The chemical constituents of Cydonia oblonga (Quince) seeds were investigated using chromatographic methods, including silica gel, Sephadex LH-20 and semi-preparative-HPLC. Eleven compounds were isolated and their structures were elucidated on 1D or 2D NMR, and HR-ESI-MS. These analyses have lead to identification of 5, 7-dihydroxy-2-n-pentacosanyl chromen-4-one (1), ursolic acid (2), tormentic acid (3), oleanolic acid (4), β-daucosterol (5), β-sitosterol (6), amygdalin (7), rutin (8), kaempferol (9), quercetin (10) and isoquercitrin (11). Among these, compound 1 is a new compound, and compounds 3, 5 were isolated from this plant for the first time. Compounds 1-4 and 8 showed significant PTP1B inhibitory activities with IC50 value of 0.465, 16.14, 28.96, 23.78 and 11.70 μmol·L-1, respectively.

采用硅胶、Sephadex LH-20及半制备液相等多种色谱方法从榅桲种子中分离得到11个化合物,利用UV、1D、2D NMR、HR-ESI-MS等波谱技术分别鉴定为5,7-二羟基-2-正二十五烷基色烯-4-酮(1)、乌苏酸(2)、委陵菜酸(3)、齐墩果酸(4)、β-胡萝卜苷(5)、β-谷甾醇(6)、苦杏仁苷(7)、芦丁(8)、山柰酚(9)、槲皮素(10)和异槲皮素(11)。其中化合物1为新化合物,35为首次从榅桲属中分离得到,其他成分均为首次从榅桲籽中分离得到。用体外PTP1B抑制活性实验,化合物1~48对PTP1B有抑制活性,其IC50值分别为0.465、16.14、28.96、23.78和11.70 μmol·L-1

The chemical constituents of Cydonia oblonga (Quince) seeds were investigated using chromatographic methods, including silica gel, Sephadex LH-20 and semi-preparative-HPLC. Eleven compounds were isolated and their structures were elucidated on 1D or 2D NMR, and HR-ESI-MS. These analyses have lead to identification of 5, 7-dihydroxy-2-n-pentacosanyl chromen-4-one (1), ursolic acid (2), tormentic acid (3), oleanolic acid (4), β-daucosterol (5), β-sitosterol (6), amygdalin (7), rutin (8), kaempferol (9), quercetin (10) and isoquercitrin (11). Among these, compound 1 is a new compound, and compounds 3, 5 were isolated from this plant for the first time. Compounds 1-4 and 8 showed significant PTP1B inhibitory activities with IC50 value of 0.465, 16.14, 28.96, 23.78 and 11.70 μmol·L-1, respectively.

榅桲籽为蔷薇科榅桲属(Cydonia oblonga)植物榅桲的种子。榅桲原产中亚西亚、高加索等地, 在我国分布于陕西、福建、新疆等地。榅桲籽在《全国中草药汇编》中记载: “主治伤署、呕吐、腹泻、消化不良、关节疼痛、腓肠肌痉挛”[1]。具有清除胃肠干热、生津止咳, 用于烦躁不安、咳嗽、肺结核等多种功效[2]。该药材在民间制成不同配方的糖浆和浸膏用于治疗消化器官疾病、糖尿病及心脑肝等器官疾病[3]。目前, 已从榅桲籽中发现酚酸类、黄酮类、三萜类、萜类、脂肪酸类化合物[4-6], 现代药理研究表明, 具有抗氧化[7]、抗溶血[8]、抗肾癌细胞增殖[9]、抗皮肤毒性[10]等作用。本课题组在前期研究工作中发现, 榅桲籽不同提取部位对蛋白酪氨酸磷酸酯酶1B (PTP1B)有不同程度的抑制活性[11]。为进一步寻找活性成分, 利用生物活性导向, 采用硅胶、Sephadex LH-20及半制备液相等多种色谱方法, 从榅桲种籽中分离得到11个化合物, 经UV、1D、2D NMR、HR-ESI-MS等波谱技术分别鉴定为: 5, 7-二羟基-2-正二十五烷基色烯-4-酮(1)、乌苏酸(2)、委陵菜酸(3)、齐墩果酸(4)、β-胡萝卜苷(5)、β-谷甾醇(6)、苦杏仁苷(7)、芦丁(8)、山柰酚(9)、槲皮素(10)和异槲皮素(11)。其中, 化合物1为新化合物, 35为首次从榅桲属植物中分离得到, 其他成分均为首次从榅桲籽中分离得到, 结构见图 1。化合物1~48展示出较好的PTP1B抑制活性, 其IC50值分别为0.465、16.14、28.96、23.78和11.70 μmol·L-1
