Objective:

To establish a simple and efficient method for detecting telomerase activity for evaluating tumorigenic risk of cell products.

Methods:

Specific primers and probes were designed for the conserved domain of telomerase catalytic subunit (TERT), and the primers and probes of TERT gene were optimized and screened, and the primers and probes of internal reference genes were set up. Multiple quantitative PCR reaction was performed using two-color fluorescent probes in a reaction system, and RT-qPCR probe method was established. The expression of TERT gene in human mesenchymal stem cells HMSC was used to determine whether telomerase activity existed in the cells indirectly.

Results:

TERT gene of 293T/17 positive control cells could be stably and specifically detected by this method, with a mean Ct value of 23.96 and a RSD of 1.5. The internal reference gene GAPDH could be detected successfully, the mean Ct value was 14.13, the RSD was 1.3%. The reference gene GAPDH in MRC-5 could be detected in negative control cells, the mean Ct value was 12.81, the RSD was 0.46%, the TERT gene was not detected, and the telomerase activity was negative. The reference gene GAPDH in HMSC could be detected, the Ct mean value was 13.01, and the RSD was 3.8%, whereas, the telomerase activity of HMSC was negative.

Conclusion:

The real-time fluorescent quantitative RT-qPCR probe method established in this study can accurately detect the expression of catalytic subunit mRNA in telomerase positive cells with good repeatability and high specificity. It can be used to analyze telomerase activity of stem cells and indirectly evaluate tumorigenic risk of cell products derived from stem cells.

目的:

建立一种简便、高效的端粒酶活性检测方法,用于评价间充质干细胞产品的成瘤性风险。

方法:

针对端粒酶催化亚基(TERT)的保守结构域设计特异性引物和探针,并对TERT基因的引物和探针进行优化筛选,同时设置内参基因的引物和探针,在一个反应体系内使用双色荧光探针进行多重定量PCR反应,建立RT-qPCR探针法。应用该法对人间充质干细胞中是否表达TERT基因来间接判断细胞中是否存在端粒酶活性。

结果:

该方法可稳定特异地检测到阳性对照细胞293T/17的TERT基因,Ct平均值为23.96,RSD为1.5%。内参基因GAPDH均可以正常检出,Ct平均值为14.13,RSD为1.3%。阴性对照细胞MRC-5内参基因GAPDH可以正常检出,Ct平均值为12.81,RSD为0.46%,TERT基因未检出,该细胞端粒酶活性为阴性。人间充质干细胞HMSC内参基因GAPDH可以正常检出,Ct平均值为13.01,RSD为3.8%,TERT基因未检出,人间充质干细胞HMSC的端粒酶活性为阴性。

结论:

本研究建立的RT-qPCR探针法重复性好,特异性高,能够准确检测端粒酶阳性细胞催化亚基mRNA的转录。可用于间充质干细胞的端粒酶活性分析,间接评估间充质干细胞成瘤性风险。

目的:

建立一种简便、高效的端粒酶活性检测方法,用于评价间充质干细胞产品的成瘤性风险。

方法:

针对端粒酶催化亚基(TERT)的保守结构域设计特异性引物和探针,并对TERT基因的引物和探针进行优化筛选,同时设置内参基因的引物和探针,在一个反应体系内使用双色荧光探针进行多重定量PCR反应,建立RT-qPCR探针法。应用该法对人间充质干细胞中是否表达TERT基因来间接判断细胞中是否存在端粒酶活性。

结果:

该方法可稳定特异地检测到阳性对照细胞293T/17的TERT基因,Ct平均值为23.96,RSD为1.5%。内参基因GAPDH均可以正常检出,Ct平均值为14.13,RSD为1.3%。阴性对照细胞MRC-5内参基因GAPDH可以正常检出,Ct平均值为12.81,RSD为0.46%,TERT基因未检出,该细胞端粒酶活性为阴性。人间充质干细胞HMSC内参基因GAPDH可以正常检出,Ct平均值为13.01,RSD为3.8%,TERT基因未检出,人间充质干细胞HMSC的端粒酶活性为阴性。

结论:

本研究建立的RT-qPCR探针法重复性好,特异性高,能够准确检测端粒酶阳性细胞催化亚基mRNA的转录。可用于间充质干细胞的端粒酶活性分析,间接评估间充质干细胞成瘤性风险。

Objective:

To establish a simple and efficient method for detecting telomerase activity for evaluating tumorigenic risk of cell products.

