Seven compounds were isolated from Onychium japonicum by macroporous resin, silica gel, ODS, Sephadex LH-20 column chromatography and semi-preparative HPLC. Their structures were identified by NMR, MS and other spectroscopic methods as onychone A (1), quercetin (2), quercetin-3-O-α-L-rhamnoside (3), kaempferol-7-O-β-D-glucopyranoside (4), kaempferol-3-O-α-L-rhamnopyranoside (5), (-)-prunin (6), and norathyriol (7). Compound 1 is a novel macrocyclic flavonoid, and all the others are reported from this plant for the first time. In vitro cytotoxic activities of compounds 1-7 were evaluated by MTS testing with five cancer cell lines. Compound 7 exhibited weak cytotoxicity against tumor cell lines A549, SMMC-7721, and SW480.

采用大孔树脂、硅胶、反相硅胶、葡聚糖凝胶及高效液相等色谱法, 从野雉尾金粉蕨中分离纯化得到7个化合物。通过核磁共振谱、质谱等波谱学方法分别鉴定为金粉蕨酮A (1)、槲皮素(2)、槲皮素-3-O-α-L-吡喃鼠李糖苷(3)、山柰酚-7-O-β-D-葡萄糖苷(4)、山柰酚-3-O-α-L-吡喃鼠李糖苷(5)、(-)-prunin (6) 和1, 3, 6, 7-四羟基酮(7), 其中化合物1为结构新颖的大环黄酮类化合物, 其余化合物均为首次从该植物中分离得到。采用MTS法测试化合物对5种肿瘤细胞株的细胞毒活性, 结果显示化合物7对肺癌A549、肝癌SMMC-7721和结肠癌SW480细胞具有弱的细胞增殖抑制活性。

采用大孔树脂、硅胶、反相硅胶、葡聚糖凝胶及高效液相等色谱法, 从野雉尾金粉蕨中分离纯化得到7个化合物。通过核磁共振谱、质谱等波谱学方法分别鉴定为金粉蕨酮A (1)、槲皮素(2)、槲皮素-3-O-α-L-吡喃鼠李糖苷(3)、山柰酚-7-O-β-D-葡萄糖苷(4)、山柰酚-3-O-α-L-吡喃鼠李糖苷(5)、(-)-prunin (6) 和1, 3, 6, 7-四羟基酮(7), 其中化合物1为结构新颖的大环黄酮类化合物, 其余化合物均为首次从该植物中分离得到。采用MTS法测试化合物对5种肿瘤细胞株的细胞毒活性, 结果显示化合物7对肺癌A549、肝癌SMMC-7721和结肠癌SW480细胞具有弱的细胞增殖抑制活性。

Seven compounds were isolated from Onychium japonicum by macroporous resin, silica gel, ODS, Sephadex LH-20 column chromatography and semi-preparative HPLC. Their structures were identified by NMR, MS and other spectroscopic methods as onychone A (1), quercetin (2), quercetin-3-O-α-L-rhamnoside (3), kaempferol-7-O-β-D-glucopyranoside (4), kaempferol-3-O-α-L-rhamnopyranoside (5), (-)-prunin (6), and norathyriol (7). Compound 1 is a novel macrocyclic flavonoid, and all the others are reported from this plant for the first time. In vitro cytotoxic activities of compounds 1-7 were evaluated by MTS testing with five cancer cell lines. Compound 7 exhibited weak cytotoxicity against tumor cell lines A549, SMMC-7721, and SW480.

