Non-small cell lung cancer (NSCLC) is the primary pathological type of lung cancer. Osimertinib, as a third-generation EGFR-TKI (epidermal growth factor receptor-tyrosine kinase inhibitor) targeted drug, effectively prolong the progression-free survival of patients with EGFR-mutated NSCLC. However, drug resistance limits the efficacy of osimertinib. Traditional Chinese medicine can effectively delay the resistance to EGFR-TKIs. In this study, Cucurbitacin B (CuB) was investigated to analyze its pharmacological effects in osimertinib-resistant NSCLC cells through cell viability, migration, and invasion experiments. Public databases were used to screen potential targets of CuB in osimertinib-resistant NSCLC cells, and the interactions between CuB and potential targets were verified through molecular docking, cellular thermal shift assay (CETSA), and microscale thermophoresis (MST). Western blot was used to detect the effects of CuB on downstream pathways of the targets. The results showed that CuB significantly reduced the proliferation activity of osimertinib-resistant NSCLC cells and inhibited the migration and invasion abilities of tumor cells. Mechanistically, CuB inhibited the expression of ERK and AKT molecules by binding to the tyrosine kinase receptor AXL. In summary, CuB exhibits resistance to osimertinib-resistant NSCLC by inhibiting the migration and invasion of resistant cells through regulating the abnormal activation of the AXL-ERK/AKT axis. This study provides a basis for the pre-clinical in vitro efficacy of CuB in the treatment of osimertinib targeted resistance in NSCLC and theoretical support for the development of clinical drug combinations.

非小细胞肺癌(non-small cell lung cancer, NSCLC) 是肺癌的主要病理类型, 奥希替尼作为第三代表皮生长因子受体酪氨酸激酶抑制剂(epidermal growth factor receptor-tyrosine kinase inhibitor, EGFR-TKI) 靶向药物, 能有效延长EGFR突变NSCLC患者的无进展生存期。然而, 耐药性限制了奥希替尼的疗效, 而中医药能有效延缓EGFR-TKIs的耐药性。本研究以葫芦素B (cucurbitacin B, CuB) 为研究对象, 通过细胞活力检测和细胞迁移、侵袭实验分析葫芦素B在奥希替尼耐药NSCLC细胞中的药效作用; 使用公共数据库筛选葫芦素B在奥希替尼耐药NSCLC细胞中的潜在靶点, 并通过分子对接、细胞热位移实验(cellular thermal shift assay, CETSA) 和微量热泳动实验(microscale thermophoresis, MST) 验证葫芦素B与潜在靶点之间的相互作用; 免疫印迹实验检测葫芦素B对靶点下游信号通路的影响。