Studies on chemical constituents in the rhizome of Dalbergia rimosa Roxb. The chemical constituents from the ethyl acetate part of D. rimosa were isolated and purified by silica gel, MCI gel, Sephadex LH-20 gel and semi-preparative HPLC, and the stuctures were identified by spectral method. Thirteen compounds were isolated from the ethyl acetate part of the rhizome of D. rimosa and identified as dalbergiaisoflavones A, B (1, 2), formononetin (3), 7,4′-dimethoxyisoflavone (4), 4′,6,7-trimethoxyisoflavone (5), biochanin A (6), prunetin (7), 7-O-methyltectorigenin (8), 3′-hydroxydaidzein (9), orobol 7,3′-dimethyl ether (10), 2′,7-dihydroxy-4′,5′- dimethoxyisoflavone (11), pruinosanone E (12), caviunin (13). Compounds 1 and 2 are new compounds, and compounds 3-13 were isolated from this plant for the first time. Compounds 9 and 11 had remarkable scavenging effect on DPPH free radicals.

研究多裂黄檀Dalbergia rimosa Roxb.根茎的化学成分。采用硅胶、MCI和Sephadex LH-20柱色谱以及半制备高效液相色谱等技术进行分离纯化, 通过波谱数据鉴定化合物结构。从多裂黄檀根茎的乙酸乙酯部位中分离得到13个化合物, 分别鉴定为多裂异黄酮A、B (12)、刺芒柄花素(3)、7,4′-二甲氧基异黄酮(4)、4′,6,7-trimethoxyisoflavone (5)、鹰嘴豆芽素A (6)、樱黄素(7)、7-O-methyltectorigenin (8)、3′-hydroxydaidzein (9)、orobol 7,3′-dimethyl ether (10)、2′,7-dihydroxy-4′,5′-dimethoxyisoflavone (11)、pruinosanone E (12)、caviunin (13)。其中, 化合物12为新化合物, 化合物3~13为首次从该植物中分离得到。化合物911对DPPH自由基有显著清除作用。

研究多裂黄檀Dalbergia rimosa Roxb.根茎的化学成分。采用硅胶、MCI和Sephadex LH-20柱色谱以及半制备高效液相色谱等技术进行分离纯化, 通过波谱数据鉴定化合物结构。从多裂黄檀根茎的乙酸乙酯部位中分离得到13个化合物, 分别鉴定为多裂异黄酮A、B (12)、刺芒柄花素(3)、7,4′-二甲氧基异黄酮(4)、4′,6,7-trimethoxyisoflavone (5)、鹰嘴豆芽素A (6)、樱黄素(7)、7-O-methyltectorigenin (8)、3′-hydroxydaidzein (9)、orobol 7,3′-dimethyl ether (10)、2′,7-dihydroxy-4′,5′-dimethoxyisoflavone (11)、pruinosanone E (12)、caviunin (13)。其中, 化合物12为新化合物, 化合物3~13为首次从该植物中分离得到。化合物911对DPPH自由基有显著清除作用。

Studies on chemical constituents in the rhizome of Dalbergia rimosa Roxb. The chemical constituents from the ethyl acetate part of D. rimosa were isolated and purified by silica gel, MCI gel, Sephadex LH-20 gel and semi-preparative HPLC, and the stuctures were identified by spectral method. Thirteen compounds were isolated from the ethyl acetate part of the rhizome of D. rimosa and identified as dalbergiaisoflavones A, B (1, 2), formononetin (3), 7,4′-dimethoxyisoflavone (4), 4′,6,7-trimethoxyisoflavone (5), biochanin A (6), prunetin (7), 7-O-methyltectorigenin (8), 3′-hydroxydaidzein (9), orobol 7,3′-dimethyl ether (10), 2′,7-dihydroxy-4′,5′- dimethoxyisoflavone (11), pruinosanone E (12), caviunin (13). Compounds 1 and 2 are new compounds, and compounds 3-13 were isolated from this plant for the first time. Compounds 9 and 11 had remarkable scavenging effect on DPPH free radicals.

