Objective:

To establish a dual-wavelength method for the determination of amylose and amylopectin in potato starch in Tibet.

Methods:

The determination wavelengths of amylopectin and amylopectin in Tibetan potato were 623 and 557 nm,respectively,and the reference wavelengths were 498 and 729 nm. The contents in 11 batches of starch samples collected were determined and analyzed.

Results:

The results showed that the mass concentration of amylose and amylopectin showed a good linear relationship (r=0.999 9,r=0.999 8) in the range of 0- 66 μg·mL-1 and 0-90 μg·mL-1,respectively. The average recoveries of amylose and amylopectin were 102.8% (RSD=1.8%) and 98.5% (RSD=2.2%). The precision (RSD=0.071%,RSD=0.31%),repeatability (RSD=0.26%,RSD=2.4%) and stability (RSD=0.14%,RSD=1.4%) were all in compliance with regulations. The contents of amylose and amylopectin in Tibetan potato starch were 36.74% and 45.87%,respectively. Compared with the commercially available potato starch,the amylose content of Tibetan potato starch was significantly increased (P<0.001),while the amylopectin content was significantly decreased (P<0.05).

Conclusion:

The method is proved to be suitable for the quality evaluation of potato starch in Tibet. There are significant differences in the contents of amylopectin and amylopectin between potato starch in Tibet and potato starch in the market,which provides an experimental basis for the establishment of quality standards of potato starch in Tibet and the development of high-value products.

目的:

建立紫外双波长法测定西藏马铃薯淀粉中直链淀粉和支链淀粉的含量。

方法:

按照双波长测定的等吸收点作图法确定西藏马铃薯直链淀粉和支链淀粉测定波长分别为623、557 nm,参比波长分别为498、729 nm,对收集的11批次淀粉样品进行含量测定及分析。

结果:

直链淀粉、支链淀粉质量浓度分别在0~66、0~90 μg·mL-1范围内呈良好的线性关系(r=0.999 9、0.999 8),6份样品中直链淀粉和支链淀粉的平均加样回收率分别为102.8%(RSD=1.8%)、98.5%(RSD=2.2%),精密度(RSD=0.071%、RSD=0.31%)、重复性(RSD=0.26%、RSD=2.4%)、稳定性(RSD=0.14%、RSD=1.4%)均符合规定。西藏马铃薯淀粉中直链淀粉、支链淀粉含量分别为36.74%、45.87%,与市售马铃薯淀粉相比,西藏马铃薯淀粉的直链淀粉含量显著性增高(P<0.001),支链淀粉含量显著性降低(P<0.05)。

结论:

经方法学验证,该方法可用于西藏马铃薯淀粉的质量评价,西藏马铃薯淀粉与市售的马铃薯淀粉的直链淀粉、支链淀粉含量存在显著性差异,本法为西藏马铃薯淀粉的质量标准建立以及高值产品开发提供了实验依据。

按照双波长测定的等吸收点作图法确定西藏马铃薯直链淀粉和支链淀粉测定波长分别为623、557 nm,参比波长分别为498、729 nm,对收集的11批次淀粉样品进行含量测定及分析。

结果:

直链淀粉、支链淀粉质量浓度分别在0~66、0~90 μg·mL-1范围内呈良好的线性关系(r=0.999 9、0.999 8),6份样品中直链淀粉和支链淀粉的平均加样回收率分别为102.8%(RSD=1.8%)、98.5%(RSD=2.2%),精密度(RSD=0.071%、RSD=0.31%)、重复性(RSD=0.26%、RSD=2.4%)、稳定性(RSD=0.14%、RSD=1.4%)均符合规定。西藏马铃薯淀粉中直链淀粉、支链淀粉含量分别为36.74%、45.87%,与市售马铃薯淀粉相比,西藏马铃薯淀粉的直链淀粉含量显著性增高(P<0.001),支链淀粉含量显著性降低(P<0.05)。

结论:

经方法学验证,该方法可用于西藏马铃薯淀粉的质量评价,西藏马铃薯淀粉与市售的马铃薯淀粉的直链淀粉、支链淀粉含量存在显著性差异,本法为西藏马铃薯淀粉的质量标准建立以及高值产品开发提供了实验依据。

Objective:

To establish a dual-wavelength method for the determination of amylose and amylopectin in potato starch in Tibet.

