Three compounds were obtained from the crude extract of the fermentation broth of endophyte Myrothecium roridum IFB-E008 in Trachelospermum jasminoides by silica gel column chromatography, Sephadex LH-20 gel column chromatography and high performance liquid chromatography. They were determined as 3′-iso-isororidin A (1), verrol (2) and N-acetyltryptamine (3), respectively, through HR-MS, 1D and 2D NMR and literature comparison. 3′-iso-isororidin A (1) is a new trichothecene macrolide that has never been reported in the literature. The in vitro cytotoxicity assay showed that compound 2 had certain cytotoxicity against the human gastric cell line SGC-7901 with half inhibition concentration (IC50) of 59.79 μg·mL-1 (158.1 μmol·L-1), while IC50 value of the positive control cisplatin was 6.58 μg·mL-1 (21.9 μmol·L-1).

采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱以及高效液相色谱法从络石(Trachelospermum jasminoides) 内生菌Myrothecium roridum IFB-E008发酵液的粗浸膏中分离得到3个化合物。经高分辨质谱、一维和二维核磁共振谱及文献比对等方法, 鉴定为: 3′-iso-isororidin A (1)、verrol (2) 和N-乙酰色胺(3), 其中, 3′-iso-isororidin A (1) 是一个未见文献报道的新单端孢霉烯大环内酯。体外细胞毒活性测定表明, 化合物2对人胃癌细胞株SGC-7901具有一定的抑制活性, 半数抑制浓度(half inhibition concentration, IC50) 为59.79 μg·mL-1 (158.1 μmol·L-1), 阳性对照顺铂的IC50值为6.58 μg·mL-1 (21.9 μmol·L-1)。

采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱以及高效液相色谱法从络石(Trachelospermum jasminoides) 内生菌Myrothecium roridum IFB-E008发酵液的粗浸膏中分离得到3个化合物。经高分辨质谱、一维和二维核磁共振谱及文献比对等方法, 鉴定为: 3′-iso-isororidin A (1)、verrol (2) 和N-乙酰色胺(3), 其中, 3′-iso-isororidin A (1) 是一个未见文献报道的新单端孢霉烯大环内酯。体外细胞毒活性测定表明, 化合物2对人胃癌细胞株SGC-7901具有一定的抑制活性, 半数抑制浓度(half inhibition concentration, IC50) 为59.79 μg·mL-1 (158.1 μmol·L-1), 阳性对照顺铂的IC50值为6.58 μg·mL-1 (21.9 μmol·L-1)。

Three compounds were obtained from the crude extract of the fermentation broth of endophyte Myrothecium roridum IFB-E008 in Trachelospermum jasminoides by silica gel column chromatography, Sephadex LH-20 gel column chromatography and high performance liquid chromatography. They were determined as 3′-iso-isororidin A (1), verrol (2) and N-acetyltryptamine (3), respectively, through HR-MS, 1D and 2D NMR and literature comparison. 3′-iso-isororidin A (1) is a new trichothecene macrolide that has never been reported in the literature. The in vitro cytotoxicity assay showed that compound 2 had certain cytotoxicity against the human gastric cell line SGC-7901 with half inhibition concentration (IC50) of 59.79 μg·mL-1 (158.1 μmol·L-1), while IC50 value of the positive control cisplatin was 6.58 μg·mL-1 (21.9 μmol·L-1).

