Objective:

To identify the structure and determine the source of unknown impurities present at levels greater than 0.1% in representative pharmaceutical excipient benzalkonium chloride samples, in accordance with ICH Q3A guidelines.

Methods:

A new LC-MS method suitable for the structure prediction of related substances of benzalkonium chloride was established. The separation was performed on an Acclaim 120 C18 column (250 mm×4.6 mm, 5 μm) using a mobile phase consisting of methanol (mobile phase A) and 20 mmol·L-1 ammonium formate aqueous solution (pH 3.5, mobile phase B) with gradient elution at a flow rate was 1 mL·min-1. The post-column split was set to 1 :3. MS data were collected in positive ion mode using an electrospray ionization (ESI) source. The structures of unknown impurities were inferred using a “Diagnostic fragment ion extension strategy” and confirmed by comparing the chromatographic and mass spectrometric behaviors of the impurities with those of reference substances. Additionally, the source attribution and genotoxicity prediction of the detected impurities were performed.

Results:

A total of five unknown impurities were detected in a representative sample of benzalkonium chloride by the newly established LC-MS method, and their structures were inferred. The structures of four impurities were confirmed using two commercially available reference substances and two reference substances synthesized in a directionally oriented manner. Impurity D was identified as N-benzyl-N,N-dimethyl-1-phenylmethanaminium chloride. Impurity E as N-benzyl-N-methyl-1-phenylmethanamine. The impurity G as N-benzyl-N-methyldodecan-1-amine, and impurity H as N,N-dibenzyl-N-methyldodecane-1-ammonium chloride. Impurity F was hypothesized to be an isomer of the formula C15H17N. Predictions using Nexus 2.6.0 software indicated that all the above impurities fell into category 5 and had no genotoxic potential. Furthermore, the method successfully located four unknown impurities detected by the USP method in benzalkonium chloride related substances using impurity reference standards.

Conclusion:

This study systematically examines the structure and safety risks of potential impurities in benzalkonium chloride, providing valuable insights for enhancing quality control standards and pharmacopoeia criteria both domestically and internationally.

目的:

参照ICH Q3A的要求,对代表性药用辅料苯扎氯铵样品中含量>0.1%的未知杂质进行结构鉴定及来源归属。

方法:

运用新建立的适合于苯扎氯铵有关物质结构推定的LC-MS方法,采用Acclaim 120 C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇(A)-20 mmol·L-1的甲酸铵水溶液(pH 3.5) (B)为流动相,梯度洗脱,流速1 mL·min-1,柱后分流(1 :3);采用电喷雾离子源,选择正离子模式进行扫描,采集代表性样品中主成分和未知杂质的质谱信息,借助“诊断碎片离子延伸策略”对未知杂质结构进行推定,通过比较未知杂质与对照品的色谱质谱行为,对未知杂质进行结构确证,并对色谱系统中检出杂质进行来源归属和毒性预测。

结果:

采用新建立的LC-MS方法,在苯扎氯铵代表性样品中共检出5个未知杂质,并推定了结构,利用商业途径获得的2个杂质对照品及定向合成的另外2个杂质对照品对其中4个杂质结构进行了确证,杂质D为二苄基二甲基氯化铵,杂质E为甲基二苄胺,杂质G为N-甲基-N-十二烷基苄胺,杂质H为NN-二苄基-N-甲基十二烷-1-氯化铵,杂质F可能是分子式为C15H17N的同分异构体。Nexus 2.6.0软件预测结果显示,上述杂质均为5类杂质,无基因毒性;此外,还利用杂质对照品,对USP苯扎氯铵有关物质测定方法中检测的4个未知杂质进行了定位。

结论:

本文对苯扎氯铵潜在杂质的结构及安全风险进行了系统研究,对完善国内外药典标准及提高苯扎氯铵的质量控制水平具有指导意义。

目的:

参照ICH Q3A的要求,对代表性药用辅料苯扎氯铵样品中含量>0.1%的未知杂质进行结构鉴定及来源归属。

方法:

运用新建立的适合于苯扎氯铵有关物质结构推定的LC-MS方法,采用Acclaim 120 C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇(A)-20 mmol·L-1的甲酸铵水溶液(pH 3.5) (B)为流动相,梯度洗脱,流速1 mL·min-1,柱后分流(1 :3);采用电喷雾离子源,选择正离子模式进行扫描,采集代表性样品中主成分和未知杂质的质谱信息,借助“诊断碎片离子延伸策略”对未知杂质结构进行推定,通过比较未知杂质与对照品的色谱质谱行为,对未知杂质进行结构确证,并对色谱系统中检出杂质进行来源归属和毒性预测。

结果:

采用新建立的LC-MS方法,在苯扎氯铵代表性样品中共检出5个未知杂质,并推定了结构,利用商业途径获得的2个杂质对照品及定向合成的另外2个杂质对照品对其中4个杂质结构进行了确证,杂质D为二苄基二甲基氯化铵,杂质E为甲基二苄胺,杂质G为N-甲基-N-十二烷基苄胺,杂质H为NN-二苄基-N-甲基十二烷-1-氯化铵,杂质F可能是分子式为C15H17N的同分异构体。Nexus 2.6.0软件预测结果显示,上述杂质均为5类杂质,无基因毒性;此外,还利用杂质对照品,对USP苯扎氯铵有关物质测定方法中检测的4个未知杂质进行了定位。

结论:

本文对苯扎氯铵潜在杂质的结构及安全风险进行了系统研究,对完善国内外药典标准及提高苯扎氯铵的质量控制水平具有指导意义。

Objective:

To identify the structure and determine the source of unknown impurities present at levels greater than 0.1% in representative pharmaceutical excipient benzalkonium chloride samples, in accordance with ICH Q3A guidelines.

Methods:

A new LC-MS method suitable for the structure prediction of related substances of benzalkonium chloride was established. The separation was performed on an Acclaim 120 C18 column (250 mm×4.6 mm, 5 μm) using a mobile phase consisting of methanol (mobile phase A) and 20 mmol·L-1 ammonium formate aqueous solution (pH 3.5, mobile phase B) with gradient elution at a flow rate was 1 mL·min-1. The post-column split was set to 1 :3. MS data were collected in positive ion mode using an electrospray ionization (ESI) source. The structures of unknown impurities were inferred using a “Diagnostic fragment ion extension strategy” and confirmed by comparing the chromatographic and mass spectrometric behaviors of the impurities with those of reference substances. Additionally, the source attribution and genotoxicity prediction of the detected impurities were performed.

Results:

A total of five unknown impurities were detected in a representative sample of benzalkonium chloride by the newly established LC-MS method, and their structures were inferred. The structures of four impurities were confirmed using two commercially available reference substances and two reference substances synthesized in a directionally oriented manner. Impurity D was identified as N-benzyl-N,N-dimethyl-1-phenylmethanaminium chloride. Impurity E as N-benzyl-N-methyl-1-phenylmethanamine. The impurity G as N-benzyl-N-methyldodecan-1-amine, and impurity H as N,N-dibenzyl-N-methyldodecane-1-ammonium chloride. Impurity F was hypothesized to be an isomer of the formula C15H17N. Predictions using Nexus 2.6.0 software indicated that all the above impurities fell into category 5 and had no genotoxic potential. Furthermore, the method successfully located four unknown impurities detected by the USP method in benzalkonium chloride related substances using impurity reference standards.

Conclusion:

This study systematically examines the structure and safety risks of potential impurities in benzalkonium chloride, providing valuable insights for enhancing quality control standards and pharmacopoeia criteria both domestically and internationally.