Methods:

Specific primers and probes were designed for the conserved domain of telomerase catalytic subunit (TERT), and the primers and probes of TERT gene were optimized and screened, and the primers and probes of internal reference genes were set up. Multiple quantitative PCR reaction was performed using two-color fluorescent probes in a reaction system, and RT-qPCR probe method was established. The expression of TERT gene in human mesenchymal stem cells HMSC was used to determine whether telomerase activity existed in the cells indirectly.

Results:

TERT gene of 293T/17 positive control cells could be stably and specifically detected by this method, with a mean Ct value of 23.96 and a RSD of 1.5. The internal reference gene GAPDH could be detected successfully, the mean Ct value was 14.13, the RSD was 1.3%. The reference gene GAPDH in MRC-5 could be detected in negative control cells, the mean Ct value was 12.81, the RSD was 0.46%, the TERT gene was not detected, and the telomerase activity was negative. The reference gene GAPDH in HMSC could be detected, the Ct mean value was 13.01, and the RSD was 3.8%, whereas, the telomerase activity of HMSC was negative.

Conclusion:

The real-time fluorescent quantitative RT-qPCR probe method established in this study can accurately detect the expression of catalytic subunit mRNA in telomerase positive cells with good repeatability and high specificity. It can be used to analyze telomerase activity of stem cells and indirectly evaluate tumorigenic risk of cell products derived from stem cells.

