One undescribed diterpenoid acid and six compounds were isolated from the 95% ethanol fraction of Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui resin by using various chromatographic methods, including MCI Gel, Sephadex LH-20, ODS, silica gel and semi-preparative HPLC. The planar structures were identified by spectroscopy methods (1D, 2D NMR, UV, IR, MS, etc.), and the absolute configuration of the new compound was determined by ECD calculation. Compound 1 is a new compound, and compounds 2, 5-7 were isolated from Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui for the first time.

运用MCI Gel、Sephadex LH-20、ODS和硅胶等柱色谱, 结合半制备液相、TLC等分离方法, 从思茅松松脂的95%乙醇部位分离纯化得到7个化合物。运用现代波谱学方法(1D、2D NMR、UV、IR、MS等) 对所分离得到的化合物进行结构鉴定, 并运用计算ECD的方法确定了新化合物的绝对构型。其中, 化合物1为新化合物, 化合物25~7为首次从思茅松中分离得到。

One undescribed diterpenoid acid and six compounds were isolated from the 95% ethanol fraction of Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui resin by using various chromatographic methods, including MCI Gel, Sephadex LH-20, ODS, silica gel and semi-preparative HPLC. The planar structures were identified by spectroscopy methods (1D, 2D NMR, UV, IR, MS, etc.), and the absolute configuration of the new compound was determined by ECD calculation. Compound 1 is a new compound, and compounds 2, 5-7 were isolated from Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui for the first time.

思茅松(Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui) 为松科(Pinaceae) 松属植物[1], 主要生长在云南省, 生态适应性强, 产脂量高[2]。松脂为松属植物中渗出的树脂, 味苦性温, 归脾经, 主治疥疮、皮癣[3]。松脂除了药用外[4], 也可用于油墨、建材、造纸、涂料[5]、合成杀虫剂等[6]。因松脂应用广泛, 故研究人员十分关注其化学成分以及药理活性。现有研究表明, 松脂中的化学成分主要由单萜、倍半萜、二萜酸类化合物组成[7]。松脂中的松香酸及其衍生物在抗菌、抗炎、抗病毒、抗肿瘤和降血糖等方面疗效显著[8]。为了进一步从松脂中寻找结构新颖的化学成分, 丰富思茅松松脂的化学组成, 本研究从其95%乙醇部位中分离纯化得到了4个二萜酸、1个黄酮和2个酚酸类化合物(图 1), 其中, 化合物1为新化合物, 化合物25~7为首次从思茅松中分离得到。
