Ten compounds were isolated from the 95% ethanol extract of the whole plant of Gerbera piloselloides by silica gel column chromatography, MCI column chromatography and semi-preparative HPLC methods. Their structures were identified on the basis of physicochemical properties, spectral data (UV, IR, MS and NMR), circular dichroism (CD) spectra and single crystal X-ray diffraction analysis as 3′R-gerpilosecoumarin A (1a), 3′S-gerpilosecoumarin A (1b), gymnastone (2), gerberinside (3), divaricataester C (4), luteolin (5), caffeic acid methyl ester (6), ethyl chlorogenate (7), 6-(β-D-glucopyranosyloxy)-7-methoxy-5-benzoranpropanoic acid methyl ester (8) and glucozaluzanin C (9). Among them, new compounds 1a and 1b were new compounds and optical enantiomers, which were obtained by chiral resolution, and their absolute configurations were determined by quantum chemical calculation ECD. Compounds 1 and 1a/1b significantly increased the survival of IEC-6 in rat small intestinal crypt epithelial cells after LPS injury.

采用硅胶柱色谱、MCI柱色谱、半制备高效液相色谱等技术从毛大丁草(Gerbera piloselloides) 全草95%乙醇提取物中分离纯化得到10个化合物。根据其理化性质、波谱数据(UV、IR、MS、NMR)、圆二色谱(CD) 和单晶X射线衍射等技术鉴定化合物结构, 分别为3′R-毛大丁香豆素A (1a)、3′S-毛大丁香豆素A (1b)、gymnastone (2)、大丁苷(3)、divaricataester C (4)、木犀草素(5)、咖啡酸甲酯(6)、绿原酸乙酯(7)、6-(β-D-glucopyranosyloxy)-7-methoxy-5-benzoranpropanoic acid methyl ester (8) 和glucozaluzanin C (9)。其中化合物1a1b为新化合物且为光学对映异构体, 通过手性拆分得到, 并经量子化学计算ECD确定了其绝对构型。化合物11a/1b能显著提高LPS损伤后的大鼠小肠隐窝上皮细胞IEC-6的存活率。

采用硅胶柱色谱、MCI柱色谱、半制备高效液相色谱等技术从毛大丁草(Gerbera piloselloides) 全草95%乙醇提取物中分离纯化得到10个化合物。根据其理化性质、波谱数据(UV、IR、MS、NMR)、圆二色谱(CD) 和单晶X射线衍射等技术鉴定化合物结构, 分别为3′R-毛大丁香豆素A (1a)、3′S-毛大丁香豆素A (1b)、gymnastone (2)、大丁苷(3)、divaricataester C (4)、木犀草素(5)、咖啡酸甲酯(6)、绿原酸乙酯(7)、6-(β-D-glucopyranosyloxy)-7-methoxy-5-benzoranpropanoic acid methyl ester (8) 和glucozaluzanin C (9)。其中化合物1a1b为新化合物且为光学对映异构体, 通过手性拆分得到, 并经量子化学计算ECD确定了其绝对构型。化合物11a/1b能显著提高LPS损伤后的大鼠小肠隐窝上皮细胞IEC-6的存活率。

Ten compounds were isolated from the 95% ethanol extract of the whole plant of Gerbera piloselloides by silica gel column chromatography, MCI column chromatography and semi-preparative HPLC methods. Their structures were identified on the basis of physicochemical properties, spectral data (UV, IR, MS and NMR), circular dichroism (CD) spectra and single crystal X-ray diffraction analysis as 3′R-gerpilosecoumarin A (1a), 3′S-gerpilosecoumarin A (1b), gymnastone (2), gerberinside (3), divaricataester C (4), luteolin (5), caffeic acid methyl ester (6), ethyl chlorogenate (7), 6-(β-D-glucopyranosyloxy)-7-methoxy-5-benzoranpropanoic acid methyl ester (8) and glucozaluzanin C (9). Among them, new compounds 1a and 1b were new compounds and optical enantiomers, which were obtained by chiral resolution, and their absolute configurations were determined by quantum chemical calculation ECD. Compounds 1 and 1a/1b significantly increased the survival of IEC-6 in rat small intestinal crypt epithelial cells after LPS injury.

