Six compounds including three new compounds were obtained from the fruiting bodies of Ganoderma lucidum in Baoshan area. These structures were identified as baosacid A (1), baosside A (2), ethyl 2,5-dihydroxy-γ-oxobenzenebutanoate (3), australins A (4), 2,5-dihydroxy-γ-oxobenzenebutanoic acid (5), and methyl 2,5-dihydroxy-γ-oxobenzenebut anoate (6) on the basis of spectroscopic methods. In addition, compound 5 is a newly occurring natural product.

利用各种色谱技术, 从保山产赤芝中得到6个化合物。结合波谱方法鉴定了它们的结构, 分别为baosacid A (1)、baosside A (2)、ethyl-2,5-dihydroxy-γ-oxobenzenebutanoate (3)、australins A (4)、2,5-dihydroxy-γ-oxobenzenebutanoic acid (5) 和methyl 2,5-dihydroxy-γ-oxobenzenebutanoate (6)。其中, 化合物1~3为新的酚类化合物, 化合物5为新天然产物。

Six compounds including three new compounds were obtained from the fruiting bodies of Ganoderma lucidum in Baoshan area. These structures were identified as baosacid A (1), baosside A (2), ethyl 2,5-dihydroxy-γ-oxobenzenebutanoate (3), australins A (4), 2,5-dihydroxy-γ-oxobenzenebutanoic acid (5), and methyl 2,5-dihydroxy-γ-oxobenzenebut anoate (6) on the basis of spectroscopic methods. In addition, compound 5 is a newly occurring natural product.

灵芝(Ganoderma lucidum), 担子菌纲、灵芝科、灵芝属的药食两用真菌, 载于《神农本草经》, 性平, 味甘, 具有补中益气、滋外强壮、扶正固本、延年益寿等功效[1], 是我国传统的中药材[2]。2020版《中国药典》记载, 灵芝为多孔菌科真菌赤芝Ganoderma lucidum (Leyss. ex Fr.) Karst.或紫芝Ganoderma sinense Zhao, Xu et Zhang的干燥子实体[3]。其主要化学成分为多糖类、三萜、生物碱、甾体、核苷、蛋白质和微量元素等[4-7], 具有抗炎[5]、抗肿瘤[8, 9]、抗氧化[10, 11]、抗肾纤维化[12, 13]等多种药理活性。近年来, 灵芝因其显著功效而备受关注, 其化学成分的研究也一直被重点关注。尽管如此, 安神、延缓衰老等作用的物质基础远未能解决。因此, 进一步深入挖掘灵芝的化学成分还具有重大意义。本实验在对其的深入研究中分离鉴定了6个酚类化合物(图 1), 其中化合物1~3为新的酚类化合物, 化合物4~6为已知化合物, 化合物5为新天然产物。
化合物1为黄色固体。根据13C NMR谱数据和HR-ESI-MS数据(m/z 295.058 8 [M+Na]+, calcd. for 295.057 7), 确定该化合物的分子式为C15H12O5, 不饱和度为10。1H NMR (600 MHz, CD3OD) 谱(表 1) 显示芳香区有6个质子信号[δH 7.96 (1H, d, J = 8.0 Hz, H-6′), 7.42 (1H, dd, J = 8.0, 1.7 Hz, H-5′), 7.16 (1H, d, J = 1.7 Hz, H-3′), 6.95 (1H, dd, J = 8.9, 3.0 Hz, H-4), 6.82 (1H, d, J = 8.9 Hz, H-3), 6.50 (1H, d, J = 3.0 Hz, H-6)]。