Thirteen isoflavones were separated and purified from an ethanol extract of the rhizome of Dalbergia benthamii Prain by using silica gel, Sephadex LH-20, recrystallization et al. Their structures were identified by physicochemical properties and spectral analysis such as MS, 1D/2D-NMR as dalbergibenthamin (1), butesuperin A (2), xanthocercin A (3), butesuperin B (4), di-O-methylalpinum isoflavone (5), 2′-deoxgisoaunculutin (6), robustone (7), 4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8), formononetin (9), 6″-O-rhamnosyldaidzin (10), 3′, 4′-di-O-methylene-5-hydroxy-7-methoxy-6-isopentenyl isoflavone (11), derrubone dimethyl enter (12), and derrubone (13). Compound 1 is a pair of new isoflavonoid enantiomers, compound 12 is a new natural product and compounds 1-7 and 10-13 were obtained from D. benthamii Prain for the first time. In vitro cytotoxic activities of the compounds were explored by MTS testing with HL-60, A-549, SMMC-7721, MCF-7 and SW480 cell lines. Results show that compound 8 significantly inhibited cellular proliferation. The IC50 of compound 8 in A-549 and SW480 cells was 16.68 ± 0.19 and 15.21 ± 0.60 μmol·L-1.

运用硅胶、Sephadex LH-20柱色谱及重结晶等方法对两粤黄檀Dalbergia benthamii Prain根茎的乙醇提取物进行分离纯化, 得到13个异黄酮类化合物, 通过理化性质和MS、1D/2D NMR等波谱数据确定化合物的结构, 分别鉴定为两粤黄檀素(1)、butesuperin A (2)、xanthocercin A (3)、butesuperin B (4)、di-O-methylalpinum isoflavone (5)、2′-deoxgisoaunculutin (6)、robustone (7)、4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8)、芒柄花素(9)、6″-O-rhamnosyldaidzin (10)、3′, 4′-di-O-methylene-5-hydroxy-7-methoxy-6-isopentenyl isoflavone (11)、derrubone dimethyl enter (12) 和derrubone (13)。其中化合物1为一对新的异黄酮对映异构体, 化合物12为新的天然产物, 化合物1~710~13为首次从该植物中分离得到。采用MTS测试了化合物2~13对5种细胞株(人急性白血病细胞株HL-60、人非小细胞肺癌细胞株A-549、人肝癌细胞株SMMC-7721、人乳腺癌细胞株MCF-7、人结肠癌细胞株SW480) 的体外细胞毒活性, 结果表明, 化合物8对人非小细胞肺癌细胞株A-549和人结肠癌细胞株SW480显示出较强的生长抑制活性, IC50值分别为16.68 ± 0.19和15.21 ± 0.60 μmol·L-1

运用硅胶、Sephadex LH-20柱色谱及重结晶等方法对两粤黄檀Dalbergia benthamii Prain根茎的乙醇提取物进行分离纯化, 得到13个异黄酮类化合物, 通过理化性质和MS、1D/2D NMR等波谱数据确定化合物的结构, 分别鉴定为两粤黄檀素(1)、butesuperin A (2)、xanthocercin A (3)、butesuperin B (4)、di-O-methylalpinum isoflavone (5)、2′-deoxgisoaunculutin (6)、robustone (7)、4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8)、芒柄花素(9)、6″-O-rhamnosyldaidzin (10)、3′, 4′-di-O-methylene-5-hydroxy-7-methoxy-6-isopentenyl isoflavone (11)、derrubone dimethyl enter (12) 和derrubone (13)。其中化合物1为一对新的异黄酮对映异构体, 化合物12为新的天然产物, 化合物1~710~13为首次从该植物中分离得到。采用MTS测试了化合物2~13对5种细胞株(人急性白血病细胞株HL-60、人非小细胞肺癌细胞株A-549、人肝癌细胞株SMMC-7721、人乳腺癌细胞株MCF-7、人结肠癌细胞株SW480) 的体外细胞毒活性, 结果表明, 化合物8对人非小细胞肺癌细胞株A-549和人结肠癌细胞株SW480显示出较强的生长抑制活性, IC50值分别为16.68 ± 0.19和15.21 ± 0.60 μmol·L-1

Thirteen isoflavones were separated and purified from an ethanol extract of the rhizome of Dalbergia benthamii Prain by using silica gel, Sephadex LH-20, recrystallization et al. Their structures were identified by physicochemical properties and spectral analysis such as MS, 1D/2D-NMR as dalbergibenthamin (1), butesuperin A (2), xanthocercin A (3), butesuperin B (4), di-O-methylalpinum isoflavone (5), 2′-deoxgisoaunculutin (6), robustone (7), 4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8), formononetin (9), 6″-O-rhamnosyldaidzin (10), 3′, 4′-di-O-methylene-5-hydroxy-7-methoxy-6-isopentenyl isoflavone (11), derrubone dimethyl enter (12), and derrubone (13). Compound 1 is a pair of new isoflavonoid enantiomers, compound 12 is a new natural product and compounds 1-7 and 10-13 were obtained from D. benthamii Prain for the first time. In vitro cytotoxic activities of the compounds were explored by MTS testing with HL-60, A-549, SMMC-7721, MCF-7 and SW480 cell lines. Results show that compound 8 significantly inhibited cellular proliferation. The IC50 of compound 8 in A-549 and SW480 cells was 16.68 ± 0.19 and 15.21 ± 0.60 μmol·L-1.

