Macroporous adsorption resin, MCI, Toyopearl HW-40C and silica gel column chromatography combined with the semi-preparative HPLC were used to isolate and purify the water extract of Cornus officinalis. And the structures of compounds were identified by HRESIMS, NMR, IR, UV and ECD. A total of twelve compounds were isolated from the fruits of Cornus officinalis. They were identified as neolignan B (1), 2, 3-dihydro-7-methoxy-2-(4′-hydroxy-3′-methoxyphenyl)-3a-O-β-D-xylopyranosyloxymethyl-5-benzofuranpropanol (2), cornucadinoside A (3), 3, 4-dihydroxyacetophenone (4), n-butyl gallate (5), 3-hydroxybenzoic acid (6), n-butyl quininate (7), 5-hydroxymaltol (8), 2-furolic acid (9), 5-hydroxymethylfurfural (10), 5-(methoxycarbonyl)-2-pyrrolidone (11), and dimethyl malate (12). Compound 1 is a new biphenyl lignan, named as neolignan B. Compounds 2, 4, 5, 7-9 and 11 were isolated from Cornus officinalis for the first time.

采用大孔吸附树脂、MCI、Toyopearl HW-40C以及硅胶等柱色谱技术并结合半制备液相对山茱萸水提物进行分离纯化, 通过HRESIMS、NMR、IR、UV和ECD等方法对分离得到的化合物进行结构鉴定。从山茱萸果实中共分离得到12个化合物, 分别鉴定为neolignan B (1)、2, 3-dihydro-7-methoxy-2-(4′-hydroxy-3′-methoxyphenyl)-3a-O-β-D-xylopyranosyloxymethyl-5-benzofuranpropanol (2)、cornucadinoside A (3)、3, 4-二羟基苯乙酮(4)、没食子酸正丁酯(5)、间羟基苯甲酸(6)、奎宁酸正丁酯(7)、5-羟基麦芽酚(8)、2-糠酸(9)、5-羟甲基糠醛(10)、5-(甲氧基羰基)-2-吡咯烷酮(11)、苹果酸二甲酯(12)。化合物1为新联苯类木脂素, 命名为neolignan B。化合物2457~911为首次从山茱萸中分离得到。

采用大孔吸附树脂、MCI、Toyopearl HW-40C以及硅胶等柱色谱技术并结合半制备液相对山茱萸水提物进行分离纯化, 通过HRESIMS、NMR、IR、UV和ECD等方法对分离得到的化合物进行结构鉴定。从山茱萸果实中共分离得到12个化合物, 分别鉴定为neolignan B (1)、2, 3-dihydro-7-methoxy-2-(4′-hydroxy-3′-methoxyphenyl)-3a-O-β-D-xylopyranosyloxymethyl-5-benzofuranpropanol (2)、cornucadinoside A (3)、3, 4-二羟基苯乙酮(4)、没食子酸正丁酯(5)、间羟基苯甲酸(6)、奎宁酸正丁酯(7)、5-羟基麦芽酚(8)、2-糠酸(9)、5-羟甲基糠醛(10)、5-(甲氧基羰基)-2-吡咯烷酮(11)、苹果酸二甲酯(12)。化合物1为新联苯类木脂素, 命名为neolignan B。化合物2457~911为首次从山茱萸中分离得到。

Macroporous adsorption resin, MCI, Toyopearl HW-40C and silica gel column chromatography combined with the semi-preparative HPLC were used to isolate and purify the water extract of Cornus officinalis. And the structures of compounds were identified by HRESIMS, NMR, IR, UV and ECD. A total of twelve compounds were isolated from the fruits of Cornus officinalis. They were identified as neolignan B (1), 2, 3-dihydro-7-methoxy-2-(4′-hydroxy-3′-methoxyphenyl)-3a-O-β-D-xylopyranosyloxymethyl-5-benzofuranpropanol (2), cornucadinoside A (3), 3, 4-dihydroxyacetophenone (4), n-butyl gallate (5), 3-hydroxybenzoic acid (6), n-butyl quininate (7), 5-hydroxymaltol (8), 2-furolic acid (9), 5-hydroxymethylfurfural (10), 5-(methoxycarbonyl)-2-pyrrolidone (11), and dimethyl malate (12). Compound 1 is a new biphenyl lignan, named as neolignan B. Compounds 2, 4, 5, 7-9 and 11 were isolated from Cornus officinalis for the first time.

