In this study, the pharmacodynamic substance basis of the therapeutic activity of different origin sources of the Tibetan medicinal herb Zha xun was evaluated, and the protective effect of the Zha xun, from Habahe county of Altay region, Xinjiang Uygur Autonomous Region; Gilgit region, Pakistan; Lhozhag county of Lhozhag city, Tibet Autonomous Region; Lhorong county of Chamdo city, Tibet Autonomous Region; and Jiulong county of Ganzi Tibetan Autonomous Prefecture, Sichuan Province, on 0.2% carbon tetrachloride (CCl4)-induced acute liver injury in ICR mice was evaluated. The results showed that different sources of Zha xun significantly reduced serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) in the CCl4-induced acute oxidative liver injury model, improved liver histopathological damage. Among them, Zha xun from Habahe County, Altay Region, Xinjiang Uygur Autonomous Region; Gilgit Region, Pakistan; and Lhorong County, Chamdo City, Tibet Autonomous Region significantly reduced the malondialdehyde (MDA) content in liver tissues (P < 0.05), increased the glutathione (GSH) content (P < 0.01), improved the oxidative stress in liver tissues. The preliminary evaluation of the hepatoprotective activity of Zha xun from different origins was carried out using the model of HepG2 cell injury induced by acetaminophen (APAP). The results showed that Zha xun from different origins could significantly inhibit APAP-induced hepatocellular injury and improve the survival rate of hepatocytes. The present study demonstrated that the different origins of Zha xun had definite hepatoprotective activities in vivo and ex vivo, among which, Zha xun from Lhorong County, Chamdo City, Tibet Autonomous Region, had stronger hepatoprotective activities. The animals used in this experiment and the related dispositions were in accordance with the requirements of animal welfare, and the experiment was approved by the Committee of Laboratory Animal Management and Use of the Institute of Pharmaceutical Sciences of the Chinese Academy of Medical Sciences (approval No. 00004024) before the experiment was carried out.

对来自不同产地的藏药材渣驯进行肝病治疗活性比较研究, 在本研究中, 首先评价了产自新疆维吾尔自治区阿勒泰地区哈巴河县、巴基斯坦吉尔吉特地区、西藏自治区山南市洛扎县、西藏自治区昌都市洛隆县、四川省甘孜藏族自治州九龙县等5个地区的藏药材渣驯对0.2%四氯化碳(CCl4) 诱导ICR小鼠急性肝损伤的保护作用。结果显示, 5个不同产地来源的藏药材渣驯在CCl4诱导的小鼠急性氧化性肝损伤模型中, 均可显著降低肝损伤小鼠血清谷丙转氨酶(alanine aminotransferase, ALT)、谷草转氨酶(aspartate aminotransferase, AST)、乳酸脱氢酶(lactate dehydrogenase, LDH) 水平; 改善肝组织病理损伤; 其中, 新疆维吾尔自治区阿勒泰地区哈巴河县、巴基斯坦吉尔吉特地区、西藏自治区昌都市洛隆县三地来源的渣驯可显著降低肝组织丙二醛(malondialdehyde, MDA) 含量(P < 0.05), 升高谷胱甘肽(glutathione, GSH) 含量(P < 0.01), 改善肝组织氧化应激。进而利用对乙酰氨基酚(acetaminophen, APAP) 诱导的HepG2细胞损伤模型, 评价了产自不同产地渣驯药材的体外保肝活性, 结果显示, 不同产地来源的藏药材渣驯均能显著抑制APAP诱导的肝细胞损伤, 提高肝细胞存活率。本研究表明, 不同产地来源的渣驯均具有明确的体内外肝保护活性, 其中, 来源于西藏自治区昌都市洛隆县的渣驯药材肝保护活性相对略优。本实验所用动物及相关处置符合动物福利的要求, 实验开展前经过中国医学科学院药物研究所实验动物管理和使用委员会的审查批准(批准号: 00004024)。

