Electronic tongue is one kind of bionic detection technologies, which can objectively reflect the taste of drugs based on electrochemical principle. In this paper, the development histories of electronic tongue both of potential type and voltammetry type were introduced, including their detection principles and key innovation technologies. In order to comprehensively improve the understanding of electronic tongue, its technological progresses, such as the study of dedicated sensors or biosensors for specific tastes, and the development of miniaturized or hybrid devices, were also discussed in detail. And the challenges and countermeasures in the application of electronic tongue were analyzed to provide some suggestions for its further technology promotion.

Electronic tongue is one kind of bionic detection technologies, which can objectively reflect the taste of drugs based on electrochemical principle. In this paper, the development histories of electronic tongue both of potential type and voltammetry type were introduced, including their detection principles and key innovation technologies. In order to comprehensively improve the understanding of electronic tongue, its technological progresses, such as the study of dedicated sensors or biosensors for specific tastes, and the development of miniaturized or hybrid devices, were also discussed in detail. And the challenges and countermeasures in the application of electronic tongue were analyzed to provide some suggestions for its further technology promotion.

在口服制剂的开发过程中, 口感评价是一项重要的研究内容。目前, 国内外常用的口服制剂口感评价方法有电子舌(electronic tongue) 和溶出度实验等体外评价方法、人尝法和动物味觉喜好实验等体内评价方法[1]。其中, 电子舌是近年来最常用且安全的口感评价技术[2]
