The chemical constituents in dried roots of Atractylodes macrocephala were investigated in this study. Through utilizing normal-phase silica gel and ODS column chromatography, TLC, and semi-preparative HPLC, 4 new sesquiterpenoids were purified from the ethyl acetate extract of A. macrocephala. By various spectroscopic techniques, such as 1D and 2D NMR, HR-ESI-MS, IR, UV, and CD, their structures were identified as atractylmacron A (1), 9β-hydroxyasterolide (2), atractylenolide H (3), atractylenolide J (4). Compound 1 possesses a rare 6/7 bicyclic skeleton.

对苍术属植物白术Atractylodes macrocephala Koidz.的干燥根进行化学成分研究。运用正相硅胶、ODS、半制备HPLC等色谱方法从白术乙酸乙酯部位分离纯化得到4个倍半萜类化合物, 采用1D和2D NMR、HR-ESI-MS、IR、UV、CD等技术手段鉴定其结构为白术酮A (1)、9β-羟基紫菀内酯(2)、白术内酯H (3)、白术内酯J (4)。化合物1~4均为新的倍半萜类化合物, 其中化合物1具有一个少见的6/7双环骨架结构。

对苍术属植物白术Atractylodes macrocephala Koidz.的干燥根进行化学成分研究。运用正相硅胶、ODS、半制备HPLC等色谱方法从白术乙酸乙酯部位分离纯化得到4个倍半萜类化合物, 采用1D和2D NMR、HR-ESI-MS、IR、UV、CD等技术手段鉴定其结构为白术酮A (1)、9β-羟基紫菀内酯(2)、白术内酯H (3)、白术内酯J (4)。化合物1~4均为新的倍半萜类化合物, 其中化合物1具有一个少见的6/7双环骨架结构。

The chemical constituents in dried roots of Atractylodes macrocephala were investigated in this study. Through utilizing normal-phase silica gel and ODS column chromatography, TLC, and semi-preparative HPLC, 4 new sesquiterpenoids were purified from the ethyl acetate extract of A. macrocephala. By various spectroscopic techniques, such as 1D and 2D NMR, HR-ESI-MS, IR, UV, and CD, their structures were identified as atractylmacron A (1), 9β-hydroxyasterolide (2), atractylenolide H (3), atractylenolide J (4). Compound 1 possesses a rare 6/7 bicyclic skeleton.