化合物1  为白色粉末, mp 101~103 ℃, 溶于氯仿。10% H2SO4-乙醇溶液显淡黄色, FeC13反应呈阳性, 可能有酚羟基。HR-ESI-MS m/z: 527.808 1 (18) [M-H]- (计算值为527.808 3), 190.1 (100) [M-H-C24H49]-和203.1 (53)等其他碎片离子峰, 结合1H NMR和13C NMR数据推知分子式为C34H56O4, 不饱和度7, 含有苯环。1H NMR (600 MHz, C5D5N)图谱中:在低场出现2个羟基质子信号, 分别为δH 13.3 (5-OH)和δH 13.6 (7-OH); 3个芳香环质子δH 6.24 (1H, s, H-3)、δH 6.72 (1H, d, J = 1.6 Hz, H-8)和δH 6.75 (1H, d, J = 1.6 Hz, H-6), 后两个(H-8和H-6)显示为色酮环上两个间位偶合的氢; δH 2.48 (2H, t, J = 7.2 Hz, H-1'), 1.60 (2H, m, H-2')提示结构中有不连氧的亚甲基信号; δH 1.27~1.33 (44H, m, H-3'~H-24')提示结构中有长链亚甲基质子信号; δH 0.89 (3H, t, J = 7.2 Hz, H-25')提示结构中含有长链末端甲基信号; 13C NMR结合DEPT谱显示出34个碳信号: 1个伯碳[δC 14.7 (C-25')], 24个仲碳[δC 34.5 (C-1')、27.4 (C-2')、29.6~32.6 (C-3'~C-24')], 推测该化合物含有长链-CH2-(CH2)23-CH3基片段, 3个叔碳[δC 108.5 (C-3)、100.4 (C-6)、95.2 (C-8)]和6个季碳[δC 183.3 (C-4)、171.1 (C-2)、166.3 (C-9)、163.7 (C-5)、159.4 (C-7)、105.3 (C-10)]信号。在13C NMR (150 MHz, C5D5N)谱中, δC 183.3为羰基碳信号; δC 166.3、163.7、159.4为3个含氧芳碳信号; δC 108.5、105.3、100.4、95.2为4个芳香碳信号; 在δC 27.4~34.5区域内给出了侧链上的若干碳信号。利用HSQC、HMBC谱对化合物11H NMR、13C NMR谱数据进行了归属(表 1)。HSQC谱中, δH 6.24与δC 108.5, δH 6.75与δC 100.4, δH 6.72与δC 95.2, δH 2.48与δC 34.5, δH 1.60与δC 27.4, δH 1.27~1.33与δC 29.6~32.6, δH 0.89与δC 14.7相关。根据以上数据确定化合物1的母核为烷基取代的5, 7-二羟基色烯-4-酮。在HMBC谱谱中, δH2.48 (H-1')与δC 171.1 (C-2)相关外还与С-3、C-2'、С-(3'-24')有远程相关(图 2), 推测化合物结构中长链-CH2-(CH2)23-CH3基连接位置在δC 171.1 (C-2)碳上。因此, 确定化合物1结构式如图 2所示, 命名为5, 7-二羟基-2-正二十五烷基色烯-4-酮。
通过体外PTP1B活性检测实验, 测定化合物1~58的抑制PTP1B活性。结果, 化合物1~48表现出明显的抑制活性。
INOVA-400型核磁共振仪(美国VARIAN公司); Varian VNMRS-600 NMR核磁共振波谱仪(美国VARIAN公司); QSTAR Elite Hybrid LC/MS/MS质谱仪(美国惠氏公司); Shimadzu 2550型紫外可见光谱仪(日本岛津公司); 半制备高效色谱仪(DIONEX公司); X-BridgeTM半制备C18柱(5 μm, 10 mm×250 mm, 美国Waters公司); 薄层硅胶HSGF254 (山东烟台市芝罘黄务硅胶开发实验厂); 柱色谱硅胶(100~200目, 200~300目, 青岛海洋化工厂); Sephadex LH-20 (Pharmacia Biotech公司); Buchi B-540熔点测定仪(瑞士Buchi公司); ESJ-180A型电子天平(沈阳龙腾有限公司); KQ-100B型超声波清洗仪(江苏省昆山市超声仪器有限公司); ELISA酶标仪(美国); HPLC用甲醇、乙腈为色谱纯(美国Fisher公司); 水为哇哈哈纯净水; 提取分离用试剂(石油醚、乙酸乙酯、氯仿、二氯甲烷、丙酮、甲醇、乙醇等)均为分析纯。
榅桲籽为2015年11月采集于新疆阿图什市上阿图什乡, 经中国科学院新疆生态与地理研究所植物分类研究室沈观冕研究员鉴定为蔷薇科植物榅桲(Cydonia oblonga MILL.)的成熟种子。