对来自不同产地的藏药材渣驯进行肝病治疗活性比较研究, 在本研究中, 首先评价了产自新疆维吾尔自治区阿勒泰地区哈巴河县、巴基斯坦吉尔吉特地区、西藏自治区山南市洛扎县、西藏自治区昌都市洛隆县、四川省甘孜藏族自治州九龙县等5个地区的藏药材渣驯对0.2%四氯化碳(CCl4) 诱导ICR小鼠急性肝损伤的保护作用。结果显示, 5个不同产地来源的藏药材渣驯在CCl4诱导的小鼠急性氧化性肝损伤模型中, 均可显著降低肝损伤小鼠血清谷丙转氨酶(alanine aminotransferase, ALT)、谷草转氨酶(aspartate aminotransferase, AST)、乳酸脱氢酶(lactate dehydrogenase, LDH) 水平; 改善肝组织病理损伤; 其中, 新疆维吾尔自治区阿勒泰地区哈巴河县、巴基斯坦吉尔吉特地区、西藏自治区昌都市洛隆县三地来源的渣驯可显著降低肝组织丙二醛(malondialdehyde, MDA) 含量(P < 0.05), 升高谷胱甘肽(glutathione, GSH) 含量(P < 0.01), 改善肝组织氧化应激。进而利用对乙酰氨基酚(acetaminophen, APAP) 诱导的HepG2细胞损伤模型, 评价了产自不同产地渣驯药材的体外保肝活性, 结果显示, 不同产地来源的藏药材渣驯均能显著抑制APAP诱导的肝细胞损伤, 提高肝细胞存活率。本研究表明, 不同产地来源的渣驯均具有明确的体内外肝保护活性, 其中, 来源于西藏自治区昌都市洛隆县的渣驯药材肝保护活性相对略优。本实验所用动物及相关处置符合动物福利的要求, 实验开展前经过中国医学科学院药物研究所实验动物管理和使用委员会的审查批准(批准号: 00004024)。

In this study, the pharmacodynamic substance basis of the therapeutic activity of different origin sources of the Tibetan medicinal herb Zha xun was evaluated, and the protective effect of the Zha xun, from Habahe county of Altay region, Xinjiang Uygur Autonomous Region; Gilgit region, Pakistan; Lhozhag county of Lhozhag city, Tibet Autonomous Region; Lhorong county of Chamdo city, Tibet Autonomous Region; and Jiulong county of Ganzi Tibetan Autonomous Prefecture, Sichuan Province, on 0.2% carbon tetrachloride (CCl4)-induced acute liver injury in ICR mice was evaluated. The results showed that different sources of Zha xun significantly reduced serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) in the CCl4-induced acute oxidative liver injury model, improved liver histopathological damage. Among them, Zha xun from Habahe County, Altay Region, Xinjiang Uygur Autonomous Region; Gilgit Region, Pakistan; and Lhorong County, Chamdo City, Tibet Autonomous Region significantly reduced the malondialdehyde (MDA) content in liver tissues (P < 0.05), increased the glutathione (GSH) content (P < 0.01), improved the oxidative stress in liver tissues. The preliminary evaluation of the hepatoprotective activity of Zha xun from different origins was carried out using the model of HepG2 cell injury induced by acetaminophen (APAP). The results showed that Zha xun from different origins could significantly inhibit APAP-induced hepatocellular injury and improve the survival rate of hepatocytes. The present study demonstrated that the different origins of Zha xun had definite hepatoprotective activities in vivo and ex vivo, among which, Zha xun from Lhorong County, Chamdo City, Tibet Autonomous Region, had stronger hepatoprotective activities. The animals used in this experiment and the related dispositions were in accordance with the requirements of animal welfare, and the experiment was approved by the Committee of Laboratory Animal Management and Use of the Institute of Pharmaceutical Sciences of the Chinese Academy of Medical Sciences (approval No. 00004024) before the experiment was carried out.

