To investigate the metabolites of a new synthetic cannabinoid 3, 3-dimethyl-2-[1-(4-cyanobutyl)indazole-3-formamimino]methyl butyrate (4CN-MDMB-BUTINACA) in vitro, a human liver microsome incubation model was established to analyze the metabolic biotransformation of synthetic cannabinoids using ultra-high performance liquid chromatography coupled to quadrupole-orbitrap high-resolution mass spectrometry. Nontarget metabolomic results showed that the metabolites of 4CN-MDMB-BUTINACA included hydroxylation, ester hydrolysis, ester hydrolysis with hydroxylation reaction, pentane oxidation and ester hydrolysis with pentane oxidation reaction, among which M1-a, M2 and M4 were potential metabolic markers. The research results provide a theoretical basis and technical support for the biomonitoring and metabolic characterisation of the cannabinoid 4CN-MDMB-BUTINACA.

为研究新型合成大麻素3, 3-二甲基-2-[1-(4-氰基丁基)吲唑-3-甲酰氨基]丁酸甲酯(4CN-MDMB-BUTINACA) 的体外代谢产物, 本研究通过体外人肝微粒体孵育模型, 采用高效液相色谱串联Q exactive质谱检测合成大麻素原形药及其代谢产物。肝微粒体实验发现4CN-MDMB-BUTINACA通过羟基化、酯水解、酯水解加羟基化、戊烷氧化为戊酸反应代谢物和酯水解加戊烷氧化为戊酸反应代谢物等代谢途径共产生7种代谢产物。本研究解析了合成大麻素体外代谢途径及其代谢产物, 其中羟基化反应代谢产物(M1-a)、酯水解反应代谢产物(M2) 以及戊烷氧化为戊酸反应代谢物(M4) 为潜在的代谢标志物。研究结果为合成大麻素4CN-MDMB-BUTINACA的司法鉴定和吸食认定提供技术支撑。

为研究新型合成大麻素3, 3-二甲基-2-[1-(4-氰基丁基)吲唑-3-甲酰氨基]丁酸甲酯(4CN-MDMB-BUTINACA) 的体外代谢产物, 本研究通过体外人肝微粒体孵育模型, 采用高效液相色谱串联Q exactive质谱检测合成大麻素原形药及其代谢产物。肝微粒体实验发现4CN-MDMB-BUTINACA通过羟基化、酯水解、酯水解加羟基化、戊烷氧化为戊酸反应代谢物和酯水解加戊烷氧化为戊酸反应代谢物等代谢途径共产生7种代谢产物。本研究解析了合成大麻素体外代谢途径及其代谢产物, 其中羟基化反应代谢产物(M1-a)、酯水解反应代谢产物(M2) 以及戊烷氧化为戊酸反应代谢物(M4) 为潜在的代谢标志物。研究结果为合成大麻素4CN-MDMB-BUTINACA的司法鉴定和吸食认定提供技术支撑。

To investigate the metabolites of a new synthetic cannabinoid 3, 3-dimethyl-2-[1-(4-cyanobutyl)indazole-3-formamimino]methyl butyrate (4CN-MDMB-BUTINACA) in vitro, a human liver microsome incubation model was established to analyze the metabolic biotransformation of synthetic cannabinoids using ultra-high performance liquid chromatography coupled to quadrupole-orbitrap high-resolution mass spectrometry. Nontarget metabolomic results showed that the metabolites of 4CN-MDMB-BUTINACA included hydroxylation, ester hydrolysis, ester hydrolysis with hydroxylation reaction, pentane oxidation and ester hydrolysis with pentane oxidation reaction, among which M1-a, M2 and M4 were potential metabolic markers. The research results provide a theoretical basis and technical support for the biomonitoring and metabolic characterisation of the cannabinoid 4CN-MDMB-BUTINACA.