野雉尾金粉蕨Onychium japonicum (Thumb.) Kunze是中国蕨科金粉蕨属植物, 又称小叶金花草、解毒蕨、解毒草、野鸡尾等, 广泛分布于我国华东、华中、东南及西南地区, 日本、菲律宾、印度尼西亚等也有分布[1, 2]。野雉尾金粉蕨在广西壮族地区是常用的解毒药(壮药名: Gutgaijdoeg棍盖冬), 壮医认为其具有调龙路(人体血液循环的通路), 通气道(人体与环境进行气体交换功能的通道)、谷道(人体消化、吸收食物并排出残渣的通道), 解毒除湿, 止血的功效, 用于治疗贫痧(感冒)、能蚌(黄疸)、阿意咪(痢疾)、林得叮相(跌打损伤) 和农药中毒等疾病[3]。该药临床上常单用或配伍用于治疗癌症等疾病, 是正在研制的抗肿瘤新药“壮药复方铁草胶囊(国家科技支撑计划2006BAI06A17)”的主要药物之一[4]。“壮药复方铁草胶囊”是广西国际壮医医院用于治疗肝癌、肺癌等恶性肿瘤的临床经验方, 具有消肿散结、驱邪扶正之功, 临床效果显著[5]。同时, 现代药理研究亦表明野雉尾金粉蕨的醇提物能明显抑制小鼠体内移植性肿瘤小鼠肉瘤S180和小鼠肝癌H22的生长, 而且呈现量效关系[6]。文献报道野雉尾金粉蕨的主要成分有黄酮[7, 8]、蕨素倍半萜[9]及鸟巢烷二萜[10]等类成分。为了进一步阐明野雉尾金粉蕨的化学成分, 筛选出结构新颖具有抗肿瘤活性的天然产物, 本文对野雉尾金粉蕨乙酸乙酯部位的化学成分进行研究, 从中分离鉴定了7个黄酮类成分, 分别鉴定为金粉蕨酮A (1)、槲皮素(2)、槲皮素-3-O-α-L-吡喃鼠李糖苷(3)、山柰酚- 7-O-β-D-葡萄糖苷(4)、山柰酚-3-O-α-L-吡喃鼠李糖苷(5)、(-)-prunin (6) 和1, 3, 6, 7-四羟基酮(7), 其中化合物1为结构罕见的大环黄酮类化合物, 其他化合物均为首次从该植物中分离得到(图 1)。活性测试结果显示, 化合物7对肺癌A549、肝癌SMMC-7721和结肠癌SW480细胞具有一定的细胞增殖抑制活性。
化合物1  黄色固体, [α]$ {}_{\mathrm{D}}^{20} $ +769.7 (c 0.1, MeOH)。HR-ESI-MS显示准分子离子峰m/z: 737.171 7 [M-H] (计算值737.171 8), 提示该化合物分子式为C36H34O17, 不饱和度为20。
1H NMR谱中显示有10个芳香或双键质子信号(表 1), 推测可能含有1个ABX耦合体系的苯环[δH 8.03 (1H, d, J = 2.1 Hz, H-2″″), 7.38 (1H, dd, J = 8.3, 2.3 Hz, H-6″″), 6.97 (1H, d, J = 8.2 Hz, H-5″″)]; 1个AA′BB′耦合体系的苯环[δH 7.54 (2H, d, J = 8.3 Hz, H-2′, -6′), 6.68 (2H, d, J = 8.3 Hz, H-3′, -5′)]; 1个孤立的芳香质子信号δH 6.35 (1H, s, H-8) 及2个反式双键质子信号[δH 7.74 (1H, d, J = 16.0 Hz, H-7″″), 6.69 (1H, d, J = 16.1 Hz, H-8″″)]; 以及11个连氧次甲基或亚甲基和1个甲基质子信号δH 1.33 (3H, d, J = 6.1 Hz, H-6″), 推测可能为2个糖基的质子信号。13C NMR谱、DEPT谱结合HSQC谱显示34个碳信号(包含2个重叠的碳信号δC 133.2和116.5), 包括14个季碳, 其中δC 179.0、168.1为2个羰基信号; 20个次甲基碳。其中δC 145.2、117.2为2个烯碳信号; 1个连氧亚甲基信号δC 64.0及1个甲基信号δC 18.1。1H-1H COSY谱显示H-5″″/H-6″″相关, H-7″″/H-8″″相关, HMBC谱显示H-2″″与C-4″″、C-6″″、C-7″″相关, H-6″″与C-2″″、C-4″″、C-7″″相关, H-7″″与C-2″″、C-6″″、C-9″″相关, H-8″″与C-1″″相关, 确证了对羟基桂皮酰结构片段的存在(图 2)。