苯扎氯铵(benzalkonium chloride,BAC)是通式为[C6H5CH2N(CH32R]Cl的烷基苄基二甲基氯化铵的混合物,其中R表示烷基的混合物,主要由n-C12H25(C12-BAC)和n-C14H29(C14-BAC)组成,苯扎氯铵兼具亲水及疏水性,可通过静电吸附和离子交换达到抑菌效果[1-2],因其抗菌谱广,起效快,广泛用于滴眼剂中的抑菌剂[3],并被多国药典收载。
近年来,随着化学原料药及制剂的杂质谱研究的深入,药用辅料的杂质研究也越来越受到关注[4-5],USP-NF(offical as of 01-Aug-2023)[6]和EP 11.2[7]中,均收载了苯扎氯铵中已知杂质A、B、C及检测方法,苯扎氯铵及其已知杂质的化学结构见图1。杂质B和杂质C是国外药典指定严格控制的杂质,但2020年版《中华人民共和国药典》[8]收载的苯扎氯铵品种未设置该检查项,缺少全面控制苯扎氯铵质量的关键一环[9],彭琳等[10]曾报道发现1个未知杂质,但未进行定性研究。日常使用USP方法检测国内苯扎氯铵药用辅料过程中还发现,不同厂家的产品中,除USP规定的3个杂质外,均可检出较多的未知杂质,且不同厂家间未知杂质含量存在明显差异,药用辅料中杂质可能会影响药品的稳定性、安全性和有效性[11-12],故对其杂质谱的系统了解很有必要。但迄今为止,未见对苯扎氯铵中的杂质谱研究的系统报道,本文拟对苯扎氯铵的未知杂质进行分离和结构鉴定。由于目前药用辅料还没有杂质研究的指导原则,研究过程中参考ICH Q3A,对含量≥0.1%的杂质进行研究。
本研究开发了1个LC-MS方法,可实现对苯扎氯铵药用辅料中未知杂质的在线分离和质谱检测,共分离出5个未知杂质,通过比较杂质对照品和样品中杂质的色谱和质谱行为确证了其中4个未知杂质的结构,并将其应用于USP方法中分离出的未知杂质的确认。该研究还推测了未知杂质可能的来源,为提高产品质量控制水平提供一定指导意义。
LC-MS-IT-TOF高效液相色谱(PDA检测器)-离子阱-飞行时间质谱仪(Shimadzu公司);Avance HD核磁共振波谱仪(Bruker公司);Bransonic 3510 EMT超声仪(Branson公司);Milli-Q超纯水机(Millipore公司);DZF-6022真空干燥箱(上海一恒科学仪器有限公司)。
对照品苯甲醇(批号190019-202205,含量100%)、苯甲醛(批号11650-202106,含量99.6%)均购自中国食品药品检定研究院;甲基二苄胺(批号C15363842,含量97%),购自上海麦克林生化科技股份有限公司;十二烷基苄胺(批号DPC988,含量98.8%),购自上海毕得医药科技有限公司;N-甲基-N-十二烷基苄胺(含量97%),购自天津易创成医药技术有限公司;苯扎氯铵(A厂家,批号20230601;B厂家,批号20230602;C厂家,批号20230603;D厂家,批号20230604);色谱级甲醇(ACS公司)。
采用Acclaim 120 C18(250 mm×4.6 mm,5 μm,Thermo Fisher Scientific)色谱柱,以甲醇(A)-20 mmol·L-1的甲酸铵水溶液(甲酸调节pH至3.5)(B)为流动相,梯度洗脱(0~19 min,35%A;19~21 min,35%A→75%A;21~54 min,75%A;54~55 min,75%A→80%A;55~65 min,80%A;65~66 min,80%A→35%A;65~75 min,35%A),流速1 mL·min-1,柱后分流(1 :3),柱温30 ℃,检测波长210 nm,进样体积20 μL。