“电子舌”这一概念, 最早源于二十世纪八九十年代仿生学技术的兴起。1989年, 日本九州大学Kiyoshi Toko教授设想通过一种传感器感知信息反映样品的味觉值, 开启了味觉传感器的研发; 1993年, Toko教授团队推出了世界上第一台味觉分析仪, 即电子舌的前身。1995年, 圣彼得堡大学Andrey Legin教授团队以表征人舌的味觉识别功能作为味觉分析仪的目标, 类比电子鼻(electronic nose, 始于1982年) 的概念, 提出了“电子舌”这一形象化的名词[3, 4]。2005年, 基于交互响应的液敏传感器为主流的电子舌仪器, 国际纯粹与应用化学联合会(International Union of Pure and Applied Chemistry, IUPAC) 将电子舌定义为[5]: 一种低选择、非特异性的交互敏感传感器阵列, 采用适当的模式识别方式或多元统计方法, 进行定性定量分析的现代化分析检测仪器。但随着味觉传感器技术的不断革新, 现在对电子舌的定义具有了更宽泛的含义: 一种在某种程度上可以表征或代替人舌对样品进行味觉指标定性或定量分析的检测仪器。
电子舌模拟生物味觉工作机制对复杂液体中多种物质进行有效测量和表征。根据传感器工作方式不同, 可分为电位型、伏安型、阻抗谱型、光寻址型以及物理型电子舌[6]。目前商品化的电子舌仅有电位型和伏安型两大类, 因此本文主要对这两大类电子舌的发展简史、新型电子舌技术动态做详细介绍, 以增进相关研究人员对电子舌技术的了解, 为药品口感评价提供技术支撑。
目前已经商品化的电子舌仪器主要来自日本、法国和中国, 其中日本和法国的电子舌技术为电位型, 而中国的电子舌技术为伏安型。
20世纪80年代, 日本九州大学Kiyoshi Toko等[7]研究发现不同化学成分修饰的脂质膜会与溶液状态下的呈味物质发生有差异化的静电或疏水作用, 产生类似于味蕾受体细胞上生物膜与溶于唾液的呈味物质之间发生的味觉反应, 由此逐步开展以脂质膜为主体、味觉值测量为目的的传感器研制。
1990年, Toko教授团队开始与Intelligent Sensor Technology (INSENT) 公司Ikezaki Hidekazu合作研究, 于1993年成功研制出第一台具有实用价值的味觉分析产品——Taste Sensor (TSS) SA401, 开启了味觉传感器的研究工程。SA401由8根附有不同脂质/聚合物敏感膜的电极(图 1)组成传感器阵列, 每个传感器由银/氯化银(Ag/AgCl) 电极芯、内部电解液(100 mmol·L-1氯化钾溶液)、敏感膜形成稳定的内部回路; 传感器分为正极传感器和负极传感器, 即分别在其电极芯上通正、负电使得敏感膜表面带正电或负电; 当测试样品溶液时, 不同敏感膜上面的脂质材料与溶液界面中的不同分子或离子发生疏水作用或离子交换反应, 改变敏感膜内外两相原有的电荷分布而产生电位差, 该电位差与样品溶液中不同离子浓度存在相关性, 以此通过组合响应将样品味道的整体差异区分出来, 同时还可以考察由呈味物质的数量变化而引起的味道变化[8]
2000年, Toko教授团队开创性推出了具有选择性的传感器, 即不断优化传感器双层脂质膜的成分、配比、极性和表面电荷密度等以实现某一根传感器仅对某一类呈味物质产生特异响应, 由此构建了酸、甜、苦、咸、鲜、涩这些基本味道的专属味觉传感器; 每种味觉传感器只检测对应的味觉指标, 可以独立使用和更换。另外, 为了让传感器响应与人舌感受相匹配, 开发以下内容: ①依据人舌阈值浓度范围, 如天然苦味物质0.01~0.1 mmol·L-1、天然糖20~1 000 mmol·L-1, 进行传感器响应信号的线性区域筛选, 以提高味觉传感器的特异选择性; ②依据韦伯费希纳定律, 当味物质浓度每增加20%时人舌可以感受到味道差异, 把这20%的增量定义为一个味觉单位, 则人舌一般可感知味觉差异的浓度范围, 即阈值的1~100倍(1.20~1.225) 可表征为0~25个味觉单位, 其中“阈值”为人舌感受到某种呈味物质的味道所需要的最低浓度; ③选定代表性呈味物质, 根据传感器线性响应区域, 确定测量浓度最低值、最高值(若最高值为最低值的N倍, 则1.2X = N, X为上述浓度范围下的味觉单位数), 测量对应浓度的响应值V1V2, 则该传感器味觉值的转换系数为K = X/(V2-V1), 可根据转换系数K和实际样品响应值V3直接计算得到样品味觉值T = T0+K×V3, 但初始味觉值T0需通过大量人尝结果拟合得到。