毛大丁草(Gerbera piloselloides syn Piloselloides hirsute) 为菊科大丁草属(广义) 多年生草本植物, 又名白眉、白头翁等。该属在全球约80种植物, 主要分布在非洲, 次为亚洲东部, 国内约有20种, 主要分布于西藏、云南、四川、贵州、广西等地[1]。毛大丁草在国内常作为民族用药, 其全草入药, 具有宣肺止咳、发汗利水、行气活血作用, 常用于治疗伤风咳嗽、哮喘、水肿、小儿积食等[2, 3]。除其药用外, 毛大丁草常与三叉苦根茎混合使用用于发酵酿酒, 让酒具有特殊的香味, 所以毛大丁草还是一种药食同源性植物[4]。此外, 从毛大丁草中分离到的化学成分主要为香豆素类、糖苷类、黄酮类、萜类等, 但已有成分研究多侧重于GC-MS分析的挥发油类成分[5]。现代药理研究表明该植物具有止咳、抗菌、抗肿瘤、子宫镇痛、美白等多种活性[6-10], 但大多局限于其提取物的活性研究, 而对其化学成分活性研究较少。本课题组前期曾对毛大丁草的化学成分进行了研究, 分离并鉴定了12个非挥发性成分[11]。为进一步研究毛大丁草的化学成分, 发现结构新颖的活性成分, 本实验对毛大丁草95%乙醇提取物中的化学成分进行了研究, 从其乙酸乙酯萃取部位分离得到了10个化合物, 分别鉴定为: 3′R-毛大丁香豆素A (1a)、3′S-毛大丁香豆素A (1b)、gymnastone (2)、大丁苷(3)、divaricataester C (4)、木犀草素(5)、咖啡酸甲酯(6)、绿原酸乙酯(7)、6-(β-D-glucopyranosyloxy)-7-methoxy-5-benzoranpropanoic acid methyl ester (8)、glucozaluzanin C (9)。其中化合物1a/1b为新化合物, 结构如图 1所示, 本文着重介绍新化合物的分离纯化、手性拆分、结构鉴定和活性评价。
化合物1  无色针状晶体, mp 120~123 ℃。易溶于甲醇, 254 nm下显示为暗斑, 10%浓硫酸乙醇溶液加热显色为黄色(105 ℃)。UV (MeOH) λmax (logε)/nm: 214 (3.89), 260 (3.72), 315 (3.88); IR图谱中显示有羟基(3 370 cm-1)、羰基(1 714 cm-1)、双键(1 568和1 621 cm-1) 等基团特征吸收峰; HR-ESI-MS给出m/z: 279.086 4 [M-H]- (计算值279.086 9, C14H15O6), 提示分子式为C14H16O6, 不饱和度为7。在CD3OD中, 该化合物的1H NMR谱(表 1) 显示苯环四取代的芳香族质子信号7.03 (1H, d, J = 8.6 Hz, H-5)、6.96 (1H, d, J = 8.6 Hz, H-6); 一对顺式取代的2个双键质子信号δH 6.19 (1H, d, J = 9.4 Hz, H-3)、7.79 (1H, d, J = 9.4 Hz, H-4); 2个甲基氢信号: δH 1.24 (3H, s, H-5′)、1.20 (3H, s, H-6′)。13C NMR谱(表 1) 给出14个碳共振信号, 结合HSQC谱, 提示该化合物1含有6个季碳, 包括1个酯羰基碳δC 163.1 (C-2), 4个芳香季碳δC 151.4 (C-7)、144.1 (C-8)、115.5 (C-4a)、135.2 (C-8a), 1个连氧季碳72.6 (C-4′); 5个次甲基碳, 包括1个连氧次甲基碳δC 77.5 (C-3′)、2个烯烃次甲基碳δC 113.7 (C-3)、146.3 (C-4) 和2个芳香次甲基碳; 1个连氧亚甲基碳δC 71.9 (C-2′); 2个甲基碳δC 25.0 (C-5′)、27.0 (C-6′) (表 1)。