The excipient processing is an essential part of traditional Chinese medicine processing, and understanding its scientific connotations is a critical scientific issue that urgently needs resolution. Building upon a foundation where the composition of traditional Chinese medicine substances is fundamentally clear, this paper applies the techniques and methods of chemoinformatics to the study of the excipient processing mechanism. Relevant information on traditional Chinese medicines processed with four kinds of excipients (wine, vinegar, salt and honey) was collected, including properties, taste, meridian tropism, chemical components, etc. Molecular descritors and skeletons corresponding to each chemical component were calculated using chemoinformatics to characterize the properties and structural features of the components. Characteristic components associated with the four excipients (wine, vinegar, salt and honey) were explored through multivariate statistical analysis and Murcko skeleton analysis. Further analysis, taking honey-processed Astragali Radix as an example, the focus was on the impact of the processing excipient honey on the solubility and permeability of its characteristic pharmacologically active components (isoflavones). It was discovered that the excipient honey can increase their solubility and permeability, emphasizing that the impact of processing excipients on the pharmacological classification properties is a key breakthrough in elucidating the mechanism of excipient processing. In summary, this study analyzed the characteristic components associated with the processing excipients "wine, vinegar, salt and honey" based on their composition characteristics, providing data support for the research on the mechanism of excipient processing from a biopharmaceutical perspective.

辅料炮制是中药炮制的重要组成部分, 其科学内涵解析是一个亟待解决的关键科学问题。在现代中药物质成分构成基本清晰的基础上, 本文将化学信息学的技术和方法应用到辅料炮制机制研究中。收集酒、醋、盐、蜜4种辅料炮制中药饮片的相关信息, 包括性味、归经、化学成分等。采取化学信息学手段计算各化学成分相应的分子描述符和骨架, 表征成分的性质和结构特征, 并且通过多元统计分析和Murcko骨架分析挖掘酒、醋、盐、蜜4种辅料关联的特征组分。进一步以蜜炙黄芪为例, 聚焦炮制辅料蜜对其特征药效组分(异黄酮组分)溶解度和渗透性的影响, 发现辅料蜜可增加其溶解度和渗透性, 明确炮制辅料对生物药剂学分类性质的影响是阐明辅料炮制机制的关键突破口。综上, 本文基于组分构成特征解析了炮制辅料“酒醋盐蜜”关联的特征组分, 为从生物药剂学角度解析炮制机制提供数据支撑。

辅料炮制是中药炮制的重要组成部分, 其科学内涵解析是一个亟待解决的关键科学问题。在现代中药物质成分构成基本清晰的基础上, 本文将化学信息学的技术和方法应用到辅料炮制机制研究中。收集酒、醋、盐、蜜4种辅料炮制中药饮片的相关信息, 包括性味、归经、化学成分等。采取化学信息学手段计算各化学成分相应的分子描述符和骨架, 表征成分的性质和结构特征, 并且通过多元统计分析和Murcko骨架分析挖掘酒、醋、盐、蜜4种辅料关联的特征组分。进一步以蜜炙黄芪为例, 聚焦炮制辅料蜜对其特征药效组分(异黄酮组分)溶解度和渗透性的影响, 发现辅料蜜可增加其溶解度和渗透性, 明确炮制辅料对生物药剂学分类性质的影响是阐明辅料炮制机制的关键突破口。综上, 本文基于组分构成特征解析了炮制辅料“酒醋盐蜜”关联的特征组分, 为从生物药剂学角度解析炮制机制提供数据支撑。

The excipient processing is an essential part of traditional Chinese medicine processing, and understanding its scientific connotations is a critical scientific issue that urgently needs resolution. Building upon a foundation where the composition of traditional Chinese medicine substances is fundamentally clear, this paper applies the techniques and methods of chemoinformatics to the study of the excipient processing mechanism. Relevant information on traditional Chinese medicines processed with four kinds of excipients (wine, vinegar, salt and honey) was collected, including properties, taste, meridian tropism, chemical components, etc. Molecular descritors and skeletons corresponding to each chemical component were calculated using chemoinformatics to characterize the properties and structural features of the components. Characteristic components associated with the four excipients (wine, vinegar, salt and honey) were explored through multivariate statistical analysis and Murcko skeleton analysis. Further analysis, taking honey-processed Astragali Radix as an example, the focus was on the impact of the processing excipient honey on the solubility and permeability of its characteristic pharmacologically active components (isoflavones). It was discovered that the excipient honey can increase their solubility and permeability, emphasizing that the impact of processing excipients on the pharmacological classification properties is a key breakthrough in elucidating the mechanism of excipient processing. In summary, this study analyzed the characteristic components associated with the processing excipients "wine, vinegar, salt and honey" based on their composition characteristics, providing data support for the research on the mechanism of excipient processing from a biopharmaceutical perspective.

