Objective:

To establish a multiple fingerprint analysis method for polysaccharides from Sanghuangporus sanghuang, providing a reference for quality evaluation.

Methods:

High-performance gel filtration chromatography(HPGFC-RID) was used with a TSK-GEL®G3000 PWXL column (7.8 mm×30 cm, 7 μm). The mobile phase was 20 mmol·L-1 HAc-NaAc buffer (pH 5.7) at a flow rate of 0.5 mL·min-1. The injection volume was 15 μL, column temperature was 35 ℃, and detection was carried out using a RID detector. Fourier transform infrared(FT-IR) spectroscopy was performed in the range of 4 000 to 400 cm-1 with a resolution of 4 cm-1 and 16 scans. HPLC-UV monosaccharide profiling was done using an Agilent 5 HC-C18 column (250 mm×4.6 mm, 5 μm), with acetonitrile-0.02 mol·L-1 ammonium acetate (20:80) as the mobile phase, at a flow rate of 1 mL·min-1, detection wavelength of 250 nm, an injection volume of 10 μL, and column temperature of 35 ℃. An UV detector was used for detection. Multiple fingerprints of Sanghuangporus polysaccharides were established, and the mass average molar mass, characteristic absorptive functional groups, and monosaccharide compositions of 29 batches of Sanghuangporus polysaccharides were compared and analyzed for their intraspecific and interspecific variations by combining with chemometrics and principal component analysis (PCA).

Results:

HPGFC-RID monosaccharide profiling revealed that the polysaccharides from Sanghuangporus sanghuang (SH), Sanghuangporus vaninii (YH), Sanghuangporus baumii (BH), and Phellinus pini (SHH) all exhibited two major chromatographic peaks (P1, P2). For P1, BH polysaccharides had the highest molecular weight, followed by SSH, SH, and YH polysaccharides. For P2, YH polysaccharides had the highest molecular weight, followed by SSH, SH, and BH polysaccharides. FT-IR analysis indicated that SH, YH, BH, and SSH polysaccharides shared similar infrared absorption peaks, with no significant differences between species or within species. However, the intensity of the main uptake peaks in the range of 1 800-900 cm-1 was different between bagged and linden cultivated Sanghuangporus polysaccharides. HPLC-UV monosaccharide profiling combined with chemometrics revealed that Sanghuangporus polysaccharides consist of mannose, rhamnose, glucose, xylose, and fucose. Glucose content was the highest in SH, YH (linden cultivation), BH, and SSH polysaccharides, while xylose was the highest in YH (bagged cultivation). Monosaccharide composition varied significantly among different Sanghuangporus varieties. PCA grouped YH (bagged cultivation) as one class, and SH, YH (linden cultivation), BH, and SSH as another, indicating that cultivation methods also influence Sanghuangporus quality.

Conclusion:

The multiple fingerprint analysis method for Sanghuangporus polysaccharides effectively evaluates the quality of different Sanghuangporus varieties, providing a foundation for quality control.

目的:

建立桑黄多糖多重指纹图谱法,为桑黄质量评价提供参考。

方法:

结果:

HPGFC-RID糖谱分析显示,桑树桑黄(SH)、杨树桑黄(YH)、暴马桑黄(BH)和松木层孔菌(松树桑黄)(SSH)多糖均有2个主要色谱峰(P1、P2)。对于主要色谱峰P1,BH多糖的重均分子量最高,其次依次为SSH、SH和YH多糖;对于主要色谱峰P2,YH多糖的重均分子量最高,其次依次为SSH、SH和BH多糖。FT-IR糖谱分析表明,SH、YH、BH和SSH多糖具有相似的红外光谱吸收峰,种内、种间差异不明显,但袋料栽培与椴木栽培的桑黄多糖在1 800~900 cm-1范围内主要吸收峰强度不同。HPLC-UV糖谱结合化学计量学分析显示,桑黄多糖由甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖组成,其中SH、YH(椴木栽培)、BH和SSH多糖中葡萄糖含量最高,YH(袋料栽培)多糖中木糖含量最高。各单糖组成分布结果表明,不同品种桑黄单糖含量存在明显差异。此外,PCA将YH(袋料栽培)聚为一类,SH、YH(椴木栽培)、BH和SSH聚为一类,说明栽培方式的不同也会影响桑黄的质量。