化合物1为黄色油状物, [α]$ {}_{\mathrm{D}}^{20} $ -5.62 (c 0.11, MeOH), HR-ESI-MS给出准分子离子峰m/z 349.200 6 [M+H]+, 计算值为349.201 0, 推测分子式为C20H28O5, 不饱和度为7。紫外光谱显示在202、238 nm处有最大吸收峰。红外光谱显示存在羟基(3 445 cm-1)、羰基(1 697 cm-1) 和碳碳双键(1 651 cm-1) 等特征吸收峰。化合物11H NMR (CD3OD, 500 MHz) 谱(表 1) 显示1个双键特征氢信号δH 6.51 (1H, s, H-14)、两个连氧次甲基氢信号峰[δH 4.26 (1H, dd, J = 10.5, 5.4 Hz, H-7), δH 5.37 (1H, d, J = 6.7 Hz, H-11)]、1个次甲基氢信号峰δH 3.11 (1H, d, J = 6.7 Hz, H-12) 和4个甲基的信号峰[δH 1.62 (3H, s, H-17), 1.13 (3H, s, H-16), 1.14 (3H, s, H-19), 0.69 (3H, s, H-20)]。分析其13C NMR (CD3OD, 125 MHz) 谱并结合HSQC谱发现, 化合物1中存在20个碳, 其中δC 198.6 (C-13) 为羰基碳信号峰, δC 181.6 (C-18) 为羧基碳信号峰, 两个双键碳信号分别为[δC 168.5 (C-8), 122.2 (C-14)], 由C-8不寻常的高化学位移和C-14较小的化学位移推测双键可能和羰基共轭, 一个连氧季碳信号和两个连氧次甲基碳信号分别为[δC 86.9 (C-15), 72.7 (C-7), 72.2 (C-11)]。此外, 在中高场区(60~10之间) 还观察到13个碳信号, 分别为4个甲基碳信号[δC 25.9 (C-16), 29.7 (C-17), 16.8 (C-19), 16.4 (C-20)]、4个亚甲基碳信号[δC 38.2 (C-1), 19.2 (C-2), 38.1 (C-3), 37.2 (C-6)]、3个次甲基碳信号[δC 47.6 (C-5), 55.5 (C-9), 53.1 (C-12)] 和2个季碳信号[δC 48.1 (C-4), 41.0 (C-10)]。将以上数据与文献对比, 发现化合物1和pinyunins A[9]的平面结构具有相似的骨架, 同为含有α, β不饱和酮的二萜酸。二者的区别在于化合物1的C-7被羟基取代, 这一点可以由HMBC谱(图 2) 中δH 4.26 (H-7) 与δC 37.2 (C-6), 168.5 (C-8), 122.2 (C-14) 存在的相关信号, 以及碳谱中C-7 (δC 72.7) 的化学位移推定; 此外, 化合物1中C环与D环的骈合位置以及α, β不饱和酮的位置也发生了改变, 这一点可以由δH 5.37 (H-11) 与δC 168.5 (C-8), 41.0 (C-10), 53.1 (C-12), 198.6 (C-13)、δH 3.11 (H-12) 与δC 72.2 (C-11), 122.2 (C-14), 86.9 (C-15), 25.9 (C-16), 29.7 (C-17) 存在的HMBC相关信号证明, 另外, 1H-1H COSY谱中H-1/H-2/H-3, H-5/H-6/H-7, H-9/H-11/H-12存在的一系列相关信号进一步证实了以上推断。根据以上信息确定了化合物1的平面结构。
化合物1的相对构型是通过分析耦合常数和NOESY谱(图 3) 确定的。根据H-5和Hb-6与文献相似的大耦合常数(3JH5, H6 = 12.9 Hz) 判断出H-5为直立键[10], H-7和Hb-6的大耦合常数(3JH6, H7 = 10.5 Hz) 判断出H-7为直立键[11], H-11和H-12与文献一致的较大耦合常数(3JH11, H12 = 6.7 Hz) 判断出H-11和H-12位于同侧[9]。进一步分析NOESY谱发现, 在位于六元环轴向位置的CH3-20与CH3-19存在相关, CH3-20与H-11相关, H-5与H-7/H-9存在相关信号, 说明H-5/H-7/H-9位于环平面同侧, 而H3-20/H3-19/H-11/H-12位于另一侧, 同时由于H-5/H-7/H-9与H3-20/H3-19之间均没有NOESY相关信号, 可以确定H-5/H-7/H-9为α取向, H3-20/H3-19/H-11/H-12为β取向, 所以确定相对构型为(4R*, 5R*, 7S*, 9R*, 10S*, 11S*, 12R*)。化合物1的绝对构型是通过电子圆二色性(TDDFT ECD) 计算与实验ECD对比来确定的, 在TDDFT/B3LYP/6-31G (d, p) 水平下使用SMD模式并添加色散校正计算。结果显示实验ECD曲线与构型(4R, 5R, 7S, 9R, 10S, 11S, 12R) 的曲线吻合较好(图 4), 所以确定了化合物1的绝对构型并将其命名为Rosin J。

