GNPS-based mass spectrum-molecular networks is an effective strategy for rapidly identifying known natural products and discovering novel structures. The chemical diversity of azaphilones from the fermentation extracts of Talaromyces sp. HK1-18 was studied by molecular network technique. Three linear tricyclic azaphilones, sequoiamonacins A-C (2a, 2b, 1), were isolated by silica gel column chromatography and high performance liquid chromatography from the extracts of the fungal strain of HK1-18, and their structures were identified by nuclear magnetic resonance and high-resolution mass spectrometry. Guided by the mass spectra of sequoiamonacins A-C (2a, 2b, 1), the cluster of sequoiamonacinoid analogues was discovered from the full molecular networking of HK1-18. By analyzing the MS/MS fragments of each parent ion in this cluster, 7 azaphilones (3-9) included 6 new ones (4-9) were predicted successfully. Then the MS/MS cracking regularity of this type of azaphilones was revealed. Compound 1 showed anti-inflammatory activity, which can inhibit the production of interleukin-1α (IL-1α) in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7, with an inhibitory rate of 29% at the concentration of 12.5 μg·mL-1.

基于GNPS的质谱-分子网络策略是快速识别已知天然产物和发现新颖化合物的有效方法。本文利用分子网络技术研究了海洋真菌Talaromyces sp. HK1-18发酵产物中的嗜氮酮类化合物的化学多样性。综合采用硅胶柱色谱和高效液相色谱等色谱分离技术以及核磁共振和高分辨质谱等波谱解析技术从真菌HK1-18发酵产物中分离并鉴定了3个线型三环嗜氮酮化合物sequoiamonacins A~C (2a2b1); 继而以其质谱为指导, 从该真菌的全分子网络中提取了sequoiamonacin类嗜氮酮化合物的分子聚簇; 通过解析该聚簇中各母离子的MS/MS (二级质谱) 碎片, 成功预测了7个嗜氮酮化合物的化学结构(3~9), 其中sequoiamonacins E~J (4~9) 为新化合物, 并揭示了该类化合物的MS/MS裂解规律。化合物1具有一定的抗炎活性, 其能抑制脂多糖(LPS) 诱导的小鼠巨噬细胞RAW264.7中白细胞介素1α (IL-1α) 的产生, 质量浓度为12.5 μg·mL-1时, 抑制率达29%。

基于GNPS的质谱-分子网络策略是快速识别已知天然产物和发现新颖化合物的有效方法。本文利用分子网络技术研究了海洋真菌Talaromyces sp. HK1-18发酵产物中的嗜氮酮类化合物的化学多样性。综合采用硅胶柱色谱和高效液相色谱等色谱分离技术以及核磁共振和高分辨质谱等波谱解析技术从真菌HK1-18发酵产物中分离并鉴定了3个线型三环嗜氮酮化合物sequoiamonacins A~C (2a2b1); 继而以其质谱为指导, 从该真菌的全分子网络中提取了sequoiamonacin类嗜氮酮化合物的分子聚簇; 通过解析该聚簇中各母离子的MS/MS (二级质谱) 碎片, 成功预测了7个嗜氮酮化合物的化学结构(3~9), 其中sequoiamonacins E~J (4~9) 为新化合物, 并揭示了该类化合物的MS/MS裂解规律。化合物1具有一定的抗炎活性, 其能抑制脂多糖(LPS) 诱导的小鼠巨噬细胞RAW264.7中白细胞介素1α (IL-1α) 的产生, 质量浓度为12.5 μg·mL-1时, 抑制率达29%。

GNPS-based mass spectrum-molecular networks is an effective strategy for rapidly identifying known natural products and discovering novel structures. The chemical diversity of azaphilones from the fermentation extracts of Talaromyces sp. HK1-18 was studied by molecular network technique. Three linear tricyclic azaphilones, sequoiamonacins A-C (2a, 2b, 1), were isolated by silica gel column chromatography and high performance liquid chromatography from the extracts of the fungal strain of HK1-18, and their structures were identified by nuclear magnetic resonance and high-resolution mass spectrometry. Guided by the mass spectra of sequoiamonacins A-C (2a, 2b, 1), the cluster of sequoiamonacinoid analogues was discovered from the full molecular networking of HK1-18. By analyzing the MS/MS fragments of each parent ion in this cluster, 7 azaphilones (3-9) included 6 new ones (4-9) were predicted successfully. Then the MS/MS cracking regularity of this type of azaphilones was revealed. Compound 1 showed anti-inflammatory activity, which can inhibit the production of interleukin-1α (IL-1α) in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7, with an inhibitory rate of 29% at the concentration of 12.5 μg·mL-1.

