Objective:

To establish HPLC fingerprint and determine caffeic acid,isoquercitrin,quercitrin,rosmarinic acid,lithospermic acid and salvianolic acid B in Tournefortia sibirica Linnaeus,and to provide evidence for quality control of Tournefortia sibirica Linnaeus.

Methods:

The chromatographic separation was performed on an Agilent Eclipse Plus C18 column (250 mm×4.6 mm,5 μm) with gradient elution (0-60 min,9%B→38%B) of 0.2% phosphoric acid(A) and acetonitrile(B) . The detection wavelength was 330 nm. The column temperature was kept at 40 ℃ and the flow rate was 1.0 mL·min-1. The HPLC fingerprint of all batches of Tournefortia sibirica Linnaeus was established using Similarity Evaluation Software for Chromatographic Fingerprint of Traditional Chinese Medicine (2012 edition) and the common peaks were identified by reference standards. Six constituents in Tournefortia sibirica Linnaeus were quantified.

Results:

Eleven common peaks were confirmed,and 6 common peaks were identified by reference standards including caffeic acid,isoquercetin,quercetin,rosmarinic acid,purple oxalic acid,and salvianolic acid B. The similarities of 17 batches samples were 0.931 to 0.996. By the methodology examination,RSDs for the precision test were 1.9%,1.0%,1.4%,0.19%,1.1% and 0.32%,respectively. RSDs for the reproducible test were 3.5%,2.3%,3.3%,0.14%,1.1% and 0.19%,respectively. RSDs for the stability test were 0.80%,1.1%,1.7%,0.52%,0.54% and 0.78%,respectively. Caffeic acid,isoquercitrin,quercitrin,rosmarinic acid,lithospermic acid and salvianolic acid B had good separation and showed good linearity in their respective linear ranges. The average recoveries ranged from 95% to 105%,and the contents (calculated with reference to the dried drug) were between 0.004% to 0.013%,0.030% to 0.259%,0.032% to 0.256%,0.256% to 1.246%,0.018% to 0.072% and 0.062% 0.499%,respectively.

Conclusion:

The established HPLC fingerprint and quantification method is stable and reliable,which can provide basis for the quality control of Tournefortia sibirica Linnaeus.

目的:

建立不同产地蒙药材砂引草Tournefortia sibirica Linnaeus的HPLC指纹图谱并测定其6个成分(咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B)的含量,为砂引草质量控制提供依据。

方法:

采用Agilent Eclipse Plus C18色谱柱(250 mm×4.6 mm,5 μm),以0.2%磷酸溶液(A)-乙腈(B)为流动相,进行梯度洗脱(0~60 min,9%B→38%B),检测波长330 nm。对不同产地砂引草进行分析,采用国家药典委员会“中药色谱指纹图谱相似度评价系统软件”(2012版),建立砂引草指纹图谱,同时测定6个成分含量。

结果:

HPLC指纹图谱共标定了不同产地砂引草中11个共有峰,指认了咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B 6个成分,17批砂引草药材与对照指纹图谱之间的相似度在0.931~0.996。经方法学考察,咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸6个成分精密度试验RSD依次为1.9%、1.0%、1.4%、0.19%、1.1%和0.32%;重复性试验RSD依次为3.5%、2.3%、3.3%、0.14%、1.1%和0.19%;稳定性试验RSD依次为0.80%、1.1%、1.7%、0.52%、0.54%和0.78%。咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B在0.003 3~0.029 5、0.011 9~0.107 3、0.014 8~0.132 8、0.092 8~0.835 2、0.068 9~0.105 8、0.068 8~0.619 9 μg·mL-1浓度范围内线性关系良好(均为r=1.000);平均回收率在93.9%~102.3%,按干燥品计含量在0.004%~0.013%、0.030%~0.259%、0.032%~0.256%、0.256%~1.246%、0.018%~0.072%和0.062%~0.499%。

结论:

建立的砂引草HPLC指纹图谱及6个成分含量测定方法稳定、可靠、重复性好,可为砂引草质量控制及开发研究提供参考。

目的:

建立不同产地蒙药材砂引草Tournefortia sibirica Linnaeus的HPLC指纹图谱并测定其6个成分(咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B)的含量,为砂引草质量控制提供依据。

方法:

采用Agilent Eclipse Plus C18色谱柱(250 mm×4.6 mm,5 μm),以0.2%磷酸溶液(A)-乙腈(B)为流动相,进行梯度洗脱(0~60 min,9%B→38%B),检测波长330 nm。对不同产地砂引草进行分析,采用国家药典委员会“中药色谱指纹图谱相似度评价系统软件”(2012版),建立砂引草指纹图谱,同时测定6个成分含量。

结果:

HPLC指纹图谱共标定了不同产地砂引草中11个共有峰,指认了咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B 6个成分,17批砂引草药材与对照指纹图谱之间的相似度在0.931~0.996。经方法学考察,咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸6个成分精密度试验RSD依次为1.9%、1.0%、1.4%、0.19%、1.1%和0.32%;重复性试验RSD依次为3.5%、2.3%、3.3%、0.14%、1.1%和0.19%;稳定性试验RSD依次为0.80%、1.1%、1.7%、0.52%、0.54%和0.78%。咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B在0.003 3~0.029 5、0.011 9~0.107 3、0.014 8~0.132 8、0.092 8~0.835 2、0.068 9~0.105 8、0.068 8~0.619 9 μg·mL-1浓度范围内线性关系良好(均为r=1.000);平均回收率在93.9%~102.3%,按干燥品计含量在0.004%~0.013%、0.030%~0.259%、0.032%~0.256%、0.256%~1.246%、0.018%~0.072%和0.062%~0.499%。

结论:

建立的砂引草HPLC指纹图谱及6个成分含量测定方法稳定、可靠、重复性好,可为砂引草质量控制及开发研究提供参考。

The chromatographic separation was performed on an Agilent Eclipse Plus C18 column (250 mm×4.6 mm,5 μm) with gradient elution (0-60 min,9%B→38%B) of 0.2% phosphoric acid(A) and acetonitrile(B) . The detection wavelength was 330 nm. The column temperature was kept at 40 ℃ and the flow rate was 1.0 mL·min-1. The HPLC fingerprint of all batches of Tournefortia sibirica Linnaeus was established using Similarity Evaluation Software for Chromatographic Fingerprint of Traditional Chinese Medicine (2012 edition) and the common peaks were identified by reference standards. Six constituents in Tournefortia sibirica Linnaeus were quantified.

Results:

Eleven common peaks were confirmed,and 6 common peaks were identified by reference standards including caffeic acid,isoquercetin,quercetin,rosmarinic acid,purple oxalic acid,and salvianolic acid B. The similarities of 17 batches samples were 0.931 to 0.996. By the methodology examination,RSDs for the precision test were 1.9%,1.0%,1.4%,0.19%,1.1% and 0.32%,respectively. RSDs for the reproducible test were 3.5%,2.3%,3.3%,0.14%,1.1% and 0.19%,respectively. RSDs for the stability test were 0.80%,1.1%,1.7%,0.52%,0.54% and 0.78%,respectively. Caffeic acid,isoquercitrin,quercitrin,rosmarinic acid,lithospermic acid and salvianolic acid B had good separation and showed good linearity in their respective linear ranges. The average recoveries ranged from 95% to 105%,and the contents (calculated with reference to the dried drug) were between 0.004% to 0.013%,0.030% to 0.259%,0.032% to 0.256%,0.256% to 1.246%,0.018% to 0.072% and 0.062% 0.499%,respectively.

Conclusion:

The established HPLC fingerprint and quantification method is stable and reliable,which can provide basis for the quality control of Tournefortia sibirica Linnaeus.

