Objective:

To establish an HPLC-MS/MS method for simultaneous determination of 11 components(harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ)in Gengnianning.

Methods:

HPLC assay was performed on C18 column(100 mm×2.1 mm,1.9 μm)with a mixture of methanol and 0.1% formic acid as the mobile phase in gradient elution at a flow rate of 0.3 mL·min-1. The column temperature was 25 ℃ and the injection volume was 1 μL. Detection was carried out on a triple quadrupole mass spectrometer in positive ion mode(harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone and harpagoside)and negative ion mode(vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ)using an electrospray ion source(ESI). Multiple reaction monitoring(MRM)mode was employed.

Results:

The calibration curves were linear within the ranges of 1.485-29.71 μg·mL-1,1.620-32.40 μg·mL-1,7.801-156.0 μg·mL-1,0.518-10.35 μg·mL-1,0.167-3.333 μg·mL-1,0.359-7.179 μg·mL-1,1.455-29.10 μg·mL-1,1.520-30.40 μg·mL-1,0.160-3.205 μg·mL-1,0.143-2.864 μg·mL-1 and 0.157-3.136 μg·mL-1 for harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ,respectively. All 11 components showed good linearity(r≥0.998 0). The average recoveries(n=6)were in the range of 95.9%-102.6% with RSDs within 0.90%-3.0%. The contents of harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ and atractylenolide Ⅰ in 10 tested samples from 5 manufactures were 14.8-104.5,37.6-288.5,335.8-1 332.8,6.2-10.1,6.6-61.8,13.7-75.1,57.4-132.8,16.9-70.6,11.8-33.9,3.4-15.4 and 6.5-12.9 μg·g-1.

Conclusion:

The developed method is accurate and sensitive. It can be used in quality control of Gengnianning.

目的:

方法:

采用C18色谱柱(100 mm×2.1 mm,1.9 μm),以甲醇-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.3 mL·min-1,柱温为25 ℃,进样量为1 μL;质谱采用电喷雾离子源(ESI),电离模式为ESI-(哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷)和ESI+(王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ),多反应监测(MRM)模式进行定量分析。

11个成分的线性范围分别为1.485~29.71、1.620~32.40、7.801~156.0、0.518~10.35、0.167~3.333、0.359~7.179、1.455~29.10、1.520~30.40、0.160~3.205、0.143~2.864、0.157~3.136 μg·mL-1r均≥0.998 0;平均回收率(n=6)在95.9%~102.6%,RSD在0.90%~3.0%。5个企业生产的10批更年宁样品中上述11个成分的含量分别为14.8~104.5、37.6~288.5、335.8~1 332.8、6.2~10.1、6.6~61.8、13.7~75.1、57.4~132.8、16.9~70.6、11.8~33.9、3.4~15.4、6.5~12.9 μg·g-1

结论:

本方法灵敏、准确,可用于更年宁的质量控制。

目的:

建立HPLC-MS/MS法同时测定更年宁中哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ共11个成分的含量。

方法:

采用C18色谱柱(100 mm×2.1 mm,1.9 μm),以甲醇-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.3 mL·min-1,柱温为25 ℃,进样量为1 μL;质谱采用电喷雾离子源(ESI),电离模式为ESI-(哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷)和ESI+(王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ),多反应监测(MRM)模式进行定量分析。

结果:

11个成分的线性范围分别为1.485~29.71、1.620~32.40、7.801~156.0、0.518~10.35、0.167~3.333、0.359~7.179、1.455~29.10、1.520~30.40、0.160~3.205、0.143~2.864、0.157~3.136 μg·mL-1r均≥0.998 0;平均回收率(n=6)在95.9%~102.6%,RSD在0.90%~3.0%。5个企业生产的10批更年宁样品中上述11个成分的含量分别为14.8~104.5、37.6~288.5、335.8~1 332.8、6.2~10.1、6.6~61.8、13.7~75.1、57.4~132.8、16.9~70.6、11.8~33.9、3.4~15.4、6.5~12.9 μg·g-1

结论:

本方法灵敏、准确,可用于更年宁的质量控制。

Objective:

To establish an HPLC-MS/MS method for simultaneous determination of 11 components(harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ)in Gengnianning.

