Six compounds were isolated from the ethyl acetate fraction of Citri Sarcodactylis Fructus by using various column chromatographic methods, such as MCI Gel CHP-20, ODS, Sephadex LH-20, silica gel and semipreparative HPLC. Their structures were identified to be citrusin G (1), citrusin H (2), citrusin E (3), syringin (4), coniferin (5), methylconiferin (6) by NMR, HR-ESI-MS, UV, IR spectra. Compounds 1 and 2 were new secondary metabolism products and 3-6 were obtained from the titled material for the first time. Compound 1 showed anti-renal fibrosis activity in TGF-β1-induced kidney proximal tubular cells.

采用MCI Gel CHP-20、ODS、Sephadex LH-20和硅胶等方法, 结合制备薄层和半制备液相色谱对佛手85%醇提物的乙酸乙酯部位进行分离纯化, 从中得到6个化合物, 通过NMR、HR-ESI-MS、UV和IR等方法鉴定其结构为citrusin G (1)、citrusin H (2)、citrusin E (3)、紫丁香酚苷(4)、松柏苷(5)、甲基松柏苷(6)。其中化合物12为新化合物, 3~6均为首次从佛手中分离得到。化合物1在TGF-β1诱导大鼠肾小管上皮细胞(NRK-52e) 肾纤维化体外模型上显示抗肾纤维化活性。

佛手中的两个新糖苷类化合物

采用MCI Gel CHP-20、ODS、Sephadex LH-20和硅胶等方法, 结合制备薄层和半制备液相色谱对佛手85%醇提物的乙酸乙酯部位进行分离纯化, 从中得到6个化合物, 通过NMR、HR-ESI-MS、UV和IR等方法鉴定其结构为citrusin G (1)、citrusin H (2)、citrusin E (3)、紫丁香酚苷(4)、松柏苷(5)、甲基松柏苷(6)。其中化合物12为新化合物, 3~6均为首次从佛手中分离得到。化合物1在TGF-β1诱导大鼠肾小管上皮细胞(NRK-52e) 肾纤维化体外模型上显示抗肾纤维化活性。

Six compounds were isolated from the ethyl acetate fraction of Citri Sarcodactylis Fructus by using various column chromatographic methods, such as MCI Gel CHP-20, ODS, Sephadex LH-20, silica gel and semipreparative HPLC. Their structures were identified to be citrusin G (1), citrusin H (2), citrusin E (3), syringin (4), coniferin (5), methylconiferin (6) by NMR, HR-ESI-MS, UV, IR spectra. Compounds 1 and 2 were new secondary metabolism products and 3-6 were obtained from the titled material for the first time. Compound 1 showed anti-renal fibrosis activity in TGF-β1-induced kidney proximal tubular cells.

中药佛手(Citri Sarcodactylis Fructus) 又名佛手柑、五指橘、飞穰等[1], 为芸香科植物佛手(Citrus medica L. var. sarcodactylis Swingle) 的干燥果实, 味辛、苦、酸、温, 归肝、脾、胃、肺经[2], 为传统药食同源中药材之一。历代本草中记载佛手具有疏肝理气、和中止痛、化痰止咳等功效[3]。现代药理及化学研究表明, 佛手具有抗炎[4]、抗肿瘤[5]、降血脂[6]、抗抑郁[7]、抗菌[8]和调节血糖[9]等多方面的药理活性, 主要含有黄酮类、香豆素类、多糖、挥发油类等成分[10]。佛手作为食品和药品应用广泛, 其炮制品“老香黄”在粤东地区家喻户晓, 称为“潮州三宝”之一。对于生品其药效及化学研究多集中于黄酮、挥发油和总多糖等成分, 功效相关物质不够明确, 而炮制品的特有药效物质更不清楚, 因此本课题以生佛手为研究对象, 从其乙酸乙酯部位分离纯化得到的6个糖苷类化合物(图 1), 化合物12为新化合物, 3~6均为首次从佛手中分离得到。
