Fourteen flavonoids were isolated and purified from Epimedium sagittatum by various chromatography techniques such as macroporous adsorbent resin, silica gel, ODS, Sephadex LH-20, HW-40C and semi-preparative HPLC. Their structures were identified by analysis of physicochemical properties and spectral data, and determined as 3'-hydroxy-baohuoside-Ⅱ (1), huazhongilexone-7-O-β-D-glucopyranoside (2), kaempferol-3-O-α-L-rhamnoside (3), baohuoside-Ⅱ (4), icariside-Ⅱ (5), kaempferol 3, 7-di-O-α-L-rhamnopyranoside (6), (+)-aromadendrin (7), kaempferol 3-O-(2-O-β-D-apiofuranosyl)-α-L-rhamnopyranoside (8), sagittatoside A (9), 2″-O-rhamnosyl icariside-Ⅱ (10), apigenin-7-O-β-D-glucoside (11), quercetin 3-O-β-D-apiofuranoyl-(1→2)-α-L-rhamnopyranoside (12), kaempferol (13), icariin (14). Among them, compound 1 is a new compound, while compounds 2, 6-8, 11, and 12 were isolated from E.sagittatum for the first time.

利用现代柱色谱技术,如大孔吸附树脂、硅胶、ODS、Sephadex LH-20和HW-40C柱色谱及半制备液相等对箭叶淫羊藿正丁醇部位化学成分进行分离纯化,得到14个黄酮类化合物。根据化合物的理化性质和波谱特征,确定其结构分别为3'-羟基宝藿苷-Ⅱ(1)、huazhongilexone-7-O-β-D-glucopyranoside(2)、kaempferol-3-O-α-L-rhamnoside(3)、宝藿苷-Ⅱ(4)、淫羊藿次苷-Ⅱ(5)、kaempferol 3,7-di-O-α-L-rhamnopyranoside(6)、(+)-aromadendrin(7)、kaempferol 3-O-(2-O-β-D-apiofuranosyl)-α-L-rhamnopyranoside(8)、箭藿苷A(9)、2″-O-rhamnosyl icariside-Ⅱ(10)、apigenin-7-O-β-D-glucoside(11)、quercetin 3-O-β-D-apiofuranoyl-(1→2)-α-L-rhamnopyranoside(12)、山柰酚(13)、淫羊藿苷(14)。其中化合物1为新化合物,化合物26~81112为首次从箭叶淫羊藿中分离得到。

利用现代柱色谱技术,如大孔吸附树脂、硅胶、ODS、Sephadex LH-20和HW-40C柱色谱及半制备液相等对箭叶淫羊藿正丁醇部位化学成分进行分离纯化,得到14个黄酮类化合物。根据化合物的理化性质和波谱特征,确定其结构分别为3'-羟基宝藿苷-Ⅱ(1)、huazhongilexone-7-O-β-D-glucopyranoside(2)、kaempferol-3-O-α-L-rhamnoside(3)、宝藿苷-Ⅱ(4)、淫羊藿次苷-Ⅱ(5)、kaempferol 3,7-di-O-α-L-rhamnopyranoside(6)、(+)-aromadendrin(7)、kaempferol 3-O-(2-O-β-D-apiofuranosyl)-α-L-rhamnopyranoside(8)、箭藿苷A(9)、2″-O-rhamnosyl icariside-Ⅱ(10)、apigenin-7-O-β-D-glucoside(11)、quercetin 3-O-β-D-apiofuranoyl-(1→2)-α-L-rhamnopyranoside(12)、山柰酚(13)、淫羊藿苷(14)。其中化合物1为新化合物,化合物26~81112为首次从箭叶淫羊藿中分离得到。

箭叶淫羊藿  /  黄酮苷  /  3'-羟基宝藿苷-Ⅱ  /  化学成分  /  提取分离

Fourteen flavonoids were isolated and purified from Epimedium sagittatum by various chromatography techniques such as macroporous adsorbent resin, silica gel, ODS, Sephadex LH-20, HW-40C and semi-preparative HPLC. Their structures were identified by analysis of physicochemical properties and spectral data, and determined as 3'-hydroxy-baohuoside-Ⅱ (1), huazhongilexone-7-O-β-D-glucopyranoside (2), kaempferol-3-O-α-L-rhamnoside (3), baohuoside-Ⅱ (4), icariside-Ⅱ (5), kaempferol 3, 7-di-O-α-L-rhamnopyranoside (6), (+)-aromadendrin (7), kaempferol 3-O-(2-O-β-D-apiofuranosyl)-α-L-rhamnopyranoside (8), sagittatoside A (9), 2″-O-rhamnosyl icariside-Ⅱ (10), apigenin-7-O-β-D-glucoside (11), quercetin 3-O-β-D-apiofuranoyl-(1→2)-α-L-rhamnopyranoside (12), kaempferol (13), icariin (14). Among them, compound 1 is a new compound, while compounds 2, 6-8, 11, and 12 were isolated from E.sagittatum for the first time.