两粤黄檀(Dalbergia benthamii Prain) 为豆科黄檀属植物, 树干或根部心材为其药用部位, 主要分布在广西、广东、海南等地, 它作为壮医临床常用药, 资源丰富, 可通经活血, 主治跌打损伤、筋骨疼痛等症状[1-3]。目前国内外对两粤黄檀的研究较少。前期, 本课题组从两粤黄檀中发现异黄酮、香豆素和甾醇等成分[4], 为了进一步阐明两粤黄檀的物质基础, 寻找具有生物活性的化合物, 本实验对两粤黄檀的异黄酮类化学成分进行研究, 采取多种柱色谱分离手段, 从其乙醇提取物中分离到了13个异黄酮类化合物, 分别鉴定为两粤黄檀素(1)、butesuperin A (2)、xanthocercin A (3)、butesuperin B (4)、di-O-methylalpinum isoflavone (5)、2′-deoxgisoaunculutin (6)、robustone (7)、4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8)、刺芒柄花素(9)、6″-O-rhamnosyldaidzin (10)、3′, 4′-di-O-methylene-5-hydroxy-7-methoxy-6-isopentenyl isoflavone (11)、derrubone dimethyl enter (12) 和derrubone (13), 结构见图 1。其中化合物1为新化合物且为一对光学对映异构体, 分别命名为(1″R, 2″R)-两粤黄檀素和(1″S, 2″S)-两粤黄檀素, 化合物12为新的天然产物, 化合物1~710~13为首次从该植物中分离得到。MTS法显示化合物81213具有体外肿瘤细胞毒活性。
化合物1  无色粉末, 能溶于氯仿、乙酸乙酯, 难溶于甲醇。HR-ESI-MS给出准分子离子峰m/z: 435.104 8 [M+Na]+ (C22H20O8Na, 计算值为435.105 6), 推测其分子式为C22H20O8, 计算其不饱和度为13。1H NMR谱显示芳香区有5个质子, 其中δH 8.02 (1H, s) 是异黄酮2-位质子的特征信号, 利用HSQC谱中确定其对应的碳信号为δC 151.8, 结合13C NMR中的δC 174.3 (异黄酮的C-4) 推断该化合物为异黄酮类化合物; δH 7.44 (1H, d, J = 1.7 Hz)、δH 7.17 (1H, dd, J = 8.0, 1.7 Hz) 和δH 6.97 (1H, d, J = 8.0 Hz) 的信号显示芳香区存在一个ABX耦合系统, 分别为黄酮B环的2′-、6′-和5′-H, 通过HSQC谱确定它们对应的碳位置: δC 110.4、δC 108.5、δC 122.8; δH 5.97、5.98 (2H, d, J = 1.3 Hz) 为-OCH2O-的氢信号, δH 6.24 (1H, dd, J = 6.5, 3.5 Hz)、δH 5.12 (1H, d, J = 3.5 Hz) 为2个连氧基团质子信号; δH 3.74 (3H, s) 为1个甲氧基质子信号, δH 1.67 (3H, s)、δH 1.64 (3H, s) 分别为甲基质子信号。13C NMR谱显示该化合物有22个碳, 其中δC 174.3为异黄酮4-位羰基碳信号。HMBC谱中显示δH 5.970 (1H, d, J = 1.3 Hz)、δH 5.974 (1H, d, J = 1.3 Hz) 与δC 147.9、δC 148.0有相关, 提示该CH2通过两个氧原子与苯环的δC 147.9、δC 148.0相连, H-2/C-9相关, H-8/C-6、C-10相关, H-1″/C-5、C-3″相关, H-2″/C-6、C-5、C-4″、C-5″相关, H-7′/C-3′、C-4′远程相关, H-4″/C-2″相关, H-5″/C-2″相关, H-3″-OH/C-4″、C-5″相关, H-1″-OH/C-1″、C-6相关。