1H-1H COSY谱显示两组糖残基自旋耦合片段: H-1″/H-2″/H-3″/H-4″/H-5″/H3-6″和H-1‴/H-2‴/H-3‴/H-4‴/H-5‴/H2-6‴, 推测可能存在一个鼠李糖和一个葡萄糖单元; 通过酸水解实验, 并结合糖端基质子耦合常数[δH 5.44 (1H, d, J = 1.3 Hz, H-1″), 3.36 (1H, d, J = 7.7 Hz, H-1‴)], 最终确定为α-L-鼠李糖和β-D-葡萄糖[11]。HMBC谱显示H-3″与C-1‴相关, H-6‴与C-9″″相关, 说明鼠李糖的C-3″位通过氧原子与葡萄糖的端基C-1‴位相连, 葡萄糖基的C-6‴通过酯键与对羟基桂皮酸片段的C-9″″相连。
至此, 该化合物剩余15个碳原子, 1H-1H COSY谱显示H-2′ (H-6′)/H-3′ (H-5′) 相关, HMBC谱显示H-2′ (H-6′) 与C-2相关, H-8与C-6、C-7、C-10相关, 结合化学位移和分子式, 说明该化合物还存在一个黄酮母核。HMBC谱显示, 鼠李糖的H-1″与C-3相关, 对羟基桂皮酰的H-2″″与C-6相关(图 2), 说明鼠李糖的端基C-1″通过氧原子连接在黄酮的C-3位, 对羟基桂皮酸的C-3″″连接在黄酮的C-6位, 形成了如图 1所示的大环黄酮结构。
化合物1黄酮的A环与桂皮酰基的苯环间存在联苯轴手性分子的特征, 进一步采用TDDFT方法, 计算了该化合物简化模型的ECD谱, 发现计算ECD谱与实验ECD谱(图 3) Cotton吸收峰一致, 从而确定化合物1的绝对构型为aS, 命名为金粉蕨酮A (onychone A)。该化合物属于结构罕见的大环黄酮类, 大环母核中由对羟基桂皮酰、葡萄糖、鼠李糖以及黄酮的A和B环构成。
体外细胞毒活性测试结果表明(表 2), 化合物7对肺癌A549、肝癌SMMC-7721和结肠癌SW480细胞显示弱的细胞毒活性; 在50 μmol·L-1浓度下, 化合物1~6对测试的5种肿瘤细胞增殖均无显著抑制作用。

野雉尾金粉蕨药材于2020年5月由广西仙茱中药科技有限公司提供, 经广西中医药大学廖月葵高级实验师鉴定为中国蕨科金粉蕨属植物野雉尾金粉蕨Onychium japonicum (Thumb.) Kunze的干燥全草。植物标本(编号JDC-001) 保存于广西中医药大学药学院。
干燥的野雉尾金粉蕨全草18 kg, 用75%乙醇加热回流提取2次, 每次2 h。合并提取液后减压浓缩得总浸膏2 076 g, 总浸膏用等量水悬浮, 依次用石油醚、乙酸乙酯和正丁醇萃取, 回收溶剂, 得石油醚部位浸膏110 g, 乙酸乙酯部位浸膏1 023 g, 正丁醇部位浸膏217 g和水部位浸膏726 g。
乙酸乙酯部位浸膏950 g, 经AB-8大孔树脂柱色谱分离, 依次以甲醇-水(30%~100%) 梯度洗脱, 得到5个组分Fr. A~Fr. E。Fr. B经硅胶柱色谱分离, 以二氯甲烷-甲醇(50∶1→30∶1→10∶1→5∶1→3∶1→1∶1→0∶1) 系统梯度洗脱, 得到11个组分Fr. B.1~Fr. B.11。Fr. B.8经C18反相中低压柱色谱(甲醇-水, 10%~100%) 得到8个组分Fr. B.8.1~Fr. B.8.8。Fr. B.8.5用硅胶柱色谱分离, 以二氯甲烷-甲醇(50∶1→30∶1→20∶1→15∶1→10∶1→5∶1→1∶1→0∶1) 为洗脱剂进行梯度洗脱, 得到8个组分Fr. B.8.5.1~Fr. B.8.5.8。Fr. B.8.5.5经半制备高效液相色谱(25%乙腈-水, 3 mL·min-1) 分离得到化合物7 (4.9 mg, tR = 24.2 min)、化合物2 (4.2 mg, tR = 37.1 min)。Fr.B.8.5.8经MCI柱色谱(甲醇-水, 30%~100%) 得到6个组分Fr. B.8.5.8.1~Fr. B.8.5.8.6。Fr. B.8.5.8.4经Sephadex LH-20柱色谱(甲醇) 分离, 再经半制备高效液相色谱(22%乙腈-水, 3 mL·min-1) 分离得到化合物5 (29.3 mg, tR = 33.0 min)。Fr. B.9经C18柱色谱(甲醇-水, 10%~100%) 得到11个组分Fr. B.9.1~Fr.B.9.11。Fr. B.9.5经半制备高效液相色谱(43%甲醇-水, 3 mL·min-1) 分离得到化合物6 (8.5 mg, tR = 17.5 min)、4 (3.2 mg, tR = 22.0 min)、3 (7.0 mg, tR = 30.0 min)。Fr.B.9.8经半制备高效液相色谱(28%乙腈-水, 3 mL·min-1) 分离得到化合物1 (5.9 mg, tR= 19.5 min)。