山茱萸为山茱萸科植物山茱萸Cornus officinalis的干燥成熟果实, 为河南省道地药材之一。河南省西峡县为山茱萸的主产区, 因其产量大, 品质佳, 素有“山茱萸之乡”的美誉。目前文献[1,2]报道表明, 山茱萸中富含环烯醚萜及其苷、木脂素、黄酮、倍半萜、鞣质以及酚酸等多种化学成分。研究发现, 山茱萸具有广泛的药理作用, 包括降血糖[3]、抗炎[4]、神经保护[5]、抗菌[6]、抗氧化[7]、抗抑郁[8]、肝肾保护[9]以及抗肿瘤[10]等活性。山茱萸具有较高的药用价值, 在我国已有千年以上的用药历史[11], 同时其作为观赏性树种, 可广泛应用于城市绿化[12], 具有较好的开发前景。因此本实验对山茱萸水提物进行系统的分离研究, 以期发现更多结构新颖、生物活性显著的化合物, 丰富山茱萸化学成分研究内容, 为山茱萸的进一步开发利用奠定物质基础。本实验利用各种柱色谱方法以及半制备液相, 从山茱萸中分离得到12个化合物, 分别鉴定为neolignan B (1)、2, 3-dihydro-7-methoxy-2-(4′-hydroxy-3′-methoxyphenyl)-3a-O-β-D-xylopyranosyloxymethyl-5-benzofuranpropanol (2)、cornucadinoside A (3)、3, 4-二羟基苯乙酮(4)、没食子酸正丁酯(5)、间羟基苯甲酸(6)、奎宁酸正丁酯(7)、5-羟基麦芽酚(8)、2-糠酸(9)、5-羟甲基糠醛(10)、5-(甲氧基羰基)-2-吡咯烷酮(11)、苹果酸二甲酯(12)。化合物1为新联苯类木脂素, 命名为neolignan B。化合物2457~911为首次从山茱萸中分离得到。
化合物1 灰色无定形粉末。[α]$ {}_{\mathrm{D}}^{20} $ -10 (c 0.04, CH3OH); CD (CH3OH) λmaxε): 208 (-0.19) nm; UV (CH3OH) λmax (log ε): 209 (4.70), 282 (3.74) nm; IR νmax: 3 347, 2 946, 1 730, 1 457, 1 222, 1 119, 1 034 cm-1, 提示分子中含有酚羟基(3 347 cm-1) 和苯环(1 457, 1 222, 1 119, 1 034 cm-1); HRESIMS m/z: 429.152 5 [M+Na]+ (计算值为429.152 0), 可确定化合物1的分子式为C21H26O8, 不饱和度为9。1H NMR谱(表 1) 中, 芳香区显示两组苯环上氢信号δH 6.72 (2H, brs, H-3′, 5′), 6.47 (1H, d, J = 2.0 Hz, H-5), 6.43 (1H, d, J = 2.0 Hz, H-3); 两个连氧次甲基氢信号δH 4.85 (1H, overlapped, H-3‴), 4.00 (1H, m, H-4‴); 两个连氧亚甲基氢信号δH 3.71 (1H, dd, J = 12.4, 2.5 Hz, H-5‴a), 3.50 (1H, dd, J = 12.4, 4.5 Hz, H-5‴b), 3.56 (2H, t, J = 6.5 Hz, H-3″); 三个甲氧基氢信号δH 3.86 (9H, s, 6, 2′, 6′-OCH3) 以及两个亚甲基氢信号δH 2.59 (2H, dd, J = 8.6, 6.8 Hz, H-1″), 1.81 (2H, m, H-2″)。结合13C-NMR谱和DEPT-135谱可知, 化合物1中含有一个对称1, 3, 4, 5-四取代苯环δC 137.1 (C-1′), 149.4 (C-2′, 6′), 105.9 (C-3′, 5′), 128.7 (C-4′); δC 132.5 (C-1), 145.6 (C-2), 110.6 (C-3), 133.9 (C-4), 106.2 (C-5), 149.9 (C-6) 为另一组苯环上的碳信号。δC 77.9 (C-3‴), 79.9 (C-4‴), 62.1 (C-5‴), 62.2 (C-3″) 为两个连氧次甲基和两个连氧亚甲基碳信号; δC 56.8 (2′, 6′-OCH3), 56.7 (6-OCH3) 为三个甲氧基碳信号; δC 32.9 (C-1″), 35.5 (C-2″) 为两个亚甲基碳信号。