端粒酶是一种核糖核蛋白复合物,主要由端粒酶RNA亚基(hTR)、端粒酶催化亚基(telomerase reverse transcriptase,TERT)和端粒酶相关协助蛋白3个部分组成[1-2]。端粒酶与细胞死亡和癌变之间有着密切的关系[3]。通常在人类正常体细胞中无法检测到端粒酶活性,而在85%~95%的肿瘤细胞和具有自我更新能力的干细胞中均可以检测到端粒酶活性[4-6]
在首次开展临床试验前,需要根据干细胞产品的生物学特性、临床拟用药途径等因素,评估干细胞产品的成瘤性和/或致瘤性风险,其中端粒酶活性检测是评价干细胞产品成瘤性的体外实验之一[7-8]。因此可以通过干细胞的端粒酶活性水平来评估细胞产品的成瘤性风险[9-11]
目前关于端粒酶活性的检测方法主要分为直接检测法和间接检测法2类[12]。1994年Kim等[13]建立了端粒重复片段扩增(telomeric repeat amplification protocol,TRAP)检测法。TRAP法通过PCR技术对端粒酶合成的端粒的DNA进行扩增,该方法应用广泛,且提高了端粒酶活性检测的灵敏度[14-17]。为简化操作步骤,许多改良TRAP法被报道[12]。如TRAP-ELISA法[18]、TRAP-HPA法[19]和TRAP银染法等[12]TERT是端粒酶逆转录酶,其表达与端粒酶活性水平具有相关性[20]。因此可以通过检测TERT基因的mRNA或蛋白表达来间接检测端粒酶活性[12]
本研究使用RT-qPCR技术,通过设计TERT基因片段特异性引物和探针并优化提取和扩增条件,建立了一种间接评价端粒酶活性的方法。该方法使用双色荧光探针分别检测TERT基因和内参基因甘油醛-3-磷酸脱氢酶(glyceraldehyde-3-phosphate dehydrogenase,GAPDH)的mRNA,具有良好的重复性。
MRC-5细胞(人胚肺来源二倍体细胞,CCL-171,购自ATCC公司)、293T/17细胞[SV40T转化的人胚肾细胞(亚系),购自国家生物医学实验细胞资源库]、HeLa细胞[人宫颈癌细胞,CCL-2,购自ATCC公司]、HMSC细胞(人脐带间充质干细胞,PCS-500-010,购自ATCC公司)。细胞培养条件为37 ℃,5% CO2
提取试剂盒为市面上4种品牌的柱式法核酸提取试剂盒,分别为TaKaRa MiniBEST Universal RNA Extraction Kit(9767)(购于宝日医生物技术有限公司)、TIANamp Virus RNA Kit(DP315-R)(购于天根生化科技有限公司)、TIANamp Virus DNA/RNA Kit(DP315)(购于天根生化科技有限公司);FastPure® Viral DNA/RNA Mini Kit Pro(RC323)(购于南京诺唯赞生物科技股份有限公司),随机编号为A、B、C、D。逆转录试剂盒Hifair®Ⅱ1st Strand cDNA Synthesis Kit(gDNA digester plus)购于翌圣生物科技(上海)股份有限公司)AceQ Universal U+ Probe Master Mix V2(Q513)购于南京诺唯赞生物科技股份有限公司。根据NCBI收录的TERT基因(AB085628.1)和GAPDH基因(NC060936.1)的CDS序列,参照引物和探针的设计原则,使用SnapGene软件设计1对GAPDH基因定量引物与探针(GAPDH-F1/R1/P1)作为内参基因;设计6对TERT基因定量引物与探针(TERT-F1/R1/P1至TERT-F6/R6/P6)。每对扩增产物长度约为100 bp。定量引物及探针均由生工生物工程(上海)股份有限公司合成,TERT质粒对照品由北京诺赛基因组研究中心有限公司合成。见表1。DNA稀释液购自北京擎科生物科技股份有限公司荧光定量PCR仪(7500和Quant Studio5)购自赛默飞世尔科技(中国)有限公司。PCR扩增仪(T100)购于BIO-RAD公司。
使用端粒酶阳性细胞系293T/17细胞和端粒酶阴性细胞系MRC-5细胞的cDNA为模板。分别使用6套不同的TERT基因的引物探针对(TERT-F1/R1/P1、TERT-F2/R2/P2、TERT-F3/R3/P3、TERT-F4/R4/P4、TERT-F5/R5/P5、TERT-F6/R6/P6)与内参引物探针对(GAPDH-F1/R1/P1)混合进行扩增,比较不同引物探针检测TERT基因的效率(图1表2)。检测方法流程为cDNA 5 µL(cDNA总量约为4 μg),AceQ Universal U+Probe Master Mix V2 15 µL,10 µmol· L-1TERT定量引物各0.8 µL,10 µmol·L-1TERT探针0.4 µL,10 µmol·L-1GAPDH定量引物各0.8 µL,10 µmol·L-1GAPDH探针0.4 µL,加入ddH2O至30 µL。扩增条件为95 ℃,10 min;95 ℃,15 s;60 ℃,1 min,60 ℃采集荧光信号,共40个循环。
