Objective:

To establish an HPLC-MS method for determination of three diastereoisomers in nirmatrelvir API.

Methods:

The analytical column was an InfinityLab Poroshell SB-C18 (150 mm×3.0 mm,2.7 μm), the two columns connected in series in series. The mobile phase was buffer (0.1% formic acid) (A) -acetonitrile (B), the diluent was water-acetonitrile-methanol (61 :19.5 :19.5). The whole run carried out by gradient elution. The column temperature was 80 ℃,the injection volume was 2 μL, and the sample temperature was 5 ℃.

Results:

Nirmatrelvir was separated completely from three diastereoisomers (the resolution>1.5). The test solution was stable for at least 3 d. The LOQ of diastereoisomer 1 was 0.04%, the LOQ of diastereoisomer 2 was 0.04%, the LOQ of diastereoisomer 3 was 0.05%. The linear correlation coefficient of diastereoisomer 3 was >0.99. The linear range was LOQ-150% of specification. The average recovery (n=9) of diastereoisomer 3 was 97.2%, RSD = 3.1%. The repeatability and intermediate precision completely met the requirements. The three diastereoisomers contents in three batches of nirmatrelvir API 24 months long-term stability test were all completely met the requirements, respectively.

Conclusion:

This method is simple, rapid, sensitive and specific to be used for the determination of three diastereoisomers in nirmatrelvir API.

目的:

建立HPLC-MS法检测奈玛特韦原料药3个非对映异构体。

方法:

采用InfinityLab Poroshell SB-C18(150 mm×3.0 mm,2.7 μm)色谱柱,2根串联使用,以缓冲液(0.1%甲酸溶液)(A)-乙腈(B)为流动相,以水-乙腈-甲醇(61:19.5:19.5)为稀释液,进行梯度洗脱,柱温80 ℃,进样量2 μL,样品温度5 ℃,以电喷雾离子(ESI+)作为离子源,SIM模式扫描。

结果:

奈玛特韦与3个非对映异构体能够完全分离(分离度>1.5),供试品溶液在3 d内稳定性良好;非对映异构体1的定量限为0.04%,非对映异构体2的定量限为0.04%,非对映异构体3的定量限为0.05%;非对映异构体3线性相关系数>0.99,范围为杂质定量限浓度~150%指标浓度;非对映异构体3平均回收率(n=9)为97.2%,RSD为3.1%;重复性和中间精密度符合规定。经检测,3批奈玛特韦原料药24个月长期稳定性3个非对映异构体结果均符合质量标准。

结论:

该方法简便快速,灵敏度高,专属性强,可用于奈玛特韦原料药3个非对映异构体的测定。

目的:

建立HPLC-MS法检测奈玛特韦原料药3个非对映异构体。

方法:

采用InfinityLab Poroshell SB-C18(150 mm×3.0 mm,2.7 μm)色谱柱,2根串联使用,以缓冲液(0.1%甲酸溶液)(A)-乙腈(B)为流动相,以水-乙腈-甲醇(61:19.5:19.5)为稀释液,进行梯度洗脱,柱温80 ℃,进样量2 μL,样品温度5 ℃,以电喷雾离子(ESI+)作为离子源,SIM模式扫描。

结果:

奈玛特韦与3个非对映异构体能够完全分离(分离度>1.5),供试品溶液在3 d内稳定性良好;非对映异构体1的定量限为0.04%,非对映异构体2的定量限为0.04%,非对映异构体3的定量限为0.05%;非对映异构体3线性相关系数>0.99,范围为杂质定量限浓度~150%指标浓度;非对映异构体3平均回收率(n=9)为97.2%,RSD为3.1%;重复性和中间精密度符合规定。经检测,3批奈玛特韦原料药24个月长期稳定性3个非对映异构体结果均符合质量标准。

结论:

该方法简便快速,灵敏度高,专属性强,可用于奈玛特韦原料药3个非对映异构体的测定。

Objective:

To establish an HPLC-MS method for determination of three diastereoisomers in nirmatrelvir API.