海洋天然产物由于其结构多样性和强生物活性受到广泛关注, 是发现新型药物分子的重要来源之一; 在过去的几十年里, 从海洋天然产物中已发现了大量的药物先导物[1-4]。然而, 随着已知化合物的重复分离率逐年增加, 结构新颖的活性天然产物的高效发掘变得极具挑战性[5]。基于全球天然产物社会分子网络(global natural products social molecular networking, GNPS) 的分子网络技术是实现天然产物去重复和新颖化合物快速发现的一种有效策略[6-9]。由于结构相似的分子具有相似的MS/MS (二级质谱) 碎片模式, GNPS-分子网络通过计算MS/MS碎片的相似性, 从而实现相似分子的聚类和可视化。如果分子聚簇中一个节点的化学结构被阐明, 那么其他节点可以基于质量差和特征离子碎片解析进行结构预测, 从而实现新颖化合物的高效发掘。
嗜氮酮(azaphilone) 是一类以高度氧化的吡喃-醌双环骨架为核心的聚酮类真菌色素, 具有多种生物活性, 如抗菌、抗病毒、抗肿瘤、抗炎等; 多由子囊菌和担子菌纲的真菌产生, 包括曲霉属(Aspergillus)、青霉属(Penicillium)、毛壳菌属(Chaetomium)、蓝状菌属(Talaromyces) 等, 是国内外天然药物化学研究的热点[10]。本课题组长期从事特殊生境微生物活性次级代谢产物研究, 也发现了一系列具有显著抗MRSA (耐甲氧西林金葡菌) 活性的新型嗜氮酮化合物[11, 12]。本文对一株海洋真菌Talaromyces sp. HK1-18的次级代谢产物展开研究, 又分离获得了一类具有线型三环骨架的嗜氮酮类化合物sequoiamonacins A~C (2a2b1), 并利用基于GNPS的分子网络技术对三环嗜氮酮类化合物的化学多样性进行挖掘, 从嗜氮酮分子簇中成功预测了7个化合物的化学结构(3~9), 其中4~9是新化合物。下文对上述嗜氮酮类化合物的分离、结构鉴定、抗炎活性、分子网络以及MS/MS裂解途径进行详细报道。
化合物1是黄色油状物。其核磁共振氢谱(1H NMR) 中, 出现了一组反式烯氢信号δH 7.06 (d, J = 15.6 Hz), 6.24 (d, J = 15.6 Hz), 一个烯氢信号δH 6.12 (s), 两个化学不等价的连氧亚甲基信号δH 4.90 (d, J = 13.2 Hz), 4.81 (d, J = 13.2 Hz), 两个甲基信号δH 1.34 (s), 0.87 (t, J = 6.6 Hz); 其碳谱(13C NMR) 中显示有三个羰基碳信号δC 191.4, 173.8, 170.2, 六个烯族碳信号δC 156.6, 150.2, 135.1, 124.1, 118.3, 111.1, 三个连氧碳信号δC 82.2, 67.1, 63.3, 两个甲基碳信号δC 16.9, 14.0。该化合物的以上氢谱、碳谱特征与文献[13]报道极为相似, 因此鉴定化合物1为具有线型三环结构的嗜氮酮化合物sequoiamonacin C, 是一种天然黄色色素[14]。化合物1的NMR和MS数据如下: 1H NMR (600 MHz, CDCl3) δ 7.06 (1H, d, J = 15.6 Hz, H-3), 6.24 (1H, d, J = 15.6 Hz, H-2), 6.12 (1H, s, H-5), 4.90 (1H, d, J = 13.2 Hz, Ha-14), 4.81 (1H, d, J = 13.2 Hz, Hb-14), 3.71 (1H, brd, J = 9.0 Hz, H-15), 3.04 (1H, m, H-9), 2.80 (1H, td, J = 10.8, 3.6 Hz, H-8), 2.67 (1H, dd, J = 18.0, 4.2 Hz, Ha-7), 2.61 (1H, dd, J = 18.0, 11.4 Hz, Hb-7), 1.74 (1H, m, Ha-16), 1.44 (3H, m, Hb-16, H-17), 1.34 (3H, s, H-23), 1.26 (8H, brs, H-18~H-21), 0.87 (3H, t, J = 7.2 Hz, H-22); 13C NMR (150 MHz, CDCl3) δ 191.4 (C, C-12), 173.8 (C, C-10), 170.2 (C, C-1), 156.6 (C, C-4), 150.2 (C, C-6), 135.1 (CH, C-3), 