Eight compounds were isolated and purified from the ethyl acetate part of 70% acetone extract of Rehmannia glutinosa by various chromatographic techniques such as silica gel, MCI gel CHP-20, ODS, Toyopearl HW-40C, combined with TLC and semi-preparative HPLC. Their structures were elucidated by modern spectroscopy techniques (NMR, MS, UV, IR), and identified as neomartynoside A (1), osmanthuside B6 (2), martynoside (3), isomartynoside (4), (E)-p-hydroxycinnamic acid (5), caffeic acid (6), ferulic acid (7), and methyl caffeate (8). Compound 1 is a new phenylethanol glycoside, which was identified as neomartynoside A. Compound 2 was isolated from Rehmannia glutinosa for the first time. In addition, compounds 2, 6 and 7 significantly increased relative glucose consumption, showing potential hypoglycemic activity.

采用硅胶、MCI gel CHP-20、ODS、Toyopearl HW-40C等柱色谱技术, 并结合TLC和半制备液相等手段对覃怀1号鲜地黄70%丙酮提取物的乙酸乙酯部位进行分离纯化, 得到8个单体化合物。运用现代波谱学技术 (NMR、MS、UV、IR) 鉴定化合物的结构, 分别鉴定为地黄新苷A (1)、osmanthuside B6 (2)、地黄苷 (3)、异马蒂罗苷 (4)、反式对羟基肉桂酸 (5)、咖啡酸 (6)、阿魏酸 (7)、咖啡酸甲酯 (8)。其中, 化合物1为新苯乙醇苷, 鉴定为地黄新苷A, 化合物2为首次从地黄中分离得到。药理活性研究表明, 化合物267可以显著增加相对葡萄糖消耗量, 表现出潜在的降糖活性。

采用硅胶、MCI gel CHP-20、ODS、Toyopearl HW-40C等柱色谱技术, 并结合TLC和半制备液相等手段对覃怀1号鲜地黄70%丙酮提取物的乙酸乙酯部位进行分离纯化, 得到8个单体化合物。运用现代波谱学技术 (NMR、MS、UV、IR) 鉴定化合物的结构, 分别鉴定为地黄新苷A (1)、osmanthuside B6 (2)、地黄苷 (3)、异马蒂罗苷 (4)、反式对羟基肉桂酸 (5)、咖啡酸 (6)、阿魏酸 (7)、咖啡酸甲酯 (8)。其中, 化合物1为新苯乙醇苷, 鉴定为地黄新苷A, 化合物2为首次从地黄中分离得到。药理活性研究表明, 化合物267可以显著增加相对葡萄糖消耗量, 表现出潜在的降糖活性。

Eight compounds were isolated and purified from the ethyl acetate part of 70% acetone extract of Rehmannia glutinosa by various chromatographic techniques such as silica gel, MCI gel CHP-20, ODS, Toyopearl HW-40C, combined with TLC and semi-preparative HPLC. Their structures were elucidated by modern spectroscopy techniques (NMR, MS, UV, IR), and identified as neomartynoside A (1), osmanthuside B6 (2), martynoside (3), isomartynoside (4), (E)-p-hydroxycinnamic acid (5), caffeic acid (6), ferulic acid (7), and methyl caffeate (8). Compound 1 is a new phenylethanol glycoside, which was identified as neomartynoside A. Compound 2 was isolated from Rehmannia glutinosa for the first time. In addition, compounds 2, 6 and 7 significantly increased relative glucose consumption, showing potential hypoglycemic activity.