上述味觉转换算法, 使得该传感器阵列与人舌的感知阈值和趋势具有较好的一致性, 不仅可以检测样品苦味、酸味、鲜味、咸味、甜味、涩味这些基本味道指标值, 还可以额外度量苦味、鲜味、涩味的回味指标值, 在一定程度上真正实现人体味觉感受的电子化、数字化[9]。但受限于内置转换算法建立时代表性呈味物质的选择范围, 应恰当使用和解释产生的味觉值差异。建议明确样品目标组分和干扰组分在传感器上的线性响应情况, 若在某传感器上目标组分有线性响应、且干扰组分无响应, 则该传感器适用于目标组分的味觉测量。
在上述味觉传感器技术革新以及软硬件升级改造的基础上, 该品牌电子舌型号已陆续升级至SA402B (2000年) 和TS-5000Z (2006年)。目前, 基于人工类脂膜的味觉传感器技术仍在不断研究和优化中, 以进一步提升味觉传感器的灵敏度和选择性。
1993年开始, 法国Alpha MOS公司Mifsud博士团队为了解决液体基质样品的味觉质量控制快速检测问题, 基于液敏传感器技术展开研究, 发现由离子选择性敏感膜修饰场效应晶体管构建而成的传感器可对溶液样品中的离子产生选择性分配, 敏感膜在电荷分离时产生电位变化, 进而引发可测量的电化学信号(图 2); 当上述非特定性或弱特定性传感器在有效组合后, 可全面反映液体样品之间的离子差异, 以及样品味觉差异。
2000年, Alpha MOS公司基于上述研究推出第一代电子舌Astree。该设备由Ag/AgCl参比电极和第一套传感器组合(ZZ\BA\BB\CA\GA\HA\JB) 组成, 可以对不同液体样品进行区分。随后传感器阵列逐步升级为第二套药用传感器组合(ZZ\AB\BA\BB\CA\DA\JE)、第三套绝对苦味传感器组合(BD\EB\JA\JG\KA\OA\OB)、第五套味觉筛选传感器组合(SRS\GPS\STS\UMS\ SPS\SWS\BRS) 和第六套味觉筛选传感器组合(AHS\PKS\CTS\NMS\CPS\ANS\SCS)。随着硬件和软件的升级迭代, 该品牌电子舌型号已陆续升级至Astree Ⅱ (2012年) 和Astree Ⅱ V5 (2021年)。
针对Astree Ⅱ V5, 配套使用最新的第六套传感器, 信号稳定性进一步提升, 既可以实现样品的整体差异分析和距离比较, 也可以将样品中酸、咸、鲜3种特定滋味的相对强度进行0~12排序; 另外, 基于非特异性传感器组合的特点, 7根传感器不仅对酸、甜、苦、咸、鲜呈味物质有响应, 还对如辣、辛、麻、涩、醇厚等呈味物质有不同程度的响应, 增加了该电子舌的检测范围。另外, 针对制药领域, Alpha soft软件中载入了苦味掩盖效率模型, 主要用于精准快速地筛选对不良滋味掩盖效果最好的掩味剂。
整体来看, 电位型电子舌具有一定的味觉特异检测性能, 但其检测样品局限在强电解质溶液, 传感器寿命也严重受脂质膜易产生不可逆吸附的影响。
相较于电位型电子舌的离子选择性敏感膜修饰电极, 伏安型电子舌一般采用三电极体系(即工作电极、对电极和参比电极) 作为传感器阵列进行检测。
1997年开始, 瑞典林雪平大学Winquist F课题组[10, 11]率先开展伏安型电子舌研究, 在具有交互敏感作用的贵金属裸电极上, 配以大幅脉冲采集电流响应值, 然后通过模式识别获得产品间的整体差异。由于其特殊的激发机制, 在施加的电位下, 溶液中所有具有电化学活性的物质均会产生响应电流, 造成采集的信号数据量巨大, 给后端的模式识别造成一定的难度。因此, 伏安型电子舌后期的研究重点集中在激发信号的改进、模式识别方式的选择, 以实现特征值的有效提取[12]
2001年开始, 浙江工商大学邓少平、田师一课题组[13]在激发信号改进技术上不断突破, 利用虚拟仪器平台, 在常规大幅脉冲信号的基础上扩充了频率的变化, 即通过1、10、100 Hz三个频率段结合脉冲电压技术实现组合脉冲弛豫谱思想[14]
