It aimed to research the chemical constituents of Solidago canadensis Lour. A new sucrose derivative was isolated from the the methanol extract of Solidago canadensis Lour. with the technologies of silica gel, microporous resin, Rp-18, and Sephadex LH-20 column chromatography. It was identified as 2, 4, 6, 1′, 3′, 6′-O-hexaisovaleryl sucrose by 1D NMR, 2D NMR, high-resolution mass spectrometry, and acid hydrolysis method. Its mass spectrometry fragmentation behaviors were analyzed using high-resolution mass spectrometry and found that compound 1 was easy to break the glycosidic bond to produce the fragment at m/z 415. The ion at m/z 415 continued to eliminate the isovaleryl group as neutral or non-neutral form to generate a series of daughter ions. Compound 1 could inhibit NO production on LPS-induced neuroinflammatory responses in BV2 microglial cells at the concentrations of 5, 10, and 20 μmol·L-1 to exhibit neuroprotective effect. Its half maximal inhibition rate of NO was measured to be 13.96 ± 0.78 μmol·L-1.

本文对加拿大一枝黄花的化学成分进行研究, 运用硅胶、微孔树脂、Rp-18、Sephadex LH-20柱色谱等技术, 从加拿大一枝黄花的甲醇提取物中分离得到一个新的蔗糖衍生物。利用一维、二维核磁共振波谱、高分辨质谱和酸水解等方法将其鉴定为2, 4, 6, 1′, 3′, 6′-O-六异戊酰基蔗糖(2, 4, 6, 1′, 3′, 6′-O-hexaisovaleryl sucrose)。并运用高分辨质谱对其质谱裂解行为进行解析, 发现化合物1容易断裂糖苷键, 产生分子量为m/z 415的碎片离子。碎片离子m/z 415继续以中性或者非中性的形式丢失异戊酰基碎片, 产生一系列子离子。化合物1在5、10和20 μmol·L-1浓度下能够抑制脂多糖(lipopolysaccharide, LPS) 诱导的小胶质细胞(BV2) 炎症损伤模型的NO生成, 表现出神经保护作用, 测得其NO半数抑制率(IC50) 为13.96 ± 0.78 μmol·L-1

本文对加拿大一枝黄花的化学成分进行研究, 运用硅胶、微孔树脂、Rp-18、Sephadex LH-20柱色谱等技术, 从加拿大一枝黄花的甲醇提取物中分离得到一个新的蔗糖衍生物。利用一维、二维核磁共振波谱、高分辨质谱和酸水解等方法将其鉴定为2, 4, 6, 1′, 3′, 6′-O-六异戊酰基蔗糖(2, 4, 6, 1′, 3′, 6′-O-hexaisovaleryl sucrose)。并运用高分辨质谱对其质谱裂解行为进行解析, 发现化合物1容易断裂糖苷键, 产生分子量为m/z 415的碎片离子。碎片离子m/z 415继续以中性或者非中性的形式丢失异戊酰基碎片, 产生一系列子离子。化合物1在5、10和20 μmol·L-1浓度下能够抑制脂多糖(lipopolysaccharide, LPS) 诱导的小胶质细胞(BV2) 炎症损伤模型的NO生成, 表现出神经保护作用, 测得其NO半数抑制率(IC50) 为13.96 ± 0.78 μmol·L-1

It aimed to research the chemical constituents of Solidago canadensis Lour. A new sucrose derivative was isolated from the the methanol extract of Solidago canadensis Lour. with the technologies of silica gel, microporous resin, Rp-18, and Sephadex LH-20 column chromatography. It was identified as 2, 4, 6, 1′, 3′, 6′-O-hexaisovaleryl sucrose by 1D NMR, 2D NMR, high-resolution mass spectrometry, and acid hydrolysis method. Its mass spectrometry fragmentation behaviors were analyzed using high-resolution mass spectrometry and found that compound 1 was easy to break the glycosidic bond to produce the fragment at m/z 415. The ion at m/z 415 continued to eliminate the isovaleryl group as neutral or non-neutral form to generate a series of daughter ions. Compound 1 could inhibit NO production on LPS-induced neuroinflammatory responses in BV2 microglial cells at the concentrations of 5, 10, and 20 μmol·L-1 to exhibit neuroprotective effect. Its half maximal inhibition rate of NO was measured to be 13.96 ± 0.78 μmol·L-1.

