The study aims to provide an important basis for IMH020 production process and quality control and develop an HPLC method for the determination of related substances in IMH020. The analysis was performed on a Waters Symmetry C18 column (250 mm × 4.6 mm, 5 µm). The mobile phase A was acetonitrile and the mobile phase B was water-0.2% glacial acetic acid solution with isocratic elution. The flow rate was 1.0 mL·min-1 and the column temperature was maintained at 30 ℃. The wavelength was 270 and 300 nm. The injection volume was 10 μL. The results show that all the related substances gained a completely chromatographic separation. Good linear relationships of all the related substances (I-1, I-2, I-3a, I-3b, I-4 and I-5) were obtained (r ≥ 0.998 9) and recoveries were in the range of 93.1%-97.8% (RSD ≤ 2%, n = 9). Only I-3a and I-3b were detected below the limit for specified impurities in the three batches of samples, and the largest unspecified single impurity and the total impurities were within the limit. The HPLC method established in this paper is simple, sensitive and accurate, and can be used for the determination of related substances of IMH020.

为了对IMH020生产工艺过程和质量控制提供重要依据, 建立IMH020中有关物质的高效液相色谱测定法。色谱柱为Waters Symmetry C18 (250 mm × 4.6 mm, 5 µm) 柱, 流动相A为乙腈, 流动相B为体积分数0.2%的乙酸水溶液, 进行等度洗脱; 流速为1.0 mL·min-1; 柱温为30 ℃; 检测波长为270和300 nm; 进样量为10 μL。结果表明, IMH020与有关物质I-1、I-2、I-3a、I-3b、I-4、I-5及未知杂质均能良好分离; 各有关物质在考察范围内线性关系良好, r ≥ 0.998 9, 回收率在93.1%~97.8%之间(RSD ≤ 2%, n = 9)。三批样品中已知杂质仅检出I-3a和I-3b, 并低于限度, 最大未知杂质及总杂质含量质量分数均低于限度。建立的HPLC方法简便、灵敏、准确, 可用于IMH020的有关物质检测。

The study aims to provide an important basis for IMH020 production process and quality control and develop an HPLC method for the determination of related substances in IMH020. The analysis was performed on a Waters Symmetry C18 column (250 mm × 4.6 mm, 5 µm). The mobile phase A was acetonitrile and the mobile phase B was water-0.2% glacial acetic acid solution with isocratic elution. The flow rate was 1.0 mL·min-1 and the column temperature was maintained at 30 ℃. The wavelength was 270 and 300 nm. The injection volume was 10 μL. The results show that all the