For quantitative analysis of related substances in TSD-1 active pharmaceutical ingredient, structures of prepared impurities were confirmed by NMR and UHPLC-MS, and a high performance liquid chromatographic method was established to determine the related substances in TSD-1. The analytical column was an Agilent ZORBAX Eclipe XDB-C8 (250 mm × 4.6 mm, 5 µm). The mobile phase A was 50 mmol·L-1 ammonium acetate solution (adjusted pH to 5.8 with acetic acid) and the mobile phase B was acetonitrile. The whole run was carried out by gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was set at 240 nm and the column temperature was 30 ℃. The resolutions among peaks of TSD-1, impurity A, impurity B, TSD-D, and TSD-F were good. The calibration curves (n = 7) of TSD-1, impurity A, impurity B, TSD-D and TSD-F were linear in their respective weight ranges of 0.242-48.4 µg·mL-1 (r = 1.000 0), 0.244-9.75 µg·mL-1 (r = 0.999 9), 0.244-4.80 µg·mL-1 (r = 0.999 9), 0.254-1.02 µg·mL-1 (r = 0.999 9), and 0.247-0.987 µg·mL-1 (r = 0.999 9). The lower limits of quantitation were 0.244, 0.244, 0.254, and 0.247 µg·mL-1 for impurity A, impurity B, TSD-D, and TSD-F, respectively, and the average recovery of each impurity ranged from 99.08% to 103.00% with high accuracy. TSD-D and TSD-F were not detected in the three batches of TSD-1 active pharmaceutical ingredients, and impurity A and impurity B were not detected beyond the limit. The established HPLC method is simple, accurate, and suitable for determination of related substances of TSD-1, which can provide a valuable reference for the subsequent development of TSD-1.

为了对TSD-1原料药中有关物质进行定量分析, 本文采用核磁共振波谱仪和超高效液相-质谱联用仪对制备的杂质进行结构确证, 并建立一种高效液相色谱法测定TSD-1中有关物质。采用Agilent ZORBAX Eclipe XDB-C8 (250 mm × 4.6 mm, 5 µm) 色谱柱, 以50 mmol·L-1乙酸铵溶液(乙酸调节至pH 5.8) 为流动相A, 乙腈为流动相B, 梯度洗脱, 流速1.0 mL·min-1, 检测波长240 nm, 柱温30 ℃。结果表明, TSD-1与杂质A、杂质B、TSD-D和TSD-F的色谱峰均能良好分离, 并分别在0.242~48.4 µg·mL-1 (r = 1.000 0)、0.244~9.75 µg·mL-1 (r = 0.999 9)、0.244~4.80 µg·mL-1 (r = 0.999 9)、0.254~1.02 µg·mL-1 (r = 0.999 9)、0.247~0.987 µg·mL-1 (r = 0.999 9) 范围内线性关系良好(n = 7)。杂质A、杂质B、TSD-D和TSD-F的定量下限分别为0.244、0.244、0.254、0.247 μg·mL-1; 各杂质的平均回收率在99.08%~103.00%之间, 准确度高。3批TSD-1原料药中TSD-D、TSD-F均未检出, 杂质A、杂质B检出量均未超限。建立的HPLC方法简便、准确, 适合TSD-1的有关物质测定, 可为TSD-1的后续研究与开发提供有价值的参考。