加拿大一枝黄花(Solidago canadensis Lour.) 为菊科一枝黄花属的多年生草本植物[1]。原产北美洲, 于20世纪30年代作为观赏花卉引入中国, 后来逸生野外, 成为一种具有较强入侵性的恶性杂草, 破坏本地生态平衡并对本地生物多样性造成威胁[2, 3], 2023年初被列入中国《重点管理外来入侵物种名录》。研究表明, 加拿大一枝黄花含有二萜、三萜、黄酮、皂苷、苯丙素、多炔、酚酸和挥发油等多种次生代谢产物[4-6]。加拿大一枝黄花在欧洲被用于治疗尿结石、慢性肾炎、膀胱炎和糖尿病等疾病[7, 8]。研究发现加拿大一枝黄花具有抗菌[9]、抗炎[10]、抗突变[9, 11]、抗肿瘤[12]、抗氧化[13]、抗抑郁[14]、肝肾脏保护[15]、抗寄生虫[16-19]和化感作用[20]等多种生物活性。加拿大一枝黄花因其较强的繁殖能力和生存适应能力, 入侵后对当地的自然生态系统平衡和生物多样性造成了较大危害和经济损失[21]。因此, 国内外学者对加拿大一枝黄花侧重于在植物保护及环境科学方面开展研究, 对加拿大一枝黄花的化学成分及生物活性研究较少, 不利于加拿大一枝黄花药用价值的挖掘, 阻碍了其开发利用。
本课题组长期致力于挖掘天然产物中具有神经保护作用的活性分子, 并对迷迭香[22]、高良姜[23]、益智[24]、樟叶越桔[25]等药用植物的化学成分及神经保护作用进行了系统性研究, 从中发现了多个具有神经保护作用的天然活性小分子, 为开发对神经退行性疾病具有潜在治疗作用的药物奠定了基础。为进一步促进加拿大一枝黄花的开发利用, 本研究对加拿大一枝黄花的化学成分和生物活性开展研究, 从中发现了一个新的六取代异戊酰基蔗糖衍生物(图 1A), 对其质谱裂解行为进行解析, 利用LPS诱导的BV2炎症损伤模型评价其神经保护作用, 并测定IC50。本文报道了该化合物的分离纯化、结构鉴定过程、质谱裂解行为以及神经保护作用。
化合物1为无色油状物, 高分辨电喷雾质谱(HR-ESI-MS) 测得m/z 864.496 6 [M+NH4]+ (计算值为864.495 1), 预测其分子式为C42H70O17, 不饱和度为8, 化合物11H NMR (CDCl3, 600 MHz) 在高场区显示了12个重叠的甲基上的氢信号δH 0.95 (36H, overlapped), 6个次甲基氢信号δH 2.09 (6H, overlapped), 以及6个亚甲基上的氢信号δH 2.24 (12H, overlapped)。分析其 13C NMR (CDCl3, 150 MHz) 图谱, 结合DEPT谱低场区显示6个数值相近的羰基碳信号, 依次为δC 173.4×2, 172.9×2, 172.8, 172.2; 高场区显示6个化学位移相近的次甲基碳信号, 依次为δC 43.4, 43.3, 43.2×2, 43.1, 43.0; 还有6个化学位移相近的亚甲基碳信号, 依次为δC 25.9×2, 25.8, 25.7×2, 25.6; 同时还有多个化学位移相近, 信号重叠的甲基碳信号, 结合1H NMR信号, 应当为12个化学环境相似的甲基。由此推断化合物1的结构中含有6个结构相同, 化学环境类似的取代基, 综合氢谱和碳谱数据, 推测该基团为异戊酰基。