化合物1为淡黄色粉末, HR-ESI-MS [M+Na]+ m/z 411.126 0 (计算值C17H24O10Na 411.126 2) 推测分子式为C17H24O10。其1H NMR (CD3OD, 500 MHz) 谱(表 1) 中显示两个芳香氢信号[δH 6.45 (1H, d, J = 2.3 Hz, H-5), 6.23 (1H, d, J = 2.3 Hz, H-3)], 两个甲氧基氢信号[δH 3.78 (3H, s, 4-OCH3), 3.76 (3H, s, 6-OCH3)], 两个亚甲基氢信号[δH 2.91 (2H, m, H-7), 2.38 (2H, m, H-8)] 和一组葡萄糖氢信号[δH 4.83 (1H, d, J = 7.5 Hz, H-1′), 3.90 (1H, dd, J = 12.0, 2.3 Hz, H-6′a), 3.68 (1H, dd, J = 12.0, 5.9 Hz, H-6′b), 3.34~3.53 (4H, m, H-2′-5′)]; 13C NMR (CD3OD, 125 MHz) 谱(表 1) 结合HMBC谱共观察到17个碳信号, 包括一个羰基信号[δC 179.1 (C-9)]、一个苯环碳信号[δC 112.5 (C-1), 158.1 (C-2), 93.8 (C-3), 160.1 (C-4), 94.9 (C-5), 160.9 (C-6)]、两个亚甲基碳信号[δC 20.3 (C-7), 36.7 (C-8)]和两个甲氧基碳信号[δC 56.0 (4-OCH3), 55.8 (6-OCH3)] 以及一组葡萄糖碳信号[δC 103.1 (C-1′), 74.9 (C-2′), 78.3 (C-3′), 71.5 (C-4′), 78.1 (C-5′), 62.6 (C-6′)]。在1H-1H COSY谱中出现H2-7 (δH 2.91) 与H2-8 (δH 2.38) 的相关信号(图 2), 说明存在自旋偶合系统-CH2-CH2-。HMBC谱中存在H2-8 (δH 2.38) 与C-7 (δC 20.3) 和C-9 (δC 179.1) 的相关信号, 以及H2-7 (δH 2.91) 与C-1 (δC 112.5), C-2 (δC 158.1) 和C-8 (δC 36.7) 的相关信号, 证明了苯环C-1位丙酸基片段的存在。通过HMBC谱中H-1′ (δH 4.83)/C-2 (δC 158.1), 4-OCH3 (δH 3.78)/C-4 (δC 160.1), 6-OCH3 (δH 3.76)/C-6 (δC 160.9) 的相关信号确定了葡萄糖基和两个甲氧基的位置。通过酶水解确定葡萄糖绝对构型为D型, 同时根据端基氢的偶合常数J = 7.5 Hz推断为β构型。由此确定化合物1结构如图 1所示, 命名为citrusin G。
化合物2为淡黄色粉末, HR-ESI-MS [M+Na]+ m/z 425.141 9 (计算值C18H26O10Na 425.141 8) 推测分子式为C18H26O10。比较化合物12的1D和2D NMR数据, 发现二者结构相似, 区别在于化合物2的C-9位羟基被甲氧基取代。1H NMR (CD3OD, 500 MHz) 谱(表 1) 中[3.64 (3H, s, 9-OCH3)] 和13C NMR (CD3OD, 125 MHz) 谱(表 1) 中[52.1 (9-OCH3)] 结合HMBC谱中, H2-7 (δH 2.92) 与C-1 (δC 111.6), C-2 (δC 158.1) 和C-8 (δC 34.8) 存在相关信号, H2-8 (δH 2.47) 和9-OCH3 (δH 3.64) 均与C-9 (δC 176.5) 存在相关信号, 提示丙酸基的羧基被甲基化且C7-C9片段与苯环的C-1位相连; H-1′ (δH 4.84) 与C-2 (δC 158.1) 存在HMBC相关信号, 提示葡萄糖片段与苯环的C-2位相连; H-5 (δH 6.45) 和C-1 (δC 111.6)、C-3 (δC 93.8) 以及H-3 (δH 6.24) 和C-1 (δC 111.6)、C-5 (δC 95.1) 的HMBC相关信号证明了3/5位两个芳氢的存在。4-OCH3 (δH 3.78) 与C-4 (δC 160.1), 6-OCH3 (δH 3.76) 与C-6 (δC 161.2) 存在相关信号证明了两个甲氧基的位置(图 2)。通过酶水解确定葡萄糖绝对构型为D型, 同时根据端基氢的偶合常数J = 7.3 Hz推断为β构型。综上所述确定化合物2结构如图 1所示, 命名为citrusin H。