地黄为玄参科植物地黄 (Rehmannia glutinosa Libosch.) 的新鲜或干燥块根, 始载于《神农本草经》, 具有悠久的用药历史。地黄以鲜品入药习称鲜地黄, 其性寒, 味甘、苦, 归心、肝、肾经, 具有清热生津、凉血散瘀、解毒止痛等功效[1]。地黄中含有丰富的化学成分, 主要为环烯醚萜类、紫罗兰酮类和苯乙醇类化合物, 此外还包括三萜类、黄酮类、木脂素类、酚酸类等其他类化合物[2]。多样的化学成分为其药理学研究奠定坚实的基础。现代药理学研究表明, 地黄具有调节血糖和血脂[3]、抗衰老[4]、抗肿瘤[5]、抑菌[6]和抗抑郁[7]等多种生物活性。为了进一步寻找结构新颖的化学成分, 丰富其化学组成, 本课题组对覃怀1号鲜地黄70%丙酮提取物的乙酸乙酯部位进行系统的化学成分研究, 分离鉴定得到8个化合物, 包括4个苯乙醇苷类化合物: 地黄新苷A (1)、osmanthuside B6 (2)、地黄苷 (3)、异马蒂罗苷 (4); 4个苯丙素类化合物: 反式对羟基肉桂酸 (5)、咖啡酸 (6)、阿魏酸 (7)、咖啡酸甲酯 (8), 具体结构见图1。其中, 化合物1为新化合物, 化合物2首次从地黄中分离得到。
化合物1: 白色无定形粉末, 易溶于甲醇。根据准分子离子峰HR-ESIMS m/z 615.204 4 [M+Na]+ (计算值615.204 8), 推断其分子式为C29H36O13, 不饱和度为12。根据1H NMR和13C NMR (表1)、DEPT-135和HSQC谱图所提供的信息, 化合物1的结构中含有1个单取代苯环 [δH 7.21 (2H, m, H-2, 6), 7.19 (2H, overlapped, H-3, 5), 7.13 (1H, m, H-4)]; 1个ABX取代苯环 [δH 7.03 (1H, d, J = 2.0 Hz, H-2″)、6.77 (1H, d, J = 8.2 Hz, H-5″)、6.90 (1H, dd, J = 8.2, 2.0 Hz, H-6″)]; 一组反式双键氢信号 [δH 7.57 (1H, d, J = 15.9 Hz, H-7″)、6.29 (1H, d, J = 15.9 Hz, H-8″)]; 1个连氧亚甲基氢信号 [δH 3.99 (1H, m, H-8a)、3.78 (1H, m, H-8b)]; 1个亚甲基氢信号 [δH 2.92 (2H, td, J = 7.4, 2.5 Hz, H-7)]。根据13C NMR谱中出现的2个连氧次甲基碳信号 (δC 104.4、102.7) 以及1H NMR谱中2个连氧次甲基氢信号 [δH 4.34 (1H, d, J = 8.0 Hz, H-1′)、5.18 (1H, d, J = 1.6 Hz, H-1‴)], 推测化合物1的结构中含有两个单糖。结合1H NMR谱中一组连氧亚甲基氢信号 [δH 4.49 (1H, dd, J = 11.9, 2.2 Hz, H-6′a)、4.38 (1H, m, H-6′b), δC 64.6], 推断结构中含有1个单糖为葡萄糖; 结合双峰甲基氢信号 [δH 1.25 (3H, d, J = 6.2 Hz, H-6‴), δC 17.9] 和端基氢信号 [δH 5.18 (1H, d, J = 1.6 Hz, H-1‴), δC 102.7], 推断另一个单糖为鼠李糖。
进一步采用2D NMR谱将化合物1中的结构片段进行连接。1H-1H COSY谱 (图2) 中, H-7 (δH 2.92) 和H-8 (δH 3.99/3.78) 相关, 确定结构中含有一个乙氧基结构片段。HMBC谱 (图2) 中, H-2, 6 (δH 7.21) 和C-3, 5 (δC 129.3)、C-4 (δC 127.2) 相关, H-3, 5 (δH 7.19) 和C-1 (δC 139.9) 相关, H-7 (δH 2.92) 和C-1 (δC 139.9)、C-2, 6 (δC 130.0) 相关, H-8 (δH 3.99/3.78) 和C-1 (δC 139.9) 相关, 表明化合物1的苷元部分为单取代苯乙醇结构。