Lipopolysaccharides (LPS) are major outer membrane components of Gram-negative bacteria. Unlike most Gram-negative bacteria, Acinetobacter baumannii can still survive and acquire polymyxin resistance after complete loss of LPS. Previous studies of LPS-deficient Acinetobacter baumannii mostly focused on LPS-deficient strains induced by polymyxin, whose background was complex and unstable. To investigate LPS loss-mediated polymyxin resistant-Acinetobacter baumannii, this study constructed a stable and clear background LPS-deficient strain by knocking out lpxC gene in Acinetobacter baumannii ATCC 19606 with CIRSPR/Cas9, and then studied the phenotypic changes of the lpxC-deficient strain including morphology, growth rate, antibiotic susceptibility, virulence, membrane permeability, and membrane potential. Animal experiments were approved by the Animal Care and Welfare Committee Institute of Medicinal Biotechnology, CAMS and PUMC (approval number: IMB-20240119D9). The results indicated that the lpxC gene of Acinetobacter baumannii ATCC 19606 was successfully knocked out. After losing the lpxC, the strain underwent the morphological change from rod-shaped to spherical. Furthermore, it leads to reduced growth rate, enhanced membrane permeability, decreased membrane potential, lower virulence, and increased antibiotic susceptibility to β-lactams, quinolones, aminoglycosides, macrolides, glycopeptides. The lpxC deletion results in significant changes in membrane homeostasis and adaptability of Acinetobacter baumannii. Understanding the phenotypic changes of colistin-resistant Acinetobacter baumannii mediated by LPS loss is useful for exploring the resistance mechanism of Acinetobacter baumannii and developing new therapeutic strategies.