2007年, 邓少平、田师一课题组[15]基于上述原理, 将电子舌系统进行集成化设计, 构建了初代电子舌商品机, 命名为智舌(Smartongue), 这是继日本、法国后中国生产的首款电子舌产品。智舌的电化学信号产生原理如图 3所示。首先, 由信号激励模块生成多频大幅脉冲激励电压并传输至用于检测液体样品的惰性贵金属传感器阵列。其次, 由于传感器界面会周期性带正电或不带电, 导致溶液中的正负离子发生定向移动形成充电电流; 与此同时, 溶液中的部分分子还会发生氧化还原反应, 形成氧化还原电流。最后, 氧化还原电流和充电电流共同形成了多频大幅脉冲弛豫响应曲线。由于不同溶液中正负离子和发生氧化还原的分子的种类和数量均不同, 其产生的响应曲线也会不同。因此, 可以采集响应曲线的顶点和拐点电流值作为不同溶液样品的特征值, 用于后续数据分析。
2007年至今, 智舌技术主要在样品特征信号采集后的智能模式识别算法上进行革新, 并经历如下3个阶段。阶段1: 基于主成分分析方法的样品整体差异性区分和检验; 阶段2: 基于偏最小二乘法、判别因子分析等方法的产品真伪辨识和特殊物质(单一味觉物质、农残、抗生素等) 定性和半定量分析; 阶段3: 基于线性判别分析、支持向量机、BP神经网络、最近邻算法等智能算法的阶段样品感官属性以及理化指标的快速反演、货架期评价、多重品质等级识别等。
相较于电位型电子舌的选择性检测, 以智舌为代表的伏安型电子舌对溶液中所有具有电化学活性的物质产生响应电流, 更加适用于样品间的整体差异评价; 另外, 智舌采用惰性贵金属传感器阵列, 对于腐蚀性较强的样品或是油类、脂类等电位型电子舌无法检测的样品也能正常检测。
近年来的研究表明, 通过电极表面修饰不同化学材料, 可实现伏安型传感器的优化匹配。其中, 导电聚合物具有活性基团浓度高、电化学响应信号大以及化学、机械和电化学稳定性好的特点, 可通过电化学氧化方法制备并覆盖在铂、金、玻碳等惰性导电材料表面, 构建具有高交互感应性的传感器阵列, 采用循环伏安法对流体进行检测, 可获得良好的测量效果[16-18]。但该类传感器是一个化学性、物理性和电学性共同存在且相互影响的复杂体系, 系统的重现性和稳定性方面还需要进行大量的研究[19]
近十年来, 电子舌在药品领域的应用增长得益于传感器适用性的提高, 主要集中在苦味、甜味和酸味传感器[20]
传统的味觉传感器无法测量不带电的苦味物质, 如咖啡因、茶碱、可可碱等。Toko团队通过研究发现, 当不带电的苦味物质与羟基、二羟基和三羟基苯甲酸等芳香族羧酸的羟基结合时, 芳香族羧酸内部的羟基和羧基发生氢键断裂, 导致传感器膜上的电势发生变化。基于上述原理, 该团队将传感器电极浸入含有2, 6-二羟基苯甲酸的改性溶液中, 对传感器进行表面改性。后期实验证实, 该传感器的响应随着咖啡因等相关物质的浓度增加而增加, 且检测阈值和增加趋势与人尝法一致, 可客观评估不带电苦味物质引起的苦味强度[21]
田师一课题组利用四溴十二烷基铵、2-硝基苯辛醚、聚四氟乙烯和四氢呋喃构建了能够检测异α-酸浓度的人工类脂膜, 并将其分别制备成丝网印刷苦味味觉传感器和石英微天平苦味味觉传感器。进一步地, 通过基于感官品评员的感官试验证明, 上述传感器能够良好地反映出异α-酸的感官苦味强度。
甜味传感器较难开发, 原因是: 甜味剂在水溶液中的电荷分为无电荷、负电荷和正电荷三类, 当使用膜电位测量时, 味觉传感系统需要三种类型的脂质膜[22]。现有商用甜味传感器一般适用于不带电的甜味剂[7, 23], 如INSENT公司的甜味传感器GL1[24]。后续, Toko团队[22, 25]通过比较不同脂质/聚合物膜, 确定了对带正电荷甜味剂具有高度选择性的成分——磷酸二辛酯(脂质) 和2-丁氧基油酸乙酯(增塑剂) 以及对带负电荷甜味剂具有高度选择性的成分——磷酸三辛酯和四溴十二烷基铵, 专门开发了用于阿斯巴甜等带正电荷、糖精钠和安赛蜜等带负电荷甜味剂的甜味传感器。
Mao等[26]构建了基于Ni(OH)2/Ni的甜味传感器, 能够特异性地对甜味物质响应的同时不受到酸味、咸味、苦味和鲜味物质影响, 具有良好的稳定性。该甜味传感器能够对蔗糖、葡萄糖、果糖、乳糖、麦芽糖、木糖醇、山梨糖醇、赤藓糖醇等八种天然甜味剂进行广谱响应, 响应电流与对应甜味剂的浓度呈现出良好的线性关系。该甜味传感器的特征值与蔗糖、葡萄糖、果糖、乳糖和麦芽糖的甜感指数具有良好的相关性。
