The column chromatography and semi-preparative liquid phase chromatography with several chromatographic packing materials, including macroporous adsorbent resin, silica gel, ODS, and Sephadex LH-20, were used for the separation and purification of n-butanol-soluble portion of ethanol extract of Leonurus japonicus Houtt. The structures of isolates were identified by HR-MS, IR, NMR, and acid hydrolysis reaction. Six phenylethanol glycosides were obtained from L. japonicus and identified as leonoside G (1), leonoside E (2), leonoside B (3), leonoside F (4), cistanoside G (5), and salidroside (6). Compound 1 is a new phenylethanol glycoside.

采用大孔吸附树脂、硅胶、ODS、葡聚糖凝胶等色谱填料, 运用柱色谱、半制备液相色谱等分离技术, 对益母草(Leonurus japonicus Houtt.) 乙醇提取物的正丁醇部位进行分离纯化, 利用高分辨质谱、红外、核磁共振等波谱技术和酸水解反应鉴定化合物的结构。从益母草中分离得到6个苯乙醇苷类化合物, 分别为leonoside G (1)、leonoside E (2)、leonoside B (3)、leonoside F (4)、cistanoside G (5)、红景天苷(6), 其中化合物1为从益母草中获得的新的苯乙醇苷类化合物。

The column chromatography and semi-preparative liquid phase chromatography with several chromatographic packing materials, including macroporous adsorbent resin, silica gel, ODS, and Sephadex LH-20, were used for the separation and purification of n-butanol-soluble portion of ethanol extract of Leonurus japonicus Houtt. The structures of isolates were identified by HR-MS, IR, NMR, and acid hydrolysis reaction. Six phenylethanol glycosides were obtained from L. japonicus and identified as leonoside G (1), leonoside E (2), leonoside B (3), leonoside F (4), cistanoside G (5), and salidroside (6). Compound 1 is a new phenylethanol glycoside.

益母草为唇形科益母草属植物益母草(Leonurus japonicus Houtt.) 的新鲜或干燥地上部分, 全国各地均有分布[1]。益母草始载于《神农本草经》, 以全草入药, 具有活血调经、利水消肿、清热解毒的传统功效。由于其在妇产科疾病中的疗效显著, 被誉为“经产良药”和“胎产圣药”, 现代药理研究发现益母草具有调节子宫平滑肌[2, 3]、促血管生成[4]、抗血小板聚集[5]、舒张血管[6]、凝血[7]、细胞毒[8]、抗菌[9]和肝保护[10]等药理活性。植化研究发现益母草主要有生物碱[2, 3, 11]、二萜[6, 12]、三萜[13]、黄酮[14]和苯乙醇苷[15]等类型化合物。其中, 化学成分研究多集中于生物碱类和二萜类成分[2], 而香豆素、苯乙醇苷等报道相对较少。苯乙醇苷类成分由于苷元和单糖的不同导致该类化合物的结构变化较为丰富, 至今苯乙醇苷类化合物已报道570余个[16]。现代药理研究发现, 苯乙醇苷类化合物具有肝保护[17]、神经保护[18]、抗炎[19]、抗肿瘤[20]、心肌保护[19]等药理活性。本课题组前期从益母草中发现了系列生物碱、二萜和黄酮类化合物[3, 6], 为进一步丰富益母草中的化学成分, 本研究采用多种分离技术得到6个苯乙醇苷类化合物, 运用高分辨质谱、红外、核磁共振等波谱技术和酸水解反应鉴定化合物的结构, 其中化合物1为1个新的苯乙醇苷类化合物。
