Objective:

To determine the color of Arnebiae Radix, and the contents of six main purple pigment components (acetylshikonin,β-acetoxyisovalerylalkannin, deoxyshikonin, isobutylshikonin,β,β’-dimethylacrylalkannin and isovalerylshikonin) in Arnebiae Radix, and to study on the correlation between the color of Arnebiae Radix and the contents of six main purple pigment components.

Methods:

The La,and b values of the sample powder were determined using a spectrophotometer to characterize the color of Arnebiae Radix. The International Commission on Illumination (CIE) had developed a Lab color model,which was a digital description of human vision. A higher L value indicated greater brightness,a higher a value indicated redness and a lower a value indicates greenness,and a higher b value indicated yellowing and a lower b value indicates blueness. The contents of purple pigment components were determined using high performance liquid chromatography (HPLC),and the correlation between La,and b values and the contents of six main purple pigments was calculated using SPSS software.

Results:

The contents of acetylshikonin in 135 batches of samples ranged from 0.01% to 3.39%. The contents of β-acetoxyisovalerylalkannin ranged from 0.00% to 1.95%. The contents of deoxyshikonin ranged from 0.00% to 0.23%. The contents of isobutylshikonin ranged from 0.01% to 1.13%. And the contents of isovalerylshikonin ranged from 0.02% to 2.88%. The contents of β,β’-dimethylacrylalkannin ranged from 0.01% to 2.17%. There was a significant negative correlation between the contents of acetylshikonin,β-acetoxyisovalerylalkannin and isobutylshikonin and the L (black_white) chromaticity value of Arnebiae Radix,with a Spearman correlation coefficients between -0.138 and -0.222. The chromaticity value of a (red_green) was related to the five components other than acetylshikonin,which were β-acetoxyisovalerylalkannin,deoxyshikonin,isobutylshikonin,β,β’-dimethylacrylalkannin,and isovalerylshikonin,with a spearman correlation coefficients between 0.176 and 0.355;b (blue_yellow) chromaticity value was related to the five components other than β-acetoxyisovalerylalkannin,which were acetylshikonin,deoxyshikonin,and isobutylshikonin,β,β’-dimethylacrylalkannin. β,β’-dimethylacrylalkannin was positively correlated with a coefficient of 0.290,and negatively correlated with the other four components with a coefficients between -0.325 and -0.633.

Conclusion:

It is recommended that the assay limits of Arnebiae Radix be revised to β,β’-dimethylacrylalkannin not less than 0.30% and isovalerylshikonin not less than 0.29%.

目的:

测量紫草的色泽,测定紫草中6个主要紫色素类成分(乙酰紫草素、β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰紫草素)的含量,研究新疆紫草色泽与6个主要紫色素成分含量的相关性。

方法:

采用分光测色仪测定样品粉末的Lab值用于表征紫草的色泽。国际照明委员会(CIE)制定了Lab颜色模型,是人类视觉的数字化描述,L值越大表示亮度越大,a值增大表示偏红减小表示偏绿,b值增大表示偏黄减小表示偏蓝;采用高效液相色谱法(HPLC)测定紫色素类成分的含量,使用SPSS软件计算Lab值与6个主要紫色素含量的相关程度。

结果:

目的:

测量紫草的色泽,测定紫草中6个主要紫色素类成分(乙酰紫草素、β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰紫草素)的含量,研究新疆紫草色泽与6个主要紫色素成分含量的相关性。

方法:

采用分光测色仪测定样品粉末的Lab值用于表征紫草的色泽。国际照明委员会(CIE)制定了Lab颜色模型,是人类视觉的数字化描述,L值越大表示亮度越大,a值增大表示偏红减小表示偏绿,b值增大表示偏黄减小表示偏蓝;采用高效液相色谱法(HPLC)测定紫色素类成分的含量,使用SPSS软件计算Lab值与6个主要紫色素含量的相关程度。

结果:

135批样品乙酰紫草素的含量为0.01%~3.39%,β-乙酰氧基异戊酰阿卡宁含量为0.00%~1.95%,去氧紫草素含量为0.00%~0.23%,异丁酰紫草素含量为0.01%~1.13%,异戊酰紫草素含量为0.02%~2.88%,β,β’-二甲基丙烯酰阿卡宁含量为0.01%~2.17%。紫草药材的L(黑_白)色度值与乙酰紫草素、β-乙酰氧基异戊酰阿卡宁和异丁酰紫草素3个成分的含量呈显著负相关关系,斯皮尔曼相关系数在-0.138和-0.222之间;a(绿_红)色度值与β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰阿卡宁的含量均呈现显著的正相关,斯皮尔曼相关系数在0.176和0.355之间;b(蓝-黄)色度值与乙酰紫草素、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰阿卡宁均呈现显著的相关性,其中,与β,β’-二甲基丙烯酰阿卡宁呈正相关,系数为0.290,与其余4个成分呈负相关,系数在-0.325和-0.633之间。

结论:

建议紫草(新疆紫草)的含量测定项修订为β,β’-二甲基丙烯酰阿卡宁不得少于0.30%并且异丁酰紫草素不得少于0.29%。

Objective:

To determine the color of Arnebiae Radix, and the contents of six main purple pigment components (acetylshikonin,β-acetoxyisovalerylalkannin, deoxyshikonin, isobutylshikonin,β,β’-dimethylacrylalkannin and isovalerylshikonin) in Arnebiae Radix, and to study on the correlation between the color of Arnebiae Radix and the contents of six main purple pigment components.

