A new benzoic acid derivative, spinulacid (1), together with six known compounds ascomindone A (2), ascomindone C (3), monomethylsulochrin (4), barceloneic acid A (5), flufuran (6), and 5-hydroxymethyl-furaldehyde (7) were obtained from the rice fermentation extract of an endophytic fungus Penicillium spinulosum isolated from Emmenopterys henryi Oliv. by various chromatographic techniques. Their structures were elucidated by the analysis of NMR data, MS and comparison with literature. The biological activity results showed that compounds 1, 2, 5, and 6 exhibited significant proangiogenic activity in transgenic zebrafish at concentrations of 20 and 40 μmol·L-1 (the animal experiment was approved by the Animal Ethical and Welfare Committee of Biology Institute, Shandong Academy of Science, the approval number is SWS20240611). Compounds 1 and 4 displayed moderate or strong antifungal activity against phytopathogen Fusarium graminearum with minimum inhibitory concentration (MIC) values of 12.5, and 6.25 μg·mL-1, respectively. Besides, compound 4 was also found to show obvious antibacterial activity against Staphylococcus aureus and Erwinia carotovora with MIC values of 6.25 and 12.5 μg·mL-1, respectively.

本研究采用多种色谱技术从一株香果树内生真菌Penicillium spinulosum大米发酵产物中获得7个次级代谢产物, 经核磁、质谱等波谱学方法结合文献数据对这些化合物进行了结构鉴定, 分别为spinulacid (1), ascomindone A (2), ascomindone C (3), monomethylsulochrin (4), barceloneic acid A (5), flufuran (6) 和5-hydroxymethyl-furaldehyde (7), 其中化合物1为新苯甲酸类衍生物。药理活性评价显示, 化合物1256在20和40 μmol·L-1浓度下对转基因斑马鱼具有显著的促血管生成活性(动物实验获得山东省科学院生物研究所实验动物福利伦理委员会批准, 批准号为SWS20240611); 化合物14对禾谷镰刀菌表现出中等或强的抗真菌活性, 其最小抑菌浓度(MIC) 分别为12.5和6.25 μg·mL-1; 此外, 化合物4对金黄色葡萄球菌和欧文杆菌也显示出明显的抗菌活性, 其MIC值分别为6.25和12.5 μg·mL-1

A new benzoic acid derivative, spinulacid (1), together with six known compounds ascomindone A (2), ascomindone C (3), monomethylsulochrin (4), barceloneic acid A (5), flufuran (6), and 5-hydroxymethyl-furaldehyde (7) were obtained from the rice fermentation extract of an endophytic fungus Penicillium spinulosum isolated from Emmenopterys henryi Oliv. by various chromatographic techniques. Their structures were elucidated by the analysis of NMR data, MS and comparison with literature. The biological activity results showed that compounds 1, 2, 5, and 6 exhibited significant proangiogenic activity in transgenic zebrafish at concentrations of 20 and 40 μmol·L-1 (the animal experiment was approved by the Animal Ethical and Welfare Committee of Biology Institute, Shandong Academy of Science, the approval number is SWS20240611). Compounds 1 and 4 displayed moderate or strong antifungal activity against phytopathogen Fusarium graminearum with minimum inhibitory concentration (MIC) values of 12.5, and 6.25 μg·mL-1, respectively. Besides, compound 4 was also found to show obvious antibacterial activity against Staphylococcus aureus and Erwinia carotovora with MIC values of 6.25 and 12.5 μg·mL-1, respectively.

