Objective:

To establish an in vitro permeation method for crisaborole ointment and to provide a reference for the bioequivalence of crisaborole ointment.

Methods:

The HPLC method was used to determine the permeability of crisaborole ointment, and the vertical Franz diffusion pool method was used to study the release characteristics of the ointment in vitro. The chromatographic column was Thermo BDS Hypersil-C18(150 mm×4.6 mm, 5 μm), with 0.1% phosphoric acid solution-acetonitrile (60:40) as the mobile phase, flow rate of 1.5 mL·min-1, column temperature of 30 ℃, detection wavelength of 254 nm, and injection volume of 10 μL.

Results:

The peak shape of crisaborole in the chromatogram of the test sample was good. There was no peak at the position of crisaborole in the negative sample chromatogram, which did not interfere with the determination. The linear relationship of crisaborole was good within the range of 0.026-21.02 μg·mL-1 (r≥0.999). In six repeated experiments, the mean maximum transmittance (Jmax) of the self-made product and the reference formulation were compared (odds ratio: 0.94). Simultaneously calculate the confidence interval for Jmax, with a 90% confidence interval ranging from 87.9% to 102.0%. The mass conservation percentage of each sample was 94.1%-101.1%(n=12). The reference solution was left at room temperature for 26 h, with a peak area RSD of 0.44%. The self-made product and reference preparation were left in a water bath at room temperature and 32 ℃ for 26 h, with peak area RSD ranging from 0.78% to 1.1%, indicating good stability of the solution. For the three batches of samples tested, the ratio of the mean Jmax values of the self-made product and the reference formulation was within the range of 0.92-0.97. Simultaneously calculate the confidence interval for Jmax, with a 90% confidence interval ranging from 84.8%-100.7%.

Conclusion:

This method is simple and convenient to operate, with high selectivity, strong exclusivity and high accuracy.

目的:

建立克立硼罗软膏体外透皮方法,为克立硼罗软膏生物等效性提供参考依据。

方法:

结果:

样品色谱图中克立硼罗峰形良好;阴性样品色谱图中在克立硼罗位置未出峰,不干扰测定;克立硼罗质量浓度在0.026~21.02 μg·mL-1的范围内线性关系良好(r≥0.999);重复性6份试验中,将自制品与参比制剂最大渗透速率(Jmax)的均值进行比较,比值为0.94;同时对Jmax进行置信区间计算,考察90%置信区间为87.9%~102.0%;各样品质量守恒百分比为94.1%~101.1%(n=12);对照品溶液室温放置26 h,峰面积的RSD为0.44%,自制品及参比制剂分别于室温、32 ℃水浴放置26 h,峰面积的RSD为0.78%~1.1%,说明溶液稳定性较好。对3批样品检测,自制品和参比制剂的Jmax均值的比值均在0.92~0.97范围内,同时对Jmax进行置信区间计算,考察90%置信区间为84.8%~100.7%。

结论:

该方法操作简单、便捷,方法选择性高,专属性强,准确度较高。

目的:

建立克立硼罗软膏体外透皮方法,为克立硼罗软膏生物等效性提供参考依据。

方法:

采用HPLC法测定克立硼罗软膏渗透量,采用垂直式的Franz扩散池法研究软膏体外释放特性。采用Thermo BDS Hypersil-C18(150 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸溶液-乙腈(60:40)为流动相,流速1.5 mL·min-1,柱温30 ℃,检测波长254 nm,进样量10 μL。

结果:

样品色谱图中克立硼罗峰形良好;阴性样品色谱图中在克立硼罗位置未出峰,不干扰测定;克立硼罗质量浓度在0.026~21.02 μg·mL-1的范围内线性关系良好(r≥0.999);重复性6份试验中,将自制品与参比制剂最大渗透速率(Jmax)的均值进行比较,比值为0.94;同时对Jmax进行置信区间计算,考察90%置信区间为87.9%~102.0%;各样品质量守恒百分比为94.1%~101.1%(n=12);对照品溶液室温放置26 h,峰面积的RSD为0.44%,自制品及参比制剂分别于室温、32 ℃水浴放置26 h,峰面积的RSD为0.78%~1.1%,说明溶液稳定性较好。对3批样品检测,自制品和参比制剂的Jmax均值的比值均在0.92~0.97范围内,同时对Jmax进行置信区间计算,考察90%置信区间为84.8%~100.7%。

结论:

该方法操作简单、便捷,方法选择性高,专属性强,准确度较高。

Objective:

To establish an in vitro permeation method for crisaborole ointment and to provide a reference for the bioequivalence of crisaborole ointment.

