Seven triterpenoids were isolated and purified from the 95% aqueous EtOH extract whole plants of P. villosa by various chromatographic techniques, such as silica gel, ODS, Sephadex LH-20 gel column chromatography and preparative high performance liquid chromatography. Based on physicochemical properties and spectral analyses, the structures of the seven compounds were identified as 29-acetoxyoleanolic acid-3-O-α-L-arabinopyranoside (1), oleanolic acid (2), 3β-hydroxy-24-norursa-4(23), 12, 20(30)-trien-28-oic acid (3), 3β-hydroxy-24-nor-urs-4(23), 12-dien-28-oic acid (4), ursolic acid (5), hederagenin (6), oleanolic acid 3-O-arabinoside (7). Compound 1 is a new compound. Compounds 3, 4, 6, and 7 are isolated from P. villosa for the first time. All compounds were assayed for their anti-inflammatory activity by the prodution of NO in lipopolysaccharide-stimulated RAW 264.7 cells. The results showed that compounds 1, 2, 3, 4, 6, and 7 significantly inhibited the NO release.

采用硅胶柱色谱、中压液相色谱、葡聚糖凝胶Sephadex LH-20柱色谱及制备液相色谱等多种色谱技术, 从白花败酱95%乙醇提取物中分离得到7个三萜类化合物。根据化合物的理化性质和波谱数据确定了它们的结构, 分别为29-乙酰氧基齐墩果酸-3-O-α-L-阿拉伯糖苷(1)、齐墩果酸(2)、3β-羟基-24-降-乌苏-4(23), 12(13), 20(30)-三烯-28-酸(3)、3β-羟基-24-降-乌苏-4(23), 12-二烯-28-酸(4)、熊果酸(5)、常春藤皂苷元(6)、齐墩果酸3-O-阿拉伯糖苷(7)。化合物1为新化合物, 化合物3467为首次从该植物中分离得到。通过对脂多糖(LPS) 诱导的RAW 264.7细胞(小鼠单核巨噬细胞) 释放的一氧化氮(NO) 影响实验测定了化合物的抗炎活性, 结果表明化合物123467有显著抑制NO作用。

采用硅胶柱色谱、中压液相色谱、葡聚糖凝胶Sephadex LH-20柱色谱及制备液相色谱等多种色谱技术, 从白花败酱95%乙醇提取物中分离得到7个三萜类化合物。根据化合物的理化性质和波谱数据确定了它们的结构, 分别为29-乙酰氧基齐墩果酸-3-O-α-L-阿拉伯糖苷(1)、齐墩果酸(2)、3β-羟基-24-降-乌苏-4(23), 12(13), 20(30)-三烯-28-酸(3)、3β-羟基-24-降-乌苏-4(23), 12-二烯-28-酸(4)、熊果酸(5)、常春藤皂苷元(6)、齐墩果酸3-O-阿拉伯糖苷(7)。化合物1为新化合物, 化合物3467为首次从该植物中分离得到。通过对脂多糖(LPS) 诱导的RAW 264.7细胞(小鼠单核巨噬细胞) 释放的一氧化氮(NO) 影响实验测定了化合物的抗炎活性, 结果表明化合物123467有显著抑制NO作用。

Seven triterpenoids were isolated and purified from the 95% aqueous EtOH extract whole plants of P. villosa by various chromatographic techniques, such as silica gel, ODS, Sephadex LH-20 gel column chromatography and preparative high performance liquid chromatography. Based on physicochemical properties and spectral analyses, the structures of the seven compounds were identified as 29-acetoxyoleanolic acid-3-O-α-L-arabinopyranoside (1), oleanolic acid (2), 3β-hydroxy-24-norursa-4(23), 12, 20(30)-trien-28-oic acid (3), 3β-hydroxy-24-nor-urs-4(23), 12-dien-28-oic acid (4), ursolic acid (5), hederagenin (6), oleanolic acid 3-O-arabinoside (7). Compound 1 is a new compound. Compounds 3, 4, 6, and 7 are isolated from P. villosa for the first time. All compounds were assayed for their anti-inflammatory activity by the prodution of NO in lipopolysaccharide-stimulated RAW 264.7 cells. The results showed that compounds 1, 2, 3, 4, 6, and 7 significantly inhibited the NO release.