结论:

桑黄多糖多重指纹图谱分析方法可对不同品种桑黄质量进行有效评价,为桑黄质量控制奠定基础。

目的:

建立桑黄多糖多重指纹图谱法,为桑黄质量评价提供参考。

方法:

采用高效凝胶色谱法(HPGFC-RID法),色谱柱为TSK-GEL®G3000 PWXL(7.8 mm×30 cm,7 μm),以20 mmol·L-1 醋酸-醋酸钠缓冲液(pH 5.7)为流动相,流速0.5 mL·min-1,进样量15 μL,柱温35 ℃,RID检测器;采用红外光谱法(FT-IR法),测定范围为4 000~400 cm-1,分辨率4 cm-1,扫描16次;采用HPLC-UV糖谱法,色谱柱为Agilent 5 HC-C18(2)(250 mm×4.6 mm,5 μm),以乙腈-0.02 mol·L-1乙酸铵(20:80)为流动相,流速1 mL·min-1,检测波长250 nm,进样量10 μL,柱温35 ℃,UV检测器。建立桑黄多糖多重指纹图谱,对29批桑黄多糖的重均分子量、特征吸收官能团、单糖组成进行比较,并结合化学计量学和主成分分析(PCA),分析其种内及种间差异。

结果:

HPGFC-RID糖谱分析显示,桑树桑黄(SH)、杨树桑黄(YH)、暴马桑黄(BH)和松木层孔菌(松树桑黄)(SSH)多糖均有2个主要色谱峰(P1、P2)。对于主要色谱峰P1,BH多糖的重均分子量最高,其次依次为SSH、SH和YH多糖;对于主要色谱峰P2,YH多糖的重均分子量最高,其次依次为SSH、SH和BH多糖。FT-IR糖谱分析表明,SH、YH、BH和SSH多糖具有相似的红外光谱吸收峰,种内、种间差异不明显,但袋料栽培与椴木栽培的桑黄多糖在1 800~900 cm-1范围内主要吸收峰强度不同。HPLC-UV糖谱结合化学计量学分析显示,桑黄多糖由甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖组成,其中SH、YH(椴木栽培)、BH和SSH多糖中葡萄糖含量最高,YH(袋料栽培)多糖中木糖含量最高。各单糖组成分布结果表明,不同品种桑黄单糖含量存在明显差异。此外,PCA将YH(袋料栽培)聚为一类,SH、YH(椴木栽培)、BH和SSH聚为一类,说明栽培方式的不同也会影响桑黄的质量。

结论:

桑黄多糖多重指纹图谱分析方法可对不同品种桑黄质量进行有效评价,为桑黄质量控制奠定基础。

Objective:

To establish a multiple fingerprint analysis method for polysaccharides from Sanghuangporus sanghuang, providing a reference for quality evaluation.

Methods:

High-performance gel filtration chromatography(HPGFC-RID) was used with a TSK-GEL®G3000 PWXL column (7.8 mm×30 cm, 7 μm). The mobile phase was 20 mmol·L-1 HAc-NaAc buffer (pH 5.7) at a flow rate of 0.5 mL·min-1. The injection volume was 15 μL, column temperature was 35 ℃, and detection was carried out using a RID detector. Fourier transform infrared(FT-IR) spectroscopy was performed in the range of 4 000 to 400 cm-1 with a resolution of 4 cm-1 and 16 scans. HPLC-UV monosaccharide profiling was done using an Agilent 5 HC-C18 column (250 mm×4.6 mm, 5 μm), with acetonitrile-0.02 mol·L-1 ammonium acetate (20:80) as the mobile phase, at a flow rate of 1 mL·min-1, detection wavelength of 250 nm, an injection volume of 10 μL, and column temperature of 35 ℃. An UV detector was used for detection. Multiple fingerprints of Sanghuangporus polysaccharides were established, and the mass average molar mass, characteristic absorptive functional groups, and monosaccharide compositions of 29 batches of Sanghuangporus polysaccharides were compared and analyzed for their intraspecific and interspecific variations by combining with chemometrics and principal component analysis (PCA).

