Objective:

To collect 37 batches of Scrophulariae Radix decoction pieces from the market as the research object, and to establish a quality grade standard of Scrophulariae Radix decoction pieces based on the correlation between quantitative appearance characteristics and intrinsic quality.

Methods:

According to the traditional quality evaluation method, 37 batches of Scrophulariae Radix decoction pieces were divided into two grades—A and B. Their appearance characteristics were quantified while their moisture, ash, leaching and intrinsic quality indexes were detected, respectively. Finally, the independent sample t test analysis, orthogonal partial least squares discriminant analysis (OPLS-DA) and Pearson correlation analysis were carried out to reveal the correlation between appearance characteristics and intrinsic quality.

Results:

On the basis of meeting the quality control standard of Scrophulariae Radix recorded in the 2020 edition of the Pharmacopoeia of the People's Republic of China(referred to as Chinese Pharmacopoeia, ChP), it was found that water-soluble extractives and alcohol-soluble extractives were the key indexes for the grades classification of Scrophulariae Radix decoction pieces. In summary, the lower the chromatic value and the greater the density(ρ)of Scrophulariae Radix decoction pieces, the higher the water-soluble extractives and the lower the alcohol-soluble extractives. The grading quality standard of “selected goods” and “gradeless and uniformly priced goods” of Scrophulariae Radix decoction pieces were preliminarily drawn up: the chromatic value(E*ab)of “selected goods” of Scrophulariae Radix decoction pieces should be less than or equal to 5.5, the ρ of Scrophulariae Radix decoction pieces should be greater than or equal to 1.4 g·cm-3, and the content of water-soluble extractives should be greater than or equal to 77%, the value of E*ab of “gradeless and uniformly priced goods” of Scrophulariae Radix decoction pieces should be between 5.5 and 14, the value of ρ should be between 0.8 and 1.4 g·cm-3, and the content of water-soluble extractives should be greater than or equal to 60%.

Conclusion:

The appearance characteristics of Scrophulariae Radix decoction pieces are related with the internal quality, which is consistent with the traditional quality evaluation method, and it is convenient to scientifically judge the internal quality of Scrophulariae Radix decoction pieces from its appearance characteristics.

目的:

以市场上收集的37批玄参饮片为研究对象,初步建立基于量化的外观性状与内在质量相关联的玄参饮片质量等级评价方案。

方法:

按传统质量评价方法将玄参饮片分别分为A、B两等,在量化A、B两等玄参饮片外观性状指标的同时,开展水分、灰分、浸出物和内在质量指标成分检测,并进行独立样本t检验分析、正交偏最小二乘判别分析(OPLS-DA)和Pearson相关性分析。

结果:

在满足2020年版《中华人民共和国药典》(简称《中国药典》)玄参质控标准的基础上,发现水溶性浸出物和醇溶性浸出物为玄参饮片分类的关键指标,并且玄参饮片色度值越小,密度(ρ)越大,则其水溶性浸出物含量越高,醇溶性浸出物含量越低。初步拟定玄参饮片“选货”和“统货”的等级划分标准:玄参饮片“选货”色泽(E*ab)≤5.5,ρ≥1.4 g·cm-3,水溶性浸出物含量≥77%;玄参饮片“统货”E*ab分布需在5.5~14,ρ大小应位于0.8~1.4 g·cm-3,水溶性浸出物含量≥60%。

结论:

玄参饮片的外观性状与内在质量相关,且符合玄参“大而肉坚黑润者佳”的传统观点,便于从外观性状对玄参饮片的内在质量做出科学合理的判断。

目的:

以市场上收集的37批玄参饮片为研究对象,初步建立基于量化的外观性状与内在质量相关联的玄参饮片质量等级评价方案。

方法:

按传统质量评价方法将玄参饮片分别分为A、B两等,在量化A、B两等玄参饮片外观性状指标的同时,开展水分、灰分、浸出物和内在质量指标成分检测,并进行独立样本t检验分析、正交偏最小二乘判别分析(OPLS-DA)和Pearson相关性分析。

结果:

在满足2020年版《中华人民共和国药典》(简称《中国药典》)玄参质控标准的基础上,发现水溶性浸出物和醇溶性浸出物为玄参饮片分类的关键指标,并且玄参饮片色度值越小,密度(ρ)越大,则其水溶性浸出物含量越高,醇溶性浸出物含量越低。初步拟定玄参饮片“选货”和“统货”的等级划分标准:玄参饮片“选货”色泽(E*ab)≤5.5,ρ≥1.4 g·cm-3,水溶性浸出物含量≥77%;玄参饮片“统货”E*ab分布需在5.5~14,ρ大小应位于0.8~1.4 g·cm-3,水溶性浸出物含量≥60%。

结论:

玄参饮片的外观性状与内在质量相关,且符合玄参“大而肉坚黑润者佳”的传统观点,便于从外观性状对玄参饮片的内在质量做出科学合理的判断。

Objective:

To collect 37 batches of Scrophulariae Radix decoction pieces from the market as the research object, and to establish a quality grade standard of Scrophulariae Radix decoction pieces based on the correlation between quantitative appearance characteristics and intrinsic quality.

Methods:

According to the traditional quality evaluation method, 37 batches of Scrophulariae Radix decoction pieces were divided into two grades—A and B. Their appearance characteristics were quantified while their moisture, ash, leaching and intrinsic quality indexes were detected, respectively. Finally, the independent sample t test analysis, orthogonal partial least squares discriminant analysis (OPLS-DA) and Pearson correlation analysis were carried out to reveal the correlation between appearance characteristics and intrinsic quality.

Results:

On the basis of meeting the quality control standard of Scrophulariae Radix recorded in the 2020 edition of the Pharmacopoeia of the People's Republic of China(referred to as Chinese Pharmacopoeia, ChP), it was found that water-soluble extractives and alcohol-soluble extractives were the key indexes for the grades classification of Scrophulariae Radix decoction pieces. In summary, the lower the chromatic value and the greater the density(ρ)of Scrophulariae Radix decoction pieces, the higher the water-soluble extractives and the lower the alcohol-soluble extractives. The grading quality standard of “selected goods” and “gradeless and uniformly priced goods” of Scrophulariae Radix decoction pieces were preliminarily drawn up: the chromatic value(E*ab)of “selected goods” of Scrophulariae Radix decoction pieces should be less than or equal to 5.5, the ρ of Scrophulariae Radix decoction pieces should be greater than or equal to 1.4 g·cm-3, and the content of water-soluble extractives should be greater than or equal to 77%, the value of E*ab of “gradeless and uniformly priced goods” of Scrophulariae Radix decoction pieces should be between 5.5 and 14, the value of ρ should be between 0.8 and 1.4 g·cm-3, and the content of water-soluble extractives should be greater than or equal to 60%.

Conclusion:

The appearance characteristics of Scrophulariae Radix decoction pieces are related with the internal quality, which is consistent with the traditional quality evaluation method, and it is convenient to scientifically judge the internal quality of Scrophulariae Radix decoction pieces from its appearance characteristics.