Methods:

HPLC assay was performed on C18 column(100 mm×2.1 mm,1.9 μm)with a mixture of methanol and 0.1% formic acid as the mobile phase in gradient elution at a flow rate of 0.3 mL·min-1. The column temperature was 25 ℃ and the injection volume was 1 μL. Detection was carried out on a triple quadrupole mass spectrometer in positive ion mode(harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone and harpagoside)and negative ion mode(vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ)using an electrospray ion source(ESI). Multiple reaction monitoring(MRM)mode was employed.

Results:

The calibration curves were linear within the ranges of 1.485-29.71 μg·mL-1,1.620-32.40 μg·mL-1,7.801-156.0 μg·mL-1,0.518-10.35 μg·mL-1,0.167-3.333 μg·mL-1,0.359-7.179 μg·mL-1,1.455-29.10 μg·mL-1,1.520-30.40 μg·mL-1,0.160-3.205 μg·mL-1,0.143-2.864 μg·mL-1 and 0.157-3.136 μg·mL-1 for harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ,and atractylenolide Ⅰ,respectively. All 11 components showed good linearity(r≥0.998 0). The average recoveries(n=6)were in the range of 95.9%-102.6% with RSDs within 0.90%-3.0%. The contents of harpagide,salidroside,nuezhenide,lobetyolin,wedelolactone,harpagoside,vaccarin,6-gingerol,atractylenolide Ⅲ,atractylenolide Ⅱ and atractylenolide Ⅰ in 10 tested samples from 5 manufactures were 14.8-104.5,37.6-288.5,335.8-1 332.8,6.2-10.1,6.6-61.8,13.7-75.1,57.4-132.8,16.9-70.6,11.8-33.9,3.4-15.4 and 6.5-12.9 μg·g-1.

Conclusion:

The developed method is accurate and sensitive. It can be used in quality control of Gengnianning.