脂多糖(lipopolysaccharides, LPS) 是革兰阴性菌细胞膜外膜重要组成成分, 与多数革兰阴性菌不同, 鲍曼不动杆菌在LPS缺失后仍可存活并获得对多黏菌素的耐药性。有关LPS缺失鲍曼不动杆菌的研究多是针对多黏菌素诱导产生的LPS缺失株, 背景复杂且不稳定。为深入研究LPS缺失介导的多黏菌素耐药鲍曼不动杆菌, 本研究通过双质粒CRISPR/Cas9系统敲除鲍曼不动杆菌ATCC 19606的lpxC基因, 构建稳定且背景清晰的LPS缺失株, 并对缺失株的形态、生长速率、抗生素的敏感性、毒力、膜通透性和膜电势进行分析。动物实验经中国医学科学院、北京协和医学院医药生物技术研究所实验动物管理与动物福利委员会批准(批准号: IMB-20240119D9)。结果显示, 鲍曼不动杆菌ATCC 19606的lpxC基因被成功敲除, lpxC缺失后菌株失去外膜LPS, 形态由杆状变为球状, 细菌生长速率变慢, 膜通透性升高, 膜电势降低, 对β-内酰胺类、喹诺酮类、氨基糖苷类、大环内酯类、糖肽类抗生素的敏感性增强, 并且体内毒力大幅下降。lpxC缺失引起鲍曼不动杆菌的膜稳态、适应性发生明显改变, 了解LPS缺失介导的多黏菌素耐药鲍曼不动杆菌的特征变化有利于探究鲍曼不动杆菌耐药机制、寻找治疗新策略。

脂多糖(lipopolysaccharides, LPS) 是革兰阴性菌细胞膜外膜重要组成成分, 与多数革兰阴性菌不同, 鲍曼不动杆菌在LPS缺失后仍可存活并获得对多黏菌素的耐药性。有关LPS缺失鲍曼不动杆菌的研究多是针对多黏菌素诱导产生的LPS缺失株, 背景复杂且不稳定。为深入研究LPS缺失介导的多黏菌素耐药鲍曼不动杆菌, 本研究通过双质粒CRISPR/Cas9系统敲除鲍曼不动杆菌ATCC 19606的lpxC基因, 构建稳定且背景清晰的LPS缺失株, 并对缺失株的形态、生长速率、抗生素的敏感性、毒力、膜通透性和膜电势进行分析。动物实验经中国医学科学院、北京协和医学院医药生物技术研究所实验动物管理与动物福利委员会批准(批准号: IMB-20240119D9)。结果显示, 鲍曼不动杆菌ATCC 19606的lpxC基因被成功敲除, lpxC缺失后菌株失去外膜LPS, 形态由杆状变为球状, 细菌生长速率变慢, 膜通透性升高, 膜电势降低, 对β-内酰胺类、喹诺酮类、氨基糖苷类、大环内酯类、糖肽类抗生素的敏感性增强, 并且体内毒力大幅下降。lpxC缺失引起鲍曼不动杆菌的膜稳态、适应性发生明显改变, 了解LPS缺失介导的多黏菌素耐药鲍曼不动杆菌的特征变化有利于探究鲍曼不动杆菌耐药机制、寻找治疗新策略。

Lipopolysaccharides (LPS) are major outer membrane components of Gram-negative bacteria. Unlike most Gram-negative bacteria, Acinetobacter baumannii can still survive and acquire polymyxin resistance after complete loss of LPS. Previous studies of LPS-deficient Acinetobacter baumannii mostly focused on LPS-deficient strains induced by polymyxin, whose background was complex and unstable. To investigate LPS loss-mediated polymyxin resistant-Acinetobacter baumannii, this study constructed a stable and clear background LPS-deficient strain by knocking out lpxC gene in Acinetobacter baumannii ATCC 19606 with CIRSPR/Cas9, and then studied the phenotypic changes of the lpxC-deficient strain including morphology, growth rate, antibiotic susceptibility, virulence, membrane permeability, and membrane potential. Animal experiments were approved by the Animal Care and Welfare Committee Institute of Medicinal Biotechnology, CAMS and PUMC (approval number: IMB-20240119D9). The results indicated that the lpxC gene of Acinetobacter baumannii ATCC 19606 was successfully knocked out. After losing the lpxC, the strain underwent the morphological change from rod-shaped to