Methods:

The HPLC method was used to determine the permeability of crisaborole ointment, and the vertical Franz diffusion pool method was used to study the release characteristics of the ointment in vitro. The chromatographic column was Thermo BDS Hypersil-C18(150 mm×4.6 mm, 5 μm), with 0.1% phosphoric acid solution-acetonitrile (60:40) as the mobile phase, flow rate of 1.5 mL·min-1, column temperature of 30 ℃, detection wavelength of 254 nm, and injection volume of 10 μL.

Results:

The peak shape of crisaborole in the chromatogram of the test sample was good. There was no peak at the position of crisaborole in the negative sample chromatogram, which did not interfere with the determination. The linear relationship of crisaborole was good within the range of 0.026-21.02 μg·mL-1 (r≥0.999). In six repeated experiments, the mean maximum transmittance (Jmax) of the self-made product and the reference formulation were compared (odds ratio: 0.94). Simultaneously calculate the confidence interval for Jmax, with a 90% confidence interval ranging from 87.9% to 102.0%. The mass conservation percentage of each sample was 94.1%-101.1%(n=12). The reference solution was left at room temperature for 26 h, with a peak area RSD of 0.44%. The self-made product and reference preparation were left in a water bath at room temperature and 32 ℃ for 26 h, with peak area RSD ranging from 0.78% to 1.1%, indicating good stability of the solution. For the three batches of samples tested, the ratio of the mean Jmax values of the self-made product and the reference formulation was within the range of 0.92-0.97. Simultaneously calculate the confidence interval for Jmax, with a 90% confidence interval ranging from 84.8%-100.7%.

Conclusion:

This method is simple and convenient to operate, with high selectivity, strong exclusivity and high accuracy.

克立硼罗是一种磷酸二酯酶4(PDE4)抑制剂,可以减少特应性皮炎病理生理学中涉及的多种炎症细胞因子的产生,有效渗透皮肤屏障,实现局部给药。这种抑制剂导致细胞内环磷酸腺苷(cAMP)水平增高,用于治疗真菌感染,具体用于治疗甲癣或皮肤真菌感染。在保证患者舒适度的同时,具有可以长期应用以及用于特殊部位的优势。美国FDA批准其用于局部治疗2岁及以上患者轻度至中度特异性皮炎,为儿童提供了一款安全有效的药物,打破了外用非激素类抗炎领域没有新产品的现状,具有划时代的意义[1-3]
克立硼罗软膏属于外用药物制剂,该类制剂的质量测试分为2类:一类是评估产品质量属性的测试;另一类是评估产品性能的测试,包括体外释放试验(in vitro release test,IVRT)和体外透皮试验(in vitro permeation test,IVPT)。IVPT可用于表征外用药物制剂活性成分达到作用部位的速度和程度,可通过IVPT的等效性,证明生物利用度的速率和程度无显著差异,用于支持生物等效性论证或代替临床治疗等效性试验[4-13]
本实验采用垂直式的Franz扩散池法,建立该制剂的体外透皮测定方法,并对方法进行验证[14-15],为克立硼罗软膏质量研究提供依据。