Methods:

The analytical column was an InfinityLab Poroshell SB-C18 (150 mm×3.0 mm,2.7 μm), the two columns connected in series in series. The mobile phase was buffer (0.1% formic acid) (A) -acetonitrile (B), the diluent was water-acetonitrile-methanol (61 :19.5 :19.5). The whole run carried out by gradient elution. The column temperature was 80 ℃,the injection volume was 2 μL, and the sample temperature was 5 ℃.

Results:

Nirmatrelvir was separated completely from three diastereoisomers (the resolution>1.5). The test solution was stable for at least 3 d. The LOQ of diastereoisomer 1 was 0.04%, the LOQ of diastereoisomer 2 was 0.04%, the LOQ of diastereoisomer 3 was 0.05%. The linear correlation coefficient of diastereoisomer 3 was >0.99. The linear range was LOQ-150% of specification. The average recovery (n=9) of diastereoisomer 3 was 97.2%, RSD = 3.1%. The repeatability and intermediate precision completely met the requirements. The three diastereoisomers contents in three batches of nirmatrelvir API 24 months long-term stability test were all completely met the requirements, respectively.

Conclusion:

This method is simple, rapid, sensitive and specific to be used for the determination of three diastereoisomers in nirmatrelvir API.

奈玛特韦,一种抗病毒成分,可用于新冠病毒感染治疗。奈玛特韦是新冠病毒治疗药物奈玛特韦片/利托那韦片组合包装(Paxlovid)的组成成分之一[1-9]
目前《中华人民共和国药典》、USP、EP尚无奈玛特韦原料药的质量控制方法。采用高效液相色谱联合质谱法测定奈玛特韦已有文献报道,但并未有方法能够检测3个非对映异构体[10-14]。基于产品质量控制的需求,迫切需要开发非对映异构体检测方法。
质量研究中涉及的物质有活性成分奈玛特韦,工艺杂质非对映异构体1、非对映异构体2、非对映异构体3。杂质信息见表1
非对映异构体1、非对映异构体2、非对映异构体3的含量均≤0.15%。指标制定的依据来源于ICH Q3A[15],界定阈值≤0.15%,鉴定阈值≤0.10%。
非对映异构体1、非对映异构体2在工艺中去除效果较好,不定为已知杂质,进行限度验证,验证内容包括专属性、定量限、检测限;非对映异构体3在工艺中容易残留,定为已知杂质,进行全套方法学验证。
本文建立了奈玛特韦原料药3个非对映异构体的高效液相色谱联合质谱法,该方法重复性好,分离效率高,通过方法学验证,可用于原料药奈玛特韦的质量控制。
Agilent 1260/G6125C(Agilent公司);XPE105十万分之一电子天平(Mettler Toledo公司);色谱柱InfinityLab Poroshell SB-C18(150 mm×3.0 mm,2.7 μm);甲酸(色谱纯,阿拉丁);乙腈(色谱纯,Sigma-Aldrich公司);甲醇(色谱纯,MERCK公司);超纯水(MiLLi-Q纯水仪制备)。
对照品奈玛特韦、非对映异构体1、非对映异构体2、非对映异构体3(以上4个对照品均来自浙江华海药业股份有限公司,批号分别为2022-2177、2023-5065、2023-5064、2023-5171,含量分别为99.5%、95.2%、6.5%、99.1%)。