Zhang等[27]以石墨烯氧化物、血红素、3-氨基苯基硼酸(APBA) 为原料, 构建了一种新型的甜味传感器。该传感器可以用于检测样品中的葡萄糖、果糖、糖精和阿斯巴甜等甜味物质。
Chen等[28]设计并表征了一种基于神经元-硅界面平台的新型生物传感器。与非酸性溶液相比, 光寻址电位传感器(light-addressable potentiometric sensor, LAPS) 培养的味觉感受器细胞能够识别酸性刺激。该研究还建立了味觉感受器细胞的计算模型, 模拟味觉感受器细胞的离子电流和动作电位, 对LAPS记录的细胞外信号进行解码, 从而实现了酸味的检测。
Riul等[29]采用单一和复合纳米结构薄膜的导电聚合物作为组成电子舌的单个传感单元。该传感器能够在10~1×106 Hz的阻抗谱测量范围内, 采用超薄薄膜进行信号传导, 用于检测样品中的酸味物质, 已经在水、促味剂和酒等实际商业饮料中得到了良好的验证应用。
Mao[30]构建了基于玻璃球膜氢离子电极的酸味传感器, 其电压信号值与柠檬酸、苹果酸、酒石酸、乳酸和富马酸的浓度呈对数关系, 与它们的酸感量化值呈线性关系, 以此建立了相应的检测数学公式。基于不同酸感量化值的柠檬酸样品, 在六种不同浓度的蔗糖背景条件下, 不同酸感量化值的柠檬酸对应的电压值均没有显著差异, 说明该酸味传感器对于酸感量化值的检测不受甜味影响。说明该酸味传感器能够用于检测柠檬酸、苹果酸、酒石酸、乳酸和富马酸的酸感量化值。
1999年, Bachmann等[31]提出了生物电子舌(bioelectronic tongue) 的概念, 可直接提供味觉受体和呈味物质之间结合活动触发的味觉信号, 开发出更接近人类味觉的人工传感器, 这种高选择性和特异性可以为生物电子舌减少背景信息干扰, 显示出其在味觉转导机制研究、味道评价等方面的应用优势[32, 33]。但目前该类电子舌仍处于起步阶段, 应用报道还较少。现有的生物传感器主要包括酶、受体蛋白、味觉细胞或生物组织等类型。其中, 基于味觉受体或细胞制备的生物传感器最受关注[34]
Wu等[35]将含有苦味受体T2R4的人体血浆HEK-293细胞膜组分提取出来, 并固定在石英晶体微管上, 形成一种新型的生物传感器, 可特异性识别和捕获各种苦味刺激, 是用于苦味检测和味觉信号传导研究的宝贵工具。
Wei等[36]利用大鼠心肌细胞存在苦、鲜味内源性味觉受体, 将上述细胞有效附着在传感器表面, 可特异性检测苯甲地那铵、苯甲胺磷等苦味物质, 检测限可达1×10-6 mol·L-1
Hui等[37, 38]发现人体小肠NCI-H716细胞特异性表达G蛋白偶联甜味受体, STC-1细胞特异性表达G蛋白偶联苦味受体, 当甜味物质或苦味物质和G蛋白偶联受体结合后, 会引发钙离子流, 进而引起细胞电阻发生变化, 最终使得生物传感器的电化学阻抗值发生改变。由于不同甜味或苦味物质与G蛋白偶联受体的结合能力有强弱和快慢之分, 因此通过电化学阻抗谱频率优化, 可以实现对甜味剂和甜味混合物、苦味剂和苦味混合物的种类和浓度进行特征性识别, 为味觉机制研究提供了一种有效的手段。
Yan[39]构建了T1R2+3-Gust44-HEK 293T的稳定细胞株, 以蔗糖、木糖醇、麦芽糖醇、山梨糖醇、阿拉伯糖、木糖、阿洛酮糖七种甜味剂的12个浓度的溶液作为刺激物, 进行FlexStation钙影像分析, 建立七种甜味剂浓度对数与细胞钙流信号的关系曲线。同时, 利用细胞钙流信号曲线与上述甜味剂的感官甜度进行对应, 具有良好的对应性和稳定性。
另外, 美国Senomyx公司构建了能够稳定表达T1R2/T1R3的细胞系, 并在此基础上开发了一套用于甜味剂高通量筛选的平台SweetScreenHT, 由此开发出了多种人工甜味剂。其中, 为可口可乐公司开发的新型甜味剂已用于无糖汽水的生产[40]
上述新型生物电子舌的成功研发, 为电子舌深入开展口感评价提供新的技术支撑。
电子舌设备昂贵、传感器易损耗等因素限制了这种检测方法的普及[41]。近年来, 电子舌逐步朝着便携式、高通量芯片等小型化设备的方向发展。与传统电子舌相比, 小型化电子舌往往体积小、用样量少、检测速度快, 而且生产成本低、易集成, 可用于工业规模生产的在线监测、有毒等特殊样品的反复品尝、时间或成本限制的其他分析需求等。