砂引草为紫草科砂引草属砂引草Tournefortia sibirica Linnaeus的全草,蒙古文名浩吉格热额布苏、额日古勒根其其格,异名紫丹草、挠挠糖,其味苦,性寒,具有清热解毒、排脓、敛疮、杀粘等功效,主治瘰疬、疮疡溃破、久不收口、皮肤湿疹,收载于《内蒙古植物药志》[1]、《锡林郭勒盟草原药物诠释典》[2]。查阅相关文献可知:张欣等[3]对砂引草多糖的提取工艺进行优化;Morteza-Semnani等[4]利用GC-MS技术分析鉴定了砂引草挥发油中23个成分;高超等[5]又通过GC-MS技术从砂引草挥发油中鉴定出58个化合物;阿如汗等[6]应用LC-MS技术从砂引草中鉴定出78个化合物;Hikichi等[7]从砂引草中分离得到Lycopsamine,O-9-Angelylretronecine和Pyrrolizidine;宋志忠等[8-9]等先后从砂引草中分离得到2个木质素化合物(messerschmidin,messerschmidin)、2个木脂素衍生物(sibiricin A,sibiricin B)和1个三萜类化合物α-香脂醇二十烷酸酯;吴永江等[10]分离得到4’,5,7-三羟基黄酮醇-3-O-葡萄糖苷和3’,4’,5,7-四羟基黄酮醇-3-O-葡萄糖苷;红艳等[11]分离得到1个新生物碱成分tournesibirin;刁胜宝等[12]分离到1个新黄烷酮苷类成分(2S)-dihydrooroxylin A 7-O-[b-D-apiosyl(1fi2)]-b-D-glucoside和4个已知化合物(2”-O-乙酰基-7-O-甲基维他命、5-羟基-7-甲氧基-8-C-β-葡萄糖基黄酮、菠菜素3-O-β-吡喃葡萄糖苷和9’-甲氧基脱氢二癸二烯醇4-O-β-D-吡喃葡萄糖苷)。
以上文献表明,砂引草主要集中在化学成分研究,未发现其质量控制研究,这给本次研究提供了空间。为保证砂引草的临床用药安全有效,本研究在已报道文献基础上,结合超高效液相色谱-高分辨质谱联用(UPLC-Q TOF/MS)技术,通过分析砂引草化合物的保留时间、多级质谱信息,结合天然产物高分辨质谱数据库及相关文献[13-15],从砂引草中指认9个指定色谱峰,采用HPLC建立砂引草指纹图谱,同时测定咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B 6个成分的含量,为砂引草深入提供基础研究。
LC-20高效液相色谱仪(岛津公司);Agilent Q-TOF 6545 LC/MS高分辨串联质谱(安捷伦科技有限公司);Agilent Eclipse Plus C18色谱柱(250 mm×4.6 mm,5 μm);SQP型电子天平(赛多利斯科学仪器(北京)有限公司,0.01 mg);KQ-200VDE型三频数控超声波清洗器(昆山市超声仪器有限公司);乙腈(色谱纯,天津市大茂化学试剂厂);乙腈(质谱纯,I0928729804,Merck公司);甲酸(质谱纯,U9260048,CNW公司);水为超纯水;其他试剂均为分析纯。
对照品:咖啡酸(批号110885-201703,含量99.7%)、异槲皮苷(批号111809-201804,含量97.2%)、槲皮苷(批号111538-202007,含量93.5%)、迷迭香酸(批号111871-202007,含量98.1%)、丹酚酸B(批号111562-201917,含量96.6%),均购自中国食品药品检定研究院;紫草酸(批号10406,含量99.4%,上海诗丹德生物技术有限公司)。
17批砂引草(见表1)均采自内蒙古,由内蒙古民族大学蒙医药学院红艳副教授鉴定为砂引草Tournefortia sibirica Linnaeus的全草。
采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为0.2%磷酸溶液(A)-乙腈(B),梯度洗脱(0~60 min,9%B→38%B),检测波长330 nm,流速1.0 mL·min-1,柱温40 ℃,进样量10 μL。