玄参为玄参科玄参属植物玄参Scrophularia ningpoensis Hemsl.的干燥根,味甘、苦、咸,性微寒,归肺、胃、肾经,具有清热凉血、滋阴降火、解毒散结之效[1]。玄参作为单基源药材,其道地产区为浙江磐安,为著名的“浙八味”之一。近几十年来,随着各地经济结构调整,湖北、重庆、贵州、湖南等地相继大规模种植玄参,玄参种质混乱、质量良莠不齐的问题层出不穷[2-3]。中药材作为一种特殊商品,辨状论质、按质论价是在市场流通过程中的主要评价手段[4]。随着中药流通市场的演变与发展,中药商品规格等级逐渐成为判别中药材质量优劣的标志,“看货评级,分档议价”的经验方法也大受推崇[5]。然而,无论是1984年我国卫生部制定的《76种中药材商品规格标准》,还是近年来中华中医药学会发布的《中药材商品规格等级》团体标准,都只是规定了玄参药材的规格等级,即按照每1 kg玄参所含的支数,有无空泡,将玄参分为1~3等或统货[6-7],并无相应的法规标准对玄参饮片进行等级划分。
目前,玄参饮片多按照外形、色泽、断面等传统经验来鉴别其质量,但传统评价方法缺乏现代科学技术的支撑,不能客观、有效地评判玄参饮片质量的优劣。同时,以常规质控成分为主导的中药材及其饮片质量等级划分手段虽客观、准确,但因其烦琐、不便捷,在药材流通过程中不具优势。因此,以传统经验鉴别为主导的“辨状论质”更具实用性。“辨状论质”作为中药材及其饮片经验鉴别的精髓,可以根据药材外观性状所表现出来的特点判断中药材及其饮片的优劣真伪[8]。本研究将开展玄参饮片内在成分与传统性状划分之间的相关性研究,旨在为建立基于外观性状及内在成分相关性的玄参饮片等级分类标准提供科学依据,为提升玄参药材、饮片的质量标准水平和市场流通秩序规范化,保障中药用药安全有效起到积极的促进作用。
Agilent 1260高效液相色谱仪(Agilent公司);BSY-200T直立多功能粉碎机(永康市石柱铂欧五金厂);50目不锈钢筛(上海泰坦科技股份有限公司);OSB-2100水浴锅(上海爱郎仪器有限公司);CP313千分之一电子天平(奥豪斯仪器上海有限公司);XSE105十万分之一电子天平(梅特勒-托利多国际贸易有限公司);ADCI-60-C全自动测色色差计(实用型,北京辰泰克仪器技术有限公司);SXL-1008程控箱式电炉(上海精宏实验设备有限公司);DHG 9203A电热鼓风干燥箱(上海一恒科学仪器有限公司);Alpha1-2 LD plus冷冻干燥机(Martin Christ Gefriertrocknungsanlagen GmbH);CQ-100超声波清洗器(上海生析超声仪器有限公司);IP54电子数显卡尺(南京苏测计量仪器有限公司)。
对照品哈巴苷(批号5239)、安格洛苷C(批号5471)、哈巴俄苷(批号10463)和肉桂酸(批号6648)购于上海诗丹德生物技术公司,经HPLC检测纯度均大于98%;磷酸(85%,批号:P1558221,Fisher公司);乙腈(色谱级,上海泰坦科技股份有限公司);甲醇(色谱级,上海泰坦科技股份有限公司);水为娃哈哈纯净水;其余所用试剂均为分析纯,购于国药集团化学试剂有限公司。
通过对玄参道地产区(浙江磐安)及主要产区(湖北恩施等地)所生产的饮片进行实地采集、网络采购、药材市场采购和主要生产企业采购的方式共收集37批玄参饮片,经上海中医药大学李医明教授鉴定均为玄参饮片,具体收集情况见表1
玄参呈类圆柱形,中间略粗或上粗下细,有的微弯曲,长6~20 cm,直径1~3 cm;表面灰黄色或灰褐色,有不规则的纵沟、横长皮孔样突起、稀疏的横裂纹和须根痕;质坚实,不易折断,断面黑色,微有光泽;气特异似焦糖,味甘、微苦[1]。在药材流通市场,玄参以传统鉴别方法进行药材的分级,即按照质量大小、个头大小、是否空泡进行玄参药材规格等级划分。优质玄参根条粗壮、皮细薄、肉肥厚、体重不空泡、质坚性糯、外表灰色,断面碴口乌黑油润[9]。因此,目前虽未出台相应的法规标准对玄参饮片进行质量等级划分,仍可相对应地按照玄参饮片的大小、外观形态、色泽、断面质地以及气味等传统性状特征,对玄参饮片的规格、质量进行等级划分。