藏药渣驯(Zha xun), 在藏语中“渣”通常指为“岩石”, 而“驯”泛指除去杂质的精华液。故渣驯意为岩石被挤压而渗流的精华液, 简称“渣驯”。其最早记载于藏医学典籍《月王药珍》中, 称“渣驯可干枯脓血, 主治肝病, 清诸热”。在藏医学《四部医典》[1]中通常将渣驯归类为土类或精华类药物, 其作用非常广泛, 具有匀“三因” (隆、赤巴、培根) 除胃肝肾热及延年益寿的药效作用。作为传统藏药, 渣驯应用已有3 500多年历史, 在藏药成方制剂中使用频率排名第六, 具有极高的医用价值[2]。同时, 渣驯分布地广泛[3], 除文献报道的四川省阿坝藏族羌族自治州、甘孜藏族自治州、凉山彝族自治州、西藏波密、青海省等地及世界其他地区(喜马拉雅山南麓、帕米尔高原、阿尔泰山脉、乌拉尔山脉、兴都库什山脉、不丹、埃及、蒙古国、尼泊尔、印度、挪威、巴基斯坦、俄罗斯等) 外, 项目组在近些年的实地资源考证中发现西藏的洛扎县、朗县、米林市、定日县、浪卡子县等区域蕴藏丰富的渣驯药材资源。渣驯药材的自然形成机制说法很多, 在欧洲渣驯被认为是天然的沥青, 是形成于沼泽等区域的浮状油性物, 具有延年益寿的神奇疗效; 而在藏医学中则认为渣驯是在多年的自然界变化中通过太阳和地层中的热性能量高温作用后从岩层缝隙地融挤出露的具五源精华的万能之药[4]。渣驯被誉为: 皆能治愈世上病, 尤其消肝热益寿[1]
事实上, 渣驯的实际形成过程及机制等还需进一步的考证。渣驯在自然界中形态成不规则块状, 表面有粗糙和平滑之分, 颜色根据内含物质的不同则多呈棕褐色、黑褐色、黄棕色、白色, 其质地坚硬, 具有油性光泽, 气味腥臭, 味辛苦[3]。由于渣驯分布产地广泛, 形成机制复杂, 其药材均一性较差, 目前国内多以其中色黑、坚硬、质重, 粪粒和石块少者质量为佳, 但仅凭外观难以判断其药材杂质含量比例, 加工为渣驯膏后均为黑色膏状物, 亦无外观判断指标, 其保肝药效更是难以评判, 同时渣驯作为能够均衡体内物质平衡的万能药物, 不同产地渣驯活性的异同, 其有效物质基础以及因渣驯资源匮乏而发掘替代药物是目前急需解决的研究课题。
本研究旨在对采自不同产地的藏药材渣驯药材进行保肝活性比较分析, 为临床渣驯药材的使用及渣驯药物的进一步开发提供依据。研究采用了经典的四氯化碳(CCl4) 诱导的小鼠急性氧化性肝损伤模型和对乙酰氨基酚(acetaminophen, APAP) 诱导的HepG2细胞损伤模型, 结果显示源自不同产地的渣驯均表现良好的体内外肝保护活性, 活性特点略有不同, 来自西藏自治区昌都市洛隆县产地的渣驯药材肝保护活性相对略优。
药物及主要试剂  渣驯药材来源于新疆阿勒泰地区哈巴河县(代号ZX-SI)、巴基斯坦吉尔吉特地区(代号ZX-G)、西藏山南市洛扎县(代号ZX-SNLZ)、西藏昌都市洛隆县(代号ZX-CDLL)、四川甘孜藏族自治州九龙县(代号ZX-SC), 棕色至棕黑色粉末, 详细来源信息见表 1。CCl4 (20190516) 购自北京化工厂; APAP (099K0127V) 购自西格玛-奥德里奇(上海) 贸易有限公司; 双环醇(20180428) 由北京协和药厂提供; 橄榄油(C12735529) 购自上海麦克林生化科技股份有限公司; 谷丙转氨酶(alanine aminotransferase, ALT) (210931)、谷草转氨酶(aspartate aminotransferase, AST) (210907)、乳酸脱氢酶(lactate dehydrogenase, LDH) (220921) 检测试剂盒购自中生北控生物科技股份有限公司; 谷胱甘肽(glutathione, GSH) 检测试剂盒(061923230718)、丙二醛(malondialdehyde, MDA) 检测试剂(081722230322) 购自碧云天生物技术有限公司。