Methods:

The La,and b values of the sample powder were determined using a spectrophotometer to characterize the color of Arnebiae Radix. The International Commission on Illumination (CIE) had developed a Lab color model,which was a digital description of human vision. A higher L value indicated greater brightness,a higher a value indicated redness and a lower a value indicates greenness,and a higher b value indicated yellowing and a lower b value indicates blueness. The contents of purple pigment components were determined using high performance liquid chromatography (HPLC),and the correlation between La,and b values and the contents of six main purple pigments was calculated using SPSS software.

Results:

The contents of acetylshikonin in 135 batches of samples ranged from 0.01% to 3.39%. The contents of β-acetoxyisovalerylalkannin ranged from 0.00% to 1.95%. The contents of deoxyshikonin ranged from 0.00% to 0.23%. The contents of isobutylshikonin ranged from 0.01% to 1.13%. And the contents of isovalerylshikonin ranged from 0.02% to 2.88%. The contents of β,β’-dimethylacrylalkannin ranged from 0.01% to 2.17%. There was a significant negative correlation between the contents of acetylshikonin,β-acetoxyisovalerylalkannin and isobutylshikonin and the L (black_white) chromaticity value of Arnebiae Radix,with a Spearman correlation coefficients between -0.138 and -0.222. The chromaticity value of a (red_green) was related to the five components other than acetylshikonin,which were β-acetoxyisovalerylalkannin,deoxyshikonin,isobutylshikonin,β,β’-dimethylacrylalkannin,and isovalerylshikonin,with a spearman correlation coefficients between 0.176 and 0.355;b (blue_yellow) chromaticity value was related to the five components other than β-acetoxyisovalerylalkannin,which were acetylshikonin,deoxyshikonin,and isobutylshikonin,β,β’-dimethylacrylalkannin. β,β’-dimethylacrylalkannin was positively correlated with a coefficient of 0.290,and negatively correlated with the other four components with a coefficients between -0.325 and -0.633.

Conclusion:

It is recommended that the assay limits of Arnebiae Radix be revised to β,β’-dimethylacrylalkannin not less than 0.30% and isovalerylshikonin not less than 0.29%.