根据外观质地差异,对收集到的37批玄参饮片进行初步等级划分——A等玄参饮片质坚性糯,不空泡;B等玄参饮片质空泡,存在较多孔隙。其中,SHYP-11和SHYP-12批次的玄参饮片外观较为统一,不空泡,故全划分为A等,代表性玄参饮片传统分类结果见图1
根据中药传统的“辨状论质”理论,结合2020年版《中华人民共和国药典》(《中国药典》)对玄参的形状描述及历代本草典籍对玄参药材品质的评价沿革,针对优质玄参药材“根条粗壮” “质坚性糯”以及“断面碴口乌黑油润”等特性[9],选取短径长度、密度和色度作为玄参饮片的外观性状指标进行量化测定。
在玄参加工炮制过程中,由于方法和切制角度等方面存在差异,所以各药材厂家生产的玄参饮片的长径长度会存在较大差异,但饮片的厚度差异则较小,故饮片的长径长度和厚度只可作为饮片均匀度指标而不能反映饮片质量优劣;相对而言,药材饮片的短径长度则可更为合理地反映原药材根茎的粗细,故选择短径长度作为玄参饮片的外观性状评价指标。
以2020年版《中国药典》四部0211法中的四分法,对每批玄参样品进行取样,反复数次,最终抽取5 g左右的玄参饮片进行短径长度的测量,每枚饮片平行测量5次后取平均值,最后再计算每枚饮片的平均值。结果显示,A等玄参饮片的短径长度分布在13.759~23.870 mm,B等玄参饮片的短径长度分布在11.054~26.080 mm,见图2
选取适量的100~200目硅胶粉末装于100 mL量筒中,振匀硅胶粉,使其间不存在空隙,记录体积为V1;将随机抽取的5枚玄参饮片,置于电子天平上称取药材质量为M;倾斜量筒,装入随机抽取的5枚玄参饮片,将饮片完全埋于硅胶粉中,直立量筒,振匀硅胶粉末与饮片,使其间不存在空隙,记录体积为V2,则玄参饮片密度ρ(g·cm-3)=M/(V2 - V1)。结果显示,A等玄参饮片的密度分布在1.146~3.341 g·cm-3,B等玄参饮片的密度分布在0.831~2.761 g·cm-3,见图2
CIELAB色度空间技术是基于一种颜色不可能同时既是绿色又是红色、既是蓝色又是黄色的理论而建立起来的,利用L*a*b* 3个不同坐标轴指示颜色在几何坐标图中位置的一种颜色表现技术[10]。其中,L*表示颜色明度,即L*越大则越亮,反之则越黑(当L*=100,a*b*都为0时,为白色;L*=0,a*b*都为0时,为黑色);a*代表红绿方向,a*<0,为绿色方向,a*>0,为红色方向;b*代表蓝黄方向,b*<0,为蓝色方向,b*>0,为黄色方向[11-13]
取玄参饮片粉末(过2号筛)3 g,混合均匀后,平铺于粉末测试盒内,用已进行黑白板矫正的色差仪测试口对准粉末测试盒,测定玄参饮片样品的L*a*b*,随机扫描3次,取平均值,根据公式E*ab = (L*2 + a*2 +b*2)1/2[14]计算样品的色度(E*ab)。结果显示,A等玄参饮片的L*分布在1.26~8.99,a*分布在1.04~8.04,b*分布在-0.05~6.44,E*ab分布在2.42~12.94;B等玄参饮片的L*分布在2.19~11.13,a*分布在1.69~8.00,b*分布在0.55~7.24,E*ab分布在4.56~13.71,见图2
根据2020年版《中国药典》一部玄参项下相关规定,分别按照通则0832第二法测定玄参饮片的水分,通则2302测定玄参饮片的总灰分和酸不溶性灰分,通则2201热浸法测定玄参饮片的水溶性浸出物和醇溶性浸出物,每份样品重复2次。
图3为72批A、B两等玄参饮片的水分、总灰分、酸不溶性灰分、水溶性浸出物和醇溶性浸出物等5项指标的测定结果。其中,A等玄参饮片的水分在7.28%~13.57%,B等玄参饮片的水分在6.15%~13.17%,均小于药典规定的16.0%;A等玄参饮片的总灰分在2.82%~4.60%,B等玄参饮片的总灰分在3.27%~4.68%,均低于药典规定的5.0%;A等玄参饮片的酸不溶性灰分在0.52%~1.20%,B等玄参饮片的酸不溶性灰分在0.62%~1.55%,均低于药典规定的2.0%;A等玄参饮片的水溶性浸出物含量在65.35%~81.34%,B等玄参饮片的水溶性浸出物含量在63.69%~74.31%,均高于药典规定的60.0%;A等玄参饮片的醇溶性浸出物含量在14.61%~22.00%,B等玄参饮片的醇溶性浸出物含量在15.32%~26.20%,均高于文献规定的10.0%[15]。结果显示,72批玄参饮片样品的质量常规检查结果均符合规定。