1H-1H COSY谱中显示H-5′/H-6′相关, 进一步证实了苯环ABX耦合系统的存在; H-1″/H-2″相关提示了C-1″-C-2″片段的存在, 由NOESY谱中H-1″/H-4″相关可知, 手性C-1″和C-2″的相对构型为1″S*和2″R* (结构及相关性见图 2)。结合Scifinder查询结果, 确定该化合物为新化合物, 借助于1D和2D NMR数据(图 2), 对化合物11H和13C NMR信号进行了全归属(表 1)。
由于该化合物比旋光度很小, 为[α]$ {}_{\mathrm{D}}^{20} $ +2.2 (c 0.5, CH3OH), 推测该化合物可能为外消旋体。通过手性柱在制备液相上分析, 发现有2个峰面积大约为1∶1的峰, 再通过半制备手性柱分离得到2个光学纯化合物1a1b, 实验测得比旋光度分别为23.16和-5.43, 为了确定它们的绝对构型, 分别建立RRSS两种构型的模型, 在MMFF94力场下进行构象搜索, 分别获得2种优势的构象, 再利用Gaussian16软件在B3LYP(GD3BJ)/6-31G(d) 水环境下进行结构优化和频率计算, 优化后的结构在B3LYP/aug-cc-pVTZ下进行比旋光度的计算, 发现RRSS两种构型比旋光度的正负关系及大小分别为+114.2和-114.0。而实验测得两种构型比旋光度为23.16和-5.43, 其中23.16对应的构型为RR, -5.43对应的构型为SS。因此, 化合物1a1b结构分别鉴定为(1″R, 2″R)-两粤黄檀素和(1″S, 2″S)-两粤黄檀素。
化合物12  黄色针状结晶(氯仿); EI-MS m/z: 394 [M]+; 分子式C23H22O61H NMR谱中, δH 7.79 (1H, s) 是异黄酮2-位质子的特征信号, 结合13C NMR中的δC 174.9 (异黄酮的C-4) 推断该化合物为异黄酮类化合物; δH 7.08 (1H, d, J = 1.7 Hz)、δH 6.94 (1H, dd, J = 8.0, 1.7 Hz)、δH 6.85 (1H, d, J = 8.0 Hz) 的信号显示芳香区存在一个ABX耦合系统, 分别为黄酮B环的2′-、6′-和5′-H; δH 6.64 (1H, s) 为苯环五取代的芳氢信号; δH 5.14~5.18 (1H, m)、δH 3.41 (2H, d, J = 6.7 Hz)、δH 1.66 (3H, s)、δH 1.79 (3H, s) 提示有1个异戊烯基存在; δH 5.97 (2H, s) 为-OCH2O-的氢信号; δH 3.91 (3H, s)、δH 3.85 (3H, s) 为2个甲氧基质子信号。13C NMR谱中, 共显示23碳信号, 结合DEPT谱发现, 其中δC 174.9为酮羰基碳信号、δC 101.1为-OCH2O-碳信号; 1个三取代苯环上的碳信号[δC 147.6 (C-3′)、147.5 (C-4′)、125.9 (C-1′)、122.4 (C-6′)、110.1 (C-2′)、108.3 (C-5′)], 1个五取代苯环上的碳信号[δC 162.2 (C-7)、158.1 (C-5)、157.8 (C-9)、113.0 (C-10)、95.1 (C-8)、122.6 (C-6)], 2个甲氧基碳信号[δC 62.3 (5-OCH3)、56.0 (7-OCH3)], 2个甲基碳信号[δC 25.8 (C-4″)、17.9 (C-5″)]。在HMBC谱中, H-2/C-9、C-4、C-3相关, H-2′、H-6′/C-3相关, 确定为异黄酮类化合物; H-7′/C-4′、C-3′相关, 提示该CH2通过两个氧原子与苯环的δC 147.6、δC 147.5相连; δH 3.91和3.85为2个甲氧基氢分别与C-5和C-7相关, 表明2个甲氧基分别连在C-5和C-7上, H-1″、H-4″、H-5″分别与C-2″、C-3″有相关, 提示存在一个异戊烯基。
