Fifteen compounds were isolated from fruits of Cornus officinalis by various chromatographic techniques such as Toyopearl HW-40C, Sephadex LH-20, silica gel, and the semi-preparative HPLC. Their chemical structures were identified by analysis of physicochemical properties and spectral data, and determined as neolignan A (1), caffeic acid (2), trans-p-hydroxy cinnamic acid (3), esculetin (4), scopoletin (5), benzyl-7-O-β-D-glucopyranoside (6), tachioside (7), 6-O-(4-hydroxybenzoyl) arbutin (8), 2-(3′, 4′-dihydroxyphenyl)-1, 3-benzodioxole-5-carboxaldehyde (9), (-)-pinoresinol-4-O-β-D-glucopyranoside (10), (7S, 8R)-dihydrodehydrodiconiferyl alcohol-9-O-β-D-glucopyranoside (11), (7S, 8R)-dihydrodehydrodiconiferyl alcohol-9′-O-β-D-glucopyranoside (12), (+)-lyoniresinol (13), (+)-isolariciresinol-9-O-β-D-glucopyranoside (14), and isolariciresinol-9′-O-β-D-glucopyranoside (15). Compound 1 was a new compound and named as neolignan A, and compounds 6-9 and 14 were isolated from Cornus officinalis for the first time. Compounds 2, 3 and 15 efficiently alleviated the PC12 cells injury induced by Aβ25-35, suggesting their potential anti-Alzheimer's disease activity.

采用Toyopearl HW-40C、Sephadex LH-20、硅胶和半制备液相等多种色谱学技术从山茱萸果实中分离得到15个化合物。根据理化性质与波谱数据鉴定了其结构, 分别为新木脂素A (1)、咖啡酸(2)、反式对羟基桂皮酸(3)、马栗树皮素(4)、东莨菪素(5)、benzyl-7-O-β-D-glucopyranoside (6)、它乔糖苷(7)、6-O-(4-hydroxybenzoyl) arbutin (8)、2-(3′, 4′-二羟苯基)-1, 3-胡椒环-5-醛(9)、(-)-松脂素-4-O-β-D-吡喃葡萄糖苷(10)、(7S, 8R)-二氢去氢二愈创木基醇-9-O-β-D-吡喃葡萄糖苷(11)、(7S, 8R)-二氢去氢二松柏醇-9′-O-β-D-吡喃葡萄糖苷(12)、(+)-南烛木树脂酚(13)、(+)-异落叶松树脂醇-9-O-β-D-吡喃葡萄糖苷(14)、异落叶松树脂醇-9′-O-β-D-吡喃葡萄糖苷(15)。化合物1为新化合物, 命名为新木脂素A, 化合物6~914首次从山茱萸中分离得到。化合物2315可以有效缓解Aβ25-35诱导的PC12细胞损伤, 表明具有潜在的抗阿尔茨海默症活性。