124.1 (CH, C-2), 118.3 (C, C-13), 111.1 (CH, C-5), 82.2 (C, C-11), 67.1 (CH, C-15), 63.3 (CH2, C-14), 47.5 (CH, C-9), 42.6 (CH, C-8), 33.2 (CH2, C-16), 31.3 (CH2, C-20), 28.9 (CH2, C-7), 28.7 (CH2, C-19), 28.7 (CH2, C-18), 25.9 (CH2, C-17), 22.1 (CH2, C-21), 16.9 (CH3, C-23), 14.0 (CH3, C-22); HRESIMS m/z 417.194 4 ([M-H], calcd for C23H29O7 417.191 9)。
化合物2a/2b为黄色粉末, 是C-4构型相反的一对差向异构体。采用多种色谱分离方法对2a/2b进行分离, 结果均难以拆分。其氢谱和碳谱中呈现出两套强度接近的核磁信号, 大部分信号完全重叠, 只有少数靠近C-4位置的信号有细微差异。同时也与化合物1的氢谱和碳谱数据相似, 与化合物1显著不同之处在于, 化合物2a/2b的氢谱中均呈现出一组顺式烯氢信号δH 7.69 (d, J = 5.4 Hz), 6.46 (d, J = 5.4 Hz), 同时少了一个单峰烯族氢信号而多出一组亚甲基信号。基于以上信号特征, 进一步结合文献[13, 15]鉴定化合物2a/2b为sequoiamonacins A/B。化合物2a/2b谱图的其中一组NMR数据归属如下: 1H NMR (600 MHz, DMSO-d6) δ 7.69 (1H, d, J = 5.4 Hz, H-3), 6.46 (1H, d, J = 5.4 Hz, H-2), 4.45 (1H, d, J = 15.6 Hz, Ha-14), 4.27 (1H, d, J = 15.6 Hz, Hb-14), 3.70 (1H, m, H-15), 3.02 (1H, m, H-9), 3.01 (1H, m, Ha-5), 2.82 (1H, m, H-8), 2.55~2.65 (3H, m, H-7, Hb-5), 1.74 (1H, m, Ha-16), 1.44 (3H, m, Hb-16, H-17), 1.38 (3H, s, H-23), 1.26 (8H, brs, H-18~H-21), 0.87 (3H, t, J = 7.2 Hz, H-22); 13C NMR (150 MHz, DMSO-d6) δ 191.8 (C, C-12), 173.6 (C, C-10), 169.8 (C, C-1), 154.5 (CH, C-3), 150.3 (C, C-6), 123.4 (CH, C-2), 125.8 (C, C-13), 82.2 (C, C-11), 67.1 (CH, C-15), 60.4 (CH2, C-14), 47.2 (CH, C-9), 42.5 (CH, C-8), 33.2 (CH2, C-16), 31.3 (CH2, C-7), 31.2 (CH2, C-20), 28.9 (CH2, C-19), 28.7 (CH2, C-18), 25.9 (CH2, C-17), 22.1 (CH2, C-21), 16.5 (CH3, C-23), 14.0 (CH3, C-22)。
Sequoiamonacins A~C具有对卤虫(brine shrimp) 的致死作用[13]。本研究采用联科生物Mouse IL-1α ELISA Kit检测试剂盒(酶联免疫吸附法) 测定了化合物的抗炎活性, 选用的细胞种类是小鼠巨噬细胞RAW264.7。结果表明(表 1), sequoiamonacin C (1) 表现出一定的抗炎活性, 其能抑制脂多糖(LPS) 诱导的小鼠巨噬细胞RAW264.7中白细胞介素1α (IL-1α) 的产生, 当质量浓度为12.5 μg·mL-1时, 抑制率达29%, 而阳性对照药地塞米松(DEX) 的抑制率为35%。
从蓝状菌属真菌HK1-18中分离发现的sequoiamonacins A~C (2a2b1) 是一类较为少见的具有线型三环骨架的嗜氮酮化合物[16], 结构相似的化合物还包括从其他蓝状菌属真菌中发现的talaromyacins A~C[15]以及具有抗结肠癌活性的talaroilones A、B[17]。该类线型三环嗜氮酮化合物的化学多样性值得进一步挖掘。本文采用分子网络技术对真菌HK1-18发酵产物中的sequoiamonacins A~C类似物进行了结构预测。
