Objective:

To establish the ultra performance liquid chromatography (UPLC) characterization profile of Styrax, and to determine the contents of three components (cinnamic acid, cinnamyl cinnamate, 3-phenylpropyl cinnamate) in Styrax, the aim was to study the quality of commercially available sulforaphane of different origins and forms, and to provide a reference for the quality control of imported medicinal herbs Styrax.

Methods:

The UPLC method was performed on an Acquity UPLC BEH C18 (100 mm×2.1 mm, 1.7 µm) column with acetonitrile-1% acetic acid solution as the mobile phase in a gradient elution. The volume flow rate was 0.3 mL·min-1. The detection wavelength was 277 nm. and the temperature of the column was 35 ℃. Characteristic chromatograms of styrax were established, and the contents of three major components of Styrax from different sources were determined and the data were statistically analyzed by hierarchical cluster analysis (HCA) and principal component analysis (PCA) with ChemPattern software.

Results:

The results showed that the UPLC characteristic chromatograms of Styrax was established with 10 common peaks and 4 identified components. The linear relationship of 3 components in their respective ranges was good (r≥0. 999), and the average recoveries (n=6) were 101.8% (RSD=1.3%), 105.8% (RSD=1.2%), 99.2% (RSD=1.8%), respectively. The content of 3 components were 2.74%-3.69%, 28.21%-30.63%, 16.89%-20.98%, respectively.

Conclusion:

The method is simple, accurate and reproducible, and can provide an overall quality control basis for the quality standard of Styrax. Combined with the information of origin research, it has important reference significance for the expansion of the origin of Styrax, as well as the harmonization and improvement of the standards of Styrax in different countries.

目的:

建立苏合香药材的超高效液相色谱(UPLC)特征图谱,并测定苏合香中3个成分(肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯)的含量,旨在研究不同产地、不同形态市售苏合香的质量,为进口药材苏合香的质量控制提供参考。

方法:

采用UPLC法,使用Acquity UPLC BEH C18(100 mm×2.1 mm, 1.7 µm)色谱柱,以乙腈-1%乙酸溶液为流动相,梯度洗脱,体积流量0.3 mL·min-1,检测波长277 nm,柱温35 ℃。建立苏合香药材特征图谱,并测定不同来源苏合香药材的3个主要成分的含量,采用ChemPattern软件进行聚类分析(HCA)、主成分分析(PCA)等化学计量方法对苏合香进行质量评价。

结果:

12批苏合香药材特征图谱共确定10个共有峰,指认了4个特征峰;3个成分(肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯)在各自范围内线性关系良好(r≥0.999);平均回收率(n=6)分别为101.8%(RSD=1.3%)、105.8%(RSD=1.2%)、99.2%(RSD=1.8%)。12批苏合香中3个成分含量的分别为2.74%~3.69%、28.21%~30.63%、16.89%~20.98%。

结论:

该方法简便易行,准确度高,重复性好,可为苏合香药材质量标准提供整体质量控制依据;结合产地调研信息,对苏合香基原的扩充以及不同国家苏合香标准的协调、提高具有重要的参考意义。

目的:

建立苏合香药材的超高效液相色谱(UPLC)特征图谱,并测定苏合香中3个成分(肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯)的含量,旨在研究不同产地、不同形态市售苏合香的质量,为进口药材苏合香的质量控制提供参考。

方法:

采用UPLC法,使用Acquity UPLC BEH C18(100 mm×2.1 mm, 1.7 µm)色谱柱,以乙腈-1%乙酸溶液为流动相,梯度洗脱,体积流量0.3 mL·min-1,检测波长277 nm,柱温35 ℃。建立苏合香药材特征图谱,并测定不同来源苏合香药材的3个主要成分的含量,采用ChemPattern软件进行聚类分析(HCA)、主成分分析(PCA)等化学计量方法对苏合香进行质量评价。

结果:

12批苏合香药材特征图谱共确定10个共有峰,指认了4个特征峰;3个成分(肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯)在各自范围内线性关系良好(r≥0.999);平均回收率(n=6)分别为101.8%(RSD=1.3%)、105.8%(RSD=1.2%)、99.2%(RSD=1.8%)。12批苏合香中3个成分含量的分别为2.74%~3.69%、28.21%~30.63%、16.89%~20.98%。

结论:

该方法简便易行,准确度高,重复性好,可为苏合香药材质量标准提供整体质量控制依据;结合产地调研信息,对苏合香基原的扩充以及不同国家苏合香标准的协调、提高具有重要的参考意义。

Objective:

To establish the ultra performance liquid chromatography (UPLC) characterization profile of Styrax, and to determine the contents of three components (cinnamic acid, cinnamyl cinnamate, 3-phenylpropyl cinnamate) in Styrax, the aim was to study the quality of commercially available sulforaphane of different origins and forms, and to provide a reference for the quality control of imported medicinal herbs Styrax.