结果显示, 葫芦素B能显著降低奥希替尼耐药NSCLC细胞的生长活力, 抑制细胞的迁移和侵袭能力; 机制上, 葫芦素B通过结合AXL进而抑制其下游细胞外信号调节激酶(extracellular regulated protein kinases, ERK) 和蛋白激酶B (protein kinase B, AKT) 的激活。综上所述, 葫芦素B具有缓解NSCLC奥希替尼治疗抵抗的潜在作用, 其可通过下调AXL-ERK/AKT轴的异常激活, 抑制耐药细胞的迁移、侵袭。本研究为葫芦素B治疗NSCLC奥希替尼靶向耐药提供了临床前体外药效的依据, 也为临床联合用药开发提供了理论支持。

非小细胞肺癌(non-small cell lung cancer, NSCLC) 是肺癌的主要病理类型, 奥希替尼作为第三代表皮生长因子受体酪氨酸激酶抑制剂(epidermal growth factor receptor-tyrosine kinase inhibitor, EGFR-TKI) 靶向药物, 能有效延长EGFR突变NSCLC患者的无进展生存期。然而, 耐药性限制了奥希替尼的疗效, 而中医药能有效延缓EGFR-TKIs的耐药性。本研究以葫芦素B (cucurbitacin B, CuB) 为研究对象, 通过细胞活力检测和细胞迁移、侵袭实验分析葫芦素B在奥希替尼耐药NSCLC细胞中的药效作用; 使用公共数据库筛选葫芦素B在奥希替尼耐药NSCLC细胞中的潜在靶点, 并通过分子对接、细胞热位移实验(cellular thermal shift assay, CETSA) 和微量热泳动实验(microscale thermophoresis, MST) 验证葫芦素B与潜在靶点之间的相互作用; 免疫印迹实验检测葫芦素B对靶点下游信号通路的影响。结果显示, 葫芦素B能显著降低奥希替尼耐药NSCLC细胞的生长活力, 抑制细胞的迁移和侵袭能力; 机制上, 葫芦素B通过结合AXL进而抑制其下游细胞外信号调节激酶(extracellular regulated protein kinases, ERK) 和蛋白激酶B (protein kinase B, AKT) 的激活。综上所述, 葫芦素B具有缓解NSCLC奥希替尼治疗抵抗的潜在作用, 其可通过下调AXL-ERK/AKT轴的异常激活, 抑制耐药细胞的迁移、侵袭。本研究为葫芦素B治疗NSCLC奥希替尼靶向耐药提供了临床前体外药效的依据, 也为临床联合用药开发提供了理论支持。

Non-small cell lung cancer (NSCLC) is the primary pathological type of lung cancer. Osimertinib, as a third-generation EGFR-TKI (epidermal growth factor receptor-tyrosine kinase inhibitor) targeted drug, effectively prolong the progression-free survival of patients with EGFR-mutated NSCLC. However, drug resistance limits the efficacy of osimertinib. Traditional Chinese medicine can effectively delay the resistance to EGFR-TKIs. In this study, Cucurbitacin B (CuB) was investigated to analyze its pharmacological effects in osimertinib-resistant NSCLC cells through cell viability, migration, and invasion experiments. Public databases were used to screen potential targets of CuB in osimertinib-resistant NSCLC cells, and the interactions between CuB and potential targets were verified through molecular docking, cellular thermal shift assay (CETSA), and microscale thermophoresis (MST). Western blot was used to detect the effects of CuB on downstream pathways of the