采用Agilent公司Extend-C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)~0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~8 min,3%A→6%A;8~18 min,6%A→15%A;18~25 min,15%A→20%A;25~35 min,20%A→25%A;35~38 min,25%A→47%A;38~45 min,47%A→75%A;45~50 min,75%A→80%A),流速1 mL·min-1,检测波长210 nm(哈巴苷)、280 nm(哈巴俄苷和肉桂酸)、330 nm(安格洛苷C),柱温30 ℃,进样量20 μL。
取玄参样品粉末(过3号筛)约0.5 g,精密称定,置具塞锥形瓶中,精密加入50%甲醇水溶液50 mL,密塞,称定,浸泡1 h,超声(功率500 W,频率40 kHz)处理45 min,放冷,50%甲醇水溶液补足减失的量,摇匀,过滤,取续滤液,过0.22 μm微孔滤膜,即得。
分别取哈巴苷、安格洛苷C、哈巴俄苷和肉桂酸的对照品适量,精密称定,用30%甲醇水溶液配制成每1 mL含哈巴苷0.497 mg、安格洛苷C 0.167 mg、哈巴俄苷0.183 mg、肉桂酸0.063 mg的混合对照品溶液。
图4可知,HPLC图谱中哈巴苷(1)、安格洛苷C(2)、哈巴俄苷(3)和肉桂酸(4)出峰时间稳定,峰形佳,分离度良好,均大于1.5,专属性良好。
分别精密吸取“2.4.3”项下的混合对照品溶液,用30%甲醇水溶液依次按照1:1、2:3、1:1、1:1、2:3、1:1、1:1的比例稀释,按“2.4.1”项下的色谱条件进行测定,以对照品质量浓度X为横坐标,色谱峰峰面积Y为纵坐标,绘制标准曲线并进行线性回归,结果见表2
按信噪比S/N=3和S/N=10时分别计算哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸的检测限(limit of detection,LOD)和定量限(limit of quantification,LOQ),结果见表2
精密吸取“2.4.3”项的混合对照品溶液,按“2.4.1”项下的色谱条件重复进样6次,测定峰面积并计算RSD。结果哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸峰面积的RSD(n=6)分别为0.57%、0.061%、0.064%、0.25%,表明仪器精密度良好。
按“2.4.2”项下方法制备供试品溶液(样品批号SHYP-13-A),分别在0、3、6、9、12、15、18、21、24 h,按“2.4.1”项下的色谱条件进样测定峰面积,并计算RSD。结果哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸峰面积的RSD分别为0.46%、0.40%、0.37%、2.59%,表明供试品溶液在24 h内稳定性良好。
精密称取同一批号样品(批号SHYP-13-A)6份,分别按“2.4.2”项下方法制备供试品溶液,并在“2.4.1”的色谱条件下进样测定峰面积,分别计算含量和RSD。结果哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸的含量值分别为7.025、1.246、0.875、1.222 mg·g-1,RSD分别为0.44%、0.80%、0.82%、0.71%,表明该方法的重复性良好。