为了对TSD-1原料药中有关物质进行定量分析, 本文采用核磁共振波谱仪和超高效液相-质谱联用仪对制备的杂质进行结构确证, 并建立一种高效液相色谱法测定TSD-1中有关物质。采用Agilent ZORBAX Eclipe XDB-C8 (250 mm × 4.6 mm, 5 µm) 色谱柱, 以50 mmol·L-1乙酸铵溶液(乙酸调节至pH 5.8) 为流动相A, 乙腈为流动相B, 梯度洗脱, 流速1.0 mL·min-1, 检测波长240 nm, 柱温30 ℃。结果表明, TSD-1与杂质A、杂质B、TSD-D和TSD-F的色谱峰均能良好分离, 并分别在0.242~48.4 µg·mL-1 (r = 1.000 0)、0.244~9.75 µg·mL-1 (r = 0.999 9)、0.244~4.80 µg·mL-1 (r = 0.999 9)、0.254~1.02 µg·mL-1 (r = 0.999 9)、0.247~0.987 µg·mL-1 (r = 0.999 9) 范围内线性关系良好(n = 7)。杂质A、杂质B、TSD-D和TSD-F的定量下限分别为0.244、0.244、0.254、0.247 μg·mL-1; 各杂质的平均回收率在99.08%~103.00%之间, 准确度高。3批TSD-1原料药中TSD-D、TSD-F均未检出, 杂质A、杂质B检出量均未超限。建立的HPLC方法简便、准确, 适合TSD-1的有关物质测定, 可为TSD-1的后续研究与开发提供有价值的参考。

For quantitative analysis of related substances in TSD-1 active pharmaceutical ingredient, structures of prepared impurities were confirmed by NMR and UHPLC-MS, and a high performance liquid chromatographic method was established to determine the related substances in TSD-1. The analytical column was an Agilent ZORBAX Eclipe XDB-C8 (250 mm × 4.6 mm, 5 µm). The mobile phase A was 50 mmol·L-1 ammonium acetate solution (adjusted pH to 5.8 with acetic acid) and the mobile phase B was acetonitrile. The whole run was carried out by gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was set at 240 nm and the column temperature was 30 ℃. The resolutions among peaks of TSD-1, impurity A, impurity B, TSD-D, and TSD-F were good. The calibration curves (n = 7) of TSD-1, impurity A, impurity B, TSD-D and TSD-F were linear in their respective weight ranges of 0.242-48.4 µg·mL-1 (r = 1.000 0), 0.244-9.75 µg·mL-1 (r = 0.999 9), 0.244-4.80 µg·mL-1 (r = 0.999 9), 0.254-1.02 µg·mL-1 (r = 0.999 9), and 0.247-0.987 µg·mL-1 (r = 0.999 9). The lower limits of quantitation were 0.244, 0.244, 0.254, and 0.247 µg·mL-1 for impurity A, impurity B, TSD-D, and TSD-F, respectively, and the average recovery of each impurity ranged from 99.08% to 103.00% with high accuracy. TSD-D and TSD-F were not detected in the three batches of TSD-1 active pharmaceutical ingredients, and impurity A and impurity B were not detected beyond the limit. The established HPLC method is simple, accurate, and suitable for determination of related substances of TSD-1, which can provide a valuable reference for the subsequent development of TSD-1.