此外, 13C NMR及DEPT谱上还有12个其他碳原子信号, 分别是位于δC 63.9 (C-6), 62.3 (C-1′) 和63.8 (C-6′) 的3个仲碳信号, 位于δC 89.2 (C-1), 72.3 (C-2), 70.4 (C-3), 70.9 (C-4), 68.6 (C-5), 78.3 (C-3′), 73.3 (C-4′), 80.6 (C-5′) 的8个叔碳信号, 以及位于δC 102.6 (C-2′) 的1个季碳信号。同时分析1H NMR谱中低场区从δH 3.99到δH 5.57的系列连氧次甲基信号, 结合文献[26, 27], 除去6个异戊酰基后剩余的部分结构信号为蔗糖的特征信号(表 1)。说明该化合物为6个异戊酰基取代的蔗糖衍生物(图 1A)。在1H-1H COSY谱中, H-1/H-2/H-3/H-4/H-5/H-6相关, H-3′/H-4′/H-5′/H-6′相关; 在HMBC谱中, H-1和C-5/C-2′相关, H-2和C-4相关, H-3和C-6相关, H-4和C-5相关, H-5和C-3相关, H-6和C-4相关, H-1′和C-3′相关, H-3′和C-4′相关, H-4′和C-3′/C-5′相关, H-5′和C-3′相关, H-6′和C-4′相关, 由此确定了蔗糖上的碳原子位置信息。同时, HMBC谱中H-2/H-4/H-6/H-1′/H-3′/H-6′与羰基碳原子信号簇δC 173.4×2, 173.0, 172.9×2, 172.8, 172.3相关, 可知6个异戊酰基分别与蔗糖的C-2/C-4/C-6/C-1′/C-3′/C-6′位置连接(图 1B)。由于6个羰基碳原子信号值差异较小, 无法具体区分, 但是不影响结构解析。
运用酸水解试验, 通过带有蒸发光散射检测器的高效液相色谱仪进行分析, 与糖标准品进行对照, 确定了糖单元的组成为葡萄糖和果糖[28, 29]。综上所述, 将该化合物鉴定为2, 4, 6, 1′, 3′, 6′-O-六异戊酰基蔗糖(2, 4, 6, 1′, 3′, 6′-O-hexaisovaleryl sucrose)。
对化合物1进行MS/MS分析, 解析其质谱裂解规律(表 2)。化合物的[M+NH4]+峰为m/z 864.496 7, 表明其分子式为C42H70O17 (图 2A)。以m/z 864.496 7为母离子进行MS/MS分析, 发现其容易产生碎片离子m/z 415.228 4。该碎片为蔗糖断裂糖苷键所得, 即C-1与C-2′之间断裂(图 2B)。该糖苷键断裂以后生成的两部分碎片离子的分子式和分子量一致, 均为m/z 415 (C21H35O8), 是异戊酰基葡萄糖或异戊酰基果糖碎片。该碎片离子的异戊酰基以中性丢失方式裂解(-C5H10O2), 或进一步丢失一分子水, 产生m/z 313.159 1, 211.092 8, 295.149 4和109.026 0等碎片; 或者异戊酰基以非中性丢失的方式裂解(-C5H8O), 产生m/z 127.036 2等碎片。m/z 85.062 7和57.068 1为异戊酰基自身裂解所得。以上MS/MS裂解行为进一步证明了化合物1的结构是正确的。
建立LPS诱导的BV2炎症损伤模型评价化合物1的神经保护作用。首先通过Cell Counting Kit-8 (CCK-8) 法测试化合物1对BV2细胞的细胞毒性。在给予化合物1后, 在低、中、高三个浓度下BV2的细胞存活率均达到80%以上, 说明化合物1对BV2细胞没有毒性(图 3A)。神经保护作用测试表明: LPS组和对照组对比, NO显著升高(图 3B), 说明LPS诱导BV2细胞炎症模型造模成功。与LPS组相比, 阳性药米诺环素(MINO) 和化合物1作用于模型细胞后, NO含量明显降低(图 3B), 且呈剂量依赖关系, 表明化合物1对LPS诱导BV2细胞具有保护作用, 呈现抗神经炎症功能。进一步测试化合物1在多个浓度下对LPS诱导BV2细胞神经炎症保护作用, 测得其抑制NO释放的半数抑制率为13.96 ± 0.78 μmol·L-1 (图 3C), 阳性对照为米诺环素, 其IC50为16.26 ± 4.97 μmol·L-1