新精神活性物质又称为“策划药”, 一般通过对现有毒品的化学结构进行修改获得, 不但具有类似列管毒品的麻醉、兴奋或致幻作用, 而且能逃避法律的管制, 因此此类物质的生产、贩卖和滥用形势日益严峻[1]。合成大麻素是目前全球种类最多、滥用最为严重的新精神活性物质。合成大麻素种类繁多、化学结构变化多样, 十余年时间已从第一代萘甲酰胺类合成大麻素发展至第八代吲哚/吲唑类合成大麻素[2]。过量摄入合成大麻素会导致焦虑、幻觉、思维混乱等症状, 产生暴力行为、自杀倾向, 严重危害人体健康和社会安全稳定[3]。近年来, 合成大麻素通常添加在电子烟油中以合法商品的外观出现在非法交易市场上[4-6]。在我国越来越多的省市出现添加了合成大麻素的“上头电子烟”案件, 对人民群众特别是青少年的身体健康造成了巨大的安全隐患。
3, 3-二甲基-2-[1-(4-氰基丁基)吲唑-3-甲酰氨基]丁酸甲酯(4CN-MDMB-BUTINACA, 图 1), 属于吲唑甲酰胺类的新型合成大麻素, 以氰丁基和3, 3-二甲基丁酸甲酯为侧链, 是国内首次发现的新型合成大麻素衍生物。由于吲唑甲酰胺类合成大麻素具有吸食量低、代谢速度快, 尿液中几乎无原形药检出等特点, 给司法鉴定和毒品犯罪认定带来了极大的困难[7]。因此, 急需确定这类物质的代谢途径和代谢标志物。由于人体阳性样本收集困难, 对真实案件进行代谢分析的报道较少[8, 9]。动物实验可用于代谢分析, 但其操作复杂、成本较高[10, 11]。肝微粒体体外代谢实验是评价新精神活性物质代谢物及其代谢途径的重要方式, 具有简便、快捷、成本低等优势, 已广泛应用于法庭科学领域[12-15]。目前缺乏4CN-MDMB-BUTINACA的信息, 代谢物的详细信息更是鲜有报道。
本研究通过人肝微粒体模型结合高效液相色谱串联Q-exactive组合型四级杆orbitrap高分辨质谱技术分析了4CN-MDMB-BUTINACA体外Ⅰ相代谢产物及代谢途径, 并确定了潜在的代谢标志物, 从而为新型合成大麻素4CN-MDMB-BUTINACA的相关案件检验鉴定提供技术支撑。
仪器  Scientific Dionex Ultimate 3000超高效液相色谱串联四极杆-静电场轨道离子阱Q Exactive质谱、ST16R高速离心机和反渗透纯水机, 美国Thermo Fisher公司; KH-250DE型数控超声波清洗器, 昆山禾创超声仪器有限公司。
材料与试剂  4CN-MDMB-BUTINACA对照品(上海市刑事科学技术研究院); 男性肝微粒体(20 mg·mL-1, 上海瑞德肝脏疾病研究有限公司); 氧化型辅酶Ⅱ钠盐(NADPNa2)、葡萄糖-6-磷酸脱氢酶(G-6-PDH)、6-磷酸葡萄糖二钠(G-6-P), 北京索莱宝科技有限公司; 二甲基亚砜(≥ 99.8%), 美国西格玛公司; MgCl2 (≥ 99.0%), 上海沪试试剂公司; 磷酸盐缓冲溶液(PBS, 7.6 > pH > 7.2), 博士德生物有限公司; 乙腈(HPLC级), 美国Merck公司; 甲酸(HPLC级), 美国ROE Scientific INC公司。
色谱与质谱条件  色谱柱为Waters T3 (100 mm × 2.1 mm, 1.8 μm), 流动相A为0.1%甲酸水溶液、B为0.1%甲酸乙腈溶液; 梯度洗脱程序为0~1 min, 5% B; 1~8.5 min, 5%~100% B; 8.5~11 min, 100% B; 11~11.1 min, 100%~5% B; 11.1~13 min, 5% B。柱温为30 ℃, 流速为0.3 mL·min-1, 进样量为2 µL。采用电喷雾电离源(ESI), 离子源参数: 喷射电压: 3 800 V, 碰撞气: 氮气, 雾化温度: 320 ℃, 雾化气压力: 35 arb, 辅助气压力: 15 arb, 毛细管温度: 350 ℃。采用Full MS-ddMS2扫描模式采集数据; 全扫描数据采集参数, Full MS分辨率: 140 000, 最大注射时间(maximum IT): 50 ms, 扫描范围: m/z 160~600; 二级质谱数据采集参数, 分辨率: 35 000, 碰撞能量为25 eV。
肝微粒体孵育实验  取10 µL G-6-P、10 µL NADPNa2、8 µL G-6-PDH和66 µL MgCl2混合均匀, 37 ℃金属浴孵育5 min, 获得NADPH再生系统。另取1 mg·mL-1 4CN-MDMB-BUTINACA对照品溶液1 µL, 加入PBS缓冲盐溶液96 µL和20 mg·mL-1肝微粒体10 µL, 混合均匀后37 ℃金属浴条件下预孵育5 min, 然后加入NADPH再生系统混合均匀, 开始启动体外孵育反应, 37 ℃金属浴孵育60 min。待孵育结束之后向孵育体系内加入乙腈200 µL, 涡旋2 min, 10 000 r·min-1离心10 min, 取上清液200 µL待测。分别将未加入4CN-MDMB-BUTINACA和未加入肝微粒体的两组孵育系统作为对照分析, 计算4CN-MDMB-BUTINACA在人肝微粒体孵化体系中的代谢转化率以及在阴性对照样品中的稳定性。