离子源为ESI源,离子扫描质量范围为m/z 50~500,加热模块温度200 ℃,曲形脱溶剂管(CDL)温度200 ℃,雾化气流速1.5 L·min-1,干燥气体压力94.0 kPa,正离子模式检测。
精密称取苯扎氯铵样品适量,用甲醇溶解并稀释成质量浓度为5 mg·mL-1的溶液,作为供试品溶液,分别按“2”项下条件测定A、B、C和D厂家的各1批样品,其中A厂家产品中检出杂质最多,其色谱图见图2,共得到5个未知杂质的质谱信息,根据出峰顺序依次命名为杂质D~H,苯扎氯铵及其未知杂质的质谱信息和化学结构分别见表12
通过液相色谱定位及质谱定性,并与市售对照品对比鉴定了C12-BAC和C14-BAC的结构。主成分碎片离子可能的裂解模式见图3
C12-BAC在m/z 304.329 1处有[M-Cl]+离子峰,与离子式C21H38N+相应。m/z 212.244 7和m/z 136.112 3是分别失去了1个甲苯(m/z 92)和十二烷基(m/z 168)形成的。
C14-BAC的m/z 332.329 3处有[M-Cl]+离子峰,与离子式C23H42N+相应。m/z 240.266 7和136.111 9是分别失去1个甲苯(m/z 92)和十四烷基(m/z 196)形成的。
使用Formula Predictor预测未知杂质的元素组成,通过已知物质(如C12-BAC等)与未知杂质多级质谱行为的异同性推测结构。未知杂质可能的裂解途径见图4
杂质D可能的裂解模式见图4-D,在m/z 226.157 0处有[M-Cl]+ 离子峰,与离子式C16H20N+相应,较C12-BAC的m/z [M-Cl]+少78,m/z 91.053 0 表明杂质D存在苄基,故杂质D无十二烷基。碎片离子m/z 181.100 3是丢失1个二甲胺并重排形成的,m/z 166.078 8较m/z 181.100 3少1个甲基(m/z 15)。m/z 134.096 3是丢失1个甲苯(m/z 92)形成的。故推断该结构为二苄基二甲基氯化铵,CAS为100-94-7。
杂质E可能的裂解模式见图4-E,在m/z 212.141 4 处有[M+H]+离子峰,与离子式C15H18N+相应,较杂质D的m/z 226.157 0少14,即1个亚甲基。m/z 181.101 1、m/z 91.053 4与杂质D碎片离子相似,提示可能存在2个苄基。而m/z 120.082 4为丢失1个甲苯(m/z 92)形成的,较杂质D的碎片离子m/z 134.096 3少14,表明比杂质D缺少1个亚甲基。故推测结构为甲基二苄胺,CAS为102-05-6。
杂质F可能的裂解模式见图4-F,在m/z 276.266 8处有[M+H]+离子峰,与离子式C19H34N+相应,较C12-BAC的m/z [M-Cl]+少28,说明缺少2个亚甲基。m/z 91.052 7说明杂质F存在苄基,m/z 184.205 8为母离子失去1个甲苯(m/z 92)形成的,较C12-BAC的碎片离子m/z 212.244 7少2个亚甲基(m/z 28)。根据主成分结构推断该杂质为十二烷基苄胺,CAS为1687-68-9。
杂质G可能的裂解模式见图4-G,在m/z 290.282 7处有[M+H]+ 离子峰,与离子式C20H36N+相应,较C12-BAC的m/z [M-Cl]+少14;MS/MS谱图中最丰富的离子位于m/z 198.220 8处,为丢失1个甲苯(m/z 92)形成的,较C12-BAC的碎片离子m/z 212.244 7少14,即1个亚甲基;m/z 91.050 5表明杂质G存在苄基。故推断杂质G的结构为N-甲基-N-十二烷基苄胺,CAS为68397-57-9。
