Objective:

To establish a quantitative analysis of multi-components by single marker (QAMS) for simultaneous determination of 7 flavanoids (5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin,eupatilin and casticin) in Artemisiae Argyi Folium.

Methods:

The HPLC system consisted of the Agilent ZORBAX SB-C18 column (150 mm×4.6 mm,5 μm) column with gradient elution of acetonitrile and 0.2% phosphoric acid as the mobile phase at a flow rate of 1.0 mL·min-1,a detection wavelength of 350 nm,and a column temperature of 30 ℃. Eupatilin was selected as the internal reference substance,the relative correction factors between eupatilin and the other 6 flavanoids were established,and the contents of these 7 constituents in samples were calculated to realize QAMS. At the same time,compared with the external standard method to verify the accuracy and feasibility of the QAMS method.

Results:

Within a certain linear range,the relative correction factors between eupatilin and 5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin as well as casticin were 0.958,1.387,1.000,0.950,0.957 and 1.297,respectively (RSDs of RCFs were less than 2.0%). The contents of 5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin,eupatilin,casticin in 20 batches of Artemisiae Argyi Folium were 0.031 4-0.623 5 mg·g-1,0.000 9-0.092 6 mg·g-1,0.020 6-0.170 7 mg·g-1,0.011 0-0.184 7 mg·g-1,0.011 7-0.864 0 mg·g-1,0.253 2-2.555 0 mg·g-1 and 0.015 6-0.250 7 mg·g-1,respectively.

Conclusion:

Using eupatilin as the internal reference,QAMS method for 7 flavanoids is established. The method is accurate and reliable,and can be used for quality control and quantitative analysis of Artemisiae Argyi Folium.

目的:

建立同时测定艾叶中7个黄酮类成分(5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素)的一测多评含量测定方法(QAMS法)。

方法:

采用高效液相色谱法,Agilent ZORBAX SB-C18色谱柱(150 mm×4.6 mm,5 μm),以乙腈-0.2%磷酸水溶液为洗脱流动相,进行梯度洗脱,流速1.0 mL·min-1,检测波长350 nm,柱温30 ℃。以异泽兰黄素为内参物,建立与其他6个黄酮类成分的相对校正因子,并计算7个待测成分的含量,实现一测多评。同时与外标法进行比较,以验证QAMS法的准确性和可行性。

结果:

在一定的线性范围内,异泽兰黄素与5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、蔓荆子黄素的相对校正因子值分别为0.958、1.387、1.000、0.950、0.957、1.297(相对校正因子的RSD<2.0%),并与常规外标一点法比较,20批艾叶中5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素的含量测定结果分别为0.031 4~0.623 5、0.000 9~0.092 6、0.020 6~0.170 7、0.011 0~0.184 7、0.011 7~0.864 0、0.253 2~2.555 0、0.015 6~0.250 7 mg·g-1

结论:

以异泽兰黄素为内参物,建立艾叶中7个黄酮类成分的QAMS法准确、可行,可用于艾叶的定量分析及质量控制。

目的:

建立同时测定艾叶中7个黄酮类成分(5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素)的一测多评含量测定方法(QAMS法)。

方法:

采用高效液相色谱法,Agilent ZORBAX SB-C18色谱柱(150 mm×4.6 mm,5 μm),以乙腈-0.2%磷酸水溶液为洗脱流动相,进行梯度洗脱,流速1.0 mL·min-1,检测波长350 nm,柱温30 ℃。以异泽兰黄素为内参物,建立与其他6个黄酮类成分的相对校正因子,并计算7个待测成分的含量,实现一测多评。同时与外标法进行比较,以验证QAMS法的准确性和可行性。

结果:

在一定的线性范围内,异泽兰黄素与5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、蔓荆子黄素的相对校正因子值分别为0.958、1.387、1.000、0.950、0.957、1.297(相对校正因子的RSD<2.0%),并与常规外标一点法比较,20批艾叶中5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素的含量测定结果分别为0.031 4~0.623 5、0.000 9~0.092 6、0.020 6~0.170 7、0.011 0~0.184 7、0.011 7~0.864 0、0.253 2~2.555 0、0.015 6~0.250 7 mg·g-1

结论:

以异泽兰黄素为内参物,建立艾叶中7个黄酮类成分的QAMS法准确、可行,可用于艾叶的定量分析及质量控制。

Objective:

To establish a quantitative analysis of multi-components by single marker (QAMS) for simultaneous determination of 7 flavanoids (5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin,eupatilin and casticin) in Artemisiae Argyi Folium.