血栓是引发心脑血管疾病的主要因素之一, 抗血小板药能够在一定程度上降低动脉血栓疾病的发生, 已成为临床治疗急性冠状动脉综合征的主要手段[1]。P2Y12受体是P2受体家族的一员, 可被二磷酸腺苷(adenosine diphosphate, ADP) 激活, 诱导血小板聚集和血栓形成。P2Y12受体拮抗剂能够抑制血管损伤部位的持续血小板聚集, 是急性冠状动脉综合征患者常用的预防血栓事件发生的抗血小板药物之一[2, 3]
氯吡格雷是噻吩并吡啶类P2Y12受体拮抗剂的典型代表, 是目前临床上广泛应用、安全性较优的抗血小板聚集药物[4, 5]。它是前药, 本身并无活性, 口服经肠道吸收后经两步代谢成为有活性的巯基代谢物, 起到抗血栓形成的作用[6, 7]。随着临床研究的不断深入, 该药物被发现存在很多问题。一方面, 由于甲酯水解, 造成给药量中的绝大部分成为无活性的氯吡格雷酸, 生成活性代谢物的比例只有1%左右, 生物利用度极低; 另一方面, 大约有11%~44%的患者在治疗期间出现“氯吡格雷抵抗”现象, 目前研究表明这与服用者的CYP2C19基因多态性密切相关[8]。鉴于此, 天士力控股集团有限公司研究开发出具有自主知识产权的1.1类新药氘代噻吩并吡啶衍生物TSD-1 (图 1, 专利号: CN107698620A)。TSD-1是氯吡格雷活性代谢中间体的前药, 结构中羧酸甲酯氘代后, 可降低酯基水解失活的比例。同时, TSD-1经酯酶直接水解成活性代谢物2-氧-氯吡格雷-d3, 不依赖CYP2C19代谢酶, 避免了因CYP2C19酶弱代谢造成的“抵抗”。药代动力学研究结果表明, TSD-1在大鼠体内活性产物暴露量约是氯吡格雷的10倍。临床前药理学研究表明, 与氯吡格雷比较, 在同等药效基础上, TSD-1在延长正常小鼠凝血时间、抑制ADP及U46619诱导的大鼠血小板聚集率方面具有剂量小且出血风险小的优势, 目前正在申报临床研究。本研究旨在建立TSD-1原料药中有关物质检查的HPLC分析方法, 结合合成工艺以及强制降解实验, 对主要有关物质结构进行了分析和鉴定。这些研究结果可为TSD-1原料药的生产工艺过程和质量控制提供重要依据。
仪器与试药  Agilent 1260型HPLC色谱仪(美国Agilent公司); BRUKER AV-400型核磁共振波谱仪(瑞士Bruker公司); Thermo Ultimate 3000 UHPLC-TSQ Quantis三重四极杆液质联用仪(美国Thermo Fisher Scientific公司); XS105DU型十万分之一、XPE26型百万分之一电子分析天平(瑞士Mettler-Toledo公司); Waters 2545型半制备液相色谱仪(美国Waters公司)。
TSD-1原料药(批号: 181104、181105、181106)、TSD-1对照品(批号: 180323, 纯度99.5%)、杂质A对照品(批号: 190124, 纯度98.2%)、杂质B对照品(批号: 190426-1, 纯度96.7%)、TSD-D (批号: 180305, 纯度99.9%) 和TSD-F (批号: 1801261, 纯度99.2%) 均由天津天士力控股集团研究院化学药品开发中心自制(表 1); 乙腈(批号: JB084230, 色谱纯) 购于默克公司; 乙酸铵(批号: C10351235, 色谱纯)、冰乙酸(批号: 180626, 色谱纯) 购于麦克林公司; 其他试剂均为市售分析纯。
色谱条件  色谱柱: Agilent ZORBAX Eclipse XDB-C8色谱柱(250 mm × 4.6 mm, 5 µm); 流动相A: 50 mmol·L-1乙酸铵溶液(用乙酸调节至pH 5.8), 流动相B: 乙腈, 梯度洗脱(0~30 min, 25% B→70% B; 30~50 min, 70% B; 50~50.1 min, 70% B→25% B; 50.1~55 min, 25% B); 柱温: 30 ℃; 流速: 1.0 mL·min-1; 检测波长: 240 nm; 进样量: 40 µL。
对照品溶液  分别精密称取TSD-1、杂质A、杂质B、TSD-D和TSD-F的对照品适量, 加流动相A-流动相B (75∶25) 溶解并稀释, 制成每1 mL中分别含TSD-1、杂质A、杂质B、TSD-D和TSD-F为500.0、50.0、25.0、5.0和5.0 µg的溶液, 作为TSD-1和各杂质对照品储备液; 精密量取各杂质对照品储备液适量, 加流动相A-流动相B (75∶25) 定量稀释, 制成每1 mL中含杂质A、杂质B、TSD-D和TSD-F分别为5.0、2.5、0.5和0.5 µg的混合溶液, 作为杂质对照品溶液。