通过建立TGF-β1诱导的大鼠肾小管上皮细胞(NRK-52e) 肾纤维化体外模型, 对佛手乙酸乙酯部位中分离得到的化合物12的生物学效应进行测试。纤维连接蛋白(fibronectin)、Ⅰ型胶原蛋白(collagen Ⅰ) 和α-平滑肌肌动蛋白(α-SMA) 为本实验的特征标记物。首先使用CCK-8试剂盒测定化合物处理过的细胞活性, 结果表明化合物12在20 μmol·L-1时对细胞活力影响较小(图 3A), 因此用20 μmol·L-1测定抗肾纤维化作用。用Western blot检测化合物12对TGF-β1诱导的NRK-52e细胞的保护作用, 实验结果表明, 化合物1在20 μmol·L-1浓度下显著抑制了fibronectin和collagen Ⅰ的表达(图 3B)。
BrukerAM-500MHz超导核磁共振仪(TMS内标, 德国Bruker公司); Triple TOF 6600型高效液相色谱-四极杆飞行时间高分辨质谱联用仪(美国AB SCIEX公司); EVO300紫外分光光度计和Nicolet IS10红外光谱仪(美国Thermo Scientific公司); Autopol Ⅳ全自动旋光仪(美国Rudolph公司); LC-52型半制备液相色谱仪(赛谱锐思北京科技有限公司); N-1100型旋转蒸发仪和EYELACA-3310型低温冷凝循环泵(东京理化器械株式会社); 原子型1820C型纯水机(重庆摩尔水处理设备有限公司); Cholester柱(250 mm × 10 mm, 日本COSMOSIL有限公司); MCIGel CHP-20柱色谱材料(日本三菱化学公司); SephadexLH-20柱色谱材料(美国GE Healthcare公司); 正相硅胶柱色谱材料(200~300目) 和GF254硅胶薄层板(中国青岛海洋化工厂)。
佛手购自广东省肇庆市, 经韩山师范学院郑玉忠教授鉴定为芸香科植物佛手(Citrus medica L. var. sarcodactylis Swingle) 的干燥果实, 干燥标本保存于河南中医药大学中药药效物质基础研究室(标本号: FS202204ZQ)。
20 kg佛手6倍量85%乙醇浸泡三次, 所得提取液减压浓缩后得到总浸膏(10 kg), 加水分散后依次用石油醚、乙酸乙酯、正丁醇萃取, 浓缩萃取液得到乙酸乙酯部位(170 g)、正丁醇部位(2 kg) 和水部位(8 kg)。乙酸乙酯部位上MCI柱, 用乙醇-水系统(20∶80~95∶5) 梯度洗脱后, 依次得到20%乙醇部位(49 g)、30%乙醇部位(14 g)、40%乙醇部位(19 g)、50%乙醇部位(17 g)、60%乙醇部位(23 g)、70%乙醇部位(24 g)、80%乙醇部位(5 g)和95%乙醇部位(20 g)。20%乙醇部位(49 g) 经Toyopearl HW-40C柱, 以甲醇-水(0∶100~100∶0) 梯度洗脱得到5个组分Fr.A.1~Fr.A.5。Fr.A.1 (25 g) 经SephadexLH-20柱色谱用甲醇-水(0∶100~100∶0) 梯度洗脱, 得到Fr.A.1.1~Fr.A.1.4共4个组分。Fr.A.1.1 (20 g) 经硅胶柱色谱(200~300目) 用二氯甲烷-甲醇(100∶0~0∶100) 梯度洗脱, 得到Fr.A.1.1.1~Fr.A.1.1.10共10个组分。Fr.A.1.1.8 (2 g) 经硅胶柱色谱(200~300目) 用二氯甲烷-甲醇(50∶1~0∶100) 梯度洗脱, 得到Fr.A.1.1.8.1~Fr.A.1.1.8.5共5个组分。Fr.A.1.1.8.3 (1 g) 上ODS柱色谱, 以甲醇-水(5∶95~100∶0) 梯度洗脱, 得到10个组分Fr.A.1.1.8.3.1~Fr.A.1.1.8.3.10。Fr.A.1.1.8.3.10 (25 mg) 经半制备高效液相纯化, 得到化合物4 (乙腈-水12∶88, 3.0 mL·min-1, tR = 17.7 min, 2 mg)。Fr.A.1.1.9 (2 g) 经硅胶柱色谱(200~300目) 用二氯甲烷-甲醇(50∶1~0∶100) 梯度洗脱, 得到Fr.A.1.1.9.1~Fr.A.1.1.9.4共4个组分。Fr.A.1.1.9.3 (134 mg) 经半制备高效液相纯化, 得到化合物2 (乙腈-水24∶76, 3.0 mL·min-1, tR = 28.3 min, 5 mg) 和化合物3 (乙腈-水24∶76, 3.0 mL·min-1, tR = 20.8 min, 35 mg)。Fr.A.1.1.9.4 (134 mg) 经ODS柱色谱, 以甲醇-水(5∶95~100∶0) 梯度洗脱, 得到10个组分Fr.A.1.1.9.4.1~Fr.A.1.1.9.4.10。