spherical. Furthermore, it leads to reduced growth rate, enhanced membrane permeability, decreased membrane potential, lower virulence, and increased antibiotic susceptibility to β-lactams, quinolones, aminoglycosides, macrolides, glycopeptides. The lpxC deletion results in significant changes in membrane homeostasis and adaptability of Acinetobacter baumannii. Understanding the phenotypic changes of colistin-resistant Acinetobacter baumannii mediated by LPS loss is useful for exploring the resistance mechanism of Acinetobacter baumannii and developing new therapeutic strategies.

鲍曼不动杆菌(Acinetobacter baumannii, A. baumannii) 是临床一种常见的革兰阴性条件致病菌, 2021年CHINET统计数据显示鲍曼不动杆菌占据主要临床分离菌株前五位, 常对临床上大多抗生素耐药, 亟需针对鲍曼不动杆菌耐药机制的研究[1]。脂多糖(lipopolysaccharide, LPS) 是革兰阴性菌细菌细胞壁外膜的重要组成成分, 是大多数革兰阴性细菌生存所不可或缺的部分。LPS分子由脂质A、核心多糖、O-抗原三部分组成, LPS生物合成过程中的第一阶段Kdo2-lipidA九种基因(lpxAlpxClpxDlpxHlpxBlpxKKdtAlpxLlpxM) 参与反应, 其中lpxC基因表达的去乙酰化酶(UDP-3-O-acyl-N-acetylglucosamine deacetylase, LpxC) 对脂质A的合成起到关键作用[2]。鲍曼不动杆菌lpxC基因突变引起外膜LPS缺失对多黏菌素类药物高度耐药[3]。CRISPR/Cas9系统是目前最热门的分子生物学技术之一, 能够简洁、高效、无痕地实现基因编辑, 提高对细菌基因功能研究的准确性。
本研究通过CRISPR/Cas9基因编辑技术对鲍曼不动杆菌的lpxC基因全敲除, 研究lpxC基因缺失介导多黏菌素耐药的鲍曼不动杆菌的表型特征, 为进一步探讨细菌耐药机制、发现新型药物靶点提供了基础, 也为临床治疗细菌耐药提供参考材料。

CRISPR/Cas9基因编辑技术敲除lpxC基因  本研究参考季泉江课题组的方法[4], 在此基础上, 重新设计并构建携带lpxC基因sgRNA和同源重组模板的pSGAb-km质粒, CRISPR/Cas9基因编辑原理见图 1。利用CRISPOR网站(http://crispor.tefor.net/) 设计针对lpxC的spacer序列, 选择PAM位点(5'-NGG-3') 的前20 bp设计3对寡链核苷酸spacer-lpxC spacer-F/R序列, 通过Gloden Gate无缝克隆将lpxC spacer-F/R连接到经BsaI酶切过的pSGAb-km质粒中。质粒转化到E. coli DH5α后, 通过50 µg·mL-1卡那霉素抗性平板筛选阳性克隆, 以lpxC spacer-F/M13R为PCR引物, 经菌落PCR和桑格测序验证后获得携带lpxC基因sgRNA的pSGAb-lpxC spacer-km质粒。接着通过限制性内切酶NotI-HF、EcoRI-HF将pSGAb-lpxC spacer-km质粒线性化, 以lpxC基因上下游约500 bp的同源臂lpxC-up和lpxC-down为同源重组模板, 采用Gibson组装的方式设计带有overlap的引物对lpxC-up F/R、lpxC-down F/R, 以细菌基因组为模板通过PCR获得lpxC-up、lpxC-down, 经Gibson组装将上下游同源臂无缝连接到线性化的pSGAb-lpxC spacer-km质粒中, 经转化、卡那霉素抗性筛选、PCR、测序验证后获得携带lpxC基因sgRNA和同源重组模板的pSGAb-lpxC spacer-HR-km质粒。所有的引物序列见表 1。将pCasAb-apr质粒电转到鲍曼不杆菌ATCC 19606中, 以pCasAb-apr上携带的安普霉素抗性基因(ApmR) 为模板设计引物对ApmR-F/R (表 1), 通过PCR验证、测序获得并保存带有pCasAb-apr质粒的菌株, 通过IPTG诱导表达2 h后, 制备成感受态细胞。