质子信号的峰型和耦合常数表明结构中存在两个典型的ABX系统。此外, 在高场区还有1个甲基信号[δH 2.45 (3H, s, H-9′)]。根据甲基质子信号的化学位移推断其为连在苯环或双键上的甲基。由13C NMR (150 MHz, CD3OD) 谱(表 1) 和HMBC谱可知结构中存在15个碳信号, 包括8个非质子碳(芳香区或烯区) [δC 205.4 (C-1′), 170.4 (C-8′), 156.7 (C-2), 150.3 (C-5), 143.8 (C-4′), 142.0 (C-2′), 130.5 (C-7′), 121.7 (C-1)], 6个次甲基(芳香区或烯区) [δC 131.3 (C-6′), 131.2 (C-5′), 128.6 (C-3′), 125.4 (C-4), 119.3 (C-3), 118.1 (C-6)] 和1个甲基[δC 21.4 (C-9′)]。1H-1H COSY谱中(图 2), H-3/H-4的相关证明了环A中ABX的取代方式。此外, 1H-1H COSY谱中(图 2) H-5′/H-6′的相关和HMBC谱(图 2) 中H3-9′/C-3′, C-4′, C-5′, H-6′/C-7′, C-8′ (δC 170.4) 和H-3′/C-2′, C-7′相关证明了另一个ABX系统环B的存在, 且甲基和羧基分别连接于C-4′和C-7′位。最后, HMBC谱(图 2) 中H-6, H-3′/C-1′ (δC 205.4) 的相关表明环A和环B通过C-1′连接。综上所述, 化合物1的结构被确定, 并将其命名为baosacid A。
化合物2为褐色固体。根据13C NMR谱数据和HR-ESI-MS数据(m/z 235.060 2 [M+H]+, calcd. for 235.060 1), 确定该化合物的分子式为C12H10O5, 不饱和度为8。1H NMR (500 MHz, CD3OD) 谱(表 1) 显示芳香区或烯区有4个质子信号[δH 7.78 (1H, s, H-6), 7.29 (1H, d, J = 3.5 Hz, H-3′), 6.35 (1H, s, H-3), 6.34 (1H, d, J = 3.5 Hz, H-4′)], 此外, 在高场区还有1个甲基信号[δH 2.47 (3H, s, H-9′)]。13C NMR (150 MHz, CD3OD) 谱(表 1) 显示出12个碳信号, 结合DEPT谱可知, 结构中存在7个非质子碳(芳香区或烯区) [δC 183.8 (C-1′), 161.5 (C-2), 159.4 (C-5′), 155.9 (C-4), 152.3 (C-2′), 139.6 (C-5), 111.4 (C-1)], 4个次甲基(芳香区或烯区) [δC 122.7 (C-3′), 117.1 (C-6), 110.1 (C-4′), 104.2 (C-3)], 1个甲基碳[δC 13.9 (C-9′)]。HMBC谱(图 2) 中H-3/C-1, C-2 (δC 161.5), C-4 (δC 155.9), C-5 (δC 139.6) 和H-6/C-1, C-2, C-4, C-5的相关, 结合C-2, C-4, C-5的化学位移表明C-2, C-4, C-5位含氧的1,2,4,5-四取代苯环的存在。在1H-1H COSY谱(图 2) 中H-3′/H-4′的相关和HMBC谱(图 2) 中H-3′, H-4′/C-5′, H-3′, H-4′/C-2′和H3-6′/C-4′, C-5′, C-2′ (weak) 表明环B的存在。最后, HMBC谱中H-6, H-3′/C-1′的相关表明两个环通过C-1′进行连接。因此, 化合物2的结构被确定, 并将其命名为baosside A。
化合物3为黄色固体。根据13C NMR谱数据和HR-ESI-MS数据(m/z 261.073 4 [M+Na]+, calcd. for 261.073 3), 确定该化合物的分子式为C12H14O5, 不饱和度为6。1H NMR (600 MHz, CD3OD) 谱(表 1) 显示芳香区有3个质子信号[δH 7.27 (1H, d, J = 2.9 Hz, H-6), 7.01 (1H, dd, J = 8.9, 2.9 Hz, H-4), 6.80 (1H, d, J = 8.9 Hz, H-3)]。