紫草为紫草科植物新疆紫草Arnebia euchroma (Royle) Johnst.或内蒙紫草Arnebia guttata Bunge的干燥根[1]。《中华人民共和国药典》(简称ChP)最早收载紫草含量测定项的是ChP 1990[2],使用紫外分光光度法检测左旋紫草素最大吸收波长516 nm处的吸收度,沿用至ChP 2020。ChP 2005[3]增设含量测定项(2),使用高效液相色谱法检测β,β’-二甲基丙烯酰阿卡宁(C21H22O6)这一萘醌类成分的含量,沿用至ChP 2020。ChP 1963[1]首次收载紫草品种,并提出紫草“……色紫者为佳”。历版药典[1-10]对紫草各部位的描述均围绕颜色展开,使用的词详见表1
经统计,上述10个版本ChP的紫草标准正文中共提到紫色相关的词99次,其中,紫红色44次、紫褐色19次、深紫色18次、紫黑色11次、暗紫色7次;提到黄白色相关的词56次,其中黄白色26次、黄色20次、灰黄色10次,这些颜色词汇比较接近,肉眼观察难以区分,有很大的主观性。近年来,药材市场上出现了紫草的混伪品,但这些混伪品按照药典含量测定项方法检测大多都合格,因此,很多研究者对紫草含量测定提出了改进意见,建议增加羟基萘醌为母核的天然紫色素类成分指标,如乙酰紫草素、β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素或异戊酰紫草素等。因此,如何客观地描述紫草的颜色,并且探究备选多个紫色素类指标成分与颜色之间的相关性,最终做到“辨色论质”,成为紫草质量评价亟待解决的问题。
色泽是药材鉴别的重要内容,通过色泽可直观快速地判断药材的优劣。研究表明,色泽的测定数据可以用于药材的质量评价[11-12]。现代色彩分析技术的发展,特别是测色仪器的使用,使得色泽测量和分析更加精密和准确[13-14],且可以将人的主观判断客观量化。在传统的色泽评价指标中引入现代的色彩分析技术,对于中药材及饮片的质量评价具有极为重要的意义。
近年来,紫草内在质量控制方面开展了较多的研究[15-18],但同时结合传统性状进行客观分析尚未见报道。本文采用分光测色仪测定紫草的色泽,采用HPLC法测定乙酰紫草素、β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁及异戊酰紫草素含量,对色泽与化学成分之间相关性进行分析,为紫草质量的分析、评价与提高奠定了基础。
DC850分光测色仪(德塔公司);Waters e2695型液相色谱仪-2998二极管阵列检测器(沃特世科技有限公司);MSE125S型十万分之一电子天平、QUINTIX313-1CN型万分之一电子天平(Sartorius公司);KQ-500DV型超声波清洗器(江苏昆山市超声仪器有限公司);Milli-Q去离子水发生器(密理博公司)。
紫草(新疆紫草)对照药材(批号121430-201705)和β,β’-二甲基丙烯酰阿卡宁对照品(批号111689-201805,纯度≥98%)由中国食品药品检定研究院提供;对照品β-羟基异戊酰紫草素(批号STC9420105,纯度≥98%)、乙酰紫草素(批号ST78820120,纯度≥98%)、β-乙酰氧基异戊酰阿卡宁(批号ST17190120,纯度≥98%)、去氧紫草素(批号ST17160120,纯度≥98%)由上海诗丹德科技有限公司提供;对照品异丁酰紫草素(批号T02D6Z6828,纯度≥98%)和异戊酰紫草素(批号Z1357B20931,纯度≥98%)由上海源叶生物技术有限公司提供。135批药材(标记为001~135)中,001~077批为2022年国家药品抽检项目的合格样品;078~135批为市场收集及实地采集的样品,经中国食品药品检定研究院郑健研究员鉴定为紫草科软紫草属新疆紫草Arnebia euchroma(Royle)Johnst.的干燥根。
色度测量:样品粉碎后过6号筛,取样量2 g,使用D65光源,30 mm孔径,平行测量2次,取平均值。
色谱条件:采用文献[17]的方法,使用Waters Xbridge C18色谱柱(4.6 mm×250 mm,5 μm),流动相为乙腈-0.05%甲酸水溶液(70:30),流速1.0 mL·min-1,柱温30 ℃,检测波长275 nm,进样量10 μL。
135批样品CIE色彩空间Lab值测量结果:L值(黑_白)最小值19.65,最大值41.41;a值(绿_红)最小值5.85,最大值15.79;b值(蓝_黄)最小值3.46,最大值10.54。含量测定结果:乙酰紫草素含量为0.01%~3.39%,β-乙酰氧基异戊酰阿卡宁含量为0.00%~1.95%,去氧紫草素含量为0.00%~0.23%,异丁酰紫草素含量为0.01%~1.13%,异戊酰紫草素含量为0.02%~2.88%,β,β’-二甲基丙烯酰阿卡宁含量为0.01%~2.17%。
使用SPSS 25.0软件中的相关分析法,分别计算上述紫草样品中紫草素类成分的含量与其b值(蓝_黄)、L值(黑_白)、a值(绿_红)之间的相关系数,结果见表23
从上述相关分析可见,紫草(新疆紫草)的L(黑_白)色度值与乙酰紫草素、β-乙酰氧基异戊酰阿卡宁和异丁酰紫草素3个成分的含量呈显著负相关,斯皮尔曼相关系数在-0.138到-0.222之间;a(绿_红)色度值与除乙酰紫草素之外的5个成分(β-乙酰氧基异戊酰阿卡宁、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰阿卡宁)均呈现显著的正相关,斯皮尔曼相关系数在0.176到0.355之间;b(蓝-黄)色度值与除β-乙酰氧基异戊酰阿卡宁之外的5个成分(乙酰紫草素、去氧紫草素、异丁酰紫草素、β,β’-二甲基丙烯酰阿卡宁、异戊酰阿卡宁)均呈现显著的相关性,其中,与β,β’-二甲基丙烯酰阿卡宁呈正相关,相关系数为0.290,与其余4个成分呈负相关,相关系数在-0.325到-0.633之间。
按照色彩混合原理,紫色为红色加蓝色的混合色,因此,与ChP描述新疆紫草“紫红色或紫褐色”“深紫色”等词汇接近的色度应为L值(黑_白)低、a值(绿_红)高和b值(蓝-黄)低的模式。6个紫草素类成分中,β,β’-二甲基丙烯酰阿卡宁与a值(绿_红)呈显著的正相关,与L值(黑_白)存在负相关,说明药典选择该成分作为紫草含量测定项的指标成分在颜色相关性上有一定合理性,与传统观点紫草“色紫者佳”有一致之处。但是,β,β’-二甲基丙烯酰阿卡宁与L值(黑_白)的负相关关系不显著,且与b值(蓝-黄)呈显著的正相关,因此,建议增加与L值(黑_白)呈显著负相关且与b值(蓝-黄)呈显著负相关的成分,即乙酰紫草素和异丁酰紫草素作为含量测定项的指标,同时,考虑到只有异丁酰紫草素与a值(绿_红)同时呈显著的正相关,因此,新增的最佳含量测定指标成分为异丁酰紫草素。
根据135批样品的含量计算异丁酰紫草素含量平均值为0.27%,同时,计算β,β’-二甲基丙烯酰阿卡宁不少于0.30%的115批样品,其异丁酰紫草素含量平均值为0.29%。综合考虑,建议紫草(新疆紫草)的含量测定项修订为β,β’-二甲基丙烯酰阿卡宁不得少于0.30%并且异丁酰紫草素不得少于0.29%。