取已知含量的玄参粉末样品(批号SHYP-13-A)50.00 mg共6份,精密称定,分别按已知含量的100%浓度水平精密加入对照品溶液,再加入50%甲醇水溶液。密塞,称定,浸泡1 h,超声(功率500 W,频率40 kHz)处理45 min,放冷,用50%甲醇水溶液补足减失的量,摇匀,滤过,取续滤液,过0.22 μm微孔滤膜,即得加样回收试液。按“2.4.1”项下色谱条件进样测定峰面积,计算加样回收率和RSD。结果哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸的平均加样回收率分别为99.3%、99.3%、101.3%、103.0%,RSD分别为2.4%、2.6%、3.0%、0.85%,表明该方法准确度良好。
按“2.4.2”项下方法制备72批玄参饮片的供试品溶液,并按“2.4.1”项下色谱条件进样测定峰面积,计算各批次玄参饮片中哈巴苷、安格洛苷C、哈巴俄苷、肉桂酸的含量,每份样品重复2次,结果见图5。批号为SHYP-3-A、SHYP-4-A、SHYP-6-A、SHYP-16-A和SHYP-16-B的样品中,哈巴苷和哈巴俄苷总量没有达到药典所规定的下限(0.45%),因此,在后续的分析过程中剔除相应不合格玄参饮片样品批次(SHYP-3、SHYP-4、SHYP-6及SHYP-16)所对应的A、B两等玄参饮片样品。
将目前获得的33批合格玄参饮片样品的外观性状及内在质量数据进行汇总和整理,即33组A等玄参饮片和31组B等玄参饮片,进行后续研究。
利用SPSS 26.0软件对64组玄参饮片的外观性状指标和内在质量指标进行独立样本t检验,分析A等和B等玄参饮片的短径长度、密度、色度、水分、总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、哈巴苷、安格洛苷C、哈巴俄苷和肉桂酸含量的差异性,结果如表3所示,A等和B等玄参饮片的密度、色度、总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、安格洛苷C、哈巴俄苷和肉桂酸含量差异具有统计学意义(P<0.05),而A等和B等玄参饮片的短径长度、水分和哈巴苷含量差异不存在统计学意义(P>0.05)。因此,考虑将密度、色度、总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、安格洛苷C、哈巴俄苷和肉桂酸含量作为玄参饮片等级划分的分级指标。
正交偏最小二乘判别分析(orthogonal partial least squares discrimination analysis,OPLS-DA)是一种有监督的判别分析统计方法,是在已知样品分组关系的前提下,对数据进行降维处理,可以将不同组别样品的差异最大化,从而更好地筛选出区分样品的指标成分[16-17]
将7个在A等和B等玄参饮片中存在显著性差异的内在质量指标(总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、安格洛苷C含量、哈巴俄苷、肉桂酸含量)数据导入Simca 14.0软件中,采用OPLS-DA统计法对其进行分析,结果见图6。该模型的R2X=0.549,R2Y=0.792,说明该模型对变量X的解释能力为54.9%、对因变量Y的解释能力为79.2%,Q2 = 0.741则表示该模型的预测能力较为理想。通过OPLS-DA分析,A等和B等玄参饮片可以明显分为2组,这表明A等和B等的玄参饮片在总灰分、酸不溶性灰分及水溶性浸出物、醇溶浸出物、安格洛苷C、哈巴俄苷和肉桂酸含量方面存在一定的差异,而这7个成分指标的差异可以将A等和B等的玄参饮片分为2组。图7中的变量投影重要度(variable importance for the projection,VIP)代表了每个指标在对每组样品进行区分时所做出的贡献程度,并且VIP>1的指标在对样品进行分类时起到主导作用[16]。因此,可得知水溶性浸出物含量(VIP=1.341)和醇溶性浸出物含量(VIP=1.213)这2个指标在对A等和B等的玄参饮片进行分类时起到关键性作用,可为后续筛选玄参饮片等级划分的外在性状指标提供依据。