箭叶淫羊藿(Epimedium sagittatum), 又名三枝九叶草, 始载于《神农本草经》, 是中药淫羊藿的药源植物之一, 主要分布于安徽、江西、湖南和湖北等地[1]。淫羊藿具有补肾阳、强筋骨、祛风湿等功效[2], 临床上可用于治疗骨质疏松、肾虚、更年期综合征、乳房肿块、冠心病、高血压等疾病[3]。研究表明, 淫羊藿中主要含有黄酮类、木脂素类、多糖类、生物碱类等活性成分[4], 具有抗肿瘤[5]、抗骨质疏松[6]、调节免疫[7]、抗抑郁[8]等多种药理活性。作为药食两用的传统中药之一, 近年来淫羊藿也用于多种保健产品的开发, 而箭叶淫羊藿是中药淫羊藿中最早被药典收录且分布最广的品种, 其有着巨大的开发潜力和应用前景[9]。本实验以箭叶淫羊藿为研究对象, 对其化学成分进行系统的分离纯化, 从中分离得到14个化合物, 且均为黄酮类化合物(图 1), 其中化合物1 (图 2) 为新化合物, 化合物26~81112为首次从箭叶淫羊藿中分离得到。
化合物1为黄色粉末, [α]$ {}_{\mathrm{D}}^{20} $ -75 (c 0.086, MeOH); HR-ESI-MS m/z 539.154 4 [M+Na]+ (计算值539.152 4), 提示化合物的分子式为C26H28O11, 计算不饱和度为13。1H NMR (CD3OD, 500 MHz) 谱中, δH 7.38 (1H, d, J = 2.2 Hz, H-2′)、7.31 (1H, dd, J = 2.2, 8.4 Hz, H-6′) 和6.92 (1H, d, J = 8.4 Hz, H-5′) 为一组苯环ABX系统特征信号峰; 低场区δH 6.25 (1H, s, H-6) 为苯环上的一个氢质子信号, 推测其可能存在一个五取代苯环; δH 5.34 (1H, d, J = 1.7 Hz, H-1‴), 结合0.94 (3H, d, J = 6.4 Hz, H-6‴), 提示分子中可能存在一个鼠李糖的结构片段; 由δH 5.20 (1H, m, H-2″)、3.48 (2H, m, H-1″)、1.73 (3H, s, H-5″) 和1.66 (3H, s, H-4″) 的氢质子信号, 推测分子中可能存在一个异戊烯基的结构片段。分析13C NMR (CD3OD, 125 MHz) 谱并结合HSQC谱可知, 化合物1中共有26个碳。根据化学位移δC 99.3~180.0内存在18个碳信号, 结合1H NMR数据, 提示该化合物可能为黄酮类化合物, 其中由δC 136.1 (C-3) 和179.9 (C-4) 的碳信号推测该化合物为黄酮醇类化合物。δC 132.6 (C-3″)、123.6 (C-2″)、25.9 (C-4″)、22.4 (C-1″)、18.1 (C-5″) 为一组异戊烯基上碳的特征信号峰。δC 103.6 (C-1‴)、71.9 (C-2‴)、72.1 (C-3‴)、73.3 (C-4‴)、72.0 (C-5‴), 17.7 (C-6‴) 为一组鼠李糖上碳的特征信号峰。化合物1经酸水解后, 水解产物经HPLC分析, 表明该化合物中的糖为L-鼠李糖, 且根据C-3‴和C-5‴的化学位移, 确定其为α构型。在HMBC谱中, δH 5.34 (H-1‴) 与δC 136.1 (C-3) 具有远程相关, 表明鼠李糖连在黄酮的C-3位上。δH 3.48 (H-1″) 与δC 163.1 (C-7)、105.9 (C-8) 以及155.7 (C-9) 具有远程相关, 表明异戊烯基连在黄酮的C-8位上。化合物1的核磁数据与文献报道的已知化合物宝藿苷-Ⅱ的核磁数据对比[10], 化合物1的C-3′位上多了一个酚羟基。综上所述, 确定化合物1结构为3′-羟基宝藿苷-Ⅱ, 为新化合物, 数据归属见表 1