中药白术为菊科(Compositae) 苍术属植物白术(Atractylodes macrocephala Koidz.) 的干燥根茎, 载于《神农本草经》, 性苦、甘、温, 具有健脾益气, 燥湿利水、止汗、安胎的功效, 主要用于治疗脾虚食少、腹胀泄泻、痰饮眩悸、水肿、自汗、胎动不安等症。白术为临床常用大宗药材, 主产于江苏、浙江、福建等地, 是中国药典(2020年版) 收载品种[1, 2]。现代药理学研究表明, 白术具有免疫调节、抗肿瘤、抗氧化、抗衰老、对胃肠运动的双向调节作用、神经系统保护、保肝、调节血糖和血脂等活性[3-12]。白术的化学成分主要包括倍半萜类、聚炔类、三萜、香豆素、苯类衍生物、寡糖、多糖等[2, 3]。对倍半萜类化合物的研究一直以来都是研究的热点[13-15]。近年来, 由于其重要的临床应用价值, 从白术中发现一系列的倍半萜及其二聚体、杂聚体等结构新颖的化合物[16-18], 为白术的进一步开发和利用提供了重要依据。为进一步揭示白术的化学组成, 本文从白术乙酸乙酯部位分离得到4个新的倍半萜类化合物(图 1), 白术酮A (1)、9β-羟基紫菀内酯(2)、白术内酯H (3)、白术内酯J (4); 其中, 化合物1具有一个少见的6/7双环骨架结构。
化合物1为白色无定形粉末, HR-ESI-MS谱中显示其准分子离子峰m/z 273.146 1 [M+Na]+ (calcd. for C15H22O3Na+ 273.146 2), 结合氢谱和碳谱数据(表 1), 推断其分子式为C15H22O3, 不饱和度为5。1H NMR谱显示一组末端双键氢信号δH 4.82 (1H, d, J = 1.0 Hz, H-15a)、4.49 (1H, d, J = 1.0 Hz, H-15b), 一个连氧次甲基质子信号δH 4.72 (1H, d, J = 2.0 Hz, H-8), 一个次甲基质子信号δH 2.90 (1H, ddd, J = 12.6, 6.2, 2.0 Hz, H-7), 两组甲基质子信号δH 2.25 (3H, s, H-13)、0.72 (3H, s, H-14), 此外, 在高场区还有11个烷烃质子信号。13C NMR谱显示14个碳信号, 结合高分辨质谱数据分析其可能有两个碳信号重叠。通过分析HSQC谱数据, 将14个碳信号归为酮羰基碳信号δC 210.3 (C-9, 12), 双键季碳信号δC 148.0 (C-4), 季碳信号δC 40.4 (C-11), 连氧次甲基碳信号δC 75.3 (C-8), 次甲基碳信号δC 52.5 (C-7)、47.7 (C-5), 双键亚甲基碳信号δC 107.5 (C-15), 亚甲基碳信号δC 55.8 (C-10)、41.4 (C-1)、36.6 (C-3)、24.9 (C-6)、23.0 (C-2), 甲基碳信号δC 26.4 (C-13)、17.1 (C-14)。以上信息提示化合物1具有类似桉叶烷型倍半萜的骨架结构。
通过1H-1H COSY和HMBC谱进一步确认化合物1的平面结构(图 2)。HMBC谱中, H-15 (δH 4.82, 4.49) 与C-3 (δC 36.6)、C-5 (δC 47.7) 有相关, 提示C4=C15环外双键的存在。H-14 (δH 0.72) 与C-1 (δC 41.4)、C-5 (δC 47.7)、C-10 (δC 55.8)、C-11 (δC 40.4)有相关, 提示11位角甲基的存在。结合H-1 (δH 1.50) 与C-2 (δC 23.0) 的HMBC相关以及H-2b (δH 1.54) 与H-3a (δH 2.39) 的 1H-1H COSY相关, 确证了含环外双键的甲基取代环己烷结构片段。H-13 (δH 2.25) 与C-12 (δC 210.3)、C-7 (δC 52.5) 的HMBC相关提示7位乙酰基取代。仅与H-7有偶合的H-8 (δH 4.72) 与C-6 (δC 24.9)、C-7 (δC 52.5)、C-9 (δC 210.3)、C-12 (δC 210.3) 有HMBC相关, H-10 (δH 2.29) 与C-9 (δC 210.3) 有HMBC相关, 提示8位被羟基取代和δC 210.3为两个重合的酮羰基碳信号, 羰基分别位于C-9和C-12。以上信息表明化合物具有少见的6/7双环骨架结构, 可能是通过桉叶烷型倍半萜重排而成。白术中桉叶烷型倍半萜的角甲基和H-5通常为反式构型, H-5与H-7的NOE相关(图 3) 说明H-7与H-5位于七元环的同侧, H-7与H-8的偶合常数2.0 Hz提示两个氢位于七元环的同侧。H-13与H-14的NOE相关说明CH3-14与7位乙酰基位于七元环的同侧。由此, 确定了化合物1的相对构型。进而通过比对化合物1的实测CD谱与计算CD谱(图 4), 确定化合物1的绝对构型为5S, 7S, 8S, 11R。最终确定了化合物1的结构, 将其命名为白术酮A (atractylmacron A)。