此外, HMBC谱中, H-5″ (δH 6.77) 和C-1″ (δC 127.7) 相关, H-2″ (δH 7.03) 和C-3″ (δC 149.6)、C-4″ (δC 146.8) 相关, H-6″ (δH 6.90) 和C-2″ (δC 115.1) 相关, H-8″ (δH 6.29) 和C-1″ (δC 127.7)、C-9″ (δC 169.1) 相关, 推测化合物1中含有1个咖啡酸的结构片段。糖基间的连接及其在苷元上的连接位置, 可进一步通过HMBC谱 (图2) 确定。在HMBC谱中, H-8 (δH 3.99/3.78) 和C-1′ (δC 104.4) 相关, 表明葡萄糖端基碳和苯乙醇的C-8位相连形成苷键; 同时, H-6′ (δH 4.49/4.38) 和C-9″ (δC 169.1) 相关, 表明葡萄糖的6′位羟基与9″位羰基成酯; 此外, 葡萄糖的3′位碳向低场位移至84.0, 且H-3′ (δH 3.52) 和C-1‴ (δC 102.7) 相关, 进一步确定鼠李糖连接在葡萄糖的C-3′位。
根据氢谱中端基氢的偶合常数确定单糖的相对构型。在1H NMR中, 根据端基氢的偶合常数JH1′-H2′ = 8.0 Hz, 可知葡萄糖的端基碳构型为β-型; 根据端基氢小的偶合常数JH1‴-H2‴ = 1.6 Hz, 可知鼠李糖的端基碳构型为α-型[8]。最后, 采用酸水解反应鉴定化合物1中的单糖分别为D-葡萄糖和L-鼠李糖, 其糖链为α-L-鼠李糖基(1→3)-β-D-葡萄糖基, 最终确定化合物1的结构如图1所示, 命名为地黄新苷A。
本实验对化合物18进行降糖活性筛选, 结果如图3图4所示。CCK-8实验结果显示: 化合物18可以提高胰岛素诱导的HepG2细胞抵抗模型的细胞活力, 表明化合物18无细胞毒作用。相对葡萄糖消耗量实验结果显示: 与正常组相比, 模型组的相对葡萄糖消耗量显著降低 (P < 0.05), 表明胰岛素抵抗模型造模成功; 与模型组相比, 化合物267可以显著增加HepG2细胞的相对葡萄糖消耗量 (P < 0.05或P < 0.01), 表明化合物267具有潜在的降糖活性。
覃怀1号鲜地黄由河南中医药大学与仲景宛西制药股份有限公司以“北京3号”为母本, 与“金九”“狮子头”“怀丰”“85-5”“红薯王”等品种经集团杂交系统选育而成, 2021年12月通过河南省中药材品种鉴定专业委员会鉴定为地黄新品种 (鉴定编号2021028)。实验用覃怀1号鲜地黄于2021年11月采自河南省温县, 经河南中医药大学药学院董诚明教授鉴定为玄参科植物地黄Rehmannia glutinosa Libosch.的新鲜块根, 植物标本 (No. 20211220) 保存于河南省中药开发工程技术研究中心。
AVANCE Ⅲ 500型核磁共振仪、maxis HD型飞行时间质谱 (德国布鲁克公司); EVO300紫外分光光度计型紫外检测器、Nicolet IS 10傅里叶变换红外光谱仪 (美国Thermo Scientific公司); ZF-90型暗箱式紫外投射仪 (上海宝山顾村电光仪器厂); CA-1115A型冷冻水循环装置、OSB-2200型旋转蒸发仪、NVP-1000型隔膜真空泵 (上海爱朗仪器有限公司); LC-20AT型半制备液相色谱仪 (日本岛津公司); COSMOSIL 5C18-MS-Ⅱ色谱柱 (10 mm × 250 mm, 5 µm; 日本Tesque公司); Toyopearl HW-40C (日本TOSOH公司); MCI gel CHP-20 (日本三菱化学公司); C18 ODS (日本YMC有限公司); 柱色谱硅胶 (100~200和200~300目, 青岛海洋化工厂); 甲醇、乙腈 (色谱纯, 河南宏材化工有限公司); 分析纯试剂 (天津富宇精细化工有限公司)。
取覃怀1号鲜地黄70 kg, 使用70%丙酮组织破碎提取两次, 合并滤液, 减压浓缩得总浸膏19.18 kg。总浸膏加入适量蒸馏水分散, 依次使用石油醚、乙酸乙酯、正丁醇萃取, 得到石油醚部位、乙酸乙酯部位、正丁醇部位和水部位。