化合物1: 白色无定形粉末, 易溶于甲醇。比旋度[α]$ {}_{\mathrm{D}}^{20} $ +50.0 (c 0.02, MeOH)。根据准分子离子峰(+)-HR-ESI-MS m/z: 615.189 7 [M+Na]+ (计算值为C25H36O16Na, 615.190 1), 确定化合物1的分子式为C25H36O16, 不饱和度为8。UV (MeOH) λmax (log ε) 224 (3.35), 281 (3.45) nm; IR数据提示含有-OH (3 434 cm-1)、-CH- (2 921, 2 853 cm-1)、-CO- (1 129, 1 059 cm-1), 在1 628 cm-1处的伸缩振动提示有苯环存在。
13C NMR谱(表 1) 和DEPT谱显示化合物1 (图 1) 共有25个碳信号, 可归属为1个甲基(δC 17.9)、3个连氧亚甲基(δC 62.4, 66.9, 72.4)、18个次甲基[含15个连氧次甲基(δC 69.7, 69.8, 70.0, 71.8, 72.8, 74.1, 74.3, 78.7, 79.8, 80.0, 82.1, 83.2, 99.1, 100.6, 107.4) 和3个芳环次甲基(δC 115.4, 116.4, 119.8)]、3个芳香季碳[含2个芳香连氧季碳(δC 146.4, 146.7)] 信号。1H NMR谱(表 1) 显示, 低场有3个芳环质子信号[δH 6.85 (1H, d, J = 1.8 Hz), 6.76 (1H, d, J = 7.8 Hz) 和6.72 (1H, dd, J = 7.8, 1.8 Hz)], 是一个典型的ABX系统, 推测化合物1含有一个1, 3, 4-三取代苯环。根据edit-HSQC谱对化合物1的氢碳信号进行了准确归属。在13C NMR谱中的3个连氧次甲基碳信号(δC 99.1, 100.6和107.4) 以及在1H NMR谱中2个明显的连氧次甲基质子信号[δH 5.37 (1H, d, J = 1.8 Hz) 和4.46 (1H, d, J = 7.8 Hz)], 推测化合物1含有三个单糖。结合1H NMR谱中一组连氧亚甲基信号[δH 3.89 (1H, dd, J = 12.0, 1.8 Hz) 和3.72 (1H, overlapped), δC 62.4] 推断其中1个单糖为葡萄糖; 结合[δH 1.21 (3H, d, J = 6.0 Hz), δC 17.9] 的双峰甲基信号和[δH 5.37 (1H, d, J = 1.8 Hz), δC 100.6] 的端基质子信号推断另一个单糖为鼠李糖; 综合上述已确定片段和剩下的NMR数据, 推测该化合物还含有1个阿拉伯糖。
进一步采用2D NMR对化合物1的结构进行了鉴定。在HMBC谱中, H-2与C-4、C-6相关, H-5与C-1、C-3相关, H2-8与C-1相关, 以及H-7与C-1、C-2、C-6、C-8相关, 表明化合物1的苷元部分为1, 3, 4-三取代苯乙醇结构。质谱显示化合物1的不饱和度为8, 除去苯环和3个单糖共7个不饱和度以外, 还剩1个不饱和度, 推测苷元与糖基部分的羟基相连而成环, 而HMBC谱显示H-7与C-2′相关, 同时与文献报道的taraffinisoside A[21]和cuneataside A[22]的波谱数据十分类似, 表明葡萄糖的2′-OH与苷元上的C-7成环。在HMBC谱中, H2-8与C-1′相关, 表明葡萄糖端基质子与苯乙醇的C-8位羟基相连形成苷键。同时H-1″与C-3′有HMBC相关, 表明鼠李糖连接在葡萄糖的C-3′上; 此外, 阿拉伯糖的端基质子H-1‴与C-2″有HMBC相关, 表明阿拉伯糖连接在鼠李糖的C-2″位。
葡萄糖的相对构型根据1H NMR中其端基质子的JH1′-H2′ = 7.8 Hz, 可以确定为β-葡萄糖; 阿拉伯糖的相对构型通过其端基质子的JH1‴-H2‴ = 7.2 Hz, 确定为α-阿拉伯糖[22, 23]。鼠李糖的H-1″为e键, 其相对构型无法通过偶合常数确定, 根据鼠李糖的端基碳化学位移[24], 以及利用HETLOC实验[25]测得JH1″-C1″ = 175 Hz[26], 从而确定为α-鼠李糖。另外, 在NOESY光谱中可以看到H-7与H-2′有相关, 表明H-7为β构型, 且偶合常数(JH7-H8b = 10.8 Hz) 也验证了该结果。最后, 采用酸水解反应鉴定化合物1中的单糖分别为D-葡萄糖、L-鼠李糖和L-阿拉伯糖, 其糖链为α-L-阿拉伯糖基(1→2)-α-L-鼠李糖基(1→3)-β-D-葡萄糖基, 化合物1的结构最终确定如图 1所示, 命名为leonoside G。
通过波谱数据与文献进行对照, 鉴定了5个已知化合物, 分别为leonoside E (2)[15]、leonoside B (3)[27]、leonoside F (4)[15]、cistanoside G (5)[28]和红景天苷(6)[29]