加拿大一枝黄花次生代谢产物种类丰富, 生物活性多样, 在欧洲药用历史悠久, 被收录于欧洲药典[30]。加拿大一枝黄花在我国的民间药用情况暂无报道, 但其同属植物一枝黄花(Solidago decurrens Lour.) 被收录于2020版中国药典中, 具有清热解毒、疏风散热的功效, 用于治疗喉痹、乳蛾、咽喉肿痛、疮疖肿毒和风热感冒[31]。这提示加拿大一枝黄花的基础研究依然不够深入, 致使其开发利用受到限制。随着人口老龄化加剧, 神经退行性疾病的患病率逐年增加[32]。神经退行性疾病是一类由神经元的退行性损害导致神经系统功能逐渐丧失的疾病, 包括帕金森病、阿尔茨海默病、亨廷顿舞蹈病和肌萎缩性脊髓侧索硬化症等, 这类疾病损害患者的认知功能或运动协调能力, 严重危害人类的健康和生活质量[33-35]。氧化应激和神经炎症是导致神经退行性疾病的主要机制, 另外还与患者年龄、兴奋性毒素、线粒体功能障碍等因素相关[36]。目前临床上治疗神经退行性疾病的药物主要是改善症状, 无法逆转或者阻止疾病的进一步发展, 同时其安全性和有效性还有待考证, 开发满足神经退行性疾病治疗需求的新药至关重要[37]。天然活性产物因为其复杂多变的结构和丰富的药理活性成为了新药研发过程中先导化合物的重要来源。从天然活性产物中发现能够改善细胞内部环境, 降低氧化应激和减轻神经炎症的神经保护剂, 有望为治疗神经退行性疾病的新药研究开发提供新的思路和参考。
本研究从加拿大一枝黄花中分离鉴定出一个新的化合物: 2, 4, 6, 1′, 3′, 6′-O-六异戊酰基蔗糖, 同时通过高分辨质谱对化合物1的MS/MS裂解规律进行了解析, 发现在正离子模式下化合物1容易断裂葡萄糖和果糖之间的糖苷键, 产生异戊酰基葡萄糖或者异戊酰基果糖的碎片, 继续以中性或者非中性的形式丢失异戊酰基碎片, 产生一系列子离子, 为后续解析该类型化合物提供参考。该化合物在5、10和20 μmol·L-1浓度下具有较好的神经保护作用且均无细胞毒性, 测得其NO的IC50为13.96 ± 0.78 μmol·L-1。在后续工作中, 将持续对加拿大一枝黄花开展研究, 丰富加拿大一枝黄花的化学成分, 挖掘具有神经保护作用的活性小分子, 推动加拿大一枝黄花和治疗神经退行性疾病药物的研究。
加拿大一枝黄花药材于2021年9月采自云南省昆明市滇池周边区域, 经云南中医药大学中药学院邱斌教授鉴定为菊科植物加拿大一枝黄花(Solidago canadensis Lour.) 的干燥全株, 样品编号(No. 2021-0920), 保存于云南中医药大学中药学院暨云南省南药可持续利用研究重点实验室。
LC-MS-Q-TOF G6530高分辨率质谱、1260 Infinity II高效液相色谱仪(美国安捷伦科技有限公司); Bruker AV 600/800型核磁共振波谱仪[布鲁克(北京) 科技有限公司]; NICOLET iS10型红外光谱仪[赛默飞世尔科技(中国) 有限公司]; Jascomodel 1020旋光仪(日本Horiba公司); ME204E分析天平(上海梅特勒-托利多仪器有限公司); 蒸发光散射检测器ELSD 6100来自昆明德冠科技有限公司; 葡萄糖标准品(S17HB195198) 购自上海源叶生物科技有限公司; 果糖标准品(BBP04081) 购自云南西力生物技术股份有限公司; 氘代试剂购自北京伊诺凯有限公司; 薄层色谱硅胶板和柱色谱硅胶购自上海皓鸿生物医药科技有限公司; BV2细胞株来自普诺赛科技有限公司; 常用有机试剂为国产AR级试剂购自云南利妍科技有限公司; 显色方法为10%硫酸-乙醇溶液加热后显色。