更年宁是由柴胡、黄芩、白芍、墨旱莲、人参、党参、郁金、醋香附、当归、薄荷、川芎、玄参、茯苓、法半夏、石菖蒲、牡丹皮、陈皮、干姜、麸炒白术、丹参、炒王不留行、酒女贞子22味药材制成的中药制剂,具有疏肝解郁、益气养血、健脾安神的功效,用于更年期引起的心悸气短、烦躁易怒、眩晕失眠、阵热汗出、胸乳胀痛、月经紊乱等症[1]。更年宁现行质量标准收载于《卫生部药品标准》中药成方制剂第六册,仅有显微鉴别项和薄层鉴别项,未对药味进行含量测定控制。目前,更年宁的含量测定相关研究较少,仅见芍药苷、阿魏酸、黄芩苷和丹皮酚的含量测定报道[2-4]
本实验在前期指纹图谱研究、HPLC法含量测定和GC-MS法含量测定的基础上,选择更年宁中玄参、酒女贞子、党参、墨旱莲、炒王不留行、干姜、麸炒白术中的指标成分哈巴苷、哈巴俄苷[5-6]、红景天苷、特女贞苷[7-8]、党参炔苷[9]、蟛蜞菊内酯[10]、王不留行黄酮苷[11]、6-姜辣素[12]、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ [13-14],采用HPLC-MS/MS技术建立含量测定方法,以期为全面、客观评价更年宁质量提供数据支持。
TSQ Altis三重四极杆液相色谱-质谱联用仪,Thermo Fisher公司;Mettler XPE205十万分之一电子天平,Mettler Toledo公司;KQ-300DA型数控超声波清洗仪(超声功率:300 W;频率:40 kHz),昆山市超声仪器有限公司。
分析纯甲醇(国药集团化学试剂有限公司);质谱纯甲醇(Merck公司);质谱纯甲酸(Fisher公司);实验用水为Milli-Q系统制备。
对照品哈巴苷(批号111729-201707,含量以96.8%计)、红景天苷(批号110818-202009,含量以98.6%计)、特女贞苷(批号111926-201906,含量以95.0%计)、党参炔苷(批号111732-201908)、蟛蜞菊内酯(批号111885-202105,含量以97.2%计)、哈巴俄苷(批号111730-202110,含量以96.8%计)、王不留行黄酮苷(批号111853-201303,含量以92.6%计)、6-姜辣素(批号111833-202007,含量以99.3%计)、白术内酯Ⅲ(批号111978-201501,含量以99.9%计)、白术内酯Ⅱ(批号111976-201501,含量以99.9%计)、白术内酯Ⅰ(批号111975-201501,含量以99.9%计)均来自中国食品药品检定研究院。10批更年宁样品均为2022年国家药品抽检样品,来自5个企业(相关信息:A企业,批号21030536、21033791;B企业,批号210602、210201;C企业,批号200610、210802;D企业,批号200703、201202;E企业,批号191001、191002)。
采用Thermo Fisher Hypersil GOLD VANQUISH C18色谱柱(100 mm×2.1 mm,1.9 μm),以甲醇(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱(0~5 min,20%A;5~25 min,20%A→60%A),流速0.3 mL·min-1,柱温25 ℃,进样量1 μL。
采用电喷雾离子源(ESI),分段正负离子化,多反应监测(MRM)模式,离子喷雾电压4 kV(正离子)、2.5 kV(负离子),鞘气50 Arb,辅助气10 Arb,蒸发温度350 ℃,离子传输管温度325 ℃。各成分质谱参数见表1,各成分总离子流图见图1
分别取哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ对照品适量,精密称定,加甲醇制成每1 mL各含400 μg的对照品储备液。
取本品水蜜丸适量,研碎,取约2.0 g,精密称定,置具塞锥形瓶中,精密加入80%甲醇25 mL,密塞,称量,超声处理(功率300 W,频率40 kHz)30 min,放冷,再称量,用80%甲醇补足减失的量,摇匀,滤过,取续滤液,即得。
分别精密量取哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ的对照品储备液2、2、10、1、0.2、0.5、2、2、0.2、0.2、0.2 mL,置于同一25 mL量瓶中,加甲醇定容,即得混合对照品储备液。继续逐级稀释2、4、10、20倍,即得上述5个浓度的系列混合对照品溶液,按“2.1”项下条件进样测定,以进样浓度(X,μg·mL-1)为横坐标,峰面积积分值(Y)为纵坐标对各成分进行线性回归,11个成分的线性范围、回归方程、相关系数见表2,各成分在各自浓度范围内线性关系良好。基于响应值标准偏差和标准曲线斜率法计算各成分的检测限和定量限(以截距的标准偏差作为响应值标准偏差),结果见表2
取哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ质量浓度为7.43、8.10、39.01、2.59、0.83、1.80、7.28、7.60、0.80、0.72、0.78 μg·mL-1的混合对照品溶液,按“2.1”项下条件连续进样6次,测得上述11个成分峰面积的RSD分别为2.0%、1.2%、1.7%、1.8%、0.89%、1.5%、2.4%、1.7%、1.6%、0.98%、0.77%,表明仪器精密度良好。