乙酸乙酯部位 (179.04 g) 经硅胶柱色谱分离, 以二氯甲烷-甲醇 (200∶1→1∶1) 梯度洗脱, 得到11个组分E1~E11。E9 (10.33 g) 采用Toyopearl HW-40C凝胶柱, 以甲醇-水 (80∶20→100∶0) 梯度洗脱, 得到7个组分E9-1~E9-7。E9-4 (7.0 g) 采用C18 ODS柱分离, 使用甲醇-水 (40∶60→100∶0) 梯度洗脱, 得到7个组分E9-4-1~E9-4-7; 其中E9-4-4 (2.6 g) 经硅胶柱色谱分离, 得到12个组分E9-4-4-1~E9-4-4-12。E9-4-4-4 (150 mg) 经半制备液相 (乙腈-水, 23.5∶76.5) 进一步纯化得到化合物1 (8.50 mg, tR = 44.8 min)。E9-4-4-6 (70 mg) 经半制备液相 (甲醇-水, 40∶60) 纯化得到化合物3 (5.35 mg, tR = 32.7 min) 和4 (4.28 mg, tR = 36.8 min)。E9-4-6 (2.25 g) 经硅胶柱色谱分离, 二氯甲烷-甲醇 (50∶1→1∶1) 梯度洗脱, 得到10个组分E9-4-6-1~E9-4-6-10。E9-4-6-4 (55 mg) 经半制备液相 (乙腈-水, 22∶78) 纯化得到化合物2 (2.85 mg, tR = 42.5 min)。E4 (22.5 g) 上硅胶柱分离, 使用二氯甲烷-甲醇 (200∶1→1∶1) 进行梯度洗脱, 得到8个组分E4-1~E4-8。E4-5 (4.5 g) 采用MCI gel CHP-20柱色谱分离, 甲醇-水 (10∶90→100∶0) 梯度洗脱, 得到8个组分E4-5-1~E4-5-8。E4-5-4 (1.85 g) 经硅胶柱色谱分离, 使用二氯甲烷-甲醇 (50∶1→1∶1) 进行梯度洗脱, 得到10个组分E4-5-4-1~E4-5-4-10。E4-5-4-4 (50 mg) 经半制备液相 (甲醇-水, 42∶58) 纯化得到化合物5 (8.65 mg, tR = 39.6 min)。E4-5-4-7 (85 mg) 经半制备液相 (乙腈-水, 21∶79) 分离纯化得到化合物7 (3.50 mg, tR = 39.5 min) 和8 (2.82 mg, tR = 42.2 min)。E5 (12.2 g) 上ODS柱, 使用甲醇-水 (20∶80→100∶0) 梯度洗脱, 得到8个组分E5-1~E5-8; 其中E5-3 (2.5 g) 经硅胶柱分离, 得到10个组分E5-3-1~E5-3-10。E5-3-6 (70 mg) 经半制备液相 (甲醇-水, 30∶70) 纯化得到化合物6 (10.5 mg, tR = 39.5 min)。
化合物1 (1.0 mg) 加入2 mol·L-1盐酸水溶液2.5 mL, 80 ℃下加热3 h。反应混合物中加入乙酸乙酯萃取3次, 每次3 mL。低温减压条件下浓缩水层, 浓缩过程中反复加水稀释, 得到中性残留物。将残留物和D-葡萄糖、L-鼠李糖对照品溶于色谱甲醇 (1.5 mL), 采用以下条件进行高效液相分析: CHIRALPAK AD-H色谱柱为250 mm × 4.6 mm, 5 μm; 流动相为正己烷-无水乙醇 (82∶18); 流速为0.5 mL·min-1; 蒸发光散射检测器漂移管温度为75 ℃, 气体压力为20.0 psi (1 psi ≈ 6.9 kPa), 柱温为30 ℃。通过比较相同液相条件下D-葡萄糖 (tR = 21.4 min) 和L-鼠李糖 (tR = 23.5 min) 的保留时间, 确定化合物1中的单糖分别为D-葡萄糖和L-鼠李糖。

化合物5  无色油状物。ESI-MS m/z 163 [M-H]-, 分子式为C9H8O31H NMR (500 MHz, CD3OD) δH: 7.60 (1H, d, J = 15.8 Hz, H-8), 7.44 (2H, d, J = 8.7 Hz, H-2, 6), 6.81 (2H, d, J = 8.7 Hz, H-3, 5), 6.28 (1H, d, J = 15.8 Hz, H-7); 13C NMR (125 MHz, CD3OD) δC: 146.6 (C-1), 131.1 (C-2, 6), 116.8 (C-3, 5), 161.1 (C-4), 127.2 (C-7), 115.6 (C-8), 171.1 (C-9)。以上数据与文献[12]报道基本一致, 鉴定化合物5为反式对羟基肉桂酸 [(E)-p-hydroxycinnamic acid]。