榅桲种子7 kg粉碎后, 加石油醚浸泡3次, 每次24 h, 脱脂。将浸泡液减压浓缩, 得到榅桲籽石油醚部位(100 g)。脱脂后的榅桲籽用甲醇加热回流提取3次, 每次2 h。提取液减压浓缩至无醇味, 加水混悬, 依次用乙酸乙酯、正丁醇萃取减压浓缩得到各部分浸膏。其中取乙酸乙酯部分(80 g)进行硅胶柱色谱分离, 以石油醚-二氯甲烷-甲醇(V/V, 100:0:0, 90:10:0, 80:20:0, 70:30:0, 1:1:0, 30:70:0, 0:100:0, 0:95:5, 0:90:10, 0:80:20)梯度洗脱, 合并相同馏分, 依次得到10个馏分(Fr.1~10)。其中Fr.1 (8 g)经GC-GC/MS分离鉴定5个化合物。Fr.2 (17 g)经硅胶柱色谱(正己烷-乙酸乙酯) (V/V, 100:0, 90:10, 80:20, 70:30)梯度洗脱, TLC检测合并为4个馏分(Fr.2.1~2.4)。Fr.2.2 (1.5 g)用Sephadex LH-20等度洗脱(V/V, 氯仿-甲醇1:1)分离纯化得到化合物1 (12 mg)。Fr.8 (5.5 g)用Sephadex LH-20等度洗脱(V/V, 氯仿-甲醇1:1)分离纯化得到化合物2 (30 mg)和3 (130 mg)。Fr.9 (4.2 g)经硅胶柱色谱、Sephadex LH-20等度洗脱(V/V, 氯仿-甲醇1:1)分离纯化得到化合物4 (8.4 mg)、5 (80 mg)和6 (12mg)。
正丁醇部位(200 g), 其中取120 g加水溶解, 过滤, 上大孔吸附树脂AB-8, 依次用水、10%、30%、50%、70%、90%乙醇-水梯度洗脱得到6个洗脱组分。其中50%和70%乙醇-水洗脱组分经TLC检测, 组分成分基本一致, 合并(33 g), 经硅胶柱色谱, 以氯仿-甲醇(V/V, 50:1, 40:1, 30:1, 10:1, 1:1)梯度洗脱, 分离得到5个组分(Fr.1~Fr.5)。Fr.4组分经过Sephadex LH-20柱色谱(甲醇洗脱)得到化合物7 (100 mg), Fr.5通过Sephadex LH-20柱色谱(甲醇), 半制备高效液相色谱(甲醇-水, 8:2)纯化, 得到化合物8 (12 mg)。
取正丁醇部位80 g采用硅胶柱色谱, 以氯仿-丙酮(V/V, 80:1, 50:1, 30:1, 10:1, 5:1, 1:1)、氯仿-甲醇(5:1)梯度洗脱, TLC检测将相同组分合并得到7个组分(Fr.1~Fr.7)。Fr.2组分(7 g)经过硅胶柱色谱(氯仿-丙酮5:1), Sephadex LH-20 (甲醇)洗脱分离, 半制备高效液相(甲醇-水8:2)色谱纯化, 得到化合物9 (15 mg)和化合物10 (9 mg)。Fr.5组分(4 g)经硅胶柱(氯仿-甲醇1:1), Sephadex LH-20柱色谱(甲醇洗脱)分离纯化, 得化合物8 (24 mg)和化合物11 (5 mg)。
化合物1   白色粉末(氯仿), mp 101~103 ℃, 10% H2SO4乙醇溶液显色为淡黄色; ESI-MS m/z: 527.8 [M-H]-, 190.1 [M-H-C24H49]- (100), 203.1 (53), 217.3 (6), 231.0 (8), 246.4 (5), 259.1 (16), 273.0 (10), 287.2 (9), 303.4 (5), 315.2 (9), 329.1 (5), 343.0 (6), 357.1 (8), 371.2 (8), 399.4 (3), 413.0 (3), 441.1 (3), 427.3 (6), 455.2 (3), 469.2 (7), 497.4 (5); HR-ESI-MS m/z: 527.808 1 [M-H]- (计算值C34H55O4 527.808 3), 确定分子式为C34H56O4, 不饱和度为4。NMR数据见表 1
以对-硝基苯基磷酸二钠(pNPP)作为底物, 根据PTP1B水解pNPP的磷酸基团而产生颜色反应来测定PTP1B的活性[12, 13]。将化合物1~58和阳性对照品钒酸钠(NaVO3)用DMSO溶解, 取1 μL加入至200 μL反应体系中(pH 6.0, 1 mmol·L-1 EDTA, 0.1 mol·L-1 NaCl, 3 mmol·L-1 DDT, 50 mmol·L-1柠檬酸), 加入底物pNPP至2 mmol·L-1, 加入人重组PTPlB酶0.3 µg开始反应, 37 ℃保温30 min, 加入5 mmol·L-1 NaOH 20 µL终结反应, 用酶标仪在405 nm测定吸收度, 以原钒酸钠作为阳性对照, 以不加PTP1B为空白对照, 计算抑制率, 应用Origin软件计算IC50值。抑制率=(A405空白-A405样品)/A405空白×100%。
结果显示, 化合物1~48表现出明显的PTP1B抑制活性, IC50分别为0.465、16.14、28.96、23.78和11.70 μmol·L-1
本研究以PTP1B生物活性为导向, 从榅桲籽中分离得到11个化合物, 5个化合物具有抑制PTP1B的活性。抑制PTP1B活性是防治糖尿病及肥胖等代谢性疾病的有效途径[14]。因此, 本实验为进一步研究或能够找到潜在的用于治疗或改善2型糖尿病和肥胖症的新的PTP1B抑制剂提供科学依据。