取本品粉末(过3号筛)约0.2 g,精密称定,置具塞锥形瓶中,精密加入甲醇25 mL,密塞,称量,超声(功率250 W,频率40 kHz)处理0.5 h,放冷,再称量,用甲醇补足减失的量,摇匀,滤过,即得。
取咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B的对照品适量,精密称定,加甲醇制成每1 mL含咖啡酸0.001 6 mg、异槲皮苷0.006 0 mg、槲皮苷0.007 4 mg、迷迭香酸0.046 0 mg、紫草酸0.005 9 mg和丹酚酸B 0.034 4 mg的混合溶液,即得。
精密称定S1号样品,按“2.2.1”项下方法制备供试品溶液,按“2.1”项下色谱条件连续进样6次,以6号色谱峰(迷迭香酸峰)为参照峰,计算指纹图谱中各共有峰相对保留时间及相对峰面积,结果各共有峰相对保留时间的RSD在0.050%~0.74%,共有峰相对峰面积的RSD在1.2%~2.7%,表明仪器精密度良好。
精密称定S1号样品,按“2.2.1”项方法制备供试品溶液,按“2.1”项色谱条件,分别在0、2、4、6、8、12 h进样测定,以6号色谱峰(迷迭香酸峰)为参照峰,计算指纹图谱中各共有峰相对保留时间及相对峰面积,结果各共有峰相对保留时间的RSD在0.45%~0.73%,共有峰相对峰面积的RSD在1.5%~2.2%,说明供试品溶液在12 h内稳定。
精密称定S1号样品,按“2.2.1”项方法平行制备6份供试品溶液,按“2.1”项色谱条件进样测定,以6号色谱峰(迷迭香酸峰)为参照峰,计算指纹图谱中各共有峰相对保留时间及相对峰面积,结果各共有峰相对保留时间的RSD在0.59%~1.2%,共有峰相对峰面积的RSD在1.8%~2.9%,表明所用方法重复性良好。
采用UPLC-Q TOF/MS技术,通过分析砂引草化合物的保留时间、多级质谱信息,并结合天然产物高分辨质谱数据库及相关文献,从砂引草中指认9个指定色谱峰,除8号峰未推断出具体结构外,其余8个色谱峰鉴定结果分别为:1号峰Schizotenuin A,2号峰异槲皮苷,3号峰槲皮素3-O-丙二酰基葡萄糖苷,4号峰迷迭香酸,5号峰紫草酸,6号峰为丹酚酸H,7号峰丹酚酸B,9号峰迷迭香酸甲酯异构体(见表2图1)。取“2.2.1”项下供试品溶液(S1号样品)和“2.2.2”项下混合对照品溶液,按“2.1”项色谱条件进样,通过保留时间和UV紫外图谱对照分析,确定1号峰为咖啡酸,3号峰为异槲皮苷,5号峰为斛皮苷,6号峰为迷迭香酸,7号峰为紫草酸,10号峰为丹酚酸B,砂引草供试品溶液及混合对照品的HPLC色谱图见图2
取17批(S1~S17)不同产地砂引草,按“2.2.1”项下方法制备供试品溶液,按“2.1”项色谱条件进样并记录色谱数据,得17批次砂引草HPLC色谱图(图3)。将其导入国家药典委员会中药色谱指纹图谱相似度评价系统软件(2012.130723版),以样品(S1)图谱为参照指纹图谱,中位数法生成对照指纹图谱,多点校正得到砂引草样品的指纹图谱与对照图谱并进行相似度分析,共得到11个共有峰,6号色谱峰有较好的峰形,且峰面积大,故以6号峰为参照峰。结果显示17批砂引草药材与对照指纹图谱之间的相似度在0.931~0.996。说明17批次砂引草药材总体相似度较高,药材质量稳定。
以17批砂引草样品的11个共有峰峰面积为变量,通过SPSS 27.0软件,运用组间对比法进行系统聚类分析(CA),以平方欧氏距离为标尺计算样品的相似程度,连接方法为组间连接,17批药材的聚类分析树状图见图4。可看出,不同地区的砂引草可以被较好地分开。当距离为5时,样品可被划分为4类:第Ⅰ类为S2、S4、S5、S7~S9、S11、S13;第Ⅱ类为S6、S10、S14、S16;第Ⅲ类为S1、S3、S12、S15;第Ⅳ类为S17。当距离为20时,样品可被划分为2类:第Ⅰ类为S1~S16;第Ⅱ类为S17。