MCI反相柱色谱材料(日本三菱公司); Sephadex LH-20柱色谱材料(Parmacia Biotech公司); ODS反相柱色谱材料; 正相硅胶色谱填料(青岛海洋化工厂); YMC-Pack ODS-A半制备柱(250 mm × 20 mm, 10 μm); Agela ODS半制备柱(250 mm × 10 mm, 5 μm), 色谱纯及分析纯试剂(天津四友精细化学品有限公司)。
松脂采自中国云南省思茅市景谷县, 经云南中医药大学邱斌教授鉴定为松科植物思茅松(Pinus kesiya var. langbianensis (A.Chev.) Gaussen ex Bui) 松脂, 标本号: CHYX0708, 保存于河南中医药大学中药药效物质基础研究室。
将25 kg松脂粉碎后, 先用3倍体积石油醚溶解3 h (3次), 然后将上清液浓缩得到石油醚部位。底部未溶解的大极性样品用95%乙醇充分溶解, 除去底部少量泥沙后浓缩成浸膏, 共6.83 kg, 得到95%乙醇部位。95%乙醇部位上MCI柱, 用乙醇-水系统(0:100~95:5) 梯度洗脱后, 依次得到Fr.A1 (34.8 g)、Fr.A2 (119.1 g)、Fr.B (344.5 g)、Fr.C (863.8 g) 和Fr.D (4 523 g)。Fr.A1 (34.8 g) 采用凝胶柱(4 cm × 43 cm), 以甲醇-水(0:100~100:0) 为洗脱剂进行梯度洗脱, 得到Fr.A1-1~Fr.A1-7。将Fr.A1-2与Fr.A1-3合并(17.3 g), 上ODS柱色谱, 用甲醇-水梯度洗脱(12:88~100:0), 依次得到Fr.A1-2-1~Fr.A1-2-10。Fr.A1-2-3 (1.5 g) 过硅胶柱色谱(200~300目; 二氯甲烷-甲醇100:0~0:100), 依次得到Fr.A1-2-3-1~Fr.A1-2-3-10。Fr.A1-2-3-4经半制备高效液相(甲醇: 水= 38:62) 纯化, 从中分离出化合物7 (tR = 18.3 min; 2.78 mg)。Fr.A1-2-5 (4.65 g) 过硅胶柱色谱(200~300目; 二氯甲烷-甲醇100:0~0:100), 依次得到Fr.A1-2-5-1~Fr.A1-2-5-8。Fr.A1-2-5-4经半制备液相(乙腈: 水= 26:74) 纯化, 从中分离出化合物1 (tR = 25.1 min; 4.70 mg)。Fr.A1-2-6 (1.92 g) 过硅胶柱色谱(200~300目; 二氯甲烷-甲醇100:0~0:100), 依次得到Fr.A1-2-6-1~Fr.A1-2-6-5, Fr.A1-2-6-2依次经凝胶、TLC、半制备高效液相(甲醇: 水= 77:23) 纯化, 从中分离出化合物2 (tR = 25.7 min; 25.40 mg)。Fr.A1-2-8 (498 mg) 过硅胶柱色谱(200~300目; 二氯甲烷-甲醇100:0~0:100), 依次得到Fr.A1-2-8-1~Fr.A1-2-8-8, Fr.A1-2-8-6经TLC、半制备高效液相(甲醇: 水= 85:15) 纯化, 从中分离出化合物4 (tR = 42.0 min; 5.26 mg)。Fr.A1-4 (1.15 g) 过凝胶柱, 依次得到Fr.A1-4-1~Fr.A1-4-5。Fr.A1-4-1 (0.95 g) 过硅胶柱色谱(200~300目; 二氯甲烷-甲醇100:0~0:100), 依次得到Fr.A1-4-1-1~Fr.A1-4-1-9。Fr.A-4-1-5经TLC、半制备高效液相(甲醇: 水= 83:17) 分离出化合物3 (tR = 22.6 min; 9.66 mg)。Fr.A1-4-2 (96.7 mg) 经半制备高效液相(甲醇: 水= 72:28) 纯化, 从中分离出化合物6 (tR = 12.3 min; 2.13 mg)。Fr.B (344.5 g) 依次过MCI、硅胶、ODS柱色谱, 得到Fr.B-5-6-9。Fr.B-5-6-9 (152.2 mg) 经TLC、半制备液相(甲醇: 水= 75:25) 纯化, 得到化合物5 (tR = 12.0 min; 7.24 mg)。
化合物1  黄色油状物; [α]$ {}_{\mathrm{D}}^{20} $ -5.62 (c 0.11, MeOH); UV (MeOH) λmax (log ε) 202 (3.51)、238 (3.58) nm; IR νmax 3 444、2 932、1 697、1 651、1 197、1 134 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1; 正离子HR-ESI-MS m/z 349.200 6 [M+H]+ (计算值为C20H29O5+, 349.201 0)。