杂质H可能的裂解模式见图4-H,在m/z为380.328 9处有[M-Cl]+离子峰,与离子式C27H42N+相应,较C12-BAC的m/z [M-Cl]+多76,较杂质D母离子多154,即十一烷基,m/z 198.221 3与杂质G碎片离子相似,说明杂质H存在十二烷基;较杂质G多1个苄基(m/z 90)。m/z 134.095 6、m/z 166.076 4、m/z 181.101 2与杂质D碎片离子相似,且m/z 289.270 6是丢失1个甲苯(m/z 92)形成的,198.221 3为丢失1个苄基(m/z 90)并结合了1个氢形成的,说明存在2个苄基。故推断杂质H的结构为NN-二苄基-N-甲基十二烷-1-氯化铵,CAS为108789-53-3。
取对照品NN-二甲基苄胺3.0 g和氯化苄3.0 g置于顶空瓶1,取对照品N-甲基-N-十二烷基苄胺5.0 g和氯化苄2.5 g置于顶空瓶2,密封,常温放置48 h,当顶空瓶中液体变为乳白色凝胶块,即得粗品。将粗品溶于乙酸乙酯中,于4 ℃冰箱重结晶12 h,取乙醚300 mL分3次洗涤过滤,50 ℃真空干燥12 h,即得杂质D和杂质H的精制品。
取“3.4.1”项下制备纯化的杂质精制品,制成杂质质量浓度为0.005 mg·mL-1的甲醇溶液,按“2.1”项下液相色谱条件进样6次,采用峰面积归一化法计算,结果所制备的杂质D纯度(n=6)为97.1%,杂质H纯度(n=6)为97.8%。
红外吸收光谱(IR):取除水干燥后的“3.4.1”项下杂质精制品约5 mg与溴化钾粉末500 mg研磨均匀,压片,置于红外光谱仪中测定。
质谱(MS):取“3.4.1”项下杂质精制品适量,制成杂质质量浓度分别为0.005 mg·mL-1的甲醇溶液,液质联用仪直接进样,以80%甲醇水溶液为流动相,流速0.3 mL·min-1;离子扫描质量范围为m/z 100~600,采用ESI源进行正离子模式和负离子模式扫描。
氢谱(1H-NMR):取“3.4.1”项下杂质精制品各约10 mg,杂质D用氘代氯仿(CDCl3)溶解,杂质H用氘代二甲亚砜(DMSO-d6)溶解,分别转移至5 mm内径的核磁管中。核磁共振波谱仪采用zg30脉冲序列,重复扫描16次,工作频率为400 MHz进行1H-NMR的扫描。
碳谱(13C-NMR):取“3.4.1”项下杂质精制品各约50 mg,分别用DMSO-d6溶解,并转移至5 mm内径的核磁管中。核磁共振波谱仪采用zgpg30脉冲序列,重复扫描512次,工作频率为400 MHz进行13C-NMR的扫描。
IR图见图5-a,3 027.96 cm-1处的中强峰为苯环C-H的伸缩振动吸收峰,2 961.89、2 806.98 cm-1为甲基和亚甲基的伸缩振动峰,1 995.83、1 929.37、1 848.24、1 734.75 cm-1为单取代苯环上的C-H面外弯曲振动吸收峰,1 597.55、1 479.30 cm-1为苯环C=C骨架伸缩振动吸收峰,1 454.24 cm-1为季铵盐的特征吸收峰。
MS图见图5-b,以正离子方式检测的一级全扫描质谱得到m/z 226.160 8 [M-Cl]+,与未知杂质D的一级质谱信息一致;m/z 487.475 8 [2M-Cl]+m/z 489.359 4的丰度比为3 :1,提示有1个氯离子。负离子模式检测得到m/z 296.386 1 [M+Cl]-。因此可以确认该化合物质谱的相对分子质量为261.61,这与二苄基二甲基氯化铵的相对分子质量是一致的。