293T/17细胞和MRC-5细胞使用6套混合的引物探针对均可以稳定检出GAPDH基因表达(表2)。293T/17细胞使用TERT-F2/R2/P2引物探针无法检测到TERT基因表达,说明该引物探针不适合本检测方法(图1-B)。293T/17细胞使用TERT-F1/R1/P1,TERT-F3/R3/P3,TERT-F4/R4/P4,TERT-F6/R6/P6引物探针均能够检测到TERT基因表达,Ct值在25.76至29.42范围内,但是MRC-5细胞使用这4组引物和探针会产生非特异性扩增曲线,表明这4组引物探针可能产生了非特异性扩增,不适合该方法(图1-A,C,D,F)。使用TERT-F5/R5/P5引物探针检测端粒酶阳性对照细胞293T/17的TERT基因有明显的扩增曲线,Ct值在26.40至26.91范围内,同时阴性对照MRC-5细胞TERT基因无明显扩增曲线。综上,从扩增效率上和检测特异性上分析,TERT-F5/R5/P5引物探针特异性更好,适用于细胞样品TERT基因表达产物mRNA的检测(图1-E)。
取细胞数为1×106 cells的细胞悬液1 mL,2 000 r·min-1离心5 min,弃上清,立即使用或置于-80 ℃保存备用。分别使用4种核酸提取试剂盒(A、B、C、D)提取293T/17细胞的总RNA。具体操作步骤按照试剂盒说明书进行。使用NanoDrop对抽提的RNA进行浓度和纯度(A260/A280)检测。从总体积50 µL中取RNA模板3 µL进行逆转录。以cDNA为模板,使用内参引物探针对GAPDH-F1/R1/P1和TERT-F5/R5/P5引物探针对混合后进行扩增,通过荧光定量PCR,比较不同核酸提取试剂盒对TERT基因提取效率的影响,选择最合适的核酸提取试剂盒。荧光定量PCR步骤同“2.1”项。
通过荧光定量RT-PCR检测结果显示,GAPDH基因和TERT基因均能够检测到明显的扩增曲线,TERT基因检测为阳性,Ct值在24.74至29.86范围内(如表3所示)。其中试剂盒A、B、C和D提取RNA后检测的TERT基因的Ct值均值分别为24.80、26.38、29.62和25.39。A、B、D这3种试剂盒提取效率相似,优于试剂盒C。本检测方法可使用A、B和D任意1种试剂盒提取RNA,在本研究中,选择了试剂盒A进行了后续方法建立。
分别使用不同浓度(100、200、300、400、500 nmol·L-1)的上下游引物和不同浓度(50、100、150、200、250 nmol·L-1)的探针,对293T/17细胞样品和MRC-5细胞样品进行TERT基因的检测。使用内参引物探针对GAPDH-F1/R1/P1和TERT-F5/R5/P5引物探针对混合后进行扩增,RT-qPCR步骤同“2.1”项。
使用293T/17细胞为模板,固定TERTGAPDH 2个基因的探针终浓度为100 nmol·L-1,比较上下游引物终浓度在100、200、300、400、500 nmol·L-1的Ct值。如表4所示,GAPDH基因均能够稳定检出。TERT基因的上下游引物终浓度为100、200、300、400、500 nmol·L-1时,Ct值分别为25.71,25.67,25.70,25.77,25.95。其中引物浓度在200 nmol·L-1时,Ct值最低,提示在该引物浓度时扩增效率相对其他浓度较高。本研究中上下游引物浓度选择200 nmol·L-1为目的浓度。
使用293T/17细胞为模板,固定TERTGAPDH 2个基因的引物终浓度为200 nmol·L-1,比较探针终浓度在50、100、150、200、250 nmol·L-1 的Ct值。如表5所示,GAPDH基因均能够稳定检出。TERT基因的探针终浓度为50、100、150、200、250 nmol·L-1时,Ct值分别为27.22,26.92,26.89,26.84,26.87。其中探针浓度在200 nmol·L-1 时,Ct值最低,提示在该探针浓度时扩增效率相对其他浓度较高。本研究中上下游探针浓度选择200 nmol·L-1为目的浓度。
(1)阳性对照内参基因Ct值在14±3,TERT基因Ct值在25±3。(2)阴性对照内参基因Ct值在14±3,TERT基因无Ct值或无明显扩增曲线。(3)无模板对照内参基因和TERT基因均无Ct值或无明显扩增曲线。
(1)待测样品内参基因Ct值在14±3,TERT基因Ct≤35且有明显扩增曲线,则样品端粒酶活性为阳性。(2)待测样品内参基因Ct值在14±3,TERT基因无Ct值或无明显扩增曲线,则样品端粒酶活性为阴性。(3)待测样品内参基因Ct值在14±3,TERT基因在临界值35<Ct<40,复检若Ct≥40,则为阴性;复检若35<Ct<40,则为阳性。
检测2种TERT特异性样品(HeLa细胞和293T/17细胞)和2种TERT非特异性样品(HMSC细胞和MRC5细胞),同时检测无模板对照(NTC)样品。采用优化后的反应体系进行检测,每个样本重复检测3次。考察分析方法的专属性。优化后的反应体系为:AceQ Universal U+ Probe Master Mix V2 15 µL,10 µmol·L-1TERT定量引物(F5/R5)各0.6 µL,10 µmol·L-1TERT探针(P5)0.6 µL,10 µmol·L-1GAPDH定量引物(F1/R1)各0.6 µL,10 µmol·L-1GAPDH探针0.6 µL,模板5 µL,加入ddH2O至30 µL。反应程序为95 ℃,10 min;95℃,5 s,60 ℃,1 min,60 ℃采集荧光信号,共45个循环。