纳米材料, 在电子舌小型化研制中较为受欢迎, 特别是在生物电子舌方面[42]。Akifulhaque等[43]综述了纳米材料在生物电子舌方面的应用, 并指出纳米材料的使用可以增加纳米生物传感器的灵敏度和生产可及性。Song等[44]将亚胺类甜味受体嵌入纳米囊泡, 利用单壁碳纳米晶体管的管场效应, 开发了一种生物电子舌, 其检测灵敏度和选择性均有所提升。由于人体味觉受体细胞的味觉检测通常发生在细胞膜的离子通道或受体上, Liu等[45]将短杆菌肽整合到高度有序的纳米脂质双层膜中形成离子通道, 为某些特殊离子的味道检测提供了可能。
聚二甲基硅氧烷(polydimethylsiloxane, PDMS), 是一种疏水的有机硅物料, 广泛用于微流体芯片制造。Daikuzono等[46]在PDMS基质中嵌入微通道, 并覆在金电极上, 然后通过逐层沉积得到双层阴、阳离子膜修饰电极, 得到一种新的微流控电子舌。该电子舌检测速度比常规电子舌快三倍, 可用于临床快速检测。Shimizu等[47]在5根非功能化的不锈钢细丝上分别涂覆金、铂、镍、铁和氧化铝薄膜, 然后插入PDMS微流控芯片中, 结合阻抗谱技术, 成功应用于浓度低于人类味觉阈值的基本味道分类。
目前, 其他小型化电子舌的技术报道不多, 但该研究方向仍在持续推进中。Toko团队开发了一种便携式味觉感知系统, 其核心部件为高通量芯片, 该芯片是将参比和工作电极嵌入低成本、多功能的聚碳酸酯基板上制作而成, 在保证传感器实用性的同时, 设备成本也有所下降[48, 49]。Litvineoko等[50]利用硅表面光能载流子寿命的二维成像开发了一种新的电子舌, 可对不同溶液创建特征电子指纹; 并可设置不同的偏置电压以提高设备的可靠性。Shi等[51]开发了一种便携式伏安电子舌(VE-tongue), 可用于陈皮等中草药的年限鉴定。
传感器可分为电化学、光学、生物等类型[52-54]。Nery等[55]在分析酒样品时发现, 仅仅采用几个单一类型的交叉敏感传感器组成的电子舌, 其预测能力低于混合选择性的电子舌。
随着模式识别技术的不断进步, 不同类型的传感数据能够实现多维融合, 避免同一类型传感器数量过多造成的数据冗余、过度拟合的问题[56]。Winquist等课题组基于伏安型电子舌数据复杂、电位型电子舌对带电分子敏感的缺陷, 将这两种技术进行优势互补, 形成一种混合型电子舌, 成功应用于发酵牛奶分类[57]、啤酒发酵分析[58]、碎肉中钠盐或钾盐水平分析[59]、三硝基甲苯检测[60]等。
随着电子舌的硬件与软件技术的日益完善、研发理念的逐步提升, 基于电子舌的口感评价结果的准确性和可靠性与日俱增。然而, 作为能够广泛应用于高校、企业乃至于普通消费者场景而言, 目前的电子舌技术仍存在以下挑战: ①电子舌产品的设计原理多样, 测样方式多样且复杂, 仪器稳定性有限, 很可能造成不同传感器阵列或同一传感器阵列在不同检测时期获得差异性结果, 即存在漂移现象。提示该检测方法标准化困难, 很难形成统一的指导原则或技术文件, 这也是电子舌应用面临的重大挑战和技术瓶颈; ②仿生传感获得的结果往往需要人体感官评价进行验证, 但对于药品而言, 有时不建议甚至不能采用基于感官评价人员的测评方法, 导致该电子舌检测方法的结果可靠性难以得到有效论证。
针对上述电子舌面临的挑战, 可以考虑采用以下方式进行尝试面对: ①利用表面改性、电化学沉积、微电极阵列等传感器技术和支持向量机、人工神经网络、决策树等人工智能算法来提升电子舌的检测准确性和识别准确性; ②利用盐酸奎宁或盐酸小檗碱(苦味)、蔗糖(甜味)、柠檬酸(酸味)、氯化钠(咸味)、谷氨酸钠(鲜味) 和单宁酸(涩味) 等基本味觉的代表性物质建立人体感官强度曲线, 再与电子舌检测结果进行标定, 从而得到标准的味觉数据库; ③利用类器官生物技术, 构建能够真实反映人感官强度的味觉细胞传感器, 通过味觉细胞传感器来对药物分子和制剂成品进行检测并得到相应的评价结果, 从而解决人无法评价特殊药品的问题。