单端孢霉烯是一类以12, 13-epoxytrichothec-9-ene (EPT) 为骨架特征的倍半萜烯化合物, 其主要是由位于不同染色体上的16个TRI基因(TRI1~TRI14TRI16TRI101) 组成的基因簇所调控合成。一分子的法尼基焦磷酸(farnesyl pyrophosphate, FPP) 经TRI5编码的萜烯合成酶—单端孢霉烯合成酶(trichodiene synthase) 催化环化形成单端孢霉二烯(trichodiene, TDN); TDN随后经历由TRI1TRI4TRI11TRI13编码的单端孢霉二烯氧化酶—细胞色素P450单加氧酶催化的一系列氧化反应(包括: C-2、C-3、C-11以及C-12位的氧化反应) 以及由TRI3TRI7TRI16TRI101编码的酰基转移酶催化的酯化反应, 有时还会发生其他修饰反应, 从而产生具有EPT骨架特征的单端孢霉烯化合物[1]。若C-4和/或C-15继续发生衍生化反应, 就形成复杂的单端孢霉烯化合物, 包括A~D四种结构类型[1, 2]; 其中, type D为单端孢霉烯大环内酯, 其从结构上可分为verrucarins (主要是C27化合物) 和roridins (主要C29化合物) 两大类。Roridins和verrucarins结构非常相似, roridins的C-6′处C2侧链氧化裂解即生成相应的verrucarins; roridins先后经历C-6′, 12′缩合成环、C-12′, 14′缩合成环可生成satratoxins、mytoxins、roritoxins和vertisporins等衍生物[3]。C-9, 10双键和C-12, 13-epoxy是单端孢霉烯的典型结构特征, 但一些内共生菌却能产生10, 13-碳环单端孢霉烷衍生物[4, 5], 内共生对单端孢霉烯生物合成的影响值得关注。
单端孢霉烯类化合物具有丰富的结构和广泛的药理活性, 其中, 体外抗肿瘤活性开展较多。Lakornwong等[6]Myrothecium roridum中分离得到2个新单端孢霉烯大环内酯6′, 12′-epoxymyrotoxin A和7′-hydroxymytoxin B以及4个已知类似物myrotoxin B、myrotoxin D hydrate、2′, 3′-epoxymyrothecine A和miotoxin A。进一步研究发现, 除2′, 3′-epoxymyrothecine A外, 其他5个化合物对疟原虫Plasmodium falciparum均表现出显著的抗疟活性; 同时具有强体外细胞毒活性, 对KB细胞和NCI-H187细胞的半数抑制浓度(half inhibition concentration, IC50) 为0.60~112.28 nmol·L-1, 对Vero细胞的IC50值为1.50~46.51 nmol·L-1。Parvatkar等[7]研究发现, 单端孢霉烯化合物anguidin (又名: anguidine) 和verrucarin A对恶性疟原虫无性血液阶段表现出良好的抗疟活性, 半数有效浓度(half effective concentration, EC50) 分别为6.16和0.45 nmol·L-1。值得一提的是, anguidin曾作为抗肿瘤药物进入二期临床研究[8-10]
研究发现, 单端孢霉烯大环内酯是漆斑菌Myrothecium sp.、毛壳菌Chaetomium sp.、黑色葡萄状穗菌Stachybotrys sp.、拟茎点霉菌Phomopsis sp.等真菌的特征次生代谢产物。Myrothecium roridum IFB-E008是从传统药用植物络石(Trachelospermum jasminoides) 中分离获得的一株内生真菌, 前期曾从其发酵液中分离得到一个新单端孢霉烯大环酯myrothecine D[5], 本文经进一步分离纯化又得到1个新单端孢霉烯大环内酯3′-iso-isororidin A (1) 以及2个已知物verrol[11] (2) 和N-乙酰色胺(N-acetyltryptamine) [12] (3) (图 1)。体外细胞毒活性测定表明, 化合物2对人胃癌细胞株SGC-7901具有一定的增殖抑制活性。