通过以上数据, 可推测1为香豆素类化合物。在1H-1H COSY谱中(图 2), 存在3个自旋耦合系统(H-3/H-4, H-5/H-6, H-2′/H-3′), 在HMBC谱(图 2) 中, H-4与C-2, C-5, C-8a有相关, H-5与C-7, C-8a有相关, H-6与C-8, C-4a有相关, 推测出该结构的A片段如图 1所示。此外, H-5′, H-6′分别与C-4′, C-3′有相关, H-2′与C-4′, C-7有相关, 由此推测出该结构的B片段如图 1, 且结构片段B通过O原子与片段A的7位相连接。因此, 化合物1的平面结构如图 1所示。本研究获得了化合物1的晶体, 培养溶剂为二氯甲烷-甲醇(1:1, v/v), 通过单晶X射线衍射(图 3) 进一步证实了化合物1的平面结构, 但其Flack参数为N, 结合旋光值接近于0, 且ECD光谱中无明显的Cotton效应, 表明化合物1为外消旋体。随后, 经Chiralpak®IBN-5手性柱对其拆分, 得到一对对映异构体1a/1b, 比例约为1:1, 为了确定它们的绝对构型, 采取量子化学计算的方法[B3LYP-D3(BJ)/6-31G*] 计算了1a1b的ECD谱, 结果表明计算ECD谱与实验值吻合较好(图 4), 从而确定1a1b的绝对构型分别为3′R和3′S。因此, 化合物1a1b结构鉴定为3′R-毛大丁香豆素A (3′R-gerpilosecoumarin A) (1a) 和3′S-毛大丁香豆素A (3′S-gerpilosecoumarin A) (1b)。
所获晶体为斜方晶(0.12 mm × 0.09 mm × 0.01 mm), 培养溶剂为: 二氯甲烷-甲醇(1:1, v/v), 晶体数据为: C14H18O7, M = 298.28, T = 100.00 K, 空间群是P-1, 晶胞参数: a = 7.180 6 (3) Å, b = 9.557 0 (3) Å, c = 11.483 0 (3) Å, α = 112.128 1 (3)°, β = 93.259 (3)°, γ = 107.820 (3)°, V = 681.59 (4) Å3, Z = 2, ρ = 1.322 g·cm-3, μ (Cu Kα) = 0.997 mm-1; 最终一致因子[I > 2σ (I)]: R1 = 0.048 4, 和wR2 = 0.145 9; Flack参数为N。最后, 单晶数据提交保存至Cambridge Crystallo-graphic Data Centre (CCDC) 数据库, CCDC号为2143556。
采用CCK-8法对化合物11a/1b进行了肿瘤细胞毒活性测试, 结果显示, 化合物11a/1b对人乳腺癌细胞MCF-7和胃癌细胞SGC-7091均未表现出明显的抑制作用(IC50 > 100 μmol·L-1)。此外, 还采用CCK-8法考察了化合物11a/1b对大鼠小肠隐窝上皮细胞IEC-6增殖的影响, 结果表明, 化合物11a/1b在50 μmol·L-1剂量下对IEC-6未表现出毒性作用。在LPS诱导的IEC-6细胞损伤模型中, 当剂量为25 μmol·L-1时, 化合物11a/1b组的细胞存活率分别提高了1.08倍、1.09倍和1.07倍; 当剂量为50 μmol·L-1时, 化合物11a/1b组的细胞存活率分别提高了1.16倍、1.17倍和1.15倍(图 5)。结果显示, 化合物11a/1b能显著提高LPS损伤后的IEC-6细胞存活率, 并呈现出一定程度的剂量依赖关系, 表明化合物11a/1b能显著提高LPS损伤后IEC-6细胞的存活率。