实验动物及饲养条件  无特定病原体级(specific pathogen free, SPF) 雄性ICR小鼠(20~22 g), 由北京华阜康生物科技股份有限公司提供, 动物许可证号: SCKX (京) 2019-0008。动物饲养于中国医学科学院药物研究所GLP动物实验中心。室温22~24 ℃, 相对湿度40%~60%。最小换气次数为每小时20次, 间隔12 h开灯光照。动物饲养于聚丙烯小鼠群养笼中, 每笼4~5只。所有动物均由培训合格的人员进行饲养管理, 整个饲养过程中保持动物饮食和活动自由。本实验所用动物及相关处置符合动物福利的要求, 实验开展前经过中国医学科学院药物研究所实验动物管理和使用委员会(IACUC) 的审查批准(批准号: 00004024)。
0.2% CCl4模型: 动物适应环境后根据体重随机分为13组, 分别为空白对照组、0.2% CCl4模型组、阳性对照组双环醇(bicyclol) 200 mg·kg-1组、ZX-SI 50 mg·kg-1组; ZX-SI 200 mg·kg-1组; ZX-G 50 mg·kg-1组; ZX-G 200 mg·kg-1组; ZX-SNLZ 50 mg·kg-1组; ZX-SNLZ 200 mg·kg-1组; ZX-CDLL 50 mg·kg-1组; ZX-CDLL 200 mg·kg-1组; ZX-SC 50 mg·kg-1组; ZX-SC 200 mg·kg-1组, 每组8~9只。化合物分别于造模前5天灌胃给药, 每天1次, 双环醇200 mg·kg-1组于造模前一天的下午、当天的上、下午各灌胃给药1次, 空白对照组及模型组动物给予同等剂量的生理盐水。末次给药2 h后, 除空白对照组, 各组小鼠腹腔注射0.2% CCl4 10 mL·kg-1 (花生油配制) 一次, 空白对照组腹腔注射同等剂量花生油。小鼠禁食不禁水16 h后, 收集动物血清。
细胞系  人肝癌HepG2细胞为本实验室传代保存, 用含100 u·mL-1青霉素(penicillin)、100 µg·mL-1链霉素(streptomycin) 及10%灭活新生牛血清的DMEM高糖培养基, 在37 ℃、含5% CO2细胞培养箱中培养。细胞约每隔2~3天传代。取对数生长期的细胞开始实验。
渣驯药物提取方式  渣驯原药材样品, 除杂粉碎成粗粉; 加5倍量去离子水, 加热回流提取3次, 每次2 h; 减压浓缩至干燥, 即得渣驯提取物。
渣驯药物配制方式  动物给药: 使用电子天平(GL2242-ISCN, 德国赛多利斯) 精密称取一定质量不同产地来源的渣驯药物, 加入相应体积双蒸水, 充分震荡及超声混匀, 配制成50、200 mg·kg-1渣驯药物溶液。细胞给药: 使用电子天平精密称取一定质量不同产地来源的渣驯药物, 加入相应体积双蒸水, 充分震荡及超声混匀, 配制成10、25、50、100 mg·mL-1渣驯药物溶液, 细胞加药前使用0.22 μm滤膜(SLGPR33R8, 美国密理博) 充分滤过后使用。
血清转氨酶检测  收集小鼠血清, 采用全自动生化分析仪(TBA-40FR, 日本东芝株式会社) 检测血清中ALT、AST、LDH含量。
肝组织GSH、MDA含量检测  取肝组织, 用生理盐水制成10%肝组织匀浆, 采用微量GSH和MDA检测试剂盒检测肝组织匀浆内GSH和MDA含量, 同时应用BCA蛋白定量试剂盒测定各样品蛋白含量对结果进行校正。