青霉菌(Penicillium) 由300多种真菌组成, 是最大的真菌群之一, 特别是作为抗生素的重要来源[1]。在过去的20年里, 对内生青霉菌的研究已经远远超越了它们的抗生素潜力, 目前已发现的内生青霉菌超过130多种, 其中280多个活性次级代谢产物被陆续报道出来, 结构类型涉及聚酮类、萜类、生物碱类、甾体类等[2, 3], 这些化合物表现出抗微生物、抗病毒、抗肿瘤、抗氧化、免疫抑制等多种生物活性[4], 是新药开发的重要宝库。
Penicillium spinulosum是从濒危植物香果树(Emmenopterys henryi Oliv.) 的根部分离获得的一株内生真菌, 对其次级代谢产物进行了化学成分和药理活性研究。P. spinulosum的大米发酵产物经乙酸乙酯提取获得粗浸膏, 经硅胶柱层析、Sephadex LH-20凝胶柱层析及半制备高效液相色谱等现代分离技术, 共分离得到7个化合物, 包括1个新的苯甲酸类衍生物, 命名为spinulacid (1), 以及6个已知化合物2~7 (图 1)。对分离获得的化合物进行了体内促斑马鱼血管生成活性、抗农业致病真菌活性及抗细菌活性评价。本文对上述化合物的分离、结构鉴定及活性评价进行详细报道。
化合物1为白色无定形固体。根据高分辨质谱HR-ESI-MS m/z 259.094 4 [M+Na]+, 结合1D核磁, 推断其分子式为C13H16O4, 不饱和度为6。化合物11H NMR谱中, 显示三个芳香质子信号δH 7.89 (dd, J = 8.6, 1.9 Hz), 7.80 (d, J = 1.9 Hz) 和6.99 (d, J = 8.6 Hz), 一个烯氢信号δH 5.57 (t, J = 7.4 Hz), 两个亚甲基信号δH 3.97 (s), 3.38 (d, J = 7.4 Hz), 一个甲氧基信号δH 3.97 (s) 和一个甲基信号δH 1.76 (s)。化合物113C NMR和HSQC谱揭示了13个碳信号, 包括5个sp2季碳, 4个sp2次甲基, 2个亚甲基, 1个甲氧基和1个甲基信号。在1H-1H COSY谱中, 显示H-5/H-6及H-8/H-9的相关性。仔细对比化合物1和化合物rhizopycnis acid A[5]的核磁数据发现其结构非常相似, 其差异性仅在于11位羧基被还原成醇。在HMBC谱中, H-12和C-9, C-10和C-11相关, 以及H-9和C-8, C-11和C-12相关也证实了上述推测(表 1)。因此, 化合物1的结构得到确定, 为一个新的苯甲酸类化合物, 命名为spinulacid。
利用转基因斑马鱼Tg(vegfr2:GFP) 模型[6]对分离获得的化合物125~7进行了体内促血管生成活性评价(图 23)。与PTK787抑制斑马鱼节间血管的模型组相比, 发现化合物1256在浓度为20和40 μmol·L-1时表现出显著的促血管生成活性。其中, 化合物1在10、20和40 μmol·L-1浓度下以浓度依赖的方式发挥促血管生成作用, 且化合物1在40 μmol·L-1浓度下表现出与阳性药阿魏酸(ferulic acid) 相当的显著活性。化合物7未表现出活性, 对比化合物6的结构, 表明羧基对该活性至关重要; 此外, 分析化合物1256的结构, 其结构共同点均含有羧基片段, 也表明羧基是重要的药效团。
对分离获得的化合物1~7还进行了抗农业致病真菌活性及抗细菌活性评价, 农业致病真菌包括链格孢菌Alternaria alternata、禾谷镰刀菌Fusarium graminearum和尖孢镰刀菌Fusarium oxysporum。细菌菌株包括2株革兰阳性菌株金黄色葡萄球菌Staphylococcus aureus和欧文杆菌Erwinia carotovora, 和1株革兰阴性菌株肺炎克雷伯菌Klebsiella pneumonia, 其结果如下表 23所示。抗农业致病真菌结果显示, 化合物14对禾谷镰刀菌表现出中等或强的抗真菌活性, MIC值分别为12.5和6.25 μg·mL-1; 化合物1对尖孢镰刀菌表现出弱的抗真菌活性, MIC值为50 μg·mL-1。抗细菌活性显示, 化合物2~4对金黄色葡萄球菌和欧文杆菌表现出一定的抗菌活性, MIC值为6.25~100 μg·mL-1; 其中化合物4的抗菌活性最为显著, MIC值分别为6.25和12.5 μg·mL-1
Bruker UltrashiedTM 400 MHz Plus核磁共振谱仪(德国布鲁克公司); Dionex Ultimate 3000型高效液相色谱仪(美国赛默飞戴安公司); Waters 1525型半制备型高效液相色谱仪、Micromass Q-TOF高分辨质谱仪(美国Waters公司); 清博华PUC0050高效液相色谱仪(北京清博华科技有限公司); SHIMADZU UV-1900i紫外光谱仪(日本岛津公司); 薄层色谱硅胶和正相柱色谱硅胶(青岛海洋化工有限公司); 反相柱色谱硅胶(美国Unicorn公司); Sephadex LH-20凝胶(美国Amersham Biosciences公司); 提取分离所用分析纯试剂石油醚、氯仿、乙酸乙酯和甲醇(天津科密欧试剂公司); 高效液相所用色谱乙腈(美国Tedia“天地”试剂公司); 环丙沙星(货号E021013)、制霉菌素(货号E121990) 购买于上海安耐吉化学有限公司。