毛大丁草于2019年11月采自广西省玉林市, 经广西中医药研究院中药资源研究所黄云峰研究员鉴定为菊科大丁草属植物毛大丁草Gerbera piloselloides (L.) Cass干燥全草。标本(SCI-NCL006379-1) 现存于中国医学科学院药用植物研究所国家中药化合物库。
毛大丁草全草30 kg, 粉碎后用10倍量的95%乙醇加热回流提取2次, 每次2 h, 过滤后合并提取液, 减压浓缩得总浸膏2.1 kg, 浸膏加水混悬至5 L, 分别用石油醚、乙酸乙酯、正丁醇、水进行萃取, 最后得到石油醚部位Fr. A (171 g)、乙酸乙酯部位Fr. B (1 067 g)、正丁醇部位Fr. C (287 g)、水部位Fr. D (343 g)。
取乙酸乙酯部位Fr.B (1 045 g) 进行硅胶柱色谱(60~100目) 分离, 以石油醚-乙酸乙酯系统(40:1~0:100) 梯度洗脱, 通过TLC检识合并得到6个组分, 即Fr.B1~Fr.B6。Fr.B5 (202 g) 经硅胶柱色谱(100~200目) 分离, 以二氯甲烷-甲醇系统(80:1~0:100) 洗脱, 通过TLC检识合并得到4个组分, 即Fr.B5.1~Fr.B5.4。Fr.B5.4 (56 g) 经硅胶柱色谱(200~300目) 分离, 以二氯甲烷-甲醇系统(25:1) 等度洗脱, 通过TLC合并得到2个组分, 即Fr.B5.4.1和Fr.B5.4.2。Fr.B5.4.1 (26 g) 经MCI柱色谱后再经半制备高效液相(甲醇-水50:50) 分离得到6个组分, 即Fr.B5.4.1.1~Fr.B5.4.1.6。Fr.B5.4.1.2经半制备高效液相分离得到7个组分, 即Fr.B5.4.1.2.1~Fr.B5.4.1.2.7。Fr.B5.4.1.2.3经半制备高效液相分离得到化合物2 (甲醇-水35:65, tR = 20.5 min, 10.5 mg)。Fr.B5.4.1.2.4经半制备高效液相分离得到化合物3 (甲醇-水35:65, tR = 30.3 min, 78 mg)。Fr.B5.4.1.2.6经半制备高效液相分离得到化合物1 (甲醇-水35:65, tR = 33.0 min, 19 mg), 1经Chiralpak®IBN-5手性柱(5 μm, 4.6 mm × 250 mm) 拆分得到化合物1a (正己烷-异丙醇80:20, tR = 14.2 min, 1.5 mg) 和化合物1b (正己烷-异丙醇80:20, tR = 17.2 min, 1.3 mg)。Fr.B5.4.1.4经半制备高效液相分离得到6个组分, 即Fr.B5.4.1.4.1~Fr.B5.4.1.4.6。Fr.B5.4.1.4.4经半制备高效液相分离得到化合物4 (甲醇-水23:77, tR = 77.0 min, 7.0 mg)。Fr.B5.4.1.5经半制备高效液相分离得到化合物5 (甲醇-水50:50, tR = 33.5 min, 4.8 mg)。Fr.B5.4.2 (30 g) 经MCI柱色谱分离, 以甲醇-水系统(70:1~0:100) 洗脱, 通过TLC合并得到4个组分, 即Fr.B5.4.2.1~Fr.B5.4.2.4。Fr.B5.4.2.2经硅胶柱色谱(200~300目) 分离, 以二氯甲烷-甲醇系统(30:1~1:1) 洗脱, 通过TLC检识合并得到4个组分, 即Fr.B5.4.2.2.1~Fr.B5.4.2.2.4。Fr.B5.4.2.2.1经半制备高效液相(乙腈-水25:75) 分离得到3个组分, 即Fr.B5.4.2.2.1.1~Fr.B5.4.2.2.1.3。Fr.B5.4.2.2.1.1经半制备高效液相分离得到化合物6 (乙腈-水25:75, tR = 45.4 min, 60 mg)。