多裂黄檀为豆科(Fabaceae) 黄檀属(Dalbergia) 植物多裂黄檀(Dalbergia rimosa Roxb.) 的干燥根茎, 分布于广西各地, 壮药名为Gengpeng (更彭) 和Meikouwai (美扣外)。壮医认为其性热味辣, 具有调龙路、除湿毒、消肿痛的作用, 可用于治疗巧尹(头痛), 叶用于呗农(痈疮)、呗叮(疗疮)[1, 2]。目前, 国内外对多裂黄檀化学成分和药理研究较少, 本实验运用现代色谱和波谱技术, 从多裂黄檀乙酸乙酯部位中分离纯化得到13个化合物, 分别鉴定为: 多裂异黄酮A、B (12)、刺芒柄花素(3)、7,4′-二甲氧基异黄酮(4)、4′,6,7-trimethoxyisoflavone (5)、鹰嘴豆芽素A (6)、樱黄素(7)、7-O-methyltectorigenin (8)、3′-hydroxydaidzein (9)、orobol 7,3′-dimethyl ether (10)、2′,7-dihydroxy-4′,5′-dimethoxyisoflavone (11)、pruinosanone E (12)、caviunin (13), 结构如图 1所示。其中, 化合物911对DPPH自由基有显著的清除作用。
化合物1  黄色油状物, 易溶于甲醇。HR-ESI-MS给出准分子离子峰m/z: 489.227 7 [M-H]- (calcd for C30H33O6 489.227 7), 推测化合物的分子式为C30H34O6, 计算其不饱和度为14。1H NMR (500 MHz, CD3OD) 谱中, 芳香区出现4个氢质子信号, 其中δH 7.98 (1H, s) 为异黄酮2位质子的特征信号; δH 7.05 (2H, s) 和δH 6.37 (1H, s) 提示结构中存在一个1, 3, 4, 5-四取代苯环和一个1, 2, 3, 4, 5-五取代苯环, 通过HSQC谱确定它们对应的碳原子分别出现在δC 129.0、δC 93.9; δH 3.89 (2H, s) 提示分子中一个存在CH2OH片段, 通过HSQC谱确定对应的碳原子出现在δC 69.1; δH 5.55~5.50 (1H, m)、5.36~5.31 (2H, m)、3.39 (2H, d, J = 7.3 Hz)、3.35 (4H, d, J = 7.3 Hz)、1.82 (3H, s)、1.74 (6H, s)、1.73 (6H, s) 为3个异戊烯基的氢质子信号, 通过HSQC谱确定它们对应的碳原子分别出现在δC 124.6、123.7、21.9、29.7、13.8、25.9、17.9。13C NMR (125 MHz, CD3OD) 谱中显示了该化合物有30个碳信号, 包括14个季碳、7个次甲基碳、4个亚甲基碳以及5个甲基碳信号, 其中δC 182.4为异黄酮4位羰基碳特征信号, 结合1H NMR谱信号, 推断该化合物为异黄酮类化合物。HMBC谱中, δH 7.05和δC 153.7、129.9、125.1、29.7相关, 进一步提示有1, 3, 4, 5-四取代的苯环; δH 3.35与δC 129.9、129.0、153.7相关, 从而确定C-3′和C-5′位分别取代一个异戊烯基; δH 6.37和δC 163.8、157.7、112.5、106.1相关, δH 3.39和δC 163.8、160.3、112.5相关, δH 3.89和δC 135.8、124.6、13.8相关, 从而确定C-6取代一个4-羟基异戊烯基。1H-1H COSY谱中显示H-1′′/H-2′′相关, H-1′′′/H-2′′′、1′′′′/2′′′′相关进一步证实了异戊烯基结构的存在。ROESY谱中显示H-5′′/H-1′′相关, H-5′′/H-4′′相关, 提示4-羟基异戊烯基上双键构型为E型。综合以上信息, 确定化合物的结构为5, 7,4′-trihydroxy-6-(4-hydroxy-3-methyl-2-butenyl)-3′, 5′-diprenyl isoflavone (图 1), 经Scifinder检索为未见文献报道的新化合物, 命名为多裂异黄酮A (dalbergiaisoflavone A), 借助于1D和2D NMR数据(图 2), 对化合物11H NMR和13C NMR信号进行了全归属(表 1)。