Results:

HPGFC-RID monosaccharide profiling revealed that the polysaccharides from Sanghuangporus sanghuang (SH), Sanghuangporus vaninii (YH), Sanghuangporus baumii (BH), and Phellinus pini (SHH) all exhibited two major chromatographic peaks (P1, P2). For P1, BH polysaccharides had the highest molecular weight, followed by SSH, SH, and YH polysaccharides. For P2, YH polysaccharides had the highest molecular weight, followed by SSH, SH, and BH polysaccharides. FT-IR analysis indicated that SH, YH, BH, and SSH polysaccharides shared similar infrared absorption peaks, with no significant differences between species or within species. However, the intensity of the main uptake peaks in the range of 1 800-900 cm-1 was different between bagged and linden cultivated Sanghuangporus polysaccharides. HPLC-UV monosaccharide profiling combined with chemometrics revealed that Sanghuangporus polysaccharides consist of mannose, rhamnose, glucose, xylose, and fucose. Glucose content was the highest in SH, YH (linden cultivation), BH, and SSH polysaccharides, while xylose was the highest in YH (bagged cultivation). Monosaccharide composition varied significantly among different Sanghuangporus varieties. PCA grouped YH (bagged cultivation) as one class, and SH, YH (linden cultivation), BH, and SSH as another, indicating that cultivation methods also influence Sanghuangporus quality.

Conclusion:

The multiple fingerprint analysis method for Sanghuangporus polysaccharides effectively evaluates the quality of different Sanghuangporus varieties, providing a foundation for quality control.

桑黄始载于《神农本草经》,名曰“桑耳”,为桑黄孔菌属真菌桑黄Sanghuangporus sanghuang (Sheng H. Wu, T. Hatt & Y.C.Dai) Sheng H. Wu.的子实体[1],具有活血止血、化饮、止泻之功效[2]。现代研究表明,桑黄中的活性成分有桑黄多糖、黄酮类、萜类、多肽类以及微量元素,在肿瘤、炎症、氧化应激和高血糖等多种疾病的治疗中体现出很好的药用价值[3-6]。桑黄孔菌属真菌分布广泛,多产于中国的东北、华北、华东等地[7]。但由于生长环境和生长年限的制约,野生桑黄难以满足市场的需求,如今多通过人工栽培来解决资源短缺的问题[8]。目前市场流通的桑黄主要有桑树桑黄(Sanghuangporus sanghuang,SH)、杨树桑黄(Sanghuangporus vaninii,YH)、暴马桑黄(Sanghuangporus baumii,BH)和松树桑黄(Phellinus pini,SSH)等。由于桑黄品种繁多,质量存在差异且质控标准缺乏从而导致市场桑黄质量良莠不齐,因此建立桑黄质量控制标准很有必要[9]
桑黄多糖作为桑黄的主要活性成分,具有免疫调节、降血糖、调血脂、抗炎、抗氧化、抗肿瘤、降血压、抗衰老等药理作用[10-12]。目前,桑黄暂无国家级质控标准[13]。已颁布并实施的省级质量标准中,均将桑黄多糖列为重要质控指标,且多数相关研究表明桑黄多糖是评价不同来源桑黄质量的重要指标[14-15]。由于桑黄多糖结构复杂及相对分子质量多分散性,使得桑黄多糖质量难以控制[16],因此,亟需建立一种简便快速,准确性好,特异性高的方法对桑黄多糖进行定性定量分析。本研究基于高效凝胶色谱法(HPGFC-RID法)、红外光谱法(FT-IR法)和HPLC-UV糖谱法,建立桑黄多糖多重指纹图谱,对29批桑黄药材中桑黄多糖的重均分子量、特征吸收官能团和单糖组成进行比较,明确不同品种桑黄多糖差异,以期填补桑黄多糖标准化研究的不足,为桑黄质量控制与合理开发提供参考。