Rudolph Autopol Ⅰ自动旋光仪, 美国Rudolph公司; Bruker Avance NEO 600 M核磁共振波谱仪和Bruker timsTOF质谱仪, 德国Bruker公司; Applied Photophysics Chirascan圆二色光谱仪, 英国Applied Photophysics公司; Spectrum One FT-IR spectrometer光谱仪, 美国PerkinElmer公司; Agilent 1220高效液相色谱仪和Varian Dynamax色谱柱(250 mm × 21.4 mm, 8 μm), 美国Agilent公司; Welch Ultimate XB-C18色谱柱(250 mm × 10 mm, 5 μm), 月旭科技(上海) 股份有限公司; Kromasil C18色谱柱(250 mm × 4.6 mm, 5 μm), 瑞典Akzo Nobel公司; 旋转蒸发仪, 上海贤德实验仪器有限公司; SQP型分析天平, 赛多利斯科学仪器(北京) 有限公司。柱层析硅胶(200~300目), 青岛海浪硅胶干燥剂有限公司; 高效薄层色谱硅胶板(GF254硅胶), 安徽良臣硅源材料有限公司; Sephadex LH-20凝胶, 瑞典Amershan Pharmaci公司; 甲醇(色谱级), 美国Sigma公司; 其他所用试剂均为分析纯(成都市科隆化学品有限公司)。L-阿拉伯糖(批号: WP23012907) 和L-鼠李糖标准品(批号: WP23080409), 四川省维克奇生物科技有限公司; D-葡萄糖标准品(批号: MUST-19032905), 成都曼思特生物科技有限公司。
样品于2012年5月采自四川省冕宁县, 由成都中医药大学药学院李敏教授鉴定为益母草(L. japonicus), 标本现存于成都中医药大学西南特色药材创新药物成分研究所, 标本号为(LJ20130522)。
取干燥粉碎的益母草20 kg, 用8倍量的95%乙醇回流提取3次, 每次2 h。提取液经减压浓缩得浸膏1.2 kg, 将浸膏混悬于水中, 依次用乙酸乙酯和正丁醇萃取, 回收溶剂, 得正丁醇部位180 g。正丁醇部位经大孔树脂分离, 以不同浓度乙醇(10%、30%、50%、70%、95%) 进行梯度洗脱, 回收溶剂得到5个洗脱部分(A~E), A (30 g) 经反相C18减压柱分离, 以甲醇-水溶液(8%~100%) 洗脱得到5个亚组分(A1~A5)。A1采用Sephadex LH-20凝胶柱(60%甲醇-水溶液), 得到8个亚组分(A1-1~A1-8), A1-4经过VARIAN Dynamax制备柱得到8个亚组分(A1-4-1~A1-4-8), A1-4-4采用硅胶柱色谱(二氯甲烷-甲醇溶液, 50∶1~3∶1)和半制备高效液相色谱(40%甲醇-水溶液) 分离得到化合物2 (5.3 mg, tR = 30.8 min) 和3 (3.0 mg, tR = 40.3 min)。A1-4-6经半制备高效液相色谱(50%甲醇-水溶液) 分离得到化合物4 (3.3 mg, tR = 20.8 min) 和6 (2.0 mg, tR = 30.5 min)。A2采用Sephadex LH-20凝胶柱(50%甲醇-水溶液) 分离得到8个亚组分(A2-1~A2-8), A2-3用Sephadex LH-20凝胶柱(40%甲醇-水溶液), 得到6个亚组分(A2-3-1~A2-3-6), A2-3-5经半制备高效液相色谱(35%甲醇-水溶液) 纯化得到化合物1 (12.0 mg, tR = 45.3 min), A2-3-6经半制备高效液相色谱(40%甲醇-水溶液) 纯化得到化合物5 (2.2 mg, tR = 25.5 min)。
化合物1 (2.0 mg) 用0.5 mol·L-1盐酸溶液在90 ℃下水解2 h, 将水解液减压干燥, 残渣用4 mL水溶解, 用3倍量乙酸乙酯萃取5次, 收集水层[30]。将水层干燥后用500 μL纯水溶解, 吸取100 μL与1-苯基-3-甲基-5-吡唑酮(PMP) 反应, 同时精密吸取D-葡萄糖、L-鼠李糖和L-阿拉伯糖标准品制成的混标溶液100 μL与PMP反应[31]。采用Kromasil C18色谱柱(250 mm × 4.6 mm, 5 μm) 进行高效液相(HPLC) 分析, 流动相为: A相磷酸盐缓冲液(0.05 mol·L-1, pH 6.7), B相乙腈; 洗脱梯度为: 0~17 min, VA/VB = 82∶18; 17~20 min, VA/VB = 72∶28; 20~45 min, VA/VB = 72∶28; 柱温30 ℃, 流速1.0 mL·min-1, 检测波长为254 nm。通过比较单糖标准品衍生物的色谱峰和样品水解液中单糖衍生物的色谱峰的保留时间, 确定化合物1中的单糖分别为D-葡萄糖、L-鼠李糖和L-阿拉伯糖。