肝组织HE染色  取肝大叶, 4%多聚甲醛固定, 石蜡进行包埋。将石蜡组织块切成4 µm切片。进行苏木精-伊红(HE) 染色, 并评估肝脏损伤程度。
细胞毒性分析  采用MTT法检测ZX-SI、ZX-G、ZX-SNLZ、ZX-CDLL、ZX-SC的细胞毒性。取对数生长期HepG2细胞以8 000个/孔接种于96孔板中, 培养24 h后, 加入终质量浓度10、25、50、100 μg·mL-1 ZX-SI、ZX-G、ZX-SNLZ、ZX-CDLL和ZX-SC作用细胞24 h, 弃去培养液, 每孔加入100 µL 0.5 mg·mL-1 MTT, 继续培养4 h, 弃去MTT液, 每孔加入150 µL二甲基亚砜(DMSO), 于酶标仪(Multiskan FC) 570 nm波长处测定吸光度(OD) 值。细胞存活率(%) = (给药细胞OD平均值/溶剂对照细胞OD平均值) ×100%。
细胞存活率检测  HepG2细胞以8 000个/孔接种于96孔细胞培养板, 培养24 h后, 加入终质量浓度10、25、50、100 μg·mL-1 ZX-SI、ZX-G、ZX-SNLZ、ZX-CDLL和ZX-SC, 同时设阳性对照组双环醇10 mmol·L-1、溶剂对照组及APAP模型组, 药物预孵育12 h。随后除溶剂对照组外, 其余各组均加入终浓度为8 mmol·L-1 APAP继续作用细胞24 h。弃去培养液, 采用MTT法检测细胞存活率。
统计学分析  使用GraphPad Prism 8进行单因素方差分析(one way ANOVA) 以确定组间差异显著性。数据显示为平均值±标准误(SEM), 并且在P < 0.05时被认为具有统计学意义。
ZX-SI、ZX-G、ZX-SNLZ、ZX-CDLL和ZX-SC (50和200 mg·kg-1) 灌胃给药5天, 各组小鼠状态正常。与空白对照组比较, 0.2% CCl4有降低小鼠体重的趋势, 亦可显著升高小鼠肝脏指数(P < 0.001), 对脾脏指数无明显影响。与模型组比较, 双环醇200 mg·kg-1和ZX-CDLL 50 mg·kg-1给药显著升高小鼠体重(P < 0.001, P < 0.05), 改善小鼠一般状态(图 1A)。同时与模型组比较, ZX-SNLZ 200 mg·kg-1能显著降低小鼠肝脏指数(P < 0.05); 双环醇200 mg·kg-1能显著增加小鼠肝脏指数(P < 0.001), 与前期课题组研究结果相一致; 不同产地来源的渣驯各给药组脾脏指数较模型组相比无显著变化(图 1BC)。
CCl4所致急性肝损伤模型其肝组织病理学特征与人类病毒性肝炎极为相似, 为研究保肝药物药效的经典模型。肝脏CYP450酶系可将CCl4代谢为·CCl3, 引起生物膜脂质过氧化, 以及细胞内活性氧(reactive oxygen species, ROS) 积聚, DNA突变、断裂, 从而导致肝细胞严重损伤[5]
在本研究中, CCl4所致急性肝损伤模型组小鼠血清肝损伤生物标记物ALT、AST水平较空白对照组显著升高(P < 0.001)。