炮制辅料是在炮制过程中添加的具有辅助主药达到炮制目的的附加物料[1, 2], 是中药炮制的核心组成部分。炮制辅料与中药功效之间有着密切联系, 适宜的炮制辅料可起增效减毒作用, 增强中药在临床应用中的灵活性、有效性与安全性[3]。目前, 炮制辅料对饮片临床疗效的影响已有较为详细的记载, 但对于更深层面的其与化学成分性质及结构特征间关联规律仍缺乏系统认识。
近些年, 中药药效物质基础的整体性逐渐得到重视, 形成诸多以组分为代表的研究思路与方法[4-6]。组分的体内生物药剂学过程是影响其临床疗效的关键环节, 而炮制辅料可通过影响组分的溶解度及渗透性, 进而影响其体内吸收及临床疗效[7-9]。作者所在课题组前期研究发现炮制辅料“羊脂油”可促进生物体内自组装纳米胶束形成, 增加活性黄酮的溶解度和渗透性, 促进肠吸收转运, 达到增效作用[10-12]。然而, 目前炮制辅料关联的组分结构特征及对其生物药剂学性质的研究仍相对薄弱, 有待进一步深入研究。
本研究基于数据驱动的研究模式, 对《中国药典》2020年版收载加辅料炮制中药饮片的化学成分性质与结构特征进行分析, 挖掘酒、醋、盐、蜜4种辅料在炮制应用中的关联特征组分, 并初步探讨炮制辅料对特征组分溶解度和渗透性的影响, 为中药辅料炮制的科学内涵研究提供理论和数据支撑。
数据来源  以“加辅料炮制”作为数据纳入标准, 在《中国药典》2020年版“药材和饮片”中检索得到中药饮片的名称、性味、归经等信息; 通过TCMSP[13] (https://old.tcmsp-e.com/tcmsp.php)、ETCM[14] (http://www.tcmip.cn/ETCM/)、TCMID[15] (http://47.100.169.139/tcmid/search/)3个数据库检索中药成分信息, 获得“中药饮片-中药成分”信息共11 061条, 查重合并, 包含中药饮片81种、中药成分5 954种, 均具有明确的2D结构; 在Pubchem中检索化学成分结构; 利用RDKit化学信息学分析平台(https://github.com/openforcefield/openff-toolkit)计算获得可表征中药成分结构特征的200个2D分子描述符, 包括分子组成描述符[分子质量(molecular weight, MolWt)、官能团数、环数、氢键供体/受体数等]、分子性质描述符[定量评估类药性(quantitative estimate of drug-likeness, QED)、油水分配系数(partition coefficient, LogP)、摩尔折射率等] 和拓扑指数[分子连接性指数(molecular connectivity index, MCI)、拓扑极性表面积(topological polar surface area, TPSA)、分子虚拟表面积(virtual surface area, VSA)等]; 进一步提取中药化学成分的环结构, 保留连接环结构最少的键(如果环结构上的原子直接连接双键, 则保留双键), 得到成分的Murcko骨架。
数据处理及特征组分解析  根据辅料类型, 分类统计加辅料炮制中药饮片的性味、归经、成分等信息(动物、矿物药不纳入分析), 并对所纳入中药饮片的性味进行统一, 如“微苦”、“微温”统一为“苦”、“温”; 对各辅料炮制中药饮片性味、归经进行频数分析; 使用SIMCA 14.1软件, 以200个2D分子描述符为变量, 对不同辅料炮制中药饮片的特有成分性质进行正交偏最小二乘判别分析(OPLS-DA), 并结合Murcko骨架分析挖掘特征组分构成特征; 骨架结构绘制均在ChemDraw软件上完成; 数据图表的绘制均在Origin 2021软件(美国Origin Lab公司)上完成。