供试品溶液  精密称取TSD-1原料药约12.5 mg, 置25 mL量瓶中, 加流动相A-流动相B (75∶25) 溶解并稀释, 制成每1 mL中含TSD-1为0.5 mg的供试品溶液。
加标供试品溶液  精密称取TSD-1约25.0 mg, 置50 mL量瓶中, 分别加入各杂质对照品储备液5.0 mL, 加流动相A-流动相B (75∶25) 定量稀释, 摇匀, 即得加标供试品溶液。
系统适用性溶液  精密称取TSD-1原料药及4个杂质对照品适量, 加流动相A-流动相B (75∶25) 溶解并稀释, 制成每1 mL中含TSD-1、杂质A、杂质B、TSD-D和TSD-F分别为500.0、5.0、5.0、5.0和5.0 µg的系统适用性溶液。
线性系列对照品溶液  精密量取各对照品储备液适量, 加流动相A-流动相B (75∶25) 定量稀释, TSD-1质量浓度分别为50.00、25.00、15.00、5.00、2.50、0.50和0.25 µg·mL-1的线性溶液, 制成杂质A质量浓度分别为10.00、8.00、5.00、3.00、1.00、0.50和0.25 µg·mL-1的线性溶液; 制成杂质B质量浓度分别为5.00、3.00、2.50、1.00、0.75、0.50和0.25 µg·mL-1的线性溶液; 制成TSD-D、TSD-F质量浓度分别为1.00、0.85、0.65、0.50、0.45、0.35和0.25 µg·mL-1的线性溶液。
回收率加标溶液  精密称取TSD-1约25.0 mg, 置50 mL量瓶中, 平行制备9份, 分为3组, 分别加入各杂质对照品储备液适量, 用流动相A-流动相B (75∶25) 定量稀释制成各杂质浓度分别为限度浓度的LOQ、100%和150%, 即得。
TSD-1储备溶液  称取TSD-1原料药100.0 mg, 置100 mL量瓶中, 加流动相B溶解并稀释至刻度, 摇匀即得。
酸破坏TSD-1  量取TSD-1储备溶液5.0 mL, 加1 mol·L-1盐酸溶液1.0 mL, 常温放置72 h, 加1 mol·L-1氢氧化钠溶液1.0 mL中和。
碱破坏TSD-1  量取TSD-1储备溶液5.0 mL, 加1 mol·L-1氢氧化钠溶液1.0 mL, 60 ℃水浴中放置8 h, 加1 mol·L-1盐酸溶液1.0 mL中和。
氧化破坏TSD-1  量取TSD-1储备溶液5.0 mL, 加入3%过氧化氢溶液1.0 mL, 常温放置4 d。
高温破坏TSD-1  量取TSD-1储备溶液5.0 mL, 60 ℃水浴中放置4 h。
光照破坏TSD-1  量取TSD-1储备溶液5.0 mL, 光照强度4 500 lx的光照箱中照射24 h。
取上述各破坏后的样品溶液, 分别转移至10 mL量瓶中, 加流动相B稀释至刻度, 摇匀即得。
杂质的制备  根据本品的合成工艺以及可能的降解途径分析, 结合多批次样品的有关物质检测结果及影响因素实验考察结果, 研究出本品中可能存在的有关物质有4个, 分别为杂质A、杂质B、TSD-D和TSD-F。其中杂质A为合成过程中的副产物, 杂质B为热降解杂质, TSD-D和TSD-F为合成中间体; 经半制备液相分离纯化得到杂质A、杂质B、TSD-D和TSD-F。
系统适用性  取系统适用性溶液在建立的HPLC方法下进样测定, 考察方法的系统适用性。
专属性  分别取各破坏实验溶液在建立的HPLC方法下进样测定, 考察方法的专属性。
线性关系考察  分别取各线性系列对照品溶液在建立的HPLC方法下进样测定, 以浓度为横坐标, 峰面积为纵坐标, 绘制标准曲线, 计算回归方程。
检测下限和定量下限  精密量取各对照品储备溶液适量, 逐级稀释, 在建立的HPLC方法下进样测定, 得到信噪比大于3的浓度为杂质的检测下限, 信噪比大于10的浓度为杂质的定量下限。
重复性  平行制备供试品溶液和加标供试品溶液各6份, 在建立的HPLC方法下进样测定, 记录峰面积, 采用外标法计算各杂质含量, 并分别计算6组测定所得含量的RSD (n = 6), 考察其重复性。
中间精密度  为考察随机变动因素对精密度的影响, 在同一实验室, 由不同实验人员在不同日期, 平行制备供试品溶液和加标供试品溶液各6份, 在建立的HPLC方法下进样测定, 记录峰面积, 采用外标法计算各杂质的含量, 并分别与重复性项下6组杂质的含量合并, 计算12组测定所得含量的RSD (n = 12), 考察其中间精密度。
溶液稳定性  系统适用性溶液和供试品溶液在制备后置于室温放置0、2、4、6、8、10和12 h和在制备后置于冷藏(4~8 ℃) 放置0、2、4、6、8、10、12、14、16、18、20、22和24 h, 在建立的HPLC方法下进样测定, 考察杂质峰面积随放置时间的变化情况。
准确度  取回收率加标溶液在建立的HPLC方法下进样测定并计算其回收率(即为准确度), 计算公式为: 回收率(%) = (回收率加标溶液测得量- 本底原有量) / 对照品加入量× 100%。
耐用性  将建立的HPLC色谱条件进行微小改变, 如在流速(1.0 ± 0.1 mL·min-1)、柱温(30 ± 2 ℃)、流动相A的pH (5.8 ± 0.1) 等发生微小变动时, 取供试品溶液进行测定, 记录峰面积, 以外标法计算各杂质的含量, 考察方法的耐用性。