采用Toyopearl HW-40C、Sephadex LH-20、硅胶和半制备液相等多种色谱学技术从山茱萸果实中分离得到15个化合物。根据理化性质与波谱数据鉴定了其结构, 分别为新木脂素A (1)、咖啡酸(2)、反式对羟基桂皮酸(3)、马栗树皮素(4)、东莨菪素(5)、benzyl-7-O-β-D-glucopyranoside (6)、它乔糖苷(7)、6-O-(4-hydroxybenzoyl) arbutin (8)、2-(3′, 4′-二羟苯基)-1, 3-胡椒环-5-醛(9)、(-)-松脂素-4-O-β-D-吡喃葡萄糖苷(10)、(7S, 8R)-二氢去氢二愈创木基醇-9-O-β-D-吡喃葡萄糖苷(11)、(7S, 8R)-二氢去氢二松柏醇-9′-O-β-D-吡喃葡萄糖苷(12)、(+)-南烛木树脂酚(13)、(+)-异落叶松树脂醇-9-O-β-D-吡喃葡萄糖苷(14)、异落叶松树脂醇-9′-O-β-D-吡喃葡萄糖苷(15)。化合物1为新化合物, 命名为新木脂素A, 化合物6~914首次从山茱萸中分离得到。化合物2315可以有效缓解Aβ25-35诱导的PC12细胞损伤, 表明具有潜在的抗阿尔茨海默症活性。

Fifteen compounds were isolated from fruits of Cornus officinalis by various chromatographic techniques such as Toyopearl HW-40C, Sephadex LH-20, silica gel, and the semi-preparative HPLC. Their chemical structures were identified by analysis of physicochemical properties and spectral data, and determined as neolignan A (1), caffeic acid (2), trans-p-hydroxy cinnamic acid (3), esculetin (4), scopoletin (5), benzyl-7-O-β-D-glucopyranoside (6), tachioside (7), 6-O-(4-hydroxybenzoyl) arbutin (8), 2-(3′, 4′-dihydroxyphenyl)-1, 3-benzodioxole-5-carboxaldehyde (9), (-)-pinoresinol-4-O-β-D-glucopyranoside (10), (7S, 8R)-dihydrodehydrodiconiferyl alcohol-9-O-β-D-glucopyranoside (11), (7S, 8R)-dihydrodehydrodiconiferyl alcohol-9′-O-β-D-glucopyranoside (12), (+)-lyoniresinol (13), (+)-isolariciresinol-9-O-β-D-glucopyranoside (14), and isolariciresinol-9′-O-β-D-glucopyranoside (15). Compound 1 was a new compound and named as neolignan A, and compounds 6-9 and 14 were isolated from Cornus officinalis for the first time. Compounds 2, 3 and 15 efficiently alleviated the PC12 cells injury induced by Aβ25-35, suggesting their potential anti-Alzheimer's disease activity.

山茱萸为山茱萸科植物山茱萸(Cornus officinalis) 的干燥成熟果肉, 广泛分布于河南、陕西、浙江等地[1]。其性味酸、涩、微温, 临床中一般用于治疗腰膝酸软、大汗虚脱、遗精遗尿等滑脱之症, 在《神农本草经》《汤液本草》《普济方》等中医典籍中均有详细记载, 是我国名贵的滋补类药材[2]。现代药理学研究表明, 山茱萸具有降血糖[3]、改善认知能力[4]、抗炎[5]等功效。此外, 相关研究表明, 山茱萸主要含有环烯醚萜及其苷类、三萜、有机酸、黄酮、鞣质等多种有效成分[6, 7]。为了进一步阐明山茱萸的药效物质基础, 促进其作为药食两用中药的充分合理利用, 本实验对山茱萸中的化学成分进行系统分离与鉴定, 得到15个化合物, 包括7个木脂素类化合物(110~15)、2个苯丙素类化合物(23)、2个香豆素类化合物(45)、4个酚酸类化合物(6~9), 结构见图 1。化合物1为新化合物, 命名为新木脂素A, 化合物6~914首次从山茱萸中分离得到。体外抗阿尔茨海默症活性筛选实验表明, 化合物2315对Aβ25-35诱导的PC12细胞损伤具有显著的保护作用。