药品和试剂  本研究使用的蜜为产自河北邢台的荆条蜜; 异黄酮组分(批号: 20200710, 纯度: 68.3%, 实验室自制); 刺芒柄花素(formononetin, FMN, 批号: H06S9Z69494, 纯度: ≥ 98%)、芒柄花苷(ononin, ON, 批号: R04J6F2, 纯度: ≥ 98%)、毛蕊异黄酮-7-O-β-D-葡萄糖苷(calycosin-7-O-β-D-glucoside, CAG, 批号: J19HB174062, 纯度: ≥ 98%)均购自上海源叶生物科技有限公司; 毛蕊异黄酮(calycosin, CA, 批号: PS0524-0020, 纯度: > 98%)购自普思生物科技股份有限公司; 蛋黄卵磷脂(批号: K22171056)、十二烷(批号: E2318821)购自上海阿拉丁生化科技有限公司; 0.45 μm Immobilon PVDF膜购自德国Merck公司; 色谱级乙腈(批号: 23065278)购自安徽天地高纯溶剂有限公司; 纯水为纯水仪自制, 其余试剂均为分析纯。
仪器设备  XPR2百万分之一电子天平(梅特勒-托利多国际股份有限公司); PX125DZH型十万分之一电子天平(奥豪斯仪器有限公司); Alliance E2695型高效液相色谱仪和2489 UV/Vis检测器(美国Waters公司); SVEATM C18 Gold色谱柱(5 μm, 4.6 mm × 250 mm, 瑞典Nanologica公司); Milli-Q超纯水系统(美国Millipore公司); CHA-S型数显气浴恒温振荡箱(金坛市双捷实验仪器厂); TPY-2药物透皮扩散试验仪(上海黄海药检有限公司); TG16A高速离心机(上海卢湘仪离心机仪器有限公司); PHS-3C雷磁pH计(上海仪分科学仪器有限公司)。
色谱条件  采用SVESTM C18 Gold (4.6 mm × 250 mm, 5 μm)色谱柱; 流动相A为纯水, 流动相B为乙腈, 梯度洗脱程序为: 0~2 min, 20%~23% B; 2~4 min, 23%~24% B; 4~5 min, 24% B; 5~8 min, 24%~25% B; 8~14 min, 25%~35% B; 14~18 min, 35%~55% B; 18~20 min, 55% B; 20~24 min, 55%~90% B; 流速: 1.0 mL·min-1; 柱温: 30 ℃; 检测波长: 254 nm; 进样量: 10 μL。
异黄酮组分溶解度测定  采用饱和摇瓶法测定纯水环境下炮制辅料蜜对异黄酮组分(CAG、ON、CA或FMN)平衡溶解度的影响。取异黄酮组分12 mg, 精密称定, 加纯水或蜜水溶液(蜜浓度为0.025 g·mL-1) 1 mL制备过饱和溶液, 涡旋2 min后置于37 ℃水浴中振摇24 h (120 r·min-1), 使之充分达到平衡, 取出样品放冷至室温, 13 000 r·min-1离心10 min, 精确吸取上清液0.2 mL, 加入0.8 mL甲醇, 13 000 r·min-1离心10 min, 取上清液, 测定成分含量(每组平行3份)。
异黄酮组分渗透性测定  采用平行人工膜渗透模型(PAMPA)探讨炮制辅料蜜对异黄酮组分体外渗透行为的影响, 课题组前期已证明该装置测量渗透性的可行性[16-18]。取异黄酮组分0.26 g, 精密称定, 加入PBS溶液或蜜-PBS溶液(蜜浓度为0.025 g·mL-1) 13 mL, 摇匀溶解, 制备上室药液。将PVDF膜夹在立式透皮扩散池上下两室中间以模拟生物膜, 膜上涂覆500 μL澄清4% (W/V) (蛋黄卵磷脂/十二烷)的人工膜溶液, 下室接收室中加8 mL空白接收液, 上室供给室中加2 mL药物溶液, 透皮仪设置温度为37 ℃、转速为300 r·min-1, 孵育24 h, 分别于2、4、6、8、12、24 h时吸取下室渗透药液0.3 mL, 并补充0.3 mL PBS溶液。将取出药液用适量甲醇稀释, 测定成分含量, 计算有效透过率(每组平行3份)。

Vd: 供给板液体加入量(2 mL); Va: 接收板液体加入量(8 mL); [drug]a: 时间为t时接收板药物浓度; [drug]e: 时间为t时的理论平衡浓度; [drug]d: 时间为t时供给板药物浓度。