化合物1为白色固体, 其高分辨电喷雾质谱(HR-ESI-MS) 显示m/z 549.270 9 [M+OH]-, 567.236 8 [M+Cl]-, 说明其相对分子质量为532, 分子式为C29H40O9 (C29H41O10理论值为549.270 5), 具有10个不饱和度。化合物11H和13C NMR谱显示单端孢霉烯大环内酯化合物的特征信号[5]1H NMR谱中, 一对相互偶合的氢信号δH-13a 3.10和δH-13b 2.81 (J = 3.6 Hz) 说明其结构中存在12, 13-环氧环, 另有2个甲基单峰δH-14 0.89和δH-16 1.75, 2个甲基双峰δH-12′ 0.91 (J = 7.2 Hz) 和δH-14′ 1.17 (J = 6.6 Hz); 13C NMR谱显示29个碳信号, 包括: 6个双键碳信号δC-9 140.8、δC-10 118.2、δC-7′ 137.0、δC-8′ 127.0、δC-9′ 141.1和δC-10′ 118.9, 2个羰基碳信号δC-1′ 175.3和δC-11′ 167.2。上述NMR数据说明化合物1是roridin型的单端孢霉烯大环内酯。进一步分析化合物11H和13C NMR谱发现, 其与roridin A[13, 14]和isororidin A[15] (两者分子式皆为C29H40O9) 比较相似, 但也存在明显的差异, 其可能是roridin A和isororidin A的异构体。
与roridin A和isororidin A相比, 化合物1的C-3′和C-12′的化学位移皆向高场显著移动(ΔδC-3′ = -4.1和-3.7; ΔδC-12′ = -4.0和-3.7); 同时, 与roridin A相比, 化合物1的H-3′的化学位移向低场显著移动(ΔδH-3′ = 0.38), H-12′的化学位移向高场显著移动(ΔδH-12′ = -0.18)。这些信息提示, 化合物1可能为roridin A和isororidin A的C-3′手性异构体, 即: 化合物1的H-3′处于α位、C-3′为S型。与roridin A和isororidin A相比, 化合物1的C-5′、C-6′、C-7′和C-13′的化学位移皆明显向高场移动(ΔδC-5′ = -2.7和-2.0; ΔδC-6′ = -2.6和-0.9; ΔδC-7′ = -2.4和-2.0; ΔδC-13′ = -2.9和-1.8); 同时, 与roridin A相比, H-6′和H-13′的化学位移也向低场移动(ΔδH-6′ = 0.37; ΔδH-13′ = 0.18)。这说明化合物1与roridin A和isororidin A还可能存在C-6′和C-13′的手性异构。化合物11H和13C NMR谱中, J6′, 7′ = 2.4 Hz和ΔδC-7′/C-8′ = 10, 说明H-6′处于β位、C-6′为R[16, 17]。随后HSQC、HMBC、COSY确证化合物1的平面结构如图1所示。在NOESY谱中, 观察到δH-14′ 1.17与δH-6′ 3.89、δH-7′ 6.02相关, δH-6′ 3.89和δH-7′ 6.02相关, 说明13′-CH3处于β位、H-13′处于α位, 因此, C-13′为S型; 还观察到, δH-12′ 0.91与δH-2′ 4.21、δH-5′ 3.52相关、δH-5′ 3.52与δH-6′ 3.89相关, 说明H-2′处于β位, C-2′为S型(图 2)。再结合单端孢霉烯大环内酯的生物合成途径, 将化合物1的其他手性中心构型确定为2R, 4R, 5S, 6R, 9S, 10S, 11R, 12R。因此, 化合物1被确定为3′-iso-isororidin A (图 1), 该结构尚未见文献报道, 其1H和13C NMR谱归属列于表 1
MTT法测定显示, 化合物2对人胃癌细胞株SGC-7901具有明显的增殖抑制活性, 其IC50值为59.79 μg·mL-1 (158.1 μmol·L-1), 而阳性对照顺铂对SGC-7901细胞增殖抑制的IC50值为6.58 μg·mL-1 (21.9 μmol·L-1)。
AVANCE600核磁共振仪、UHR-TOF-Maxis超高分辨飞行时间质谱仪(德国Bruker公司); Hitachi Primaide高效液相色谱仪(日本日立公司); P200Ⅱ高效液相色谱仪(大连依利特分析仪器有限公司); Jasco J-810圆二色谱仪(日本分光公司); Thermo Series Ⅱ CO2培养箱(美国Thermo Fisher Scientific公司); SYNERGY2多功能酶标仪(美国BioTek公司) 等。