Methods:

The HPLC system consisted of the Agilent ZORBAX SB-C18 column (150 mm×4.6 mm,5 μm) column with gradient elution of acetonitrile and 0.2% phosphoric acid as the mobile phase at a flow rate of 1.0 mL·min-1,a detection wavelength of 350 nm,and a column temperature of 30 ℃. Eupatilin was selected as the internal reference substance,the relative correction factors between eupatilin and the other 6 flavanoids were established,and the contents of these 7 constituents in samples were calculated to realize QAMS. At the same time,compared with the external standard method to verify the accuracy and feasibility of the QAMS method.

Results:

Within a certain linear range,the relative correction factors between eupatilin and 5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin as well as casticin were 0.958,1.387,1.000,0.950,0.957 and 1.297,respectively (RSDs of RCFs were less than 2.0%). The contents of 5-hydroxy-6,7,3’,4’-tetramethoxyflavone,apigenin,hispidulin,kaempferol,jaceosidin,eupatilin,casticin in 20 batches of Artemisiae Argyi Folium were 0.031 4-0.623 5 mg·g-1,0.000 9-0.092 6 mg·g-1,0.020 6-0.170 7 mg·g-1,0.011 0-0.184 7 mg·g-1,0.011 7-0.864 0 mg·g-1,0.253 2-2.555 0 mg·g-1 and 0.015 6-0.250 7 mg·g-1,respectively.

Conclusion:

Using eupatilin as the internal reference,QAMS method for 7 flavanoids is established. The method is accurate and reliable,and can be used for quality control and quantitative analysis of Artemisiae Argyi Folium.