1H-1H COSY谱(图 1) 中, H-2″ (δH 1.81) 和H-1″ (δH 2.59)/H-3″ (δH 3.56) 相关, 确定化合物1中含有一个正丙醇基结构片段。H-4‴ (δH 4.00) 和H-3‴ (δH 4.85)/H-5‴ (δH 3.71, 3.50) 相关, 表明C-3‴, C-4‴, C-5‴相连。HMBC谱(图 1) 中, δH 4.85 (H-3‴) 和δC 128.7 (C-4′)/105.9 (C-3′, 5′) 存在远程相关信号, 表明C-3‴和苯环通过C-4′相连。δH 3.86 (6, 2′, 6′-OCH3) 和δC 149.4 (C-2′, 6′)/149.9 (C-6) 存在远程相关信号, 表明甲氧基分别连在苯环的6, 2′和6′位。此外, δH 2.59 (H-1″) 和δC 110.6 (C-3)/133.9 (C-4) 存在远程相关信号, 进一步确定正丙醇基结构片段连在苯环的C-4位上。通过与文献数据进行对比, 化合物1与从棒节石斛中分离得到的6″-de-O-methyldendrofindlaphenol A[13]非常相似, 仅缺少一个对羟基苯乙基结构片段, 多出一个正丙醇基结构片段。NOESY谱(图 1) 中, H-3‴和H-5‴b相关, 确定H-3‴和H-4‴处于异侧。化合物1在208 nm处显示负Cotton效应, 274 nm处显示正Cotton效应, 通过对比实测ECD和计算ECD图谱(图 2), 确定该化合物的绝对构型为3‴S, 4‴S。最终, 化合物1的结构鉴定为4ʹ-((3S, 4S)-4-(hydroxymethyl)-1, 2-dioxetan-3-yl)-4-(3-hydroxypropyl)-2ʹ, 6, 6ʹ-trimethoxy-[1, 1ʹ-biphenyl]-2-ol (图 1), 为1个新化合物, 命名为neolignan B。
山茱萸是我国临床常用中药材, 具有补益肝肾、收敛固涩的功效, 其果实具有较高的营养价值, 可开发成山茱萸酱、山茱萸饮料和山茱萸酒等一系列保健产品, 具有广泛的开发前景。本实验从山茱萸水提物中分离得到12个化合物, 其中化合物1为新化合物, 化合物2457~911首次从山茱萸中分离得到, 结构类型涉及木脂素、倍半萜和其他类。本实验进一步丰富了山茱萸化学成分的多样性, 为其后续研究奠定了一定的物质基础。
山茱萸药材于2019年9月采自河南省西峡县, 经河南中医药大学药学院董诚明教授鉴定为山茱萸科山茱萸属植物山茱萸(Cornus officinalis) 的果肉, 药材标本(No.20190903) 保存于河南省中药开发工程技术研究中心。
Bruker AVANCE Ⅲ 500型核磁共振仪、Bruker maxis HD型飞行时间质谱(德国布鲁克公司); Thermo EVO300紫外分光光度计型紫外检测器, Thermo Nicolet IS 10傅立叶变换红外光谱仪(美国Thermo Scientific公司); Applied PhotophysicsChirascanqCD圆二色光谱仪(英国Applied Photophysics公司); Autopol Ⅳ全自动旋光仪(美国鲁道夫公司); N-1111型冷冻水循环装置、N-1001型旋转蒸发仪(上海埃朗仪器有限公司); 岛津LC-16型半制备液相色谱仪(日本岛津公司); COSMOSIL 5C18-MS-Ⅱ色谱柱(10 mm × 250 mm, 5 µm; 日本Tesque公司); MCI gel CHP-20、Diaion HP-20、D101大孔吸附树脂(日本三菱化学公司); Toyopearl HW-40C (日本TOSOH公司); 柱色谱硅胶(100~200目, 200~300目, 青岛海洋化工厂); 甲醇、乙腈(色谱纯, 河南宏材化工有限公司)。