Methods:

The UPLC method was performed on an Acquity UPLC BEH C18 (100 mm×2.1 mm, 1.7 µm) column with acetonitrile-1% acetic acid solution as the mobile phase in a gradient elution. The volume flow rate was 0.3 mL·min-1. The detection wavelength was 277 nm. and the temperature of the column was 35 ℃. Characteristic chromatograms of styrax were established, and the contents of three major components of Styrax from different sources were determined and the data were statistically analyzed by hierarchical cluster analysis (HCA) and principal component analysis (PCA) with ChemPattern software.

Results:

The results showed that the UPLC characteristic chromatograms of Styrax was established with 10 common peaks and 4 identified components. The linear relationship of 3 components in their respective ranges was good (r≥0. 999), and the average recoveries (n=6) were 101.8% (RSD=1.3%), 105.8% (RSD=1.2%), 99.2% (RSD=1.8%), respectively. The content of 3 components were 2.74%-3.69%, 28.21%-30.63%, 16.89%-20.98%, respectively.

Conclusion:

The method is simple, accurate and reproducible, and can provide an overall quality control basis for the quality standard of Styrax. Combined with the information of origin research, it has important reference significance for the expansion of the origin of Styrax, as well as the harmonization and improvement of the standards of Styrax in different countries.

2020年版《中华人民共和国药典》 (简称《中国药典》)一部规定苏合香(Styrax)为金缕梅科植物苏合香树Liquidambar orientalis Mill.树干渗出的香树脂经加工精制而成,为半流动性的浓稠液体,具有开窍、辟秽、止痛的功效。用于中风痰厥、猝然昏倒、胸痹心痛、胸腹冷痛、惊痫[1]。主要产于土耳其西南部等地。然而,由于环境的变化和山火等问题,苏合香树Liquidambar orientalis Mill.品种濒危,资源紧缺,2011年土耳其海关出口管理制度中明确规定禁止出口苏合香树Liquidambar orientalis Mill.树脂到中国。USP 43规定苏合香(Storax)来源于苏合香树Liquidambar orientalis Mill.和其同属植物北美枫香Liquidambar styraciflua Linné的树脂,前者称为亚洲苏合香(Levant Storax),后者称为美洲苏合香(American Storax),后者主产于中美洲、北美洲的洪都拉斯和墨西哥等地[2-3]。在苏合香基原的问题上,诸多文献认为二者虽然一些成分的含量存在一定的差异,但是化学成分种类相近,认为二者均为苏合香树脂的来源[4-8]。同时,有国内学者对苏合香品种变迁进行了研究,建议可以考虑将北美枫香树Liquidambar styraciflua Linné作为苏合香的第二基原纳入《中国药典》,或者作为“美洲苏合香”纳入《中国药典》[9]