取400 ng构建好的pSGAb-lpxC spacer-HR-km质粒加到携带pCasAb-apr质粒的鲍曼不杆菌ATCC 19606的感受态细胞中进行电转化。电击完成后, 加入1 mL BHI液体培养基并置于37 ℃摇床100 r·min-1震荡复苏16 h。取100 μL菌液涂布在含有20 μg·mL-1多黏菌素B的BHI固体培养基平板上培养16~20 h筛选阳性克隆。在lpxC基因上下游同源臂设计引物对lpxC-CF/CR (表 1) 进行PCR。产物通过琼脂糖凝胶电泳检测并送样测序, 验证目的基因敲除结果。通过含有5%蔗糖的BHI固体培养平板去除lpxC缺失菌株中基因编辑的质粒, 挑选菌落通过PCR验证质粒去除结果, 以ApmR-F/R引物验证pCasAb-apr质粒, 以pSG-F/R引物验证pSGAb-km质粒, 引物见表 1
LPS的提取与鉴定  在已知质量的15 mL离心管中加入12 mL预冷的PBS (pH 7.2), 刮取平板上的菌苔到PBS中混匀, 控制菌液OD600为0.6左右。4 ℃下重复离心两次(2 500 ×g, 10 min) 获得菌体沉淀, 按沉淀重量的10倍加入裂解液, 利用超声波细胞破碎仪进行破碎(破碎3 s, 间歇3 s, 40%功率, 共10 min)。离心取上清, 加入蛋白酶K (终浓度为0.1 mg·mL-1), 60 ℃孵育60 min后离心, 取带有LPS的上清。将提取的LPS样品进行SDS-PAGE电泳, 140 V电压下运行55 min, 通过考马斯亮蓝染色。
细菌膜电势、膜通透性比较分析  将过夜培养的菌液调整到0.5 MCF左右, 取10 μL稀释后的菌液加入到200 μL Dulbecco's磷酸缓冲液(Dulbecco's phosphate-buffered saline, DBS) 缓冲液中, 加入染料DiBAC4(3)、EB、PI, 终浓度分别为10、10、5 μg·mL-1。将样品在25 ℃下孵育30 min, 然后通过BD Accuri C6 Flow Cytometer流式细胞仪检测, 采用488 nm作为激发光, 检测器分别在 > 630 nm、575 nm的波长测量PI荧光、EB荧光反映膜通透性, 525 nm处测量DiBAC4(3) 荧光反映膜电势。
扫描电镜和透射电镜  取过夜培养的菌液, 离心去上清后用0.2 mol·L-1 PBS重复洗涤离心两次, 再将菌体转移到1.5 mL离心管中, 沿管壁加入1 mL 2.5%戊二醛缓慢加入, 轻轻吹打悬浮1 min后离心成团, 2 500 ×g离心5 min, 加满戊二醛后, 4 ℃冰箱避光过夜(> $ $12 h), 送样品到中科百测公司通过扫描电镜、透射电镜观察菌株外貌形态。
药敏分析  VITEK2-compact全自动细菌药敏分析系统通过革兰阴性细菌药敏卡片(VITEK2 AST-GN13) 检测鲍曼不动杆菌ATCC 19606野生型(wild type)、敲除株(∆lpxC) 的药敏。同时根据CLSI标准, 通过肉汤微稀释法测定菌株对多黏菌素B、黏菌素、卡那霉素、安普霉素、头孢他啶、头孢哌酮、头孢呋辛、美罗培南、氨曲南、舒巴坦、阿奇霉素、阿米卡星、左氧氟沙星、万古霉素、四环素、替加环素、达托霉素的敏感性, 药物浓度范围为0.125~256 μg·mL-1, 接种菌液浓度约5×105 CUF·mL-1, 置于37 ℃培养16~20 h, 肉眼观察无细菌生长的最低药物浓度为最低抑菌浓度。
生长速率测定  取过夜培养至饱和的菌液, 按照1∶1 000稀释, 通过BIOSCREEN全自动生长曲线分析仪, 37 ℃连续震荡培养菌株48 h, 每4 min检测OD600的数值以得到菌株的生长曲线, 使用GraphPad Prism 10对生长曲线数据进行双因素方差分析, 采用Bonferroni事后多重检验, 当P < 0.05时,有统计学意义。
小鼠全身感染[5, 6]  小鼠按体重随机分组, 每组10只, 雌雄各半。实验菌液用5%高活性干酵母稀释成, 每只小鼠腹腔注入约2×106 CFU的菌液(A. baumannii ATCC 19606 1.6×106 CFU、A. baumannii ATCC 19606 ΔlpxC 2.1×106 CFU), 观察并记录7天内各组动物死亡数, 使用Fisher精确检验进行统计分析; 应用Prism软件Kaplan-Meier的乘积极限法创建生存曲线, 并使用对数秩检验(log-rank) 比较生存曲线,当P < 0.05时认为两组具有显著性差异。