4CN-MDMB-BUTINACA分子式为C20H26N4O3, 在正离子模式下, [M+H]+m/z 371.207 70, 图 2为4CN-MDMB-BUTINACA带有特征碎片结构的质谱图。二级质谱显示[M+H]+离子经裂解丢失一分子C2H4O形成碎片离子m/z 311.186 58, 进一步发生酰胺键的断裂并丢失一分子C5H11N形成碎片离子m/z 226.097 47。[M+H]+离子另一条裂解路径是N1-C1b键发生断裂并丢失一分子C5H7N形成碎片离子m/z 145.039 55。详细质谱裂解途径如图 3所示。
采用超高效液相质谱进行分析, 发现并鉴定了7种4CN-MDMB-BUTINACA代谢产物, 其中羟基化代谢物2种、酯水解反应代谢物1种、酯水解和羟基化代谢物2种、戊烷氧化为戊酸反应代谢物1种、酯水解和戊烷氧化为戊酸反应代谢物1种。4CN-MDMB-BUTINACA在人肝微粒体孵化体系中的代谢转化率为98.94%, 在阴性对照样品中的稳定性为94.54%。
代谢产物提取离子流图见图 4, 在正离子模式下获得的4CN-MDMB-BUTINACA中7个代谢物的一级质谱数据及其二级质谱碎片离子信息见表 1图 5为7种代谢物的质谱图。
M1-a和M1-b的保留时间分别为7.38和8.01 min, 一级质谱图中m/z 387.202 68为[M+H]+离子, 分子式为C20H26N4O4。M1-a和M1-b比原形药4CN-MDMB-BUTINACA增加了16 Da。图 5A为M1-a的二级质谱图, 出现的主要碎片离子为m/z 355.176 36、327.181 52和226.097 46; M1-a产生碎片离子m/z 327.181 52相对于原形药产生的碎片离子m/z 311.186 58增加16 Da, 而碎片离子m/z 226.097 46对应原形药m/z 226.097 47的碎片离子未发生变化, 碎片离子297.170 96是由碎片离子m/z 327.181 52的侧链戊烷结构继续丢失一分子HCHO, 减少30 Da得到的。因此, 推测M1-a为原形药在C-3a侧链戊烷结构上发生羟基化反应, 而1-氰丁基-1H-吲唑-3-甲酰结构没有发生改变。图 5B为M1-b的二级质谱图, 二级质谱图中M1-b主要的碎片离子为m/z 355.176 48、327.181 55、242.092 38和145.039 69, 其中碎片离子m/z 145.039 69对应原形药中的碎片离子m/z 145.039 55, 推测1H-吲唑-3-甲酰结构并未发生变化, 而碎片离子m/z 355.176 48、327.181 55和242.092 38相较于原形药产生的碎片离子m/z 339.181 52、311.186 58和226.097 47均增加16 Da。因此, 推测M1-b为原形药的N1侧链氰丁基结构上发生羟基化反应。
M2的保留时间为7.55 min, 一级质谱图中m/z 357.192 12为[M+H]+离子, 分子式为C19H24N4O3, 相对原形药少了一个CH2图 5C为M2的二级质谱图, 主要的碎片离子为m/z 311.186 55、226.097 46和145.039 54。M2碎片离子m/z 311.186 55、226.097 46和145.039 54分别对应原形药中的碎片离子m/z 311.186 58、226.097 47和145.039 55, 而M2质子化离子的m/z为357.192 12与原形药相比减少14 Da, 这是由于酯基8a处发生水解缺失CH2产生的, M2其他碎片离子均与原形药相同, 酯基以外的结构未发生改变。因此, 推测M2为原形药的酯基8a处发生水解反应。