与模型组相比, 各不同产地来源的渣驯给药[ZX-SI 50 mg·kg-1 (P < 0.05, P > 0.05)、ZX-SI 200 mg·kg-1 (P < 0.01, P < 0.05)、ZX-G 50 mg·kg-1 (P < 0.01)、ZX-G 200 mg·kg-1 (P < 0.01, P < 0.05)、ZX-SNLZ 50 mg·kg-1 (P < 0.001, P < 0.01)、ZX-SNLZ 200 mg·kg-1 (P < 0.001)、ZX-CDLL 50 mg·kg-1 (P < 0.01, P > 0.05)、ZX-CDLL 200 mg·kg-1 (P < 0.01)、ZX-SC 50 mg·kg-1 (P < 0.05, P < 0.01)、ZX-SC 200 mg·kg-1 (P < 0.001, P < 0.01)] 均能显著降低小鼠血清中ALT、AST含量(图 1DE)。与对照组相比, 模型组小鼠血清LDH显著升高(P < 0.01)。与模型组相比, 各不同产区的渣驯给药[ZX-SI 50 mg·kg-1 (P < 0.01)、ZX-SI 200 mg·kg-1 (P < 0.05)、ZX-G 50 mg·kg-1 (P < 0.01)、ZX-G 200 mg·kg-1 (P < 0.05)、ZX-SNLZ 50 mg·kg-1 (P < 0.05)、ZX-SNLZ 200 mg·kg-1 (P < 0.01)、ZX-CDLL 50 mg·kg-1 (P < 0.05)、ZX-SC 50 mg·kg-1 (P < 0.05)、ZX-SC 200 mg·kg-1 (P < 0.01)] 均能显著降低小鼠血清中LDH含量(图 1F)。不同产地来源的藏药材渣驯在CCl4所致急性肝损伤模型中对小鼠血清ALT、AST、LDH的降低作用无显著差异。
肝组织病理检查对鉴别诊断及准确、全面评估肝组织损伤程度至关重要。对肝组织进行HE染色病理分析, 与空白对照组比较, 模型组小鼠肝组织可见肝细胞坏死, 并有炎症细胞弥散性浸润。与模型组相比, 200 mg·kg-1 ZX-SI、ZX-G、ZX-SNLZ、ZX-CDLL和ZX-SC均可降低肝细胞坏死水平和肝组织炎症(图 2)。
ROS如超氧自由基阴离子(O2·) 和过氧化物非自由基二阴离子(O22-), 既可以通过酶产生, 也可以通过分子氧(O2) 产生。ROS具有高度反应性, 可攻击其附近的各种生物分子, 包括蛋白质、DNA和脂质, 如多聚不饱和脂肪酸(polyunsaturated fatty acid, PUFAs), 这种现象通常被称为氧化应激。其中, PUFA花生四烯酸通过过氧化, 最终形成MDA[6], 因此, 检测MDA可以反映体内氧化应激的程度。在本研究中, 与空白对照组相比, 模型组小鼠肝组织MDA含量显著升高(P < 0.01)。与模型组相比, 200 mg·kg-1 ZX-SI、ZX-G和ZX-CDLL给药能显著降低小鼠肝组织MDA含量(P < 0.05), 改善肝组织氧化应激(图 3A), 并且效果优于阳性药双环醇。
GSH是一种普遍存在的三肽, 通过多种机制参与细胞稳态的调节。GSH的主要已知作用是抗氧化能力, 有助于维持细胞的氧化还原循环[7]。在本研究中, 与空白对照组相比, 模型组小鼠肝组织GSH含量显著下降(P < 0.01)。与模型组相比, 不同产地来源的藏药材渣驯(200 mg·kg-1) 均能显著升高小鼠肝组织GSH含量(P < 0.01, 图 3B)。
为进一步分析不同产地渣驯的肝保护活性, 开展了体外活性评价研究, 细胞毒性研究结果显示, ZX-SI 25~100 μg·mL-1作用HepG2细胞24 h, 能够浓度依赖性地降低细胞存活率, 表现细胞毒性作用。ZX-SI 10 μg·mL-1、ZX-G 25~100 μg·mL-1、ZX-SNLZ 10~100 μg·mL-1、ZX-CDLL 10~100 μg·mL-1、ZX-SC 10~100 μg·mL-1对HepG2细胞存活均无显著影响(图 4A~E), 采用无毒浓度开展体外肝保护活性研究。