为了进一步探究玄参饮片的外观性状与内在质量之间的相关性,将玄参饮片的外观性状指标和内在质量指标导入SPSS 26.0软件进行两两相关的Pearson相关性分析,结果见图8。表和图中的数字为这2个指标之间的Pearson相关系数,数值为负数代表负相关,数值为正数代表正相关,*代表2个指标之间有显著性关系(P<0.05),**代表2个指标之间的关系极为显著(P<0.01)。结果显示,玄参饮片的短径长度与酸不溶性灰分、安格洛苷C含量呈显著负相关,与哈巴俄苷含量呈极显著负相关,表明玄参饮片越大,则酸不溶性灰分及安格洛苷C、哈巴俄苷含量越低;玄参饮片的密度与醇溶性浸出物含量呈显著负相关,与总灰分、安格洛苷C含量呈极显著负相关,与水溶性浸出物含量呈显著正相关,表明玄参饮片越“质坚”,则其水溶性浸出物含量越高,而醇溶性浸出物含量、总灰分、安格洛苷C含量越低;玄参饮片的色度与酸不溶性灰分呈显著正相关,与醇溶性浸出物、哈巴苷含量呈极显著正相关,与水分和水溶性浸出物含量呈极显著负相关,表明玄参饮片色度越暗,即色泽越黑,其水分和水溶性浸出物含量越高,而其酸不溶性灰分及醇溶性浸出物、哈巴苷的含量则越低。从相关性分析结果可知,玄参饮片的外观性状(短径长度、密度及色度)与玄参饮片的内在指标(水分、总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、安格洛苷C和哈巴俄苷的含量)存在着一定的显著相关性,结合独立样本t检验和OPLS-DA结果,选择密度和色度作为快速辨别玄参饮片内在质量的外观性状指标。同时,密度、色度与水溶性浸出物含量的显著性关系最符合玄参“大而肉坚黑润者佳”的传统经验描述。因此可知,通过对玄参饮片进行快速的外观性状考察,可以有效地评价其内在质量的好坏。
结合上述的分析结果,最终选择密度和色度作为快速辨别玄参饮片内在质量的外观性状指标,并将与二者存在显著相关性的水溶性浸出物含量作为玄参饮片“统货”和“选货”的关键划分指标。结合行业内药材分级情况和实际应用的可行性,以玄参饮片样品数前20%的比例要求制定标准,本研究拟定玄参饮片“选货”和“统货”的等级划分标准,具体见表4。其中最低标准参照2020年版《中国药典》和该检查项实验数据最小值。
明代《药品化义》记载,玄参为“取大而肉坚黑润者佳”[18];1963年版《中国药典》评价玄参“以支条肥大、皮细、质坚、芦头修尽,内色乌黑者为佳,支条小、皮粗糙、带短芦者质次,支条很小、芦头大、折断显柴性者不可入药。”[19];《中华本草》描述玄参“以条粗壮、质坚实、断面色黑者为佳”[20]。可见,历代对于优质玄参的划分均以个头大、质坚实、断面黑润为佳。因此,为了与传统优质玄参划分标准相一致,本课题在探究玄参饮片的等级划分标准时,首先对多途径收集到的37批玄参饮片进行简单的基于外观的分类,并对其外观性状——短径长度、密度和色度,展开量化研究。
在质控成分方面,2020年版《中国药典》规定玄参药材、饮片均仅以哈巴苷和哈巴俄苷为指标性成分,但前期研究和大量的文献报道表明,玄参中的安格洛苷C和肉桂酸在抗氧化、抗炎等多方面有显著的活性[21-22]。因此,本研究将哈巴苷、安格洛苷C、哈巴俄苷和肉桂酸这4个活性成分作为探讨玄参饮片等级的指标性成分,同时依据2020年版《中国药典》玄参饮片的质控标准,剔除不达标的玄参饮片。OPLS-DA结果表明,水溶性浸出物和醇溶性浸出物的含量是对玄参等级起到关键分类作用的2个内在指标,尤其以水溶性浸出物的含量与中药煎煮过程紧密相关,因此本研究着重将水溶性浸出物含量作为玄参饮片“统货”和“选货”的关键内在划分指标。
最后,对64批玄参饮片进行外观性状与内在质量指标的相关性分析。结果显示,玄参饮片的外观性状(短径长度、密度及色度)与玄参饮片的内在指标(水分、总灰分、酸不溶性灰分及水溶性浸出物、醇溶性浸出物、哈巴苷、安格洛苷C和哈巴俄苷的含量)存在着一定的显著相关性,并且尤以密度和色度与水溶性浸出物含量的显著性关系最符合玄参“大而肉坚黑润者佳”的传统经验描述。
综上,将玄参饮片的密度和色度作为快速辨别饮片质量优劣的外在性状特征,并以与其存在显著相关性的水溶性浸出物含量作为关键的内在质量等级划分指标。较传统的等级标准而言,此类等级划分标准研究能够充分考虑到药材饮片的外观性状与内在质量之间的相关性,更符合实际情况,结果也更有科学性,有助于日常中药材流通过程中对玄参饮片的优质优价做出快速的等级划分指导。