真菌HK1-18发酵产物所形成的全分子网络如图 1A所示。该分子网络中共含有300个节点, 节点数≥ 3个的分子簇有18个。经与GNPS数据库检索匹配, 可知部分分子簇包含甾醇、脂肪酸、联苯醚、吡喃酮及生物碱等化合物, 未发现嗜氮酮化合物的单独分子簇。Sequoiamonacins A~C的低分辨质谱均为m/z 419 [M+H]+, 而分子网络中有两个直接相连的节点均显示为m/z 419; 分别提取其MS/MS图, 发现两者谱图高度相似, 主要特征信号均为m/z 401, 375, 357, 对应于sequoiamonacins A~C分子的脱水、脱羧等裂解过程, 表明这两个节点分别代表C-4侧链开环的化合物1和C-4侧链成环的化合物2a/2b, 但无法明确指定。
以两个m/z 419节点为中心, 截取了与之形成聚簇的m/z > 400的局部分子网络(图 1B), 共获得13个节点, 其中包括直接相连的MS/MS谱图高度相似的两个m/z 435节点以及通过m/z 461间接相连的MS/MS谱图相似的两个m/z 449节点。这两对节点均为C-4侧链开环和成环分别形成的两对化合物。而相距较远的两个m/z 433节点的MS/MS谱图有显著差异, 推测两者结构差异较大。此外, 节点m/z 441为[418+Na]+信号, 而m/z 457则为[434+Na]+信号。因此, 以sequoiamonacins A~C结构为指导, 通过仔细分析图 1中各节点的MS/MS谱图, 以及参考相似的嗜氮酮类化合物结构及其生物合成途径报道[10], 从分子簇B中共预测了如图所示的7个嗜氮酮结构, 除化合物3为已报道的talaroilone A[17], 其余(4~9) 均为新化合物, 被依次命名为sequoiamonacins E~J。值得注意的是, 化合物3~9也可能为C-4侧链成环结构, 本文统一以开链结构作为代表。
仔细剖析化合物1~9的MS/MS特征碎片离子(表 2), 发现该类嗜氮酮化合物的破碎途径呈现出规律性: 首先均从羧基端开始脱水[M-H2O+H]+、脱羧[M-CO2+H]+, 对应于质谱损失18 Da或44 Da; 然后C-15-OH发生脱氢或脱水; 最后连接于C-4和C-9的两个侧链分别氧化断裂, 形成三环嗜氮酮母核碎片。图 2推测了化合物3的MS/MS裂解途径, 代表了此类化合物的裂解规律。此外, 化合物56均出现了特有的脱乙酰基碎片峰(分别对应于m/z 433和m/z 417)。化合物78虽然分子量相同, MS/MS中也同时出现了m/z 415和m/z 389信号, 但是化合物7还出现了特有的“脱水+脱甲基”信号(m/z 401) 以及“脱羧+脱水”信号(m/z 357), 表明化合物7的C-15-OH被甲基化; 而化合物8则出现了特有的“脱水+脱氢”信号(m/z 413) 以及“脱羧+脱水”信号(m/z 371), 结合嗜氮酮类化合物C-9侧链结构特征[10, 13], 推测化合物8的C-16位连接了一个甲基。化合物9的MS/MS中出现了多出一个氧的三环母核信号(m/z 267), 表明其母核上多出一个羟基, 并且最有可能连接于C-7[10]。这些化合物的MS/MS碎片特征为化合物结构预测提供了依据。
本文将分子网络策略成功应用于真菌的新颖次级代谢产物挖掘, 从海洋真菌Talaromyces sp. HK1-18发酵产物所形成的分子网络中提取了线型三环嗜氮酮sequoiamonacin类似物的分子聚簇, 并以分离鉴定的sequoiamonacins A~C结构为基础, 从该分子簇中成功预测了7个嗜氮酮化合物的结构(3~9), 其中4~9为新化合物。该类化合物的MS/MS裂解呈现出规律性, 均出现脱水、脱羧、脱氢、氧化断链等特征碎片, 最终保留三环嗜氮酮母核。Sequoiamonacin C (1) 具有抗炎活性。本文为新颖嗜氮酮化合物的高效发掘提供参考, 为后续新化合物的靶向分离提供依据。真菌HK1-18的PDB培养基发酵产物中的嗜氮酮类化合物产量较低, 虽然已成功分离获得了sequoiamonacins A~C, 但新化合物4~9由于产量太低无法分离。本课题组将继续优化真菌HK1-18的培养条件, 提高嗜氮酮类化合物的产量, 以期成功分离获得其中的新化合物, 为该类化合物的抗炎活性评价以及构效关系研究提供基础。
