外灯(上海嘉鹏科技有限公司), 柱层析硅胶、薄层色谱硅胶GF254 (青岛海洋化工厂), Sephadex LH-20 (上海如吉生物科技发展有限公司), 常用有机试剂均购自成都科龙化工有限公司。
独活药材于2019年3月采购于重庆市巫山县官阳镇, 由袁吕江教授鉴定为伞形科重齿毛当归Angelica pubescens Maxim. f. biserrata Shan et Yuan.的根。样品标本(20190302) 存放于西南大学药学院。
独活药材(20 kg) 干燥粉碎后, 用75%的乙醇回流提取三次, 每次2 h。滤液合并后减压浓缩得浸膏4.2 kg。将浸膏悬浮于水中, 依次采用等体积的石油醚、二氯甲烷、乙酸乙酯、正丁醇萃取5次, 萃取液浓缩后分别得到石油醚部位(342 g), 二氯甲烷部位(600 g), 乙酸乙酯部位(64 g), 正丁醇部位(128 g)。二氯甲烷部位经硅胶柱色谱分离, 以石油醚-乙酸乙酯(100∶0→0∶100) 进行梯度洗脱, 分段收集洗脱液, 经TLC分析合并, 得到11个组分Fr. C1~Fr. C11。Fr. C11经Sephadex LH-20柱色谱分离, 以二氯甲烷-甲醇(1∶1) 进行洗脱, 得到3个组分Fr. C11-1~Fr. C11-3。其中Fr. C11-2再经半制备HPLC纯化, 50%甲醇为流动相, 流速为3.0 mL·min-1, 得到化合物1 (tR = 25.8 min, 7.8 mg), 化合物2 (tR = 28.3 min, 3.4 mg)。
取适量化合物12, 加少量甲醇溶解后, 分别用5% HCl于90 ℃水解5 h, 冷却后用乙酸乙酯萃取, 萃取液减压浓缩后得到苷元部分, 利用圆二色谱判断苷元的绝对构型; 水相用NaHCO3中和后浓缩, 用旋光仪检测糖的比旋度, 与D-葡萄糖标准品对照, 确定化合物12中葡萄糖的绝对构型均为D型。
利用Gaussian 16软件, 甲醇为溶剂, 根据密度泛函理论(DFT), 采用B3LYP泛函结合6-31G(d) 基组对分子结构进行几何优化。获得优化后的几何构型后, 采用PCM隐式溶剂模型, 结合B3LYP/6-311+G**基组计算分子的ORD。