1H-NMR图见图5-c1H-NMR图谱共出现4组峰(除溶剂峰外),积分比为2 :3 :2 :3。由谱图化学位移值可得δ(7.717,t,2 H)(7.445,m,3 H),苯环上的氢;δ(5.221,s,2 H),连接苯环和氮正离子的亚甲基;δ(3.219,s,3 H),氮正离子相连的甲基。这与Chemdraw软件预测的二苄基二甲基氯化铵的氢谱图一致。
13C-NMR图和碳原子编号分别见图5-d图6,δ:133.683 5(C-5/9/12/16)、130.693 5(C-4/11)、129.329 3(C-6/8/13/15)、128.712 8(C-7/14)、67.093 2(C-3/10)、48.393 8(C-1/2)。这与Chemdraw软件预测的二苄基二甲基氯化铵的碳谱图一致。综上,合成的杂质D即是二苄基二甲基氯化铵。
IR图见图7-a,2 923.47、2 852.84 cm-1为甲基和亚甲基的伸缩振动峰,1 997.69、1 916.95、1 834.05、1 796.75 cm-1为单取代苯环上的C-H面外弯曲振动吸收峰,1 597.55、1 479.30 cm-1为苯环C=C骨架伸缩振动吸收峰,1 454.24 cm-1为季铵盐的特征吸收峰。
MS图见图7-b,以正离子模式检测的一级全扫描质谱得到m/z [M-Cl]+ 380.330 47,与未知杂质H的一级质谱信息一致。负离子模式检测得到m/z 470.327 79,推测[M-Cl]+结合了1个苄基又失去了1个氢。因此可以确认该化合物质谱的相对分子质量为415.78,这与NN-二苄基-N-甲基十二烷-1-氯化铵的相对分子质量是一致的。
1H-NMR图见图7-c,共有9组峰(除溶剂峰外),积分比为4 :6 :2 :2 :2 :3 :2 :18 :3。由谱图化学位移值可得δ:(7.604 4,t,4 H)(7.532 4,t,6 H),苯环上的氢;(4.728 8,t,2 H)(4.575 5,t,2 H),连接吸电子基和苯环的亚甲基;(3.104 0,d,2 H),连接氮正离子的亚甲基;(2.909 8,d,3 H),连接氮正离子的甲基;(1.887 1,s,2 H),连接烷基的亚甲基;(1.257 3,d,18 H),连接烷基的亚甲基;(0.862 6,t,3 H),连接烷基的甲基。这与Chemdraw软件预测的NN-二苄基-N-甲基十二烷-1-氯化铵的氢谱图一致。
13C-NMR图和碳原子编号分别见图7-d图8,δ:133.622 9(C-16/20/23/27),130.697 5(C-15/22),129.388 3(C-17/19/24/26),128.504 8(C-18/25),65.007 9(C-14/21),60.366 5(C-12),46.598 6(C-13),31.765 4(C-11),29.485 1(C-5/6),29.394 0(C-7),29.226 9(C-8),29.191 9(C-9),28.968 3(C-10),26.317 6(C-4),22.571 4(C-3),22.194 7(C-2),14.433 0(C-1)。这与Chemdraw软件预测的NN-二苄基-N-甲基十二烷-1-氯化铵的碳谱图一致。综上,可得合成的杂质H即是NN-二苄基-N-甲基十二烷-1-氯化铵。
将利用商业途径购买的杂质E(甲基二苄胺)、杂质F(十二烷基苄胺)、杂质G(N-甲基-N-十二烷基苄胺)对照品及定向合成的杂质D(二苄基二甲基氯化铵)和杂质H(NN-二苄基-N-甲基十二烷-1-氯化铵)对照品,根据“2.1”和“2.2”项下的条件进样测定,结果表明样品中杂质D、E、G、H和相应杂质对照品的色谱和质谱行为一致,而杂质F不一致,表明杂质F不是十二烷基苄胺。故杂质D为二苄基二甲基氯化铵;杂质E为甲基二苄胺;杂质F可能是分子式为C15H17N的同分异构体;杂质G为N-甲基-N-十二烷基苄胺;杂质H为NN-二苄基-N-甲基十二烷-1-氯化铵。