Fr.B5.4.2.2.3经半制备高效液相(乙腈-水21:79) 分离得到5个组分, 即Fr.B5.4.2.2.3.1~Fr.B5.4.2.2.3.5。Fr.B5.4.2.2.3.2经半制备高效液相分离得到化合物7 (乙腈-水24:76, tR = 32.2 min, 24 mg)。Fr.B5.4.2.2.3.4经半制备高效液相分离得到化合物8 (乙腈-水30:70, tR = 24.4 min, 2.5 mg) 和化合物9 (乙腈-水25:75, tR = 33.1 min, 2.0 mg)。

化合物9  无色针晶, mp 103~105 ℃, 易溶于甲醇, HR-ESI-MS m/z: 407.169 9 [M-H]- (计算值407.170 6, C21H27O8), 推测分子式为C21H28O81H NMR (600 MHz, CD3OD) δH 6.07 (1H, d, J = 3.6 Hz, H-13), 5.53 (1H, d, J = 3.6 Hz, H-13), 5.40 (1H, br s, H-15), 5.32 (1H, br s, H-15), 4.99 (1H, br s, H-14), 4.88 (1H, br s, H-14), 4.62 (1H, m, H-3), 4.43 (1H, d, J = 7.9 Hz, H-1′), 4.25 (1H, dd, J = 10.3, 9.0 Hz, H-6), 2.98 (1H, dd, J = 17.2, 9.0 Hz, H-5), 2.86 (1H, m, H-7), 2.75 (1H, m, H-1), 2.48 (1H, m, H-9), 2.31 (1H, m, H-2), 2.25 (1H, m, H-8), 2.24 (1H, m, H-9), 1.93 (1H, m, H-2), 1.42 (1H, m, H-8); 13C NMR (150 MHz, CD3OD) δC 46.4 (C-1), 38.6 (C-2), 81.3 (C-3), 150.8 (C-4), 51.8(C-5), 85.2 (C-6), 46.2 (C-7), 31.6 (C-8), 34.4 (C-9), 150.1 (C-10), 142.1 (C-11), 172.3 (C-12), 120.4 (C-13), 114.7 (C-14), 113.6 (C-15), 103.1 (C-1′), 75.3 (C-2′), 78.2 (C-3′), 71.8 (C-4′), 77.9 (C-5′), 62.8 (C-6′)。以上数据与文献[19]报道基本一致, 故鉴定为化合物9为glucozaluzanin C。
采用CCK-8法检测化合物1和化合物1a/1b对乳腺癌细胞MCF-7和胃癌细胞SGC-7091的细胞毒活性。
取对数生长期IEC-6细胞以每孔1×104接种于96孔板中, 培养24 h后吸出培养上清, 加入不同浓度(25、50 μmol·L-1) 的化合物11a/1b, 对照组加入等量含0.1%的DMSO的完全培养基; 在CO2孵育箱中继续培育24 h后, 每孔加CCK-8 10 μL, 培养1 h, 450 nm波长处测定每孔吸光度值, 计算细胞增殖率。
取对数生长期IEC-6细胞以每孔1×104接种于96孔板中, 培养24 h。加入LPS (10 μg·mL-1) 和不同浓度(25、50 μmol·L-1) 的化合物11a/1b, 模型组加不含药物的等量含0.1%的DMSO完全培养基; 空白对照组加入等量含0.1%的DMSO完全培养基; 在CO2孵育箱中继续培育24 h, 各孔加CCK-8 10 μL, 培养1 h, 450 nm波长处测定每孔吸光度值, 计算细胞增殖率。