化合物2  白色固体, ESI-MS m/z 333 [M+H]+, 分子式为C20H28O41H NMR (CDCl3, 500 MHz) δH 6.17 (1H, s, H-11), 5.24 (1H, d, J = 6.8 Hz, H-14), 2.99 (1H, d, J = 6.7 Hz, H-13), 2.51~2.37 (2H, m, H-7), 2.09~2.11 (2H, m, H-5, H-8), 2.00 (1H, m, H-1a), 1.80 (1H, m, H-3a), 1.61~1.70 (4H, m, H-2a, H-2b, H-3b, H-6a), 1.59 (3H, s, H-16), 1.56~1.58 (1H, m, H-6b), 1.35 (1H, m, H-1b), 1.15 (3H, s, H-17), 1.12 (3H, s, H-19), 0.65 (3H, s, H-20); 13C NMR (CDCl3, 125 MHz) δC 37.3 (C-1), 18.1 (C-2), 37.0 (C-3), 47.3 (C-4), 48.8 (C-5), 27.1 (C-6), 37.6 (C-7), 56.1 (C-8), 164.3 (C-9), 40.4 (C-10), 125.7 (C-11), 196.5 (C-12), 51.4 (C-13), 70.7 (C-14), 85.7 (C-15), 25.7 (C-16), 29.7 (C-17), 184.0 (C-18), 16.2 (C-19), 16.2 (C-20), 以上数据与文献[9]报道基本一致, 确定化合物为pinyunins A。
化合物3  白色无定形固体, ESI-MS m/z 317 [M+H]+, 分子式为C20H28O31H NMR (CD3OD, 500 MHz) δH 7.18 (1H, overlapped, H-11), 7.18 (1H, overlapped, H-12), 7.10 (1H, s, H-14), 2.87 (2H, m, H-7), 2.34 (1H, m, H-1a), 2.17 (1H, dd, J = 12.3, 1.5 Hz, H-5), 1.77~1.90 (3H, m, H-2a, H-3a, H-6a), 1.60~1.72 (2H, m, H-2b, H-3b), 1.53 (1H, m, H-6b), 1.48 (6H, s, H-16/H-17), 1.42 (1H, m, H-1b), 1.25 (3H, s, H-19), 1.19 (3H, s, H-20); 13C NMR (CD3OD, 125 MHz) δC 39.5 (C-1), 19.5 (C-2), 38.1 (C-3), 49.0 (C-4), 46.6 (C-5), 22.9 (C-6), 31.3 (C-7), 135.5 (C-8), 148.9 (C-9), 38.1 (C-10), 123.2 (C-11), 126.0 (C-12), 147.6 (C-13), 126.0 (C-14), 72.8 (C-15), 31.8 (C-16), 31.9 (C-17), 183.2 (C-18), 17.2 (C-19), 19.7 (C-20), 以上数据与文献[11]报道基本一致, 确定为化合物15-hydroxydehydroabietic acid。
化合物4  白色粉末, ESI-MS m/z 315 [M+H]+, 分子式为C20H26O31H NMR (CD3OD, 500 MHz) δH 7.82 (1H, d, H-14, J = 1.8 Hz), 7.50 (1H, dd, H-12, J = 8.1, 1.8 Hz), 7.41 (1H, d, H-11, J = 8.1 Hz), 2.94 (1H, m, H-15), 2.60~2.85 (2H, m, H-6), 2.45 (1H, d, J = 12.8 Hz, H-5), 2.38 (1H, m, H-1a), 1.75~1.95 (3H, m, H-2a, H-3a, H-3b), 1.58~1.62 (2H, m, H-1b, H-2b), 1.35 (3H, s, H-19), 1.30 (3H, s, H-20), 1.26 (3H, d, J = 6.9 Hz, H-16), 1.26 (3H, d, J = 6.9 Hz, H-17); 13C NMR (CD3OD, 125 MHz) δC 38.9 (C-1), 19.2 (C-2), 38.6 (C-3), 47.5 (C-4), 45.5 (C-5), 37.9 (C-6), 200.9 (C-7), 131.6 (C-8), 155.1 (C-9), 38.5 (C-10), 125.2 (C-11), 134.2 (C-12), 148.2 (C-13), 125.6 (C-14), 34.9 (C-15), 24.2 (C-16), 24.2 (C-17), 181.3 (C-18), 23.8 (C-19), 16.8 (C-20), 以上数据与文献[11]报道基本一致, 确定化合物为7-oxocallitrisic acid。
化合物5  白色粉末, ESI-MS m/z 279 [M+Na]+, 分子式为C15H12O41H NMR (CD3OD, 500 MHz) δH 7.50 (2H, d, J = 7.3 Hz, H-2′, H-6′), 7.42 (2H, t, J = 7.4 Hz, H-3′, H-5′), 7.37 (1H, m, H-4′), 5.94 (1H, d, J = 2.1 Hz, H-8), 5.91 (1H, d, J = 2.1 Hz, H-6), 5.47 (1H, dd, J = 12.7, 3.0 Hz, H-2), 3.10 (1H, dd, J = 17.1, 12.8 Hz, H-3), 2.78 (1H, dd, J = 17.1, 3.1 Hz, H-3); 13C NMR (CD3OD, 125 MHz) δC 80.5 (C-2), 44.2 (C-3), 197.3 (C-4), 164.7 (C-5), 97.2 (C-6), 168.4 (C-7), 96.2 (C-8), 165.5 (C-9), 103.4 (C-10), 140.4 (C-1′), 127.3 (C-2′, C-6′), 129.7 (C-3′, C-5′), 129.6 (C-4′)。通过对比旋光数据[α]$ {}_{\mathrm{D}}^{20} $ -38.33 (c 0.10, MeOH), 以及ECD计算(图 4) 确定了5的立体构型, 以上数据与文献[12]报道基本一致, 确定化合物为pinocembrin。
化合物6  黄色固体, ESI-MS m/z 169 [M+H]+, 分子式为C8H8O41H NMR (CD3OD, 500 MHz) δH 7.58 (1H, m, H-2), 7.57 (1H, m, H-6), 6.85 (1H, d, J = 8.7 Hz, H-5), 3.91 (3H, s, 3-OCH3)。13C NMR (CD3OD, 125 MHz) δC 123.8 (C-1), 114.5 (C-2), 147.2 (C-3), 151.2 (C-4), 112.3 (C-5), 123.9 (C-6), 168.8 (C-7), 54.9 (C-OCH3)。以上数据与文献[13]报道基本一致, 确定化合物为香草酸。
化合物7  黄色固体, ESI-MS m/z 153 [M+H]+, 分子式为C8H8O31H NMR (CD3OD, 500 MHz) δH 9.77 (1H, s, -CHO), 7.46 (H, m, H-6), 7.44 (H, m, H-2), 6.96 (1H, d, H-5, J = 8.0 Hz), 3.94 (3H, s, -OCH3)。13C NMR (CD3OD, 125 MHz) δC 131.1 (C-1), 111.1 (C-2), 148.8 (C-3), 154.7 (C-4), 115.7 (C-5), 127.9 (C-6), 56.3 (-OCH3)。以上数据与文献[14]报道基本一致, 确定化合物为香草醛。
化合物1和化合物5的绝对构型是通过(TDDFT ECD) 计算与实验ECD对比来确定的。将化合物1和化合物5的构象导入Gaussview 6.0.16软件中的MMFF94s程序, 使用10 kcal·mol-1的能量窗进行构象搜索。对搜索出来的优势构象使用Gaussian 16软件进行优化B3LYP/6-31G (d, p), 优化后的构象在B3LYP/6-31G (d, p) 水平下以甲醇为溶剂进行ECD计算, 然后在SpecDis 1701软件中对ECD计算结果进行拟合。
