The chemical constituents of Viburnum taitoense Hayata were investigated using column chromatography silica gel and Sephadex LH-20, etc. Seven pentacyclic triterpenoids were isolated and their structures were elucidated by spectral data and physicochemical properties as 3β, 6β-dihydroxy olean-11, 13(18)-dien-28-acid (1), 3β-hydroxy olean-11, 13(18)-diene-28-acid (2), 12-ene-olean-28-acid-3β-palmitate (3), 3β-acetylcocodiol (4), corosolic acid (5), uvaol (6) and ursolic acid (7). Among them, compound 1 is a new oleanane type triterpenoid and its absolute configuration was confirmed by single-crystal X-ray diffraction data. Compounds 2-5 were isolated from this genus for the first time. All the compounds were evaluated for their anti-inflammatory activities in vitro, and compound 5 showed significant inhibitory activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW264.7 macrophage cells with an IC50 of 25.52 ±0.56 μmol·L-1 when compared to the positive control, quercetin (IC50 of 25.46 ±0.62 μmol·L-1).

采用硅胶柱色谱、Sephadex LH-20柱色谱等色谱方法,对台东荚蒾乙酸乙酯部位进行分离,从中分离纯化得到7个三萜类化合物,根据其波谱数据和理化性质鉴定为3β,6β-二羟基齐墩果-11,13(18)-二烯-28-酸(1)、3β-羟基齐墩果-11,13(18)-二烯-28-酸(2)、12-烯-齐墩果-28-酸-3β-棕榈酸酯(3)、3β-乙酰古柯二醇(4)、科索酸(5)、熊果醇(6)和熊果酸(7)。其中,化合物1为新化合物,其绝对构型通过单晶衍射得以证实。化合物2~5为首次从荚蒾属中分离得到。采用LPS诱导的RAW264.7释放NO的细胞模型,评价了化合物1~7的体外抗炎活性,结果显示,与阳性对照槲皮素(IC50值为25.46 ±0.62 μmol·L-1)相比,化合物5显示出较强的NO抑制活性,其IC50值为25.52 ±0.56 μmol·L-1

采用硅胶柱色谱、Sephadex LH-20柱色谱等色谱方法,对台东荚蒾乙酸乙酯部位进行分离,从中分离纯化得到7个三萜类化合物,根据其波谱数据和理化性质鉴定为3β,6β-二羟基齐墩果-11,13(18)-二烯-28-酸(1)、3β-羟基齐墩果-11,13(18)-二烯-28-酸(2)、12-烯-齐墩果-28-酸-3β-棕榈酸酯(3)、3β-乙酰古柯二醇(4)、科索酸(5)、熊果醇(6)和熊果酸(7)。其中,化合物1为新化合物,其绝对构型通过单晶衍射得以证实。化合物2~5为首次从荚蒾属中分离得到。采用LPS诱导的RAW264.7释放NO的细胞模型,评价了化合物1~7的体外抗炎活性,结果显示,与阳性对照槲皮素(IC50值为25.46 ±0.62 μmol·L-1)相比,化合物5显示出较强的NO抑制活性,其IC50值为25.52 ±0.56 μmol·L-1

The chemical constituents of Viburnum taitoense Hayata were investigated using column chromatography silica gel and Sephadex LH-20, etc. Seven pentacyclic triterpenoids were isolated and their structures were elucidated by spectral data and physicochemical properties as 3β, 6β-dihydroxy olean-11, 13(18)-dien-28-acid (1), 3β-hydroxy olean-11, 13(18)-diene-28-acid (2), 12-ene-olean-28-acid-3β-palmitate (3), 3β-acetylcocodiol (4), corosolic acid (5), uvaol (6) and ursolic acid (7). Among them, compound 1 is a new oleanane type triterpenoid and its absolute configuration was confirmed by single-crystal X-ray diffraction data. Compounds 2-5 were isolated from this genus for the first time. All the compounds were evaluated for their anti-inflammatory activities in vitro, and compound 5 showed significant inhibitory activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW264.7 macrophage cells with an IC50 of 25.52 ±0.56 μmol·L-1 when compared to the positive control, quercetin (IC50 of 25.46 ±0.62 μmol·L-1).