白花败酱Patrinia villosa Juss.为败酱科败酱属多年生草本植物, 其全草作为败酱草基原植物之一, 被中国药典(2020版) 第四部收载。败酱草性辛、苦, 微寒, 属于清热解毒类中药, 具有清热解毒、排脓、活血化瘀等功效[1, 2]。败酱草作为我国传统的常用中药, 在我国华中、华南、华东及西南等地区广泛分布, 主产于江西、福建、湖南、四川、浙江等省; 在韩国、日本也有分布[3]。白花败酱草化学成分的研究表明该类药材中主要含有黄酮类、环烯醚萜类、苯丙素类、挥发油、酚酸类、三萜(皂苷) 类、脂肪酸类、甾醇类、及香豆素类等化合物[4]。课题组前期对其全草提取物进行了化学成分研究, 共分离得到了37个化合物, 包括8个环烯醚萜、1个单萜、6个木脂素、4个紫罗兰酮、2个黄酮、2个香豆素、4个甾醇、8个酚酸及2个其他类成分[5, 6]。为进一步对白花败酱全草的化学成分进行系统研究, 探讨其活性物质基础, 为白花败酱的开发利用提供依据, 本研究利用多种色谱分离技术对其三萜类成分进行了分离和鉴定, 共得到7个三萜类化合物。分别鉴定为: 29-乙酰氧基齐墩果酸-3-O-α-L-阿拉伯糖苷(1)、齐墩果酸(2)、3β-羟基-24-降-乌苏-4(23), 12(13), 20(30)-三烯-28-酸(3)、3β-羟基-24-降-乌苏-4(23), 12-二烯-28-酸(4)、熊果酸(5)、常春藤皂苷元(6)、齐墩果酸3-O-阿拉伯糖苷(7), 其中化合物1为新化合物, 化合物3467均为首次从该植物中分离得到。并通过体外细胞实验对分离得到的化合物进行了生物活性筛选。
化合物1  为白色结晶, 易溶于甲醇, [α]$ {}_{\mathrm{D}}^{20} $-139 (c 0.1, MeOH)。通过高分辨质谱(HRESIMS) 给出准分子离子峰为m/z 645.402 4 [M-H]- (计算值为645.400 8) 及1H NMR、13C NMR波谱数据可确定其分子式为C37H58O9, 不饱和度为9。红外图谱中发现3 381、2 932、1 716、1 456 cm-1处有吸收, 表明该化合物中有羟基、羰基及双键的存在。
1H NMR谱中显示一个烯烃质子信号δH 5.26 (1H, brs, H-12), 6个甲基单峰信号δH 1.15 (3H, s, H-27), 1.03 (3H, s, H-23), 0.99 (3H, s, H-30), 0.94 (3H, s, H-25), 0.84 (3H, s, H-24), 0.81 (3H, s, H-26)。13C NMR谱中共显示37个碳信号, 结合DEPT谱, 可知其中包括一个羧基碳δC 181.4 (C-28), 两个烯烃碳δC 144.8 (C-13), 124.1 (C-12), 一个连氧次甲基δC 90.6 (C-3), 一个连氧亚甲基δC 75.8 (C-29), 6个甲基δC 28.6 (C-23), 26.4 (C-27), 19.5 (C-30), 17.7 (C-26), 17.0 (C-24), 15.9 (C-25)。此外, 化合物11H NMR和13C NMR谱中还显示一个糖的端基信号δH 4.26 (1H, d, J = 6.7 Hz, H-1′); δC 107.2 (C-1′), 和一个乙酰基信号δH 2.04 (3H, s, H-2″); δC 173.0 (C-1″), 20.7 (C-2″)。以上波谱数据提示, 化合物1为含有1个5碳糖的三萜皂苷类化合物。综合化合物11H NMR、13C NMR、HMQC和HMBC谱信息, 对其碳氢信号进行了全归属, 见表 1。将化合物1的核磁数据与文献报道ciwujianoside D3的碱水解产物29-hydroxyoleanolic acid-3-O-α-L-arabinopyranoside[7]的核磁数据进行比较, 除了化合物1多了一个乙酰基, 以及C-29位碳信号从δC 73.8位移到δC 75.8外, 两者数据基本一致, 并表明乙酰基连接在C-29位上, 结合文献和端基氢耦合常数可知连接的糖结构为阿拉伯糖, 提示化合物1的结构为29-acetoxyoleanolic acid-3-O-α-L-arabinopyranoside。化合物1经酸水解及衍生化后HPLC分析结果表明, 化合物1中存在的单糖保留时间与α-L-阿拉伯糖对照品基本一致, 提示化合物1所连接的糖为α-L-阿拉伯糖。