对照品D-无水葡萄糖(批号110833-201908,纯度以99.8%计),购自中国食品药品检定研究院;对照品D-甘露糖(批号G1072059,纯度>98%)、D-木糖(批号G1152942,纯度>98%),购自Dr. Ehrenstorfer公司;对照品L-鼠李糖(批号FS1607340,纯度以98%计),购自广州硕谱生物科技有限公司;对照品D(+)岩藻糖(批号D06HB201374,纯度≥98%)、葡聚糖(平均分子质量分别为2 000、20 000、40 000、100 000、200 000、500 000),购自上海源叶生物科技有限公司。溴化钾为光谱纯,购自上海麦克林生化科技有限公司;95%乙醇、无水乙醇、三氯甲烷、正丁醇、乙酸、乙酸钠、浓盐酸、氢氧化钠、1-苯基-3-甲基-5-吡唑啉酮(PMP)、乙酸铵均为分析纯,购自国药集团化学试剂有限公司。
本研究于市场及原产地共收集桑黄样品29批,包括桑树桑黄(SH)8批、杨树桑黄(YH)15批、暴马桑黄(BH)3批、松木层孔菌(松树桑黄)(SSH)3批,经安徽省食品药品检验研究院张亚中主任中药师鉴定分别为桑树桑黄Sanghuangporus sanghuang(Sheng H. Wu,T. Hatt & Y. C. Dai)Sheng H. Wu、杨树桑黄(Sanghuangporus vaninii Ljub.)L.W. Zhou &Y. C. Dai、暴马桑黄Sanghuangporus baumii(Pilát)L. W. Zhou &Y.C.Dai及松树桑黄Phellinus pini(Thore: Fr.)A. Ames的干燥子实体。样品产地信息见表1,性状图见图1。SH外观为耳形或马蹄形,颜色鲜黄,比重轻;YH外观呈球形或椭圆形,颜色棕褐色或灰黄色,比重轻;BH年轮有明显角度,有扎手感,整体饱和度低;SSH呈宽马蹄形,大小中等,有独特的松香味,桑黄的质量可能因其品种不同存在差异。
取样品适量,粉碎过4号筛,烘干,置于烧杯中,加入4倍体积95%乙醇浸提,过夜,3 500 r·min-1离心5 min得滤渣,重复1次,滤渣置37 ℃烘箱中烘干。取滤渣5 g,置圆底烧瓶中,加水100 mL,加热回流提取2 h,冷却,3 500 r·min-1离心5 min,取上清液,重复1次。合并上清液,加热浓缩至20 mL,冷却至室温,加无水乙醇至醇浓度为80%,于4℃下静置12 h,进行沉淀。3 500 r·min-1离心5 min,收集沉淀物,复溶到20 mL纯水中,得粗多糖溶液,备用。
在粗多糖溶液中,按体积比5:1加入Sevage试剂[三氯甲烷-正丁醇(5:1)],充分振摇。3 500 r·min-1离心5 min后收集上层糖液,重复上述步骤,直至出现薄薄的蛋白层。将糖液转移至透析袋中进行透析,透析后糖液置50 mL顶空瓶中,冷冻干燥,得到桑黄粗多糖冻干粉。
采用TSK-GEL®G3000 PWXL色谱柱(7.8 mm×30 cm,7 μm),以20 mmol·L-1醋酸-醋酸钠缓冲液(pH 5.7)为流动相,流速0.5 mL·min-1,进样量15 μL,柱温35 ℃,检测器为示差检测器(RID)。
取桑黄粗多糖冻干粉适量,用流动相配成质量浓度为5 mg·mL-1的供试品溶液。取葡萄糖和葡聚糖对照品(平均分子质量分别为2 000、20 000、40 000、100 000、200 000、500 000)适量,用流动相配制成1 mg·mL-1的对照品溶液。
采用傅里叶变换红外光谱仪,使用OMNIC 5.0软件进行数据采集。称取冻干粉2 mg,用溴化钾压片法进行红外光谱分析,光谱测定范围4 000~400 cm-1,光谱分辨率为4 cm-1,扫描16次。
取桑黄粗多糖冻干粉5 mg,精密称量,置10 mL安瓿瓶中,加入纯水1 mL和3 mol·L-1盐酸(取浓盐酸24.9 mL,加水稀释至100 mL,即得)500 μL。封口后,放置于110 ℃鼓风干燥箱中水解1 h,取出,冷却至室温,用3 mol·L-1氢氧化钠溶液(取氢氧化钠12 g,加水溶解并稀释至100 mL,即得)调节pH至中性。