对于资源紧缺的中药材品种,尤其是进口药材,为保障其可持续使用和发展,在中医药基本理论的指导下,结合现代研究成果,以主治类同品种替代、扩大药用部位、近缘品种替代为主寻找濒危中药替代品,是缓解中药资源紧缺压力的重要途径之一[10-11]。在植(动)物长期的演化过程中,亲缘相近的种类由于遗传上的联系,其生理生化特性也相似,特别是次生代谢产物。从基原亲缘相近的品种中寻找替代品,发现新资源,是濒危中药材替代品寻找的重要有效途径之一,是值得大力提倡和研究的,也是中药替代品研究的重要方向。
苏合香大多由芳香族化合物和萜类化合物组成,芳香族化合物主要为桂皮酸及其酯类,其中游离桂皮酸含量为17%~23%,结合桂皮酸含量为24%~25%,酯类包括乙酸肉桂酯、棕榈酸、辛基肉桂酸、苯甲酸苄酯等。萜类化合物包括挥发性的单萜,倍半萜类化合物和三萜类化合物;尚含有部分不饱和脂肪酸,如亚油酸等。孙健等[12]采用气相色谱(GC)法建立特征图谱,并测定肉桂酸-3-苯丙酯的含量,结果表明苏合香真伪品的特征图谱和肉桂酸-3-苯丙酯的含量均具有一定的差异。刘莉等[13]基于苏合香中的挥发性成分,采用GC法同时测定苏合香中的苯甲酸、苯甲酸苄酯、肉桂酸苄酯、肉桂酸肉桂酯4个成分,评价了苏合香的质量。黄晓婧等[14]采用HPLC波长切换法同时测定苏合香中7个具有代表性的化学成分的含量,并且建立苏合香的特征图谱,结合化学计量学统计分析,对苏合香进行整体质量评价。与亚洲苏合香相比,美洲苏合香的研究较少,二者物质基础的研究主要集中在挥发性成分和三萜类成分[15-17]。由于该药材为树脂加工品,生产加工过程不清,且市场上还充斥着固体块状的标识为苏合香的样品,很难通过该树脂来鉴定基原。目前,现有标准无较好的真伪鉴别方法,并且质控指标较为单一,专属性不强,不乏有企业为满足标准而掺伪掺假。因此,本文采用UPLC建立苏合香药材特征图谱,对市售的不同种类的苏合香药材进行比较分析,结合化学计量学统计分析结果和苏合香的成分特点,对肉桂酸(游离肉桂酸)及其酯类成分进行含量测定,对亚洲苏合香和美洲苏合香进行比较,探究美洲苏合香扩充为苏合香基原的可能性,同时为苏合香质量标准的提高提供参考。
Acquity H-Class型超高效液相色谱仪(沃特世公司);KQ-500DE超声仪(昆山市超声仪器有限公司);XS205DU电子天平(十万分之一,梅特勒-托利多仪器公司);ChemPattern化学计量学软件(科迈恩(北京)科技有限公司)。
对照品肉桂酸(批号110786-201604,纯度98.8%,购自中国食品药品检定研究院)、肉桂酸苄酯(批号B25438,纯度≥98%,购自上海源叶生物科技有限公司)、肉桂酸肉桂酯(批号B30311,纯度≥98%,购自上海源叶生物科技有限公司)、肉桂酸-3-苯基丙酯(批号1ST005037,纯度≥98%,购自天津阿尔塔科技有限公司);乙腈为色谱纯(Thermo Fisher),冰乙酸、乙酸乙酯、无水乙醇(分析纯,均购自国药集团化学试剂有限公司);水为Mili-Q纯水(实验室自制)。
苏合香代表性样品见图1,样品具体信息见表1,样品均经中国食品检定研究院魏锋研究员鉴定。
采用Acquity UPLC BEH C18(100 mm×2.1 mm,1.7 µm)色谱柱,以1%乙酸溶液(A)-乙腈(B)为流动相,梯度洗脱(0~5 min, 80%A→75%A; 5~10 min, 75%A→45%A; 10~16 min, 45%A→42%A; 16~20 min, 42%A→35%A),流速0.3 mL·min-1,柱温35℃,检测波长277 nm,进样量1 µL。
称取苏合香约0.5 g,精密称定,置于锥形瓶中,加入乙酸乙酯40 mL,超声(300 W)30 min至完全溶解,放冷,将溶液转移至50 mL量瓶中,用乙酸乙酯定容至刻度。精密吸取1 mL至25 mL量瓶中,用无水乙醇定容至刻度,摇匀,通过0.22 µm滤膜过滤,即得。
精密称取对照品肉桂酸15.99 mg于10 mL量瓶A中,用无水乙醇溶解并定容,再精密吸取2 mL于25 mL量瓶B中;分别精密称取对照品肉桂酸肉桂酯33.13 mg、肉桂酸-3-苯基丙酯29.42 mg于25 mL量瓶B,用无水乙醇溶解并定容,即得混合对照品溶液。
另精密称取对照品肉桂酸苄酯10.34 mg于10 mL量瓶中,用无水乙醇溶解并定容;再精密吸取2 mL于25 mL量瓶中,用无水乙醇定容,得肉桂酸苄酯对照品溶液。