化合物6  无色油状物。ESI-MS m/z 179 [M-H]-, 分子式为C9H8O41H NMR (500 MHz, CD3OD) δH: 7.53 (1H, d, J = 15.8 Hz, H-7), 7.04 (1H, d, J = 1.8 Hz, H-2), 6.93 (1H, dd, J = 8.1, 1.8 Hz, H-6), 6.78 (1H, d, J = 8.1 Hz, H-5), 6.22 (1H, d, J = 15.8 Hz, H-8); 13C NMR (125 MHz, CD3OD) δC: 127.8 (C-1), 115.1 (C-2), 146.7 (C-3), 149.4 (C-4), 116.5 (C-5), 122.8 (C-6), 147.0 (C-7), 115.6 (C-8), 171.1 (C-9)。以上数据与文献[13]报道基本一致, 鉴定化合物6为咖啡酸 (caffeic acid)。
化合物7  白色无定形粉末。ESI-MS m/z 217 [M+Na]+, 分子式为C10H10O41H NMR (500 MHz, CD3OD) δH: 7.59 (1H, d, J = 15.9 Hz, H-7), 7.17 (1H, d, J = 1.8 Hz, H-2), 7.06 (1H, dd, J = 8.3, 1.8 Hz, H-6), 6.81 (1H, d, J = 8.3 Hz, H-5), 6.31 (1H, d, J = 15.9 Hz, H-8), 3.89 (3H, s, 3-OCH3); 13C NMR (125 MHz, CD3OD) δC: 127.8 (C-1), 116.4 (C-2), 150.5 (C-3), 149.3 (C-4), 116.0 (C-5), 123.9 (C-6), 146.8 (C-7), 111.7 (C-8), 171.1 (C-9), 56.4 (3-OCH3)。以上数据与文献[14]报道基本一致, 鉴定化合物7为阿魏酸 (ferulic acid)。
化合物8  白色无定形粉末。ESI-MS m/z 193 [M-H]-, 分子式为C10H10O41H NMR (500 MHz, CD3OD) δH: 7.54 (1H, d, J = 15.9 Hz, H-7), 7.03 (1H, brs, H-2), 6.94 (1H, d, J = 8.2 Hz, H-6), 6.77 (1H, d, J = 8.1 Hz, H-5), 6.26 (1H, d, J = 15.9 Hz, H-8), 3.75 (3H, s, 3-OCH3); 13C NMR (125 MHz, CD3OD) δC: 127.7 (C-1), 115.1 (C-2), 146.9 (C-3), 149.6 (C-4), 122.9 (C-5), 116.5 (C-6), 146.9 (C-7), 114.8 (C-8), 169.7 (C-9), 52.0 (9-OCH3)。以上数据与文献[15]报道基本一致, 鉴定化合物8为咖啡酸甲酯 (methyl caffeate)。
HepG2细胞使用含10%胎牛血清的高糖DMEM培养基, 置于37 ℃、5% CO2恒温培养箱中培养。将HepG2细胞接种于96孔板, 密度每孔为1×105个, 每组设置6个复孔。培养24 h后, 更换为无血清培养基配置的胰岛素 (2×10-6 mol·L-1, 每孔100 μL), 同时加入二甲双胍 (20 μmol·L-1, 每孔100 μL) 或含单体化合物18 (20 μmol·L-1, 每孔100 μL), 继续孵育24 h。
细胞处理后, 加入CCK-8试剂10 μL, 37 ℃孵育1 h, 使用微孔板分光光度计在450 nm处测定每孔的光密度 (OD) 值。
培养结束后, 收集各组培养基, 使用葡萄糖测定试剂盒测定葡萄糖浓度。原DMEM培养基的葡萄糖含量减去实验组培养基的葡萄糖含量, 得到葡萄糖消耗量 (GC)。相对葡萄糖消耗量 (RGC) 计算公式为: RGC = GC / OD。