分别精密称取同一更年宁样品(批号21033791)6份,按“2.2.2”项下方法制备供试品溶液,按“2.1”项下条件进样测定,测得哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ的平均含量分别为99.9、111.5、374.1、8.8、50.6、62.9、67.0、60.2、21.0、5.0、9.3 μg·g-1,RSD分别为2.4%、2.8%、1.8%、2.4%、0.89%、1.2%、1.1%、1.6%、1.8%、1.3%、0.95%,表明方法重复性良好。
取“2.3.3”项下同一供试品溶液,分别于0、2、4、8、12、24 h按“2.1”项下条件进样,测定峰面积,结果哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ峰面积的RSD分别为2.9%、2.6%、1.6%、2.2%、0.88%、1.4%、2.0%、1.4%、1.9%、1.9%、0.65%,表明供试品溶液在24 h内稳定性良好。
依次精密量取哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ的对照品储备液5、5、20、0.75、2.5、2.5、4、3、1、0.25、0.5 mL,置于同一250 mL量瓶中,加甲醇定容,得到回收率混合对照品溶液(上述11个成分的质量浓度依次为3.71、4.05、15.60、0.39、2.08、1.79、2.91、2.28、0.80、0.18、0.39 μg·mL-1)。精密称取已测得11个成分含量的更年宁样品(批号21033791)粉末1.0 g共6份,分别精密加入上述回收率混合对照品溶液各25 mL,按“2.2.2”项下方法平行制备6份供试溶液,按“2.1”项下条件进样测定,结果哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ的平均回收率分别为95.9%、97.7%、98.3%、102.6%、96.7%、97.9%、97.1%、98.2%、96.5%、100.9%、96.4%,RSD分别为1.2%、1.3%、1.2%、2.8%、1.8%、0.80%、2.1%、1.1%、2.0%、2.2%、3.0%。
取10批更年宁样品各2份,分别按“2.2.2”项下方法制备供试品溶液,按“2.1”项下条件进样测定,以随行标准曲线计算含量,结果见表3
更年宁处方中含22味药,成分复杂,各药味占比小,采用HPLC法进行检测时方法灵敏度低,不能满足低含量成分定量分析的需求。前期,本研究已采用HPLC-PDA建立了更年宁中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、芍药苷、橙皮苷、丹酚酸B的含量测定方法,对处方中黄芩、白芍、牡丹皮、陈皮、丹参的投料情况进行评价。本实验选择采用HPLC-PDA法难以检测的成分哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ为研究对象,采用专属性强、灵敏度高的HPLC-MS/MS法对其进行含量测定,评价处方中玄参、酒女贞子、党参、墨旱莲、炒王不留行、干姜、麸炒白术的投料质量。
实验中曾考察了不同提取溶剂(甲醇、80%甲醇水溶液、50%甲醇水溶液)、提取方式(回流、超声)、提取时间(回流30、60 min,超声15、30、45 min)的效果。结果表明,80%甲醇水溶液提取效果优于甲醇或50%甲醇水溶液,超声提取30 min结果最优。故供试品溶液制备采用80%甲醇水溶液,超声处理30 min。
文献研究表明,女贞子酒炙过程中,随着炮制时间的延长,红景天苷含量显著增加,特女贞苷含量稍有下降,但整体趋于稳定;红景天苷含量与炮制时间显著相关,炮制不足者,红景天苷含量较低[15-17]。2015年版《中华人民共和国药典》(以下简称《中国药典》)规定酒女贞子“含特女贞苷(C31H42O17)不得少于0.70%”,2020年版《中国药典》规定酒女贞子“含红景天苷(C14H20O7)不得少于0.20%”。按处方量折算,更年宁中特女贞苷的理论值为238 μg·g-1,10批样品特女贞苷含量为理论值的140.6%~558.1%,均符合拟定限度;红景天苷的理论值为68 μg·g-1,10批样品红景天苷含量为理论值的55.1%~423.1%,C、D和E企业样品不符合拟定限度。提示各企业所用酒女贞子原料质量存在一定差异,部分企业应关注2020年版《中国药典》酒女贞子质量控制指标的变化。
2020年版《中国药典》规定玄参“含哈巴苷和哈巴俄苷的总量不得少于0.45%”、墨旱莲“含蟛蜞菊内酯不得少于0.04%”、炒王不留行“含王不留行黄酮苷不得少于0.15%”、干姜“含6-姜辣素不得少于0.60%”。按处方量折算,更年宁中哈巴苷与哈巴俄苷的总量、蟛蜞菊内酯、王不留行黄酮苷、6-姜辣素的理论值分别为153、13、38、51 μg·g-1。除A企业2批样品外,其他4个企业8批更年宁样品哈巴苷与哈巴俄苷总量均未达到拟定限度,提示生产企业应重点关注玄参质量。C企业样品蟛蜞菊内酯、6-姜辣素含量均未达到拟定限度,提示该企业应关注墨旱莲、干姜投料质量。各批样品王不留行黄酮苷均符合拟定限度,认为炒王不留行投料情况较好。
2020年版《中国药典》暂未收载党参、麸炒白术含量测定项。对比5个企业10批样品党参炔苷及白术内酯Ⅲ、白术内酯Ⅱ和白术内酯Ⅰ总量的测定结果,各批次间党参炔苷含量及白术内酯总量差异较小。
本文建立了同时测定更年宁中哈巴苷、红景天苷、特女贞苷、党参炔苷、蟛蜞菊内酯、哈巴俄苷、王不留行黄酮苷、6-姜辣素、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ共11个成分的HPLC-MS/MS方法,该方法简便快速,稳定可靠,灵敏度高,专属性强,可为更年宁的质量评价及控制提供科学依据。