取S3样品,精密称定,按“2.2”项方法制备供试品溶液,按“2.1”项色谱条件连续进样6次,记录色谱图,以肉桂酸峰为参照,计算各特征峰相对保留时间和相对峰面积,结果相对保留时间的RSD为0.06%~0.09%,相对峰面积的RSD为0.39%~0.52%,表明仪器的精密度良好。
取S3样品,精密称定,按“2.2”项方法制备供试品溶液,按“2.1”项色谱条件,分别在0、2、4、8、12、24 h进样测定,以肉桂酸峰为参照,计算各特征峰相对保留时间和相对峰面积,结果相对保留时间的RSD为0.67%~0.95%,相对峰面积的RSD为0.86%~1.8%,表明供试品溶液在24 h内稳定。
取S3样品6份,精密称定,分别按“2.2”项方法制备供试品溶液,按“2.1”项色谱条件进行测定,记录色谱图,以肉桂酸峰为参照,计算各特征峰相对保留时间和相对峰面积,结果相对保留时间的RSD为0.14%~0.26%,相对峰面积的RSD为0.41%~0.59%,表明该方法重复性良好。
取18批苏合香样品,按“2.2”项方法制备供试品溶液,按“2.1”项色谱条件进行测定,记录色谱图。将18批样品特征图谱数据导入ChemPattern软件中,经过保留时间自动对齐校正后,浓稠液态样品(S1~S12)在全谱峰匹配模式下,标记10个共有峰,并生成共有模式对照特征图谱,见图2图3。同时计算相似度,S1、S2样品图谱与对照图谱的相似度均>0.92,S3~S12样品图谱与对照图谱的相似度均>0.99。而固体块状样品(S13~S18)图谱与对照图谱的相似度为0。说明其不仅在性状上不符合标准规定,在成分及含量上与苏合香差异更明显。
对照品指认,确定了苏合香特征图谱中峰2为肉桂酸,峰7为肉桂酸苄酯,峰9为肉桂酸肉桂酯,峰10为肉桂酸-3-苯基丙酯,见图4。以肉桂酸峰(S峰)为参照峰,以参照峰的保留时间,计算S1~S12共计12批苏合香药材中峰7、峰9、峰10的平均相对保留时间分别为2.13(±0.06)、2.52(±0.07)、2.63(±0.07);RSD分别为0.12%、0.13%、0.17%。
采用ChemPattern化学计量学软件对不同产地的18批苏合香样品中10个共有峰峰面积为原始数据进行聚类分析。将样品图谱标准化后,以街区距离为度量,采用加权平均距离法进行聚类分析,见图5。由图5可以看出18批样品中,亚洲苏合香(S1~S2)聚为一类,美洲苏合香(S3~S12)聚为一类,同时6批固态块状样品(S13~S18)单独聚为一类,固态块状样品与浓稠液态样品差异明显。
利用Chem Pattern软件将样品测定数据标准化后进行主成分分析,见图6。第一主成分(PC1)的贡献率为92.19%,第二主成分(PC2)的贡献率为7.60%,第三主成分(PC3)的贡献率为0.20%,累计达99.99%,说明这些成分可代表苏合香特征图谱的绝大部分信息。主成分载荷图给出各色谱峰在主成分中所占的比例,PC1-PC2图中,离X=0坐标轴距离越远的点对PC1的贡献越大,为肉桂酸肉桂酯;而离Y=0坐标轴距离越远的点对PC2的贡献越大,为峰5。PC1~PC3图中离X=0坐标轴距离越远的点对PC1的贡献越大,为肉桂酸肉桂酯;离Y=0坐标轴距离越远的点对PC3的贡献越大,为肉桂酸、肉桂酸-3-苯基丙酯,峰5。综合分析,肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯、峰5为影响不同产地苏合香药材差异性的主要化学成分。
取“2.3”项下混合对照品溶液,用无水乙醇按照倍半稀释法连续稀释,每个对照品对应5个浓度,按“2.1”项下色谱条件进样,以对照品质量浓度(X)为横坐标,峰面积(Y)为纵坐标,作线性回归。回归方程、相关系数、线性范围及定量限结果如表2所示。结果表明,3个成分质量浓度在各自的线性范围内与峰面积呈现良好的线性关系。
取S3样品,精密称定,按“2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件连续进样6次,肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯峰面积的RSD分别为0.46%、0.19%、0.15%,表明仪器的精密度良好。
取S3样品,精密称定,按“2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件,分别在制备后0、2、4、8、12、24 h进样测定,肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯峰面积的RSD分别为0.79%、1.4%、2.0%,表明供试品溶液在24 h内稳定。