Methods:

The determination wavelengths of amylopectin and amylopectin in Tibetan potato were 623 and 557 nm,respectively,and the reference wavelengths were 498 and 729 nm. The contents in 11 batches of starch samples collected were determined and analyzed.

Results:

The results showed that the mass concentration of amylose and amylopectin showed a good linear relationship (r=0.999 9,r=0.999 8) in the range of 0- 66 μg·mL-1 and 0-90 μg·mL-1,respectively. The average recoveries of amylose and amylopectin were 102.8% (RSD=1.8%) and 98.5% (RSD=2.2%). The precision (RSD=0.071%,RSD=0.31%),repeatability (RSD=0.26%,RSD=2.4%) and stability (RSD=0.14%,RSD=1.4%) were all in compliance with regulations. The contents of amylose and amylopectin in Tibetan potato starch were 36.74% and 45.87%,respectively. Compared with the commercially available potato starch,the amylose content of Tibetan potato starch was significantly increased (P<0.001),while the amylopectin content was significantly decreased (P<0.05).

Conclusion:

The method is proved to be suitable for the quality evaluation of potato starch in Tibet. There are significant differences in the contents of amylopectin and amylopectin between potato starch in Tibet and potato starch in the market,which provides an experimental basis for the establishment of quality standards of potato starch in Tibet and the development of high-value products.

