药性是中药的“身份证”，而归经又是破解中药药性理论的密钥。本实验基于分子热力学，从微观的能量释放与吸收角度研究中药与离体器官相互作用的效应变化，以期构建表征归经理论的新思路。以黄芩为例，应用等温滴定量热技术（isothermal titration calorimetry，ITC）分别测定黄芩与其主要活性成分黄芩苷与离体组织脑、心、肺、脾、肾的相互作用过程中能量的变化，比较结合常数（association constant，K_a）、解离常数（disassociation constant，K_d）、结合计量比（stoichiometry ratio，n）、焓变（enthalpy change，ΔH）、熵变（entropy change，ΔS）和吉布斯自由能（Gibbs free energy，ΔG）等热力学参数。结果发现，黄芩与肺的作用强度明显高于其他脏器，此结论与古籍黄芩归经一致；此外，主要活性成分黄芩苷也显现与黄芩一致的作用规律。热力学参数分析结果表明该作用为熵和焓共同驱动的弱键诱导的自发化学结合反应。比旋度测量结果进一步证明黄芩苷与肺存在相互作用，并与ITC滴定结果相吻合，表明ITC能用于表征中药归经理论。动物实验获得北京中医药大学实验动物伦理委员会批准。因此，基于ITC建立的以中药水煎液与其活性成分分别与离体组织相互作用的能量变化大小表征中药归经的方法是科学、可行的，此实验为探讨中药及其功效成分归经理论提供了新思路。

The property of medicine is the "identity card" of traditional Chinese medicine (TCM), and the key to crack the theory of property of TCM. Based on molecular thermodynamics, the effects of interaction between TCM and organs in vitro were studied from the perspective of micro-energy release and absorption in order to construct a new idea of characterizing meridian theory. Scutellaria baicalensis, for example, application of isothermal titration calorimetry (ITC) were used to determine the energy changes during the interaction of Scutellaria baicalensis and its main active ingredient baicalin with brain, heart, lung, spleen and kidney in vitro, comparison including the association constant (K_a) and disassociation constant (K_d), combined with thermodynamic parameters, such as stoichiometry ratio (n), enthalpy change (ΔH), entropy change (ΔS), Gibbs free energy (ΔG), it is found that the interaction intensity between Scutellaria baicalensis and lung is significantly stronger than that of other organs, which is consistent with the theory of the return of Scutellaria baicalensis in ancient books. In addition, baicalin, the main active ingredient, showed the same action pattern as Scutellaria baicalensis. The thermodynamic parameters analysis showed that the action was a weak bond-induced spontaneous chemical binding reaction driven by both entropy and enthalpy. The results of specific curl measurement further proved the interaction between baicalin and lung, and were consistent with the results of ITC titration, indicating that ITC could be used to characterize the meridian tropism of TCM. Therefore, based on ITC, it is scientific and feasible to characterize the meridian of TCM by the energy change of the interaction between the decoction of TCM and its active components and the in vitro tissues respectively. This experiment provides a new idea for the discussion of meridian of TCM.

自《神农本草经》开始, 中医药界对于归经的探讨从未停歇。仅就归经的理论释义而言, 各家解释层出不穷^{[1, 2]}, 现代研究方法自然也呈现出百家争鸣的局面, 目前有效成分的体内观测法^[3]、微量元素体内观测法^[4]、环核苷酸检测法^[5]、受体效应^[6]和超分子化学^[7-10]等均从新角度对归经理论做出了一定的解释, 推动了归经理论进一步发展。然而, 缺乏统一、规范的标准阻碍了归经理论国际化的发展脚步, 本实验试图从分子热力学的角度对归经理论做出一定的解释。

生物的整个生命过程中永恒地伴随着热量的变化, 植物、动物甚至于人类自身都是一座巨大的热量源, 每时每刻都在与环境发生热量交换。从热力学角度而言, 中药-人体的作用像是特异性的热量调节剂, 中药有效成分吸收后选择性地作用于特定的组织或器官, 改变人体能量的发生、传导和消亡过程, 矫正偏离正常轨道的吸收代谢途径, 维持机体的健康, 这也是中药归经的一种体现。目前, 从中药与动物的整体热量变化角度研究归经理论存在较大的难度和误差, 因此, 本实验尝试从中药与动物不同器官的相互作用探讨归经理论的科学内涵。黄芩为清热药, 主归肺经。本课题组前期利用等温滴定量热技术(isothermal titration calorimetry, ITC)探究了黄芩质量表征的新方法^[11], 结果表明, ITC检测小檗碱滴定黄芩水煎液的能量变化幅度可以代表黄芩中黄酮类化合物的含量, 较紫外-可见分光光度法灵敏度更高, 方法简便、科学、可行。通过拆方研究解析黄连解毒汤中自沉淀来源, 在分子热力学基础上探讨水煎自沉淀的形成机制^[12]。结合前期研究成果, 利用ITC, 分别探讨黄芩及其主要成分黄芩苷与脑、心、肺、脾、肾等脏器的热力学和动力学信息, 旨在利用分子热力学表征黄芩归经理论的一致性, 同时为黄芩苷的归经研究提供一定参考(图 1)。