忍冬科(Caprifoliaceae)荚蒾属(Viburnum)植物全世界有230余种, 主要分布在亚热带和温带地区。我国有80余种, 其中药用46种, 广泛分布于各省区, 多作为民族药用于治疗咳嗽、腹泻、风湿和类风湿性关节炎以及促进骨折愈合等[1, 2]。目前从20多种荚蒾属植物中共分离得到近400个单体化合物, 主要包括二萜类、三萜类、环烯醚萜类和酚类等, 还有少量甾体、生物碱和脂肪酸等, 多具有抗肿瘤、神经保护、抗炎、抗氧化、抗菌等多方面的生物活性[3-6]。台东荚蒾(V. taitoense Hayata)是荚蒾属的药用植物, 主要在广西、贵州、云南等地作为民族药用于治疗感冒、风湿疼痛和骨折等[7, 8]。为揭示台东荚蒾的药效物质基础, 丰富荚蒾属的化学成分研究, 本实验对台东荚蒾进行系统的化学成分分离, 从中分离并鉴定了7个三萜类化合物(图 1), 并采用LPS诱导RAW264.7释放NO的细胞模型, 测试了化合物1~7的体外抗炎活性。结果表明与阳性对照槲皮素(IC50值为25.46 ± 0.62 μmol·L-1)相比, 化合物5显示出较强的NO抑制活性, 其IC50值为25.52 ± 0.56 μmol·L-1, 新化合物1显示出较弱的NO抑制活性, 其IC50值为44.16 ± 0.91 μmol·L-1
化合物1无色针晶, 10%硫酸-乙醇显色紫红色。[α]D20-111.73 (c 0.50, MeOH); UV (MeOH) λmax (log ε): 259 (4.51)、250 (4.69) nm; IR显示该化合物中含有羟基(3 435 cm-1)、羰基(1 709 cm-1)、共轭双键(1 630 cm-1)、甲基和亚甲基(2 931、2 865、1 385 cm-1)等。HR-ESI-MS m/z 493.328 7 [M+Na]+ (calcd. for C30H46NaO4 493.329 4), 提示化合物1分子式为C30H46O4, 计算不饱和度为8。
化合物11H NMR (500 MHz, pyridine-d5)谱中(表 1), 显示有2个烯烃质子信号δH 6.78 (1H, dd, J = 10.5, 3.0 Hz)和δH 5.96 (1H, d, J = 10.5 Hz); 7个甲基质子信号δH 1.76、1.75、1.66、1.49、1.17、0.96、0.95 (各3H, s); 以及2个连氧次甲基信号δH 4.88 (1H, s)和δH 3.54 (1H, dd, J = 11.5, 4.0 Hz)。其13C NMR (125 MHz, pyridine-d5)及DEPT 135显示该化合物有30个碳信号, 其中有7个甲基碳信号、6个亚甲基碳信号、8个次甲基碳信号, 其余9个为季碳信号。其中δC 179.4推测为羧基碳信号, δC 136.9、133.8、127.6、126.4推测为一对共轭双键碳信号, δC 79.1和68.4推测为连氧的次甲基碳信号。根据以上信息推测1为五环三萜酸类化合物, 含有一对共轭双键和两个羟基[9]。化合物11H NMR和13C NMR数据与化合物3β-羟基-齐墩果-11, 13(18)-二烯-28-酸(2)[10]相似, 而1在C-6位多一个羟基, 这一位置的确定可通过HMBC得以证实。
1H-1H COSY谱中, 可见质子信号δH 6.78 (H-11)与δH 5.96 (H-12)、δH 2.26 (H-9);质子信号δH 4.88 (H-6)与δH 1.06 (H-5)、δH 1.70 (H-7α)以及δH 1.90 (H-7β); 质子信号δH 3.54 (H-3)与δH 1.21 (H-1α)、δH 2.00 (H-1β)、δH 2.02 (H-2α)、δH 2.15 (H-2β); 质子信号δH 2.67 (H-22α)与δH 1.56 (H-22β)、δH 1.74 (H-21α)、δH 1.38 (H-21β)分别有偶合相关关系(图 2)。