targets. The results showed that CuB significantly reduced the proliferation activity of osimertinib-resistant NSCLC cells and inhibited the migration and invasion abilities of tumor cells. Mechanistically, CuB inhibited the expression of ERK and AKT molecules by binding to the tyrosine kinase receptor AXL. In summary, CuB exhibits resistance to osimertinib-resistant NSCLC by inhibiting the migration and invasion of resistant cells through regulating the abnormal activation of the AXL-ERK/AKT axis. This study provides a basis for the pre-clinical in vitro efficacy of CuB in the treatment of osimertinib targeted resistance in NSCLC and theoretical support for the development of clinical drug combinations.

肺癌是我国发生率和死亡率居前的恶性肿瘤之一, 而非小细胞肺癌(non-small cell lung cancer, NSCLC) 是肺癌的主要病理类型, 约占80%[1]。表皮生长因子受体(epithelial growth factor receptor, EGFR) 是NSCLC发生、发展的关键驱动基因[2]。目前, 针对EGFR突变位点开发的第三代靶向药物奥希替尼能有效延长NSCLC患者的总生存期和无进展生存期, 但耐药性的产生却制约了该药物的疗效。研究表明, EGFR非依赖性旁路激活是奥希替尼耐药性的重要机制, 其通过多种酪氨酸激酶受体的异常激活所介导[3], 比如间质表皮转化因子(mesenchymal to epithelial transition factor, MET)、原癌基因酪氨酸激酶受体(proto-oncogene tyrosine-protein kinase receptor, RET)、酪氨酸激酶受体AXL、间变性淋巴瘤激酶(anaplastic lymphoma kinase, ALK)、成纤维细胞生长因子受体(fibroblast growth factor receptor, FGFR)、胰岛素样生长因子1受体(insulin-like growth factor 1 receptor, IGF1R) 等。其中, AXL是奥希替尼耐药性通路中的关键分子之一, 其在耐药细胞中的异常高表达及激活, 诱导肿瘤细胞发生上皮间质转化(epithelial-mesenchymal transition, EMT), 促进肿瘤细胞的迁移和侵袭[4, 5]。目前, 临床研究开发的多靶点抑制剂存在一定的脱靶效应导致毒副作用的发生, 尚无有效针对AXL的特异性抑制剂。因此, 筛选AXL抑制剂并探究其药效作用, 有助于为临床治疗奥希替尼耐药且AXL高表达患者的药物开发提供新的策略和参考。
中药是我国丰富的药用资源, 毒副作用小, 在NSCLC治疗中疗效显著。如邓洪斌课题组[6]研究发现金丝桃苷通过抑制NSCLC细胞中c-Myc蛋白的表达, 抑制PD-L1转录水平的表达, 发挥抗肿瘤作用。此外, 研究显示中药可以延缓EGFR-TKI耐药的发生, 并在产生耐药性后增强EGFR-TKI的抗肿瘤效果, 起到协同增效的作用[7]。如杨梅素是一种从藤茶茎叶中或杨梅树皮提取的黄酮醇类化合物。Song等[8]发现杨梅素可以抑制β-catenin蛋白表达, 显著促进A549细胞凋亡, 增加A549细胞对厄洛替尼的敏感性, 提示杨梅素联合厄洛替尼对EGFR-TKI获得性耐药有一定的逆转作用。淫羊藿素是一种来自淫羊藿属植物的戊烯基类黄酮衍生物, Han等[9]研究发现淫羊藿素可能通过调控PI3K-AKT通路来抑制EGFR-TKIs耐药细胞的增殖, 降低其集落形成、迁移侵袭能力, 促进细胞凋亡, 从而发挥抗NSCLC EGFR-TKIs耐药的作用。此外, 知母可能通过促进氧化应激诱导细胞死亡, 逆转PI3K/AKT通路异常活化所诱导的吉非替尼耐药性[10]。然而, 目前有关中药在第三代EGFR-TKI药物奥希替尼治疗抵抗中的相关研究尚少。葫芦素B (cucurbitacin B, CuB) 是从甜瓜蒂等葫芦科植物中提取的一种四环三萜类化合物, 是葫芦素家族中含量最高的成员[11], 具有抗癌、抗炎等广泛的药理活性。Qiu等[12]在对吉非替尼耐药NSCLC PC9细胞检测发现, 葫芦素B能够显著抑制PC9吉非替尼耐药细胞的增殖, 并阻滞细胞G2/M期进程, 提示葫芦素B参与调控EGFR-TKI的耐药性。因此, 本文旨在探究葫芦素B在奥希替尼耐药NSCLC细胞中的作用及其潜在分子机制。