化合物2为白色无定形粉末, HR-ESI-MS谱中显示其准分子离子峰m/z 271.129 8 [M+Na]+ (calcd. for C15H20O3Na+ 271.130 5), 结合氢谱和碳谱数据, 推断其分子式为C15H20O3, 不饱和度为6。1H NMR谱显示一组末端双键氢信号δH 4.91 (1H, br s, H-15a)、4.64 (1H, br s, H-15b), 两个连氧次甲基质子信号δH 4.73 (1H, d, J = 8.9 Hz, H-8)、3.12 (1H, d, J = 8.9 Hz, H-9), 一个次甲基质子信号δH 1.84 (1H, dd, J = 10.8, 3.7 Hz, H-5), 两组甲基质子信号δH 1.83 (3H, s, H-13)、0.90 (3H, s, H-14)。13C NMR谱显示15个碳信号, 结合HSQC谱, 将它们分别归为一个羰基碳信号δC 174.7 (C-12), 两组双键碳信号δC 158.6 (C-7)、148.0 (C-4)、121.6 (C-11)、107.9 (C-15), 两个连氧次甲基碳信号δC 83.6 (C-8)、83.5 (C-9), 一个次甲基碳信号δC 48.6 (C-5), 一个季碳信号δC 41.7 (C-10), 四个亚甲基碳信号δC 37.0 (C-1)、36.5 (C-3)、25.6 (C-6)、22.4 (C-2), 两个甲基碳信号δC 11.1 (C-14)、8.6 (C-13)。以上碳谱数据, 除了C-1、C-5、C-8、C-9、C-10、C-14位碳的化学位移, 与化合物9α-羟基紫菀内酯[19]相似, 提示化合物2可能是桉叶烷型倍半萜内酯类化合物。
HMBC谱中H-15 (δH 4.91, 4.64) 与C-3 (δC 36.5)、C-5 (δC 48.6) 有相关, 提示C4=C15环外双键的存在。H-14 (δH 0.90) 与C-1 (δC 37.0)、C-5 (δC 48.6)、C-9 (δC 83.5)、C-10 (δC 41.7) 有相关, 提示存在10位角甲基。H-9 (δH 3.12) 与C-1 (δC 37.0)、C-8 (δC 83.6)、C-10 (δC 41.7)、C-14 (δC 11.4) 有HMBC相关, 说明9位连有羟基。H-13 (δH 1.83) 与C-12 (δC 174.7)、C-7 (δC 158.6) 的HMBC相关以及H-8 (δH 4.73) 与C-6 (δC 25.6)、C-7 (δC 158.6)、C-9 (δC 83.5)、C-11 (δC 121.6) 的HMBC相关提示α-甲基取代的α, β-不饱和内酯环的存在。确定化合物2具有与9α-羟基紫菀内酯相同的平面结构。与9α-羟基紫菀内酯(JH8/H9 = 3.6) 不同, 化合物2的氢谱中H-8与H-9的偶合常数为8.9 Hz, 提示H-8和H-9为反式构型, 即9位羟基为β构型。NOESY谱中H-8与H-14、H-5和H-9的相关确证了这一推断。进一步通过比较化合物2的计算和实测CD谱确定其绝对构型, 命名为9β-羟基紫菀内酯(9β-hydroxyasterolide)。
化合物3为白色无定形粉末, 根据其HR-ESI-MS谱中显示的准分子离子峰m/z 301.140 7 [M+MeOH+Na]+ (calcd. for C16H22O4Na+, 301.141 6), 结合氢谱和碳谱数据, 推断其分子式为C15H18O3, 不饱和度为7。1H NMR谱显示一个双键氢信号δH 5.54 (1H, br s, H-9), 一组连氧亚甲基质子信号δH 3.62 (1H, d, J = 10.9 Hz, H-8)、3.49 (1H, d, J = 10.9 Hz, H-9), 两组甲基质子信号δH 1.90 (3H, s, H-13)、1.25 (3H, s, H-14)。13C NMR谱显示15个碳信号, 结合HSQC谱, 将其归为一个羰基碳信号δC 171.3 (C-12), 两组双键碳信号δC 148.2 (C-7)、1478 (C-8)、121.1 (C-11)、120.4 (C-9), 一个次甲基碳信号δC 46.6 (C-5), 两个季碳信号δC 73.1 (C-4)、36.5 (C-10), 五个亚甲基碳信号δC 53.3 (C-15)、38.9 (C-1)、36.6 (C-3)、19.9 (C-6)、17.4 (C-2), 两个甲基碳信号δC 21.2 (C-14)、8.7 (C-13)。以上数据, 除了C4=C15环外双键变为三元氧环上连氧的季碳和亚甲基外, 其他与白术内酯Ⅰ[20]相似, 提示化合物3可能是桉叶烷型倍半萜内酯类化合物。