此外, 在中高场区存在3个亚甲基[δH 4.14 (2H, q, J = 7.1 Hz, H-5′), 3.33 (2H, t, J = 6.3, Hz, H-2′), 2.71 (2H, t, J = 6.3 Hz, H-3′)], 1个甲基信号[δH 1.25 (3H, t, J = 7.1 Hz, H-6′)]。结合质子信号的峰型和耦合常数确定结构中存在一个典型的ABX系统。13C NMR (150 MHz, CD3OD) 谱和DEPT谱(表 1) 显示12个碳信号, 包括5个非质子碳(烯区或芳香区) [δC 156.4 (C-2), 150.7 (C-5), 120.4 (C-1), 205.5 (C-1′), 174.6 (C-4′)], 3个芳香次甲基[δC 125.8(C-4), 119.7 (C-3), 115.4 (C-6)], 3个亚甲基[δC 61.7(C-5′), 34.3 (C-2′), 28.8 (C-3′)], 1个甲基碳[δC 14.5 (C-6′)]。在1H-1H COSY谱(图 2) 中H-3/H-4的相关证实了ABX系统的存在。此外, 1H-1H COSY谱(图 2) 中H2-2′/H2-3′和H2-5′/H3-6′的相关及HMBC谱(图 2) 中H-6, H2-2′, H2-3′/C-1′和H2-2′, H2-3′, H2-5′/C-4′的相关表明C2′-C3′-C4′-O-C5′-C6′片段与苯环通过C-1′连接。综上所述, 化合物3的结构被确定, 并将其命名为ethyl 2,5-dihydroxy-γ-oxobenzenebutanoate。
SP 5030型半制备高效液相色谱仪(北京赛谱锐思科技有限公司); AB SCIEX triple TOF X500R高分辨质谱仪(AB SCIEX公司); Bruker AV-500 MHz及AV-600 MHz核磁共振波谱仪(以TMS为内标, 德国Bruker公司); MCI gel CHP 20P填料(70~150 μm, 日本三菱公司); YMC gel ODS-A-HG填料(12 nm S-50 μm, 日本YMC公司); 半制备SEP Basic C18色谱柱(120A 5 μm, 10 mm × 250 mm) 和Thermo EVO 300型紫外分光光度计(北京赛谱锐思科技有限公司)。
灵芝药材于2018年4月购自云南省保山市滇滩镇胜利村, 由中国科学院昆明植物研究所杨祝良教授鉴定为赤芝的干燥子实体, 样品标本(CHYX0619) 已存放在广东省深圳市深圳大学药学院。
取500.0 kg保山产赤芝干燥子实体, 粉碎后于室温下用80%乙醇回流提取(3 000 L × 2, 3 h), 提取液减压浓缩, 得浸膏24.6 kg。其加适量水混悬后, 加等体积的乙酸乙酯萃取4次, 减压浓缩, 得浸膏11.5 kg。乙酸乙酯浸膏经MCI gel CHP 20P反相色谱柱(甲醇/水, 40%~100%) 梯度洗脱, TLC监测, 合并相同的部分, 得Fr.A~Fr.Q共17个组分。
Fr.E (83.0 g) 经葡聚糖凝胶柱色谱(Sephadex LH-20) 洗脱, TLC检测合并相同成分, 得Fr.E.1和Fr.E.2两个亚组分。Fr.E.2 (79.9 g) 经MCI gel CHP 20P反相色谱柱(甲醇/水, 30%~100%) 梯度洗脱, 得Fr.E.2.1~Fr.E.2.4四个组分。Fr.E.2.1 (44.4 g) 再次经Sephadex LH-20色谱洗脱得Fr.E.2.1.1~Fr.E.2.1.3三个组分。Fr.E.2.1.1 (23.2 g) Sephadex LH-20凝胶色谱柱(甲醇/水, 70%) 洗脱得Fr.E.2.1.1.1~Fr.E.2.1.1.4四个组分。因Fr.E.2.1.1.2和Fr.E.2.1.1.3差别不大, 遂将其合并为Fr.E.2.1.1.b一个组分。Fr.E.2.1.1.b (35.0 g), 经MCI gel CHP 20P反相色谱柱(甲醇/水, 24%~100%) 梯度洗脱得Fr.E.2.1.1.b.1~Fr.E.2.1.1.b.4四个组分。Fr.E.2.1.1.b.2 (26.6 g) 经Sephadex LH-20色谱柱进行洗脱后得Fr.E.2.1.1.b.2.1~Fr.E.2.1.1.b.2.4。Fr.E.2.1.1.b.2.4 (1.1 g) 经Sephadex LH-20色谱柱(甲醇/水, 70%) 洗脱后得Fr.E.2.1.1.b.2.4.1~ Fr.E.2.1.1.b.2.4.7七个亚组分。