HMBC谱中可以观察到H3-23, H3-24与C-3、C-4、C-5, H3-25与C-1、C-5、C-9、C-10, H3-26与C-7、C-8、C-9、C-14, H3-27与C-8、C-13、C-14、C-15, H3-30与C-19、C-20、C-21、C-29, H2-29与C-19、C-20、C-21、C-30、C-1″有相关信号, 其中H2-29与C-1′′的相关信号确认乙酰基连接在C-29位上。HMBC谱中尚可见糖端基质子H-1′与C-3有相关, 表明阿拉伯糖基与C-3相连。以上证实了上述化合物1的结构推断。
化合物1的NOESY谱中, 可观察到H-3与H-5和Me-23相关, H-5与H-3和H-9相关, 而H-3和H-5与Me-24无相关峰。H-3 (δ 3.12, dd, J = 11.6, 4.5 Hz) 与H2-2的偶合常数较大, 表明H-3处于环己烷椅式构象的直立键上, 为α取向[8]。故H-5, H-9和Me-23与H-3处于同一面为α取向, 而Me-24为β取向。齐墩果烷型三萜的D\E环为顺式稠合, H-18为β取向[8], NOESY谱中还可观察到, H-18β与Me-30和H-12; H-12与H-11β和H-18β的NOE相关, 而与Me-27, H2-29无NOE交叉峰。进一步尚可观察到H-11β与Me-25和Me-26的相关信号。以上表明, Me-25, 26和30为β取向, Me-27和H2-29为α取向。确定化合物1与经典齐墩果烷三萜化合物的相对构型一致[9] (图 12)。
综上所述, 化合物1的结构鉴定为29-乙酰氧基齐墩果酸-3-O-α-L-阿拉伯糖苷(29-acetoxyoleanolic acid-3-O-α-L-arabinopyranoside), 为一新的齐墩果酸型三萜化合物。
6个已知化合物分别鉴定为齐墩果酸(2)[10]、3β-羟基-24-降-乌苏-4(23), 12(13), 20(30)-三烯-28-酸(3)[11]、3β-羟基-24-降-乌苏-4(23), 12-二烯-28-酸(4)[12]、熊果酸(5)[13]、常春藤皂苷元(6)[14]、齐墩果酸3-O-阿拉伯糖苷(7)[15]。化合物3467为首次从该植物中分离得到。化合物结构见图 1
结果表明, 与模型组相比, 化合物1467能显著抑制LPS诱导RAW 264.7释放NO量, 其半数抑制浓度IC50分别为26.48、25.65、25.75、16.45 μmol·L-1; 化合物23对LPS诱导的RAW 264.7细胞释放的NO具有抑制作用, 半数抑制浓度IC50值分别为47.51、31.14 μmol·L-1; 阳性药氨基胍盐酸盐IC50值为14.25 μmol·L-1

白花败酱P. villosa全草于2018年8月采自安徽省岳西县包头乡, 并由上海中医药大学吴力宏教授鉴定。凭证标本BHBJ20180807保存于上海中医药大学中药研究所。
干燥的白花败酱全草15 kg, 粉碎成粗粉, 用95%乙醇室温渗漉提取, 减压浓缩得总浸膏约4 kg。将白花败酱总浸膏混悬于热水中, 依次用石油醚、二氯甲烷、乙酸乙酯、正丁醇萃取, 减压回收溶剂后得到不同极性的萃取部位。
二氯甲烷萃取部位316 g经200~300目硅胶层析色谱柱, 石油醚-乙酸乙酯系统(100∶1~0∶1) 不同比例梯度洗脱, TLC检测合并相同组分, 得到7个组分(Fr. 1~Fr. 7)。Fr. 3经硅胶色谱柱, 石油醚-乙酸乙酯(20∶1~0∶1) 梯度洗脱得到Fr. 3.1~Fr. 3.6, Fr. 3.3经过ODS柱色谱, 水-甲醇(3∶7~0∶1) 洗脱得到Fr. 3.3.1~Fr. 3.3.7, Fr. 3.3.6经Sephadex LH-20凝胶色谱柱甲醇洗脱和制备液相色谱(30%~95%) 梯度洗脱, 再经薄层色谱制备(二氯甲烷-乙酸乙酯20∶1) 纯化得到化合物2 (35 mg)。Fr.5经硅胶色谱柱, 二氯甲烷-甲醇系统(1∶0~0∶1) 梯度洗脱得到Fr. 5.1~Fr. 5.9, Fr. 5.3经ODS柱色谱, 水-甲醇(9∶1~0∶1) 洗脱得到Fr. 5.3.1~Fr. 5.3.10, Fr. 5.3.7和Fr.5.3.8再经Sephadex LH-20凝胶色谱柱(石油醚-二氯甲烷-甲醇3∶3∶1) 洗脱, 再经制备薄层色谱(二氯甲烷-乙酸乙酯1∶1) 纯化得到化合物3 (13 mg)、4 (15 mg)、5 (2 mg)。