化合物2  黄色油状物, 易溶于甲醇。HR-ESI-MS给出准分子离子峰m/z: 489.227 5 [M-H]- (calcd for C30H33O6 489.227 7), 推测化合物的分子式为C30H34O6, 计算其不饱和度为14。比较化合物2与化合物1的1D和2D NMR数据(图 3), 发现二者结构相似, 仅是4-羟基异戊烯基取代位置不同。HMBC谱显示δH 6.27 (H-6) 和δC 163.7 (C-7)、161.5 (C-5)、106.3 (C-8) 相关, δH 3.48 (H-1′′) 和δC 163.7、156.9 (C-9)、106.3相关, 因此确定4-羟基异戊烯基取代在C-8位上。ROESY谱中显示H-5′′/H-1′′相关, H-5′′/H-4′′相关, 提示4-羟基异戊烯基上双键构型为E型。从而确定化合物的结构为5, 7,4′-trihydroxy-8-(4-hydroxy-3-methyl-2-butenyl)-3′, 5′-diprenyl isoflavone, 命名为多裂异黄酮B (dalbergiaisoflavone B), 其1H NMR和13C NMR信号归属如表 1
以化合物1~13对DPPH自由基的清除能力评价各化合物的抗氧化活性。结果显示, 化合物911具有显著的DPPH清除能力, 其IC50分别为(17.66 ± 0.04)、(14.68 ± 0.08) μg·mL-1, 接近于阳性对照L-抗坏血酸(Vitc), 其IC50为(11.99 ± 0.17) μg·mL-1
Agilent G6230型飞行时间质谱仪(美国Agilent公司); 布鲁克AVANCE Ⅲ 500 MHz型核磁共振波谱仪(德国Bruker公司); 赛默飞世尔NICOLET iS10型中红外光谱仪, Hypersil GOLDTM C18柱(250 mm × 10 mm, 5 μm, 美国赛默飞世尔公司); 岛津LC-20AR、岛津LC-2030 PLUS型高效液相色谱仪、岛津UV2401PC型紫外可见分光光度仪(日本岛津公司); BP211D型电子分析天平(德国赛多利斯公司); Multiskan Sky 1530型全波长酶标仪(美国Thermo Fisher Scientific公司); 旋转蒸发仪(EYELA OSB-2100、OSB-2200, 上海爱朗仪器有限公司); ZF-6型三用紫外分析仪(上海嘉鹏科技有限公司); CHP P20小孔树脂(日本三菱化学公司); LH-20羟丙基葡聚糖凝胶(Sephadex LH-20, 美国Pharmacia公司); 柱色谱硅胶(100~200目, 国药集团化学试剂有限公司); 硅胶薄层板(50 mm × 100 mm等, 青岛海洋化工有限公司); 甲醇、乙腈(色谱纯, Fisher科技有限公司); L-抗坏血酸(纯度 > 99%, 上海麦克林生化科技有限公司); 1, 1-二苯基-2-三硝基苯(DPPH, 纯度 > 98%, 美国Sigma公司); 所用常规试剂均为分析纯(四川西陇科学有限公司)。
多裂黄檀药材38.0 kg, 以10倍量80%乙醇回流3次(2.0、1.5、1 h), 减压回收溶剂得总浸膏(1.84 kg)。取总浸膏(1.64 kg), 温水悬浮后, 依次用石油醚、乙酸乙酯、正丁醇萃取, 减压回收溶剂后得到石油醚部位(36.0 g), 乙酸乙酯部位(179.4 g), 正丁醇部位(504.9 g) 和水部位(773.0 g)。