样品测定  取3批TSD-1原料药(批号: 181104、181105、181106), 制备供试品溶液并进行测定, 以外标法计算各杂质的含量。
杂质A、杂质B、TSD-D和TSD-F分别经HR-MS、1H NMR、13C NMR确证化学结构, 化合物结构图谱数据见表 1
在建立的HPLC方法下, TSD-1与相邻杂质峰分离良好, 各色谱峰与相邻色谱峰分离度均大于1.5, 理论板数按TSD-1峰计算不低于5 000, 且满足系统适用性要求。
在建立的HPLC方法下, TSD-1在光照破坏、高温破坏条件下较稳定, 杂质个数、总量无明显变化, 而在酸破坏、氧化破坏、碱破坏条件下均有不同程度的降解。各破坏条件下所产生的降解产物均与TSD-1能很好分离, 峰纯度分析显示各破坏溶液色谱图中TSD-1均为单一峰, 且不干扰已知杂质的测定; 以未破坏溶液总峰面积为100%计, 各破坏溶液物料平衡率为95%~105%, 物料均守恒, 表明该色谱条件的专属性良好(图 2)。
线性回归方程、检测下限和定量下限结果如表 2所示。结果表明, TSD-1在0.242~48.4 µg·mL-1、杂质A在0.244~9.75 µg·mL-1、杂质B在0.244~4.80 µg·mL-1、TSD-D在0.254~1.02 µg·mL-1、TSD-F在0.247~0.987 µg·mL-1浓度内与峰面积呈良好的线性关系。按照有关物质测定供试品溶液质量浓度0.50 mg·mL-1计算, 供试品质量浓度相当于定量下限的2 000倍, 相当于检测下限的6 666倍; 各杂质的LOD和LOQ均远低于其限度, 该方法的灵敏度满足检测要求。
6组供试品溶液中检出杂质A、杂质B含量的RSD (n = 6) 分别为1.87%和0, TSD-D和TSD-F均未检出; 6组加标供试品溶液中检出杂质A、杂质B、TSD-D和TSD-F含量的RSD (n = 6) 分别为1.33%、0.28%、0.36%和1.45%; 方法重复性良好。
12组供试品溶液中检出杂质A、杂质B含量的RSD (n = 12) 分别为4.43%和0, TSD-D和TSD-F均未检出; 12组加标供试品溶液中检出杂质A、杂质B、TSD-D和TSD-F含量的RSD (n = 12) 分别为1.01%、0.23%、1.06%和1.17%; 方法中间精密度良好。
系统适用性溶液在室温保存条件下, TSD-1、杂质A、杂质B、TSD-D和TSD-F峰面积的RSD分别为0.17%、0.43%、2.67%、0.28%和0.37%; 系统适用性溶液在冷藏(4~8 ℃) 保存条件下, TSD-1、杂质A、杂质B、TSD-D和TSD-F峰面积的RSD分别为0.14%、0.50%、0.46%、0.48%和0.41%。供试品溶液在室温保存条件下, TSD-1、杂质A和杂质B峰面积的RSD分别为0.17%、1.10%和0.43%, TSD-D和TSD-F均未检出; 供试品溶液在冷藏(4~8 ℃) 保存条件下, TSD-1、杂质A和杂质B峰面积的RSD分别为0.14%、0.58%和0.32%, TSD-D和TSD-F均未检出。结果表明, 系统适用性溶液和供试品溶液在室温保存条件下, 12 h内稳定; 系统适用性溶液和供试品溶液在冷藏(4~8 ℃) 保存条件下, 24 h内稳定。
经计算, 样品中杂质A、杂质B、TSD-D和TSD-F的平均回收率(n = 9) 分别为103.00%、99.08%、100.92%和100.74%, RSD均小于3%, 表明方法准确度较高。
在流速、柱温、流动相A的pH等发生微小变动时, 供试品检出的杂质含量基本无变化, 各组分均能得到较好的分离, 方法耐用性良好。
样品测定结果如表 3所示, TSD-D、TSD-F均未检出, 杂质A、杂质B检出量均未超限。
本研究杂质限度的确定参考ICH Q3A (R2) 附件中对杂质限度的要求和《中华人民共和国药典》2020年版(四部) 通则《药品杂质分析指导原则》[9, 10]。本品日服用最大剂量为50 mg, 依据药学、毒理研究等安全性数据, 结合本品的稳定性和大生产的可行性, 将杂质A的限度暂定为1.0%, 杂质B的限度暂定为0.5%, TSD-D、TSD-F的限度暂定为0.1%, 最大未知杂质的限度暂定为0.1%, 总杂质的限度暂定为2.0%。
在药品研发的过程中, 安全性和有效性是评价药物质量研究的重要手段, TSD-1作为创新药, 质量研究不可或缺。本文建立一种有效检测TSD-1原料药中有关物质的高效液相色谱法, 并对该方法进行验证。结果表明, 本方法专属性好, 灵敏度高, 线性关系及重复性良好, 可为TSD-1原料药有关物质研究提供检测方法与可靠的实验数据, 同时也为TSD-1制剂的开发提供保障。
TSD-1是应用氘代技术对氯吡格雷结构进行改造的产物, 这是一次很好的尝试。在新药开发成功率低下的今天, 氘代策略既能优化已上市药品, 又能有效地挽救临床试验失败的药品, 同时也可以促进新药的研发。与非氘代药物相比, 氘代药物能有效改善药物的安全性及药代动力学特性, 使患者获益显著。