related substances gained a completely chromatographic separation. Good linear relationships of all the related substances (I-1, I-2, I-3a, I-3b, I-4 and I-5) were obtained (r ≥ 0.998 9) and recoveries were in the range of 93.1%-97.8% (RSD ≤ 2%, n = 9). Only I-3a and I-3b were detected below the limit for specified impurities in the three batches of samples, and the largest unspecified single impurity and the total impurities were within the limit. The HPLC method established in this paper is simple, sensitive and accurate, and can be used for the determination of related substances of IMH020.

高脂血症(hyperlipidemia, HLP) 作为一种复杂的脂质代谢异常, 其特征是总胆固醇(total cholesterol, TC)、甘油三酯(triglyceride, TG) 和低密度脂蛋白胆固醇(low-density lipoprotein cholesterol, LDL-C) 水平升高[1, 2]。LDL-C水平升高与动脉粥样硬化性心血管疾病(atherosclerotic cardiovascular disease, ASCVD) 的风险增加密切相关[3]。他汀类药物是最常用的血脂调节药物, 其通过降低LDL-C水平来减少ASCVD发生风险[4, 5]。然而, 随着他汀类药物在临床上的广泛应用, 其潜在的不良反应也逐渐引起人们的重视, 近10%的患者表现出对他汀类药物不耐受[6, 7]。此外, 他汀类药物的广泛应用也引发了人们对与其他心血管药物相互作用的担忧, 并增加了他汀类药物相关疾病和肝毒性的风险, 尤其是在接受多种药物治疗的患者中[8, 9]
为了开发具有降低LDL-C作用的非他汀类小分子新药, 寻找可以调节与LDL-C密切相关的其他脂蛋白类别的新靶点至关重要。腺苷酸活化蛋白激酶(AMP-activated protein kinase, AMPK) 在细胞内能量代谢平衡和脂质调控中扮演着重要角色, 被认为是治疗高脂血症的潜在靶点之一[10, 11]。IMH020是我国自主研发的口服AMPK直接激动剂, 以虫草素为先导化合物, 经过传统药物化学原理、计算机辅助设计方法和结构优化而得。前期研究已经表明, IMH020能够通过激活AMPK来减少脂质代谢的紊乱, 具有调节血脂的功效, 因此, 有望成为治疗高脂血症的药物之一。
目前, IMH020原料药尚未在各国药典收载, 也暂无公开的质量标准。同时, 对于IMH020及其结构类似物, 尚未发现关于其有关物质测定的文献报道。本研究旨在建立IMH020原料药中有关物质的分析方法, 对6个杂质进行定量测定和方法学验证, 为其生产工艺过程和质量控制提供重要依据。
根据对IMH020合成过程进行全面分析, 并结合降解途径对IMH020制备过程和储藏过程中可能存在的杂质进行分析, 分析结果及6个杂质结构见图 1。有关物质I-1、I-2、I-3a、I-3b和I-4均为IMH020的降解产物, 均为脱乙酰基产物; 此外, I-5为合成中间体, 可能残留至IMH020中, 因此将其纳入质量标准。
仪器  Agilent 1260型HPLC色谱仪(美国Agilent公司); XS105DU型十万分之一电子分析天平(瑞士Mettler-Toledo公司); Milli-Q Direct 8纯水机(美国Millipore公司)。
试药  IMH020原料药(批号: 210101、210102和210103)、IMH020对照品(批号: 210223, 含量质量分数99.8%)、有关物质对照品I-1 (批号: 210323-1, 含量质量分数98.1%)、I-2 (批号: 210323-2, 含量质量分数98.3%)、I-3 (批号: 210511-3, 含量质量分数98.7%)、I-4 (批号: 210516-4, 含量质量分数99.1%)、I-5 (批号: 210607-5, 含量质量分数98.9%) 均由天士力医药集团股份有限公司自制。乙腈、乙酸为色谱纯, 水为超纯水, 其余试剂均为分析纯。