Fr.A.1.1.9.4.1 (75 mg) 经半制备高效液相纯化, 得到化合物5 (乙腈-水14∶86, 3.0 mL·min-1, tR = 11.748 min, 9 mg)。Fr.A.1.2 (2 g) 经硅胶柱色谱(200~300目) 用二氯甲烷-甲醇(50∶1~0∶100) 梯度洗脱, 得到Fr.A.1.2.1~Fr.A.1.2.6共6个组分。Fr.A.1.2.6 (77 mg) 经半制备高效液相纯化, 得到化合物1 (乙腈-水19∶81, 3.0 mL·min-1, tR = 19.1 min, 13 mg)。Fr.A.2 (7.1 g) 经硅胶柱色谱(200~300目) 用二氯甲烷-甲醇(50∶1~0∶100) 梯度洗脱, 得到Fr.A.2.1~Fr.A.2.4共4个组分。Fr.A.2.4 (3 g) 上ODS柱色谱, 以甲醇-水(5∶95~100∶0) 梯度洗脱, 得到8个组分Fr.A.2.4.1~Fr.A.2.4.8。Fr.A.2.4.1 (54 mg) 经半制备高效液相纯化, 得到化合物6 (甲醇-水35∶65, 3.0 mL·min-1, tR = 14.9 min, 2 mg)。
称取化合物1.0 mg, 加入2.0 mg蜗牛酶和3 mL蒸馏水, 37 ℃水浴加热48 h。向干燥后的混合物中加1 mL蒸馏水, 用等体积乙酸乙酯萃取3次。水层浓缩干燥后加L-半胱氨酸甲酯盐酸盐1 mg, 吡啶1 mL, 在60 ℃下加热1.5 h。加入邻甲苯异硫氰酸酯20 μL, 60 ℃加热1.5 h。LC-MS分析混合物, 色谱条件: Agilent Eclipse XDS-C18 (3.5 μm, 3.0 mm×150 mm); 等度洗脱: 水(A)-甲醇(B), 0~28 min, 40% B; 柱温40 ℃, 流速0.3 mL·min-1; 质谱条件: 正离子模式。化合物1 (tR = 11.91 min)、2 (tR = 11.93 min) 与D-葡萄糖标准品(tR = 12.00 min) 的保留时间基本一致, 鉴定化合物12中葡萄糖的绝对构型为D型。
化合物1为淡黄色粉末, [α]$ {}_{\mathrm{D}}^{20} $ -25.0 (c 0.10, MeOH); IR (MeOH) vmax 3 244、1 613、1 456、1 202 cm-1; UV (MeOH) λmax (log ε): 209.0 (2.536) nm, HR-ESI-MS m/z 411.126 0 [M+Na]+, 计算值为411.126 2。1H NMR和13C NMR数据见表 1
化合物2为淡黄色粉末, [α]$ {}_{\mathrm{D}}^{20} $ -37.0 (c 0.10, MeOH); IR (MeOH) vmax 3 411、1 612、1 659、1 607、1 458、1 177、1 035 cm-1; UV (MeOH) λmax (log ε): 211.0 (2.814) nm, HR-ESI-MS m/z 425.141 9 [M+Na]+, 计算值为425.141 8。1H NMR和13C NMR数据见表 1
化合物3为淡黄色结晶, 易溶于甲醇, ESI-MS m/z 395.130 9 [M+Na]+, 化合物分子式为C17H24O91H NMR (500 MHz CD3OD) 谱中, δH 7.02 (1H, d, J = 8.4 Hz, H-5), 6.78 (1H, d, J = 2.5 Hz, H-2), 6.51 (1H, dd, J = 8.4, 2.5 Hz, H-6), 4.88 (1H, d, J = 7.1 Hz, H-1′), 3.90 (1H, dd, J = 12.1, 2.2 Hz, H-6′a), 3.75 (3H, s, 9-OCH3), 3.69 (1H, dd, J = 12.1, 5.8 Hz, H-6′b), 3.63 (3H, s, 3-OCH3), 3.42~3.48 (4H, m, H-2′-5′), 2.90 (2H, m, H-7), 2.61 (2H, m, H-8); 13C NMR (CD3OD, 125 MHz) 谱中, δC 131.3 (C-1), 108.5 (C-2), 160.9 (C-3), 157.7 (C-4), 103.2 (C-5), 123.2 (C-6), 35.5 (C-7), 26.3 (C-8), 175.9 (C-9), 52.0 (3-OCH3), 55.8 (9-OCH3), 102.7 (C-1′), 74.9 (C-2′), 78.2 (C-3′), 71.4 (C-4′), 78.2 (C-5′), 62.6 (C-6′)。以上NMR数据与文献[11]报道的citrusin E基本一致。