柱色谱硅胶(200~300目, 青岛海洋化工厂分厂); Sephadex LH-20 (瑞典Pharmacia Biotech公司); Apollo C18色谱柱(150 mm × 4.6 mm, 5 μm, 美国Alltech科技有限公司); Hypersil ODS2柱(250 mm × 4.6 mm, 5 μm, 大连依利特分析仪器有限公司); CDCl3 (美国Aldrich公司, 生产批号: MW09413); CD3OD (青岛腾龙微波科技有限公司, 生产批号: MFSA2005T06); 色谱甲醇(美国TEDIA化学试剂有限公司, 生产批号: 18080613); 人胃癌细胞株SGC-7901 (上海生命科学研究院细胞库); RPMI 1640培养基(美国Gibco公司, 生产批号: 8123499); 四甲基偶氮唑蓝(MTT, 美国Amresco公司, 生产批号: 1107S056); 顺铂注射液(江苏豪森药业股份有限公司, 生产批号: 181103) 等。
络石内生真菌Myrothecium roridum IFB-E008 (Genbank号: EF211124) 采用査氏培养基(蔗糖30 g、硝酸钠3 g、磷酸氢二钾1 g、酵母膏1 g、氯化钾0.5 g、硫酸镁0.5 g、硫酸亚铁0.01 g和水1 000 mL) 进行液体发酵, 共发酵300瓶(500 mL培养基/1 000 mL三角瓶), 28 ℃、150 r·min-1培养10天。发酵液经乙酸乙酯萃取3次, 减压去除溶剂得粗浸膏12.2 g。
发酵液粗浸膏经硅胶柱分离, 梯度洗脱(氯仿-甲醇, 100∶0→0∶100), 经TLC合并得到6个极性组分(Fr.1~6)。组分Fr.2先后经硅胶柱和Sephadex LH-20凝胶柱分离得到Fr.2-1~2-3, 其中, Fr.2-3进一步经HPLC (Apollo C18色谱柱, 220 nm, 甲醇-水, 53∶47, 1.1 mL·min-1) 分离纯化得到化合物1 (1.8 mg, tR = 25.0 min)。组分Fr.3经硅胶柱分离, 再进一步经HPLC (Hypersil ODS2柱, 220 nm, 甲醇-水, 57∶43, 0.5 mL·min-1) 分离纯化得到化合物3 (9.7 mg, tR = 10.4 min)。组分Fr.4经硅胶柱分离得到Fr.4-1~4-3, 其中, Fr.4-2再经Sephadex LH-20凝胶柱和HPLC (Hypersil ODS2柱, 220 nm, 甲醇-水, 5∶5, 1 mL·min-1) 分离纯化得到化合物2 (1.8 mg, tR = 13.2 min)。
3′-iso-Isororidin A (1) 为白色固体, 分子式C29H40O9。HR-ESI-MS m/z: 549.270 9 [M+OH], 567.236 8 [M+Cl] (C29H41O10理论值: 549.270 5); UV (CH3OH): λmax (logε) = 195 (4.08), 256 (3.71) nm; CD (CH3OH): λmaxε) = 217 (1.95), 244 (-0.66) nm; 1H NMR (CDCl3, 600 MHz) 和13C NMR (CDCl3, 150 MHz) 数据见表 1

N-乙酰色胺(3) 为白色固体, 分子式C12H14N2O。HR-ESI-MS m/z: 225.100 2 [M+Na]+, 203.117 6 [M+H]+ (C12H14N2NaO理论值: 225.099 8); 1H NMR (CD3OD, 600 MHz) δH: 7.49 (1H, d, J = 8.4 Hz, H-4), 7.26 (1H, d, J = 8.4 Hz, H-7), 7.02 (1H, d, J = 7.2, 7.8 Hz, H-5), 7.00 (1H, s, H-2), 6.93 (1H, td, J = 7.8, 6.6 Hz, H-6), 3.40 (2H, t. 参考文献[18]方法, 采用MTT法测定化合物的体外细胞毒活性, 顺铂为阳性对照。收集对数生长期肿瘤细胞, 以1×104个/孔接种于96孔板, 贴壁培养24 h后, 药物组分别加入一定量待测化合物(终质量浓度1、10、20、50和100 μg·mL-1), 阴性对照组和空白组分别加入等体积DMSO和培养液(设5个复孔)。培养48 h后, 每孔加入20 μL MTT, 继续培养4 h; 然后弃去培养液, 每孔加入150 μL DMSO, 37 ℃振荡10 min, 最后用酶标仪490 nm波长下测定各孔吸光度。