色谱条件  采用Waters Symmetry C18 (250 mm × 4.6 mm, 5 µm) 色谱柱; 以乙腈为流动相A, 以体积分数0.2%的乙酸水溶液为流动相B, 按流动相A-B (35∶65, v/v) 进行等度洗脱; 流速: 1.0 mL·min-1, 检测波长: 270 nm (I-5) 和300 nm (I-1、I-2、I-3a、I-3b、I-4), 柱温: 30 ℃, 进样体积: 10 μL。
溶液制备
空白溶液  35%乙腈水溶液。
供试品溶液  取IMH020原料药50 mg, 精密称定, 置于25 mL量瓶中, 以空白溶液溶解并稀释至刻度, 作为供试品溶液。
IMH020对照品溶液  取IMH020对照品50 mg, 精密称定, 置于25 mL量瓶中, 以空白溶液溶解并稀释至刻度, 制成每1 mL约含2 mg的IMH020对照品溶液, 即得。
对照品储备溶液  分别精密称取I-1、I-2、I-3、I-4、I-5的对照品适量, 以空白溶液溶解并稀释至刻度, 制成每1 mL中含I-1、I-2、I-3、I-4、I-5分别为20.0、20.0、40.0、20.0和20.0 µg的溶液, 作为各杂质对照品储备溶液。
对照品溶液  精密量取各杂质对照品储备溶液适量, 以空白溶液溶解并稀释至刻度, 制成每1 mL中含I-1、I-2、I-3、I-4、I-5分别为2.0、2.0、4.0、2.0和2.0 µg的杂质对照品溶液。
系统适用性溶液  分别精密量取IMH020对照品溶液、各杂质对照品储备溶液适量, 以空白溶液溶解并稀释至刻度, 制成每1 mL中含IMH020、I-1、I-2、I-3、I-4和I-5分别为2.0、2.0、2.0、4.0、2.0和2.0 µg的系统适用性溶液。
线性系列对照品溶液  精密量取IMH020对照品溶液和各杂质对照品储备溶液适量, 以空白溶液定量稀释, 制成IMH020质量浓度分别为0.5、1.0、1.6、2.0、2.4和3.0 μg·mL-1的溶液; 制成I-1质量浓度分别为0.5、1.0、1.6、2.0、2.4和3.0 μg·mL-1的溶液; 制成I-2质量浓度分别为0.5、1.0、1.6、2.0、2.4和3.0 μg·mL-1的溶液; 制成I-3质量浓度分别为1.0、2.0、3.2、4.0、4.8和6.0 μg·mL-1的溶液; 制成I-4质量浓度分别为0.5、1.0、1.6、2.0、2.4和3.0 μg·mL-1的溶液; 制成I-5质量浓度分别为0.5、1.0、1.6、2.0、2.4和3.0 μg·mL-1的溶液。
强制降解实验溶液制备
IMH020储备溶液  取IMH020原料药50 mg, 精密称定, 置25 mL量瓶中, 以空白溶液溶解并稀释至刻度, 摇匀即得。
酸破坏IMH020  量取IMH020储备溶液5.0 mL, 加1 mol·L-1盐酸溶液1.0 mL, 置于45 ℃水浴中加热1 h, 放冷至室温, 加1 mol·L-1氢氧化钠溶液1.0 mL中和。
碱破坏IMH020  量取IMH020储备溶液5.0 mL, 加1 mol·L-1氢氧化钠溶液1.0 mL, 60 ℃水浴中放置8 h, 加1 mol·L-1盐酸溶液1.0 mL中和。
氧化破坏IMH020  量取IMH020储备溶液5.0 mL, 加入2%过氧化氢溶液1.0 mL, 常温放置4 h。
光照破坏IMH020  量取IMH020储备溶液5.0 mL, 光照强度4 500 lx的光照箱中照射24 h。
高温破坏IMH020  量取IMH020储备溶液5.0 mL, 110 ℃烘箱中加热24 h。
取上述各破坏后的样品溶液, 分别转移至10 mL量瓶中, 以空白溶液溶解并稀释至刻度, 摇匀即得。
方法学考察
系统适用性  取系统适用性溶液, 按色谱条件进样, 记录色谱图, 考察方法的系统适用性。
专属性  分别取各强制降解实验溶液, 按色谱条件进样, 记录色谱图, 考察方法的专属性。
检测限及定量限  精密量取各对照品储备溶液适量, 逐级稀释, 按色谱条件进样, 记录色谱图。
线性及校正因子  取各线性系列对照品溶液, 按项下色谱条件进样, 记录色谱图, 以浓度(X, µg·mL-1) 为横坐标, 峰面积(Y) 为纵坐标, 绘制标准曲线, 计算回归方程。
重复性  制备系统适用性溶液, 按色谱条件进样, 记录色谱图。
加样回收率  取IMH020原料药50 mg, 精密称定, 置25 mL量瓶中, 平行制备9份, 分为3组, 分别加入各杂质对照品储备溶液, 以空白溶液定量稀释制成各杂质浓度分别为限度浓度的LOQ、100%和150%, 即得回收率加标溶液。按色谱条件进样, 记录色谱图, 计算回收率。
溶液稳定性  制备系统适用性溶液和供试品溶液, 于室温放置0、2、4、6、8、10和12 h, 按色谱条件进样, 记录色谱图。
耐用性  根据色谱条件, 分别考察了不同色谱柱[Waters Symmetry C18 (250 mm × 4.6 mm, 5 µm)、Agilent Zorbax SB C18 (250 mm × 4.6 mm, 5 µm) 和Resteck Pinnacle Ⅱ C18 (250 mm × 4.6 mm, 5 µm)], 不同流速(0.9、1.0和1.1 mL·min-1), 不同柱温(28、30和32 ℃), 不同流动相A乙腈比例(33%、35%和37%), 不同流动相B乙酸浓度(0.1%、0.2%和0.3%)。