已有报道称, 鲍曼不动杆菌ATCC 19606在lpxC基因缺失仍能存活, 为验证lpxC基因是否敲除成功, 本研究进行了PCR电泳分析。电泳结果显示, 野生株PCR产物的长度在1 500~2 000 bp之间, 而敲除株的DNA产物长度在1 000~1 500 bp之间(图 2A)。测序结果进一步证实, 敲除株的产物是约为1 397 bp的DNA序列, 缺失了903 bp的碱基对应lpxC基因(图 2B), 与预期结果一致, 表明lpxC基因敲除成功。由于lpxC基因是LPS合成的关键基因, 为了能更直观地反映出鲍曼不动杆菌lpxC基因与LPS的相关性, 本研究提取了野生株与敲除株的LPS进行SDS-PAGE电泳检测, 结果显示, 野生株在17~20 kDa的位置出现LPS的相应条带, 而敲除株未出现, 进一步证明了基因敲除后鲍曼不动杆菌LPS缺失(图 3)。
为了观察lpxC基因敲除对鲍曼不动杆菌外貌的影响, 本研究分别通过扫描电镜与透射电镜观察鲍曼不动杆菌形貌的变化。在扫描电镜下, 野生型菌株整体分布均匀, 单个细菌外观致密、粗糙, 形状规则两端钝圆, 呈现球杆状, 无鞭毛结构, 细菌之间界限清晰, 无黏连发生(图 4A~C)。相比之下, 敲除株的单个细菌整体外观呈现球形或椭圆形, 表面附着一些球形或长条形分泌物, 分泌物呈现网络状, 整体上细菌之间以类葡萄簇状排列, 并且部分菌之间有粘连发生, 菌与菌的界限较为模糊(图 4D~F)。
在透射电镜下, 本研究对细菌外膜结构上的变化有了更清晰的发现, 野生株单个细菌切面形状规则, 细胞壁颜色较深, 菌与菌之间分界清晰(图 4G~I)。在敲除株中, 细菌很少呈单个存在, 分泌物由外膜延伸而出, 胞质分布不均匀, 增殖过程不在中间位置内陷分裂, 不呈现典型的均匀二分裂状态, 在成簇的细胞中, 细菌之间的界限变得透明, 夹在中间的细菌形状不规则(图 4J~L)。
本研究通过流式细胞术对100 000个经过染色细菌进行检测, 分别在525、575和 > 630 nm波长下检测DiBAC4(3)、EB和PI染料的荧光强度, 以评估细菌的膜通透性和膜电势。DiBAC4(3)是一种亲脂性阴离子荧光染料, 在细胞膜电势较高的情况下, DiBAC4(3)不能轻易穿过完整的细胞膜, 因此荧光强度较低。但当膜电势降低时会增加染料的亲和力, 从而导致荧光值的增加。EB、PI是两种核酸染料, 在细胞膜完整的情况下会阻止这些染料的进入, 当膜受损、通透性增加时, EB和PI能够进入到细胞内与核酸结合发出荧光。流式细胞术的结果显示, 敲除株在各个波长下显示出比野生株更高的荧光强度, 这表明鲍曼不动杆菌lpxC基因敲除后, 膜电势降低(图 5A), 膜通透性增加(图 5BC)。
VITEK2-compact全自动细菌药敏分析显示, 在lpxC基因敲除后, 除甲氧苄啶/磺胺甲恶唑外, 鲍曼不动杆菌对各类抗生素的敏感性均有所提高, 包括氨基糖苷类、第三代头孢类和喹诺酮类(表 2)。
肉汤微稀释法的结果显示, 敲除lpxC基因后的鲍曼不动杆菌表现出对多黏菌素的高度耐药性, 但对β-内酰胺类(头孢他啶、头孢哌酮、头孢呋辛、舒巴坦、氨曲南、美罗培南)、喹诺酮类(左氧氟沙星)、氨基糖苷类(阿米卡星)、大环内酯类(阿奇霉素)、肽类(万古霉素、达托霉素)、四环素类(四环素、替加环素) 多种抗生素更加敏感(表 3)。
生长曲线的分析结果表明, lpxC基因敲除株的生长速率明显低于野生型。野生型菌株在大约12 h就达到了平台期, 而与之相比, lpxC敲除株约晚12 h才进入平台期(图 6A)。这说明lpxC基因的敲除严重影响鲍曼不动杆菌的生长动力学, 导致生长过程的延迟。
小鼠腹腔感染大约2×106 CFU的菌液后, 连续观察7天。结果显示, A. baumannii ATCC 19606 (wild type) 组小鼠死亡率为90%, A. baumannii ATCC 19606基因敲除(∆lpxC) 组小鼠死亡率为0% (图 6B)。应用Fisher精确检验进行统计分析比较显示, 两组小鼠死亡率具有极显著性差异(P < 0.01)。应用Prism软件Kaplan-Meier的乘积极限法创建生存曲线, 并使用对数秩检验(log-rank) 比较显示, 两组小鼠的生存曲线具有极显著性差异(P < 0.01)。
鲍曼不动杆菌作为临床上最常见的致病菌之一, 它的耐药率、感染率、分离率常年占据高位[1]。由于该菌能够长时间存活于物体表面, 因此在重症监护病房常引发呼吸机相关性肺炎, 而在治疗过程中, 它对常用的抗菌药物极易发生耐药[7]。自2004年起, 陆续在澳大利亚、马来西亚的临床中分离出对多黏菌素类药物高度耐药的鲍曼不动杆菌[3, 8]。这个现象为研究者敲响警钟, 迫切需要深入研究鲍曼不动杆菌的耐药机制。