HMBC谱中H-14 (δH 1.25) 与C-1 (δC 38.9)、C-5 (δC 46.6)、C-9 (δC 120.4)、C-10 (δC 36.5) 有相关, 提示存在10位角甲基。H-9 (δH 5.54) 与C-1 (δC 38.9)、C-5 (δC 46.6)、C-8 (δC 147.8) 有HMBC相关, 说明存在C9=C8双键。H-15 (δH 3.62, 3.49) 与C-3 (δC 36.6)、C-4 (δC 73.1)、C-5 (δC 46.6) 有HMBC相关, 结合C-15 (δC 53.3) 化学位移, 提示C4-C15环氧键的存在。H-13 (δH 1.90) 与C-12 (δC 171.3)、C-7 (δC 148.2)、C-11 (δC 121.1) 的HMBC相关提示α-甲基取代的α, β-不饱和内酯环的存在。至此确定化合物3的平面结构。NOESY谱中观察到H-14和H-15a与H-6β的相关, 推测C-15为β构型。最后通过比较化合物3的计算和实测CD谱确定其绝对构型为4R, 5R, 10R, 命名为白术内酯H (atractylenolide H)。
化合物4为白色无定形粉末, 根据其HR-ESI-MS谱数据(m/z 313.140 4 [M+Na]+, calcd. for C17H22O4Na+ 313.141 1) 确定其分子式为C17H22O4, 不饱和度为7。1H NMR谱显示一个双键质子信号δH 5.83 (1H, br s, H-3), 一个连氧次甲基质子信号δH 4.95 (1H, dd, J =11.7, 6.8 Hz, H-8), 一组连氧亚甲基质子信号δH 4.61 (1H, d, J = 12.3 Hz, H-15a)、4.49 (1H, d, J = 12.3 Hz, H-15b), 三组甲基质子信号δH 2.08 (3H, s, H-2′)、1.82 (3H, s, H-13)、0.97 (3H, s, H-14)。13C NMR谱显示17个碳信号, 结合HSQC谱, 将其归为两个羰基碳信号δC 174.8 (C-12)、170.8 (C-1′), 两组双键碳信号δC 162.1 (C-7)、131.8 (C-4)、129.3 (C-3)、120.9 (C-11), 一个连氧次甲基碳信号δC 78.3 (C-8), 一个次甲基碳信号δC 45.2 (C-5), 一个季碳信号δC 33.8 (C-10), 五个亚甲基碳信号δC 66.9 (C-15)、46.7 (C-9)、36.5 (C-1)、25.1 (C-6)、22.7 (C-2), 三个甲基碳信号δC 21.1 (C-2′)、16.0 (C-14)、8.4 (C-13)。以上数据与化合物15-acetoxy-iso-allo-alantolactone[21]相比, 缺少了13位末端双键的质子信号, 多了一组甲基质子信号, 提示化合物4可能是乙酰基取代的桉叶烷型倍半萜内酯类化合物。
HMBC谱中H-14 (δH 0.97) 与C-1 (δC 36.5)、C-5 (δC 45.2)、C-9 (δC 46.7)、C-10 (δC 33.8) 有相关, 提示存在10位角甲基。H-15 (δH 4.61, 4.49) 与C-3 (δC 129.3)、C-4 (δC 131.8)、C-5 (δC 45.2) 有相关, 提示C3=C4环内双键的存在。H-13 (δH 1.82)与C-12 (δC 174.8)、C-11 (δC 120.9)、C-7 (δC 162.1) 的HMBC相关以及H-8 (δH 4.95) 与C-7 (δC 162.1)、C-9 (δC 46.7)、C-11 (δC 120.9) 的HMBC相关提示α-甲基取代的α, β-不饱和内酯环的存在。H-2′ (δH 2.08) 与C-1′ (δC 170.8) 以及H-15 (δH 4.61, 4.49) 与C-1′ (δC 170.8) 的HMBC相关说明15位被乙酰氧基取代。综合以上信息确定化合物4的平面结构。NOESY谱中H-8与H-14的相关说明H-8为α构型。最终通过比较计算和实测CD谱和DP4+分析其计算碳谱数据确定化合物4的绝对构型为5R, 8S, 10R, 将其命名为白术内酯J (atractylenolide J)。