材料与方法

仪器与试剂 NANO ITC等温滴定量热仪、脱气站Degassing Station (美国TA公司); 黄芩饮片购自北京同仁堂(经北京中医药大学刘春生教授鉴定为唇形科植物黄芩Scutellaria baicalensis Georgi的干燥根), 储存于本实验室; 黄芩苷(中国食品药品检定研究院, 批号: 110715-201720, 纯度: 93.5%)。

动物 SPF级SD大鼠, 体重190~220 g, 北京维通利华实验动物技术有限公司, 合格证号: 1100111911014046。实验过程遵循北京中医药大学伦理委员会的规定。

供试品溶液的制备 称取黄芩饮片2 g, 加入10倍量水冷凝回流提取1 h, 趁热8层纱布过滤, 4 000 r·min^-1离心30 min, 取上清液稀释25倍, 经0.45 μm微孔滤膜过滤即得黄芩水煎液, 备用。精密称取黄芩苷4.46 mg加水溶解到10 mL, 配成1 mmol·L^-1黄芩苷水溶液, 备用。

组织匀浆液的制备 腹主动脉取血后, 取健康雄性大鼠心、脾、肺、肾、脑组织, 称取大鼠组织适量, 按质量(g)与体积(mL)比为1:9加生理盐水, 制成组织匀浆。匀浆液3 000 r·min^-1、4 ℃离心15 min, 取组织上清液, 备用。

紫外分光光度法测定蛋白含量 依据中国药典考马斯亮蓝法, 以牛血清白蛋白为标准品测定吸光度(A)值, 建立标准曲线。吸取组织上清液选择A值为0.400 ± 0.020的样品浓度为ITC被滴定液。

ITC测定热量变化 实验前, 黄芩水煎液和黄芩苷溶液放入脱气站中37 ℃恒温脱气处理30 min。在样品池中精确注入被滴定溶液脑、心、肺、脾、肾组织稀释液300 μL, 进样针精确吸入滴定溶液(黄芩水煎液和黄芩苷溶液) 50 μL, 37 ℃恒温条件下调整搅拌速率为250 r·min^-1, 等待仪器自动平衡, 逐次将黄芩水煎液和黄芩苷溶液分别滴定脑、心、肺、脾、肾组织稀释液, 连续滴定20滴, 每滴注入时间为2.5 s, 相邻滴定间隔时间为180 s。滴定过程中, 样品池中的热量变化曲线自动被ITC Run软件采集、保存, 使用NanoAnalyze软件对热量变化曲线进行分析, 输入实验用样品浓度, 选择Independent模型进行拟合。得到反应焓变(∆H)及平衡常数(K), 根据热力学参数关系式∆G = -RTlnK和∆G = ∆H-T∆S (∆G:吉布斯自由能; R:气体常数; T:绝对温度; ∆S:熵变), 求得∆G和∆S ^[13]。

1 紫外分光光度法测定蛋白浓度

依据考马斯亮蓝法, 以牛血清白蛋白为对照品溶液作标准曲线, 线性方程: y = 0.123 x - 0.057, R² = 0.999。调整组织液浓度, 使A值在0.400 ± 0.020区间, 肺、心、脑、脾和肾组织液的质量浓度分别为0.050、0.067、0.043、0.056和0.041 mg·mL^-1。

2 ITC能量变化

以黄芩苷溶液滴定磷酸盐缓冲溶液(phosphate buffer saline, PBS)为能量基线, 黄芩苷溶液滴定不同组织的热量变化曲线如图 2。根据以上滴定所得的能量变化数据建立滴定能量变化折线图(图 3)。由图 2可知, 黄芩苷与组织间发生的反应是一个放热过程, 较之黄芩苷滴定PBS热量释放更为明显, 显示黄芩苷确实与不同组织存在相互作用。黄芩苷滴定过程中的热量变化由高到低顺序依次为:肺 > 心 > 肾 > 脾 > 脑, 说明黄芩苷-肺的相互作用力强于其他脏器。比较黄芩与黄芩苷的滴定过程(图 3), 发现二者作用部位相似但强度有所差别, 与黄芩归肺经的中医思想一致。