一测多评(quantitative analysis of multi-components by single marker,QAMS)由王智民等[1]于2006年首次提出,是一种高效、简便的测定方法,适合中药特点的多指标质量控制评价。该方法利用在样品中各活性成分之间的关系,仅选定1个化学成分(内参物,通常为典型成分且对照品价低易得),经严格的方法学考察,建立内参物与其他多种成分间的相对校正因子,实现多个同类成分含量的测定[2]。2010年版《中华人民共和国药典》(简称《中国药典》)[3]首次收录QAMS法,但该方法仅限应用于黄连。随着该方法克服了对照品紧缺,多指标测定费用高等问题,已被广泛应用于多种中药质量评价的研究中[4-8],如在2020年版《中国药典》[9]QAMS法的应用已拓展至中药饮片、提取物及制剂等。
艾叶为菊科植物艾Artemsia argyi Lévl. et Vant.的干燥叶,常于夏季花未开时采摘,除去杂质,晒干。艾叶性辛、温,味苦,归肝、脾、肾经,具有温经止血,散寒止痛之功效,主要用于吐血、衄血、崩漏、月经过多,胎漏下血、少腹冷痛、经寒不调、宫冷不孕等疾病,外用可祛湿止痒[9]。现代药理活性研究阐明,艾叶具有抗菌、抗病毒、抗氧化、降血糖、止血、抗肿瘤等药用功效[10-13]。艾叶作为我国传统大宗药材,在中药和民间均有广泛应用[13-15],全国各地均有广泛种植,主要以湖北蕲春、河南安阳、浙江宁波和河北安国为艾叶道地药材主产区[16]。艾叶中富含挥发油类、有机酸类、黄酮类、多糖类和萜类等多种生物活性成分[17-21]。黄酮类成分作为艾叶中一类主要活性成分,已有多项研究表明艾叶黄酮类具有抗氧化、抗肿瘤、抑菌、消炎等多种药理活性[22-25],但目前对于不同产地因地区差异产生的艾叶中黄酮类成分含量内在质量控制研究相对较少。本文采用高效液相色谱法,以异泽兰黄素为内参物,建立QAMS法同时测定不同产地艾叶中5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素7个黄酮类成分的含量,并对QAMS法在中药材艾叶质量控制的研究以及科学性和可行性进行验证。
Agilent 1260高效液相系统:配有G1312B二元梯度泵,G1329B自动进样器,G1316A柱温箱,G1314F DAD检测器(安捷伦公司);Sarturius QUINTIX125D-1CN型十万分之一分析天平(赛多利斯贸易有限公司);JP-060型超声波清洗器(深圳市洁盟清洗设备有限公司);TG16G型台式高速离心机(凯达集团有限公司)。
20批艾叶药材(样品)购自全国7个省、市、自治区的药店或药材市场,湖北蕲春产6批(S1~S6),河南南阳产5批(S7~S11),山东临沂、山东日照、河北安国、安徽六安各产1批(S12~S15),陕西西安产2批(S16~S17)和黑龙江齐齐哈尔产3批(S18~S20),经齐齐哈尔大学张树军教授鉴定为菊科蒿属植物艾Artemsia argyi Lévl. et Vant.的干燥叶。
对照品:异泽兰黄素(批号A0732)、棕矢车菊素(批号A0731)、芹菜素(批号A0113)、高车前素(批号A1000)、山柰酚(批号A0129)、蔓荆子黄素(批号A0363),纯度均≥98%,购于成都曼思特生物科技有限公司。5-羟基-6,7,3’,4’-四甲氧基黄酮为实验室自制,经HPLC(面积归一化法)分析,其纯度>98%。
甲醇、乙腈、磷酸,均为色谱纯,购于上海阿拉丁生化科技股份有限公司,水为屈臣氏蒸馏水。
采用Agilent ZORBAX SB-C18色谱柱(150 mm×4.6 mm,5 μm),流动相为乙腈(A)-0.2%磷酸溶液(B),梯度洗脱时间程序(0~10 min,20%A→30%A;10~25 min,30%A→35%A;25~30 min,35%A→40%A;30~40 min,40%A→100%A),流速1.0 mL·min-1,检测波长350 nm,柱温30 ℃,进样量10 μL。
精密称取5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素的对照品适量,分别置量瓶中,加甲醇溶解,配制成质量浓度依次为10.0、1.0、2.0、2.5、10.0、25.0、5.0 μg·mL-1的混合对照品溶液,低温避光保存,备用。
艾叶样品经研磨粉碎,称取粉末(过4号筛)约0.2 g,置于具塞锥形瓶中,精密加入70%乙醇10 mL,称量,超声(140 W,42 Hz)提取60 min,再次称量,以70%乙醇为提取剂补足减失的量,摇匀,转移至离心管内,10 000 r·min-1离心5 min,取上清液,经0.22 μm微孔滤膜进行滤过,即得。