M3-a和M3-b的保留时间分别为6.63和6.96 min, 一级质谱图中m/z 373.187 03为[M+H]+离子, 分子式为C19H24N4O4图 5D为M3-a的二级质谱图, 主要的碎片离子是m/z 327.181 15、297.171 05、226.097 52, 其中m/z 327.181 15相较于原形药产生的二级碎片m/z 311.186 58增加16 Da, 碎片离子297.171 05是由碎片离子m/z 327.181 15的侧链戊烷结构继续丢失一分子HCHO, 减少30 Da得到的。m/z 226.097 52对应原形药中的二级碎片m/z 226.097 47, 故推测在C-3a侧链戊烷结构上发生羟基化反应。M3-a质子化离子的m/z为373.187 03与代谢物M1-a相比减少14 Da, 且M3-a产生的碎片离子m/z 327.181 15、297.171 05、226.097 52与M1-a生成的碎片离子一致, 故推测酯基8a处发生水解失去一分子CH2。因此, 推测M3-a为原形药的酯基8a处发生水解同时C-3a侧链戊烷结构发生羟基化形成的。图 5E为M3-b的二级质谱图, 二级质谱图中M3-b主要的碎片离子为m/z 355.175 72、327.181 27、242.092 16和145.039 72, 其中碎片离子m/z 355.175 72、327.181 27和242.092 16相较于原形药产生的二级碎片离子均增加16 Da, m/z 145.039 72对应原形药中的二级碎片m/z 145.039 55。M3-b质子化离子的m/z为373.186 89与M1-b相比减少14 Da, 且M3-b产生的碎片离子m/z 355.175 72、327.181 27、242.092 16和145.039 72与M1-b生成的碎片离子一致, 故推测酯基8a处发生水解失去一分子CH2。因此, 推测M3-b为原形药的酯基8a处发生水解同时N1侧链氰丁基结构上发生羟基化反应形成的。
M4的保留时间为7.20 min, 一级质谱图中m/z 401.181 95为[M+H]+离子, 分子式为C20H24N4O5, M4比原形药增加30 Da。图 5F为M4的二级质谱图, 主要的碎片离子为m/z 369.155 82、341.160 77和226.097 46。M4碎片离子m/z 369.155 82和m/z 341.160 77与原形药碎片离子相比增加30 Da, 而碎片离子m/z 226.097 46对应原形药产生的二级碎片离子m/z 226.097 47未发生变化。因此, 推测M4为原形药的C-3a侧链戊烷结构发生氧化反应形成羧酸。
M5的保留时间为6.67 min, 一级质谱图中m/z 387.166 30为[M+H]+离子, 分子式为C19H22N4O5图 5G为M5的二级质谱图, M5主要的碎片离子为m/z 369.192 05、226.097 46和145.039 46。其中M5碎片离子 m/z 226.097 46对应原形药的碎片离子m/z 226.097 47, 推测该结构未发生变化。M5碎片离子m/z 369.155 82和m/z 341.160 77与原形药的碎片离子相比增加30 Da, 推测C-3a侧链戊烷结构发生了氧化形成了羧酸。M5质子化离子m/z 387.166 30与M4相比减少14 Da, 且代谢物M5产生的碎片离子m/z 369.192 05、226.097 46与代谢物M4生成的碎片离子一致, 故推测酯基8a处发生水解失去一分子CH2。因此, 推测代谢物M5为原形药的酯基8a处发生水解同时C-3a侧链戊烷结构发生氧化反应形成羧酸。
4CN-MDMB-BUTINACA的Ⅰ相代谢物主要通过羟基化形成, 在代谢过程中4CN-MDMB-BUTINACA原形药首先发生羟基化, 分别在C-3a侧链戊烷结构和N1侧链氰丁基位点发生羟基化形成M1-a和M1-b。羟基化反应形成的M1-a进一步氧化成羧基, 形成M4。4CN-MDMB-BUTINACA的酯基结构在代谢过程中易发生水解形成酯水解代谢物, 原形药的酯基8a处发生酯水解后形成M2, M2为含量最高的代谢产物。M3-a和M3-b分别为酯水解代谢物M2进一步在C-3a侧链戊烷和N1侧链氰丁基结构位点上发生羟基化所得。M3-a在羟基化位点上进一步氧化成羧基形成M5。图 6为4CN-MDMB-BUTINACA代谢途径。
4CN-MDMB-BUTINACA的代谢途径主要是羟基化、酯水解和酯水解加羟基化等。经过查阅相关文献, 本研究发现的7种代谢产物尚未有报道。通过比较实验鉴定的7种代谢物色谱峰面积的大小, M1-a、M2和M4代谢物的色谱峰面积较大, 为含量最高的3种代谢物。推荐M1-a、M2及M4作为4CN-MDMB-BUTINACA的代谢标志物。今后研究可以通过实际案件中收集到的阳性生物检材加以验证。
本研究通过人肝微粒体模型结合超高效液相色谱-Q-exactive组合型四级杆orbitrap质谱分析新型合成大麻素4CN-MDMB-BUTINACA的体外代谢产物及代谢途径, 并探寻其潜在的生物标记物。结果显示4CN-MDMB-BUTINACA以羟基化、酯水解和氧化等代谢途径产生的7种代谢产物, 其中羟基化反应代谢产物(M1-a)、酯水解反应代谢产物(M2) 以及戊烷氧化为戊酸反应代谢物(M4) 可作为潜在的代谢标志物。本研究为合成大麻素类新型毒品的体内检测及代谢转化提供技术支撑。