与此同时, 对比实验选用加热失活后的组织为被滴定液, 黄芩和黄芩苷滴定相同蛋白浓度下的不同失活组织, 发现能量变化无差异, 进一步证明样品与蛋白的相互作用存在特异性, 而非简单的物理吸附。

3 黄芩苷-肺相互作用

3.1 黄芩苷滴定肺过程

分析黄芩苷滴定肺过程的热力学参数ΔG = -17.18 kJ·mol^-1 < 0, ΔH = -12.34 kJ·mol^-1 < 0, -TΔS = -5.473 kJ·mol^-1 < 0, 说明该过程为熵和焓共同驱动的自发过程, 黄芩苷和肺组织中的受体蛋白质可能发生了非共价键, 如氢键的结合^[14]。n = 2.266, 说明黄芩苷与肺组织的平均结合比例可能为2:1^{[15, 16]}。

3.2 旋光法测定旋光度

旋光仪校正稳定后, 测得肺组织旋光度[α]_D³⁷ = -0.008° (C = 0.250, PBS, C指以牛血清白蛋白标定, 肺组织液中蛋白质含量为0.250 mg·mL^-1); 黄芩苷旋光度[α]_D³⁷ = -0.001° (C = 0.150, PBS, C指黄芩苷质量浓度为0.150 mg·mL^-1); 黄芩苷与肺组织混合液旋光度[α]_D³⁷= -0.030° (C = 0.250, PBS, C指混合后肺组织液中蛋白含量为0.250 mg·mL^-1, 黄芩苷质量浓度为0.150 mg·mL^-1); 黄芩苷-肺混合溶液的旋光度与肺组织液、黄芩苷溶液比较左旋程度增加, 证明二者间发生了相互作用, 不同于机械混合, 与滴定结果一致, 二者共同验证了黄芩归肺经的药性思想。

利用等温量热滴定技术研究黄芩与不同组织的结合力, 验证黄芩与古籍归经理论的一致性。并且, 选取黄芩中的主要活性成分黄芩苷重复上述实验, 发现黄芩苷与黄芩主要作用部位相似, 但强度稍有上升。

中医脏腑和西医中的“脏”并非一一对应, 但前者总是以后者的某一个脏器为主体构成, 这也是通过中药活性成分体内分布来研究归经理论的基础。各家学说对黄芩归经情况也存在异议, 《神农本草经》言: “主诸热黄疸, 肠澼泄痢, 逐水下血闭, 恶疮疽蚀火疡”。《药性论》言: “能治热毒, 骨蒸, 寒热往来, 肠胃不利, 破拥气, 治五淋, 令人宣畅, 去关节烦闷, 解热渴。制热, 腹中疞痛, 心腹坚胀”。现今也有研究认为枯(片)芩偏清泻肺火, 条(子)芩偏清泻大肠湿热。总而言之, 黄芩多作用于中上焦, 主归肺、大肠经, 还作用于胃、脾经, 存在多脏腑联动作用。

实验结果显示, 黄芩与其主要活性成分黄芩苷主要作用于肺部。金鹏等^[17]发现静脉注射黄芩苷镁盐0.25 h后各组织中药物浓度最高, 至0.75 h目标成分质量分数迅速降低, 其中肾脏作为主要排泄器官, 分布最多, 其次是肺。肾脏作为排泄的主要器官, 药效物质在体内经代谢后集中于肾排泄, 浓度较高。排除肾脏排泄和肝脏代谢的影响, 黄芩苷在肺组织中含量最高, 与本实验结果相吻合。但本实验发现黄芩苷与肾组织相互作用的能量变化较小, 说明作用力较弱, 推测组织分布浓度的高低不一定代表相互作用力的强弱^[18]。肝脏是机体的主要代谢器官, 大多数中药成分入血后会集中于肝脏, 发生分解转化等变化, 因此黄芩苷与肝的相互作用未作为参考依据。

基于等温量热技术探讨中药归经理论是一次全新的尝试, 引入新仪器、新技术有助于多角度、全方位地解析中医药理论的科学内涵, 在现有理论基础上进行外延, 不仅赋予更多“舶来品”如玛咖以中药药性信息; 还可以探讨中药功效成分的药性参数, 为临床提供成分饮片, 扩大用药范围, 延续中医药文化。