水(屈臣氏);磷酸(天津市科密欧化学试剂有限公司,批号20230407);乙腈(MREDA公司,批号D211128);无水乙醇(FEPURE公司,批号191025);0.9%氯化钠注射液(石家庄四药有限公司,批号2304033202);聚四氟乙烯膜(津腾微孔滤膜有限公司,0.45 μm);吐温80(药用级,麦克林化学试剂有限公司,批号C15064851)。
巴马小型猪皮肤(泰州泰和生物科技有限公司);巴马小型猪皮肤(北京和邦兴业科学仪器有限公司);小型巴马香猪皮肤(北京德默高科医药技术有限公司)。
采用Thermo BDS Hypersil-C18(150 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸溶液-乙腈(60:40)为流动相,流速1.5 mL·min-1,柱温30 ℃,检测波长254 nm,进样量10 μL。
装置为垂直式的Franz扩散池,以1%吐温80溶液为接收液,接收液温度32 ℃±1.0 ℃,转速300 r·min-1,取样体积8 mL,透皮扩散面积2.2 cm2,上药量0.3 g,皮肤为巴马香猪皮肤。
称取克立硼罗对照品约10 mg,精密称定,置200 mL量瓶中,加乙腈-水(90:10)溶解并稀释至刻度,摇匀,精密量取5 mL置50 mL量瓶中,加1%吐温80溶液稀释至刻度,摇匀,即得。
将直径约25 mm的巴马香猪皮肤夹在Franz扩散池的上下两室之间,角质层朝上,称取克立硼罗软膏约300 mg,均匀涂于皮肤表面,试验参数同“2.1.2”项,分别于2、4、6、8、10、12、24 h取接收液8 mL,采用0.45 μm微孔滤膜过滤,取续滤液作为供试品溶液。
按照处方比例配制不含克立硼罗的空白样品,称取约0.3 g,按照“2.1.4”项下方法制备阴性样品溶液。
取对照品溶液、供试品溶液、阴性样品溶液各10 μL,按“2.1.1”项下方法进样测定。结果表明,供试品色谱图中克立硼罗峰形良好,与相邻峰的分离度均>1.5。阴性样品色谱图中在克立硼罗位置未出峰,不干扰测定,说明方法的专属性较好,见图1
IVPT研究中所用接收介质的组成和pH应根据其与皮肤的兼容性以及药物在接受介质中的稳定性和溶解度进行确认,EMA、PMDA要求应采用水或水性缓冲液作为接受介质[6-8],因为克立硼罗水溶性差,故考虑添加温和的表面活性剂,所以筛选0.9%氯化钠溶液、pH 7.4磷酸盐缓冲液、1%吐温80溶液、10%丙二醇溶液,考察各接受液的饱和溶解度,结果见表1
考察IVPT方法改变转速,分别为200、300、400 r·min-1,其他试验参数同“2.1.2”项,采用“2.1.3”“2.1.4”项下方法制备的对照品溶液、供试品溶液,采用“2.1.1”项下条件测定峰面积,分别计算自制品和参比制剂的Jmax,分别将自制品和参比制剂的Jmax均值进行比较,比值均在0.7~1.3范围内;同时将Jmax进行置信区间计算,考察90%置信区间为87.9%~118.7%,说明方法耐用性较好,结果见表2图2
美国FDA和EMA均要求在整个研究过程中,每个皮肤切片表面的温度应保持在32 ℃±1 ℃[5-6,9-10]。故考察体外透皮方法接收液温度,分别为31、32、33 ℃,其他试验参数同“2.1.2”项,采用“2.1.3”“2.1.4”项下方法制备的对照品溶液、供试品溶液,采用“2.1.1”项下条件测定峰面积,分别计算自制品和参比制剂的Jmax,分别将自制品和参比制剂的Jmax均值进行比较,比值均在0.7~1.3范围内;同时将Jmax进行置信区间计算,考察90%置信区间为87.6%~107.8%,说明方法耐用性较好,结果见表2图2
EMA在2018年发布的《Draft Guideline on Quality and Equivalence of Topical Products》[6]中仅推荐使用离体人类皮肤。PMDA规定可以使用离体大鼠、小鼠或猪的皮肤进行试验[7]。USP <1724>中规定,除采用离体人类皮肤外,因猪的皮肤与人类皮肤具有相对相似的形态,可作为研究的次要选择。
猪由于在解剖学、生理学和代谢等方面与人类相似,被广泛应用于生物医学领域。大量文献表明[18-19],猪皮肤的组成、渗透性及体内代谢与人体较为相似。因此,在外用制剂研究过程中,推荐采用猪的皮肤进行IVPT研究,常用的品种有巴马香猪、Landrace猪、Gottingen小型猪及Yorkshire猪等。
美国FDA、EMA与PMDA均要求在试验前需进行皮肤屏障完整性测试[5-7]。可接受的屏障完整性测试方法包括经皮水分散失法(TEWL)、电阻/电导值法。TEWL通常是首选方法,不同于其他方法,TEWL是在不受外部空气影响的条件下,测定水分通过皮肤屏障的量,相对快速和方便[20-21]
实验采用北京德默高科医药技术有限公司和北京和邦兴业科学仪器有限公司采购的皮肤,对同一批样品进行测定,对Jmax进行置信区间计算,考察90%置信区间均在87.9%~102.0%,说明皮肤的适用性良好,皮肤各项检测指标见表3,渗透率结果见表4
按照处方工艺,配制不同浓度的软膏样品,分别含主药为1%、2%、3%,考察不同浓度的受试制剂与参比制剂的渗透率,其他试验参数同“2.1.2”项,采用“2.1.3”“2.1.4”项下方法制备的对照品溶液、供试品溶液,采用“2.1.1”项下条件测定峰面积,分别计算不同处方量样品Jmax的均值并进行比较,结果表明,不同处方量样品,渗透曲线不相似,说明方法具有选择性,灵敏度较高,数据见表5图3
采用“2.1.3”项下方法制备每1 mL约含克立硼罗20 μg的对照品储备液,分别精密量取该储备液1、3、5、7 mL置20 mL量瓶中,加接收液稀释至刻度,摇匀,作为线性1号~4号溶液;精密量取该储备液7 mL置10 mL量瓶中,加接收液稀释至刻度,摇匀,作为5号溶液;量取对照品该储备液作为线性6号溶液。采用“2.1.1”项下条件测定峰面积,以浓度为横坐标,峰面积为纵坐标,绘制线性关系图。克立硼罗质量浓度在0.026~21.02 μg·mL-1范围内,线性方程为