综合以上信息, 确定化合物12结构为3′, 4′-di-O-methylene-5, 7-dimethoxy-6-isopentenyl isoflavone。经检索, 化合物12与文献[5]报道合成的产物derrubone dimethyl enter具有相同结构, 而相关文献未见其波谱数据报道, 因此确定化合物12为新天然产物, 借助于1D和2D数据(图 3), 对化合物121H和13C NMR信号进行了全归属(表 1)。
细胞毒活性测试以顺铂(DDP) 和紫杉醇(taxol) 为阳性对照, 初步检测了化合物2~13对人急性白血病细胞株(HL-60)、人非小细胞肺癌细胞株(A-549)、人肝癌细胞株(SMMC-7721)、人乳腺癌细胞株(MCF-7)、人结肠癌细胞株(SW480) 增殖的抑制作用。测试结果表明, 化合物81213对5种肿瘤细胞均具有不同程度的生长抑制活性, 其中化合物8对A-549、SW480细胞毒活性较强, 其IC50分别为16.68 ± 0.19 μmol·L-1和15.21 ± 0.60 μmol·L-1。结果见表 2

两粤黄檀于2017年7月采于广西靖西, 由广西中医药大学韦松基教授鉴定为豆科黄檀属植物两粤黄檀Dalbergia benthamii Prain的根茎。样品标本(No. DBP201707) 存放于广西中医药大学中药化学实验室。
两粤黄檀药材粗粉20 kg, 先后采用10倍量95%和60%乙醇渗漉提取, 回收溶剂, 总提取物用温水悬浮, 依次用石油醚(60~90 ℃)、氯仿、乙酸乙酯、正丁醇萃取, 回收溶剂, 得到石油醚部位浸膏66.6 g, 氯仿部位浸膏510.4 g, 乙酸乙酯部位浸膏288.9 g, 正丁醇部位浸膏580.8 g和水部位浸膏612.7 g, 各浸膏于4 ℃冰箱保存备用。
氯仿部位浸膏260 g, 经硅胶柱色谱分离, 氯仿-丙酮(100∶0、50∶1、20∶1、10∶1、5∶1、0∶100) 梯度洗脱, 得到9个组分(Fr.L1~9)。Fr.L1经硅胶柱色谱, 以石油醚∶乙酸乙酯(100∶1~1∶1) 梯度洗脱, 得3个组分; 组分1 (黄色粉末) 经Sephadex LH-20柱色谱(氯仿∶甲醇= 1∶1), 得化合物6 (4.5 mg); 组分2 (黄色粉末) 经硅胶柱色谱, 以石油醚-乙酸乙酯(40∶1~8∶1) 梯度洗脱, 得化合物7 (6.4 mg)。Fr.L3经硅胶柱色谱, 以石油醚-乙酸乙酯(20∶1~1∶1) 梯度洗脱, 得到5个组分。组分1 (1~56) 经硅胶柱色谱, 以石油醚-乙酸乙酯(100∶1~10∶1) 梯度洗脱, 流分(78~84) 经Sephadex LH-20柱色谱(氯仿-甲醇= 1∶1), 得化合物12 (3.1 mg); 流分(125~129) 经Sephadex LH-20柱色谱(氯仿-甲醇= 1∶1), 得化合物13 (3.0 mg); 流分(116~118) 用氯仿-丙酮反复重结晶, 得到化合物5 (3.2 mg)。Fr.L4硅胶柱色谱, 经石油醚-丙酮(40∶1~2∶1) 梯度洗脱, 流分(176~191) 经硅胶柱色谱, 石油醚-丙酮(1∶1) 洗脱, 得到白色粉末, 再经Sephadex LH-20 (氯仿-甲醇= 1∶1), 得化合物1 (3.3 mg), 经CHIRALCEL IC00CE-AQ059手性柱(3 μm, 4.6 mm × 250 mm) 拆分得到化合物1a (正己烷-异丙醇40∶60, tR = 13.6 min, 1.5 mg) 和化合物1b (正己烷-异丙醇40∶60, tR = 37.2 min, 1.3 mg)。