在HMBC谱(图 2)中, 质子信号δH 4.88 (H-6)与碳信号δC 56.5 (C-5)、41.0 (C-7)、41.4 (C-8)和37.3 (C-10)有远程相关; 质子信号δH 1.06 (H-5)与碳信号δC 79.1 (C-3)、40.8 (C-4)、68.4 (C-6)、37.3 (C-10)、28.4 (C-23)、18.0 (C-24)相关, 说明1在C-6位有一个羟基。化合物1结构中C-3位及C-6位羟基的相对构型, 可通过ROESY谱来确定。在化合物1的ROESY谱(图 3)中, 可见质子信号δH 3.54 (H-3)与质子信号δH 1.21 (H-1α)、δH 2.02 (H-2α)、δH 1.06 (H-5)及δH 1.49 (H-23)有NOE相关, 质子信号δH 4.88 (H-6)与质子信号δH 1.06 (H-5)、δH 1.70 (H-7α)、δH 1.90 (H-7β)及δH 1.49 (H-23)有NOE相关, 表明H-3和H-6均为α构型, 则C-3和C-6上的羟基均为β构型, 因此化合物1的结构被确定为3β, 6β-二羟基-齐墩果-11, 13(18)-二烯-28-酸, 其绝对构型(3S, 5R, 6R, 8R, 9S, 10R, 14S, 17S)通过单晶衍射得以确定(图 4)。
采用MTT法[11, 12]检测化合物1~7对LPS诱导的RAW264.7释放NO的抑制作用, 结果表明与阳性对照槲皮素(IC50值为25.46 ± 0.62 μmol·L-1)相比, 化合物5显示出较强的NO抑制活性, 其IC50值为25.52 ± 0.56 μmol·L-1, 新化合物1显示出较弱的NO抑制活性, 其IC50值为44.16 ± 0.91 μmol·L-1 (表 2)。
JASCO P-1020旋光仪(日本JASCO公司); TU 1901紫外-可见光分光光度计(北京); Spectrum 100傅里叶变换红外光谱仪(美国PerkinElmer公司); Bruker DRX-500和Bruker ARX-600核磁共振谱仪(瑞士Bruker公司); Agilent G6230飞行时间质谱仪、Agilent 1100-LC/MSD TrapSL质谱仪、SuperNova AtlasS2单晶衍射仪(美国Agilent公司)。柱色谱用硅胶(100~200目及200~300目, 青岛海洋化工厂); Sephadex LH-20 (瑞典Pharmacia公司); 显色剂为10%硫酸-乙醇溶液; 其他提取、分离所用试剂均为分析纯。
台东荚蒾新鲜枝叶采集自广西壮族自治区河池市罗城仫佬族自治县, 经广西中医药研究院黄云峰副研究员鉴定为忍冬科荚蒾属台东荚蒾Viburnum taitoense Hayata的枝叶, 药材标本(编号: JM201607)存放于广西医科大学药学院天然药物化学实验室。
台东荚蒾新鲜枝叶50 kg, 切碎, 常温下用3倍量乙酸乙酯浸提, 每次7天, 共3次。合并浸提液减压浓缩回收溶剂, 得到乙酸乙酯浸提物稠膏。药渣再用3倍量80%乙醇常温下浸提, 每次7天, 共3次, 得到的80%乙醇浸提液减压浓缩回收溶剂后得到80%乙醇浸提物浸膏, 浸膏用10 L水混悬后, 依次用等体积乙酸乙酯、正丁醇各萃取3次, 合并各萃取液并减压浓缩回收溶剂, 得到乙酸乙酯萃取物与上述乙酸乙酯浸提物合并, 得乙酸乙酯层浸膏共2.1 kg, 进行硅胶柱色谱分离, 二氯甲烷-甲醇(100:0, 50:1, 25:1, 10:1, 1:1, 0:100)梯度洗脱, 得到6个流份F1~F6。
流份F1 (490.4 g)经硅胶柱色谱分离, 环己烷-乙酸乙酯(50:1, 25:1, 10:1, 5:1, 1:1, 0:1)梯度洗脱, 得到7个流份F1A~F1G。流份F1C (64.4 g)经反复硅胶柱色谱分离, 环己烷-乙酸乙酯(100:0, 50:1, 25:1, 10:1, 1:1)梯度洗脱, 再经Sephadex LH-20 (氯仿-甲醇=1:1)纯化, 得到化合物3 (80.5 mg)、4 (30.7 mg); 流份F1D (58.4 g)经反复硅胶柱色谱, 环己烷-乙酸乙酯(50:1, 25:1, 10:1, 5:1, 1:1)梯度洗脱, 再经Sephadex LH-20 (氯仿-甲醇=1:1), 得到化合物2 (65.0 mg)、6 (87.4 mg)。