取S3样品6份,精密称定,按“2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件进样测定,肉桂酸的平均质量分数为2.78%(RSD=0.41%),肉桂酸肉桂酯的平均质量分数为28.5%(RSD=0.60%),肉桂酸-3-苯基丙酯的平均质量分数为20.5%(RSD=0.44%),表明重复性符合要求。
精密称取已知含量的S3样品0.5 g,加入样品含量100%的各对照品溶液,按“2.2”项下方法制备供试溶液各6份,按“2.1”项下色谱条件进样,肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯的平均回收率分别为101.8%、105.8%、99.2%,RSD分别为1.3%、1.2%、1.8%。表明该方法准确度良好。
取12批苏合香药材,分别按“2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件进样测定,结果见表3
12批苏合香药材中肉桂酸、肉桂酸肉桂酯、肉桂酸-3-苯基丙酯及3个成分的总含量有一定的波动,但波动不大,说明不同产地的苏合香3个成分含量相对较稳定,差异较小;3个代表性成分总含量均值高达51.35%,有足够的代表性能评价苏合香的质量。
本研究在初期考察了供试品溶液的制备方法,首先对提取溶剂进行考察,2020年版《中国药典》一部苏合香性状项下规定:本品在90%乙醇、二硫化碳、三氯甲烷或乙酸中溶解,在乙醚中微溶。通过实验得知,本品在90%的乙醇中溶解性并不好,而在乙酸乙酯中能完全溶解,因此,前处理提取溶剂选择乙酸乙酯。其次对提取方法考察,苏合香为半流动性的浓稠液体,结果显示苏合香在乙酸乙酯中通过超声15 min即可完全溶解,故选取超声作为提取方法。最后选择用不同体积分数(50%、70%、95%、100%)的乙醇作为稀释溶剂,发现无水乙醇对苏合香乙酸乙酯提取液的溶解性最好,同时考虑不同成分的含量水平,最终选取25 mL作为最佳溶剂用量。
对供试品溶液进行200~400 nm的波长扫描,发现肉桂酸和肉桂酸-3-苯基丙酯在277 nm处有最大吸收,肉桂酸肉桂酯的最大吸收波长在269 nm,综合考量各色谱峰的吸收、丰度和分离度,最终确定检测波长为277 nm,与2020年版《中国药典》一部苏合香肉桂酸含量测定项下规定的检测波长(285 nm)有所不同。此外还对流动相(乙腈-水、甲醇-1%乙酸溶液、乙腈-1%乙酸溶液)进行考察,结果发现采用乙腈-水流动相,肉桂酸峰前沿问题比较严重,需要加酸调节流动相pH以获得较好的色谱峰;以甲醇-1%乙酸溶液为流动相,肉桂酸肉桂酯和肉桂酸-3-苯基丙酯的色谱峰分离度未能达到要求(R应不小于1.5),采用乙腈-1%乙酸溶液为流动相,梯度洗脱,各色谱峰分离效果好,峰数多,峰形好,且基线平稳。
本研究共收集到12批浓稠液态的苏合香样品,其中只有2批为亚洲苏合香,由于土耳其早已禁止出口到中国,因此,该基原样品收集十分困难;据市场调研,目前市场上流通的均为来自于北美枫香的美洲苏合香(American Storax)。从特征图谱来看,二者相似度较高,均高于0.92。从含量测定结果来看,2种基原样品的3个主要成分含量测定结果一致性好,肉桂酸平均含量为3.3%;肉桂酸肉桂酯平均含量为29.66%;肉桂酸-3-苯基丙酯平均含量为18.39%。
由于国内使用的苏合香大多数源于进口,并且在国外经过初加工处理,不同产地和不同初加工方式导致苏合香的浓稠度、颜色差异较大,但其内在质量到底是否存在差异,目前尚无文献报道。根据实验结果提示,标识为“苏合香”的固体块状样品不仅性状与国家标准规定差异明显,其成分种类及含量与苏合香样品差异显著;国家相关监督管理部门应加强对此类药品的管理,提升打击力度,切实维护人民用药安全有效。
虽然苏合香现行质量标准中的总肉桂酸含量测定方法可代表肉桂酸酯类成分总量,但其含量限度远低于市场上流通的苏合香药材中肉桂酸的含量,因此本研究针对苏合香中的芳香族化合物中的代表性成分建立特征图谱并结合含量测定能更加全面、更具专属性地评价苏合香的质量。通过研究,一方面建议增加含量测定的指标并修订指标的限度;另一方面建议考虑苏合香药材的资源相关问题,增加苏合香的基原,将同属植物北美枫香树Liquidambar styraciflua Linné作为苏合香的第二基原纳入《中国药典》;同时,研究结果为制定进口药材苏合香的质量标准和促进各国质量标准的协调统一提供参考。