为更深入地探究不同批次间蒙药砂引草的质量差异,将17批药材样品中11个共有峰面积导入SPSS 27.0软件,进行主成分分析(PCA),结果以特征值>1为筛选条件,提取出4个主成分,且4个主成分特征根分别为5.199、2.068、1.276、1.038,方差贡献率分别为47.265、18.801、11.603、9.432,累计贡献率达到87.1%,表明这4类主成分包含了全部测量指标所含有的主要信息,达到了降维的目的。PCA碎石图(图5)显示,率先提取出来的4个成分的坡度较为陡峭,而后续形成的坡度较为平缓,说明这4个主成分可能就是鉴别砂引草最主要的色谱峰。
为进一步分析样品之间的相似性,用SIMCA 14.1软件绘制17批样品的主成分得分图,见图6。以11个共有峰峰面积为变量,可以直观看出各产地样品特征分布情况,17批样品分布比较集中,与CA结果大致相同。
采用SIMCA 14.1软件对17批样品进行正交偏最小二乘法-判别分析(OPLS-DA),得分矩阵图见图7。该模型R2X(cum)=0.315,R2Y (cum)=0.203,Q2=-0.616,该模型各数值均小于0.5,说明模型不是很稳定可靠,不适合作为砂引草指纹图谱的模式识别方法。通过变量重要性投影值(VIP)筛选能引起砂引草菌核成分差异的主要标记性成分,其中,拥有较大VIP值的变量,对分类的贡献越大。以VIP>1为标准,从图8可以看出,大于1者有5个,分别为峰3、峰6、峰2、峰8、峰7。通过“2.3.4”项下的指认,峰3为异槲皮苷,峰6为迷迭香酸,峰7为紫草酸。综上所述,结合OPLS-DA中VIP值筛选、对照品指认及相关药效学文献,说明异槲皮苷、迷迭香酸以及紫草酸是砂引草的差异标志物。
分别精密吸取“2.2.2”项混合对照品溶液2.0、5.0、10.0、15.0、18.0 μL,注入液相色谱仪,测定其峰面积值。以混合对照品溶液的进样量X为横坐标,峰面积积分值Y为纵坐标,进行线性分析,结果见表3。砂引草中6个化学成分在相应的质量浓度范围与峰面积线性关系良好。
精密吸取S1号样品的供试品溶液各10 μL,分别连续注入液相色谱仪6次,测定峰面积,并统计分析。结果咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B峰面积的RSD依次为1.9%、1.0%、1.4%、0.19%、1.1%和0.32%。表明该仪器精密度良好。
取S1号样品,按”2.2.1”项下方法平行制备6份供试品溶液,按“2.1”项下色谱条件进行HPLC分析,进样体积10 μL,计算含量,测得咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B的平均含量分别为0.012%、0.045%、0.033%、0.77%、0.079%和0.46%,RSD分别为3.5%、2.3%、3.3%、0.14%、1.1%和0.19%。
取S1号样品,按“2.2.1”项下方法制备供试品溶液,按“2.1”项下色谱条件,分别在0、2、4、8、10、12 h进样10 μL测定,结果咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B峰面积的RSD分别为0.80%、1.1%、1.7%、0.52%、0.54%和0.78%。表明供试品溶液在12 h内稳定。
取已知含量的S1号样品约0.1 g,共9份,精密称定,分为3组,按0.5、1、1.5的比例分别依次精密加入咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B对照品,制备供试溶液,精密吸取10 μL进样测定,计算咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B的回收率,结果见表4
取砂引草样品各约0.2 g,精密称定,按“2.2.1”项下方法制备供试品溶液,精密吸取10 μL,分别注入液相色谱仪,测定,并计算含量。结果17批次砂引草中咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B含量(以干燥品计)分别在0.004%~0.013%、0.030%~0.259%、0.032%~0.256%、0.256%~1.246%、0.018%~0.072%和0.062%~0.499%。见表5