Bruker AVANCE Ⅲ 500型核磁共振仪和Bruker maxis HD型飞行时间质谱(德国布鲁克公司); Thermo EVO300型紫外检测器和Thermo Nicolet IS10红外光谱仪(美国Thermo Scientific公司); Autopol Ⅳ全自动旋光仪(美国鲁道夫公司); Alliance HPLC-高效液相色谱仪蒸发光散射检测器(美国Waters公司); 赛谱锐思LC-52型高压制备液相色谱仪(赛谱锐思北京科技有限公司); OSB-21000型旋转蒸发仪(上海爱朗仪器有限公司); YMC-Pack ODS-A色谱柱(250 mm × 20 mm, 5 μm, 日本YMC有限公司); 柱色谱填料Sephadex LH-20 (GE Healthcare公司); Toyopearl HW-40C/F (日本TOSOH公司); ODS (50 μm, 日本YMC有限公司); 大孔树脂Diaion HP-20、MCI gel CHP-20 (日本三菱化学公司); 薄层色谱硅胶GF 254、柱色谱硅胶(100~200目, 200~300目, 青岛海洋化工厂); 甲醇(色谱纯, 天津四友精细化学品有限公司); 乙腈(色谱纯, 美国天地有限公司); 所用其他分析纯试剂均为天津市富宇精细化工有限公司及天津市致远化学试剂有限公司生产。
箭叶淫羊藿采自河南省驻马店市平舆丰惠淫羊藿种植基地, 经河南中医药大学药学院董诚明教授鉴定为淫羊藿属箭叶淫羊藿Epimedium sagittatum。植物标本(No.20200960) 保存于河南中医药大学河南省中药开发工程技术研究中心。
箭叶淫羊藿(80 kg) 进行粉碎, 用70%乙醇加热回流提取2次, 减压浓缩得到浸膏。浸膏加等量水溶解, 依次用石油醚、二氯甲烷、乙酸乙酯、正丁醇萃取, 回收溶剂后得到石油醚部位150 g、二氯甲烷部位2 kg、乙酸乙酯部位500 g、正丁醇部位4 kg。正丁醇部位上大孔吸附树脂柱DaionHP-20, 依次用水、10%乙醇、20%乙醇、30%乙醇、40%乙醇、50%乙醇、70%乙醇、95%乙醇洗脱, 依次得到对应的各个组分计作Fr.1~Fr.8。
Fr.2加水溶解, 上Sephadex LH-20凝胶柱, 以甲醇-水(0∶100→100∶0) 梯度洗脱, 通过薄层色谱检识, 合并相同流分得到5个组分(Fr.2-1~Fr.2-5)。Fr.2-5进行Sephadex LH-20凝胶柱色谱分离, 以甲醇-水(70∶30) 等度洗脱, 经薄层色谱检识得到6个组分(Fr.2-5-1~Fr.2-5-6)。Fr.2-5-4经硅胶色谱柱分离, 用二氯甲烷-甲醇(20∶1) 洗脱得到Fr.2-5-4-1~Fr.2-5-4-7。Fr.2-5-4-2运用半制备高效液相(乙腈-水, 19∶81) 等度洗脱, 得到化合物7 (tR = 45.3 min, 1.60 mg)。Fr.2-5-4-6运用半制备高效液相(甲醇-水, 60∶40→80∶20) 梯度洗脱得到化合物3 (tR = 12.0 min, 7.35 mg)、4 (tR = 27.5 min, 27.47 mg) 和1 (tR = 40.1 min, 8.56 mg)。