在“2.1”项色谱条件下,样品中7个待测成分的色谱峰峰形及分离效果良好,符合要求。混合对照品以及艾叶样品色谱图见图1
精密吸取“2.2.1”项下混合对照品溶液1、2、5、8、10、15、20 μL,按“2.1”项下色谱条件,以色谱峰峰面积Y为纵坐标,对照品质量X(μg)为横坐标制作标准曲线,得到各成分的线性回归方程、相关系数(r)以及线性范围。取混合对照品溶液,经甲醇逐级稀释后,以信噪比(S/N)约为3.0计算检测限(LOD),S/N约为10.0计算定量限(LOQ)。结果见表1
精密吸取“2.2.1”项下的混合对照品溶液,按“2.1”项下色谱条件,分别进样2、4、6、8、10、12 μL进行测定分析。按照公式fi,s=fi/fs=(mi×As)/(ms×Ai)(mi为内参物i的浓度,Ai为内参物i的峰面积,ms为内参物s的浓度,As为内参物s的峰面积),分别计算异泽兰黄素对5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、蔓荆子黄素的相对校正因子,见表2
取“2.2.1”项下混合对照品溶液,按照“2.1”项下色谱条件连续进样6次进行分析,记录各色谱峰峰面积,计算RSD。5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素峰面积的RSD(n=6)分别为0.58%、0.64%、1.0%、0.74%、0.66%、1.1%和0.77%,结果表明仪器的精密度良好。
精密称取S1样品粉末(过4号筛),按“2.2.2”项下方法制备供试品溶液,分别于配制后的0、2、4、8、12、24 h按“2.1”项下色谱条件进行进样分析,记录峰面积。5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素峰面积的RSD(n=6)分别为0.27%、0.42%、0.33%、0.46%、0.25%、0.30%和0.41%,结果表明供试品溶液在24 h内稳定性良好。
精密称取S1样品粉末(过4号筛)6份,按“2.2.2”项下方法制备供试品溶液,按照“2.1”项下色谱条件,分别进样分析,记录峰面积。5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素的平均含量(n=6)分别为0.233 3、0.092 1、0.043 0、0.087 6、0.245 6、0.764 2、0.059 4 mg·g-1,RSD分别为1.1%、1.3%、1.4%、1.2%、0.98%、1.1%和1.2%,表明该方法的重复性良好。
精密称取S15样品粉末(过4号筛)9份,每份约0.1 g,分成3组,每组3份,分别按照供试品中待测成分的量与对照品量之比为1:0.8、1:1、1:1.2,精密加入对照品适量,按照“2.2.2”项下的方法制备供试溶液,在“2.1”项色谱条件下进样测定,计算各待测物质的平均加样回收率和RSD,结果见表3,表明该方法的回收率良好。
试验分别考察2台高效液相色谱仪Agilent 1260、Ulimate 3000和2种色谱柱Agilent ZORBAX SB-C18(150 mm×4.6 mm,5 μm)、Thermo ScientificTM Hypersil GOLD C18(50 mm×2.1 mm,1.9 μm),各待测成分的相对校正因子结果见表4。表明,相对校正因子在不同的色谱柱和仪器下无显著性影响,具有较好的适用性。
采用Agilent 1260高效液相色谱仪,Agilent ZORBAX SB-C18(150 mm×4.6 mm,5 μm)色谱柱,考察流速为0.8、1.0、1.2 mL·min-1时的相对校正因子,结果流速为0.8 mL·min-1时5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、蔓荆子黄素相对校正因子分别为1.372、0.977、0.948、0.974、0.962、1.273;流速为1.0 mL·min-1时上述成分的相对校正因子分别为1.382、0.966、0.931、0.958、0.944、1.254,流速为1.2 mL·min-1时上述成分的相对校正因子分别为1.380、1.001、0.970、0.968、0.971、1.280。3种流速下求得上述成分相对校正因子的RSD(n=3)分别为0.86%、0.94%、1.0%、0.77%、1.2%和1.2%,说明相对校正因子在不同流速下具有良好适用性。