取甘露糖、鼠李糖、葡萄糖、木糖、岩藻糖各对照品适量,精密称量,置量瓶中,加纯水制成每1 mL含甘露糖0.60 mg、鼠李糖0.05 mg、葡萄糖2.00 mg、木糖0.60 mg和岩藻糖0.30 mg的混合对照品溶液。
吸取混合对照品溶液及供试品溶液各400 μL,分别置于4 mL EP管中,加入0.3 mo·lL-1氢氧化钠溶液(取氢氧化钠1.2 g,加水溶解成100 mL,即得)400 μL和0.5 mol·L-1的PMP甲醇溶液(取PMP 8.71 g,加甲醇溶解成100 mL,即得)400 μL,混匀,置70 ℃水浴100 min。反应结束后,放至室温,加入0.3 mol·L-1盐酸(取浓盐酸2.5 mL,加水稀释至100 mL,即得)500 μL,混匀。EP管中加入同等体积的三氯甲烷,于漩涡混合仪上混合洗涤,弃去三氯甲烷液,重复6次以除去多余的PMP。最后,将上述衍生化的供试品溶液及对照品溶液过0.45 μm微孔滤膜,备用。
采用Agilent 1260型高效液相色谱仪,色谱柱为Agilent 5 HC-C18(2)(250 mm×4.6 mm,5 μm),以乙腈-0.02 mol·L-1乙酸铵(20:80)为流动相,流速1 mL·min-1,检测波长250 nm,进样量10 μL,检测器为UV检测器,柱温35 ℃。
取桑黄粗多糖冻干粉,按“2.2”项方法制备供试品溶液并按“2.2.1”项条件进样测定,生成的HPGFC-RID叠加图见图2。29批桑黄样品重均分子量糖谱总体上相似,均在9~18 min产生2个主要的色谱峰P1和P2,表明桑黄多糖是由2种不同重均分子量的多糖组成的混合物。用HPGFC-RID采集不同重均分子量对照品色谱图,用GPC软件进行再分析处理,得到分子量校正曲线,相关系数(r)为0.998,加载校正曲线自动积分得到各样品重均分子量,见表2。P1的重均分子量范围为4.068×105~6.007×105,P2的重均分子量范围为1.466×104~3.880×104,2种多糖重均分子量相差1个数量级。29批桑黄多糖重均分子量散点分布图见图3。结果显示,不同品种桑黄的桑黄多糖重均分子量均值存在差异:对于主要色谱峰P1,BH多糖的重均分子量最高,其次依次为SSH、SH和YH多糖;对于主要色谱峰P2,YH多糖的重均分子量最高,其次依次为SSH、SH和BH多糖。实验结果进一步证明,HPGFC糖谱可作为多重指纹图谱之一,根据桑黄多糖重均分子量不同,用于区分不同品种桑黄,为桑黄的质量控制提供帮助。
为表征桑黄多糖化学基团和结构特征,取29批桑黄的粗多糖冻干粉,按“2.3”项条件进行FT-IR光谱测定。YH、SH、BH和SSH多糖的叠加光谱见图4。结果显示,不同官能团振动呈现出不同吸收带。除SH6和BH1多糖外,YH、SH、BH和SSH多糖的种内差异不明显,但3批袋料栽培的YH(YH13、YH14、YH15)多糖与椴木栽培的YH多糖在1 800~900 cm-1范围内的主要吸收峰强度不同,表明桑黄栽培方式不同,其质量也会存在差异。29批桑黄具有相似的红外光谱吸收峰,指纹图谱相似度较高,证明FT-IR糖谱也可以作为提高桑黄多糖质量控制的多重指纹图谱之一。
利用OMNIC 5.0软件进行谱图相加,得到YH、SH、BH和SSH多糖的平均光谱图,见图5。4 000~400 cm-1范围内具有多糖类物质的一般特征,其中3 400 cm-1附近的强吸收峰是羟基伸缩振动吸收峰,2 930 cm-1附近的吸收峰是C-H伸缩振动峰,1 638 cm-1附近的峰是糖分子中羟基弯曲振动吸收峰,1 413 cm-1附近的吸收峰是C-H弯曲振动峰,1 152 cm-1附近为吡喃糖环中醚键(C-O-C)C-O伸缩振动峰,1 100~1 010 cm-1的吸收峰是醇羟基变角振动吸收峰,1 730 cm-1附近没有糖醛酸的特征吸收峰,表明桑黄多糖可能为中性杂多糖[11-12]