结果显示线性关系良好。
依照“2.1.2”项下试验参数进行试验,按“2.1.3”“2.1.4”项下方法制备对照品溶液、供试品溶液,按“2.1.1”项条件测定峰面积,采用不同仪器、不同人员、不同样品放置位置,分别计算自制品和参比制剂的Jmax,分别将自制品和参比制剂的Jmax均值进行比较,比值均在0.94~1.02范围内;同时将Jmax进行置信区间计算,考察90%置信区间为92.7%~104.6%,说明方法重复性良好,结果见表6
将重复性项下试验后皮上剩余样品收集置50 mL离心管中,用棉签擦净皮肤表面,加入乙腈-水(90:10)20 mL,盖紧离心管,于70 ℃水浴20 min,取出离心管,趁溶液温热时立即涡旋至溶液混合均匀,放置至室温后,以4 000 r·min-1离心5 min,滤过,弃去初滤液,精密量取续滤液3 mL置10 mL量瓶中,加接收液稀释至刻度并混匀,得皮上剩余量样品溶液。将重复性项下试验后皮肤尽量剪碎,置于离心管中,加乙腈-水(90:10)25 mL,超声(频率40 kHz、功率600 W)20 min,过滤,取续滤液,得皮内滞留量样品溶液。依照“2.1.1”项色谱条件,分别精密量取上述各溶液20 µL注入液相色谱仪,记录色谱图。样品的质量守恒=(皮上剩余量+皮内滞留量+接收液总量)/上药量×100%,数据显示,各样品皮内滞留量较少,质量守恒百分比在94.1%~101.1%范围内,说明方法准确度较高,结果见表7
采用“2.1.3”项下方法制备的对照品溶液,分别取自制品软膏(批号220801)、参比制剂软膏(批号FD5696)各约300 mg,置50 mL离心管中,加接收液25 mL,加入表面积约2.5 cm2的猪皮,32 ℃水浴加热30 min,超声(频率40 kHz、功率600 W)20 min,过滤,取续滤液1 mL置50 mL量瓶中,加接收液稀释至刻度,摇匀,作为供试品溶液。依照“2.1.1”项下条件,分别精密量取各溶液20 µL注入液相色谱仪,记录色谱图。分别考察各溶液室温及32 ℃条件下的溶液稳定性,各供试品溶液26 h内,测定峰面积的RSD均在0.44%~1.1%,说明溶液稳定性良好。结果见表8
分别取3批样品各6份,依照“2.1.2”项下试验参数进行试验,按“2.1.3”“2.1.4”项下方法制备对照品溶液、供试品溶液,按“2.1.1”项条件测定峰面积,分别计算自制品和参比制剂的Jmax,分别将自制品和参比制剂的Jmax均值进行比较,比值均在0.92~0.97范围内;同时分别对Jmax进行置信区间计算,考察90%置信区间为84.8%~100.7%,均在EMA要求的80%~125%范围内,结果见表9
IVPT选择的接收液应不能影响皮肤屏障功能,不能添加有机溶剂,本品水溶性较差,经筛选,选用对皮肤影响较小的的表面活性剂吐温80,结果表明,方法专属性较好。
IVPT结果受多种因素影响,本研究针对测定方法中的转速、接收液温度、仪器差异进行耐用性考察,结果表明,方法耐用较好;试验过程需要严格控制皮肤的质量,包括来源、处理方式和保存;减少了外界因素导致的数据差异,试验采用多个供体进行平行试验,采用统计学方法对体外透皮的一致性进行评估,结果显示,自制品与参比制剂渗透率基本一致,符合EMA要求[6],可视为等效,本方法可作为克立硼罗软膏质量评价手段。
本研究开发的分析测定方法为高效液相色谱法,专属性强,方法学验证结果表明本法的精密度良好,重现性较好,准确度较高。