Fr.E.2.1.1.b.2.4.1 (150.4 mg) 经半制备高效液相色谱HPLC纯化得化合物5 (乙腈/水30%, 水中含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 16.3 min, 18.9 mg)。Fr.E.2.1.1.b.2.4.2 (179.6 mg) 经半制备HPLC纯化后得化合物6 (甲醇/水, 45%, 含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 23.5 min, 23.9 mg)。Fr.E.2.1.1.b.2.4.3 (79.4 mg) 经半制备HPLC纯化后得化合物3 (甲醇/水, 50%, 水中含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 20.3 min, 3.8 mg)。Fr.E.2.1.1.b.2.4.7 (89.1 mg) 经半制备HPLC纯化后得化合物4 (乙腈/水, 32%, 水中含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 24.7 min, 3.9 mg)。
Fr.G (1.2 kg) 经MCI gel CHP 20P反相色谱柱(甲醇/水, 40%~100%) 梯度洗脱, 得Fr.G.1~Fr.G.7七个组分。Fr.G.2 (280.9 g) 经Sephadex LH-20柱色谱进行洗脱后得Fr.G.2.1~Fr.G.2.3三个组分。Fr.G.2.3 (71.1 g) 经YMC gel ODS-A-HG反相色谱柱(甲醇/水, 30%~100%) 梯度洗脱后得Fr.G.2.3.1~Fr.G.2.3.4四个组分。Fr.G.2.3.2 (13.9 g)经Sephadex LH-20柱色谱洗脱后得到Fr.G.2.3.2.1~Fr.G.2.3.2.6六个组分。Fr.G.2.3.2.2 (6.3 g) 再次经Sephadex LH-20柱色谱(甲醇/水, 70%) 进行洗脱。得Fr.G.2.3.2.2.1和Fr.G.2.3.2.2.2两个组分。Fr.G.2.3.2.2.2 (3.2 g) 经RP-18反相色谱柱(甲醇/水, 30%) 梯度洗脱得Fr.G.2.3.2.2.2.1~Fr.G.2.3.2.2.2.3三个组分。Fr.G.2.3.2.2.2.2 (2.4 g) 经Sephadex LH-20柱色谱进行洗脱得Fr.G.2.3.2.2.2.2.1~Fr.G.2.3.2.2.2.2.4四个组分。Fr.G.2.3.2.2.2.2.3 (81.2 mg) 经半制备HPLC纯化后得化合物2 (乙腈/水, 34%, 水中含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 22.3 min, 2.0 mg)。Fr.G.2.3.3 (55.4 g) 经Sephadex LH-20柱色谱进行洗脱后得Fr.G.2.3.3.1~Fr.G.2.3.3.4四个组分。Fr.G.2.3.3.3 (1.5 g) 经RP-18反相色谱柱(甲醇/水, 30%) 梯度洗脱得Fr.G.2.3.3.3.1~Fr.G.2.3.3.3.8八个组分。Fr.G.2.3.3.3.7 (155.9 mg) 经半制备HPLC纯化后得化合物1 (乙腈/水, 36%, 水中含0.05%甲酸, 流速: 3.0 mL·min-1, tR = 18.8 min, 1.2 mg)。
化合物1    黄色固体, 溶于甲醇。[α]$ {}_{\mathrm{D}}^{20} $ -10.0 (c 0.30, MeOH)。IR (KBr): 3 372、1 754、1 697、1 476、1 229、1 185、792 cm-1。UV (MeOH) λmax (logε): 360 (3.51)、322 (3.39)、226 (4.20)、208 (4.25) nm。1H NMR (600 MHz, CD3OD) 和13C NMR (150 MHz, CD3OD) 数据见表 1, HR-ESI-MS m/z 295.058 8 [M+Na]+ (calcd. for 295.057 7, C15H12O5Na)。
化合物2    褐色固体, 溶于甲醇。IR (KBr): 3 434、1 633、1 503、1 285、1 202、1 144、830、665、513 cm-1。UV (MeOH) λmax (logε): 382 (4.05)、348 (3.89)、316 (4.15)、242 (3.76)、208 (4.20) nm。1H NMR (500 MHz, CD3OD) 和13C NMR (150 MHz, CD3OD) 数据见表 1, HR-ESI-MS m/z 235.060 2 [M+H]+ (calcd. for 235.060 1, C12H11O5)。