采用Agilent 1260高效液相色谱仪,Agilent ZORBAX SB-C18(150 mm×4.6 mm,5 μm)色谱柱,考察柱温为25、30、35 ℃时的相对校正因子,结果柱温为25 ℃时5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、蔓荆子黄素的相对校正因子分别为1.383、0.984、0.932、0.964、0.957、1.252,柱温为30 ℃时上述成分的相对校正因子分别为1.342、0.970、0.970、0.982、0.973、1.240,柱温为35 ℃时上述成分的相对校正因子分别为1.347、0.956、0.940、0.968、0.960、1.260。3种柱温下求得的相对校正因子的RSD(n=3)分别为0.94%、1.3%、1.2%、1.2%、0.95%和1.0%,说明相对校正因子在不同柱温下具有良好的适用性。
QAMS法待测成分色谱峰可以用相对保留值法Ri/s=tR(i)/tR(s)以及各色谱峰的紫外吸收特征来准确地定位。本研究考察了相对保留时间在不同色谱柱和不同品牌仪器上的重现性,结果见表5。还考察了Agilent 1260高效液相色谱仪,Agilent ZORBAX SB-C18(150 mm×4.6 mm,5 μm)色谱柱,柱温分别为25、30、35 ℃,以及流速为0.8、1.0、1.2 mL·min-1时的相对保留时间。结果显示,各成分的相对保留时间无明显差异变化,则可选用相对保留值作为目标成分色谱峰的定位指标。
取7个产地的20批艾叶,按照“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样分析,使用建立的QAMS法计算艾叶中7个黄酮类成分的含量,结果见表6
取供试品溶液,按照“2.1”色谱条件进样测定,采用外标法(ESM)计算艾叶中7个黄酮类成分的含量,并将测定的计算结果与QAMS法的计算结果进行比较,验证QAMS法用于测定艾叶中7个黄酮类成分含量的准确性,结果见表6。结果表明2种方法测定的结果无显著差异,表明QAMS法测定艾叶中7个黄酮类成分的含量具有可行性。同时对2种方法的含量测定结果进行t-检验,结果无显著性差异(P>0.05),显示QAMS法准确可行。
本研究根据黄酮类化合物的溶解性,分别采用提取溶剂(甲醇、乙醇)、提取溶剂浓度(50%、70%、80%、100%)和料液比(1:10、1:25、1:50)对艾叶中7个待测成分的含量测定进行考察,同时因超声提取法操作简单、便捷,因此选择超声提取法。根据结果显示,最终确定按照“2.2.2”项下方法制备供试品溶液。
根据各成分在流动相中的紫外吸收谱图来确定检测波长。实验采用Agilent 1260 G1314F VWD检测器,着重考察了在280、326、350、360 nm等检测波长下艾叶中7个待测成分色谱峰的响应值和分离度,当检测波长为350 nm时,色谱峰响应值较高,分离度好,故选择350 nm作为本实验QAMS法的检测波长。
在艾叶药材的7个黄酮类成分中,由于异泽兰黄素性质稳定,在样品中含量较多,易于分离提纯,可轻易获得,因此选作内参物。
正确的定位待测成分色谱峰,是QAMS法成功应用的重点。本研究以相对保留值结合待测成分的紫外吸收光谱为依据,确定艾叶中黄酮类成分色谱峰的定位。
本研究对7个产地20批艾叶进样测定,按照“2.2.2”项下的方法制备供试品溶液,在“2.1”项的色谱条件下进行样品分析测定,应用建立的QAMS法计算艾叶中7个黄酮类成分含量。20批艾叶中5-羟基-6,7,3’,4’-四甲氧基黄酮、芹菜素、高车前素、山柰酚、棕矢车菊素、异泽兰黄素、蔓荆子黄素的含量测定结果分别为0.031 4~0.623 5、0.000 9~0.092 6、0.020 6~0.170 7、0.011 0~0.184 7、0.011 7~0.864 0、0.253 2~2.555 0、0.015 6~0.250 7 mg·g-1。7个黄酮类成分含量之和为0.360 0~5.014 2 mg·g-1。湖北蕲春、河南安阳产的艾叶中7个黄酮类成分的含量之和分别为1.526~4.252 mg·g-1、2.309~3.175 mg·g-1,在7个产地中相对较高,在一定程度上说明药材质量较优。
本研究建立QAMS法同时测定艾叶中7个黄酮类成分,方法学考察表明该方法准确可行。相对校正因子考察发现其在不同仪器、不同色谱柱、不同流速、不同柱温的条件下均具有良好的适用性,因此选用相对保留值作为目标成分色谱峰的定位指标。7个产地20批药材的测定结果与标准曲线法所测值无明显的差异,说明建立不同成分之间的相对校正因子具有较高的可行性。本研究通过QAMS技术实现同时测定艾叶中的多个指标成分,为艾叶的质量控制提供了更科学、更准确、更稳定的评价模式。