化合物1  无色蜡状物, 易溶于甲醇。HR-ESI-MS谱在m/z 485.179 8 [M+Na]+ (Calcd. 485.178 2) 处显示该化合物的准分子离子峰, 确定分子式为C24H30O9, 计算不饱和度为10。IR图谱中显示有羟基(3 398 cm-1) 和双键(1 622 cm-1) 基团特征吸收峰。1H NMR (CD3OD, 500 MHz) 谱(表 1) 中, 在δH 7.15 (1H, br s, H-2′)、7.00 (1H, br d, J = 7.9 Hz, H-6′)、6.70 (1H, d, J = 7.9 Hz, H-5′) 显示一组苯环ABX系统特征信号峰; δH 7.22 (2H, d, J = 8.7 Hz, H-3, 5) 和6.74 (2H, d, J = 8.7 Hz, H-2, 6) 处的质子信号峰表明化合物中存在一个对位二取代苯环; δH 5.53 (1H, d, J = 5.8 Hz, H-7) 和3.60 (1H, m, H-8) 处为苯骈呋喃型木脂素7位和8位质子特征信号峰; δH 4.19 (1H, dd, J = 9.7, 5.8 Hz, H-9a)、3.74 (1H, dd, J = 9.7, 7.8 Hz, H-9b)、3.56 (2H, t, J = 6.6 Hz, H-9′) 处为两个连氧亚甲基质子信号峰; δH 2.62 (2H, t, J = 7.7 Hz, H-7′) 和1.80 (2H, m, H-8′) 为两个不连氧亚甲基质子信号峰; δH 4.35 (1H, d, J = 7.8 Hz, H-1″)、3.87 (1H, m, H-6″a)、3.66 (1H, dd, J = 12.1, 4.7 Hz, H-6″b) 以及δH 3.23-3.34 (4H, m, H-2″-5″) 处质子信号峰提示该化合物中含有一个糖的结构片段。13C NMR (CD3OD, 125 MHz) (表 1) 和DEPT-135谱显示22个碳信号, 结合HSQC谱可知, δC 159.3 (C-4′)、158.3 (C-4)、135.6 (C-1′)、134.3 (C-1)、129.8 (C-6′)、128.7 (C-3′)、128.3 (C-3, 5)、126.2 (C-2′)、116.1 (C-2, 6)、109.7 (C-5′) 处为两组苯环上的碳信号。δC 104.6 (C-1″)、78.2 (C-3″)、78.1 (C-5″)、75.1 (C-2″)、71.6 (C-4″)、62.8 (C-6″) 处为一组葡萄糖上碳特征信号峰。δC 88.3 (C-7) 和52.7 (C-8) 处为苯骈呋喃型木脂素7位和8位特征碳信号峰, δC 72.6 (C-9)、62.3 (C-9′)、32.5 (C-7′)、35.9 (C-8′) 处为四个亚甲基碳信号峰。以上数据与文献[8]报道的3-[2-(4-hydroxyphenyl)-3-hydroxymethyl-2, 3-dihydro-1-benzofuran-5-yl]propan-1-ol核磁数据对比后, 仅在C-8位(1, δC 52.7; 3-[2-(4-hydroxyphenyl)-3-hydroxymethyl-2, 3-dihydro-1-benzofuran-5-yl]propan-1-ol, δC 54.8) 和C-9位(1, δC 72.6; 3-[2-(4-hydroxyphenyl)-3-hydroxymethyl-2, 3-dihydro-1-benzofuran-5-yl]propan-1-ol, δC 65.1) 的化学位移存在差异, 且化合物1多了一个糖结构片段。在HMBC谱(图 2) 中, 葡萄糖端基氢δH 4.11 (H-1″) 与C-9 (δC 72.6) 远程相关, 表明葡萄糖连在C-9位上。化合物1经水解后, 水解产物经手性HPLC分析, 表明该化合物中的糖为D-葡萄糖, 且根据端基质子的偶合常数(J = 7.8 Hz) 确定其为β构型。由氢谱中H-7和H-8的偶合常数(J = 5.8 Hz) 推测H-7、H-8的相对构型为反式[9]。查阅文献[10-13]发现, 具有二氢苯骈呋喃骨架的C-7和C-8绝对构型可根据ECD谱中240~220 nm处吸收进行区分, 当ECD谱在220 nm附近显示负Cotton效应, 240 nm附近显示正Cotton效应, 绝对构型为7S, 8R; 在220 nm附近显示正Cotton效应, 240 nm附近显示负Cotton效应, 绝对构型为7R, 8S。化合物1的ECD谱在221 nm处显示正Cotton效应, 235 nm处显示负Cotton效应, 确定该化合物的绝对构型为7R, 8S。通过对比实测ECD和计算ECD曲线, 进一步证实化合物1的绝对构型为7R, 8S (图 3)。基于以上分析, 确定化合物1结构为新木脂素A, 为1个新化合物, 数据归属见表 1