统计方法  采用SPSS 26.0软件对结果进行t检验, 测量值以平均值±标准差($ \stackrel{-}{x} $ ± s)表示, P < 0.05具有统计学意义。
《中国药典》2020年版共收载炮制辅料14种, 涉及中药饮片96种, 除动物、矿物药外, 共收载由酒、醋、盐、蜜、姜汁等12种不同辅料炮制中药饮片81种, 其中约73%的中药饮片由酒、醋、盐、蜜这4种辅料炮制(图 1A), 故下文将聚焦于这4种炮制辅料进行探究。通过TCMSP、ETCM、TCMID 3个数据库检索获得加辅料炮制中药饮片成分信息11 061条, 查重合并, 酒、醋、盐、蜜4种辅料炮制中药饮片分别包含中药成分1 694、1 400、1 047、1 802个(图 1B)。
酒、醋、盐、蜜4种辅料炮制中药饮片性味、归经频数分析结果见图 2。在药性方面(图 2A), 加辅料炮制中药饮片主要为温性和寒性, 其中辅料酒偏向于炮制寒性中药饮片, 辅料醋、盐、蜜偏向于炮制温性中药饮片。在药味方面(图 2B), 中药饮片大多具有苦味, 经辅料酒、醋、盐、蜜炮制后, 可以辛制苦、以酸制苦、以咸制苦、以甘制苦, 避免药物过苦伤津耗液, 刺激胃肠道。此外, 辅料酒偏向于炮制甘味中药饮片, 辅料醋偏向于炮制辛味中药饮片, 辅料蜜偏向于炮制辛/甘味中药饮片, 而辅料盐的应用无明显偏向性。在归经方面(图 2C), 辅料酒偏向于炮制归肝/肾经中药饮片, 辅料醋偏向于炮制归肝/脾经中药饮片, 辅料盐偏向于炮制归肾经中药饮片, 辅料蜜偏向于炮制归肺经中药饮片, 这与炮制归经理论“醋制入肝”、“盐制入肾”[19-21]等基本吻合。
酒、醋、盐、蜜4种辅料炮制中药饮片化学成分复杂多样, 本研究以RDkit化学信息学分析平台计算获得表征成分性质的LogP、MCI、VSA等200个2D分子描述符为变量, 将4种辅料炮制中药饮片的特有成分两两比较, 进行正交偏最小二乘判别分析(OPLS-DA)。由得分图(图 3A)可见, 不同辅料炮制中药饮片的化学成分可被不同程度地区分开, 其中需酒炙与醋炙中药饮片化学成分的OPLS-DA分析模型的预测参数值(Q2)最大, 说明4种辅料炮制中药饮片的特有成分的整体性质存在差异, 其中需酒炙与醋炙中药饮片的成分性质差异最为明显(Q2 = 0.277), 在OPLS-DA模型得分特征空间中部分成分位置重叠, 但仍有特有成分距离较远。
进一步聚焦表征中药饮片中化学成分性质的4个典型分子描述符——MolWt、TPSA、LogP、QED, 探究4种辅料炮制中药饮片成分性质特点。由图可知(图 3B), 4种辅料炮制中药饮片中化学成分MolWt均主要分布在100~500之间; TPSA和LogP是评价药物溶解性质的重要参数[22], 其中需酒炙、盐炙、蜜炙中药饮片中化学成分的TPSA分布在0~100之间, 而需醋炙中药饮片中化学成分的TPSA主要分布在0~50之间(80%以上成分TPSA低于50), LogP基本呈正态分布, 需醋炙、蜜炙、酒炙、盐炙中药饮片中化学成分的LogP均值分别约为3.80、2.83、2.10、2.10, 说明与其他辅料相比, 需醋炙中药中疏水成分较多; QED是通过组合多个分子描述符来评估成分类药性的方法[23], 其取值范围为0~1, 值越大类药性越强, 4种辅料炮制中药饮片中类药性高的成分较少。
上述分析发现, 酒、醋、盐、蜜4种辅料炮制中药饮片的化学成分性质存在差异, 基于“结构相似性、性质相似性”, 本研究对4种辅料炮制中药饮片中化学成分结构特征进行研究。Murcko骨架可去除支链, 从化学成分分子结构中提取母核, 已运用于药物分子的形状、结构分析[24, 25]。通过成分Murcko骨架分析发现, 4种辅料炮制中药饮片中共筛选得到5 083种Murcko骨架, 除共有骨架外, 每种辅料炮制中药饮片中存在一些特有骨架(图 4A)。根据各骨架出现频数计算结果发现, 出现频数排名前50的Murcko骨架已基本包括中药常见成分骨架类型, 如黄酮骨架、皂苷骨架、生物碱骨架、香豆素骨架等(图 4B)。进一步以骨架占比≥ 60%为标准, 解析酒、醋、盐、蜜4种辅料炮制中药饮片的组分构成特征, 各骨架在4种辅料中占比见表 1。结果发现, 4种辅料炮制中药饮片中均具有占比较高的特征组分, 环烯醚萜苷、甾酮、苯乙醇苷、菲醌、蒽醌组分为需酒炙中药饮片中占比较高的特征组分, 木脂素、四环三萜、二萜、生物碱组分为需醋炙中药饮片中占比较高的特征组分, 苯丙酸组分为需盐炙中药饮片中占比较高的特征组分, 异黄酮、香豆素、联苄、紫檀烷组分为需蜜炙中药饮片中占比较高的特征组分。