乙酸乙酯萃取部位110 g经硅胶柱色谱, 石油醚-乙酸乙酯系统(100∶1~0∶1) 梯度洗脱, 得到6个组分(Fr. I~Fr. Ⅵ)。Fr. VI经MCI柱, 水-甲醇(4∶1~0∶1) 洗脱得到Fr. VI.I~Fr. VI.V, Fr. VI.IV分别经中压柱色谱(水-甲醇系统9∶1~0∶1) 洗脱, 凝胶色谱柱甲醇反复洗脱, 制备液相(20%~90%乙腈) 梯度洗脱分离纯化得到化合物6 (8 mg)、7 (67 mg)、1 (33 mg)。
精密称取α-L-阿拉伯糖标准品10 mg于10 mL量瓶中, 加入蒸馏水定容配置成1 mg·mL-1的对照品溶液; 精密称取化合物5 mg于2 mL量瓶中加入50%甲醇配置成2.5 mg·mL-1的溶液。取标准品溶液与样品溶液1 mL加入顶空瓶中, 加入0.4 mol·L-1三氟乙酸溶液1 mL, 混匀, 封口, 放入烘箱中110 ℃反应3 h后, 反复加入甲醇减压干燥至残留中无酸味, 将残渣溶解于200 μL蒸馏水中, 溶液转移至新的EP管中12 000 r·min-1离心5 min, 取上清100 μL至新的EP管中, 加入0.5 mol·L-1 NaOH溶液100 μL, 混匀后加入0.5 mol·L-1 1-苯基-3-甲基-5-吡唑啉酮(PMP) 甲醇溶液150 μL, 混匀后70 ℃加热1 h取出, 加入100 μL 0.5 mol·L-1 HCl溶液中和, 补加蒸馏水至1 mL加入等量三氯甲烷, 振摇1 min, 吸弃氯仿层, 继续重复萃取3次以除去多余的PMP, 最后过0.22 μm微孔膜, 进行HPLC分析。色谱分析条件: Thermo Ultimate 3000高效液相色谱仪(HPLC) 与Thermo U3000二极管阵列检测器(DAD) 联合使用, 液相色谱柱为YMC-Pack ODS-A柱(250 mm × 4.6 mm, 5 μm); 流动相为磷酸二氢钾缓冲液(0.1 mol·L-1 KH2PO4, 0.07 mol·L-1 NaOH) 和乙腈(17∶83), 等度洗脱, 流速0.8 mL·min-1; 柱温40 ℃; 进样量5 μL, 检测波长254 nm, 运行时间100 min。在对照品色谱图中, α-L-阿拉伯糖的保留时间为73.5 min; 化合物1色谱图单糖的保留时间为73.6 min, 与α-L-阿拉伯糖对照品的色谱峰保留时间基本一致。
本实验对分离得到的化合物通过对LPS诱导小鼠巨噬细胞RAW 264.7产生NO抑制作用来评价其抗炎活性。取生长状态良好且处于对数生长期的RAW 264.7细胞, 用DMEM完全培养基配制成单细胞悬液, 细胞计数, 以1×105个·mL-1, 每孔100 µL均匀接种于96孔板中, 于37 ℃、5% CO2细胞培养箱中培养24 h。设置空白组(仅DMEM培养基)、模型组(1 µg·mL-1 LPS)、实验组[1 µg·mL-1 LPS + (1、10、20、40、80和100 µmol·L-1化合物)], 每组3个复孔。培养24 h, 取其上清液于离心管中, 采用Griess试剂盒操作方法, 检测细胞培养液中NO的含量[16]。采用一氧化氮合酶(NOS) 抑制剂氨基胍盐酸盐作为阳性药对照。同时, 通过CCK-8试验评估RAW 264.7细胞的活力, 以排除化合物的细胞毒性干扰。用酶标仪在540 nm测定各孔吸光值(OD值), 计算抑制率和IC50值。NO抑制率(%) = (模型组OD值-实验组OD值)/(OD模型组-OD空白组) × 100%。