Thermo QE plus高分辨质谱(德国Thermo Electron公司); Shimadzu UV-2550紫外光谱仪和Shimadzu IR Tracer-100光谱仪(日本岛津公司); Bruker AV-400核磁共振仪(德国Bruker公司); Anton Paar MCP 5100旋光测定仪(澳大利亚Anton Paar公司); Chirascan VX圆二色光谱仪(英国Applied Photophysics公司); 柱色谱用硅胶(100~200、200~300目), 青岛海洋化工有限公司; 中压柱色谱用ODS-A-HG (12 nm, S-50 μm), 日本YMC公司; STI501半制备HPLC [赛智科技(杭州) 有限公司]; CDCl3购自宁波萃英化学技术有限公司; 色谱甲醇、色谱乙腈及其他分析纯试剂购自重庆市钛新化工有限公司或川东化工有限公司。
白术饮片购买自四川成都新荷花中药材交易市场, 经西藏农牧学院兰小中教授鉴定为菊科植物白术(A. macrocephala) 的根, 药材标本(2022-AM-1001) 保存在西南大学药学院中药新药筛选重庆市重点实验室。
白术饮片(100 kg) 经粉碎后, 用8倍量95%乙醇浸泡过夜, 提取3次, 回收溶剂后得到乙醇提取物4.43 kg。将乙醇提取物加适量水混悬, 用乙酸乙酯萃取, 得到乙酸乙酯部位1.80 kg。乙酸乙酯部位上硅胶柱, 石油醚-乙酸乙酯100∶0~0∶100梯度洗脱, 得到Fr. A~Fr. G, 共7个组分。Fr. C (337 g) 进行正相硅胶柱层析, 石油醚-乙酸乙酯50∶1~0∶100梯度洗脱, 得到Fr. C-1~Fr. C-22。Fr. C-19 (29.7 g) 上正相色谱柱, 石油醚-乙酸乙酯50∶1~0∶1梯度洗脱, 得到Fr. C-19-1~Fr.C-19-11, 共11个组分。Fr. C-19-4 (10.1 g) 上中压制备柱(400 mL ODS, 60%~100%甲醇, 15 mL·min-1), 得到Fr. C-19-4-1~Fr. C-19-4-25。Fr. C-19-4-4 (383 mg) 经半制备液相(Cosmosil 5C18-MS-Ⅱ 10 mm × 250 mm, 甲醇-水, 70∶30, 3 mL·min-1) 制备, 得到化合物1 (2.0 mg, tR = 21.50 min)。Fr. C-19-5 (7.6 g) 经中压液相柱(400 mL ODS, 60%~100%甲醇, 15 mL·min-1) 分离, 得到Fr. C-19-5-1~Fr. C-19-5-16。Fr. C-19-5-4 (261 mg) 经半制备液相(Cosmosil 5C18-MS-Ⅱ 10 mm × 250 mm, 甲醇-水, 56∶44, 3 mL·min-1) 得到Fr. C-19-5-4-1~Fr. C-19-5-4-2。Fr.C-19-5-4-2经半制备液相(Cosmosil cholester柱, 10 mm × 250 mm, 乙腈-水40∶60, 3 mL·min-1) 纯化, 得到2 (3.9 mg, tR = 30.52 min) 和4 (7.2 mg, tR = 44.28 min)。Fr. C-19-5-6 (197 mg) 经半制备液相(Cosmosil 5C18-MS-Ⅱ 10 mm × 250 mm, 甲醇-水, 60∶40, 3 mL·min-1; Cosmosil cholester柱10 mm × 250 mm, 乙腈-水, 43∶57, 3 mL·min-1) 制备, 得到化合物3 (2.8 mg, tR = 33.62 min)。