根据炮制辅料关联的特征组分解析结果, 利用TPSA和LogP表征特征组分的溶解度和渗透性, 结果发现特征组分存在一定的生物药剂学性质缺陷(图 5)。黄芪为“蜜制补益”的典型代表, 异黄酮类成分(CAG、ON、CA或FMN)为黄芪发挥补中益气功效的小分子活性成分, 但其溶解度不佳、渗透性不高、生物利用度较低, 严重限制了其临床应用。因此, 本研究选择以炮制辅料蜜和黄芪异黄酮组分为例, 探讨炮制辅料对特征组分溶解度和渗透性的影响。结果显示, 炮制辅料蜜可增加黄芪异黄酮组分在水溶液中的平衡溶解度, 实现改善其溶解性的目的(图 6A)。此外, 基于PAMPA模型研究发现, 炮制辅料蜜对异黄酮组分中4种成分的体外渗透行为影响趋势一致, 即可提高4种成分的有效透过率, 使其12 h内有效透过率由29.85%~38.51%提升至37.26%~43.93% (图 6B)。综上, 炮制辅料蜜对4种异黄酮组分的溶解性和渗透性均具有一定改善作用。
炮制辅料是中药炮制的重要组成分部分, 其可通过改变药性、增效减毒等方式影响中药饮片的品质。目前, 酒、醋、盐、蜜4种炮制辅料在“酒制升提”、“醋制入肝”、“盐制入肾”、“蜜制补益”等传统中医药理论指导下成为使用频数最高的炮制辅料, 在中药炮制领域发挥重要作用, 但其机制解析仍存在挑战性。
本研究聚焦于《中国药典》2020年版收载由酒、醋、盐、蜜4种辅料炮制的中药饮片, 探究其药性特点与辅料应用规律, 发现其在中药饮片性味、归经上存在炮制应用的偏向性, 该偏向性也与传统中药炮制理论及炮制增效减毒的临床实践相吻合。作者进一步采用化学信息学技术计算获得表征成分性质和结构特征的200个2D分子描述符和Murcko骨架, 通过成分性质与Murcko骨架分析, 解析酒、醋、盐、蜜4种辅料炮制中药饮片的组分构成特征, 挖掘4种辅料关联的特征组分。经研究发现, 环烯醚萜苷、甾酮、苯乙醇苷、菲醌、蒽醌组分为需酒炙中药饮片的特征组分, 木脂素、四环三萜、二萜、生物碱组分为需醋炙中药饮片的特征组分, 苯丙酸组分为需盐炙中药饮片的特征组分, 异黄酮、香豆素、联苄、紫檀烷组分为需蜜炙中药饮片中的特征组分。目前, 已有研究从化学成分变化角度证实了4种辅料与特征组分之间的内在联系, 例如辅料醋炮制可以促进甘遂中具癸二烯酰基的巨大戟烷型二萜由顺式结构转变为反式结构, 发挥“减毒存效”的作用[26]。然而, 局限于体外化学成分质变和量变的研究, 往往忽略了中药炮制后药效物质在体内的转化过程是影响药效的关键因素[27], 无法全面阐述炮制机制, 甚至有时会出现炮制后成分增减与药效相驳的悖论, 因此, 本研究尝试从生物药剂学角度探究辅料炮制机制。异黄酮类成分为黄芪发挥补中益气功效的小分子活性成分[28-30], 但其生物利用度较低且炮制前后含量变化不明, 故本研究以炮制辅料蜜和黄芪异黄酮组分为例, 初步探讨炮制辅料对特征组分生物药剂学性质的影响, 结果发现辅料蜜可改善黄芪异黄酮组分的溶解度和渗透性, 提示基于辅料作用的生物药剂学过程研究是解析炮制减毒增效机制的关键突破口。
综上所述, 本研究采用化学信息学手段, 系统探究酒、醋、盐、蜜4种炮制辅料与化学成分性质和结构特征间的关联规律, 并以蜜炙黄芪为例, 解析炮制辅料对特征药效组分生药剂学分类性质的影响规律, 为辅料炮制科学内涵研究提供新视角和数据支撑。但本研究仅围绕药物溶解度和渗透性展开, 未来仍需聚焦体内过程, 深入探讨辅料蜜对黄芪异黄酮组分胃肠吸收的影响及其机制。