Fr.L5经硅胶柱色谱, 以石油醚-丙酮(30∶1~1∶1) 梯度洗脱。其中流分31~53经硅胶柱色谱, 以石油醚-丙酮(30∶1~2∶1) 梯度洗脱, Sephadex LH-20 (氯仿-甲醇= 1∶1) 纯化, 得化合物11 (3.4 mg); 流分80~85中得到白色粉末, 经氯仿-丙酮反复重结晶和Sephadex LH-20 (氯仿-甲醇= 1∶1), 得化合物8 (3.2 mg) 和化合物9 (1.5 mg); 流分152中得到白色粉末, 经氯仿-甲醇反复重结晶和Sephadex LH-20 (氯仿-甲醇= 1∶1), 得化合物2 (3.3 mg); 流分160~166中得到白色粉末, 经丙酮反复重结晶, 得化合物4 (3.5 mg)。Fr.L6经硅胶柱色谱, 氯仿-甲醇(50∶1~3∶1) 梯度洗脱。在流分10~14中得到白色粉末, 经氯仿-丙酮反复重结晶, Sephadex LH-20 (氯仿-甲醇= 1∶1) 纯化, 得化合物3 (3.3 mg)。
正丁醇部位浸膏300 g, 大孔吸附树脂柱色谱分离, 乙醇-水系统(水、30%乙醇、50%乙醇、70%乙醇、90%乙醇) 进行洗脱, 得到5个组分(Fr.Z1~5)。Fr.Z2用MCI反相色谱水-甲醇(20%甲醇、40%甲醇、60%甲醇、甲醇) 梯度洗脱, 得到4个组分。Fr.Z2-2经制备液相色谱仪进行分离得到化合物10 (1.3 mg)。

化合物9  白色颗粒(甲醇); EI-MS m/z 268 [M]+; 分子式C16H12O41H NMR (500 MHz, CD3OD) δH 8.04 (1H, d, J = 8.8 Hz, H-5), 7.87 (1H, s, H-2), 7.40 (2H, d, J = 8.8 Hz, H-2′, 6′), 6.91 (2H, d, J = 8.8 Hz, H-3′, 5′), 6.86 (1H, dd, J = 8.8, 2.3 Hz, H-6), 6.78 (1H, d, J = 2.3 Hz, H-8), 3.78 (3H, s, 4′-OCH3)。13C NMR (125 MHz, CD3OD) δC 176.8 (C-4), 162.7 (C-7), 159.5 (C-4′), 158.3 (C-9), 152.6 (C-2), 130.2 (C-2′, 6′), 127.7 (C-5), 124.6 (C-1′), 124.3 (C-3), 117.3 (C-10), 115.3 (C-6), 113.9 (C-3′, 5′), 102.4 (C-8), 55.3 (4′-OCH3)。以上波谱数据与文献[15, 16]报道的数据基本一致, 鉴定化合物9为芒柄花素。

采用MTS法[21]进行肿瘤细胞增殖抑制活性测定, 分别将人急性白血病细胞株HL-60、人非小细胞肺癌细胞株A-549、人肝癌细胞株SMMC-7721、人乳腺癌细胞株MCF-7、人结肠癌细胞株SW480培养于含10%胎牛血清的细胞培养液中, 培养细胞至对数生长期, 将对数期生长的肿瘤细胞调整浓度为每毫升3×104~1.5×105个的细胞悬液, 接种到96孔板, 每孔接种100 μL, 于37 ℃、5% CO2培养箱中孵育24 h后加入样品(终浓度分别为40、8、1.6、0.32和0.064 μmol·L-1), 继续培养48 h后, 每孔换成培养液100 μL, 然后加入20 μL MTS溶液, 继续在37 ℃、5% CO2培养箱中孵育4 h后, 用酶标仪在492 nm波长处检测吸光度(OD值), 应用两点法(Reed and Muench法) 计算其半数抑制浓度IC50