取“2.4.3”项下PMP衍生化供试品溶液,按“2.4.4”项条件进样10 μL,记录HPLC色谱图。采用国家药典委员会中药色谱指纹图谱相似度评价系统(2012.1版)进行评价,设定SH1样品图谱为参照图谱,将其余样品色谱峰与SH1样品色谱峰进行自动匹配,生成对照指纹图谱见图6,29批桑黄药材HPLC叠加图见图7
通过中药色谱指纹图谱相似度评价系统,对29批桑黄的桑黄单糖组成指纹图谱中出峰时间基本一致的10个共有峰进行相似度分析,结果见表3。结果显示,所有样品与对照指纹图谱相似度均在0.9以上,且数值越大相似度越高,说明29批桑黄样品单糖组成具有较高相似性。
通过中药色谱指纹图谱相似度评价系统软件对29批桑黄药材单糖组成色谱图进行共有峰标识,根据各峰的峰面积、峰形和分离度,选择10个共有峰,并与对照品色谱图进行比对,最终确定1号峰为甘露糖,3号峰为鼠李糖,6号峰为葡萄糖,8号峰为木糖,10号峰为岩藻糖。
精密吸取“2.4”项下混合对照品溶液、供试品溶液及阴性对照溶液,在“2.4.4”项条件下,测定并记录色谱图,见图8。结果显示,阴性对照溶液无干扰,专属性强。
精密吸取“ 2.4.3”项PMP衍生化后混合对照品溶液适量,梯度稀释2、4、16、64倍,按“2.4.4”项色谱条件进行测定。以待测成分的浓度X为横坐标,待测成分峰面积Y为纵坐标,绘制标准曲线,计算回归方程及相关系数,结果见表4
按“2.4.4”项色谱条件,取用混合对照品溶液连续进行6次进样,记录待测成分峰面积并计算RSD。结果甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖峰面积的RSD(n=6)均<1.6%,表明使用的仪器精密度良好。
取同一份PMP衍生化后供试品溶液(YH1),分别于制样后0、2、4、6、8、10、12、24、36、48 h,按“2.4.4”项色谱条件进行测定,记录峰面积,计算甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖含量和RSD。结果5个成分含量的RSD均<2.0%,表明供试品溶液在48 h内基本稳定。
精密称取适量桑黄粗多糖冻干粉(YH1),平行6份,按“2.4.1”项方法制备供试品溶液,衍生化后,按“2.4.4”项色谱条件进行测定,记录峰面积,计算甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖的含量和RSD。结果甘露糖、鼠李糖、葡萄糖、木糖和岩藻糖的平均含量(n=6)分别为7.99%、0.49%、37.44%、0.88%、5.68%,RSD(n=6)均小于2.4%,表明本方法的重复性良好。
精密称定已知含量的桑黄多糖粗粉(YH1)2.5 mg,平行6份,分别加入对照品溶液甘露糖300 μL、鼠李糖400 μL、葡萄糖450 μL、木糖33 μL、岩藻糖500 μL,按“2.4.1”项方法制备供试溶液,衍生化后,按“2.4.4”项色谱条件进行测定,计算各个待测成分的平均回收率和RSD,结果见表5。根据统计,5个待测成分的平均加样回收率在98.1%~101.9%,RSD均小于2.0%,表明本方法可以获得较为准确的结果。
取29批样品,按“2.4.1”项下方法制备供试品溶液,按“2.4.4”项色谱条件进样测定,用外标法计算样品含量,结果见表6。29批样品中甘露糖含量范围为3.38%~10.25%,鼠李糖含量范围为0.59%~1.13%,葡萄糖含量范围为8.99%~23.45%,木糖含量范围为1.17%~4.89%,岩藻糖含量范围为1.55%~2.67%。
建立桑黄多糖中5个单糖成分含量柱状图,以相对丰度为纵坐标,直观地显示桑黄样品种内种间的差异,见图9。结果显示,29批桑黄多糖中5个单糖组成比例不同,YH(椴木栽培)、SH、BH和SSH多糖中葡萄糖含量最高,甘露糖和岩藻糖次之,木糖和鼠李糖最低,分布趋势近似平行。YH(袋料栽培)中木糖含量最高,岩藻糖、甘露糖、葡萄糖含量次之,鼠李糖含量最低,与其他样品相比具有明显差异。