流份F4 (460.9 g)经硅胶柱色谱分离, 二氯甲烷-甲醇(40:1, 30:1, 20:1, 10:1, 1:1)梯度洗脱, 得到9个流分F4A~F4I。流份F4A (16.4 g)经反复硅胶柱色谱分离, 石油醚-乙酸乙酯(50:1, 40:1, 30:1, 20:1, 10:1, 1:1)梯度洗脱, 二氯甲烷-甲醇(100:0, 50:1, 25:1, 10:1, 1:1)梯度洗脱, 再经Sephadex LH-20 (氯仿-甲醇=1:1)纯化, 得化合物7 (800 mg)。流份F4B (7.6 g)经反复硅胶柱色谱分离, 二氯甲烷-乙酸乙酯(50:1, 40:1, 30:1, 20:1, 10:1, 1:1), 再经乙酸乙酯重结晶纯化, 得化合物5 (43.1 mg)。流份F4D (74.6 g)经反复硅胶柱色谱分离, 二氯甲烷-甲醇(100:1, 50:1, 25:1, 10:1, 1:1)梯度洗脱, 再经Sephadex LH-20 (氯仿-甲醇=1:1)和无水乙醇重结晶纯化得到化合物1 (30.4 mg)。
化合物1  无色针晶, 10%硫酸乙醇显色紫红色, [α]D20-111.73 (c 0.50, MeOH); IR (KBr, cm-1) νmax: 3 435, 2 931, 2 865, 1 709, 1 630, 1 457, 1 385, 1 265, 1 193, 1 130, 1 048, 856, 772, 747, 630, 572; HR-ESI-MS给出准分子离子峰m/z 493.328 7 [M+Na]+ (计算值C30H46NaO4 493.329 4), 其1H NMR和13C NMR数据见表 1
化合物1在氯仿-甲醇(1:1)中获得晶体, 从中选取一外形大小为0.13 mm×0.12 mm×0.11 mm的单晶用于X-单晶衍射实验, 在SuperNova AtlasS2单晶衍射仪上收集衍射强度数据, Cu Kα辐射(波长λ=1.541 84 Å ), 在6.094°≤2θ≤147.278°范围内, 以ω-2θ扫描方式, 在100.00(10) K下共收集到17 681个反射点, 其中5 506个为独立可观测反射[I≧2σ(I)], R(int)=0.032 7, R(sigma)= 0.032 3。化合物1属正交晶系, 空间群为P212121, 胞晶参数: a=6.623 46(11) Å , b=14.554 1(2) Å , c=29.008 4(5) Å , α=90°, β=90°, γ=90°, V=2 796.36(8) Å 3, Z=4, Dcalcd=1.194 g·cm-3, μ(Cu Kα)=0.621 mm-1, F(000)=1104.0。通过解析其晶体结构, 用全矩阵最小二乘法修正结构, 最终的偏离因子为Rf=0.037 6, wR=0.095 9, 其最大与最小的电子密度峰的高度分别为0.29, -0.20 e/Å -3, flack参数为-0.10(8)。
化合物2~7为已知化合物, 根据其波谱数据和理化性质分别鉴定为3β-羟基齐墩果-11, 13(18)-二烯-28-酸(2)[10]、12-烯-齐墩果-28-酸-3β-棕榈酸酯(3)[13, 14]、3β-乙酰古柯二醇(4)[15]、科索酸(5)[16]、熊果醇(6)[17]和熊果酸(7)[18]
将对数生长期的RAW264.7细胞悬液调整为每毫升1×106个, 取每孔100 μL细胞悬液接种于96孔板内, 在37 ℃、5% CO2环境下培养24 h后, 首先, 以不同浓度的化合物1~7 (5~60 μmol·L-1)作用于RAW264.7细胞24 h, MTT法检测化合物对细胞活性的影响。其次, 不同浓度的化合物作用于LPS诱导细胞24 h后, 依NO试剂盒说明, 检测细胞释放NO的含量, 酶标仪检测540 nm下的吸光度值, 抑制作用按照下列公式计算: % Inhibition=[1-(Asample-Ablank)/(Amodel-Ablank)]×100, 其中槲皮素为阳性对照。