Fr.3经硅胶色谱柱分离, 用二氯甲烷-甲醇(20∶1) 洗脱得到9个组分(Fr.3-1~Fr.3-9)。Fr.3-5依次经硅胶色谱柱、Toyopearl HW-40C凝胶柱色谱分离得到Fr.3-5-4-8。Fr.3-5-4-8运用半制备高效液相(甲醇-水, 70∶30) 等度洗脱, 得到化合物5 (tR = 28.1 min, 5.86 mg)。Fr.3-8进行Sephadex LH-20凝胶柱色谱分离, 以甲醇-水(70∶30) 等度洗脱, 得到Fr.3-8-1~Fr.3-8-5。Fr.3-8-1依次进行MCI、ODS柱色谱分离, 并运用半制备高效液相(甲醇-水, 55∶45) 等度洗脱, 得到化合物14 (tR = 37.2 min, 25.26 mg)。Fr.3-8-3经硅胶色谱柱用二氯甲烷-甲醇(10∶1) 洗脱得到组分Fr.3-8-3-1~Fr.3-8-3-5。Fr.3-8-3-3运用半制备高效液相(乙腈-水, 17∶83) 等度洗脱, 得到化合物11 (tR = 33.0 min, 4.24 mg)。Fr.3-8-3-4进行Toyopearl HW-40C凝胶柱色谱分离, 以甲醇-水(50∶50) 等度洗脱, 经薄层色谱检识, 合并相同流分得到7组分, 即Fr.3-8-3-4-1~Fr.3-8-3-4-7。Fr.3-8-3-4-4运用半制备高效液相(乙腈-水, 20∶80) 等度洗脱, 得到化合物6 (tR = 13.2 min, 47.82 mg); Fr.3-8-3-4-6运用半制备高效液相(甲醇-水, 45∶55→70∶30) 梯度洗脱, 得到化合物8 (tR = 30.0 min, 10.48 mg)、9 (tR = 68.1 min, 4.91 mg) 和10 (tR = 27.5 min, 46.43 mg)。Fr.3-8-4进行Sephadex LH-20凝胶柱色谱分离, 以甲醇-水(70∶30)等度洗脱, 经薄层色谱检识, 合并相同流分得到7组分, 即Fr.3-8-4-1~Fr.3-8-4-7。Fr.3-8-4-7运用半制备高效液相(乙腈-水, 17∶83) 等度洗脱, 得到化合物2 (tR = 30.0 min, 23.56 mg)。Fr.3-9依次经Sephadex LH-20凝胶、硅胶、Toyopearl HW-40C凝胶柱色谱分离, 得到Fr.3-9-5-4-4。Fr.3-9-5-4-4运用半制备高效液相(乙腈-水, 22∶78) 等度洗脱, 得到化合物12 (tR = 30.1 min, 4.53 mg) 和13 (tR = 48.0 min, 19.89 mg)。
称取化合物1 (2.0 mg), 加入2 mol·L-1 HCl溶液2 mL, 密闭, 水浴加热(80 ℃), 反应3 h; 反应完毕后, 静置放冷, 氮吹仪吹干样品。然后加入蒸馏水(2 mL), 用等体积乙酸乙酯萃取3次, 合并水相层, 采用以下条件进行高效液相色谱分析: 采用CHIRALPAK AD-H色谱柱(250 mm × 4.6 mm), 以正己烷-乙醇-三氟乙酸(750∶250∶0.25) 为流动相(0.5 mL·min-1), 蒸发光散射检测器(ELSD, 漂移管温度: 75 ℃; 气体压力: 20.0 psi; 柱温: 30 ℃) 对化合物1的水解产物进行分离检测, 通过比较相同液相条件下L-鼠李糖的保留时间, 两者保留时间一致(tR = 21.3 min), 最终确定化合物1中单糖为L-鼠李糖。