精密称取非对映异构体1对照品7.5 mg置10 mL量瓶中,加稀释液溶解并稀释至刻度,摇匀,再精密量取1 mL该溶液置20 mL量瓶中,加稀释液稀释至刻度,摇匀,再精密量取1 mL该溶液置25 mL量瓶中,加稀释液稀释至刻度,摇匀,制成每1 mL含1.5 μL非对映异构体1的杂质储备液;同法可制非对映异构体3的杂质储备液;精密称取非对映异构体2对照品5.7 mg置10 mL量瓶中,加稀释液溶解并稀释至刻度,摇匀,再精密量取1 mL该溶液置25 mL量瓶中,加稀释液稀释至刻度,摇匀,制成每1 mL含1.5 μg非对映异构体2的杂质储备液即得。
精密称取奈玛特韦对照品20 mg置20 mL量瓶中,加稀释液溶解并稀释至刻度,摇匀,制成每1 mL含1 mg奈玛特韦的溶液即得。
精密量取奈玛特韦对照品储备液1 mL置20 mL量瓶中,加稀释液稀释至刻度,摇匀,再精密量取1 mL该溶液置100 mL量瓶中,加稀释液稀释至刻度,摇匀,再精密量取3 mL该溶液置20 mL量瓶中,加稀释液稀释至刻度,摇匀,制成每1 mL含75 ng奈玛特韦的溶液即得。
精密称取奈玛特韦原料药20 mg置20 mL量瓶中,加稀释液溶解并稀释至刻度,再精密量取1 mL该溶液置20 mL量瓶中,加稀释液稀释至刻度,摇匀,制成每1 mL含50 μg奈玛特韦的溶液即得。
精密量取“2.2.1”项下非对映异构体1、非对映异构体2的杂质储备液各1 mL,及“2.2.2”项下奈玛特韦对照品储备液1 mL,置于同一20 mL量瓶中,加稀释液稀释至刻度,摇匀,制成每1 mL含奈玛特韦50 μg、非对映异构体1 75 ng、非对映异构体2 75 ng的专属性溶液。因奈玛特韦对照品中已含有非对映异构体3,可不用额外加入。
在“2.1”项色谱条件下,精密量取专属性溶液2 μL注入高效液相色谱联合质谱仪,记录色谱图。奈玛特韦与各杂质分离度均>1.5。空白溶液、专属性溶液图谱见图1
精密量取“2.2.1”项下杂质储备液适量,并进行逐级稀释,初始配制浓度为0.05%,以0.04%、0.03%、0.02%、0.01%浓度往下稀释,最小不低于0.01%。按“2.1”项下色谱条件进样,以信噪比约为10时,各杂质的浓度作为定量限;以信噪比约为3时,各杂质的浓度作为检测限。测定结果见表3
精密量取“2.2.1”项下非对映异构体3储备液10 mL,置20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得母液。精密量取母液1 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得50%指标浓度线性溶液;精密量取母液2 mL,置于25 mL量瓶中,用稀释液稀释至刻度,摇匀,即得80%指标浓度线性溶液;精密量取母液2 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得100%指标浓度线性溶液;精密量取母液3 mL,置于25 mL量瓶中,用稀释液稀释至刻度,摇匀,即得120%指标浓度线性溶液;精密量取母液3 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得150%指标浓度线性溶液;精密量取100%指标浓度线性溶液3 mL,置于10 mL量瓶中,用稀释液稀释至刻度,摇匀,即得杂质定量限浓度线性溶液。
取线性溶液,按“2.1”项下色谱条件进样,记录色谱图。以非对映异构体3浓度为横坐标,峰面积为纵坐标,绘制标准曲线,回归方程为
Y=556.34X+1 746.09   r=0.999 9
线性范围为23.119~115.594 μg·mL-1,结果说明线性关系良好。
精密称取样品(D5448-22-001)20 mg,置20 mL量瓶中,加稀释液溶解并稀释至刻度,摇匀,即得母液。精密量取母液1 mL,置20 mL量瓶中,用稀释液稀释至刻度,摇匀,制成每1 mL含50 μg奈玛特韦的溶液,同法配制6份,按“2.1”项下色谱条件分别各进样1次;结果非对映异构体3含量的RSD(n=6)为2.9%。更换人员、色谱柱、仪器、实验日期,同法配制中间精密度溶液6份,按“2.1”项下色谱条件分别各进样1次;结果非对映异构体3含量的RSD(n=12)为4.9%,表明方法精密度好。