化合物3    黄色固体, 溶于甲醇。IR (KBr): 3 362、1 691、1 619、1 480、1 164、769、571 cm-1。UV (MeOH) λmax (logε): 362 (3.61)、288 (3.06)、256 (3.88)、226 (4.20) nm。1H NMR (600 MHz, CD3OD) 和13C NMR (150 MHz, CD3OD) 数据见表 1, HR-ESI-MS m/z 261.073 4 [M+Na]+ (calcd. for 261.073 3, C12H14O5Na)。
化合物4    黄色胶状, 溶于甲醇, ESI-MS m/z 243 [M+H]+, 分子式为C15H14O31H NMR (500 MHz, CD3OD) δH 7.34 (1H, d, J = 2.9 Hz, H-3), 7.19 (1H, t, J = 7.6 Hz, H-7′), 7.10 (1H, brs, H-4′), 7.06 (1H, overlap, H-6′), 7.06 (1H, overlap, H-8′), 7.00 (1H, dd, J = 8.9, 2.9 Hz, H-5), 6.79 (1H, d, J = 8.9 Hz, H-6), 4.24 (2H, s, H-2′), 2.32 (3H, s, H-9′); 13C NMR (150 MHz, CD3OD) δC 205.6 (C-1′), 157.1 (C-1), 150.6 (C-4), 139.4 (C-5′), 135.9 (C-3′), 131.1 (C-4′), 129.6 (C-7′), 128.6 (C-6′), 127.5 (C-8′), 126.0 (C-5), 120.1 (C-2), 119.7 (C-6), 116.3 (C-3), 46.2 (C-2′), 21.4 (C-9′)。以上数据同文献[14]所报道基本一致, 故鉴定化合物4是australins A。
化合物5    黄色固体, 溶于甲醇, ESI-MS m/z 211 [M+H]+, 分子式为C10H10O51H NMR (500 MHz, CD3OD) δH 7.27 (1H, d, J = 2.9 Hz, H-2), 7.01 (1H, dd, J = 8.9, 2.9 Hz, H-4), 6.79 (1H, d, J = 8.9 Hz, H-3), 3.33 (2H, t, J = 6.3, Hz, H-2′), 2.71 (2H, t, J = 6.3 Hz, H-3′); 13C NMR (150 MHz, CD3OD) δC 205.6 (C-1′), 176.5 (C-4′), 156.4(C-2), 150.6 (C-5), 125.8 (C-4), 120.3 (C-1), 119.6 (C-3), 115.4 (C-6), 34.3 (C-2′), 28.5 (C-3′)。以上数据同文献[15]所报道基本一致, 故鉴定化合物5是2,5-dihydroxy-γ-oxobenzenebutanoic acid。
化合物6    黄色固体, 溶于甲醇, ESI-MS m/z 225 [M+H]+, 分子式为C11H12O51H NMR (500 MHz, CD3OD) δH 7.27 (1H, d, J = 2.9 Hz, H-2), 7.01 (1H, dd, J = 8.9, 2.9 Hz, H-4), 6.79 (1H, d, J = 8.9 Hz, H-3), 3.68 (3H, s, H-5′), 3.33 (2H, t, J = 6.3, Hz, H-2′), 2.71 (2H, t, J = 6.3 Hz, H-3′); 13C NMR (150 MHz, CD3OD) δC 205.6 (C-1′), 175.0 (C-4′), 156.4 (C-2), 150.7 (C-5), 125.8 (C-4), 120.4 (C-1), 119.7 (C-3), 115.4 (C-6), 52.5 (C-5′), 34.3 (C-2′), 28.8 (C-3′)。以上数据同文献[15]所报道基本一致, 故鉴定化合物6是methyl 2,5-dihydroxy-γ-oxobenzenebutanoate。