化合物4为白色粉末, 易溶于甲醇, ESI-MS m/z 395.129 4 [M+Na]+, 化合物分子式为C17H24O91H NMR (500 MHz CD3OD) 谱中, δH: 6.75 (1H, s, H-2, 6), 6.55 (1H, d, J =15.8 Hz, H-7), 6.33 (1H, dd, J =15.8, 6.4 Hz, H-8), 4.87 (1H, s, H-1′), 4.22 (2H, dd, J =5.5, 1.6 Hz, H-9), 3.86 (6H, s, 3, 5-OCH3), 3.78 (1H, dd, J = 12.0, 2.4 Hz, H-6′a), 3.66 (1H, dd, J = 12.0, 5.2 Hz, H-6′b), 3.41-3.48 (3H, m, H-2′-4′), 3.21 (1H, dd, J = 7.5, 2.3 Hz, H-5′); 13C NMR (CD3OD, 125 MHz) 谱中, δC: 135.3 (C-1), 105.5 (C-2), 154.4 (C-3), 135.9 (C-4), 154.4 (C-5), 105.5 (C-6), 131.3 (C-7), 130.0 (C-8), δC 63.6 (C-9), 57.0 (3, 5-OCH3), 105.3 (C-1′), 75.7 (C-2′), 77.8 (C-3′), 71.3 (C-4′), 77.8 (C-5′), 62.6 (C-6′)。以上NMR数据与文献[12]报道的紫丁香酚苷基本一致。
化合物5为白色粉末, 易溶于甲醇, ESI-MS m/z 365.119 5 [M+Na]+, 化合物分子式为C16H22O81H NMR (500 MHz CD3OD) 谱中, δH 7.11 (1H, d, J = 8.4 Hz, H-5), 7.07 (1H, d, J = 2.0 Hz, H-2), 6.95 (1H, dd, J = 8.4, 2.0 Hz, H-6), 6.54 (1H, d, J = 15.9 Hz, H-7), 6.28 (1H, dd, J = 15.9, 5.7 Hz, H-8), 4.89 (1H, d, J = 7.2 Hz, H-1′), 4.21 (2H, dd, J = 5.8, 1.6 Hz, H-9), 3.87 (3H, s, 3-OCH3), 3.69 (1H, dd, J = 12.0, 4.8 Hz, H-6′b), 3.33~3.55 (4H, m, H-2′-5′); 13C NMR (CD3OD, 125 MHz) 谱中, δC 133.7 (C-1), 111.4 (C-2), 150.9 (C-3), 147.6 (C-4), 117.9 (C-5), 120.7 (C-6), 131.3 (C-7), 128.9 (C-8), 63.7 (C-9), 56.7 (3-OCH3), 102.7 (C-1′), 74.9 (C-2′), 78.2 (C-3′), 71.3 (C-4′), 77.8 (C-5′), 62.5 (C-6′)。以上NMR数据与文献[13]报道的松柏苷基本一致。
化合物6为白色粉末, 易溶于甲醇, ESI-MS m/z 379.136 2 [M+Na]+, 化合物分子式为C17H24O81H NMR (500 MHz CD3OD) 谱中, δH 7.11 (1H, d, J = 8.4 Hz, H-5), 7.08 (1H, d, J = 2.0 Hz, H-2), 6.96 (1H, dd, J = 8.4, 2.0 Hz, H-6), 6.58 (1H, d, J = 16.0 Hz, H-7), 6.23 (1H, d, J = 16.0 Hz, H-8), 4.89 (1H, d, J = 7.2 Hz, H-1′), 4.08 (1H, dd, J = 12.1, 2.2 Hz, H-6′a), 3.88 (3H, s, 3-OCH3), 3.86 (1H, dd, J = 12.1, 5.7 Hz, H-6′b), 3.69 (2H, dd, J = 12.0, 4.4 Hz, H-9), 3.33~3.50 (4H, m, H-2′-5′), 3.37 (3H, s, 9-OCH3); 13C NMR (125 MHz CD3OD) 谱中, δC 134.1 (C-1), 110.3 (C-2), 150.3 (C-3), 147.9 (C-4), 116.2 (C-5), 118.4 (C-6), 133.7 (C-7), 126.0 (C-8), 74.2 (C-9), 58.1 (3-OCH3), 56.7 (9-OCH3), 103.1 (C-1′), 75.4 (C-2′), 77.8 (C-3′), 71.3 (C-4′), 78.2 (C-5′), 63.0 (C-6′)。以上NMR数据与文献[14]报道甲基松柏苷基本一致。
抗肾纤维化活性评价实验方法与课题组之前研究[15]一致。