取“2.2.3”项下对照品溶液、“2.2.4”项下供试品溶液、“2.3.1”项下专属性溶液,室温下分别于0、3 d按“2.1”项下色谱条件进样。结果对照品溶液中奈玛特韦3 d与0 d的峰面积比值为1.00,供试品中非对映异构体3的含量无明显变化,专属性溶液中各峰的分离度均>1.5,表明对照品溶液、供试品溶液及专属性溶液在3 d内稳定性良好。
精密称取奈玛特韦对照品30 mg,置于20 mL量瓶中,加稀释液溶解并稀释至刻度,摇匀,精密量取该溶液1 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,再精密量取该溶液1 mL,置于100 mL量瓶中,用稀释液稀释至刻度,摇匀,即得母液。精密量取母液1 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得50%指标浓度准确度溶液;精密量取母液2 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得100%指标浓度准确度溶液;精密量取母液3 mL,置于20 mL量瓶中,用稀释液稀释至刻度,摇匀,即得150%指标浓度准确度溶液。上述每个浓度点的溶液同法配制3份。按“2.1”项下色谱条件进样测定并记录色谱图,计算试验结果,奈玛特韦的平均回收率(n=9)为97.2%(RSD=3.1%),说明方法准确度高。
对不同批次奈玛特韦原料药的长期稳定性样品(放置条件:T=25 ℃±2 ℃,RH=60%±5%;放置时间:0 d、3月、6月、9月、12月、18月、24月),按照“2.2.4”项下方法制备供试品溶液,在“2.1”项下色谱条件进行测定。检测结果见表4,稳定性样品3个非对映异构体结果均未见明显增长。
取不同批次奈玛特韦原料药的样品适量,按“2.2.4”项下方法制备供试品溶液,按“2.1”项下条件进行检测,结果详见表5
由于奈玛特韦结构中存在异构体位点,故在工艺生产过程中易产生非对映异构体,3个非对映异构体及主成分奈玛特韦性质极为相似,常规液相色谱分离手段无法有效将其分离,且结构中无强紫外吸收基团,常规紫外检测器亦无足够响应支持3个杂质的检测;长期稳定性样品检验数据表明奈玛特韦原料药3个非对映异构体测定结果均未见明显增长。本品性质稳定,杂质限度符合要求。
采取常温进样盘考察时,溶液不稳定,故进样盘采取5 ℃控温。
试验中发现高柱温对改善峰形及提高分离度有关键影响,通过摸索,最终确定色谱柱温80 ℃。
分别尝试用Acquity Premier Peptide CSH C18(150 mm×2.1 mm, 1.7 μm)色谱柱和Acquity UPLC Peptide CSH C18(150 mm×2.1 mm, 1.7 μm)色谱柱,以及将二者串联,均无法达到将3个杂质与主成分分离的效果;由于柱温较高,因此方法中选择能耐受高柱温的Poroshell SB-C18色谱柱,且将2根Poroshell SB-C18(150 mm×3.0 mm, 2.7 μm)色谱柱串联,可以将主峰前的3个杂质进行完全分离,且各峰间的分离度>1.5。
由于奈玛特韦与3个非对映异构体紫外吸收较弱,无法用常规高效液相色谱仪配备紫外检测器进行检测,故选择高效液相色谱联合质谱法进行实验。
综合分析,奈玛特韦原料药与涉及的3个非对映异构体差别较小,常规色谱条件较难将其分离,因此,建立能将3个非对映异构体有效分离的分析方法,是方法开发的关键;通过对不同色谱柱、不同柱温以及梯度洗脱方式等色谱条件的反复摸索,建立了奈玛特韦非对映异构体分析方法,并通过3个非对映异构体的加标实验,验证了该分析方法的专属性,保证了该方法在原料药稳定性样品检测过程中的耐用性;本实验对于奈玛特韦杂质进行了检测能力的确认(定量限、检测限),对于工艺中去除效果较好的非对映异构体1和非对映异构体2进行限度验证,对于指标杂质非对映异构体3进行了定量限、检测限、线性关系、回收率等一系列方法学验证,结果证明该分析方法精密度高,重复性好,稳定无干扰,可用于奈玛特韦原料药的质量控制。