试剂与仪器  葫芦素B (CAS-6199-67-3, 厂商编号T3404)、奥希替尼(CAS-1421373-65-0, 厂商编号T2490) 均购自上海陶术生物科技有限公司; Gibco RPMI Medium 1640培养基(批号6123192)、0.25%胰酶-EDTA溶液(货号25200-056)、青-链霉素溶液(货号15140-122)、蛋白酶/磷酸酶抑制剂(货号A32959) 均购自美国Thermo Fisher Scientific公司; Certified Fetal Bovine Serum特级胎牛血清(货号C04001-500)、磷酸盐缓冲液(PBS, 货号C3580-0500) 均购自上海逍鹏生物科技有限公司; Cell Counting Kit 8 (CCK8, 货号A311-02-AA) 购自南京诺唯赞生物科技有限公司; 二甲基亚砜(DMSO, 货号D4540) 购自上海Sigma-Aldrich公司; RIPA裂解液(中) (货号P0013C)、Western一抗稀释液(货号P0023A)、SDS-PAGE蛋白上样缓冲液(货号P0015L)、结晶紫染液(货号C0121-100mL) 均购自上海碧云天生物技术有限公司; 封闭用BSA (货号P1621) 购自北京普利莱基因技术有限公司; β-actin抗体(货号AP0060) 购自美国Bioworld公司; E-cadherin抗体(货号14472S)、vimentin抗体(货号5471S)、anti rabbit IgG (货号7074P2) 均购自美国Cell Signaling Technology公司; PAGE凝胶快速制备试剂盒(货号20324ES62) 购自上海翌圣生物科技股份有限公司; Transwell小室(货号3413) 购自美国Corning公司; CO2培养箱(型号HERA cell 150)、全波长多功能酶标仪(型号Varioskan flash) 均购自美国Thermo公司; 生物安全柜(型号Purifier Class Ⅱ) 购自美国Labconco公司; 倒置显微镜(型号CKX 41) 购自日本Olympus公司。
细胞培养与活力测试  奥希替尼耐药NSCLC细胞(PC9-OR) 用含10% FBS, 100 μg·mL-1青霉素和100 μg·mL-1链霉素的RPMI 1640培养基, 在37 ℃、5% CO2的培养箱中持续培养。取对数生长期的细胞, 以每孔5×103个细胞, 每孔100 μL体积, 接种于96孔板中。培养24 h后, 用不同浓度的化合物干预细胞, DMSO作为溶剂对照。培养48 h后, 加入10 μL CCK-8溶液继续孵育1~2 h, 用酶标仪在测定波长450 nm和参比波长630 nm处检测吸光值。细胞存活率计算公式: 存活率= (A450 nm-A630 nm) treated/(A450 nm-A630 nm)control×100%。
Transwell迁移实验  提前取处于对数生长期的PC9-OR细胞, 饥饿处理24 h。取出Transwell小室, 置于24孔板中, 加入已预热的无血清培养基, 孵育2 h。取饥饿处理后的细胞稀释成每毫升2×105个, 500 μL接种于小室, 下层加入有血清含药培养基, 培养48 h。用棉签擦拭小室上表面, PBS润洗后, 加入甲醇固定20 min; 吸除甲醇, PBS润洗后, 加入0.1%结晶紫染色30 min, 蒸馏水浸洗风干后, 显微镜观察计数。
Transwell侵袭实验  提前取处于对数生长期的PC9-OR细胞, 饥饿处理24 h。取出Transwell小室, 置于24孔板中, 加入已预热的无血清培养基, 孵育2 h。将基质胶与无血清培养基按1∶8的比例混匀, 取100 μL加入小室上层, 37 ℃孵育2 h。取饥饿处理后的细胞稀释成每毫升2×105个, 500 μL接种于小室, 下层加入有血清含药培养基, 培养48 h。用棉签擦拭小室上表面, PBS润洗后, 加入甲醇固定20 min; 吸除甲醇, PBS润洗后, 加入0.1%结晶紫染色30 min, 蒸馏水浸洗风干后, 显微镜观察计数。
蛋白免疫印迹实验  取对数生长期PC9-OR细胞, 以4×105个/孔接种于六孔板中, 培养24 h后, 加入不同浓度的葫芦素B干预48 h, DMSO作为溶剂对照, 奥希替尼为阳性药物组。药物干预48 h后, 加入含蛋白酶、磷酸酶抑制剂的RIPA裂解液, 收集细胞裂解物, 提取细胞总蛋白, 经SDS-PAGE电泳、转膜、封闭后, 一抗4 ℃孵育过夜。次日, 加入相应的二抗孵育2 h, 洗膜后, 曝光仪成像, 使用Image J分析图片。