在本研究中, 采用终浓度8 mmol·L-1 APAP刺激HepG2细胞24 h, 与溶剂对照组相比, 8 mmol·L-1 APAP显著降低HepG2细胞存活率(P < 0.001), 与模型组比较, ZX-SI 10 μg·mL-1 (P < 0.05)、ZX-G 100 μg·mL-1 (P < 0.01)、ZX-SNLZ 10, 25 μg·mL-1 (P < 0.05, P < 0.01)、ZX-CDLL 10~100 μg·mL-1 (P < 0.001)、ZX-SC 10, 25, 50 μg·mL-1 (P < 0.001, P < 0.01, P < 0.001) 均能显著提高HepG2细胞存活率, 对肝细胞产生保护作用(图 4F), 其中, 来源于西藏自治区昌都市洛隆县产地的渣驯药材(ZX-CDLL) 对肝细胞的保护作用显著优于其他产地来源渣驯药材。
CCl4是急性氧化性肝损伤动物模型的常用造模剂, CCl4可经过肝脏CYP450酶系代谢产生三氯甲基自由基(CCl3·), CCl3·可与蛋白质、脂质、核酸等结合, 引起线粒体损伤和氧化应激, 其亦可与O2反应形成CCl3OO·, 引发脂质过氧化链式反应, 破坏细胞膜, 最终导致肝脏损伤[8]。APAP是全球常用的解热镇痛药物, 在正常情况下, APAP进入体内经CYP450酶系代谢后, 生成毒性中间产物N-乙酰苯醌亚胺(NAPQI) 会与肝脏内GSH结合, 进而以无毒形式从尿中排出。APAP超剂量使用后, 毒性中间产物NAPQI生成过量, 未被GSH结合的NAPQI则会与肝细胞大分子共价结合, 从而引起肝细胞坏死, 细胞通透性增强[9]
在本研究中, 使用0.2% CCl4及APAP作为造模剂在体内外系统评价了不同产地的藏药材渣驯的肝保护活性, 明确了来源于新疆维吾尔自治区阿勒泰地区哈巴河县、巴基斯坦吉尔吉特地区、西藏自治区山南市洛扎县、西藏自治区昌都市洛隆县、四川省甘孜藏族自治州九龙县产地来源的渣驯药材肝保护药效, 为以上产地来源渣驯的临床应用提供依据。另外, 在0.2% CCl4诱导的氧化性肝损伤模型中, 渣驯对肝组织中MDA的降低作用优于双环醇, 对肝组织中GSH的升高作用双环醇更优, 提示渣驯具有与双环醇不同的作用特点, 结合课题组前期研究结果[10], 渣驯可显著抑制H2O2诱导的肝细胞损伤, 降低细胞内ROS含量, 同时抑制肝细胞凋亡, 具有良好的抗氧化活性及抗凋亡活性。双环醇作为应用了20多年的抗炎保肝药物, 抗氧化和抗炎亦是其发挥疗效的作用环节, 研究表明, 双环醇能够通过调节胆汁酸代谢、免疫反应、炎症和氧化反应以及P450等多种途径来减轻小鼠非酒精性脂肪肝[11]。渣驯肝保护活性物质基础和深入的作用机制值得进一步开展。
渣驯作为世界各大传统医学广泛使用的常用药物, 对其药物的认识和使用已具有悠久的历史[12], 但因各大医学的理论体系不一, 对其生药的归属有不一样的阐述。藏医学认为渣驯归属土类或精华类药物, 分为6种, 即金精、银精、铜精、铁精、锌精、锡(铅) 精, 其中铁精具有解肝热, 清明目药效[13]。本次实验在充分论证的情况下均选择符合铁精类性状的不同产地渣驯药材, 发现源自不同产地的渣驯确实均具有良好的肝保护活性, 说明渣驯药材作为世界性药材, 在肝保护方面具有其一致性; 但因不同产地而作用大小有一定的差异, 在体内外活性研究中, 来源于西藏自治区昌都市洛隆县产地的渣驯药材肝保护活性相对略优。