通过对化合物1~15进行抗阿尔茨海默症活性筛选, 结果如表 2所示。与正常组相比, 模型组细胞活力显著降低(P < 0.01); 与模型组相比, 化合物2315可显著提高PC12的细胞活力(P < 0.05或P < 0.01), 表明化合物2315对Aβ25-35诱导的PC12细胞损伤具有显著的保护作用, 具有潜在的抗阿尔茨海默症活性。

山茱萸于2019年9月采集于河南省西峡县, 经药学院董诚明教授鉴定为山茱萸科山茱萸属植物山茱萸(Cornus officinalis) 的果肉, 植物标本(No.20190903) 保存于河南中医药大学中药化学实验室。
取山茱萸干燥成熟果肉100.0 kg, 用10倍量水煮沸提取两次, 每次2 h。合并提取液, 减压浓缩后得到总浸膏55.1 kg。总浸膏用适量去离子水溶解后, 经D101大孔吸附树脂, 依次用水和95%乙醇进行洗脱, 95%乙醇洗脱液经减压浓缩后得浸膏21.7 kg, 用适量水分散后, 依次用石油醚、乙酸乙酯、正丁醇萃取, 回收溶剂后得到石油醚部位1.5 g、乙酸乙酯部位99.3 g、正丁醇部位4.15 kg和水部位17.4 kg。
乙酸乙酯部位(99.3 g) 经Diaion HP-20柱分离, 依次用水、20%乙醇、40%乙醇、60%乙醇和95%乙醇进行梯度洗脱, 每个梯度洗脱4个柱体积, TLC检识并合并相同流分得到5个组分Fr.1~Fr.5。Fr.3 (19.6 g) 经Sephadex LH-20凝胶柱分离, 依次用20%甲醇、50%甲醇和100%甲醇进行梯度洗脱, 得到5个组分Fr.3-1~Fr.3-5。Fr.3-2 (460.0 mg) 经半制备型高效液相色谱分离(22% MeOH, 3 mL·min-1) 得到化合物9 (tR = 16.3 min, 10.1 mg)。Fr.3-3 (860.2 mg) 经半制备型高效液相色谱分离(15% MeCN, 3 mL·min-1) 得到化合物3 (tR = 23.3 min, 79.7 mg)。Fr.3-1 (16.0 g) 经硅胶柱色谱分离, 以石油醚-丙酮(3∶1→0∶1) 梯度洗脱得到6个组分, 即Fr.3-1-1~Fr.3-1-6。Fr.3-1-4 (105.2 mg) 经半制备型高效液相色谱分离(14% MeCN, 3 mL·min-1) 得到化合物6 (tR = 15.3 min, 5.8 mg)。Fr.2 (13.26 g) 经Sephadex LH-20柱色谱分离, 依次用20%甲醇、50%甲醇和100%甲醇洗脱, 得到5个组分Fr.2-1~Fr.2-5。Fr.2-3 (80.0 mg) 经半制备型高效液相色谱分离(11% MeOH, 3 mL·min-1) 得到化合物7 (tR = 11.0 min, 7.0 mg)。
正丁醇部位经Diaion HP-20柱分离, 依次用水、20%乙醇、40%乙醇、70%乙醇和95%乙醇梯度洗脱, 每个梯度洗脱4个柱体积, 得到5个组分, 即A1~A5。A2 (201.0 g) 经MCI gel柱色谱分离, 依次用水、20%甲醇、40%甲醇、60%甲醇和100%甲醇进行梯度洗脱, 得到5个组分, 即A2-1~A2-5。A2-3 (20.1 g) 经硅胶柱色谱分离, 以二氯甲烷-甲醇(50∶1→5∶1) 梯度洗脱, 得到5个组分, 即A2-3-1~A2-3-5。A2-3-1经硅胶柱色谱分离, 以二氯甲烷-甲醇(40∶1→5∶1) 梯度洗脱, 得到9个组分, 即A2-3-1-1~A2-3-1-9。A2-3-1-3 (200.6 mg) 经半制备型高效液相色谱分离(18% MeOH, 3 mL·min-1) 得到化合物1 (tR = 62.9 min, 2.1 mg)。A2-3-1-4 (370.2 mg) 经半制备型高效液相色谱分离(13% MeCN, 3 mL·min-1) 得到化合物2 (tR = 40.1 min, 2.8 mg) 和化合物13 (tR = 26.1 min, 7.9 mg)。A2-3-1-2 (130.5 mg) 经半制备型高效液相色谱分离(42% MeOH, 3 mL·min-1) 得到化合物10 (tR = 11.5 min, 4.8 mg)。