本研究制定了蒙药材砂引草的性状、鉴别(显微鉴别和薄层色谱鉴别)、检查(水分、总灰分、酸不溶性灰分)、浸出物(醇溶性、水溶性)、指纹图谱和含量测定等内容。
本品茎圆柱形,单一或数条丛生,直立或斜升,通常分枝,长10~30 cm;表面黄绿色、黄棕色或灰棕色,密生糙伏毛或白色长柔毛;体轻,质脆,断面类白色或淡黄色。叶多脱落并破碎,展平后披针形、倒披针形或长圆形,长1~5 cm,宽6~10 mm,先端渐尖或钝,基部楔形或圆,密生糙伏毛或长柔毛,无柄或近无柄。花序顶生。气微,味淡。
本品粉末绿色。非腺毛单细胞,长可达1 000 μm,壁厚,表面有微细疣状突起。腺毛头部圆形或类圆形,柄具1~5个细胞。草酸钙簇晶直径约10 μm,单个散在或成行排列于纤维成晶纤维。花粉粒类圆形,直径约30 μm。导管主为环纹导管或螺纹导管,直径约10~30 μm。气孔多为不定式。
取本品2 g,加二氯甲烷30 mL,超声(功率250 W,频率40 kHz)处理30 min,过滤,滤液浓缩至1 mL,作为供试品溶液,滤渣备用。另取β-谷甾醇对照品,加二氯甲烷制成每1 mL含0.5 mg的溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,吸取上述2种溶液各10 μL,分别点于同一硅胶G薄层板上,以石油醚-丙酮(7:1)为展开剂,展开,取出,晾干,喷以10%硫酸乙醇溶液,加热至斑点显色清晰。供试品色谱中,在与对照品色谱相应的位置上,显相同颜色的斑点;置紫外光灯(365 nm)下检视显相同颜色的荧光斑点。
取鉴别(2)项下的滤渣,加甲醇30 mL超声(功率250 W,频率40 kHz)处理30 min,过滤,滤液浓缩至1 mL,作为供试品溶液。另取迷迭香酸对照品,加甲醇制成每1 mL含0.5 mg的溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,吸取上述2种溶液各3 μL,分别点于同一硅胶G薄层板上,以环己烷-乙酸乙酯-甲酸(8:10:1)为展开剂,展开,取出,晾干,在紫外光灯(365 nm)下检视。供试品色谱中在与对照品色谱相应的位置上,显相同颜色的荧光斑点。
参照2020年版《中华人民共和国药典》四部对17批样品进行检查和浸出物项考察测定,结果水分在7.3%~10.8%;总灰分在6.1%~9.9%;酸不溶性灰分在0.65%~1.9%;醇溶性浸出物在9.8%~14.7%;水溶性浸出物在24.2%~34.9%。
对供试品溶液进行200~400 nm的波长扫描,结果显示波长在330 nm时有较大吸收,且分离度好,故本研究检测波长选择为330 nm;分别考察了甲醇-水、乙腈-水、甲醇-0.2%磷酸水溶液、乙腈-0.2%磷酸水溶液为流动相的不同比率梯度洗脱,结果流动相为乙腈-0.2%磷酸水溶液为流动相时分离度较好,故选择了乙腈-0.2%磷酸水溶液为流动相;对选定的流动相以(1.0±0.2)mL·min-1的不同流速进行了考察试验,结果咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B色谱峰的保留时间,随流速提高依次有所前移,且不影响分离度与峰面积值,故本试验研究采用流动相流速为1.0 mL·min-1;对色谱柱温度的控制,以(30±10)℃的不同温度进行了考察试验,结果咖啡酸、异槲皮苷、槲皮苷、迷迭香酸、紫草酸和丹酚酸B色谱峰的保留时间,随柱温提高渐有前移,且不影响分离度与峰面积值,故本试验研究采用了柱温为40 ℃。
本研究考察了不同溶剂(甲醇、70%甲醇、50%甲醇)、不同提取方法(超声提取、回流提取)、不同提取时间(30、45、60 min),结果不同溶剂、提取方法及提取时间的HPLC色谱峰数、分离度以及主峰的峰面积差别不大,考虑到甲醇提取的杂质少,超声提取简便,30 min时节能,故本研究提取方法为取本品粉末(过3号筛)约0.2 g,精密称定,置具塞锥形瓶中,精密加入甲醇25 mL,密塞,称量,超声(250 W,40 kHz)处理0.5 h,放冷,再称量,用甲醇补足减失的量,摇匀,滤过,即得。