高分辨质谱仪Micromass Q-TOF (美国Waters公司); 离子阱液质联用仪Bruker amazon SL和核磁共振仪AVANCE 600/400 (德国BRUKER公司); WTF-203B暗箱式紫外分析仪(上海精科实业有限公司); QYC-2112B双层摇床(上海沪粤明科学仪器有限公司); L-2000高效液相色谱仪HPLC采用C18柱(Kromasil, 250 mm × 10 mm, 5 μm) (日本Hitachi公司); R-210旋转蒸发仪(日本Buchi公司); 硅胶薄层色谱板(烟台江友硅胶开发有限公司); 柱色谱硅胶200~300目(青岛化工厂); 凝胶Sephadex LH-20 (美国Amersham Biosciences公司); 反相硅胶ODS (美国Unicorn公司); 液相所用色谱级甲醇(天津四友精细化学品有限公司, 生产批号: B2309201); 提取和分离所用甲醇(生产批号: P2774893)、乙酸乙酯(生产批号: P2387644)、二氯甲烷(生产批号: P2387644)、石油醚(生产批号: P2152622) 等均为分析纯产品(国药集团化学试剂有限公司)。
海洋真菌HK1-18分离于海南东寨港红树林自然保护区的海桑根际土壤样品, 经分子生物学及形态学鉴定为Talaromyces sp., 其ITS-rDNA序列已经提交至Gen-Bank, 登录号为OP626088。该菌株现保存于扬州大学环境科学与工程学院海洋科学与技术研究所。
发酵: 采用PDB培养基(每1 L人工海水中加入20 g葡萄糖, 200 g土豆浸出液) 进行发酵, 每1 L锥形瓶中加400 mL培养基, 共发酵10 L, 室温静置培养30天后结束发酵。
提取: 利用纱布将菌体和发酵液分离, 其中发酵液用等体积的乙酸乙酯萃取5次, 减压浓缩后得到粗浸膏14.2 g。
分离: 上述提取物采用减压硅胶柱色谱, 以EtOAc-石油醚(0~100%)、MeOH-EtOAc (0~100%) 为溶剂进行梯度洗脱, 分为11个组分(Fr.1~Fr.11), 其中Fr.4经TLC分析显示主要包含亮黄色斑点, 经正相硅胶柱梯度洗脱(EtOAc-石油醚, 20%~100%) 分离获得Fr.4-1和Fr.4-2。Fr.4-1经Sephadex LH-20柱色谱(MeOH-CH2Cl2, 1∶1) 以及HPLC (60% MeOH-H2O) 分离获得化合物1 (5.6 mg, tR = 28.5 min)。Fr.4-2经Sephadex LH-20柱色谱(MeOH-CH2Cl2, 1∶1) 以及ODS柱色谱(50% MeOH-H2O) 分离得到 2a/2b (10.3 mg), 化合物2a/2b经HPLC分析(55% MeOH-H2O) 为单一色谱峰(tR = 45.0 min)。
首先通过细胞活力评价确定化合物的抗炎活性测试质量浓度为12.5 μg·mL-1。然后采用联科生物Mouse IL-1α ELISA Kit检测试剂盒(酶联免疫吸附法) 测定化合物的抗炎活性, 选用的细胞是小鼠巨噬细胞RAW264.7, DMSO作为空白对照, 脂多糖作为阴性对照, 地塞米松作为阳性对照, 每组设三个平行实验, 经过孵育、洗涤、显色后, 用酶标仪测试在450 nm处的吸光度, 获得化合物的炎症因子IL-1α浓度。各处理方法均包含三组平行实验, 所获得的数据使用IBM SPSS Statistics 19软件分析。利用最小显著差数法(least significant difference, LSD) 对于方差显著的因素进行多重比较。当P < 0.01时表明各分析指标有差异显著性。抗炎抑制率计算公式为抑制率= (1 - 化合物IL-1α浓度/ 脂多糖IL-1α浓度) × 100%。
采用液质联用仪对提取物进行检测。色谱柱为YMC-Park-C18 (250 mm × 4.6 mm, 5 μm)。流动相为MeOH-H2O, 流速0.8 mL·min-1, 梯度洗脱条件如下: 0~40 min, 10%~90% MeOH-H2O; 40~45 min, 90%~100% MeOH-H2O; 45~50 min, 100% MeOH-H2O。进样量为8 μL。质谱采用正离子模式, 离子源为ESI源, 扫描范围为100~2 000 Da。
将mzXML格式的质谱数据上传至GNPS分子网络平台构建分子网络。具体参数设置为: 前体离子质量误差值1.0 Da, 碎片离子质量误差值0.3 Da, 匹配的MS/MS碎片峰≥ 4, 节点之间相关余弦值≥ 0.6。最终生成的质谱分子网络图借助Cytoscape软件实现可视化。