结果如表6所示,HeLa细胞与293T/17细胞样品GAPDH基因Ct值在14±3,TERT基因Ct≤35且有明显扩增曲线,表明端粒酶活性为阳性。HMSC与MRC-5细胞样品GAPDH基因Ct值在14±3,TERT基因无Ct值或无明显扩增曲线,表明端粒酶活性为阴性。因此,该检测方法具有良好的专属性。
为验证该检测方法的耐用性,选用不同实时荧光定量PCR仪分别对阳性对照(293T/17)、阴性对照(MRC-5)、待测样品(HMSC)和无模板对照(NTC)进行检测,验证该检测方法的仪器耐用性。RT-qPCR步骤同“2.3.1”项。
仪器耐用性检测结果见表7,ABI 7500和QuantStudio 5对各样品的检测结果无明显差异。阳性对照TERT基因Ct值分别为25.95±0.08和25.10±0.08。待测样品HMSC结果为阴性。因此,当仪器存在一定变化时,该检测方法的检测结果无明显变化。
将稀释至1×1010 copies·μL-1TERT质粒对照品溶液进行倍比稀释(稀释液为DNA稀释液),取1×102~1×106 copies·μL-1质粒对照品溶液作为标准曲线各点,灵敏度对照样品质量浓度为1×10 copies·μL-1,对该检测方法的线性及灵敏度参数进行验证。RT-qPCR步骤同“2.3.1”。结果如图2所示,该检测方法的扩增效率为92.31%。标准曲线样品R2可达0.999,10个重复的灵敏度样品均可以被检测到。因此,该检测方法在1×102~1×106 copies·μL-1范围内具有良好的线性关系,并且灵敏度较高,可达10 copies·μL-1
在3次实验中检测293T/17细胞、MRC-5细胞和供试品HMSC细胞的TERT基因及其内参基因GAPDH。3次重复检测,3种细胞的内参基因GAPDH均有明显的扩增曲线,Ct值在12.34至14.68范围内。其中293T/17细胞的内参基因Ct平均值为14.13,RSD为1.3%;MRC-5细胞的内参基因Ct平均值为12.81,RSD为0.46%;HMSC细胞的内参基因Ct平均值为13.01,RSD为3.8%。阴性对照细胞MRC-5和供试品HMSC细胞均未检测到TERT基因,阳性对照细胞293T/17经过3次重复检测均稳定的检出TERT基因,且Ct值平均值为23.96,RSD为1.5%,说明本方法检测TRET基因的重复性良好(表8)。
研究表明,TERT基因在正常细胞中几乎不表达,但是在大多数类型的肿瘤细胞中表达[21]。陶志华等[22]利用RT-qPCR技术对26例前列腺癌组织和30例良性前列腺增生组织的TERT基因进行检测发现:26例前列腺癌组织中有19例(73%)可以检测到TERT基因;30份良性前列腺增生组织中TERT基因表达均为阴性。此外,TERT基因的表达还与膀胱癌、肝癌、胶质瘤和乳腺癌等的发展具有相关性[23-26]。
干细胞是具有多种分化潜能、自我更新能力和高度增殖能力的细胞[27]。有些干细胞类型具有端粒酶活性,如胚胎干细胞和诱导多能干细胞,以及某些早期代次的成体干细胞。通过对干细胞端粒酶活性进行检测可以评价干细胞来源产品的质量,减少临床应用中的成瘤性风险[9]。人间充质干细胞的(hMSCs)端粒酶活性研究结果不一致,有报道hMSCs端粒酶呈阳性[28],也有报道呈阴性[29-30]。Pittenger等[28]研究表明,hMSCs在12代内仍具有端粒酶活性,但是在他的研究中,典型的阴性对照细胞(人成纤维细胞)也呈端粒酶阳性。Zimmerman等[29]在hMSCs中没能检测到显著水平的端粒酶,这表明hMSCs端粒酶活性或者为阴性,或者低于TRAP-ELISA法的检出限。李静远等[30]使用TRAP法检测了骨髓间充质hMSCs的端粒酶活性,结果为阴性。虽然关于干细胞端粒酶活性的研究结果存在差异,但干细胞端粒酶活性与其成瘤性之间的相关性是被普遍认可的[31]。张彦等[10]测定了淋巴母细胞的端粒酶活性,并通过体内外成瘤实验证明了端粒酶活性与细胞成瘤性之间具有相关性。因此通过分析干细胞制剂的端粒酶活性从而对其成瘤性进行评估是干细胞安全性分析中非常重要的一环。
本研究针对TERT基因建立一种RT-qPCR检测方法。该方法对TERT基因的引物和探针进行优化,在一个反应内使用双色荧光探针法进行多重定量PCR反应,可稳定特异地检测到阳性对照细胞293T/17的TERT基因的转录,重复性良好。因此,本研究建立的方法可应用于干细胞库或干细胞产品端粒酶催化亚基转录水平的检测,进而定性细胞产品的端粒酶活性。
综上所述,本研究建立的RT-qPCR法可以有效地检测端粒酶催化亚基的转录水平。该方法操作简单,特异性良好,可作为干细胞样品端粒酶活性的间接检测方法,有助于体外评价干细胞的成瘤性。