样品测定  取3批IMH020 (批号: 210101、210102、210103), 按照标准制备供试品溶液, 按色谱条件进样分析, 计算各杂质含量。
系统适用性结果表明, IMH020与相邻杂质峰分离良好, 各色谱峰与相邻色谱峰分离度均大于3.0, 理论板数按IMH020峰计算不低于5 000, 且满足系统适用性要求。系统适用性图谱见图 2
专属性结果表明, IMH020在氧化破坏、高温破坏、光照破坏条件下较稳定, 杂质个数、总量无明显变化, 而在酸破坏、碱破坏条件下均有不同程度的降解。各破坏条件下所产生的降解产物均与IMH020能很好分离, 峰纯度分析显示各破坏溶液色谱图中IMH020均为单一峰, 主峰与相邻杂质峰及各杂质峰间均达到完全分离; 主峰纯度角均小于纯度阈值, 且纯度良好, 物料均守恒, 表明该色谱条件的专属性良好。
色谱结果中得到信噪比大于3的浓度为杂质的检测限, 信噪比大于10的浓度为杂质的定量限, 测得的IMH020及杂质I-1、I-2、I-3、I-4、I-5的检测限分别为0.248 1、0.251 6、0.253 9、0.509 1、0.257 7、0.256 7 µg·mL-1, 定量限分别为0.501 9、0.503 5、0.510 7、1.031 3、0.498 8、0.521 2 µg·mL-1。按照有关物质测定供试品溶液质量浓度2.0 mg·mL-1计算, 供试品质量浓度相当于定量限的4 000倍, 相当于检测限的8 000倍; 各杂质的检测限和定量限均远低于其限度, 该方法的灵敏度满足检测要求。
以浓度(X, µg·mL-1) 为横坐标, 峰面积(Y) 为纵坐标, 绘制标准曲线, 计算回归方程, 用标准曲线法计算杂质校正因子, 结果见表 1
重复性试验结果表明, 杂质I-1、I-2、I-3、I-4、I-5和IMH020峰面积的RSD值分别为0.2%、0.2%、0.3%、0.3%、0.6%, 0.1%, 均小于1%, 表明方法重复性良好。
加样回收率试验结果表明, 回收率在93.1%~97.8%之间, RSD均小于2%, 表明方法准确度较高。
系统适用性溶液在室温保存条件下, 杂质I-1、I-2、I-3、I-4、I-5和IMH020峰面积的RSD值分别为0.5%、0.2%、0.3%、0.8%、0.5%、0.7%; 供试品溶液在室温保存条件下, 杂质I-3和IMH020峰面积的RSD值分别为0.9%和0.1%, 杂质I-1、I-2、I-4和I-5均未检出。结果表明, 系统适用性溶液和供试品溶液在室温保存条件下, 17 h内稳定。
耐用性试验结果表明, 系统适用性结果均符合要求, 除保留时间稍有变化外, 各杂质含量、分离度、个数未发生明显变化, 方法的耐用性良好。
杂质含量结果见表 2
取对照品溶液和供试品溶液, 通过二极管阵列检测器(DAD) 进行扫描, 波长范围设为190~550 nm, 选择一个检出杂质峰最多及峰面积最大的波长。经测定, 无法找出一个合适的波长兼顾IMH020和杂质I-1、I-2、I-3a、I-3b、I-4、I-5。其中, IMH020和杂质I-1、I-2、I-3a、I-3b、I-4在300 nm波长附近处均有较强吸收能被有效检出, 所以将IMH020和杂质I-1、I-2、I-3a、I-3b、I-4的检测波长定为300 nm。此外, 杂质I-5在270 nm波长附近有较强吸收, 所以将杂质I-5的检测波长定为270 nm。
IMH020为1类调节血脂新药, 杂质限度的确定参考ICH Q3A (R2) 附件中对杂质限度的要求和《中华人民共和国药典》2020年版(四部) 通则《药品杂质分析指导原则》[12, 13], 结合本品生产实际情况, 对已知杂质确定不同的限量要求。其中, 有关物质I-3所含的两个有关物质峰(I-3a和I-3b), 分离困难并且相应的对照品制备后无法经过分离纯化获得, 因此按照两个色谱峰之和进行方法研究和验证。经多批次样品的检测结果, 将有关物质I-3限度暂定为0.2%, I-1、I-2、I-4、I-5的限度暂定为0.1%, 最大未知杂质的限度暂定为0.1%, 总杂质的限度暂定为0.5%。
本研究比较了甲醇-水和乙腈-水等度洗脱系统, 整体色谱峰型较好且分离度较高, 但IMH020有拖尾现象, 是由于IMH020偏酸性所致。因此进一步考察了乙腈-0.1%甲酸、乙腈-0.1%乙酸、乙腈-0.1%三氟乙酸和乙腈-0.1%磷酸洗脱系统, 最终发现, 乙腈-0.1%乙酸相比较之下, 峰型和分离度最优。后期通过优化和进一步考察, 最终确定乙腈-0.2%乙酸为最优洗脱系统。结果表明, 本方法操作简便、专属性好, 灵敏度高, 线性关系及重复性良好, 可为IMH020原料药有关物质研究提供检测方法与可靠的实验数据, 为其生产工艺过程和质量控制提供重要依据。