称取化合物1 (1.0 mg), 加入2 mol·L-1盐酸(5 mL), 密封, 于100 ℃反应4 h, 减压浓缩至干后加入5 mL蒸馏水溶解, 用乙酸乙酯萃取3次, 每次5 mL。将水解得到的单糖部分和标准糖分别溶于含有L-半胱氨酸甲酯盐酸盐(1.0 mg) 的吡啶(1 mL) 中, 并在60 ℃下加热反应1 h后加入邻甲苯异硫氰酸酯(2 µL), 并将溶液在60 ℃下继续反应1 h, 浓缩至干。对反应混合物进行HPLC分析[YMC-Pack ODS-A色谱柱(250 mm × 4.6 mm, 5 μm), 流动相为乙腈∶水= 25∶75, 流速0.8 mL·min-1, 检测波长为250 nm, 柱温为30 ℃, 进样量为5 μL]。通过比较标准糖衍生物和样品单糖衍生物的保留时间, 确定化合物中单糖的绝对构型(D-葡萄糖20.43 min, L-鼠李糖34.67 min)。

化合物2  黄色粉末; EI-MS m/z: 302 [M]+, 分子式C15H10O7; 1H NMR (500 MHz, CD3OD) δH: 7.74 (1H, d, J = 2.1 Hz, H-2′), 7.63 (1H, dd, J = 8.5, 2.1 Hz, H-6′), 6.88 (1H, d, J = 8.4 Hz, H-5′), 6.39 (1H, d, J = 2.1 Hz, H-8), 6.18 (1H, d, J = 1.9 Hz, H-6); 13C NMR (125 MHz, CD3OD) δC: 148.0 (C-2), 137.3 (C-3), 177.3 (C-4), 158.2 (C-5), 99.3 (C-6), 165.7 (C-7), 94.4 (C-8), 162.5 (C-9), 104.5 (C-10), 124.2 (C-1′), 116.0 (C-2′), 146.2 (C-3′), 148.7 (C-4′), 116.2 (C-5′), 121.6 (C-6′)。以上数据与文献[12]报道基本一致, 故鉴定化合物2为槲皮素。
化合物3  黄色粉末; ESI-MS m/z: 471 [M+Na]+, 分子式C21H20O11; 1H NMR (500 MHz, CD3OD) δH: 7.34 (1H, d, J = 2.1 Hz, H-2′), 7.31 (1H, dd, J = 8.2, 2.1 Hz, H-6′), 6.91 (1H, d, J = 8.3 Hz, H-5′), 6.37 (1H, d, J = 2.0 Hz, H-8), 6.20 (1H, d, J = 2.0 Hz, H-6), 5.35 (1H, d, J = 1.6 Hz, H-1″), 4.22 (1H, dd, J = 3.4, 1.7 Hz, H-2″), 3.75 (1H, dd, J = 9.4, 3.4 Hz, H-3″), 3.42 (1H, dq, J = 9.5, 6.2 Hz, H-5″), 3.35 (1H, m, H-4″), 0.94 (3H, d, J = 6.1 Hz, H-6″); 13C NMR (125 MHz, CD3OD) δC: 159.3 (C-2), 136.2 (C-3), 179.6 (C-4), 163.2 (C-5), 99.8 (C-6), 166.0 (C-7), 94.7 (C-8), 158.5 (C-9), 105.9 (C-10), 123.0 (C-1′), 116.9 (C-2′), 146.4 (C-3′), 149.8 (C-4′), 116.4 (C-5′), 122.8 (C-6′), 103.5 (C-1″), 71.9 (C-2″), 72.1 (C-3″), 73.2 (C-4″), 72.0 (C-5″), 17.7 (C-6″)。以上数据与文献[13]报道基本一致, 故鉴定化合物3为槲皮素-3-O-α-L-吡喃鼠李糖苷。