Lhasa Carcinogenicity Database[13]检索结果显示,杂质A对大鼠和小鼠致癌作用为阴性,杂质B和杂质C对大鼠为阳性;由国际人用药物注册技术要求协调会(ICH M7)[14]可知,杂质B有基因毒性警示结构(芳香醛),为潜在的基因毒性杂质;杂质C在体外具有致突变性和遗传毒性,虽然在哺乳动物体内不具有致突变性和遗传毒性,但其对人体的致癌性较高,为2A类致癌性物质,每日允许暴露量为44 μg。
采用Nexus 2.6.0软件,对新发现的4个未知杂质毒性进行预测,结果表明4个杂质均为5类普通杂质,无基因毒性。
USP方法使用甲醇-50 mmol·L-1磷酸二氢钠水溶液为流动相,该方法中流动相体系与质谱不兼容,且未知杂质不能达到基线分离,因此优化流动相为甲醇-20 mmol·L-1甲酸铵水溶液,各色谱峰峰形较好,分离效果较好。
苯扎氯铵为季铵盐,在溶液状态下易离子化,故离子源选择了电喷雾离子化方法,实验分别对比了正负2种模式对苯扎氯铵及其主要杂质的灵敏度,结果显示,正离子模式灵敏度远强于负离子模式,可能是因为流动相为偏酸性的甲酸铵溶液,提供氢离子,故选择正离子模式检测。实验中分别用正负离子2种模式测定杂质A、杂质B、杂质C对照品,均无质谱信号,可能是由于仪器对相对分子质量在100左右的离子灵敏度不高,故通过杂质对照品与样品中的杂质峰对应确证已知杂质结构。
为归属USP方法中苯扎氯铵的未知杂质,将杂质D、E、G、H对照品根据USP方法进样测定,色谱图见图9,杂质D、E、G、H的保留时间分别为19.412、21.378、48.748、49.727 min,A厂家样品中存在相应保留时间的杂质峰。
本研究采用USP有关物质色谱条件检测了国内4个厂家的苯扎氯铵样品,已知杂质A、B、C按照USP方法计算含量,杂质D、E、G、H除空白和已知杂质外按面积归一化法计算含量,由图10表3可见各厂家杂质含量存在明显差异。C厂家样品总杂质含量最少;A厂家杂质数量和含量最多,其中杂质D、E、G、H含量均超过0.1%,且有基因毒性和致癌性的杂质C含量远远超过USP限度(0.05%)。A厂家对于苯扎氯铵的杂质控制水平较低,生产工艺还需改善。不同杂质校正因子的测定工作还在进行中。
研究还发现各厂家烷基组成比例也存在较大差异,A厂家C12-BAC的含量高达99%以上,实际是苯度氯胺,目前市场上仍存在苯度氯胺和苯扎氯铵混用的情况,故药典应完善和增加有关物质和烷基组成比例检查项以保证产品的安全有效。
研究表明[15-16],季铵盐类化合物被热解时会产生叔胺。未知杂质可能的来源见图11,杂质E和杂质G可能为季铵盐的热解产物,杂质D和杂质H可能为C12-BAC的霍夫曼降解产物2和热解产物杂质G分别与氯化苄发生季铵化反应生成的杂质,相应的降解验证实验还在进行中。叔胺与氯化苄在常温下极易发生季铵化反应得到季铵盐杂质,因此在苯扎氯铵合成中应尽量降低工艺温度并严格控制氯化苄杂质含量以合成质量更高的苯扎氯铵产品。
本文通过液质联用的方法定性苯扎氯铵药用辅料中4个未知杂质的结构,分别为二苄基二甲基氯化铵、甲基二苄胺、N-甲基-N-十二烷基苄胺和NN-二苄基-N-甲基十二烷-1-氯化铵,本研究还推测了未知杂质的可能来源,对苯扎氯铵的产品工艺改进及质量控制水平提高具有借鉴意义且为中国药典标准的完善提供参考。