对5个单糖在29批桑黄样品中的分布进行分析,见图10。4个品种桑黄多糖的5个单糖含量相差不大。但相对而言,甘露糖、鼠李糖和木糖的含量均值在SSH样品中较高,在SH样品中较低;葡萄糖的含量均值在SH和SSH样品中较高,在YH样品中最低;岩藻糖的含量均值在SH和SSH样品中较低,5个单糖的含量分布结果可为区分不同品种的桑黄提供参考。
本研究采用HPGFC-RID方法测定桑黄多糖重均分子量,预先考察不同色谱柱(OHpak SB-804 HQ色谱柱、TSK GEL®G3000 PWXL色谱柱)、不同流动相[水、9 mg·mL-1氯化钠溶液、0.71%硫酸钠溶液、20 mmol·L-1乙酸-乙酸钠缓冲液(pH 5.7)]、不同流速(0.4、0.5、0.6 mL·min-1)及不同柱温(30、35、40 ℃)
将29批桑黄多糖样品中5个单糖成分的含量测定结果导入SIMCA-P 14.0软件进行主成分分析(PCA),以5个单糖成分的含量为变量,建立PCA模型,见图11。结果显示,3批袋料栽培的YH聚为一类,其他桑黄样品没有明显区分,与含量柱状图结果相同。进一步表明,桑黄质量可能因栽培方式不同而存在差异。对色谱峰分离效果和峰形的影响,最终确定最优色谱条件。
在进行样品多糖红外光谱测定时,为获得高质量的透射光谱,样品与溴化钾的压片比例至关重要。样品比例高时,片子不易透明,强吸收峰易超过表尺刻度,出现齐头峰,无法确定其真实峰位;样品比例低时,某些峰会消失,无法得到完整的光谱图,且易产生干涉现象。本研究通过预实验,确定最佳比例为2 mg样品多糖加300 mg KBr研磨压片。通过比较1 000~700 cm-1范围内的特征吸收峰,可以识别不同的吡喃糖和呋喃糖,有助于糖苷键及糖构型的确定[15]
若单糖衍生化后PMP萃取不彻底,HPLC图谱中PMP峰明显,影响图谱效果;若PMP萃取过度,可能会造成单糖衍生产物的损失[17]。实验预先考察了单糖衍生化后残留的PMP萃取次数,结果显示萃取6次后PMP峰已不影响谱图效果。同时,对Welch Ultimate Plus C18(250 mm×4.6 mm,5 μm)色谱柱和Agilent 5 HC-C18(2)(250 mm×4.6 mm,5 μm)色谱柱进行考察,结合分离度、峰形等因素,选择Agilent 5 HC-C18(2)(250 mm×4.6 mm,5 μm)色谱柱用于单糖组成研究。
本研究所建立的HPGFC-RID糖谱、FT-IR糖谱和HPLC-UV糖谱可对不同品种桑黄质量进行有效评价。HPGFC-RID糖谱分析表明,应有2个主要色谱峰,其重均分子量可作为桑黄多糖质量控制的参数之一,用于区分不同品种的桑黄;FT-IR糖谱分析表明,桑黄多糖具有相似的红外光谱吸收峰,种内、种间差异不明显,但袋料栽培与椴木栽培的桑黄多糖在1 800~900 cm-1范围内主要吸收峰强度不同;HPLC-UV糖谱分析表明,葡萄糖为桑黄多糖的主要单糖组分,可根据桑黄多糖的单糖含量区分不同品种的桑黄。
不同的栽培方式对桑黄的质量也存在影响,其中,袋料栽培的桑黄、椴木栽培的桑黄和野生桑黄有明显区分。已有研究表明,不同栽培基质会导致袋料栽培桑黄子实体成分的差异,包括多糖含量及粗多糖单糖组成,从而影响其有效成分活性[18]。此结果提示,尽管野生桑黄资源日渐匮乏,各地开始推广栽培桑黄,但仍需要对栽培技术、栽培桑黄的有效成分和药用功效进行更深入的研究。
吴声华等[1]通过研究确定了真正的桑黄种类为桑树桑黄,实验结果证实,目前普遍市售的另外3个桑黄品种,其多糖类成分的理化性质及单糖组成与桑树桑黄具有很高的相似性。从有效成分多糖的角度分析,收集的4种桑黄均可作为中药桑黄使用,本研究同时填补了桑黄多糖标准化研究的不足,为桑黄质量标准的建立奠定了基础。