乙酸乙酯部位浸膏145.0 g, 以硅胶柱色谱进行分离, 石油醚-乙酸乙酯(100∶0、20∶1、10∶1、5∶1、2∶1、0∶1) 梯度洗脱, 通过薄层色谱检识合并得到7个组分(Fr.1~Fr.7)。Fr.3以MCI柱色谱(甲醇-水, 50∶50~100∶0) 进行梯度洗脱, 合并得到9个组分(Fr.3-1~Fr.3-9)。其中Fr.3-8经硅胶H柱色谱(石油醚-乙酸乙酯, 15∶1~2∶1) 梯度洗脱得到10个组分(Fr.3-8-1~Fr.3-8-10)。其中, Fr.3-8-5经半制备高效液相色谱(甲醇-水, 55∶45) 纯化得化合物4 (11.2 mg, tR = 31.3 min), Fr.3-8-6经半制备高效液相色谱(甲醇-水, 68∶32) 纯化得化合物10 (1.4 mg, tR = 33.1 min)。Fr.4经MCI柱色谱(甲醇-水, 30∶70~100∶0) 梯度洗脱合并得到9个组分(Fr.4-1~Fr.4-9)。其中, Fr.4-7以硅胶柱色谱(石油醚-丙酮, 10∶1~1∶1) 以及半制备高效液相色谱(甲醇-0.1%甲酸, 52∶48) 分离纯化得化合物7 (2.9 mg, tR = 35.5 min), 化合物6 (3.5 mg, tR = 38.0 min)。Fr.5经MCI柱色谱(甲醇-水, 50∶50~100∶0) 梯度洗脱合并得到15个组分(Fr.5-1~Fr.5-15)。Fr.5-6经硅胶H柱色谱(氯仿-丙酮, 50∶1~4∶1)、Sephadex LH-20 (氯仿-甲醇, 1∶1) 以及半制备高效液相色谱(乙腈-水, 32∶68) 分离纯化得化合物12 (4.1 mg, tR=14.4 min)。Fr.5-8析出白色针状结晶, 经半制备高效液相色谱(甲醇-0.1%甲酸, 50∶50) 纯化得化合物8 (8.4 mg, tR = 27.6 min), 母液经硅胶H柱色谱(石油醚-乙酸乙酯, 3∶1~2∶3) 以及半制备高效液相色谱(甲醇-水, 45∶55) 分离纯化得化合物13 (8.5 mg, tR = 36.4 min), 化合物3 (8.8 mg, tR = 47.1 min)。Fr.5-10经硅胶H柱色谱(石油醚-乙酸乙酯, 3∶1~1∶1) 洗脱后, 经半制备高效液相色谱(甲醇-水, 53∶47) 纯化得化合物5 (5.5 mg, tR = 28.1 min)。Fr.5-11经硅胶H柱色谱(石油醚-乙酸乙酯, 8∶1~1∶1) 梯度洗脱以及半制备高效液相色谱(甲醇-水, 72∶38) 分离纯化得化合物1 (7.1 mg, tR = 42.8 min) 和化合物2 (15.1 mg, tR = 48.0 min)。Fr.6经MCI柱色谱(甲醇-水, 30∶70~100∶0) 梯度洗脱, 合并得到12个组分(Fr.6-1~Fr.6-12)。Fr.6-6经硅胶H柱色谱(氯仿-甲醇, 50∶1~15∶1)、Sephadex LH-20 (氯仿-甲醇, 1∶1) 柱色谱以及半制备高效液相色谱(甲醇-0.1%乙酸, 30∶70) 分离纯化得化合物9 (22.4 mg, tR = 37.3 min)。Fr.6-8以硅胶H柱色谱(石油醚-乙酸乙酯, 3∶1~1∶2) 梯度洗脱后, 经半制备高效液相色谱(甲醇-水, 48∶52) 分离纯化得化合物11 (36.6 mg, tR = 23.4 min)。
参考文献[3]的方法, 以甲醇溶解DPPH, 配制成0.1 mmol·L-1的DPPH贮备液, 避光冷藏备用。精确称取化合物溶解于DMSO中, 稀释成系列质量浓度备用。移取50 μL一系列浓度化合物溶液和阳性对照溶液加至96孔板中, 再向各孔中加入150 μL DPPH贮备液, 混匀, 室温黑暗中放置30 min, 反应完全后在波长517 nm处测定其吸光度A1。以不加样品的相同溶剂为空白对照A0, Vitc作为阳性对照, 每个浓度平行测定3次后取平均值, 清除率% = (A0-A1) / A0 × 100%。计算样品对DPPH自由基清除率及半数抑制浓度(IC50)。

通过文献对照, 其他已知化合物分别鉴定为刺芒柄花素(3)[4]、7,4′-二甲氧基异黄酮(4)[5]、4′,6,7-trimethoxyisoflavone (5)[6]、鹰嘴豆芽素A (6)[7]、樱黄素(7)[8]、7-O-methyltectorigenin (8)[9]、3′-hydroxydaidzein (9)[10]、orobol 7,3′-dimethyl ether (10)[11]、2′,7-dihydroxy-4′,5′-dimethoxyisoflavone (11)[12]、pruinosanone E (12)[13]、caviunin (13)[14, 15]