山茱萸干燥成熟果肉100 kg, 采用10倍量水煎煮法提取两次, 每次2 h, 减压浓缩得到总浸膏55.1 kg。总浸膏上D101大孔吸附树脂, 使用水和95%乙醇洗脱, 减压浓缩后得到95%乙醇部位(21.7 kg)。95%乙醇部位用适量水分散后, 依次用石油醚、乙酸乙酯、水饱和正丁醇萃取至溶剂变为透明, 减压浓缩后得到石油醚部位(1.5 g), 乙酸乙酯部位(99.3 g), 正丁醇部位(4.15 kg) 和水部位(17.4 kg)。
乙酸乙酯部位(99.3 g) 上Diaion HP-20大孔吸附树脂柱, 使用乙醇-水(0%→95%) 梯度洗脱, 得到5个组份Fr.1~Fr.5。Fr.2 (13.26 g) 经Sephadex LH-20柱色谱分离, 甲醇-水(20%→100%) 梯度洗脱, 得到5个组份Fr.2-1~Fr.2-5。Fr.2-5 (3.5 g) 经硅胶柱色谱分离, 二氯甲烷-甲醇(50∶1→1∶1) 梯度洗脱, 得到8个组份Fr.2-5-1~Fr.2-5-8。Fr.2-5-3经Toyopearl HW-40C柱色谱分离, 甲醇-水(40%→100%) 梯度洗脱, 得到6个组份Fr.2-5-3-1~Fr.2-5-3-6。Fr.2-5-3-3经半制备液相(乙腈-水, 9∶91) 纯化, 得到化合物8 (1.8 mg, tR = 16.2 min) 和9 (8.0 mg, tR = 18.3 min)。Fr.2-5-3-5经半制备液相(乙腈-水, 4∶96) 分离, 得到化合物10 (7.1 mg, tR = 18.0 min)。
正丁醇部位(4.15 kg) 上Diaion HP-20大孔吸附树脂柱, 使用乙醇-水(0%→100%) 梯度洗脱, 得到5个组份A1~A5。A2 (201 g) 经MCI gel柱色谱分离, 甲醇-水(20%→100%) 梯度洗脱, 得到5个组份A2-1~A2-5。A2-4 (15.5 g) 经硅胶柱色谱分离, 二氯甲烷-甲醇(30∶1→1∶1) 梯度洗脱, 得到6个组份A2-4-1~A2-4-6。A2-4-3经硅胶柱色谱分离, 二氯甲烷-甲醇(30∶1→1∶1) 梯度洗脱, 得到6个组份, 即A2-4-3-1~A2-4-3-6。A2-4-3-3经半制备液相(乙腈-水, 12∶88) 纯化, 得到化合物4 (4.4 mg, tR = 18.3 min) 和6 (3.5 mg, tR = 20.5 min)。A2-4-3-4反复经硅胶柱色谱以及半制备液相(甲醇-水, 8∶92) 纯化, 得到化合物11 (3.6 mg, tR = 17.2 min)。A2-4-3-5经半制备液相(乙腈-水, 5∶95) 纯化, 得到化合物12 (5.6 mg, tR = 17.5 min)。A3 (46.5 g) 经硅胶柱色谱分离, 二氯甲烷-甲醇(100∶1→1∶1) 梯度洗脱, 得到7个组份A3-1~A3-7。A3-5 (5.6 g) 经Toyopearl HW-40C柱色谱分离, 甲醇-水(40%→100%) 梯度洗脱, 得到12个组份A3-5-1~A3-5-12。A3-5-4反复通过硅胶柱色谱和半制备液相(甲醇-水, 38∶62) 纯化, 得到化合物1 (3.8 mg, tR = 61.3 min)。A3-5-7经半制备液相(乙腈-水, 23∶77) 分离, 得到化合物2 (3.2 mg, tR = 34.6 min) 和3 (2.9 mg, tR = 36.4 min)。