化合物4  黄色粉末; ESI-MS m/z: 447 [M-H]-, 分子式C21H20O11; 1H NMR (500 MHz, CD3OD) δH: 8.13 (2H, d, J = 9.0 Hz, H-2′, 6′), 6.91 (2H, d, J = 9.0 Hz, H-3′, 5′), 6.77 (1H, d, J = 2.2 Hz, H-8), 6.47 (1H, d, J = 2.1 Hz, H-6), 5.06 (1H, d, J = 7.3 Hz, H-1″), 3.94 (1H, dd, J = 12.1, 2.3 Hz, H-2″), 3.72 (1H, dd, J = 12.2, 5.8 Hz, H-3″), 3.55 (1H, ddd, J = 9.8, 5.8, 2.3 Hz, H-5″), 3.50 (2H, m, H-4″, 6″a), 3.40 (1H, t, J = 9.3 Hz, H-6″b); 13C NMR (125 MHz, CD3OD) δC: 148.8 (C-2), 137.6 (C-3), 177.6 (C-4), 162.2 (C-5), 100.2 (C-6), 164.5 (C-7), 95.6 (C-8), 157.8 (C-9), 106.3 (C-10), 123.6 (C-1′), 130.9 (C-2′, 6′), 116.3 (C-3′, 5′), 160.8 (C-4′), 101.6 (C-1″), 74.7 (C-2″), 77.9 (C-3″), 71.3 (C-4″), 78.4 (C-5″), 62.5 (C-6″)。以上数据与文献[14]报道基本一致, 故鉴定化合物4为山柰酚-7-O-β-D-葡萄糖苷。

化合物7  黄白色粉末; EI-MS m/z: 260 [M]+, 分子式C13H8O6; 1H NMR (500 MHz, CD3OD) δH: 7.43 (1H, s, H-8), 6.81 (1H, s, H-5), 6.28 (1H, d, J = 1.7 Hz, H-4), 6.14 (1H, d, J = 1.7 Hz, H-2); 13C NMR (125 MHz, CD3OD) δC: 164.4 (C-1), 98.7 (C-2), 166.3 (C-3), 94.7 (C-4), 159.4 (C-4a), 155.2 (C-5a), 103.5 (C-5), 144.8 (C-6), 153.2 (C-7), 109.1 (C-8), 113.9 (C-8a), 181.1 (C-9), 103.3 (C-9a)。以上数据与文献[18]报道基本一致, 故鉴定化合物7为1, 3, 6, 7-四羟基酮。
用含10%胎牛血清的培养液(DMEM或RMPI1640) 配成单个细胞悬液, 以每孔3×103~1.5×104个细胞数接种到96孔板, 每孔体积100 μL, 培养24 h后弃去旧培养液。分组给药, 每组3个复孔, 空白组加入新的完全培养液, 实验组分别加入含待测样品的完全培养液200 μL/孔, 单体化合物用DMSO溶解, 初次筛选终浓度为单体化合物50 μmol·L-1, 根据初筛结果进行复筛, 单体化合物终浓度分别为50、10、2、0.4、0.08 μmol·L-1; 并设顺铂(DDP) 和紫杉醇(Taxol) 两个阳性对照组。给药后放入培养箱中培养48 h后, 弃去贴壁细胞孔内的培养液并加入MTS溶液和培养液分别为20和100 μL; 弃去悬浮细胞孔内培养上清液100 μL, 再加入MTS溶液20 μL。设置加入MTS溶液和培养液分别为20和100 μL的3个空白复孔, 放入培养箱中继续孵育2~4 h后测定光吸收值。使用多功能酶标仪于492 nm波长处, 读取并记录各孔OD值。
细胞的生长抑制率= [1 - (实验组OD值- 空白组OD值) / (对照组OD值- 空白组OD值)] × 100%, 并利用Graphpad Prism计算半数抑制浓度(IC50)。