动物实验获得山东省科学院生物研究所实验动物福利伦理委员会批准, 批准号为SWS20240611。转基因TG (VEGFR2: GFP) 系斑马鱼和动物实验由山东省科学院生物研究所药物筛选研究室提供并完成。AXIO-V16荧光显微镜(德国ZEISS公司); 链霉蛋白酶E (北京索莱宝科技有限公司, 批号P8360); Vatalanib base (PTK787, 上海源叶生物技术有限公司, 批号S80538); 阿魏酸(上海麦克林生化科技有限公司, 批号F809521)。
真菌P. spinulosum 9-1分离自濒危植物香果树(E. henryi Oliv.) 的根部, 该植物于2022年5月采自中国湖北省神农架地区, 经形态学及分子生物学鉴定, 确定该菌株为Penicillium spinulosum (Genebank登录号: PQ577833)。
真菌P. spinulosum 9-1采用大米固体发酵, 在室温条件下发酵培养40天, 发酵45瓶。待发酵结束后, 采用乙酸乙酯进行浸泡, 反复浸提3次, 将有机相进行合并, 经减压浓缩获得粗浸膏24.3 g。粗浸膏经减压硅胶柱层析分离获得6个不同极性的组分Fr.1~Fr.6, 分别为90%石油醚-乙酸乙酯组分(3.3 g)、70%石油醚-乙酸乙酯组分(5.6 g)、50%石油醚-乙酸乙酯组分(3.5 g)、30%石油醚-乙酸乙酯组分(4.7 g)、乙酸乙酯组分和20%甲醇-乙酸乙酯组分(4.8 g)。对组分Fr.3进行正相柱分离(石油醚-乙酸乙酯, 2∶3) 和Sephadex LH-20凝胶柱分离(甲醇-二氯甲烷, 1∶1), 获得化合物4 (3.8 mg)。进一步对剩余Fr.3组分经半制备HPLC (40%乙腈水溶液) 分离获得化合物3 (4.4 mg, tR = 16 min) 和化合物5 (10.1 mg, tR = 9 min)。对组分Fr.4经过反复的正相硅胶柱分离(石油醚-乙酸乙酯, 1∶1~1∶2)、Sephadex LH-20凝胶柱分离(甲醇-二氯甲烷, 1∶1), 最后通过半制备HPLC (35%乙腈水溶液) 分离获得化合物1 (7.5 mg, tR = 12 min), 化合物2 (5.8 mg, tR = 10 min) 和化合物7 (30.0 mg, tR = 18 min)。组分Fr.6经清博华HPLC分离(乙腈-水, 10%~100%, 梯度洗脱30 min) 分离获得化合物6 (10.3 mg, tR = 6 min)。
化合物1 白色无定形固体, UV (MeOH) λmax 218 (4.00), 258 (3.82) nm; HR-ESI-MS m/z 259.094 4 [M+Na]+ (calcd for C13H16O4Na, 259.094 6); 1H NMR和 13C NMR数据见表 1所示。
化合物2 棕色晶体, 分子式为C32H26O11, 1H NMR和13C NMR数据与文献[7]报道的一致, 该化合物被鉴定为ascomindone A。
化合物3 黄色无定形固体, 分子式为C32H24O10, 1H NMR和13C NMR数据与文献[7]报道的一致, 该化合物被鉴定为ascomindone C。
化合物4 橙色无定形固体, 分子式为C18H18O7, 1H NMR和13C NMR数据与文献[8]报道的一致, 该化合物被鉴定为monomethylsulochrin。
化合物5 白色固体, 分子式为C16H16O7, 1H NMR和13C NMR数据与文献[9]报道的一致, 该化合物被鉴定为barceloneic acid A。
化合物6 黄色晶体, 分子式为C6H6O4, 1H NMR和13C NMR数据与文献[10]报道的一致, 该化合物被鉴定为flufuran。
化合物7 黄色油状, 分子式为C6H6O3, 1H NMR和13C NMR数据与文献[11]报道的一致, 该化合物被鉴定为5-hydroxymethyl-furaldehyde。
斑马鱼胚胎培养[12]: 斑马鱼在照明14 h/黑暗10 h、水温28 ℃、pH 7.2~7.5标准条件下饲养, 定时喂以人工颗粒状饵料和刚孵出的卤虫无节幼体。采卵时, 取健康性成熟的斑马鱼, 按雌雄1/1或1/2的比例放入交配缸内, 次日9~10时获得受精卵。对受精卵进行消毒和清洗后, 移入斑马鱼胚胎培养水中于28 ℃进行控光培养。
斑马鱼体节间血管数[13]的测定: 在受精卵发育24 h后, 使用1.0 mg·mL-1链酶蛋白酶E溶液脱去卵膜。挑选正常的斑马鱼胚胎, 移入24孔培养板中, 每孔10枚。设正常对照组、PTK787模型组、阳性对照组和给药治疗组(选用40 μmol·L-1浓度阿魏酸作为阳性对照, 样品浓度设置为10、20和40 μmol·L-1), 置于28 ℃光照培养箱培养。在药物作用48 h后, 用荧光显微镜观察荧光血管生长情况并记录有血流的斑马鱼节间血管(ISVs) 数。
使用GraphPad Prism 6.01软件进行数据统计与分析, 采用单因素方差分析进行差异显著性分析, P < 0.05表示差异显著。
采用微量稀释法[14, 15]对化合物1~7的抗微生物活性进行测试。微生物菌株来自中国农业科学院烟草研究所。将待测化合物溶解在DMSO中, 随后将其加入到培养基中, 包括LB细菌培养基和PDB真菌培养基, 筛选的化合物终浓度为100、50、25、12.5、6.25和3.125 μg·mL-1, DMSO的终浓度为1%。然后在各自的培养基中接种细菌和真菌培养物, 每种培养物的密度为每毫升5×105个细胞(或孢子)。细菌培养物在37 ℃下恒温培养24 h, 真菌培养物在28 ℃下恒温培养48~72 h。通过评估培养基的浊度来监测微生物的生长情况。采用环丙沙星和制霉菌素分别作为细菌和植物病原真菌的阳性对照。