LPS缺失是鲍曼不动杆菌对多黏菌素耐药的机制之一。Moffatt等[3]最早发现LPS缺失的鲍曼不动杆菌, 并首次报道了临床分离得到的LPS缺失株B0707-070。后续经临床分离得到的LPS缺失株鲜有报道, 目前LPS缺失鲍曼不动杆菌的研究多是通过多黏菌素体外诱导而来。考虑到诱导突变可能引发其他未知基因的突变[9], 背景复杂且不稳定, 本研究通过CRISPR/Cas9技术成功构建了lpxC基因全敲除菌。不同于鲍曼不动杆菌一般基因的敲除, lpxC基因的敲除会改变抗生素的敏感性, 导致CRISPR/Cas9系统质粒自带的抗性不能作为有效的筛选条件。考虑到鲍曼不动杆菌在LPS缺失后获得对多黏菌素的高度耐药性, 本研究最终采用多黏菌素B作为筛选条件, 成功获得lpxC敲除株[10]
革兰阴性菌细胞膜外膜的磷脂(phospholipids, PL) 和LPS成分维持着动态平衡, 细菌可以通过调控LPS的动态变化适应不同的环境并提高对药物的耐受性, 而一旦平衡被破坏将会对细菌产生致命威胁[11, 12]。鲍曼不动杆菌是目前发现失去LPS后仍能存活的3种细菌之一, 鲍曼不动杆菌失去LPS后仍存在外膜, 转录组学结果显示LPS的缺失造成磷脂转运体系mlaBCD和脂蛋白转运体系lolBDE基因的转录水平显著上调, 说明脂质在LPS缺失的菌株中扮演了关键调控作用[13]。有文献报道, 细菌外膜LPS缺失会导致内侧的PL翻转到外层, PL弥补了外膜LPS, 但同时破坏细菌细胞壁的刚性结构和渗透性[14], 造成细胞膜通透性增加, 疏水性物质更易进入细胞, 疏水性物质的增加以及带有负电荷LPS的减少进一步造成膜电势降低[15], 这与本研究的结果一致。从电镜结果可以看出失去LPS的鲍曼不动杆菌失去了原有的杆状形态, 变成了球状。细菌形态的改变说明细菌的细胞骨架成分发生了明显的改变, 有文献报道长链肽聚糖的合成对LPS缺失鲍曼不动杆菌的生存至关重要, 当青霉素结合蛋白1A (penicillin binding protein 1A, PBP1A) 蛋白高表达时, 鲍曼不动杆菌在失去LPS后无法存活, 因为PBP1A可能对长链肽聚糖的合成具有抑制作用[16]。在LPS缺失鲍曼不动杆菌中, 聚β-1, 6-N-乙酰葡萄糖胺(poly-β-1, 6-N-acetylglucosamin, PNAG) 的合成与转运相关的pgaABCD基因转录水平明显增加[17], 说明细胞壁的肽聚糖的合成途径可能在维持LPS缺失鲍曼不动杆菌细胞膜稳定性中发挥着关键作用。至于细胞骨架成分究竟发生了怎样的变化还需进一步的研究。以上结果反映出LPS在维持细菌形态、细菌渗透压、膜电势起至关重要作用[16]lpxC基因全敲除后, 鲍曼不动杆菌对多黏菌素的敏感性明显下降。LPS是多黏菌素的关键作用靶点, 目前多黏菌素的作用机制仍然不明确, 研究者认可度比较高的一种模型是多黏菌素二氨基丁酸残基携带的正电荷与脂质A磷酸基团携带的负电荷产生静电作用, 导致LPS之间的二价阳离子被取代, 多黏菌素的疏水性结构进一步破坏细胞外膜的稳定性[18, 19]。当鲍曼不动杆菌失去LPS后, 外膜负电荷减少, 多黏菌素失去作用靶点LPS, 导致多黏菌素敏感性下降。除多黏菌素外, 鲍曼不动杆菌对大多数的抗生素敏感性是上升的, 这可能与lpxC基因失活后, 菌株的膜通透性增加有关[20]
LPS是革兰阴性菌的标志, 也是鲍曼不动杆菌毒力的主要来源之一。鲍曼不动杆菌可以通过修饰LPS的结构从而获得对多种抗生素的耐药性、逃避宿主免疫反应, 在宿主体内大量繁殖造成败血症甚至感染性休克[21-23]。与野生株相比, LPS缺失株在感染宿主中的生长速率、毒力显著下降的同时[10, 24], 更难产生生物膜、表面运动减弱、对多种消毒剂的敏感性增加限制了临床环境中的传播, 这可能是临床上鲜有报道LPS缺失株感染的原因[25]
此外, 针对lpxC为抗菌靶点是目前抗生素研究的热门方向, 尤其是LpxC抑制剂作为针对革兰阴性细菌的潜在治疗分子已被广泛研究[26]。LpxC抑制剂与铁螯合剂或硝酸镓的组合对鲍曼不动杆菌表现出协同活性, 与单独使用的化合物相比, 细菌数量显著减少[27]。LpxC抑制剂单独使用虽然不会对鲍曼不动杆菌产生很高的抗菌活性, 但是它会增加细菌对其他抗生素的敏感性[20]。因此, 采用LpxC抑制剂联用已有抗生素治疗鲍曼不动杆菌引起的感染不仅可以提高疗效, 还能降低耐药发生率, 为未来的治疗策略开辟新的方向。
总之, 本研究通过CRISPR/Cas9系统成功构建了鲍曼不动杆菌ATCC 19606的lpxC全基因敲除株, 比较了表型上的变化, 为后续研究鲍曼不动杆菌独特的LPS缺失的膜结构、LPS缺失后菌株膜组分、相关代偿性通路变化, 以及发现潜在药物靶点提供了物质条件。