马铃薯(Solanum tuberosum L.)属茄科茄属一年生草本植物,又名土豆、洋芋等,马铃薯淀粉是从马铃薯植物中提取的由葡萄糖分子聚合而成的多糖[1],具有糊化度高、糊化温度低、吸水力强、透明度好、粘结力强、拉伸性大等特征[2],故其在工业应用中的品质和档次远高于玉米、小麦等其他淀粉,在纺织、制药、饲料和食品等领域用途也很广泛,尤其在食品、制药工业领域[3-5],马铃薯淀粉是产品附加值较高的药用淀粉、改性淀粉等淀粉产品的重要原料,应用价值大。西藏光照时间长、早晚温差大等独特的自然条件,有利于淀粉积累[7],故而马铃薯种植面积逐年增加[6],目前已经达到20余万亩,是西藏第四大粮食作物。但西藏马铃薯淀粉质量研究却鲜有报道,在食品及医药高端淀粉领域的开发应用还是空白。本研究收集了西藏高原主要产地的马铃薯,提取淀粉,建立紫外双波长法测定直链淀粉、支链淀粉的含量,并与目前市售的马铃薯淀粉产品进行对比,为西藏马铃薯淀粉质量标准的建立以及高附加值产业化开发提供实验依据。
直链淀粉标准品(批号RW23543)、支链淀粉标准品(批号CI22924 ),购自西格玛奥德里奇(上海)贸易有限公司;碘化钾(批号20211026)、碘(批号20210926)、氢氧化钾(批号20190807),购自国药集团化学试剂有限公司;无水乙醇(批号20220321),购自南通诚记化工贸易有限公司。以上均为分析纯。
样品:共收集了11批样品,详细样品信息见表1
HH-6数显恒温水浴锅(上海力辰邦西仪器科技有限公司);UV-1900紫外分光光度计(岛津公司);PTY-B5000百分之一天平(华志(福建)电子科技有限公司);XSR205DU/AC十万分之一天平(梅特勒-托利多集团公司);DHG-9070电热恒温鼓风干燥箱(常州中捷实验仪器制造有限公司);SP503A温养破壁料理机(浙江绍兴苏泊尔生活电器);HM-101X卤素水分测定仪(上海禾工科学仪器有限公司)。
选取新鲜的马铃薯150 g,切成大小约0.2 cm3的小块,置破壁机内,加水200 mL,破碎2 min,浆液用100目棉纶筛网过滤,滤渣用150 mL水搅拌洗涤3次,收集合并滤液,滤液置烧杯中,玻璃棒搅拌2 min,静置2 h,弃去上清,沉淀物加水200 mL搅拌洗涤,洗涤2次后用布氏漏斗抽滤,收集滤饼,并置鼓风干燥箱,50 ℃干燥3 h,过100目标准筛,即得。
称取碘化钾1 000 mg于50 mL量瓶中,加入双蒸水20 mL,全部溶解后再加入碘100 mg,全部溶解后再用双蒸水定容,避光保存。
精密称取直链淀粉标准品100 mg,置50 mL烧杯中,加入无水乙醇1 mL润湿,加入0.5 mol·L-1氢氧化钾溶液10 mL,80 ℃水浴10 min,放冷后用蒸馏水定容至100 mL量瓶,即得质量浓度为1 mg·mL-1的直链淀粉标准原液,依同法制备1 mg·mL-1支链淀粉标准原液。
精密称取淀粉样品100 mg,置50 mL烧杯中,加入无水乙醇1 mL润湿,加入0.5 mol·L-1的氢氧化钾溶液10 mL,80 ℃水浴10 min,放冷后用蒸馏水定容至50 mL量瓶,再精密移取2.5 mL于另一50 mL量瓶中,加双蒸水25 mL,再加0.1 mol·L-1盐酸溶液2.5 mL调pH至3左右,再加入碘试剂0.5 mL,用双蒸水定容至刻度,静置20 min,即得。
精密吸取直链淀粉标准原液1 mL,置50 mL量瓶中,加双蒸水25 mL,再加入0.1 mol·L-1盐酸溶液2.5 mL调pH至3左右,加入碘试剂0.5 mL,用双蒸水定容至刻度,静置20 min,以溶剂为空白,紫外光谱400~1 000 nm扫描,最大吸收波长为623 nm;另精密吸取支链淀粉标准原液3 mL,其他处理同直链淀粉标准原液,最大吸收波长为557 nm,见图1。根据等吸收点作图法确定直链淀粉的测定波长为623 nm,参比波长为498 nm;支链淀粉的测定波长为557 nm,参比波长为729 nm。
分别精密吸取直链淀粉标准原液0、0.3、0.9、1.5、2.1、2.7、3.3 mL,置50 mL量瓶中,加双蒸水25 mL,再加0.1 mol·L-1盐酸2.5 mL,调pH至3左右,加碘试剂0.5 mL,定容,混匀,即得质量浓度为0、6、18、30、42、54、66 μg·mL-1的直链淀粉标准溶液;分别精密吸取1 mg·mL-1支链淀粉标准原液0、2.0、2.5、3.0、3.5、4.0、4.5 mL,同法配制质量浓度为0、40、50、60、70、80、90 μg·mL-1的支链淀粉标准溶液。