生物信息学分析  使用PubChem数据库获得天然产物葫芦素B的SMILES号, 利用Swiss Target Prediction数据库预测葫芦素B的作用靶点。检索GeneCards数据库筛选NSCLC相关的疾病靶点。通过DAVID Bioinformatics Resources进行GO功能与KEGG通路富集分析。采用PDB数据库检索AXL蛋白三维结构, PubChem数据库检索天然产物葫芦素B结构, 通过Cavity-detection guided Blind Docking进行分子对接分析。
微量热泳动(microscale thermophoresis)  将AXL蛋白进行赖氨酸标记后, 测定蛋白荧光强度。在PCR管中使用结合反应缓冲液将葫芦素B梯度稀释16个浓度。取10 μL被标记的AXL蛋白分别与PCR管中不同浓度的葫芦素B溶液按照1∶1的比例等体积混合, 毛细管吸取混合液, 置于微量热泳动仪中, 收集并导出数据, 使用MO.Affinity Analysis软件进行数据分析。
细胞热位移实验(cellular thermal shift assay, CETSA)  使用DMSO和葫芦素B处理对数生长的PC9-OR细胞(每毫升2.0×108个细胞) 4 h后, PBS冲洗一次, 弃去PBS。使用细胞刮刀将细胞刮下并转移至1.5 EP管中, 1 000 r·min-1离心3 min后弃掉上清, 加入1 mL含有PMSF的PBS混匀, 分装至0.2 mL PCR管中。在PCR仪中按照设置的温度(40、44、48、52、56、60、64、68、72 ℃) 加热样品3 min, 0 ℃不加热。然后用液氮对细胞悬浮液进行3次冻融循环, 以溶解细胞。将细胞悬浮液4 ℃、10 000 r·min-1离心10 min, 将上清液转移到新的PCR小管中; 加入上样缓冲液(4× loading buffer) 混匀后, 100 ℃加热8 min。
统计学方法  采用GraphPad 9.0进行统计学分析, 计量资料以x±s表示。组间比较采用单因素方差分析, P < 0.05为差异有统计学意义。
为探究葫芦素B对奥希替尼耐药NSCLC细胞的药效影响, 本研究分别在不加药、奥希替尼单药、葫芦素B单药以及先加奥希替尼再加葫芦素B的处理条件下, 处理48 h后检测奥希替尼耐药NSCLC细胞的增殖活力变化情况。结果显示, 葫芦素B与奥希替尼联用之后能显著降低奥希替尼耐药NSCLC细胞的细胞活力(图 1)。
取对数生长期奥希替尼耐药NSCLC细胞经饥饿处理后接种于小室中, 培养24 h后加入葫芦素B处理48 h, 对细胞运动能力进行检测。Transwell结果显示, 葫芦素B给药后细胞迁移(图 2A) 和侵袭(图 2B) 的数目显著减少, 高浓度效果比低浓度效果更显著; 同时, 较葫芦素B单药处理, 葫芦素B与奥希替尼联用抑制效果显著, 提示葫芦素B能够抑制奥希替尼耐药NSCLC细胞的迁移和侵袭。
为探究葫芦素B在NSCLC奥希替尼耐药性调控中的分子机制, 通过检索Swiss Target Prediction数据库获得100个葫芦素B潜在作用靶分子; 同时, 检索GeneCards数据库筛选已知NSCLC疾病相关分子22 946个。韦恩图分析上述两个数据集, 获得94个葫芦素B在NSCLC中的潜在作用靶点。针对94个潜在作用靶点行GO分析发现, 葫芦素B主要与蛋白磷酸化、信号转导、受体酪氨酸激酶信号通路等功能密切相关(图 3A); KEGG富集分析显示, 葫芦素B可能参与EGFR酪氨酸激酶抑制剂耐药性信号通路等(图 3B)。
本课题组根据上述的筛选结果, 从94个葫芦素B潜在作用靶点中选取了与EGFR耐药最相关的前10个靶点, 分别为AXL、MET、ALK、FGFR、mTOR、MAPK1、KDR、IGF1R、RET、BCL2, 进行了分子对接模拟。如图 4A结果显示, 相较于其他9个EGFR耐药相关靶点, 葫芦素B与AXL的结合能最低(-9.1 kcal·mol-1), 提示二者结合亲和力较强。随后, 本研究对葫芦素B与AXL的结合进行进一步验证。细胞热位移实验检测发现, 葫芦素B能够显著增加AXL在不同温度下的稳定性(图 4B)。微量热泳动实验证实, 葫芦素B能直接结合AXL, 且二者的互作随葫芦素B浓度的增高而增强, 其亲和力的平衡解离常数Kd值为22.5 μmol-1 (图 4C)。综上提示, 葫芦素B可能通过直接结合AXL参与缓解NSCLC奥希替尼治疗抵抗。
研究表明, AXL通过诱导EMT发生参与促进肿瘤细胞的迁移和侵袭[13]。免疫印迹实验检测发现, 与空白对照组和葫芦素B单药处理相比, 葫芦素B联合奥希替尼处理后显著降低EMT标志分子vimentin/E-cadherin蛋白表达量比值(图 5AB), 提示葫芦素B可能通过AXL调控肿瘤迁移侵袭相关标志分子的表达。