33.0 kg干燥的加拿大一枝黄花全株粉碎后以120 L甲醇室温浸泡3 d, 共3次, 合并提取液并浓缩至无甲醇味, 得到浸膏3.2 kg, 将其分散在10 L水中, 用3倍量乙酸乙酯萃取, 共三次, 合并浓缩后得乙酸乙酯部位1.1 kg。经硅胶柱色谱(6.0 kg, 20 cm × 40 cm, 石油醚-乙酸乙酯30∶1、15∶1、9∶1、8∶2、7∶3) 洗脱获得8个流分Fr.1~Fr.8。
Fr.6 (136.0 g) 经反相MCI柱色谱(1.2 kg, 15 cm × 30 cm, 甲醇-水50∶50、60∶40、70∶30、80∶20、90∶10) 洗脱得到6个流分Fr.6-1~Fr.6-6; 其中Fr.6-4 (70.0 g) 再使用Rp-18柱色谱分离(500 g, 5 cm × 18 cm, 甲醇-水40∶60、50∶50、60∶40、70∶30、80∶20、90∶10), 洗脱得到7个流分Fr.6-4-1~Fr.6-4-7。Fr.6-4-5 (17.0 g) 通过硅胶柱色谱(170.0 g, 5 cm × 25 cm, 石油醚-乙酸乙酯30∶1、15∶1、10∶1、9∶1、8∶2、7∶3) 进一步分离纯化得到7个流分Fr.6-4-5-1~Fr.6-4-5-7。Fr.6-4-5-1 (0.5 g) 经Sephadex LH-20凝胶柱色谱分离, 甲醇洗脱得到化合物1 (24.5 mg)。
化合物1, 无色油状物, [α]$ {}_{\mathrm{D}}^{20} $ = + 11.33 (c 0.150, CH3OH); HR-ESI-MS m/z 864.496 7 [M+NH4]+ (计算值: 864.495 1), 分子式: C42H70O17。IR (KBr) νmax 3 448, 2 961, 1 744, 1 386, 1 370, 1 295, 1 255, 1 188, 1 166, 1 099, 1 019 cm-1, 核磁数据见(表 1)。
酸水解试验: 取5 mg化合物1溶于5 mL 5% KOH溶液中, 在95 ℃条件下加热回流3 h, 待冷却后将反应产物调至pH 4.0。用等量氯仿及正丁醇分别萃取三次, 减压干燥后, 得到氯仿层和正丁醇层。将正丁醇层用少量甲醇溶解后加入5 mL 2 mol·L-1盐酸, 在95 ℃的条件下加热回流4 h, 待反应产物冷却后用正丁醇萃取三次。将水层减压干燥后, 用水溶解制成待测样品, 葡萄糖和果糖标准品用超纯水溶解, 配成待测样品。用带有蒸发光检测器的高效液相色谱仪进行分析: 采用BDS HYPERSIL-C18柱(4.6 mm × 250 mm, 5 µm), 洗脱系统为: 流动相A (乙腈), 流动相B (磷酸缓冲液pH 6.8), 流速为0.8 mL·min-1, 柱温为30 ℃, 梯度洗脱条件: 起始梯度17% A, 0~22 min; 17%~19% A, 22~24 min; 19% A, 24~55 min。蒸发光检测器条件: 进样量10 μL, 增益因子1, 漂移管温度110 ℃, 气体体积流量2.7 L·min-1