取水部位(1.92 kg) 经硅胶柱色谱分离, 二氯甲烷-甲醇(100∶1→0∶1) 梯度洗脱, 得到5个组份F1~F5。F2经硅胶柱色谱分离, 二氯甲烷-甲醇(50∶1→0∶1) 梯度洗脱, 得到12个组份F2-1~F2-12。F2-7经Sephadex LH-20柱色谱分离, 甲醇-水(20%→100%) 梯度洗脱, 得到10个组份F2-7-1~F2-7-10。F2-7-4经半制备液相(甲醇-水, 48∶52) 纯化, 得到化合物5 (9.4 mg, tR = 42.8 min)。F2-7-6经半制备液相(乙腈-水, 6∶94) 纯化, 得到化合物7 (12.3 mg, tR = 44.6 min)。

化合物9 无色针状结晶。ESI-MS m/z: 135 [M+Na]+, 分子式为C5H4O31H NMR (500 MHz, CD3OD) δH: 7.72 (1H, t, J = 0.9 Hz, H-5), 7.20 (1H, d, J = 3.5 Hz, H-3), 6.59 (1H, dd, J = 3.5, 1.7 Hz, H-4); 13C NMR (125 MHz, CD3OD) δC: 146.5 (C-2), 112.9 (C-3), 119.0 (C-4), 148.0 (C-5), 161.9 (C-6)。以上数据与文献[21]报道基本一致, 鉴定化合物9为2-糠酸。
化合物10 白色无定形粉末。ESI-MS m/z: 127 [M+H]+, 分子式为C6H6O31H NMR (500 MHz, CD3OD) δH: 9.53 (1H, s, H-7), 7.38 (1H, d, J = 3.6 Hz, H-3), 6.59 (1H, d, J = 3.6 Hz, H-4), 4.61 (2H, s, H-6); 13C NMR (125 MHz, CD3OD) δC: 153.9 (C-2), 124.8 (C-3), 110.9 (C-4), 163.2 (C-5), 57.6 (C-6), 179.4 (C-7)。以上数据与文献[22]报道基本一致, 鉴定化合物10为5-羟甲基糠醛。
化合物11 白色无定形粉末。ESI-MS m/z: 166 [M+Na]+, 分子式为C6H9NO31H NMR (500 MHz, CD3OD) δH: 6.68 (1H, dd, J = 9.0, 4.4 Hz, H-5), 3.76 (3H, s, H-7), 2.74 (1H, m, H-3a), 2.15 (1H, m, H-3b), 2.33 (2H, m, H-4); 13C NMR (125 MHz, CD3OD) δC: 181.1 (C-2), 30.3 (C-3), 25.8 (C-4), 57.0 (C-5), 174.5 (C-6), 52.9 (C-7)。以上数据与文献[23]报道基本一致, 鉴定化合物11为5-(甲氧基羰基)-2-吡咯烷酮。
化合物12 无色油状物。ESI-MS m/z: 163 [M+H]+, 分子式为C6H10O51H NMR (500 MHz, CD3OD) δH: 4.53 (1H, dd, J = 7.2, 4.8 Hz, H-2), 2.81 (1H, dd, J = 15.9, 4.8 Hz, H-3a), 3.75 (3H, s, H-5), 3.69 (3H, s, H-6), 2.72 (1H, dd, J = 15.9, 7.3 Hz, H-3b); 13C NMR (125 MHz, CD3OD) δC: 174.8 (C-1), 68.3 (C-2), 39.7 (C-3), 172.4 (C-4), 52.7(C-5), 52.3 (C-6)。以上数据与文献[24]报道基本一致, 鉴定化合物12为苹果酸二甲酯。