为进一步探究葫芦素B对AXL下游信号通路的影响, 免疫印迹实验对AXL下游信号通路分子的表达与激活进行检测。结果显示, 葫芦素B联合奥希替尼处理后显著抑制AKT和ERK的蛋白磷酸化表达水平(图 6AB), 提示葫芦素B结合AXL抑制其下游信号通路的异常激活, 是介导降低奥希替尼耐药NSCLC细胞迁移侵袭能力的重要机制之一。
肺癌是全球发病率和死亡率居前的恶性肿瘤。2022年, 新增病例近250万, 占全球癌症新增病例的1/8 (占全球癌症总数的12.4%)[14]。肺癌的治疗手段主要包括手术、放化疗、靶向治疗及免疫治疗。其中, 分子靶向药是治疗非小细胞肺癌的有效手段之一, 近年来奥希替尼作为一线治疗药物, 能有效地延长患者的无进展生存期[2], 但是随着治疗时间的延长, 耐药性的出现限制了其疗效。目前, 针对奥希替尼耐药相关分子的抑制剂开发多数处于临床前I/II期研究, 且部分抑制剂伴有一定毒副作用, 因此寻找低毒高效的药物来缓解奥希替尼耐药性具有重要意义。中药种类繁多, 结构多样。近年来研究证实, 中医药作为重要的辅助治疗手段, 可以减毒增效, 改善患者的生活质量。特别是联合分子靶向治疗能有效缓解EGFR-TKIs的耐药性[15]。中医认为肺癌分子靶向治疗EGFR-TKIs获得性耐药与气阴两虚、虚热内生有关[16]。中医治疗以扶正固本、清热解毒、活血化瘀为主。Yu等[17]发现, 清热药黄芩可能通过调控NSCLC细胞氧化还原比, 提高ROS水平, 抑制NSCLC细胞干性来增强EGFR-TKIs药物吉非替尼和奥希替尼对NSCLC细胞的增殖抑制和促凋亡作用。葫芦素B是一类四环三萜类化合物, 主要分布于葫芦科中, 并在十字花科、秋海棠科和玄参科中也有发现。该药性寒, 味甘、微苦, 归肺、胃、大肠经, 具有抗癌、抗炎、抗氧化、抗病毒、降血糖、保肝、神经保护等功效。近年来, 有研究证实葫芦素B在抗肿瘤中发挥着重要作用。如Promkan等[18]发现, 葫芦素B能抑制BRCA1缺陷型乳腺癌细胞的增殖、迁移和侵袭。Shukla等[19]观察到葫芦素B对转移性NSCLC具有显著抗迁移和侵袭能力, 以及抑制肿瘤血管生成作用。Yang等[20]发现, 葫芦素B能抑制吉非替尼耐药NSCLC细胞的增殖和侵袭, 诱导其凋亡。然而, 有关葫芦素B能否缓解NSCLC奥希替尼耐药性的研究尚少。本研究发现, 葫芦素B能够降低奥希替尼耐药NSCLC细胞的活力, 同时对耐药细胞的迁移和侵袭能力具有抑制效应。
肿瘤细胞转移是目前癌症治疗中的一个难点, 其中EMT发生是癌细胞转移的重要表型。EMT是指上皮细胞丧失极性, 转变为具有迁移能力的间充质细胞, 细胞运动性增强的过程[21]。EMT是肿瘤侵袭和远处转移的第一步, 赋予肿瘤细胞在不同条件下生存的可塑性, 使其获得类似癌症干细胞的特征和特性, 如耐药性[22]。Chen等[23, 24]发现, 上调EMT标志物分子E-cadherin水平能增强头颈部鳞状细胞癌对EGFR抑制剂厄洛替尼的敏感性。此外有文献[25]报道, EMT可能引起ABC转运蛋白的过度表达, 从而促进肿瘤细胞的耐药。ABC转运蛋白即ATP结合盒式蛋白(ATP-binding cassette transporter, ABC), 对细胞生理至关重要, 肿瘤细胞中ABC转运蛋白表达的上调会导致多药耐药性, 从而限制化疗药物的治疗效果[26]。NF-κB是一个常见的参与MDR与EMT调控的信号分子, Bentires-Alj等[27]发现在肝癌细胞中NF-κB被激活并迁移到细胞核, 对P-gp (发现的第一个ABC转运蛋白) 的表达起调节作用。在文献[28]报道的肿瘤细胞中, EMT与ABC转运蛋白之间呈正相关关系。本研究结果显示, 葫芦素B与奥希替尼联用后能增加E-cadherin蛋白表达, 降低vimentin表达水平, 提示其可能参与抑制EMT发生。靶点筛选结果显示, 葫芦素B与AXL的结合亲和力较强, 尚未发现其与ABC家族成员有显著的相互作用。AXL是调控EMT表型的关键基因, 其对多种癌细胞的生存、凋亡抵抗及转移发挥促进作用[29]。在NSCLC中, AXL表达升高与EMT密切相关[30], 其介导的下游信号通路激活在EMT相关NSCLC发展中发挥重要作用[31, 32]。在本研究中, 葫芦素B通过结合AXL抑制ERK与AKT信号通路的激活参与奥希替尼耐药NSCLC细胞的迁移侵袭过程。目前尚未有文献报道ERK和AKT与ABC转运蛋白有联系, 本课题组将会在后续的实验研究中进行探索。
综上所述, 葫芦素B作为一种中药化合物, 能够缓解奥希替尼耐药NSCLC细胞的药物治疗抵抗, 其可能通过靶向结合AXL, 抑制AKT和ERK信号通路, 进而降低奥希替尼耐药NSCLC细胞迁移侵袭能力。本研究为治疗NSCLC奥希替尼耐药抑制剂的开发提供了实验支持, 也为临床中药与EGFR-TKIs的联合治疗提供了理论基础。