色谱条件: MS/MS分析在安捷伦LC-MS-Q-TOF G6530质谱仪上进行, 采用Eclipse XDB-C18柱(2.1 mm × 50 mm, 3.5 µm)。化合物1用甲醇配制成浓度为0.05 mg·mL-1备用。洗脱系统为: 流动相A: 甲酸-水(0.05∶100, v/v), 流动相B: 乙腈, 流速为0.3 mL·min-1, 柱温为30 ℃, 梯度洗脱条件: 起始梯度5%~90% B, 0~8 min; 90% B, 8~10 min; 90%~2% B, 10~11 min; 2% B, 11~14 min。
质谱条件: 离子源为ESI源, 三氟乙酸钠校正, 电喷雾负离子模式检测。分析条件如下: 喷雾电压为4.00 kV或-3.50 kV; 干燥气压力为35.0 kPa; 喷雾气为氮气, 流速为8.0 L·min-1; 曲形裂解器温度为350 ℃; 加热模块温度为280 ℃; 母离子选择范围为m/z ± 0.2 Da; 裂解能量为10、20、30和40 eV; 裂解气为氩气, 扫描范围为m/z 50~1 500, 自动模式进行MS/MS分析。Agilent Qualitative Analysis 10.0软件用于预测分子式, 其他质谱条件参照文献[25]进行设置。
使用CCK-8法测试化合物1对BV2细胞的细胞毒性, 并在LPS诱导的BV2炎症损伤模型上进行神经炎症活性测试, 米诺环素为阳性对照药。小鼠小胶质细胞接种于含有10%胎牛血清(fetal bovine serum, FBS), 100 u·mL-1青霉素和100 u·mL-1链霉素的Dulbecco's modified Eagle medium (DMEM) 高糖培养基中, 放置于5% CO2的细胞培养箱37 ℃条件下培养。接种后, 让细胞生长24 h, 直到达到80%~90%的最终密度, 用于后续研究。
将细胞分为对照组、给药组。将BV2细胞接种于96孔细胞培养板上, 密度为每毫升5×104个。①对照组分为溶剂对照组和空白对照组; ②给药组用BV2培养24 h后, 弃去旧培养基, 药物以含有不同浓度(5、10和20 μmol·L-1) 药物处理24 h, 将细胞培养基更换为孔中含有10% CCK-8的DMEM, 孵育2 h后, 使用酶标仪在450 nm处读取每个孔的吸光度值(A), 计算各组细胞存活率, 实验重复3次。用于评价化合物对神经细胞有无毒性。细胞存活率(%) = (A1-A3) / (A2-A3) × 100%, 其中A1为药物组的吸光度; A2为溶剂对照组的吸光度; A3为空白对照组的吸光度。
采用NO试剂盒检测化合物1对LPS诱导BV2细胞炎症活性。将细胞分为对照组、LPS组、MINO组和给药组。将BV2细胞接种于96孔细胞培养板上, 密度为每毫升25×104个。① LPS组用BV2培养24 h后, 弃去旧培养基, 用1 μg·mL-1 LPS处理24 h; ② MINO组使用10 μmol·L-1米诺环素预处理2 h后, 用1 μg·mL-1 LPS处理24 h; ③给药组以含有不同浓度(5、10和20 μmol·L-1) 化合物1预处理2 h, 用1 μg·mL-1LPS处理24 h。给予对应药物分组处理后收集细胞上清液, 按要求加入NO试剂盒I和II的相应溶液, 用酶标仪在540 nm处读取每个孔的A值, 将其代入标准曲线y = 163.59 (A) - 6.515 9中计算NO含量, 实验重复3次。用于评价化合物对受损的神经细胞有无降低NO浓度抗炎保护作用。
采用GraphPad Prism 9.0软件对数据进行统计分析, 结果采用平均值± SEM表示。三组数据使用单因素方差分析, 然后进行Tukey的事后检验。所有检验均采用双侧P值, 认为P < 0.05有统计学意义。