A2-3-3 (5.4 g) 经硅胶柱色谱分离, 以二氯甲烷-甲醇(20∶1→1∶1) 梯度洗脱, 得到9个组分, 即A2-3-3-1~A2-3-3-9。A2-3-3-5 (320.3 mg) 经半制备型高效液相色谱分离(38% MeOH, 3 mL·min-1) 得到化合物11 (tR = 24.1 min, 2.6 mg)。A2-4 (15.5 g) 经硅胶柱色谱(200~300目) 分离, 以二氯甲烷-甲醇(30∶1→1∶1) 梯度洗脱, 得到6个组分, 即A2-4-1~A2-4-6。A2-4-1 (300.8 mg) 经半制备型高效液相色谱分离(30% MeOH, 3 mL·min-1) 得到化合物5 (tR = 28.9 min, 1.6 mg)。A2-4-4 (40.1 g) 经Toyopearl HW-40C柱色谱分离, 依次用40%甲醇、60%甲醇和80%甲醇进行梯度洗脱, 得到9个组分, 即A2-4-4-1~A2-4-4-9。A2-4-4-6 (500.0 mg) 经半制备型高效液相色谱分离(26% MeOH, 3 mL·min-1) 得到化合物8 (tR = 34.4 min, 4.9 mg)。A2-1 (45.1 g) 经MCI gel柱, 依次用水、10%甲醇、20%甲醇、30%甲醇、40%甲醇、50%甲醇、60%甲醇、70%甲醇、80%甲醇和100%甲醇进行梯度洗脱, 得到5个组分, 即A2-1-1~A2-1-5。A2-1-3 (15.7 g) 经硅胶柱色谱分离, 以二氯甲烷-甲醇(12∶1→8∶1) 梯度洗脱, 得到5个组分A2-1-3-1~A2-1-3-5。A2-1-3-3 (60.8 mg) 经半制备型高效液相色谱分离(12% MeCN, 3 mL·min-1) 得到化合物4 (tR = 30.2 min, 6.2 mg)。A2-1-4 (8.6 g) 经硅胶柱色谱分离, 以二氯甲烷-甲醇(40∶1→2∶1) 梯度洗脱, 得到7个组分, 即A2-1-4-1~A2-1-4-7。A2-1-4-5 (95.8 mg) 经半制备型高效液相色谱分离(38% MeOH, 3 mL·min-1) 得到化合物12 (tR = 42.9 min, 5.7 mg)、14 (tR = 26.5 min, 7.9 mg) 和15 (tR = 21.2 min, 4.6 mg)。

化合物1~15在超净台内使用DMSO溶液配制成浓度为10 mmol·L-1母液, 使用0.25 μm的微孔滤膜进行过滤除菌, 之后再使用1640培养基将化合物1~15稀释为1 mmol·L-1。所有配制好的化合物均放于-20 ℃冰箱保存。
PC12细胞使用含有10% FBS的1640培养基(含青霉素、链霉素均为100 kU·L-1), 置于37 ℃、5% CO2的恒温培养箱中培养, 更换培养液两天一次。待细胞生长至对数生长期, 弃去原培养基, 用1 mL胰酶消化细胞, 加含血清培养液终止反应并传代。
将对数期生长的PC12细胞以细胞密度为4×104·mL-1接种于96孔板中, 每孔100 μL, 每组设置6个复孔, 置于37 ℃、5% CO2的恒温培养箱中培养。培养24 h后, 分为空白组(control)、模型组(model, Aβ25-35, 20 μmol·L-1) 和给药组(1~15, 10 μmol·L-1), 继续培养24 h后, 每孔加入20 μL MTT溶液(5 mg·mL-1) 孵育4 h。弃去MTT后每孔加入150 μL DMSO, 振荡10 min使其完全溶解, 并使用酶标仪在490 nm波长下测定各孔吸光度值(A), 重复3次, 计算细胞活力。
化合物1 (1.0 mg) 加入2.5 mL 2 mol·L-1 HCl-H2O中, 80 ℃下加热3 h, 反应混合物中加入EtOAc萃取3次, 每次3 mL。低温减压条件下浓缩水层, 浓缩过程中反复加H2O稀释, 最终得到中性残留物。残留物溶于色谱MeOH (1.5 mL), 进行高效液相色谱分析。色谱条件如下: 色谱柱(CHIRALPAK AD-H, 250 mm × 4.6 mm, 5 μm), 流动相(正己烷∶无水乙醇= 82∶18; 0.5 mL·min-1), 蒸发光散射检测器条件(漂移管温度: 75 ℃; 气体压力: 20.0 psi; 柱温: 30 ℃)。标准糖的保留时间为: D-葡萄糖(21.413, 22.554 min) 和L-葡萄糖(22.099, 23.648 min)。通过比较标准糖和化合物1中单糖的保留时间确定单糖的种类。