以溶剂为空白,直链淀粉标准溶液分别在623、498 nm处测得吸收度A1A2,支链淀粉标准溶液分别在557、729 nm处测得吸收度A3A4,以吸收度差值之比(A1-A2)/(A3-A4)为横坐标,质量浓度(μg·mL-1)为纵坐标,绘制标准曲线,回归方程分别为
直链、支链淀粉质量浓度分别在0~66、0~90 μg·mL-1范围内线性良好。
精密称取自提淀粉(S7)90 mg(以干品计),按“2.3.2”项下方法制备供试品溶液,以溶剂为空白,按“2.4”项下测定波长和参比波长测定吸收度,计算吸收度差值,连续测定6次,直链淀粉、支链淀粉吸收度差值的RSD分别为0.071%、0.31%,表明精密度良好。
精密称取自提淀粉(S7)90 mg(以干品计),6份,按“2.3.2”项下方法制备供试品溶液,以溶剂为空白,按“2.4”项下测定波长和参比波长测定吸收度,计算吸收度差值,直链淀粉、支链淀粉吸收度差值分别为0.573、0.194,RSD分别为0.26%、2.4%,表明重复性良好。
精密称取自提淀粉(S7)90 mg(以干品计),按“2.3.2”项下方法制备供试品溶液,以溶剂为空白,分别在0、3、5、10、15、20、30、45、60 min,按“2.4”项下测定波长和参比波长测定吸收度,计算吸收度差值,直链淀粉、支链淀粉吸收度差值的RSD分别为0.14%、1.4%,表明供试品溶液在1 h内稳定性良好。
精密称取自提淀粉(S7)45 mg(以干品计),直链淀粉含量为17.61 mg,加入直链淀粉标准品20 mg,共6份;另精密称取西藏亚东淀粉(S7)50 mg,支链淀粉含量为24.10 mg,加入支链淀粉标准品25 mg,共6份,分别按“2.3.2”项下方法制备供试溶液,以溶剂为空白,按“2.4项”下波长测定吸收度,计算吸收度差值,计算直链淀粉和支链淀粉的含量,平均回收率分别为102.8%和98.5%,RSD分别为1.8%和2.2%,表明本方法回收率较好。
不同来源的马铃薯淀粉,按“2.3.2”项下方法制备供试品溶液,以溶剂为空白,按“2.4”项下波长测定吸收度,计算吸收度差值,代入标准曲线回归方程,外标法计算供试品中直链淀粉和支链淀粉的浓度,并计算直链和支链淀粉的含量,结果见表2。用Excel和SPSS Statistics 25软件对实验数据进行整理和单因素方差分析,实验结果表明,与市售马铃薯淀粉相比,西藏马铃薯淀粉的直链淀粉含量增加约20%,并具有显著性(P<0.001),支链淀粉含量显著性降低(P<0.05)。
西藏高原特有马铃薯主要有昌果红土豆、艾玛土豆等品种[12],因此,本研究采集了上述品种的地理标志产地的马铃薯作为研究供试品,并结合我国内地马铃薯的主要产区以及目前市场上的主要淀粉辅料品种,选用了内地不同产地、不同厂家的市售马铃薯淀粉产品作为研究对照样品。因此,本研究的样品选择具有合理性。我国是世界第一大马铃薯种植国家,但消费方式单一,主要是鲜食[13]、畜禽饲料和普通工业原料,与欧美发达国家的大部分马铃薯淀粉应用于制药、造纸、纺织和建材等行业的高端改性淀粉相比,我国马铃薯淀粉产业的高、精加工依然很薄弱[14]。直链淀粉分子量相对较小且位于淀粉粒的非晶区,在淀粉改性过程中比支链淀粉更容易受到物理、化学影响而得到改性,在低升糖指数食品、可降解材料中[15]的产业化价值巨大。因此,直链淀粉占比是影响马铃薯淀粉改性的重要指标,通常马铃薯淀粉直链淀粉占20%~30%[16- 17],与本实验研究结果一致。利用转基因方法,虽能获得直链淀粉含量高达78%的马铃薯品系,但这些品系的淀粉颗粒形态高度变形,不能规模生产[18-19]。本研究结果证实了西藏马铃薯淀粉具有直链占比较高的特征,这可能与西藏高原光照强度高且持久,昼夜温差大等自然环境有关[20],本研究为转基因马铃薯的研究提供了新的样品,也为西藏马铃薯淀粉在食品、医药等领域的高端改性淀粉的开发利用提供了实验依据。
本研究建立了双波长法测定西藏马铃薯淀粉中的直链、支链的含量,传统的淀粉测定方法主要有旋光法、酶水解法和蒽酮比色法[21],这几种方法对样品的纯度要求较高,样品中残留的糖分与脂类物质会干扰测定,且无法区分直链、支链淀粉。现在最常用的是分光光度法[22],主要有单波长法、双波长法和多波长法,其原理是直链淀粉与碘的结合物呈蓝色,支链淀粉与碘的结合物呈紫色。单波长法可以测定直链、支链淀粉的含量,但直链、支链淀粉吸收峰有部分重叠,对检测结果造成了干扰[23];双波长法扣除了直链、支链淀粉吸收背景的相互影响,消除了这二者引起的干扰,提高测定的灵敏度、选择性和准确度,且能同时测定直链、支链淀粉的含量,进而计算总淀粉含量;多波长法也能同时测得直链、支链淀粉和总淀粉含量,然而由于波长数量多,结果计算相对复杂,应用不多。因此,本文选择双波长比色法进行方法学研究及含量测定,为西藏马铃薯淀粉质量检测以及质量标准研究提供了参考。
